Abstract Neurotensin-polyplex nanoparticles provide efficient gene transfection of nigral dopaminergic neurons when intracerebrally injected in preclinical trials of Parkinson’s disease because they do not cross the blood–brain barrier (BBB). Therefore, this study aimed to open BBB with focused ultrasound (FUS) on the substantia nigra to attain systemic and intranasal transfections and evaluate its detrimental effect in rats. Systemically injected Evans Blue showed that a two-pulse FUS opened the nigral BBB. Accordingly, 35 μL of neurotensin-polyplex nanoparticles encompassing the green fluorescent protein plasmid (79.6 nm mean size and + 1.3 mV Zeta-potential) caused its expression in tyrosine hydroxylase(+) cells (dopaminergic neurons) of both substantiae nigrae upon delivery via internal carotid artery, retro-orbital venous sinus, or nasal mucosa 30 min after FUS. The intracarotid delivery yielded the highest transgene expression, followed by intranasal and venous administration. However, FUS caused neuroinflammation displayed by infiltrated lymphocytes (positive to cluster of differentiation 45), activated microglia (positive to ionized calcium-binding adaptor molecule 1), neurotoxic A1 astrocytes (positive to glial fibrillary acidic protein and complement component 3), and neurotrophic A2 astrocytes (positive to glial fibrillary acidic protein and S100 calcium-binding protein A10), that ended 15 days after FUS. Dopaminergic neurons and axonal projections decreased but recuperated basal values on day 15 after transfection, correlating with a decrease and recovery of locomotor behavior. In conclusion, FUS caused transient neuroinflammation and reversible neuronal affection but allowed systemic and intranasal transfection of dopaminergic neurons in both substantiae nigrae. Therefore, FUS could advance neurotensin-polyplex nanotechnology to clinical trials for Parkinson’s disease.

[INLINE:1] Parkinsonism by unilateral, intranigral β-sitosterol β-D-glucoside administration in rats is distinguished in that the α-synuclein insult begins unilaterally but spreads bilaterally and increases in severity over time, thus replicating several clinical features of Parkinson's disease, a typical α-synucleinopathy. As Nurr1 represses α-synuclein, we evaluated whether unilateral transfected of rNurr1-V5 transgene via neurotensin-polyplex to the substantia nigra on day 30 after unilateral β-sitosterol β-D-glucoside lesion could affect bilateral neuropathology and sensorimotor deficits on day 30 post-transfection. This study found that rNurr1-V5 expression but not that of the green fluorescent protein (the negative control) reduced β-sitosterol β-D-glucoside-induced neuropathology. Accordingly, a bilateral increase in tyrosine hydroxylase-positive cells and arborization occurred in the substantia nigra and increased tyrosine hydroxylase-positive ramifications in the striatum. In addition, tyrosine hydroxylase-positive cells displayed less senescence marker β-galactosidase and more neuron-cytoskeleton marker βIII-tubulin and brain-derived neurotrophic factor. A significant decrease in activated microglia (positive to ionized calcium-binding adaptor molecule 1) and neurotoxic astrocytes (positive to glial fibrillary acidic protein and complement component 3) and increased neurotrophic astrocytes (positive to glial fibrillary acidic protein and S100 calcium-binding protein A10) also occurred in the substantia nigra. These effects followed the bilateral reduction in α-synuclein aggregates in the nigrostriatal system, improving sensorimotor behavior. Our results show that unilateral rNurr1-V5 transgene expression in nigral dopaminergic neurons mitigates bilateral neurodegeneration (senescence and loss of neuron-cytoskeleton and tyrosine hydroxylase-positive cells), neuroinflammation (activated microglia, neurotoxic astrocytes), α-synuclein aggregation, and sensorimotor deficits. Increased neurotrophic astrocytes and brain-derived neurotrophic factor can mediate the rNurr1-V5 effect, supporting its potential clinical use in the treatment of Parkinson's disease.

La población en el Ecuador desde 1950 ha incrementado su tamaño en más de cuatro veces. En el año 1952 el número de habitantes era de 3,2 millones, en el 2010 se registró 14,5 millones de habitantes, mientras que en al año 2022 la población alcanzó 18 millones de habitantes. Este crecimiento poblacional trae como consecuencia el asentamiento humano o actividades de cualquier tipo, principalmente, en zonas planas con alta susceptibilidad a inundaciones (llanuras aluviales) y/o en laderas con alta susceptibilidad a deslizamientos; amenazas que, entre los años 1970 y 2010, han cobrado 1.800 víctimas y hasta 40 mil viviendas afectadas. Como ejemplo de los desastres ocurridos, se puede señalar el aluvión suscitado en el sector de La Gasca (ciudad de Quito), así como las fuertes inundaciones en las provincias de Guayas, Esmeraldas y Manabí. Por otro lado, la geoinformación temática disponible en el país se encuentra a mediana y pequeña escala, dificultando la toma de decisiones, ya que, por ejemplo, al realizarse el plan de desarrollo y ordenamiento territorial (PDOT) se ha tenido una falsa perspectiva del fenómeno de expansión urbana. Por lo antes mencionado, el Instituto Geográfico Militar (IGM) está ejecutando el proyecto “Determinación de la capacidad de acogida del territorio con fines de desarrollo urbano mediante la generación de geoinformación temática a escala 1:5000”, generando información de 200 cabeceras cantonales (10.267,35 km2); basados, metodológicamente, en el dato espacial para realizar el análisis de las formas del relieve y uso de las tierras, y en datos tomados en campo, principalmente, de suelos. De esta forma, se está generando cartografía y datos geográficos de suelos, así como también de temáticas como geomorfología, capacidad de uso de las tierras, cobertura y uso, densidad poblacional, nivel de instrucción, disponibilidad de servicios básicos y nivel socioeconómico. A través de los cuales –sobre todo biofísica– y teniendo en cuenta la importancia del dato que se transformará en información y posteriormente en conocimiento, se produce temáticas de conocimiento geoespacial: aptitud física constructiva, conflictos de uso y capacidad de acogida (CA). Esta cartografía y datos geográficos son insumos para la formulación y ejecución objetiva de los PDOT y los planes de uso y gestión del suelo (PUGS) a nivel de gobiernos autónomos descentralizados, puesto que podrán efectuar un mejor análisis sobre la distribución y disposición del territorio, identificando zonas idóneas para levantar la infraestructura pública y los planes de vivienda de interés social; actuando de forma oportuna para prevenir y mitigar el impacto o daños ante la presencia de cualquier tipo de amenazas, beneficiando así a aproximadamente 8,7 millones de habitantes.

Recently, the excessive generation of plastic waste and its inadequate management have represented a problem worldwide. Petrochemical plastics, which account for 80% of the world's total plastic use, are the main pollutants. With the growth of the population and its needs, the generation of polystyrene waste increases considerably. Expanded polystyrene (EPS) has a very low recycling rate; it is estimated that, of 125 thousand tons generated annually in Mexico, only 0.1% is recycled. The objective of this project is to obtain a preliminary diagnosis on the generation of EPS waste in the city of Misantla, Ver., which allows its post-consumer use as recycled raw material, under a circular economy approach. The methodological proposal for the development of this research was defined in the following steps: (1) characterization of the participating actors, from the Circular Economy approach; (2) identification of businesses that generate polystyrene as waste; (3) determination of the type of polystyrene generated; (4) estimation of the quantities of waste generated; (5) analysis in comparison with the generation of other types of waste; (6) generation of post-consumer use strategies. According to the 2019 Economic Census, in Misantla the economic sector with the largest number of economic units in the municipality is retail, where it is located, the sale of food in small establishments or street stores, these agents being identified as the main generators of polystyrene waste. A survey was applied to 96 companies, mainly in the food sector. The results of these surveys show that these establishments mainly use disposable products made of expanded polystyrene such as plates and cups, and that, in some cases, the use of disposable polystyrene products such as containers and spoons was identified. With the information collected, it is concluded that the use of disposable products generates approximately 765 kilograms of EPS waste. Unlike plastic waste, misinformation about the recycling of EPS, the lack of strategies for its collection and post-consumer use, as well as the lack of awareness for its recycling, give way to it ending up in final disposal sites, continuing with the trend framed in different investigations that maintain that its final destination is incineration. After the diagnosis, there is a concrete vision of the generation of polystyrene waste and future work is given in the generation and implementation of strategies that allow the use of post-consumer unicel for its return to the value chain.

In this paper, a model-based control scheme for a DC-DC power converter as a battery charger is presented. The proposed DC-DC converter for this task is a high-gain step-down converter based on the switched inductor concept. Thus, the considered topology is able to highly reduced the output voltage on a single stage compared to traditional step-down converters. The proposed control strategy is aimed at fulfilling the requirements of the constant current and constant voltage algorithm widely used for battery charging purposes. Given the structure of the system model, the proposed control scheme results in a multi-loop controller, which is structured by a PI current control loop and a PI voltage control loop. During the constant current stage, the system is required to inject constant current into the battery while the voltage freely increases. Thus, the current control loop maintains the input battery current at a constant reference value. On the other hand, in the constant voltage stage, the battery is regulated to a desired constant value. Hence, in the end, both control loops act on the system to fulfill the objective. An external battery management system is aimed at controlling the transition between both modes, considering the physical thresholds of current and voltage required by the battery to guarantee the successful charging process. Finally, the performance of the proposed control strategy for battery charging is assessed on an experimental setup.

Abstract Objective To describe the clinical characteristics and outcomes in patients with refractory myasthenia gravis (MG) and to determine the effectiveness and side effects of the drugs used for their treatment. Methods This observational retrospective cross‐sectional multicenter study was based on data from the Spanish MG Registry (NMD‐ES). Patients were considered refractory when their MG Foundation of America post‐interventional status (MGFA‐PIS) was unchanged or worse after corticosteroids and two or more other immunosuppressive agents. Clinical and immunologic characteristics of drug‐refractory patients, efficiency and toxicity of drugs used, and outcome (MGFA‐PIS) at end of follow‐up were studied. Results We included 990 patients from 15 hospitals. Eighty‐four patients (68 of 842 anti‐acetylcholine receptor [AChR], 5 of 26 anti‐muscle‐specific tyrosine kinase [MusK], 10 of 120 seronegative, and 1 of 2 double‐seropositive patients) were drug refractory. Overexpression of neurotrophic factors in nigral dopamine neurons is a promising approach to reverse neurodegeneration of the nigrostriatal dopamine system, a hallmark in Parkinson’s disease. The human cerebral dopamine neurotrophic factor (hCDNF) has recently emerged as a strong candidate for Parkinson’s disease therapy. This study shows that hCDNF expression in dopamine neurons using the neurotensin-polyplex nanoparticle system reverses 6-hydroxydopamine-induced morphological, biochemical, and behavioral alterations. Three independent electron microscopy techniques showed that the neurotensin-polyplex nanoparticles containing the hCDNF gene, ranging in size from 20 to 150 nm, enabled the expression of a secretable hCDNF in vitro. Their injection in the substantia nigra compacta on day 21 after the 6-hydroxydopamine lesion resulted in detectable hCDNF in dopamine neurons, whose levels remained constant throughout the study in the substantia nigra compacta and striatum. Compared with the lesioned group, tyrosine hydroxylase-positive (TH+) nigral cell population and TH+ fiber density rose in the substantia nigra compacta and striatum after hCDNF transfection. An increase in βIII-tubulin and growth-associated protein 43 phospho-S41 (GAP43p) followed TH+ cell recovery, as well as dopamine and its catabolite levels. Partial reversal (80%) of drug-activated circling behavior and full recovery of spontaneous motor and non-motor behavior were achieved. Brain-derived neurotrophic factor recovery in dopamine neurons that also occurred suggests its participation in the neurotrophic effects. These findings support the potential of nanoparticle-mediated hCDNF gene delivery to develop a disease-modifying treatment against Parkinson’s disease. A frequent problem in geotechnics is soils with inadequate physical–mechanical properties to withstand construction work, incurring cost overruns caused by their engineering improvement. The need to improve the engineering properties of soils is not recent. The most common current alternatives are binders such as cement and lime. The climate change observed in recent decades and the uncontrolled emission of greenhouse gases have motivated geotechnical and geoenvironmental researchers to seek mechanisms for soil reinforcement from a more sustainable and environmentally friendly approach by proposing the use of recycled and waste materials. An alternative is natural fibers, which can be obtained as waste from many agro-industrial processes, due to their high availability and low cost. Sawdust, as a by-product of wood processing, has a rough texture that can generate high friction between the fiber and the matrix of the soils, leading to a significant increase in its shearing strength and bearing capacity. This concept of improving the properties of soils using natural fibers distributed randomly is inspired by the natural phenomenon of grass and/or plants that, when growing on a slope, can effectively stabilize the said slope.

Whether neuroinflammation leads to dopaminergic nigrostriatal system neurodegeneration is controversial. We addressed this issue by inducing acute neuroinflammation in the substantia nigra (SN) with a single local administration (5 µg/2 µL saline solution) of lipopolysaccharide (LPS). Neuroinflammatory variables were assessed from 48 h to 30 days after the injury by immunostaining for activated microglia (Iba-1 +), neurotoxic A1 astrocytes (C3 + and GFAP +), and active caspase-1. We also evaluated NLRP3 activation and Il-1β levels by western blot and mitochondrial complex I (CI) activity. Fever and sickness behavior was assessed for 24 h, and motor behavior deficits were followed up until day 30. On this day, we evaluated the cellular senescence marker β-galactosidase (β-Gal) in the SN and tyrosine hydroxylase (TH) in the SN and striatum. After LPS injection, Iba-1 (+), C3 (+), and S100A10 (+) cells were maximally present at 48 h and reached basal levels on day 30. NLRP3 activation occurred at 24 h and was followed by a rise of active caspase-1 (+), Il-1β, and decreased mitochondrial CI activity until 48 h. A significant loss of nigral TH (+) cells and striatal terminals was associated with motor deficits on day 30. The remaining TH (+) cells were β-Gal (+), suggesting senescent dopaminergic neurons. All the histopathological changes also appeared on the contralateral side. Our results show that unilaterally LPS-induced neuroinflammation can cause bilateral neurodegeneration of the nigrostriatal dopaminergic system and are relevant for understanding Parkinson’s disease (PD) neuropathology.

Abstract The spreading and accumulation of α-synuclein and dopaminergic neurodegeneration, two hallmarks of Parkinson’s disease (PD), have been faithfully reproduced in rodent brains by chronic, oral administration of β-sitosterol β-D-glucoside (BSSG). We investigated whether a single injection of BSSG (6 μg BSSG/μL DMSO) in the left substantia nigra of Wistar rats causes the same effects. Mock DMSO injections and untreated rats formed control groups. We performed immunostainings against the pathological α-synuclein, the dopaminergic marker tyrosine hydroxylase (TH), the neuroskeleton marker β-III tubulin, the neurotensin receptor type 1 (NTSR1) as non-dopaminergic phenotype marker and Fluro-Jade C (F-J C) label for neurodegeneration. Using β-galactosidase (β-Gal) assay and active caspase-3 immunostaining, we assessed cell death mechanisms. Golgi-Cox staining was used to measure the density and types of dendritic spines of striatal medium spiny neurons. Motor and non-motor alterations were also evaluated. The study period comprised 15 to 120 days after the lesion. In the injured substantia nigra, BSSG caused a progressive α-synuclein aggregation and dopaminergic neurodegeneration caused by senescence and apoptosis. The α-synuclein immunoreactivity was also present within microglia cells. Decreased density of dopaminergic fibers and dendritic spines also occurred in the striatum. Remarkably, all the histopathological changes also appeared on the contralateral nigrostriatal system, and α-synuclein aggregates were present in other brain regions. Motor and non-motor behavioral alterations were progressive. Our data show that the stereotaxic BSSG administration reproduces PD α-synucleinopathy phenotype in the rat. This approach will aid in identifying the spread mechanism of α-synuclein pathology and validate anti-synucleinopathy therapies.

Amyotrophic lateral sclerosis (ALS) and frontotemporal lobar degeneration (FTLD) lie at opposing ends of a clinical, genetic, and neuropathological continuum. In the last decade, it has become clear that cognitive and behavioral changes in patients with ALS are more frequent than previously recognized. Significantly, these non-motor features can impact the diagnosis, prognosis, and management of ALS. Partially overlapping neuropathological staging systems have been proposed to describe the distribution of TAR DNA-binding protein 43 (TDP-43) aggregates outside the corticospinal tract. However, the relationship between TDP-43 inclusions and neurodegeneration is not absolute and other pathophysiological processes, such as neuroinflammation (with a prominent role of microglia), cortical hyperexcitability, and synaptic dysfunction also play a central role in ALS pathophysiology. In the last decade, imaging and biofluid biomarker studies have revealed important insights into the pathophysiological underpinnings of extra-motor neurodegeneration in the ALS-FTLD continuum. In this review, we first summarize the clinical and pathophysiological correlates of extra-motor neurodegeneration in ALS. Next, we discuss the diagnostic and prognostic value of biomarkers in ALS and their potential to characterize extra-motor neurodegeneration. Finally, we debate about how biomarkers could improve the diagnosis and classification of ALS. Emerging imaging biomarkers of extra-motor neurodegeneration that enable the monitoring of disease progression are particularly promising. In addition, a growing arsenal of biofluid biomarkers linked to neurodegeneration and neuroinflammation are improving the diagnostic accuracy and identification of patients with a faster progression rate. The development and validation of biomarkers that detect the pathological aggregates of TDP-43 in vivo are notably expected to further elucidate the pathophysiological underpinnings of extra-motor neurodegeneration in ALS. Novel biomarkers tracking the different aspects of ALS pathophysiology are paving the way to precision medicine approaches in the ALS-FTLD continuum. These are essential steps to improve the diagnosis and staging of ALS and the design of clinical trials testing novel disease-modifying treatments.

Tilapia farming is the predominant aquaculture activity, with 4623 aquaculture farms in Mexico alone. It is relevant to apply technological alternatives to mitigate production costs, mainly those associated with supporting energy savings for aeration and water pumping in aquaculture farms. There is limited information confirming the feasibility of implementing photovoltaic systems connected to the grid (On grid-PV) in aquaculture farms. The working hypothesis proposed for the development of the work was that On Grid PV systems in Tilapia aquaculture farms in Mexico are technically feasible, economically viable and environmentally acceptable. Therefore, the objective of this research is to design a grid-connected photovoltaic system for rural Tilapia aquaculture farms in Mexico and analyze it with a feasibility assessment through technical, economic and environmental variables, as part of the link between academia and the productive sector. Methodologically, the On Grid-PV design was carried out in an aquaculture farm in Veracruz, Mexico, as a case study. It was developed in two stages: the field phase (1), where a non-participant observation guide and a survey with open questions were applied to perform the energy diagnosis, and the cabinet phase (2) where the calculation of the economic and environmental variables was carried out with the clean energy management software Retscreen expert, the engineering design was based on the Mexican Official Standard for electrical installations, and Sunny Design 5.22.5 was used to calculate and analyze the electrical parameters of the On Grid PV. The results revealed an investment cost of USD 30,062.61, the cost per KWp was of 1.36 USD/Watt, and the economic indicators were the net present value (USD 41,517.44), internal rate of return (38.2%) and cost–benefit ratio (5.6). Thus, the capital investment is recovered in 4.7 years thanks to the savings obtained by generating 2429 kW/h per month. As for the environment, it is estimated that 11,221 kg of CO2 equivalent would be released into the atmosphere without the On Grid-PV. In conclusion, the hypothesis is accepted and it is confirmed that On Grid-PV installations for Tilapia farms are technically feasible, economically viable and environmentally acceptable; their implementation would imply the possibility for aquaculture farms to produce Tilapia at a lower production cost and minimized environmental impact in terms of energy. It is recommended that aquaculture farmers in Mexico and the world implement this eco-technology that supports the sustainable development of aquaculture.

This research aims to determine whether a specific blend of phytogenic compounds (BPC) supplemented in gestating hyperprolific sow diets can promote prenatal maternal effects in terms of piglet gut function and morphology. Twenty-eight (Landrace × Yorkshire) gilts and sows (parity 0 to 7) were randomly distributed by parity number and body weight into two dietary treatments: unsupplemented Control (CON) (n = 14) or CON diet supplemented with 1 g/kg feed of BPC during gestation (n = 14). The BPC supplementation during gestation of sows downregulated the neonate piglets' jejunal genes involved in oxidation (SOD2) and nutrient transport (SLC16A1/MCT1, SLC11A2/DMT1, and SLC39A/ZIP4), while IFNG and CLDN4 related to immune response and barrier function, respectively, were upregulated (q < 0.10). In addition, the jejunal villus height and the ratio of the villus height to crypt depth tended to increase (p < 0.10), while goblet cell volume density was higher (p < 0.05) in BPC compared to CON. In conclusion, dietary supplementation of BPC in gestating diets for hyperprolific sows influences neonatal histomorphology and expression of genes related to the intestinal function and health.

Objectives: Pompe disease is a rare genetic disease produced by mutations in the GAA gene leading to progressive skeletal and respiratory muscle weakness. T1-weighted magnetic resonance imaging is useful to identify fatty replacement in skeletal muscles of late-onset Pompe disease (LOPD) patients. Previous studies have shown that replacement by fat correlates with worse results of muscle function tests. Our aim was to investigate if fat replacement of muscles involved in the ventilation process correlated with results of the spirometry and predicted respiratory muscle impairment in LOPD patients over time.Materials and Methods: We studied a cohort of 36 LOPD patients followed up annually in our center for a period of 4 years. We quantified muscle fat replacement using Mercuri score of the thoracic paraspinal and abdominal muscles and the pillars of the diaphragm. We correlated the combined Mercuri scores of these areas with spirometry results and the need of respiratory support.Results: We found a statistically significant correlation (Spearman test, p < 0.05; coefficient of correlation > 0.6) between forced vital capacity seated and lying and fat fraction score of all muscle groups studied. The group of patients who needed respiratory support had higher fat fraction scores than patients not requiring ventilatory support. Higher fat replacement in these areas correlated with worse progression in spirometry values over time.Conclusions: Fat replacement of paraspinal, abdominal, and trunk muscles correlates with results of spirometry and is able to predict worsening in respiratory muscle function tests that could lead to an emerging ventilatory dysfunction. Therefore, the identification of fat replacement in these muscle groups should lead to a closer monitorization of patients. Radiologic evaluation of diaphragm pillars in T1-weighted imaging axial sequences could also be helpful to predict respiratory insufficiency.

Objectives: Magnetization transfer (MT) imaging exploits the interaction between bulk water protons and protons contained in macromolecules to induce signal changes through a special radiofrequency pulse. MT detects muscle damage in patients with neuromuscular conditions, such as limb-girdle muscular dystrophies or Charcot-Marie-Tooth disease, which are characterized by progressive fiber loss and replacement by fatty tissue. In Pompe disease, in which there is, in addition, an accumulation of glycogen inside the muscle fibers, MT has not been tested yet. Our aim is to estimate MT ratio (MTR) in the skeletal muscle of these patients and correlate it with intramuscular fat fraction (FF) and results of muscle function tests.Methods: We obtained two-point axial Dixon and Dixon-MT sequences of the right thigh on a 1.5 Teslas MRI scanner in 60 individuals, including 29 late onset Pompe disease patients, 2 patients with McArdle disease, and 29 age and sex matched healthy controls. FF and MTR were estimated. Muscle function using several muscle function tests, including quantification of muscle strength, timed test quality of life scales, conventional spirometry obtaining forced vital capacity while sitting and in the supine position, were assessed in all patients.Results: MTR was significantly lower in Pompe patients compared with controls (45.5 ± 8.5 vs. 51.7 ± 2.3, Student T-test, p < 0.05). There was a negative correlation between the MTR and FF muscles studied (correlation coefficient: −0.65, Spearman test: p < 0.05). MTR correlated with most of the muscle function test results. We analyzed if there was any difference in MTR values between Pompe patients and healthy controls in those muscles that did not have an increase in fat, a measure that could be related to the presence of glycogen in skeletal muscles, but we did not identify significant differences except in the adductor magnus muscle (48.4 ± 3.6 in Pompe vs. 51 ± 1.3 in healthy controls, Student T-test = 0.023).Conclusions: MTR is a sensitive tool to identify muscle loss in patients with Pompe disease and shows a good correlation with muscle function tests. Therefore, the MT technique can be useful in monitoring muscle degeneration in Pompe disease in clinical trials or natural history studies.

Summary: A lymph node sinus-on-a-chip adhesion microfluidic platform that recapitulates the hydrodynamic microenvironment of the lymph node subcapsular sinus was engineered. This device was used to interrogate the effects of lymph node remodeling on cellular adhesion in fluid flow relevant to lymphatic metastasis. Wall shear stress levels analytically estimated and modeled after quiescent and diseased/inflamed lymph nodes were experimentally recapitulated using a flow-based microfluidic perfusion system to assess the effects of physiological flow fields on human metastatic cancer cell adhesion. Results suggest that both altered fluid flow profiles and presentation of adhesive ligands, which are predicted to manifest within the lymph node subcapsular sinus as a result of inflammation-induced remodeling, and the presence of lymph-borne monocytic cells may synergistically contribute to the dynamic extent of cell adhesion in flow relevant to lymph node invasion by cancer and monocytic immune cells during lymphatic metastasis.

Chronic consumption of β-sitosterol-β-D-glucoside (BSSG), a neurotoxin contained in cycad seeds, leads to Parkinson’s disease in humans and rodents. Here, we explored whether a single intranigral administration of BSSG triggers neuroinflammation and neurotoxic A1 reactive astrocytes besides dopaminergic neurodegeneration. We injected 6 μg BSSG/1 μL DMSO or vehicle into the left substantia nigra and immunostained with antibodies against tyrosine hydroxylase (TH) together with markers of microglia (OX42), astrocytes (GFAP, S100β, C3), and leukocytes (CD45). We also measured nitric oxide (NO), lipid peroxidation (LPX), and proinflammatory cytokines (TNF-α, IL-1β, IL-6). The Evans blue assay was used to explore the blood-brain barrier (BBB) permeability. We found that BSSG activates NO production on days 15 and 30 and LPX on day 120. Throughout the study, high levels of TNF-α were present in BSSG-treated animals, whereas IL-1β was induced until day 60 and IL-6 until day 30. Immunoreactivity of activated microglia (899.0±80.20%) and reactive astrocytes (651.50±11.28%) progressively increased until day 30 and then decreased to remain 251.2±48.8% (microglia) and 91.02±39.8 (astrocytes) higher over controls on day 120. C3(+) cells were also GFAP and S100β immunoreactive, showing they were neurotoxic A1 reactive astrocytes. BBB remained permeable until day 15 when immune cell infiltration was maximum. TH immunoreactivity progressively declined, reaching 83.6±1.8% reduction on day 120. Our data show that BSSG acute administration causes chronic neuroinflammation mediated by activated microglia, neurotoxic A1 reactive astrocytes, and infiltrated immune cells. The severe neuroinflammation might trigger Parkinson’s disease in BSSG intoxication.

El presente trabajo analiza las características del aprendizaje basado en investigación (ABI), con el objetivo de abordar cómo la articulación de la docencia con la investigación puede ser un factor para el fortalecimiento de los programas educativos en educación superior, concretamente en la Universidad de Quintana Roo, Unidad Académica Playa del Carmen (UAPC). Para ello, con un enfoque mixto de investigación se efectuó la revisión de documentos significativos y bibliografía especializada como técnicas cualitativas iniciales. El análisis de esta fase contribuyó al diseño de un cuestionario estructurado para llevar a cabo una encuesta en la parte cuantitativa, que permitió conocer la disposición y los obstáculos para implementar el ABI. Entre los principales hallazgos destacan el interés del profesorado por fortalecer sus capacidades investigativas, capacitarse en conocimientos teóricos sobre el ABI e incluir al alumnado en las investigaciones, para poder con todo ello hacer frente a la necesidad de formar profesionistas con creatividad; con capacidad de analizar, pensar críticamente, escribir y expresarse con eficacia; resolver problemas complejos, y sintetizar, entre otros retos de la realidad educativa del siglo XXI.

In the present research, two studies were performed to determine the effects of specific botanical compounds (BCs) on the innate feed preference and feed intake of piglets, as follows: Exp. 1 studied the innate feed preferences of post-weaning piglets using a double-choice feeding test. A total of 828 weaned piglets were distributed into 36 pens (23 pigs/pen) and assigned to three dietary pair choice feeding options (n = 12): unsupplemented prestarter diets (reference) versus reference plus D-limonene, trans-anethole, or eucalyptol. Piglets showed a preference for diets with D-limonene (53.8%) and trans-anethole (54.5%), and an aversion to eucalyptol (41.6%) (p < 0.05). Exp. 2 studied whether the prenatal and perinatal exposure to D-limonene, trans-anethole, and eucalyptol influences the feed intake and growth of newly-weaned piglets. Twenty-eight gestating and lactating sows were distributed into two dietary treatments (n = 14): unsupplemented Control diets or Control plus a blend of BCs (BBC; containing D-limonene, trans-anethole, and eucalyptol). D-limonene, trans-anethole, and eucalyptol were transferred into the placental fluid, and D-limonene and trans-anethole into the milk (p < 0.05). Furthermore, weanling piglets (n = 200; Control) and (n = 203; BBC) received the same treatment as their mothers in prestarter diets. The early response after weaning showed that piglets’ post-weaning BW gain was higher in the Control (p < 0.05) group than in those exposed to BBC. In conclusion, prenatal exposure to preferred D-limonene and trans-anethole, or familiarity to eucalyptol did not help to overcome the innate aversion to eucalyptol and its negative effect on weanling piglets’ BW.

Esta investigación utiliza el instrumento cualitativo de la Teoría del Comportamiento Planificado (TPB) para obtener las creencias hacia la implementación pedagógica de un dispositivo de interfaz humana similar a las pizarras digitales interactivas. El instrumento se aplicó a estudiantes de pedagogía que participaron de un curso sobre el uso técnico y pedagógico de pizarras digitales. El conjunto de respuestas se sometió a un análisis de contenido generando categorías de creencias en el ámbito de los factores personales, contextuales y de control, que estarían mediando en su implementación. Posteriormente, se utilizaron criterios cuantitativos para seleccionar aquellas creencias que formarán parte del instrumento metodológico principal de TPB. En total dieciséis creencias fueron seleccionadas según un criterio que contempla tanto la cobertura de sujetos como de cantidad de creencias y que finalmente conforman el conjunto modal. Estas creencias son utilizadas para diseñar una estructura de capacitación dirigida a docentes en formación.

Energy production in constructed wetlands is a little-known field, as is the operation of vertical partially saturated constructed wetlands (VPS-CWs) that promote both aerobic and anaerobic microbial interactions. By doing so, bacterial degradation is increased, becoming the main mechanism of pollutant removal in constructed wetlands (CWs). For the first time, the generation of bioelectricity, together with the production of ornamental plants in vertical partially saturated constructed wetlands during the treatment of domestic wastewater, was evaluated. Six VPS-CW systems functioned as bioelectricity generators, where the systems were filled with red volcanic gravel and activated carbon as anode and cathode. Three systems were planted with Zantedeschia aethiopica and three with Canna hybrids plants. The development was measured through mother plants and shoots produced every 60 days. The input and output of each VPS-CW was monitored using control parameters such as BOD5, phosphates (P-PO4), and total Kjeldahl nitrogen (TKN). Bioelectricity, power, voltage, and current measurements were performed every 15 days for a period of 7 months. It was found that the VPS-CWs used as biobatteries in combination with the use of domestic wastewater as a substrate improved the development of the two evaluated plant species and stimulated growth and germination of new shoots. No significant differences were found between the different treatments (p ≤ 0.05). Likewise, an average efficient removal of BOD5 (98%) for both systems without statistical differences was observed (p ≤ 0.05), but for TKN and P-PO4, significant differences (p ≤ 0.05) were found between systems planted with Z. aethiopica (TKN: 65%; P-PO4: 20%) and Canna hybrids (TKN: 69%; P-PO4: 27%). This method of water treatment and bioelectricity production with Canna hybrids was an efficient system that generated a great electric current (140 mA/m2), voltage (750 mV), and electric power (15 mW/m2), compared with those observed in systems with Z. aethiopica (60 mA/m2, 500 mV, 9 mA/m2).

El objetivo del estudio fue analizar las creencias que afectan la implementación de recursos infocomunicacionales en futuros profesores, mediante la estructura conceptual y el instrumento de evaluación que ofrece la Teoría del Comportamiento Planificado propuesta por Icek Ajzen. Se consideraron como variables: Actitud hacia la conducta, Norma Subjetiva y Control Conductual Percibido, así como las medidas indirectas basadas en las creencias. El estudio se realizó con una muestra de 115 estudiantes universitarios de la Universidad Metropolitana de Ciencias de la Educación, Chile, con edades entre los 21 y los 30 años y mayoritariamente mujeres (73.9 %). Se realizó un análisis de regresión múltiple siendo intención conductual la variable dependiente y los predictores las medidas directas de la teoría. Los resultados revelan un modelo significativo en el que las medidas directas de la teoría dan cuenta del 16% de la varianza explicada.

La investigación validó modelos geo-espaciales de carbón orgánico y de densidad aparente en la subcuenca del río Guayllabamba, obtenidos de diferentes métodos de interpolación, con base a información de 462 perfiles, para calcular la fijación de carbono. Determinándose, a través del coeficiente de determinación (r2), análisis de varianza (prueba F) y pruebas de significación (Tukey), que el método superficie móvil, fue el más adecuado, con el 90 % de confiabilidad, tanto para carbón orgánico como para densidad aparente.

Models of Parkinson’s disease with neurotoxins have shown that microglial activation does not evoke a typical inflammatory response in the substantia nigra, questioning whether neuroinflammation leads to neurodegeneration. To address this issue, the archetypal inflammatory stimulus, lipopolysaccharide (LPS), was injected into the rat substantia nigra. LPS induced fever, sickness behavior, and microglial activation (OX42 immunoreactivity), followed by astrocyte activation and leukocyte infiltration (GFAP and CD45 immunoreactivities). During the acute phase of neuroinflammation, pro- and anti-inflammatory cytokines (TNF-α, IL-1β, IL-6, IL-4, and IL-10) responded differentially at mRNA and protein level. Increased NO production and lipid peroxidation occurred at 168 h after LPS injection. At this time, evidence of neurodegeneration could be seen, entailing decreased tyrosine hydroxylase (TH) immunoreactivity, irregular body contour, and prolongation discontinuity of TH+ cells, as well as apparent phagocytosis of TH+ cells by OX42+ cells. Altogether, these results show that LPS evokes a typical inflammatory response in the substantia nigra that is followed by dopaminergic neurodegeneration.

The structural effect of neurturin (NRTN) on the nigrostriatal dopaminergic system in animals remains unknown, although NRTN has been shown to be effective in Parkinson's disease animal models. Herein, we aimed to demonstrate that NRTN overexpression in dopaminergic neurons stimulates both neurite outgrowths in the nigrostriatal pathway and striatal dendritic spines in aging rats with chronic 6-hydroxydopamine (6-OHDA) lesion. At week 12 after lesion, pTracer-mNRTN-His or pGreenLantern-1 plasmids were intranigrally transfected using the NTS-polyplex nanoparticles system. We showed that the transgenic expression in dopaminergic neurons remained until the end of the study (12 weeks). Only animals expressing NRTN-His showed recovery of tyrosine hydroxylase (TH)+ cells (28 ± 2%), their neurites (32 ± 2%) and the neuron-specific cytoskeletal marker β-III-tubulin in the substantia nigra; striatal TH(+) fibers were also recovered (52 ± 3%), when compared to the healthy condition. Neurotensin receptor type 1 levels were also significantly recovered in the substantia nigra and striatum. Dopamine recovery was 70 ± 4% in the striatum and complete in the substantia nigra. The number of dendritic spines of striatal medium spiny neurons was also significantly increased, but the recovery was not complete. Drug-activated circling behavior decreased by 73 ± 2% (methamphetamine) and 89 ± 1% (apomorphine). Similar decrease was observed in the spontaneous motor behavior. Our results demonstrate that NRTN causes presynaptic and postsynaptic restoration of the nigrostriatal dopaminergic system after a 6-OHDA-induced chronic lesion. However, those improvements did not reach the healthy condition, suggesting that NRTN exerts lesser neurotrophic effects than other neurotrophic approaches.

Estudio que indaga las habilidades científicas en un curso introductorio de robótica en la formación de profesores de física, química y matemática. La investigación se estructuró en dos fases: capacitación en aspectos propios de la robótica (electrónica y programación) mediante el uso de los kits Home-Bot® de Parallax® y el lenguaje de programación PBASIC; y una fase de resolución de problemas prácticos de navegación con robots. La recolección de información se realizó mediante una prueba escrita y una lista de cotejo, además de registros audiovisuales. El análisis se llevó a cabo utilizando una distribución de frecuencias de observación de la habilidad y sus medias estadísticas, así como las pruebas de comparación de medidas entre grupos. Al final se realizó una comparación entre datos cuantitativos y cualitativos extraídos mediante el análisis de contenido en los registros en video. Entre los principales resultados se encuentran: que las habilidades testeadas se manifiestan sin diferencia significativa entre estudiantes de las carreras de pedagogía en física, pedagogía en química y pedagogía en matemática; hechos que destacan el potencial de la robótica para desarrollar habilidades de proceso científico en la formación inicial de profesores de ciencias y matemática.
