Abstract The world's 1.4 million lakes (≥10 ha) provide many ecosystem services that are essential for human well‐being; however, only if their health status is good. Here, we reviewed common lake health issues and classified them using a simple human health‐based approach to outline that lakes are living systems that are in need of oxygen, clean water and a balanced energy and nutrient supply. The main reason for adopting some of the human health terminology for the lake health classification is to increase the awareness and understanding of global lake health issues. We show that lakes are exposed to various anthropogenic stressors which can result in many lake health issues, ranging from thermal, circulatory, respiratory, nutritional and metabolic issues to infections and poisoning. Of particular concern for human well‐being is the widespread lake drying, which is a severe circulatory issue with many cascading effects on lake health. We estimated that ∼115,000 lakes evaporate twice as much water as they gain from direct precipitation, making them vulnerable to potential drying if inflowing waters follow the drying trend, putting more than 153 million people at risk who live in close vicinity to those lakes. Where lake health issues remain untreated, essential ecosystem services will decline or even vanish, posing a threat to the well‐being of millions of people. We recommend coordinated multisectoral and multidisciplinary prevention and treatment strategies, which need to include a follow‐up of the progress and an assessment of the resilience of lakes to intensifying threats. Priority should be given to implementing sewage water treatment, mitigating climate change, counteracting introductions of non‐native species to lakes and decreasing uncontrolled anthropogenic releases of chemicals into the hydro‐, bio‐, and atmosphere.

Study region: The Upper Nerang River Catchment located in South-East Queensland (SEQ), Australia. Study focus: The Soil Water Assessment Tool (SWAT) model was applied to Upper Nerang River Catchment to test the sensitivity of catchment runoff and water quality to changes in Leaf Area Index (LAI) and Soil (topsoil) Organic Carbon (SOC) associated with climate change. These parameters are normally considered to be stationary in hydrological models, potentially constraining the accuracy of these models for catchment simulation under future climate scenarios. New hydrological insights for the region: LAI decreased by 0.4% to 16.9% and SOC by 5.9 to 11.6%, depending on land cover, future period, and emission scenario. Climate change scenarios showed an increase in extreme flows compared with a baseline (recent historical) case and were characterised by reduced return period and greater magnitude of discharge. Changes in LAI and SOC have the potential to mitigate these changes and may reduce loads of SS, TN and TP under future climate change scenarios. The findings of this study indicate that incorporating changes in LAI and SOC into hydrological models is important for modelling the catchment response to climate change. Furthermore, LAI and SOC could be managed to mitigate impacts of climate change on streamflow and losses of SS, TN and TP from catchment soils to receiving waters.

A global survey of 179 restoration practitioners spanning 65 countries identified the extent of stakeholder engagement as a key factor determining the success or failure of restoration projects. Lack of support across sectors and for funding, policy, monitoring, governance and knowledge assessment of pressures and their effects were most frequently cited as factors contributing to restoration failure.The responses indicate that, although nutrient enrichment is perceived to be the primary issue for lakes globally, the impacts of climate change, hydrological modifications and invasive species are widely recognized as pervasive anthropogenic pressures of global importance. Practitioners recognized that the ecosystem services most impacted by these pressures were recreation and tourism, although in low income countries the provisioning service, aquaculture, was considered most impacted. Ecology-based and/or pressure-related restoration targets had been set for most restoration programs in our survey. However, the strength of the evidence underpinning these targets was often weak and the effects of climate change were rarely considered when setting targets.The most effective and widely used restoration measures target nutrient loading (both catchment and in-lake) while hydrological modifications and the implementation of nature-based solutions are used to a lesser extent. Measures for the control of non-native invasive species are rarely applied and are viewed as being largely ineffective.The results of the survey provide direction for future work. New and emerging pressures, singly and in combination, may require new approaches to lake restoration: for both setting restoration targets and devising restoration strategies. The future of lake restoration depends on joined-up thinking that better integrates science into policy and practice and, most importantly, ensures strong and inclusive stakeholder engagement and collaboration across multiple sectors.

Benzalkonium chloride (BC) is widely used for disinfection in the food industry. However, Listeria monocytogenes strains with resistance to BC have been reported recently. In L. monocytogenes, the Agr communication system consists of a membrane-bound peptidase AgrB, a precursor peptide AgrD, a histidine kinase (HK) AgrC, and a response regulator (RR) AgrA. Our previous study showed that the agr genes are significantly upregulated by BC adaptation. This study aimed to investigate the role of the Agr system in BC resistance in L. monocytogenes. Our results showed that the Agr system was involved in BC resistance. However, a direct interaction between BC and AgrC was not observed, nor between BC and AgrA. These results indicated that BC could induce the Agr system via an indirect action. Both AgrBD and AgrC were required for growth under BC stress. Nevertheless, when exposed to BC, the gene deletion mutant ∆agrA strain exhibited better growth performance than its parental strain. The RR Lmo1172 played a role in BC resistance in the ∆agrA strain, suggesting that Lmo1172 may be an alternative to AgrA in the phosphotransfer pathway. Phosphorylation of Lmo1172 by AgrC was observed in vitro. The cognate HK Lmo1173 of Lmo1172 was not involved in BC stress, regardless of whether it was as the wild-type or the ∆agrA mutant strain. Our evidence suggests that the HK AgrC cross-phosphorylates its noncognate RR Lmo1172 to cope with BC stress when the cognate RR AgrA is absent. In vivo, further studies will be required to detect phosphotransfer of AgrC/AgrA and AgrC/Lmo1172.

This study combined a catchment model and one-dimensional lake model (GLM-AED) to simulate the response of hydrodynamics and water quality of subtropical Advancetown Lake (South-East Queensland, Australia) to future changing climates from 2040 to 2069 and 2070 to 2099 under Representative Concentration Pathway (RCP) 4.5 and 8.5 and increased water demand from a 50% increase in population over current levels. The simulation adequately reproduced water temperature (RMSE of 0.6 °C), dissolved oxygen (DO) (RMSE of 2 mg/L), and other water quality variables, such as nitrogen, phosphorus, and chlorophyll a (Chl-a). Warming temperatures dominated the change in thermal structure and hydrodynamic status of the lake under future climate change conditions. Projected changes in precipitation and hydrological response from the upstream catchment might, however, partly offset the warming temperatures under future climate change. Increased water withdrawal due to population growth, which involved water extraction from the epilimnion, showed antagonistic effects on water stability compared to those from climate change. Under a high emission scenario of RCP8.5 during the 2080s, there is an increased likelihood of winter turnover failure in Advancetown Lake. Nutrient concentrations were simulated to decrease from reduced catchment loads under future climate change conditions. However, Chl-a concentrations were simulated to increase, especially during the period after winter turnover, under these future conditions. The depth of the hypoxia front during stratification is expected to decrease and move towards the water surface, attributable to the warming water temperatures and prolonged thermal stratification, which might affect biogeochemical processes and exchange fluxes between the hypolimnion and bottom sediments. These potential changes may present challenges for water resource management under future conditions of climate change and population growth.

Climate and land use changes impact catchment hydrology and water quality (WQ), yet few studies have investigated the amount of land use changes required to meet specific WQ targets under future climate projections. The aim of this study was to determine streamflow and nutrient load responses to future land use change (LUC) and climate change scenarios. We hypothesized that (1) increasing forest coverage would decrease nutrient loads, (2) climate change, with higher temperatures and more intense storms, would lead to increased flow and nutrient loads, and (3) LUC could moderate potential nutrient load increases associated with climate change. We tested these hypotheses with the Soil and Water Assessment Tool (SWAT), which was applied to a lake catchment in New Zealand, where LUC strategies with afforestation are employed to address lake WQ objectives. The model was calibrated from 2002 to 2005 and validated from 2006 to 2010 using measured streamflow (Q) and total nitrogen (TN), total phosphorus (TP), nitrate (NO3-N), and ammonium (NH4-N) concentrations of three streams in the catchment. The model performance across the monitored streams was evaluated using coefficient of determination (R2) and Nash–Sutcliffe efficiency (NSE) metrics to provide a basis for model projections. Future scenarios incorporated LUC and climate change (CC) based on the Representative Concentration Pathway 8.5 and were compared to the baseline streamflow and WQ indicators. Consistent with our hypotheses, Q, TN, and TP loads were predicted to decrease with afforestation. Specifically, afforestation of 1.32 km2 in one of the monitored stream sub-catchments (subbasin 3), or 8.8% of the total lake catchment area, would result in reductions of 11.9, 26.2, and 17.7% in modeled annual mean Q, TN, and TP loads, respectively. Furthermore, when comparing simulations based on baseline and projected climate, reductions of 13.6, 22.8, and 19.5% were observed for Q, TN, and TP loads, respectively. Notably, the combined implementation of LUC and CC further decreased Q, TN, and TP loads by 20.2, 36.7, and 28.5%, respectively. This study provides valuable insights into the utilization of LUC strategies to mitigate nutrient loads in lakes facing water quality challenges, and our findings could serve as a prototype for other lake catchments undergoing LUC. Contrary to our initial hypotheses, we found that higher precipitation and temperatures did not result in increased flow and nutrient loading.

Ammonium (NH4-N) produces a paradoxical effect on submersed macrophytes because it is not only the preferred nitrogen source for the growth of plants but also threatens the growth of plants at high concentration. Whether short-term and small-scale physiological toxicity experiments at an individual level can reflect the effects of high ammonium on populations of submersed macrophytes in natural conditions is still unclear. In this study, an 18-month experiment was conducted in six 600 m2 ponds subjected to different levels of ammonium loading. The effects of high ammonium on populations of canopy-forming Myriophyllum spicatum and rosette-forming Vallisneria natans were explored. The results showed that M. spicatum and V. natans populations can develop high cover and height at high ammonium concentration (7 mg/L) at short-term exposures, and V. natans may be tolerant to 18 mg/L ammonium concentration. However, the cover of M. spicatum and the height of both species were inhibited at 2.4 mg/L at long-term exposures. The height of M. spicatum was two to six times higher than that of V. natans across all treatments and control by the end of the experiment, and the cover of M. spicatum was 7–11 times higher than that of V. natans in most NH4-N loading treatments, except the cover of M. spicatum in the highest NH4-N loading treatment with 18 mg/L NH4-N. The rosette-forming V. natans resists ammonium stress by slow growth (shoot elongation) to reduce consumption, while canopy-forming species resist ammonium stress by shoot elongation and canopy development to capture light. Although increasing ammonium concentration may induce severe stress on M. spicatum, the morphological characteristics of this species may, to some extent, release the plants from this stress. Our present study indicates that the negative effects of ammonium stress on the development of populations increased with exposure duration, and the submersed macrophyte community with stronger ability for light capture and dispersal may resist high ammonium stress. Nevertheless, in strongly ammonium-enriched systems, competition and succession cannot be neglected.

Abstract Globally, lake surface water temperatures have warmed rapidly relative to air temperatures, but changes in deepwater temperatures and vertical thermal structure are still largely unknown. We have compiled the most comprehensive data set to date of long-term (1970–2009) summertime vertical temperature profiles in lakes across the world to examine trends and drivers of whole-lake vertical thermal structure. We found significant increases in surface water temperatures across lakes at an average rate of + 0.37 °C decade−1, comparable to changes reported previously for other lakes, and similarly consistent trends of increasing water column stability (+ 0.08 kg m−3 decade−1). In contrast, however, deepwater temperature trends showed little change on average (+ 0.06 °C decade−1), but had high variability across lakes, with trends in individual lakes ranging from − 0.68 °C decade−1 to + 0.65 °C decade−1. The variability in deepwater temperature trends was not explained by trends in either surface water temperatures or thermal stability within lakes, and only 8.4% was explained by lake thermal region or local lake characteristics in a random forest analysis. These findings suggest that external drivers beyond our tested lake characteristics are important in explaining long-term trends in thermal structure, such as local to regional climate patterns or additional external anthropogenic influences.

Urban lakes are important natural assets but are exposed to multiple stressors from human activities. Submersed macrophytes, a key plant group that helps to maintain clear-water conditions in lakes, tend to be scarce in urban lakes, particularly when they are eutrophic or hypertrophic, and their loss is linked, in part, to impaired underwater light climate. We tested if enhancing the underwater light conditions using light-emitting diodes (LEDs) could restore submersed macrophytes in urban lakes. Twelve mesocosms (1000 L each) were each planted with tape grass (Vallisneria natans) and monitored over three months (22 August–7 November), using a control and three artificial light intensity treatments (10, 50, and 100 µmol m-2 s-1). Compared with the control, the high light treatment (100 µmol m-2 s-1) had higher leaf number, maximum leaf length, and average leaf length (3.9, 5.8, and 2.8 times, respectively). Shoot number, leaf number, leaf dry mass, root dry mass, and photosynthetic photon flux density in the high-light treatment were significantly greater than the control, but root length and phytoplankton chlorophyll a were not related to plant growth variables and were low in all treatments. Periphyton chlorophyll a increased significantly with the plant growth variables (i.e., shoot number, leaf number, and maximum leaf length) and was high in the light treatments but did not hamper the growth of the macrophytes. These results indicate that LED light supplementation enables the growth of V. natans under eutrophic conditions, at least in the absence of fish as in our experiment, and that the method may have potential as a restoration method in urban lakes. Lake-scale studies are needed, however, to fully evaluate LED light supplementation under natural conditions where other stressors (e.g., fish grazing) may need to be controlled for successful restoration of urban lakes.

Estuaries host unique biodiversity and deliver a range of ecosystem services at the interface between catchment and the ocean. They are also among the most degraded ecosystems on Earth. Freshwater flow regimes drive ecological processes contributing to their biodiversity and economic value, but have been modified extensively in many systems by upstream water use. Knowledge of freshwater flow requirements for estuaries (environmental flows or E-flows) lags behind that of rivers and their floodplains. Generalising estuarine E-flows is further complicated by responses that appear to be specific to each system. Here we critically review the E-flow requirements of estuaries to 1) identify the key ecosystem processes (hydrodynamics, salinity regulation, sediment dynamics, nutrient cycling and trophic transfer, and connectivity) modulated by freshwater flow regimes, 2) identify key drivers (rainfall, runoff, temperature, sea level rise and direct anthropogenic) that generate changes to the magnitude, quality and timing of flows, and 3) propose mitigation strategies (e.g., modification of dam operations and habitat restoration) to buffer against the risks of altered freshwater flows and build resilience to direct and indirect anthropogenic disturbances. These strategies support re-establishment of the natural characteristics of freshwater flow regimes which are foundational to healthy estuarine ecosystems.

Understanding the spatiotemporal trend of land cover (LC) change and its impact on humans and the environment is essential for decision making and ecosystem conservation. Land degradation generally accelerates overland flow, reducing soil moisture and base flow recharge, and increasing sediment erosion and transport, thereby affecting the entire basin hydrology. In this study, we analyzed watershed-scale processes in the study area, where agriculture and natural shrub land are the dominant LCs. The objective of this study was to assess the time series and spatial patterns of LCC using remotely-sensed data from 1973 to 2018, for which we used six snapshots of satellite images. The LC distribution in relation to watershed characteristics such as topography and soils was also evaluated. For LCC detection analysis, we used Landsat datasets accessed from the United States Geological Survey (USGS) archive, which were processed using remote sensing and Geographic Information System (GIS) techniques. Using these data, four major LC types were identified. The findings of an LC with an overall accuracy above 90% indicates that the area experienced an increase in agricultural LC at the expense of other LC types such as bushland, grazing land, and mixed forest, which attests to the semi-continuous nature of deforestation between 1973 and 2018. In 1973, agricultural land covered only 10% of the watershed, which later expanded to 48.4% in 2018. Bush, forest, and grazing land types, which accounted for 59.7%, 16.7%, and 13.5% of the watershed in 1973, were reduced to 45.2%, 2.3%, and 4.1%, respectively in 2018. As a result, portions of land areas, which had once been covered by pasture, bush, and forest in 1973, were identified as mixed agricultural systems in 2018. Moreover, spatial variability and distribution in LCC is significantly affected by soil type, fertility, and slope. The findings showed the need to reconsider land-use decision tradeoffs between social, economic, and environmental demands.

Wetlands can increase resilience to extreme climatic events and have a key role in protection and water quality improvement in coastal ecosystems. Studies in tropical coastal wetlands at a catchment scale are scarce, and most work has been undertaken on small, temperate wetlands. In this study, we tested whether natural coastal wetlands in a tropical catchment (Tully-Murray, Queensland, Australia) could ameliorate nitrogen (N) exported to the Great Barrier Reef during a flood event. We measured denitrification rates in different types of coastal wetlands (mangroves, saltmarshes, waterbodies with macrophytes, and floodplain wetlands dominated by Melaleuca spp.) to assess their potential contribution to N losses during the 6-day duration of a flood in March 2018. Denitrification potential was variable across the landscape, and we identified “hotspots” in sub-catchments with high NO3--N concentrations (0.4–0.6 mg L−1) and large areas of wetlands (>800 ha, >40% of the sub-catchment). These hotspots can denitrify up to 10 t of NO3--N per day during a flood. We used our measured denitrification rates to provide input parameters for a model that includes the main biogeochemical processes affecting N transformations within wetlands (nitrification, denitrification, plant uptake, sedimentation, anammox, and mineralization), and accounts for transport via the duration, depth, and flow of water. Model simulations of a sub-catchment of the Tully-Murray indicate that flood inundation of large areas of natural wetlands (>40% of the sub-catchment area) could potentially remove 70% of the incoming NO3--N load in the first 24 h of the flood. The management and restoration of coastal tropical wetlands could play a critical role in sustaining the health of coastal ecosystems through water quality improvement.

Australian ecosystems, particularly wetlands, are facing new and extreme threats due to climate change, land use, and other human interventions. However, more fundamental knowledge is required to understand how nutrient turnover in wetlands is affected. In this study, we deployed a mechanistic biogeochemical model of carbon (C), nitrogen (N), and sulfur (S) cycles at 0.25∘× 0.25∘ spatial resolution across wetlands in Australia. Our modeling was used to assess nutrient inputs to soil, elemental nutrient fluxes across the soil organic and mineral pools, and greenhouse gas (GHG) emissions in different climatic areas. In the decade 2008–2017, we estimated an average annual emission of 5.12 Tg-CH4, 90.89 Tg-CO2, and 2.34 × 10−2 Tg-N2O. Temperate wetlands in Australia have three times more N2O emissions than tropical wetlands as a result of fertilization, despite similar total area extension. Tasmania wetlands have the highest areal GHG emission rates. C fluxes in soil depend strongly on hydroclimatic factors; they are mainly controlled by anaerobic respiration in temperate and tropical regions and by aerobic respiration in arid regions. In contrast, N and S fluxes are mostly governed by plant uptake regardless of the region and season. The new knowledge from this study may help design conservation and adaptation plans to climate change and better protect the Australian wetland ecosystem.

Microcystins are cyclic peptides produced by multiple cyanobacterial genera. After accumulation in the liver of animals they inhibit eukaryotic serine/threonine protein phosphatases, causing liver disease or death. Accurate detection/quantification of microcystins is essential to ensure safe water resources and to enable research on this toxin. Previous methodological comparisons have focused on detection and extraction techniques, but have not investigated the commonly used biomass enrichment steps. These enrichment steps could modulate toxin production as recent studies have demonstrated that high cyanobacterial cell densities cause increased microcystin levels. In this study, three microcystin-producing strains were processed using no cell enrichment steps (by direct freezing at three temperatures) and with biomass enrichment (by centrifugation or GF/C filtration). After extraction, microcystins were analyzed using liquid chromatography-tandem mass spectrometry. All processing methods tested, except GF/C filtration, resulted in comparable microcystin quotas for all strains. The low yields observed for the filtration samples were caused by adsorption of arginine-containing microcystins to the GF/C filters. Whilst biomass enrichment did not affect microcystin metabolism over the time-frame of normal sample processing, problems associated with GF/C filtration were identified. The most widely applicable processing method was direct freezing of samples as it could be utilized in both field and laboratory environments.

Microcystins are hepatotoxic cyclic peptides produced by several cyanobacterial genera worldwide. In 2008, our research group identified eight new glycine-containing microcystin congeners in two hydro-terrestrial mat samples from the McMurdo Dry Valleys of Eastern Antarctica. During the present study, high-resolution mass spectrometry, amino acid analysis and micro-scale thiol derivatization were used to further elucidate their structures. The Antarctic microcystin congeners contained the rare substitution of the position-1 ᴅ-alanine for glycine, as well as the acetyl desmethyl modification of the position-5 Adda moiety (3S-amino-9S-methoxy-2S,6,8S-trimethyl-10-phenyldeca-4E,6E-dienoic acid). Amino acid analysis was used to determine the stereochemistry of several of the amino acids and conclusively demonstrated the presence of glycine in the microcystins. A recently developed thiol derivatization technique showed that each microcystin contained dehydrobutyrine in position-7 instead of the commonly observed N-methyl dehydroalanine.

Microcystins (MCs) are cyclic peptides produced by cyanobacteria, which can be harmful to humans and animals when ingested. Differences in the coding of the non‑ribosomal peptide synthetase/polyketide synthase enzyme complex responsible for microcystin production have resulted in more than 100 microcystin variants being reported to date. The microcystin diversity of Microcystis CAWBG11 was investigated using matrix-assisted laser desorption/ionization-time of flight mass spectrometry and liquid chromatography-mass spectrometry. This revealed that CAWBG11 simultaneously produced 21 known microcystins and six new congeners: [Asp3] MC-RA, [Asp3] MC-RAba, [Asp3] MC-FA, [Asp3] MC-WA, MC-FAba and MC-FL. The new congeners were putatively characterized by tandem mass spectrometry and chemical derivatization. A survey of the microcystin congeners produced by 49 cyanobacterial strains documented in scientific literature showed that cyanobacteria generally produce four microcystin congeners, but strains which produce up to 47 microcystin congeners have been reported. Microcystis CAWBG11 (which produces at least 27 congeners) was positioned in the top ten percentile of the strains surveyed, and showed fluidity of the amino acids incorporated into both position two and position four.

Understanding of colony specific properties of cyanobacteria in the natural environment has been challenging because sampling methods disaggregate colonies and there are often delays before they can be isolated and preserved. Microcystis is a ubiquitous cyanobacteria that forms large colonies in situ and often produces microcystins, a potent hepatotoxin. In the present study a new cryo-sampling technique was used to collect intact Microcystis colonies in situ by embedding them in a sheet of ice. Thirty-two of these Microcystis colonies were investigated with image analysis, liquid chromatography-mass spectrometry, quantitative polymerase chain reaction and high-throughput sequencing to assess their volume, microcystin quota and internal transcribed spacer (ITS) genotype diversity. Microcystin quotas were positively correlated to colony volume (R2 = 0.32; p = 0.004). Individual colonies had low Microcystis ITS genotype diversity and one ITS operational taxonomic unit predominated in all samples. This study demonstrates the utility of the cryo-sampling method to enhance the understanding of colony-specific properties of cyanobacteria with higher precision than previously possible.

Microcystins are cyclic peptides produced by cyanobacteria, which can be harmful to humans and animals when ingested. Eight of the (more than) 90 microcystin variants presently characterized, contain the amino acid tryptophan. The well-researched oxidation products of tryptophan; kynurenine, oxindolylalanine, and N-formylkynurenine, have been previously identified in intact polypeptides but microcystin congeners containing oxidized tryptophan moieties have not been reported. Liquid chromatography-tandem mass spectrometric analysis of an extract of Microcystis CAWBG11 led to the tentative identification of two new tryptophan-containing microcystins (MC‑WAba and MC-WL), as well as eight other microcystin analogs containing kynurenine, oxindolylalanine and N‑formylkynurenine (Nfk). Investigation of one of these congeners (MC‑NfkA) by nuclear magnetic resonance spectroscopy was used to verify the presence of Nfk in the microcystin. Liquid chromatography-mass spectrometry analysis of a tryptophan oxidation experiment demonstrated that tryptophan-containing microcystins could be converted into oxidized tryptophan analogs and that low levels of oxidized tryptophan congeners were present intracellularly in CAWBG11. MC-NfkR and MC-LNfk were detected in standards of MC-WR and MC-LW, indicating that care during storage of tryptophan-containing microcystins is required.
