The triple-oxygen isotope (17O/16O, 18O/16O) measurement of oxygen-bearing species represents one of the most robust tools to directly trace oxygen cycling in the environment. One particularly consequential application of this isotope system is the analysis of dissolved oxygen (O2) in aquatic environments to determine gross oxygen production. This approach assumes that photosynthesis, microbial respiration, and gas exchange are the main drivers of dissolved O2 isotope compositions, and that each process is described by predictable, consistent triple-oxygen isotope effects. However, there currently exists large disagreement in the literature on the triple-oxygen isotope effect of respiration, which carries major implications for global primary productivity estimates. Recent work has additionally highlighted the ubiquitous production of extracellular reactive oxygen species (ROS) such as superoxide (O2.-) and hydrogen peroxide (H2O2) by microorganisms; this flux may be responsible for as much as 20% of net oxygen utilization in the ocean. To examine the influence of ROS-mediated O2 recycling on the oxygen utilization isotope effect, we measured the triple-oxygen isotope fractionations and mass laws of superoxide dismutase, catalase, and iron-mediated H2O2 degradation. We incorporate these constraints into an oxygen isotope flux model to explore the influence of ROS-mediated oxygen cycling on “respiration” isotope effects in previous studies. We find that ROS-mediated oxygen cycling can reconcile the previously reported range of triple-oxygen isotope fractionation factors and that typical marine isotope effects are broadly consistent with independent estimates of superoxide-mediated oxygen loss. Lastly, we determine that O2.- production likely follows non-canonical triple-oxygen isotope mass laws (⩽0.510), indicating that this process may exert significant leverage on dissolved O2 isotope compositions and thus primary productivity estimates. Nociceptive input diminishes recovery and increases lesion area after a spinal cord injury (SCI). Recent work has linked these effects to the expansion of hemorrhage at the site of injury. The current paper examines whether these adverse effects are linked to a pain-induced rise in blood pressure and/or flow. Male rats with a low-thoracic spinal cord injury were treated with noxious input [electrical stimulation (shock) or capsaicin] soon after injury. Locomotor recovery and blood pressure were assessed throughout. Tissues were collected three hours, 24 hours, or 21 days later. Both electrical stimulation and capsaicin undermined locomotor function and increased the area of hemorrhage. Changes in blood pressure/flow varied depending on type of noxious input, with only shock producing changes in blood pressure. Providing behavioral control over the termination of noxious stimulation attenuated the rise in blood pressure and hemorrhage. Pretreatment with the alpha-1 adrenergic receptor inverse agonist, prazosin, reduced the stimulation-induced rise in blood pressure and hemorrhage. Prazosin also attenuated the adverse effect noxious stimulation has on long-term recovery. Administration of the adrenergic agonist, norepinephrine, a day after injury induced an increase in blood pressure and disrupted locomotor function, but had little effect on hemorrhage. Further, inducing a rise in blood pressure/flow using norepinephrine undermined long-term recovery and increased tissue loss. Mediational analyses suggest that the pain-induced rise in blood flow may foster hemorrhage after SCI. Increased blood pressure appears to act through an independent process to adversely affect locomotor performance, tissue sparing, and long-term recovery.

The biogeochemical fluxes that cycle oxygen (O2) play a critical role in regulating Earth’s climate and habitability. Triple-oxygen isotope (TOI) compositions of marine dissolved O2 are considered a robust tool for tracing oxygen cycling and quantifying gross photosynthetic O2 production. This method assumes that photosynthesis, microbial respiration, and gas exchange with the atmosphere are the primary influences on dissolved O2 content, and that they have predictable, fixed isotope effects. Despite its widespread use, there are major elements of this approach that remain uncharacterized, including the TOI dynamics of respiration by marine heterotrophic bacteria and abiotic O2 sinks such as the photochemical oxidation of dissolved organic carbon (DOC). Here, we report the TOI fractionation for O2 utilization by two model marine heterotrophs and by abiotic photo-oxidation of representative terrestrial and coastal marine DOC. We demonstrate that TOI slopes associated with these processes span a significant range of the mass-dependent domain (λ = 0.499 to 0.521). A sensitivity analysis reveals that even under moderate productivity and photo-oxidation scenarios, true gross oxygen production may deviate from previous estimates by more than 20% in either direction. The rise of atmospheric oxygen fundamentally changed the chemistry of surficial environments and the nature of Earth’s habitability1. Early atmospheric oxygenation occurred over a protracted period of extreme climatic instability marked by multiple global glaciations2,3, with the initial rise of oxygen concentration to above 10−5 of the present atmospheric level constrained to about 2.43 billion years ago4,5. Subsequent fluctuations in atmospheric oxygen levels have, however, been reported to have occurred until about 2.32 billion years ago4, which represents the estimated timing of irreversible oxygenation of the atmosphere6,7. Here we report a high-resolution reconstruction of atmospheric and local oceanic redox conditions across the final two glaciations of the early Palaeoproterozoic era, as documented by marine sediments from the Transvaal Supergroup, South Africa. Using multiple sulfur isotope and iron–sulfur–carbon systematics, we demonstrate continued oscillations in atmospheric oxygen levels after about 2.32 billion years ago that are linked to major perturbations in ocean redox chemistry and climate. Oxygen levels thus fluctuated across the threshold of 10−5 of the present atmospheric level for about 200 million years, with permanent atmospheric oxygenation finally arriving with the Lomagundi carbon isotope excursion at about 2.22 billion years ago, some 100 million years later than currently estimated. The majority of anaerobic biogeochemical cycling occurs within marine sediments. To understand these processes, quantifying the distribution of active cells and gross metabolic activity is essential. We present an isotope model rooted in thermodynamics to draw quantitative links between cell-specific sulfate reduction rates and active sedimentary cell abundances. This model is calibrated using data from a series of continuous culture experiments with two strains of sulfate reducing bacteria (freshwater bacterium Desulfovibrio vulgaris strain Hildenborough, and marine bacterium Desulfovibrio alaskensis strain G-20) grown on lactate across a range of metabolic rates and ambient sulfate concentrations. We use a combination of experimental sulfate oxygen isotope data and nonlinear regression fitting tools to solve for unknown kinetic, step-specific oxygen isotope effects. This approach enables identification of key isotopic reactions within the metabolic pathway, and defines a new, calibrated framework for understanding oxygen isotope variability in sulfate. This approach is then combined with porewater sulfate/sulfide concentration data and diagenetic modeling to reproduce measured 18O/16O in porewater sulfate. From here, we infer cell-specific sulfate reduction rates and predict abundance of active cells of sulfate reducing bacteria, the result of which is consistent with direct biological measurements.

The triple oxygen isotope composition (Δ'17O) of sulfate minerals is widely used to constrain ancient atmospheric pO2/pCO2 and rates of gross primary production. The utility of this tool is based on a model that sulfate oxygen carries an isotope fingerprint of tropospheric O2 incorporated through oxidative weathering of reduced sulfur minerals, particularly pyrite. Work to date has targeted Proterozoic environments (2.5 billion to 0.542 billion years ago) where large isotope anomalies persist; younger timescale records, which would ground ancient environmental interpretation in what we know from modern Earth, are lacking. Here we present a high-resolution record of the [Formula: see text]O and Δ'17O in marine sulfate for the last 130 million years of Earth history. This record carries a Δ'17O close to 0o, suggesting that the marine sulfate reservoir is under strict control by biogeochemical cycling (namely, microbial sulfate reduction), as these reactions follow mass-dependent fractionation. We identify no discernible contribution from atmospheric oxygen on this timescale. Atmospheric O2 and CO2 levels inform us of the changes in chemical and biological environments, yet the history of atmospheric compositions, and pO2 in particular, is not well-constrained. The triple oxygen isotope (16,17,18O) composition of marine SO 4 2 − has been proposed to directly record the ratio pO2/pCO2 in the contemporaneous atmosphere. To resolve this atmospheric signal, both a precise measurement of the 17O composition of sulfate and a model with which to interpret the measurement are needed. Here we present precise measurements of the triple oxygen isotope composition of modern marine sulfate and then forward a novel sulfur cycle model that deconvolves the potential atmospheric and microbial inputs to this signal. Our interpretation of marine sulfate oxygen isotope composition provides a framework for calculating atmospheric composition, relative rates of biogeochemical activity, and can be applied to geologic records of marine sulfate to constrain the pO2/pCO2 ratio over time.

Measurements of triple oxygen isotope ratios in barite horizons within post-Marinoan cap carbonates have provided some of the most compelling evidence that the Marinoan glaciation was a Snowball Earth event. However, the origin of these barite horizons remains unresolved. To constrain the Ba sources, and thus formation mechanisms of these horizons, we analyzed the Ba isotope composition of post-Marinoan barite deposits from Northwest Canada, Northern Norway, Brazil and South China. We augment these analyses with a Ba isotope survey of almost 100 modern and ancient additional barite measurements, including samples from pelagic (or ‘marine’), hydrothermal, terrestrial, Proterozoic stratiform and cold seep environments. Unlike modern cold seep or terrestrial barites, we find that globally-distributed post-Marinoan barites exhibit a relatively narrow isotopic range, suggesting a well-mixed, effectively limitless Ba source. Moreover, post-Marinoan deposits exhibit a similar mean Ba isotope composition to modern marine barites, which we interpret as evidence of a marine Ba source. Considered alongside existing geochemical, geological, and new Ba isotope survey data, we conclude that Ba in barite horizons was sourced from a well-mixed, Ba-replete but SO4-poor reservoir that accumulated during the Marinoan Snowball Earth interval. This deep Ba reservoir was then transported upward—either by ocean circulation or dolomitization of underlying cap carbonates—and was brought into contact with continental weathering-derived sulfate in a post-glacial meltwater surface layer. Thus, in addition to providing a plausible mechanism for generating globally-synchronous deposition of post-Marinoan barite horizons that reconciles all existing geochemical and geological data, our results demonstrate the utility of Ba isotopes to interrogate the origin of enigmatic barite deposits throughout the sedimentary record.

The reconstruction of modern and paleo-sulfur cycling relies on understanding the long-term relative contribution of its main actors; these include microbial sulfate reduction (MSR) and microbial sulfur disproportionation (MSD). However, a unifying theory is lacking for how MSR and MSD, with the same enzyme machinery and intimately linked evolutionary histories, perform two drastically different metabolisms. Here, we aim at shedding some light on the distribution, diversity, and evolutionary histories of MSR and MSD, with a focus on the Desulfobulbales as a test case. The Desulfobulbales is a diverse and widespread order of bacteria in the Desulfobacterota (formerly Deltaproteobacteria) phylum primarily composed of sulfate reducing bacteria. Recent culture- and sequence-based approaches have revealed an expanded diversity of organisms and metabolisms within this clade, including the presence of obligate and facultative sulfur disproportionators. Here, we present draft genomes of previously unsequenced species of Desulfobulbales, substantially expanding the available genomic diversity of this clade. We leverage this expanded genomic sampling to perform phylogenetic analyses, revealing an evolutionary history defined by vertical inheritance of sulfur metabolism genes with numerous convergent instances of transition from sulfate reduction to sulfur disproportionation.

The ability of aerobic microorganisms to regulate internal and external concentrations of the reactive oxygen species (ROS) superoxide directly influences the health and viability of cells. Superoxide dismutases (SODs) are the primary regulatory enzymes that are used by microorganisms to degrade superoxide. SOD is not one, but three separate, non-homologous enzymes that perform the same function. Thus, the evolutionary history of genes encoding for different SOD enzymes is one of convergent evolution, which reflects environmental selection brought about by an oxygenated atmosphere, changes in metal availability, and opportunistic horizontal gene transfer (HGT). In this study, we examine the phylogenetic history of the protein sequence encoding for the nickel-binding metalloform of the SOD enzyme (SodN). The genomic potential to produce SodN is widespread among bacteria, including Actinobacteriota (Actinobacteria), Chloroflexota (Chloroflexi), Cyanobacteria, Proteobacteria, Patescibacteria, and others. The gene is also present in many archaea, with Thermoplasmatota and Nanoarchaeota representing the vast majority of archaeal sodN diversity. A comparison of organismal and SodN protein phylogenetic trees reveals several instances of HGT, including multiple inter-domain transfers of the sodN gene from the bacterial domain to the archaeal domain. Nearly half of the archaeal members with sodN live in the photic zone of the marine water column. The sodN gene is widespread and characterized by apparent vertical gene transfer in some sediment- or soil-associated lineages within the Actinobacteriota and Chloroflexota phyla, suggesting the ancestral sodN likely originated in one of these clades before expanding its taxonomic and biogeographic distribution to additional microbial groups in the surface ocean in response to decreasing iron availability. In addition to decreasing iron quotas, nickel-binding SOD has the added benefit of withstanding high reactant and product ROS concentrations without damaging the enzyme, making it particularly well suited for the modern surface ocean.

Changes in the geological sulfur cycle are inferred from the sulfur isotopic composition of marine barite. The structure of the (34)S/(32)S record from the Mesozoic to present, which includes ∼50- and 100-Ma stepwise increases, has been interpreted as the result of microbial isotope effects or abrupt changes to tectonics and associated pyrite burial. Untangling the physical processes that govern the marine sulfur cycle and associated isotopic change is critical to understanding how climate, atmospheric oxygenation, and marine ecology have coevolved over geologic time. Here we demonstrate that the sulfur outgassing associated with emplacement of large igneous provinces can produce the apparent stepwise jumps in the isotopic record when coupled to long-term changes in burial efficiency. The record of large igneous provinces map onto the required outgassing events in our model, with the two largest steps in the sulfur isotope record coinciding with the emplacement of large igneous provinces into volatile-rich sedimentary basins. This solution provides a quantitative picture of the last 120 My of change in the ocean’s largest oxidant reservoir.

Significance The evolution of macroscopic animals in the latest Proterozoic Eon is associated with many changes in the geochemical environment, but the sequence of cause and effect remains a topic of intense research and debate. In this study, we use two apparently paradoxical observations—that massively phosphorus-rich rocks first appear at this time, and that the median P content of rocks does not change—to argue for a change in internal marine P cycling associated with rising sulfate levels. We argue that this change was self-sustaining, setting in motion a cascade of biogeochemical transformations that led to conditions favorable for major ecological and evolutionary change., The Ediacaran Period (635 to 541 Ma) marks the global transition to a more productive biosphere, evidenced by increased availability of food and oxidants, the appearance of macroscopic animals, significant populations of eukaryotic phytoplankton, and the onset of massive phosphorite deposition. We propose this entire suite of changes results from an increase in the size of the deep-water marine phosphorus reservoir, associated with rising sulfate concentrations and increased remineralization of organic P by sulfate-reducing bacteria. Simple mass balance calculations, constrained by modern anoxic basins, suggest that deep-water phosphate concentrations may have increased by an order of magnitude without any increase in the rate of P input from the continents. Strikingly, despite a major shift in phosphorite deposition, a new compilation of the phosphorus content of Neoproterozoic and early Paleozoic shows little secular change in median values, supporting the view that changes in remineralization and not erosional P fluxes were the principal drivers of observed shifts in phosphorite accumulation. The trigger for these changes may have been transient Neoproterozoic weathering events whose biogeochemical consequences were sustained by a set of positive feedbacks, mediated by the oxygen and sulfur cycles, that led to permanent state change in biogeochemical cycling, primary production, and biological diversity by the end of the Ediacaran Period.

Here, we describe the genome of Desulfovibrio sulfodismutans ThAc01, a Desulfobacterota member first isolated from freshwater mud and the first strain reported to be capable of growth via sulfur disproportionation. As such, this genome expands our understanding of the diversity of sulfur-disproportionating microorganisms.

Here, we report the draft genome sequence of Dissulfurirhabdus thermomarina SH388. Improved phylogenetic and taxonomic analysis of this organism using genome-level analyses supports assignment of this organism to a novel family within the phylum Desulfobacterota. Additionally, comparative genomic and phylogenetic analyses contextualize the convergent evolution of sulfur disproportionation and potential extracellular electron transfer in this organism relative to other members of the Desulfobacterota.

Here, we describe the genome of Desulfofundulus thermobenzoicus subsp. thermosyntrophicus DSM 14055, a member of the Clostridiales that is capable of sulfate reduction coupled to the oxidation of propionate, lactate, pyruvate, and H 2 /CO 2 . This genome expands our understanding of microbial sulfate reduction (MSR) in anaerobic methanogenic environments.

Here, we describe the genome sequence of Acidianus ambivalens DSM 3772, an archaeon belonging to the Sulfolobales order that was first isolated from continental solfataric fields. This thermoacidophile was sequenced because it utilizes a unique sulfur disproportionation pathway that enables this metabolism under aerobic conditions, in contrast to obligately anaerobic bacterial sulfur disproportionators.

Recent studies targeting the metabolic, physiological, and biochemical controls of sulfur isotope fractionation in microbial systems have drawn linkages between results from culture experiments and the sulfur isotope signatures observed in natural environments. Several of those studies have used newer techniques to explore the minor isotope (33S and 36S) variability in those systems, and also have attempted to place them in an ecophysiological context. Sparingly few have incorporated this newfound understanding of minor isotope behavior into natural systems (sediment pore waters, water columns) and none of them have refined existing isotope-dependent reaction-transport models to explicitly include 33S. In this study, we construct a three-isotope (32S, 33S, and 34S) reaction-transport model of pore water sulfate for a well-characterized sedimentary system within the California-Mexico Margin (Alfonso Basin). An additional goal is placing recent laboratory culture work into a natural, physical context. The model first reproduces the measured bulk geochemical characteristics of the pore water profiles of [SO42−], [CH4], dissolved inorganic carbon ([DIC]), and [Ca2+]—and predicts bulk (non-isotope-specific but depth-dependent) rates of sulfate reduction. Next, the model uses those depth-dependent bulk rates, in combination with empirically calibrated fractionation factors, to explain the minor isotope characteristics (δ34S and Δ33S values) of the 0 to 40 cm pore water SO42−. The down core, isotopic evolution of pore water sulfate requires a large fractionation associated with sulfate reduction (34eSR = 70 ± 5‰) that appears to be independent of bulk rate, but in line with low temperature thermodynamic predictions. The minor isotope characteristics (33λSR ∼ 0.5130) are also independent of rate and fall within the range expected from microbial calibrations, but differ from minor isotope predictions of thermodynamic equilibrium. The high value of 34eSR raises key questions in relating the physiological state of marine microorganisms relative to their laboratory counterparts, as well as point toward exceedingly low metabolic rates in natural marine sediments.

Iron speciation and trace metal proxies are commonly applied together in efforts to identify anoxic settings marked by the presence of free sulfide (euxinia) or dissolved iron (ferruginous) in the water column. Here, we use a literature compilation from modern localities to provide a new empirical evaluation of coupled Fe speciation and Mo concentrations as a proxy for pore water sulfide accumulation at non-euxinic localities. We also present new Fe speciation, Mo concentration, and S isotope data from the Friends of Anoxic Mud (FOAM) site in Long Island Sound, which is marked by pore water sulfide accumulation of up to 3 mM beneath oxygen-containing bottom waters. For the operationally defined Fe speciation scheme, ‘highly reactive’ Fe (FeHR) is the sum of pyritized Fe (Fepy) and Fe dominantly present in oxide phases that is available to react with pore water sulfide to form pyrite. Observations from FOAM and elsewhere confirm that Fepy/FeHR from non-euxinic sites is a generally reliable indicator of pore fluid redox, particularly the presence of pore water sulfide. Molybdenum (Mo) concentration data for anoxic continental margin sediments underlying oxic waters but with sulfidic pore fluids typically show authigenic Mo enrichments (2–25 ppm) that are elevated relative to the upper crust (1–2 ppm). However, compilations of Mo concentrations comparing sediments with and without sulfidic pore fluids underlying oxic and low oxygen (non-euxinic) water columns expose non-unique ranges for each, exposing false positives and false negatives. False positives are most frequently found in sediments from low oxygen water columns (for example, Peru Margin), where Mo concentration ranges can also overlap with values commonly found in modern euxinic settings. FOAM represents an example of a false negative, where, despite elevated pore water sulfide concentrations and evidence for active Fe and Mn redox cycling in FOAM sediments, sedimentary Mo concentrations show a homogenous vertical profile across 50 cm depth at 1 to 2 ppm. A diagenetic model for Mo provides evidence that muted authigenic enrichments are derived from elevated sedimentation rates. Consideration of a range of additional parameters, most prominently pore water Mo concentration, can replicate the ranges of most sedimentary Mo concentrations observed in modern non-euxinic settings. Together, the modern Mo and Fe data compilations and diagenetic model provide a framework for identifying paleo-pore water sulfide accumulation in ancient settings and linked processes regulating seawater Mo and sulfate concentrations and delivery to sediments. Among other utilities, identifying ancient accumulation of sulfide in pore waters, particularly beneath oxic bottom waters, constrains the likelihood that those settings could have hosted organisms and ecosystems with thiotrophy at their foundations.

Ultramafic-hosted hydrothermal systems – characterized by ongoing serpentinization reactions – exert an important influence on the global sulfur cycle. Extensive water-rock interaction causes elemental exchange between seawater and the oceanic lithosphere, effectively removing sulfate from seawater through both abiogenic and biogenic processes. Here, we use bulk rock multiple sulfur isotope signatures (32S, 33S, 34S) and in situ sulfide analyses together with petrographic observations to track the sulfur cycling processes and the hydrothermal evolution of ancient peridotite-hosted hydrothermal systems. We investigate serpentinized peridotites from the Northern Apennine ophiolite in Italy and the Santa Elena ophiolite in Costa Rica and compare those with the Iberian Margin (Ocean Drilling Program (ODP) Leg 149 and 173) and the 15°20′N Fracture Zone along the Mid-Atlantic Ridge (ODP Leg 209). In situ measurements of sulfides in the Northern Apennine serpentinites preserve a large range in δ34Ssulfide of −33.8 to +13.3‰ with significant heterogeneities within single sulfide grains and depending on mineralogy. Detailed mineralogical investigation and comparison with bulk rock Δ33Ssulfide and in situ δ34Ssulfide data implies a thermal evolution of the system from high temperatures (∼350 °C) that allowed thermochemical sulfate reduction and input of hydrothermal sulfide to lower temperatures (

A global rise in oxygen levels has been proposed to coincide with the Ediacaran to Cambrian radiation of animals, yet the precise timing and nature of this change remains unresolved. One hypothesis is that the ocean/atmosphere system became temporarily well-oxygenated in the earliest Ediacaran, directly following the Marinoan Snowball Earth glaciation (~ 635 Ma). The evidence for oxygenation is based on large enrichments of redox-sensitive trace elements in black shale from South China presumably deposited under a euxinic water column. These enrichments meet or exceed the bulk concentrations of redox-sensitive trace elements found in Phanerozoic shale deposited under euxinic water columns. Here we test the early Ediacaran post-Snowball oxygenation hypothesis with new data from a high-resolution, multi-proxy geochemical and sedimentological study of three stratigraphic sections in the earliest Ediacaran Sheepbed Formation of the Mackenzie and Wernecke Mountains, NW Canada. Iron speciation data from all sections suggest that the local water column was dominantly ferruginous, with a notable exception of probable euxinic conditions recorded in part of one section. Redox-sensitive elements show no appreciable enrichments in the basal Sheepbed Formation, with maximum concentrations of molybdenum, vanadium, uranium and chromium only slightly above world average shale values. The lack of substantial elemental enrichments within the Sheepbed Formation is consistent with results from coeval ferruginous strata in Svalbard. The dominance of local ferruginous conditions in NW Canada cannot alone account for the muted redox-sensitive element enrichments as V, U, and Cr are thought to be enriched under these conditions, and samples deposited under euxinic conditions also lack Phanerozoic-style enrichments. These contrasting results from different localities highlight a need to further investigate the veracity of trace metal enrichments in all localities as representing global redox conditions as the results have important implications for the timing of oxygenation with respect to early animal evolution.

Euxinic conditions, which are defined by the presence of sulfide in the water column, were common in ancient oceans. However, it is not clear how the presence of sulfide in the water column affects the balance between rates of sulfide oxidation and sulfate reduction, which plays a major role in regulating the net redox state of the ocean-atmosphere system. Euxinia could lead to higher rates of sulfide oxidation because sulfide may diffuse more rapidly into the oxic zone in solution than in sediment. Alternatively, sulfide oxidation could be inhibited by low overall availability of suitable oxidants in euxinic settings. Here, we constrain rates of sulfide oxidation versus sulfate reduction in four euxinic water columns in coastal ponds by modeling the evolution of the concentration and sulfur and oxygen isotope compositions of sulfate from post-hurricane, well-oxygenated conditions to modern, euxinic conditions. The results of the one-dimensional, depth-dependent models of water column sulfate geochemistry indicate that the fraction of sulfate reduced that is subsequently reoxidized is low (0.11–0.42) in euxinic systems relative to the modern well-oxygenated ocean (0.75–0.90). This implies that sulfide reoxidation rates are low in euxinic systems because of oxidant limitation and physical transport. Low fractional sulfide reoxidation in euxinic systems has important implications for exploring how oxygen levels in the ocean and atmosphere have changed through Earth history. We use a marine sulfate isotope box model to explore how low reoxidation rates in euxinic systems affect marine sulfate sulfur and oxygen isotope records. Model results indicate that marine sulfate sulfur and oxygen isotope compositions increase during the expansion of euxinia with patterns that are distinct from other isotopic changes to the marine sulfate reservoir. Thus, marine sulfate sulfur and oxygen isotope box models can be applied to ancient isotope records in order to evaluate the expansion of euxinia in ancient oceans because sulfide oxidation rates are low relative to sulfate reduction rates.

Throughout most of the sedimentary record, the marine carbon cycle is interpreted as being in isotopic steady state. This is most commonly inferred via isotopic reconstructions, where two export fluxes (organic carbon and carbonate) are offset by a constant isotopic fractionation of ~25 (termed eorg-carb ). Sedimentary deposits immediately overlying the Marinoan snowball Earth diamictites, however, stray from this prediction. In stratigraphic sections from the Ol Formation (Mongolia) and Sheepbed Formation (Canada), we observe a temporary excursion where the organic matter has anomalously heavy δ13 C and is grossly decoupled from the carbonate δ13 C. This signal may reflect the unique biogeochemical conditions that persisted in the aftermath of snowball Earth. For example, physical oceanographic modeling suggests that a strong density gradient caused the ocean to remain stratified for about 50,000 years after termination of the Marinoan snowball event, during which time the surface ocean and continental weathering consumed the large atmospheric CO2 reservoir. Further, we now better understand how δ13 C records of carbonate can be post-depostionally altered and thus be misleading. In an attempt to explain the observed carbon isotope record, we developed a model that tracks the fluxes and isotopic values of carbon between the surface ocean, deep ocean, and atmosphere. By comparing the model output to the sedimentary data, stratification alone cannot generate the anomalous observed isotopic signal. Reproducing the heavy δ13 C in organic matter requires the progressively diminishing contribution of an additional anomalous source of organic matter. The exact source of this organic matter is unclear.

The modern nitrogen cycle consists of a web of microbially mediated redox transformations. Among the most crucial reactions in this cycle is the oxidation of ammonia to nitrite, an obligately aerobic process performed by a limited number of lineages of bacteria (AOB) and archaea (AOA). As this process has an absolute requirement for O2, the timing of its evolution – especially as it relates to the Great Oxygenation Event ~2.3 billion years ago – remains contested and is pivotal to our understanding of nutrient cycles. To estimate the antiquity of bacterial ammonia oxidation, we performed phylogenetic and molecular clock analyses of AOB. Surprisingly, bacterial ammonia oxidation appears quite young, with crown group clades having originated during Neoproterozoic time (or later) with major radiations occurring during Paleozoic time. These results place the evolution of AOB broadly coincident with the pervasive oxygenation of the deep ocean. The late evolution AOB challenges earlier interpretations of the ancient nitrogen isotope record, predicts a more substantial role for AOA during Precambrian time, and may have implications for understanding of the size and structure of the biogeochemical nitrogen cycle through geologic time.

Pain (nociceptive) input caudal to a spinal contusion injury can undermine long-term recovery and increase tissue loss (secondary injury). Prior work suggests that nociceptive stimulation has this effect because it fosters the breakdown of the blood-spinal cord barrier (BSCB) at the site of injury, allowing blood to infiltrate the tissue. The present study examined whether these effects impact tissue rostral and caudal to the site of injury. In addition, the study evaluated whether cutting communication with the brain, by means of a rostral transection, affects the development of hemorrhage. Eighteen hours after rats received a lower thoracic (T11–12) contusion injury, half underwent a spinal transection at T2. Noxious electrical stimulation (shock) was applied 6 h later. Cellular assays showed that, in non-transected rats, nociceptive stimulation increased hemoglobin content, activated pro-inflammatory cytokines and engaged signals related to cell death at the site of injury. These effects were not observed in transected animals. In the next experiment, the spinal transection was performed at the time of contusion injury. Nociceptive stimulation was applied 24 h later and tissue was sectioned for microscopy. In non-transected rats, nociceptive stimulation increased the area of hemorrhage and this effect was blocked by spinal transection. These findings imply that the adverse effect of noxious stimulation depends upon spared ascending fibers and the activation of rostral (brain) systems. If true, stimulation should induce less hemorrhage after a severe contusion injury that blocks transmission to the brain. To test this, rats were given a mild, moderate, or severe, injury and electrical stimulation was applied 24 h later. Histological analyses of longitudinal sections showed that nociceptive stimulation triggered less hemorrhage after a severe contusion injury. The results suggest that brain-dependent processes drive pain-induced hemorrhage after spinal cord injury (SCI).

The sulfur isotope record provides key insight into the history of Earth's redox conditions. A detailed understanding of the metabolisms driving this cycle, and specifically microbial sulfate reduction (MSR), is crucial for accurate paleoenvironmental reconstructions. This includes a precise knowledge of the step-specific sulfur isotope effects during MSR. In this study, we aim at resolving the cellular-level fractionation factor during dissimilatory sulfite reduction to sulfide within MSR, and use this measured isotope effect as a calibration to enhance our understanding of the biochemistry of sulfite reduction. For this, we merge measured isotope effects associated with dissimilatory sulfite reduction with a quantitative model that explicitly links net fractionation, reaction reversibility, and intracellular metabolite levels. The highly targeted experimental aspect of this study was possible by virtue of the availability of a deletion mutant strain of the model sulfate reducer Desulfovibrio vulgaris (strain Hildenborough), in which the sulfite reduction step is isolated from the rest of the metabolic pathway owing to the absence of its QmoABC complex (Delta Qmo). This deletion disrupts electron flux and prevents the reduction of adenosine phosphosulfate (APS) to sulfite. When grown in open-system steady-state conditions at 10% maximum growth rate in the presence of sulfite and lactate as electron donor, sulfur isotope fractionation factors averaged -15.9 parts per thousand (1 sigma = 0.4), which appeared to be statistically indistinguishable from a pure enzyme study with dissimilatory sulfite reductase. We coupled these measurements with an understanding of step-specific equilibrium and kinetic isotope effects, and furthered our mechanistic understanding of the biochemistry of sulfite uptake and ensuing reduction. Our metabolically informed isotope model identifies flavodoxin as the most likely electron carrier performing the transfer of electrons to dissimilatory sulfite reductase. This is in line with previous work on metabolic strategies adopted by sulfate reducers under different energy regimes, and has implications for our understanding of the plasticity of this metabolic pathway at the center of our interpretation of modern and palaeo-environmental records.

Mercury (Hg) stable isotope signatures are widely used to understand Hg cycling in the environment. Sample preparation methods for determining Hg isotope ratios by CV-MC-ICP-MS vary widely among laboratory facilities and sample types. Here, we present a novel and rapid method for preparing solid samples prior to determining Hg isotope composition. We use a direct Hg analyzer (that measures total Hg) for sample combustion, amalgamation and analysis. During the thermal release of Hg from the amalgamator and following detection, the analyte gas enters a trapping solution consisting of 10% HCl/BrCl (5:1, vol/vol). We find Hg blank values are less than 1% of the Hg introduced during sample analysis, Hg detection is not altered by modifying the system, and more than 90% of the introduced Hg is recovered in the trapping solution. Hg isotope results are statistically indistinguishable from accepted values for previously published certified reference materials and uncertainty of 2σ (0.05–0.12‰) is similar to the solution standard RM8610 (2σ = 0.09‰). This new method allows for solid sample preparation for Hg isotope analysis in under 15 min. It has the additional advantage of minimizing use of sample mass during simultaneous detection and preparation.

A new approach for measuring the triple oxygen isotope composition of sulfate minerals was developed using fluorine gas and an infrared laser to generate O 2 . A correction for the mass-dependent isotope effect observed during barite fluorination was rigorously calibrated. Analyte gas purification was performed via numerous cryofocus steps and the introduction of an in-line gas chromatograph. As with previous methods, our fluorination technique still requires pairing with independent δ 18 O measurements made by TC/EA conversion to carbon monoxide, but leads to higher yields than BrF 5 -based methods. We first calibrated our method against known silicate standards (UWG-2, SCO, NBS-28). Following from this, we report high-precision triple oxygen isotope ratios for international sulfate standards (IAEA-SO-5, IAEA-S O-6, NBS-127), and an internal laboratory standard, JMG. Replicate analyses of JMG yielded a Δ ′ 17 O value of −0.057 ± 0.004‰ (standard error), demonstrating per-meg level precision that approaches instrumental limits. We provide δ 17 O and δ 18 O values that can be translated into any preferred reference frame for comparison with gases, water, or rocks/minerals. Quantification of differences in triple oxygen isotope composition of the standard reference materials further enables application of this measure and approach on environmental and geological materials.
