This chapter presents a brief analytic history of the initial 50 years of the Center for International Education (CIE) at the University of Massachusetts Amherst with the goal of understanding what made it possible and what can be learned from it for the future of Comparative and International Education programs in other universities. The chapter begins with the unusual context in which CIE was created and its commitment to a synergistic linkage between academics and managing funded, development education programs. The discussion then describes CIE’s defining characteristics, the challenges it faced, its current situation, and the insights that can be gleaned from its history. The chapter concludes with comments on the implications for the future shape of CE/IE graduate programs and centers at universities. The author is the Founding Director of CIE who has led the program for most of its 50-year history.

Potassium iron hexacyanoferrate, or Prussian blue (PB), is investigated as a cathode material for nonaqueous divalent calcium ion batteries. PB is an attractive prospect due to its high specific capacity, nontoxicity, low cost, and simple synthesis. Charge/discharge performances are examined at current densities of 23 mAg−1, 45 mAg−1, 90 mAg−1, and 125 mAg−1 that produced reversible specific capacities ranging from 150 mAhg−1 (at 23 mAg−1 current density) to over 120 mAhg−1 (at 125 mAg−1 current density). These are the highest storage capacities to date for a divalent calcium ion cathode over extended period of charge/discharge cycling and are comparable in performance to monovalent intercalating ions.

Calcium cobalt hexacyanoferrate (CaCoHCF) was synthesized and tested as a cathode material for rechargeable batteries, using divalent cations (Mg2+, Ca2+, Ba2+). CaCoHCF demonstrated reversible specific capacity and coulombic efficiency (in parentheses) of 45.49 mAh/g (99.18%) for Mg2+, 55.04 mAh/g (99.2%) for Ca2+, and 44.09 mAh/g (99.42%) for Ba2+, at a current density of 25 mA/g. Of the three ions, Ca2+ resulted in the highest absolute specific capacity as well as high specific capacity utilization. The cathodes were also subjected to rate capability measurements using current densities of 50 mA/g (30 cycles) and 0.1 A/g (100 cycles). Upon addition of 2 mL water to the non-aqueous electrolyte, the fraction of theoretical specific capacity increased to 0.55 for Mg2+, 94.8% for Ca2+, and 95.53% forBa2+. This increase has been interpreted as the ability of the cathode material to intercalate and de-intercalate more ions due to the electrostatic shielding provided by water molecules between the host lattice and the guest cations. An empirical relationship between the cation size and specific capacity utilization is presented.Calcium cobalt hexacyanoferrate (CaCoHCF) was synthesized and tested as a cathode material for rechargeable batteries, using divalent cations (Mg2+, Ca2+, Ba2+). A computationally simple yet predictive multi-scale simulation scheme is introduced to estimate zero-field charge mobilities for amorphous OSC materials. A percolating charge model is utilized to describe inhomogeneity of hopping trajectories of carriers in amorphous media. The prediction scheme is composed of the following stages: quantum chemical calculation of Marcus inner sphere reorganization energies, molecular dynamics simulations of the amorphous condensed phase bulk structure, automated quantum chemical calculations of the electronic coupling for dimer pairs in the amorphous solid, and calculation of the Marcus theory charge hopping rates and an estimated bulk mobility using the Einstein relation, corrected for the inhomogeneous hopping network of the solid. Comparisons with independent experimental measurements of hole mobility for ten OSC compounds show that this approach gives good correlation between predictions and measurements suitable for ranking systems, and useful quantitative agreement. This low-cost model with minimal complexity is well-suited for incorporation into a virtual materials discovery framework for advanced OSC solutions.

Pseudomonas aeruginosa is a virulent pathogen that has become more threatening with the emergence of multidrug resistance. The aspartate transcarbamoylase (ATCase) of this organism is a dodecamer comprised of six 37 kDa catalytic chains and six 45 kDa chains homologous to dihydroorotase (pDHO). The pDHO chain is inactive but is necessary for ATCase activity. A stoichiometric mixture of the subunits associates into a dodecamer with full ATCase activity. Unlike other known ATCases, the P. aeruginosa catalytic chain does not spontaneously assemble into a trimer. Chemical-crosslinking and size-exclusion chromatography showed that P. aeruginosa ATCase is monomeric which accounts for its lack of catalytic activity since the active site is a composite comprised of residues from adjacent monomers in the trimer. Circular dichroism spectroscopy indicated that the ATCase chain adopts a structure that contains secondary structure elements although neither the ATCase nor the pDHO subunits are very stable as determined by a thermal shift assay. Formation of the complex increases the melting temperature by about 30°C. The ATCase is strongly inhibited by all nucleotide di- and triphosphates and exhibits extreme cooperativity. Previous studies suggested that the regulatory site is located in an 11-residue extension of the amino end of the catalytic chain. However, deletion of the extensions did not affect catalytic activity, nucleotide inhibition or the assembly of the dodecamer. Nucleotides destabilized the dodecamer which probably accounts for the inhibition and apparent cooperativity of the substrate saturation curves. Contrary to previous interpretations, these results suggest that P. aeruginosa ATCase is not allosterically regulated by nucleotides.

The influence of the precursors, namely potassium ferrocyanide and potassium ferricyanide on the particles sizes of Prussian Blue (PB) and Prussian Green (PG), under identical reaction conditions have been investigated. It was found that the particle sizes influence the gravimetric capacity utilization of these materials as cathodes for aqueous potassium (K + ) ion batteries. The PG particle sizes were on the order of 50–75 nm, whereas PB particles size were on the order of 2–10 microns. The PG cathodes demonstrated a reversible capacity of 121.4 mAhr/g, with a coulombic efficiency of 98.7% compared to PB cathodes which demonstrated 53.8 mAhr/g, with a coulombic efficiency of 100%. We interpret the increased capacity of PG batteries relative to PB batteries as being a result of the smaller particle size of PG, which results in greater accessibility of the cathode to K+ ions.

Potassium barium hexacyanoferrate (K2BaFe(CN)6) was investigated as a cathode material for reversible Ca2+ ion insertion/extraction type rechargeable battery using non-aqueous electrolytes. The electrochemical performance of K2BaFe(CN)6was evaluated using cyclic voltammetry and galvanic cycling at ambient temperature. It is shown that addition of water led to significant enhancement in intercalation and de-intercalation of Ca2+ ions, leading to improved charge/discharge capacity. The enhancement in performance is attributed to formation of solvation spheres around the intercalating Ca2+ ions which provide screening from the electrostatic charges of the BaFe(CN)6 lattice. A reversible capacity of 55.8 mA hr g−1 and a coulombic efficiency of 93.8% was demonstrated at the end of 30 charge/discharge cycles.

The purpose of this prospective study was to (1) evaluate the impact of retirement, (2) monitor the change in adjustment across time, and (3) identify the resources predictive of short- and long-term adjustment in retirement. A sample of 117 male retirees was assessed on indices of physical and psychological health, perceived control, retirement satisfaction, and life satisfaction at 2-4 months preretirement, 1 year post-, and 6-7 years postretirement. The results provided support for a positive impact of retirement, as retirees evidenced increases in well-being during the first year. There was also evidence of a retirement adjustment process, in that aspects of well-being (i.e., psychological health) changed from short- to long-term retirement. Finally, physical health, income, and voluntary retirement status predicted short-term adjustment, while internal locus of control was an additional resource for long-term adjustment. Changes in resources over time also differentially predicted short- and long-term adjustment (e.g., an increase in internal locus of control predicted an increase in activity satisfaction at 1 year but not at 6-7 years postretirement). A LTHOUGH retirement has received extensive study, the •**• nature of the impact of retirement and the resources important for a successful adjustment in retirement remain unclear. The length of time in retirement may be one factor that accounts for the inconsistency in results pertaining to the impact of retirement in particular (Jonsson, 1993). For example, studies have reported the existence of a negative association of well-being with time in retirement (Bosse, Aldwin, Levenson, Spiro, & Mroczek, 1993; Conner, Dorfman, & Tompkins, 1985; Knesek, 1992). However, the process of retirement adjustment may be more complex, reflecting specific patterns of change in a retiree's well-being at different points in time (Atchley, 1976). Atchley's stage model of retirement has received some support in recent studies (Bosse, Aldwin, Levenson, & Workman-Daniels, 1991; Ekerdt,. Bosse, & Levkoff, 1985; Richardson & Kilty, 1991; The"riault, 1994). There is also indication that the predictors of well-being or potential resources for a successful adjustment differ depending on the time postretirement (Bosse et al., 1991; Cherry, Zarit, & Krauss, 1984; Romsa, Bondy, & Blenman, 1985). The present research involves a prospective, longitudinal assessment of the retiree's well-being from 2-4 months preretirement to 1 year post- and 6-7 years postretirement. This study was designed to address the effects of time on the impact and process of retirement as well as on the importance of specific resources for a successful adjustment.

International audience; Elevated hydrostatic pressure was used to probe conformational changes of Aquifex aeolicus dihydroorotase (DHO), which catalyzes the third step in de novo pyrimidine biosynthesis. The isolated protein, a 45-kDa monomer, lacks catalytic activity but becomes active upon formation of a dodecameric complex with aspartate transcarbamoylase (ATC). X-ray crystallographic studies of the isolated DHO and of the complex showed that association induces several major conformational changes in the DHO structure. In the isolated DHO, a flexible loop occludes the active site blocking the access of substrates. The loop is mostly disordered but is tethered to the active site region by several electrostatic and hydrogen bonds. This loop becomes ordered and is displaced from the active site upon formation of DHO-ATC complex. The application of pressure to the complex causes its time-dependent dissociation and the loss of both DHO and ATC activities. Pressure induced irreversible dissociation of the obligate ATC trimer, and as a consequence the DHO is also inactivated. However, moderate hydrostatic pressure applied to the isolated DHO subunit mimics the complex formation and reversibly activates the isolated subunit in the absence of ATC, suggesting that the loop has been displaced from the active site. This effect of pressure is explained by the negative volume change associated with the disruption of ionic interactions and exposure of ionized amino acids to the solvent (electrostriction). The interpretation that the loop is relocated by pressure was validated by site-directed mutagenesis and by inhibition by small peptides that mimic the loop residues.

Aspartate transcarbamoylase and dihydroorotase, enzymes that catalyze the second and third step in de novo pyrimidine biosynthesis, are associated in dodecameric complexes in Aquifex aeolicus and many other organisms. The architecture of the dodecamer is ideally suited to channel the intermediate, carbamoyl aspartate from its site of synthesis on the ATC subunit to the active site of DHO, which catalyzes the next step in the pathway, because both reactions occur within a large, internal solvent-filled cavity. Channeling usually requires that the reactions of the enzymes are coordinated so that the rate of synthesis of the intermediate matches its rate of utilization. The linkage between the ATC and DHO subunits was demonstrated by showing that the binding of the bisubstrate analog, N-phosphonacetyl-L-aspartate to the ATC subunit inhibits the activity of the distal DHO subunit. Structural studies identified a DHO loop, loop A, interdigitating between the ATC domains that would be expected to interfere with domain closure essential for ATC catalysis. Mutation of the DHO residues in loop A that penetrate deeply between the two ATC domains inhibits the ATC activity by interfering with the normal reciprocal linkage between the two enzymes. Moreover, a synthetic peptide that mimics that part of the DHO loop that binds between the two ATC domains was found to be an allosteric or noncompletive ATC inhibitor (K(i) = 22 μM). A model is proposed suggesting that loop A is an important component of the functional linkage between the enzymes.

Chemical modification of graphene by covalently functionalizing its surface potentially allows a wider flexibility in engineering electronic structure, in particular the local density of states of the carbon atoms bound to the modifier that can result in opening of the band gap. Such binding can involve covalent hydrogenation of graphene to modify hybridization of carbon atoms from sp2 to sp3 geometry [1] , [2] , [3] . Methods have also been developed to functionalize graphene covalently with molecular species [4] , [5] , [6] , [7] , [8] . Among these, perfluorophenylazide (PFPA) functionalization of graphene is well-developed using a nitrene intermediate. Films of this molecule also act as adhesion layers that allow production of long ribbons of exfoliated graphene [7] , [8] , [9] . We have developed a theory to predict electrical properties of PFPA functionalized graphene and compared it to experimental results. Conductivity of these PFPA functionalized ribbons of exfoliated graphene show good agreement with our theory.

Although the room-temperature rechargeable sodium-ion battery has emerged as an attractive alternative energy storage solution for large-scale deployment, major challenges toward practical sodium-ion battery technology remain including identification and engineering of anode materials that are both technologically feasible and economical. Herein, an antimony-based anode is developed by incorporating antimony into graphitic carbon matrices using low-cost materials and scalable processes. The composite anode exhibits excellent overall performance in terms of packing density, fast charge/discharge capability and cyclability, which is enabled by the conductive and compact graphitic network. A full cell design featuring this composite anode with a hexacyanometallate cathode achieves superior power output and low polarization, which offers the potential for realizing a high-performance, cost-effective sodium-ion battery.

The genome of the major intestinal archaeon Methanobrevibacter smithii contains a complex gene system coding for carbamoyl phosphate synthetase (CPSase) composed of both full-length and reduced-size synthetase subunits. These ammonia-metabolizing enzymes could play a key role in controlling ammonia assimilation in M. smithii, affecting the metabolism of gut bacterial microbiota, with an impact on host obesity. In this study, we isolated and characterized the small (41 kDa) CPSase homolog from M. smithii. The gene was cloned and overexpressed in Escherichia coli, and the recombinant enzyme was purified in one step. Chemical cross-linking and size exclusion chromatography indicated a homodimeric/tetrameric structure, in accordance with a dimer-based CPSase activity and reaction mechanism. This small enzyme, MS-s, synthesized carbamoyl phosphate from ATP, bicarbonate, and ammonia and catalyzed the same ATP-dependent partial reactions observed for full-length CPSases. Steady-state kinetics revealed a high apparent affinity for ATP and ammonia. Sequence comparisons, molecular modeling, and kinetic studies suggest that this enzyme corresponds to one of the two synthetase domains of the full-length CPSase that catalyze the ATP-dependent phosphorylations involved in the three-step synthesis of carbamoyl phosphate. This protein represents the smallest naturally occurring active CPSase characterized thus far. The small M. smithii CPSase appears to be specialized for carbamoyl phosphate metabolism in methanogens.

An impedimetric biosensor platform for bioaffinity assays has been developed that is based on real-time, label-free electrochemical detection performed via a direct interface to electronic digital data processing. The sensor array consists of 15 gold microelectrode pairs (Fig. 1) that are enclosed in three reaction chambers and biofunctionalized with specific probes. The impedance change caused by specific capture of target analyte molecules on the functionalized electrode surface is recorded in real time. The measuring instrument is capable of continuous and simultaneous stimulation and recording of all electrodes on the array. A corresponding mathematical algorithm and a software package for data analysis have been developed. The software performs (i) filtering of the instrument noise, and (ii) extraction of the exponential component of the impedance signal. Thus, the algorithm can quantify both rate of target to probe binding, and target to probe affinity. The described fully integrated platform can be used as a basic research tool for development of various bio-affinity impedimetric assays. To facilitate such applications, we have developed a streamlined manufacturing technology, and a set of assay protocols for detection of microbes based on nucleic acid hybridization. The assay was shown to detect and distinguish between two closely related but different Escherichia coli strains. The assay sensitivity was sufficient for reliable measurements of specific PCR products amplified from microbial genomic DNA. The sensor array platform is adaptable for detection of a wide range of analytes of practical significance, and it has potential for further integration with amplification (i.e. PCR) and sample preparation modules.

A novel, impedance-based electronic sensor format was used for label-free, real-time detection of microbial DNA on oligonucleotide probe arrays. Detection limits of 5–10 nM were achieved for single-stranded, PCR-amplified DNA targets. Hybridization selectivity yielded 9- to 12-fold signal increases for specific targets, and the sensor arrays were re-used multiple times without significant signal degradation. These and other features of the SHARP Laboratories of America (SLA) sensor array, such as its ability to acquire continuous measurements of DNA as it accumulates on the array surface, make it an attractive biosensor platform for field detection and monitoring of sentinel and/or pathogenic microorganisms.

In prokaryotes, the first three enzymes in pyrimidine biosynthesis, carbamoyl phosphate synthetase (CPS), aspartate transcarbamoylase (ATC), and dihydroorotase (DHO), are commonly expressed separately and either function independently (Escherichia coli) or associate into multifunctional complexes (Aquifex aeolicus). In mammals the enzymes are expressed as a single polypeptide chain (CAD) in the order CPS-DHO-ATC and associate into a hexamer. This study presents the three-dimensional structure of the noncovalent hexamer of DHO and ATC from the hyperthermophile A. aeolicus at 2.3 A resolution. It is the first structure of any multienzyme complex in pyrimidine biosynthesis and is a possible model for the core of mammalian CAD. The structure has citrate, a near isosteric analogue of carbamoyl aspartate, bound to the active sites of both enzymes. Three active site loops that are intrinsically disordered in the free, inactive DHO are ordered in the complex. The reorganization also changes the peptide bond between Asp153, a ligand of the single zinc atom in DHO, and Gly154, to the rare cis conformation. In the crystal structure, six DHO and six ATC chains form a hollow dodecamer, in which the 12 active sites face an internal reaction chamber that is approximately 60 A in diameter and connected to the cytosol by narrow tunnels. The entrances and the interior of the chamber are both electropositive, which suggests that the architecture of this nanoreactor modifies the kinetics of the bisynthase, not only by steric channeling but also by preferential escape of the product, dihydroorotase, which is less negatively charged than its precursors, carbamoyl phosphate, aspartate, or carbamoyl aspartate.

Carbamoyl phosphate synthetase synchronizes the utilization of two ATP molecules at duplicated ATP-grasp folds to catalyze carbamoyl phosphate formation. To define the dedicated functional role played by each of the two ATP sites, we have carried out pulse/labeling studies using the synthetases from Aquifex aeolicus and Methanococcus jannaschii, hyperthermophilic organisms that encode the two ATP-grasp folds on separate subunits. These studies allowed us to differentially label each active site with [gamma-(32)P]ATP and determine the fate of the labeled gamma-phosphate in the synthetase reaction. Our results provide the first direct demonstration that enzyme-catalyzed transfer of phosphate from ATP to carbamate occurs on the more C-terminal of the two ATP-grasp folds. These findings rule out one mechanism proposed for carbamoyl phosphate synthetase, where one ATP acts as a molecular switch, and provide additional support for a sequential reaction mechanism where the gamma-phosphate groups of both ATP molecules are transferred to reactants. CP synthesis by subunit C in our single turnover pulse/chase assays did not require subunit N, but subunit N was required for detectable CP synthesis in the traditional continuous assay. These findings suggest that cross-talk between domain N and C is required for product release from subunit C.

The flux through the de novo pyrimidine biosynthetic pathway is controlled by the multifunctional protein CAD, which catalyzes the first three steps. The cell cycle dependent regulation of pyrimidine biosynthesis is a consequence of sequential phosphorylation of CAD Thr456 and Ser1406 by the MAP kinase and PKA cascades, respectively. Coordinated regulation of the pathway requires precise timing of the two phosphorylation events. These studies show that phosphorylation of purified CAD by PKA antagonizes MAP kinase phosphorylation, and vice versa. Similar results were observed in vivo. Forskolin activation of PKA in BHK-21 cells resulted in a 8.5 fold increase in Ser1406 phosphorylation and severely curtailed the MAP kinase mediated phosphorylation of CAD Thr456. Moreover, the relative activity of MAP kinase and PKA was found to determine the extent of Thr456 phosphorylation. Transfectants expressing elevated levels of MAP kinase resulted in a 11-fold increase in Thr456 phosphorylation, whereas transfectants that overexpress PKA reduced Thr456 phosphorylation 5-fold. While phosphorylation of one site by one kinase may induce conformational changes that interfere with phosphorylation by the other, the observation that both MAP kinase and PKA form stable complexes with CAD suggest that the mutual antagonism is the result of steric interference by the bound kinases. The reciprocal antagonism of CAD phosphorylation by MAP kinase and PKA provides an elegant mechanism to coordinate the cell cycle-dependent regulation of pyrimidine biosynthesis ensuring that signals for up- and down-regulation of the pathway do not conflict.

Dihydroorotases (EC 3.5.2.3) catalyze the reversible cyclization of carbamoyl aspartate to form dihydroorotate in de novo pyrimidine biosynthesis. The X-ray structures of Aquifex aeolicus dihydroorotase in two space groups, C 222 1 and C 2, were determined at a resolution of 1.7 A. These are the first structures of a type I dihydroorotase, a class of molecules that includes the dihydroorotase domain of mammalian CAD. The type I enzymes are more ancient and larger, at 45 kDa, than the type II enzymes exemplified by the 38 kDa Escherichia coli dihydroorotase. Both dihydroorotases are members of the metallo-dependent hydrolase superfamily, whose members have a distorted “TIM barrel” domain containing the active site. However, A. aeolicus dihydroorotase has a second, composite domain, which the E. coli enzyme lacks and has only one of the two zinc atoms present in the E. coli enzyme. A. aeolicus dihydroorotase is unique in exhibiting significant activity only when complexed with aspartate transcarbamoylase, whereas the E. coli dihydroorotase and the CAD dihydroorotase domain are active as free proteins. The latency of A. aeolicus dihydroorotase can be related to two differences between its structure and that of E. coli dihydroorotase: (1) the monoclinic structure has a novel cysteine ligand to the zinc that blocks the active site and possibly functions as a “cysteine switch”; and (2) active site residues that bind the substrate in E. coli dihydroorotase are located in disordered loops in both crystal structures of A. aeolicus dihydroorotase and may function as a disorder-to-order “entropy switch”.

Dihydroorotase (DHOase) catalyzes the reversible condensation of carbamoyl aspartate to form dihydroorotate in de novo pyrimidine biosynthesis. The enzyme from Aquifex aeolicus, a hyperthermophilic organism of ancient lineage, was cloned and expressed in Escherichia coli. The purified protein was found to be a 45-kDa monomer containing a single zinc ion. Although there is no other DHOase gene in the A. aeolicus genome, the recombinant protein completely lacked catalytic activity at any temperature tested. However, DHOase formed an active complex with aspartate transcarbamoylase (ATCase) from the same organism. Whereas the kcat of 13.8 ± 0.03 s–1 was close to the value observed for the mammalian enzyme, the K mfor dihydroorotate, 3.03 ± 0.05 mm was 433-fold higher. Gel filtration and chemical cross-linking showed that the complex exists as a 240-kDa hexamer (DHO3-ATC3) and a 480-kDa duodecamer (DHO6-ATC6) probably in rapid equilibrium. Complex formation protects both DHOase and ATCase against thermal degradation at temperatures near 100 °C where the organism grows optimally. These results lead to the reclassification of both enzymes: ATCase, previously considered a Class C homotrimer, now falls into Class A, whereas the DHOase is a Class 1B enzyme. CD spectroscopy indicated that association with ATCase does not involve a significant perturbation of the DHOase secondary structure, but the visible absorption spectrum of a Co2+-substituted DHOase is appreciably altered upon complex formation suggesting a change in the electronic environment of the active site. The association of DHOase with ATCase probably serves as a molecular switch that ensures that free, uncomplexed DHOase in the cell remains inactive. At pH 7.4, the equilibrium ratio of carbamoyl aspartate to dihydroorotate is 17 and complex formation may drive the reaction in the biosynthetic direction.

Suppression of protein synthesis in the brain following an ischemic insult has been thought to occur because of inhibition of translation initiation. All eukaryotic mRNAs, with the exception of heat-shock transcripts, require the activity of eukaryotic initiation factor (eIF) 4E for formation of the translation initiation complex, and eIF-4E availability is rate-limiting. The response of brain eIF-4E concentration and phosphorylation following decapitation ischemia was studied in rat brain homogenates after electrophoresis and western blotting with antibodies against eIF-4E and phosphoserine, respectively. There was no change in level of eIF-4E after 5 min of ischemia (p = 0.82 vs. time 0), but it had decreased 32 (p = 0.01) and 57% (p = 0.006) after 10 and 20 min of ischemia, respectively. There was no loss of serine phosphorylation on eIF-4E beyond signal loss observed due to degradation of the protein itself (p = 0.31). In vitro exposure of eIF-4E to activated mu-calpain resulted in a 50% loss in 10 min of eIF-4E on western blots. If active eIF-4E is required for translation of its own mRNA, degradation of this protein during ischemia, possibly by activated mu-calpain, could be a direct mechanism of irreversible neuronal injury, and the rate of proteolysis of eIF-4E could place an upper time limit on the maximal duration of global brain ischemia compatible with neurologic recovery.

Aspartate transcarbamoylase from Pseudomonadaceae is a class A enzyme consisting of six copies of a 36-kDa catalytic chain and six copies of a 45-kDa polypeptide of unknown function. The 45-kDa polypeptide is homologous to dihydroorotase but lacks catalytic activity. Pseudomonas aeruginosa aspartate transcarbamoylase was overexpressed in Escherichia coli. The homogeneous His-tagged protein isolated in high yield, 30 mg/liter of culture, by affinity chromatography and crystallized. Attempts to dissociate the catalytic and pseudo-dihydroorotase (pDHO) subunits or to express catalytic subunits only were unsuccessful suggesting that the pDHO subunits are required for the proper folding and assembly of the complex. As reported previously, the enzyme was inhibited by micromolar concentrations of all nucleotide triphosphates. In the absence of effectors, the aspartate saturation curves were hyperbolic but became strongly sigmoidal in the presence of low concentrations of nucleotide triphosphates. The inhibition was unusual in that only free ATP, not MgATP, inhibits the enzyme. Moreover, kinetic and binding studies with a fluorescent ATP analog suggested that ATP induces a conformational change that interferes with the binding of carbamoyl phosphate but has little effect once carbamoyl phosphate is bound. The peculiar allosteric properties suggest that the enzyme may be a potential target for novel chemotherapeutic agents designed to combatPseudomonas infection.

CAD, a large multifunctional protein that carries carbamoyl phosphate synthetase (CPSase), aspartate transcarbamoylase, and dihydroorotase activities, catalyzes the first three steps of de novo pyrimidine biosynthesis in mammalian cells. The CPSase component, which catalyzes the initial, rate-limiting step, exhibits complex regulatory mechanisms involving allosteric effectors and phosphorylation that control the flux of metabolites through the pathway. Incubation of CAD with ATP in the absence of exogenous kinases resulted in the incorporation of 1 mol of P(i)/mol of CAD monomer. Mass spectrometry analysis of tryptic digests showed that Thr(1037) located within the CAD CPS.B subdomain was specifically modified. The reaction is specific for MgATP, ADP was a competitive inhibitor, and the native tertiary structure of the protein was required. Phosphorylation occurred after denaturation, further purification of CAD by SDS gel electrophoresis, and renaturation on a nitrocellulose membrane, strongly suggesting that phosphate incorporation resulted from an intrinsic kinase activity and was not the result of contaminating kinases. Chemical modification with the ATP analog, 5'-p-fluorosulfonylbenzoyladenosine, showed that one or both of the active sites that catalyze the ATP-dependent partial reactions are also involved in autophosphorylation. The rate of phosphorylation was dependent on the concentration of CAD, indicating that the reaction was, at least in part, intermolecular. Autophosphorylation resulted in a 2-fold increase in CPSase activity, an increased sensitivity to the feedback inhibitor UTP, and decreased allosteric activation by 5-phosphoribosyl-1-pyrophosphate, functional changes that were distinctly different from those resulting from phosphorylation by either the protein kinase A or mitogen-activated protein kinase cascades.

The carbamoyl phosphate synthetase domain of the multifunctional protein CAD catalyzes the initial, rate-limiting step in mammalian de novo pyrimidine biosynthesis. In addition to allosteric regulation by the inhibitor UTP and the activator PRPP, the carbamoyl phosphate synthetase activity is controlled by mitogen-activated protein kinase (MAPK)- and protein kinase A (PKA)-mediated phosphorylation. MAPK phosphorylation, both in vivo and in vitro, increases sensitivity to PRPP and decreases sensitivity to the inhibitor UTP, whereas PKA phosphorylation reduces the response to both allosteric effectors. To elucidate the factors responsible for growth state-dependent regulation of pyrimidine biosynthesis, the activity of the de novo pyrimidine pathway, the MAPK and PKA activities, the phosphorylation state, and the allosteric regulation of CAD were measured as a function of growth state. As cells entered the exponential growth phase, there was an 8-fold increase in pyrimidine biosynthesis that was accompanied by a 40-fold increase in MAPK activity and a 4-fold increase in CAD threonine phosphorylation. PRPP activation increased to 21-fold, and UTP became a modest activator. These changes were reversed when the cultures approach confluence and growth ceases. Moreover, CAD phosphoserine, a measure of PKA phosphorylation, increased 2-fold in confluent cells. These results are consistent with the activation of CAD by MAPK during periods of rapid growth and its down-regulation in confluent cells associated with decreased MAPK phosphorylation and a concomitant increase in PKA phosphorylation. A scheme is proposed that could account for growth-dependent regulation of pyrimidine biosynthesis based on the sequential action of MAPK and PKA on the carbamoyl phosphate synthetase activity of CAD.

The paper will introduce a simple new method for the synthesis of both hafnium and zirconium nitrate precursors. The intermediate product, dinitrogen pentoxide produced from the extraction of water from fuming nitric acid via phosphorus pentoxide, was condensed with a liquid nitrogen trap into a flask containing either hafnium or zironium tetrachloride. To achieve a high yield, the mixture of fuming nitric acid and phosphorus pentoxide was heated to a certain temperature, from which a large quantity of dinitrogen pentoxide had been generated. In the following steps hafnium or zirconium tetrachloride was refluxed over dinitrogen pentoxide at 30 to 35 °C for about a half-hour. The product was purified by sublimation. Yields above 95% were obtained. The cost for the hafnium nitrate precursor synthesis was estimated. The precursor was not stable at room temperature, and should be stored in refrigerator in sealed vials. No chlorine was detected from both EDS and chemical analysis. The volatility was evaluated ...

Nanostructures provide novel opportunities of studying epitaxy in nano/mesoscale and on nonplanar substrates. Epitaxial growth of silicon (Si) on the surfaces of Si nanowires along radial direction is a promising way to prepare radial p-(i)-n junction in nanoscale for optoelectronic devices. Comprehensive studies of Si radial epitaxy in micro/nanoscale reveal that morphological evolution and size-dependent radial shell growth rate for undoped and doped Si radial shells. Single crystalline Si radial p-i-n junction wire arrays were utilized to fabricate photovoltaic (PV) devices. The PV devices exhibited the photoconversion efficiency of 10%, the short-circuit current density of 39 mA/cm 2 , and the open-circuit voltage of 0.52 V, respectively.

To examine the reliability and validity of a new outcome measure, the Upper Body Musculoskeletal Assessment (UBMA).Twenty subjects physician-diagnosed as having work-related musculoskeletal disorders (WRMD) and ten healthy subjects were assessed using the UBMA on three separate occasions. All subjects with WRMD attributed their injury to equipment use on their job.The WRMD group had significantly higher UBMA scores on the side of equipment use than on the other side (p0.01), whereas the healthy group had similar scores on both sides (p0.05). UBMA scores for the WRMD group were significantly greater on both sides of the body than scores for the healthy group (p0.01). Only one test occasion was required to produce excellent reliability coefficients (ICCs0.88). Although group reliability was excellent, changes of 24% for patients with WRMD and 44% for healthy subjects would be required for confidence that UBMA scores for individual patients on the side of equipment use had changed from baseline.Although testing on one occasion produced reliable UBMA scores, healthy subjects could be distinguished from patients with WRMD, and the side of equipment use could be distinguished from the other side in patients with WRMD, prediction of individual UBMA scores was poor. In its present form, the UBMA is useful for making decisions about groups but not about individual patients. Modifications of the current UBMA are required to reduce measurement error.

The purpose of this prospective study was to (1) monitor the change in adjustment across time and life domains and (2) identify the resources predictive of short- and long-term adjustment to the university transition. A sample of 68 first-year students reported on events, resources, and well-being at four times across the year. Coping and appraisal were evaluated specifically in relation to three life domains (e.g., academic). Results showed that this transition represents a relatively acute stressor, evidencing the greatest strain for students and the largest impact on their well-being on entry. As time passed, students experienced steady improvement in most aspects of adjustment. Results also suggested that women, despite having more resources, may have greater vulnerability to this transition. Finally, the importance of a specific resource to the prediction of well-being depended on the measure of well-being assessed, the specific life domain addressed, the time of assessment, and whether or not concom...

"India's primary education glass is two-thirds full, one-third empty ... The result is that many of the huge returns to primary education are foregone. . . . India's low average educational attainment has not yet reached the critical threshold where benefits are greatest and high economic growth rates are sustained.... At the current rate of growth of schooling, some of the largest states will not reach an average of 4 to 5 years of primary education until well into the next century" (World Bank, p. 1). "No matter how one looks at it, the state of elementary education in India is dismal. ... The fact that elementary education in India remains far from universal is no secret. Yet, the enormity of the problem is often underestimated. . . . India's success with higher education ... has even created the impression that we are doing just fine" (PROBE Team, p. 8). The quotations above capture the perspectives of two recent books on the condition of primary education in India today. The necessity and enormity of the task of achieving widespread primary education in a country with over 100 million children of primary-school age makes this a topic of critical interest to educators in India and elsewhere. Either book by itself leaves the reader with a partial picture; together they produce a more complete and realistic understanding of the challenge of Education for All in India. Each of these books tries to balance the depressing reality of the educational experience for millions of rural children with careful cataloging of progress made and examples of successful interventions from various parts of India. Both books draw heavily on Indian research and scholarship, reflecting the substantial capability of both the research and the policy communities in India today. They differ radically, though, in the "voice" chosen to convey the message and in the kinds of information used to represent reality. The World Bank's Primary Education in India speaks at the macrolevel with the numbers, percentages, and computer projections of economic analysis. The Public Report on Primary Education (PROBE) portrays the reality of primary education at the village level as lived and narrated by pupils, teachers, and parents in rural communities. Primary Education in India provides a solid overview of the state of primary education in India, an extensive set of statistics on education for the fifteen major states, and a strong analysis of both the costs and financing of education. The de novo synthesis of pyrimidine nucleotides is required for mammalian cells to proliferate. The rate-limiting step in this pathway is catalysed by carbamoyl phosphate synthetase (CPS II), part of the multifunctional enzyme CAD1,2. Here we describe the regulation of CAD by the mitogen-activated protein (MAP) kinase cascade. When phosphorylated by MAP kinase in vitro or activated by epidermal growth factor in vivo , CAD lost its feedback inhibition (which is dependent on uridine triphosphate) and became more sensitive to activation (which depends upon phosphoribosyl pyrophosphate). Both these allosteric regulatory changes favour biosynthesis of pyrimidines for growth2. They were accompanied by increased epidermal growth factor-dependent phosphorylation of CAD in vivo and were prevented by inhibition of MAP kinase. Mutation of a consensus MAP kinase phosphorylation site abolished the changes in CAD allosteric regulation that were stimulated by growth factors. Finally, consistent with an effect of MAP kinase signalling on CPS II activity, epidermal growth factor increased cellular uridine triphosphate and this increase was reversed by inhibition of MAP kinase. Hence these studies may indicate a direct link between activation of the MAP kinase cascade and de novo biosynthesis of pyrimidine nucleotides.

Mammalian carbamoyl-phosphate synthetase is part of carbamoyl-phosphate synthetase-aspartate carbamoyltransferase-dihydroorotase (CAD), a multifunctional protein that also catalyzes the second and third steps of pyrimidine biosynthesis. Carbamoyl phosphate synthesis requires the concerted action of the glutaminase (GLN) and carbamoyl-phosphate synthetase domains of CAD. There is a functional linkage between these domains such that glutamine hydrolysis on the GLN domain does not occur at a significant rate unless ATP and HCO3−, the other substrates needed for carbamoyl phosphate synthesis, bind to the synthetase domain. The GLN domain consists of catalytic and attenuation subdomains. In the separately cloned GLN domain, the catalytic subdomain is down-regulated by interactions with the attenuation domain, a process thought to be part of the functional linkage. Replacement of Ser44 in the GLN attenuation domain with alanine increases thekcat/Km for glutamine hydrolysis 680-fold. The formation of a functional hybrid between the mammalian Ser44 GLN domain and the Escherichia coli carbamoyl-phosphate synthetase large subunit had little effect on glutamine hydrolysis. In contrast, ATP and HCO3− did not stimulate the glutaminase activity, indicating that the interdomain linkage had been disrupted. In accord with this interpretation, the rate of glutamine hydrolysis and carbamoyl phosphate synthesis were no longer coordinated. Approximately 3 times more glutamine was hydrolyzed by the Ser44 → Ala mutant than that needed for carbamoyl phosphate synthesis. Ser44, the only attenuation subdomain residue that extends into the GLN active site, appears to be an integral component of the regulatory circuit that phases glutamine hydrolysis and carbamoyl phosphate synthesis.

The kinetics of the coupled reactions between carbamoyl-phosphate synthetase (CPSase) and both aspartate transcarbamoylase (ATCase) and ornithine transcarbamoylase (OTCase) from the deep sea hyperthermophilic archaeon Pyrococcus abyssi demonstrate the existence of carbamoyl phosphate channeling in both the pyrimidine and arginine biosynthetic pathways. Isotopic dilution experiments and coupled reaction kinetics analyzed within the context of the formalism proposed by Ovádi et al. (Ovádi, J., Tompa, P., Vertessy, B., Orosz, F., Keleti, T., and Welch, G. R. (1989) Biochem. J. 257, 187-190) are consistent with a partial channeling of the intermediate at 37 degrees C, but channeling efficiency increases dramatically at elevated temperatures. There is no preferential partitioning of carbamoyl phosphate between the arginine and pyrimidine biosynthetic pathways. Gel filtration chromatography at high and low temperature and in the presence and absence of substrates did not reveal stable complexes between P. abyssi CPSase and either ATCase or OTCase. Thus, channeling must occur during the dynamic association of coupled enzymes pairs. The interaction of CPSase-ATCase was further demonstrated by the unexpectedly weak inhibition of the coupled reaction by the bisubstrate analog, N-(phosphonacetyl)-L-aspartate (PALA). The anomalous effect of PALA suggests that, in the coupled reaction, the effective concentration of carbamoyl phosphate in the vicinity of the ATCase active site is 96-fold higher than the concentration in the bulk phase. Channeling probably plays an essential role in protecting this very unstable intermediate of metabolic pathways performing at extreme temperatures.

The initial steps of pyrimidine biosynthesis in yeast and mammals are catalyzed by large multifunctional proteins of similar size, sequence and domain structure, but appreciable functional differences. The mammalian protein, CAD, has carbamyl phosphate synthetase (CPSase), aspartate transcarbamylase (ATCase) and dihydroorotase (DHOase) activities. The yeast protein, ura2, catalyzes the first two reactions and has a domain, called pDHO, which is homologous to mammalian DHOase, but is inactive. In CAD, only CPSase is regulated, whereas both CPSase and ATCase in the yeast protein are inhibited by UTP. These functional differences were explored by constructing a series of mammalian yeast chimeras. The isolated ATCase domain is catalytically active, but is not regulated. The inclusion of the yeast sequences homologous to the mammalian regulatory domain (B3) and the intervening pDHO domain did not confer regulation. Chimeric proteins in which the homologous regions of the mammalian protein were replaced by the corresponding domains of ura2 exhibited full catalytic activity, as well regulation of the CPSase, but not the ATCase, activities. The yeast B3 subdomain confers UTP sensitivity on the mammalian CPSase, suggesting that it is the locus of CPSase regulation in ura2. Taken together, these results indicate that there are regulatory site(s) in ura2. Channeling is impaired in all the chimeric complexes and completely abolished in the chimera in which the pDHO domain of yeast is replaced by the mammalian DHO domain.

Carbamoyl-phosphate synthetase consists of an amidotransferase domain or subunit (GLN) that hydrolyzes glutamine and transfers the ammonia to the synthetase component (CPS) where the biosynthetic reaction occurs. The CPS domain is composed of two homologous subdomains, CPS.A and CPS.B, that catalyze different ATP-dependent reactions involved in carbamoyl phosphate synthesis. When the individual CPS.A and CPS.B subdomains were individually cloned and expressed in Escherichia coli (Guy, H. I., and Evans, D. R. (1996) J. Biol. Chem. 271, 13762-13769), they were found to be functionally equivalent and could each independently catalyze carbamoyl phosphate synthesis. The proposal was advanced that, although the monomers could catalyze the individual partial reactions, overall synthesis of carbamoyl phosphate required a homodimer of CPS.A or CPS.B. To test this hypothesis, the GLN-CPS.B dimer was reversibly dissociated at 1500 bar in a high pressure cell. Dissociation was accompanied by a loss of both glutamine- and ammonia-dependent CPSase activity. Activity was recovered once the protein was returned to atmospheric pressure. If the sample was cross-linked before exposure to high pressure, there was no dissociation and no loss of biosynthetic activity. In contrast, the bicarbonate-dependent ATPase and the carbamoyl phosphate-dependent ATP synthetase activities were largely unaffected by pressure-induced dissociation. These experiments confirmed the hypothesis that the synthesis of carbamoyl phosphate requires the concerted action of the two active sites within the homodimer.
