Aim The COVID-19 pandemic has brought innumerable challenges to surgical training, especially with regard to theatre opportunities for core surgical trainees. This study aims to evaluate the implementation of training-oriented key performance indicators (KPIs) in supporting trainees to meet the Joint Committee on Surgical Training (JCST) quality indicators for core surgical training. Method A six-month long closed-loop quality improvement project. Two KPIs were developed following discussion with trainees and departmental leads, evaluation of the rota, and review of the JCST quality indicators: theatre/normal-working-day-attendance (T/A) ratio and missed-theatre-sessions/normal-working-day-attendance (M/A) ratio. Our calculations projected a T/A ratio of 1.00 and M/A ratio of 0.20 to be fully compliant with the JCST quality indicators. Data were collected prospectively from eight trainees, who provided feedback on training opportunities and allocation equitability. Results Following implementation of the KPIs, there was a significant increase in the mean T/A ratio from 0.70 (range=0.55–0.82) to 1.00 (range=0.95–1.17) (p=0.03), and a significant decrease in the mean M/A ratio from 0.55 (range=0.45–0.70) to 0.20 (range=0.11–0.22) (p=0.01). There were significant improvements in trainee satisfaction with training opportunities from 50% to 85% (p=0.02) and allocation equitability from 66% to 95% (p=0.01). Conclusions The development and implementation of training oriented KPIs led to increased allocation and fewer missed theatre sessions, enabling trainees to meet the JCST quality indicators. Furthermore, our data demonstrate smaller variances between trainees, thus, highlighting greater equitability of training opportunities. These findings are supported by the improved trainee satisfaction with training opportunities and allocation equitability reported.

A new species of the genus Tanna Distant, 1905 (Hemiptera: Cicadidae), Tanna crassa sp. nov., is described from Vietnam. Some taxonomic changes are made in Tanna. Tanna simultaneous (Chen, 1940) syn. nov. is synonymized with Tanna abdominalis (Kato, 1938). Tanna obliqua Liu, 1940 stat. rev. is resurrected from the junior synonym of Tanna auripennis Kato, 1930. Tanna shensiensis (Sanborn, 2006) is transferred to Aetanna Lee, 2014 to become Aetanna shensiensis (Sanborn, 2006) comb. nov. Tanna bakeri Moulton, 1923 is transferred to Purana Distant, 1905 to become Purana bakeri (Moulton, 1923) comb. nov. BackgroundAmong protozoan parasites in the genus Babesia, Babesia bigemina is endemic and widespread in the East African region while the status of the more pathogenic Babesia bovis remains unclear despite the presence of the tick vector, Rhipicephalus microplus which transmits both species. Recent studies have confirmed the occurrence of R. microplus in coastal Kenya and although B. bovis DNA has previously been detected in cattle blood in Kenya, no surveillance has been done to establish its prevalence. This study therefore investigated the occurrence of B. bovis in cattle in Kwale County, Kenya, where R. microplus is present in large numbers. MethodA quantitative species-specific multiplex Taqman PCR assay targeting two Babesia bovis genes, 18S ribosomal RNA and mitochondrially-encoded cytochrome b and Babesia bigemina cytochrome b gene was used to screen 506 cattle blood DNA samples collected from Kwale County for presence of Babesia parasite DNA. ResultsA total of 131 animals (25.8%) were found to have bovine babesiosis. Of these, 87 (17.2%) were positive for B. bovis while 70 (13.8%) had B. bigemina while 26 (5.1%) were observed to be co-infected with both Babesia species. A total of 61 animals (12.1%) were found to be infected with B. bovis parasites only, while 44 animals (8.7%) had B. bigemina only. B. bovis infections were detected in 11 sites while B. bigemina was detected in 9 out of the 12 sites sampled in three Kwale sub-Counties.ConclusionThese findings reveal high prevalence of pathogenic B. bovis in a Kenyan area cutting across a busy transboundary livestock trade route with neighboring Tanzania. The Babesia multiplex assay used in this study is specific and can detect and differentiate the two Babesia species and should be used for routine B. bovis surveillance to monitor the spread and establishment of the pathogen in other African countries where B. bigemina is endemic. Moreover, these findings reveal a potential threat of fatal babesiosis in the absence of endemic stability in the local cattle populations.

A sub-set of 29 B. bovis real-time PCR-positive samples were further amplified using a B. bovis-specific spherical body protein-4 (SBP-4) nested PCR and the resulting products sequenced to confirm the presence of B. bovis. Results A total of 131 animals (25.8%) were found to have bovine babesiosis based on real-time PCR. Twenty-four SBP4 nucleotide sequences obtained matched to B. bovis with a similarity of 97–100%. Of 131 infected animals, 87 (17.2%) were positive for B. bovis while 70 (13.8%) had B. bigemina and 26 (5.1%) were observed to be co-infected with both Babesia species. A total of 61 animals (12.1%) were found to be infected with B. bovis parasites only, while 44 animals (8.7%) had B. bigemina only. Authoritative film archival texts demand documentation of film restoration projects. Film archives often produce overly technical internal documentation, and although researchers occasionally publish film restoration reports, the ratio of reports to restoration projects is skewed in favour of films that stir academic debate. Film scholars can be recruited to engage with archives to write reports for lesser-known films, thereby increasing and improving film restoration documentation. However, the relationship between film scholars and film archivists must be improved, as these groups tend not to interact during the film restoration process. Based on a survey of 30 silent film restoration reports, a guide is provided to help film scholars approach archives and archivists to attain information for those reports. The guide is applied to Gräfin Küchenfee (1918), a silent film restored by EYE Filmmuseum for Il Cinema Ritrovato, to showcase usage of the guide and produce a report on the film’s restoration.

Thermal energy harvesting from natural resources and waste heat is becoming critical due to ever-increasing environmental concerns. However, so far, available thermal energy harvesting technologies have only been able to generate electricity from large temperature gradients. Here, we report a fundamental breakthrough in low-grade thermal energy harvesting and demonstrate a device based on the thermomagnetic effect that uses ambient conditions as the heat sink and operates from a heat source at temperatures as low as 24 °C. This concept can convert temperature gradients as low as 2 °C into electricity while operating near room temperature. The device is found to exhibit a power density (power per unit volume of active material) of 105 μW cm−3 at a temperature difference of 2 °C, which increases to 465 μW cm−3 at a temperature difference of 10 °C. The power density increases by 2.5 times in the presence of wind with a speed of 2.0 m s−1. This advancement in thermal energy harvesting technology will have a transformative effect on renewable energy generation and in reducing global warming.

Ovine Johne's disease is a chronic debilitating disease of sheep caused by Mycobacterium avium subsp. paratuberculosis (Mptb) which results in diarrhoea, emaciation and mortalities in infected animals. Vaccination with Gudair® has been a key strategy for controlling the disease in Australia since its approval in 2002. Previous research conducted in Australia has demonstrated that the vaccine is quite effective in reducing sheep mortalities. While some farms have also been successful in reducing the prevalence of the disease in their flocks to undetectable levels, sheep in other flocks continue to shed Mptb in faeces even after an ongoing vaccination program . This study was conducted to investigate management, husbandry and biosecurity factors associated with paratuberculosis infection in Gudair® vaccinated sheep flocks in Australia. We enrolled 64 sheep farmers and interviewed them to obtain information about their management and biosecurity practices. Pooled faecal samples were collected from sheep at each farm and cultured to create two outcome variables: Mptb positive (yes/no) and disease prevalence level (nil, < 1 %, ≥ 1 %). Binary and ordinal logistic regression analyses were conducted to evaluate the association of management, husbandry and biosecurity factors with these outcome variables. Farms were more likely to have Mptb positive sheep and a higher disease prevalence in their flocks if they: (a) provided supplementary feed on the ground (instead of in a trough); (b) had a greater number of neighbours with sheep; and (c) had introduced rams from a greater number of sources. The results suggest the effectiveness of Gudair® vaccination to control OJD can be improved if sheep producers maintain other risk management strategies and biosecurity practices. Extension agencies should advise farmers not to relax their biosecurity practices and to purchase rams from only low-risk sources, even if they are continuing to vaccinate their flocks.

Two new friedelane triterpenoids, 20,20-dimethyl-3,27-dioxo-(D:A)-friedo-olean-27,22α-lactone (20,20-dimethylglaucalactone) (1) and 3α-ethanoyloxy-30-nor-(D:A)- friedo-olean-20(29)-en-27,22α-lactone (3-acetoxyglaucalactone) (2) and four known compounds (3–6) were isolated from the methanolic extract of the roots of Caloncoba glauca (P. Beauv.) Gild (Flacourtiaceae). Their structures were determined by comprehensive spectroscopic analyses (1D and 2D NMR in addition to HRESI-MS). The relative configuration of the 22-oxymethine was determined from the NOESY spectrum. The cardiotonic effects of the crude extract and isolated compounds were tested. The results suggest that the crude extract and 3-acetoxyglaucalactone (2) induced an interesting cardiotonic activity by exerting positive inotropic and chronotropic effects, which might be derived from the direct stimulation of the β-adrenergic pathway through β1-adrenergic receptors or opening of the calcium channel.

The current study evaluated the cardioprotective effect of the aqueous extract of seeds ofDatura metel(AESDM) on the acute cardiotoxicity induced with doxorubicin in the Wistar rats. Indeed, 30 rats have been randomized then, divided into 6 groups of 5 animals each: 3 control groups (normal, negative and positive) and 3 test groups (AESDM 100, 200 and 300 mg/kg). These animals received orally, six consecutive days, 1 mL/200 g distilled water (normal and negative control), 100 mg/kg Vitamin-E (positive control), AESDM at 100, 200 and 300 mg/kg (test groups). On the seventh day of treatment, animals in the different groups excepted the normal control received a single intraperitoneal injection of 15 mg/kg doxorubicin followed respective treatments up to the tenth day of the trial period. Serum parameters (ASAT/ALAT), biochemical markers of oxidative stress (MDA, SOD, CAT, GSH) and lipid parameters (total cholesterol, triglycerides, HDL and LDL) were evaluated. Histological sections of animal hearts were prepared. The results of this study showed that treatment with AESDM resulted in a significant decrease (P ˂ 0.001) in MDA, triglycerides and LDL-cholesterol levels, ASAT/ALAT activity and a significant increase (P ˂ 0.001) in HDL-cholesterol concentration and SOD, CAT and GSH activities, compared to the negative control. Histological observation revealed that AESDM protected the heart from doxorubicin-induced damage. In conclusion, AESDM would have a cardioprotective effect that could be attributed to its antioxidant potential.&nbsp

Background: In regions with endemic Theileria orientalis, recovered cattle are carriers, leaving newborn calves and introduced stock as the most susceptible groups to develop clinical disease after tick infestation. Methods: To gain information on the kinetics of infestation and development and effects of parasitosis by virulent Theilerial genotypes ikeda and chitose and the “benign” genotype, buffeli, this study sampled at total of 150 calves from 3 farms in Dorrigo, Australia, a region with multiple genotypes of the parasite. In addition, 30 introduced beef weaners were bled and weighed for 6 months after arrival. Results: In both cohorts, parasitosis with the T.orientalis ikeda and chitose (genotypes 1 & 2) occurred within 3-4 weeks in >95% of the groups, with maximal gene copies generated around 5-7 weeks after birth or introduction, before declining. In contrast, parasitosis with T.orientalis buffeli (genotype 3), increased slowly, with around 80% of the population testing positive by PCR after 4 months. The parasitosis with the buffeli genotype did not exceed 40000 gene copies /ul and was similar to those exhibited by ikeda and chitose genotypes following the “first wave of parasitaemia” as cattle entered the carrier state. In the 30 introduced weaners, the early parasitosis with T.orientalis ikeda and chitose also caused significant reductions in haematocrit and incurred around 20Kg loss in weight gain; the latter had not been recovered by 6 months after introduction. Conclusions: The results support previous findings in other endemic regions of Theileriosis, but imply that amelioration of the early high levels of parasitosis by the virulent genotypes could reduce the initial impact of Theileriosis on production, which appears much less affected once animals enter the carrier state.

Background: An investigation aimed to confirm whether immunisation with the “benign” buffeli genotype of Theileria orientalis could reduce the parasitosis of the virulent ikeda genotype. Methods: Calves were inoculated intravenously or subcutaneously with bovine blood containing merozoites of T.orientalis buffeli. When recipients became positive, they and control animals were challenged with unfed nymphs of Haemaphysalis longicornis ticks infested as larvae with T.orientalis ikeda. Results: All calves became positive for the challenge within 12 days after tick application. In the immunised calves, the first wave of parasitosis with T.orientalis ikeda from 4-6 weeks was reduced significantly by >80% before the infestation declined into the carrier state by 9 weeks. Conclusions: The results confirm the field experience with theileriosis in endemic zones where the carrier state appears to prevent clinical disease despite repeated, seasonal tick infestations with virulent genotypes of the parasite. This method offers a means to reduce the severity of the first wave of theilerial parasitosis after tick challenge and recover associated production losses.

Background The tick vector Rhipicephalus microplus which transmits Babesia spp. and rickettsial pathogens has not been reported in Kenya since 1998. More recently, the pathogenic Babesia bovis has been detected in cattle blood DNA. The status of R. microplus in Kenya remains unknown. This study employed morphological and molecular tools to characterize R. microplus originating from Kenya and assess the genetic relationships between Kenyan and other African R. microplus genotypes. Methods Ticks were collected in south-eastern Kenya (Kwale County) from cattle and characterized to investigate the existence of R. microplus. Genetic and phylogenetic relationships between the Kenyan and other annotated R. microplus reference sequences was investigated by analysis of the cytochrome c oxidase subunit 1 (cox1) gene. To further characterize Kenyan ticks, we generated low coverage whole genome sequences of two R. microplus, one R. decoloratus and R. appendiculatus. A B. bovis specific TaqMan probe qPCR assay was used to detect B. bovis in gDNA from R. microplus ticks. Results Occurrence of R. microplus was confirmed in Kwale County, Kenya. The Kenyan R. microplus cox1 sequences showed very high pairwise identities (> 99%) and clustered very closely with reference African R. microplus sequences. We found a low genetic variation and lack of geographical sub-structuring among the African cox1 sequences of R. microplus. Four complete mitochondrial (mt) genomes for two R. microplus, one R. decoloratus and one R. appendiculatus were assembled from next generation sequence data. The mitochondrial genome sequences of the two Kenyan R. microplus ticks clustered closely with reference genome sequences from Brazil, USA, Cambodia and India forming R. microplus Clade A. No B. bovis was detected in the Kwale R. microplus DNA. Conclusions These findings confirm the presence of R. microplus in Kenya and suggest that R. microplus Clade A is prevalent in cattle in sub-Saharan Africa. These and other recent findings of widespread occurrence of R. microplus in Africa provide a strong justification for urgent surveillance to determine and monitor the spread of R. microplus and vector competence of Boophilus ticks for B. bovis in Africa, with the ultimate goal of strategic control.

This review addresses the research landscape regarding vaccines against scour worms, particularly Trichostrongylus spp. and Teladorsagia circumcincta. The inability of past research to deliver scour-worm vaccines with reliable and reproducible efficacy has been due in part to gaps in knowledge concerning: (i) host-parasite interactions leading to development of type-2 immunity, (ii) definition of an optimal suite of parasite antigens, and (iii) rational formulation and administration to induce protective immunity against gastrointestinal nematodes (GIN) at the site of infestation. Recent 'omics' developments enable more systematic analyses. GIN genomes are reaching completion, facilitating "reverse vaccinology" approaches that have been used successfully for the Rhipicephalus australis vaccine for cattle tick, while methods for gene silencing and editing in GIN enable identification and validation of potential vaccine antigens. We envisage that any efficacious scour worm vaccine(s) would be adopted similarly to "Barbervax™" within integrated parasite management schemes. Vaccines would therefore effectively parallel the use of resistant animals, and reduce the frequency of drenching and pasture contamination. These aspects of integration, efficacy and operation require updated models and validation in the field. The conclusion of this review outlines an approach to facilitate an integrated research program.

Background The tick vector Rhipicephalus (Boophilus) microplus which transmits Babesia and rickettsial pathogens has not been reported in Kenya since 1998. More recently, the pathogenic Babesia bovis has been detected in cattle blood DNA. The status of R. microplus in Kenya remains unknown. This study employed morphological and molecular tools to characterize R. microplus originating from Kenya and assess the genetic relationships between Kenyan and other African R. microplus genotypes. Methods Morphological identification and differentiation of 35 Kenyan and 11 reference tick specimens was implemented by following standard tick reference keys and identification guides. Genetic and phylogenetic relationships between the Kenyan and other annotated R. microplus reference sequences was investigated by analysis of the cytochrome c oxidase subunit I (COI) gene. The complete mitochondrial (mt) genome of four tick specimens which included two R. microplus was assembled from whole genome data to further characterize the Kenyan ticks. A B. bovis specific Taqman probe qPCR assay was used to detect B. bovis in gDNA from R. microplus ticks. Results Of the 35 Kwale tick specimens analysed, 23 were confirmed to be R. microplus by both morphology and molecular characterizations. The Kenyan R. microplus COI sequences showed very high pairwise identities (>99%) and clustered very closely with reference African sequences. We also found a low differentiation and lack of geographical sub-structuring among the African COI sequences. The mitochondrial genome sequences of the two Kenyan R. microplus ticks also clustered closely with reference genome sequences from Brazil, USA, Cambodia and India. No B. bovis was detected in the Kwale R. microplus DNA. Conclusions These findings confirm the presence of R. microplus in Kenya and suggest that a common undifferentiated genotype is prevalent in cattle in Africa. These and other recent findings of widespread occurrence of R. microplus in Africa provide a strong justification for urgent surveillance to determine and monitor the spread of R. microplus and vector competence of Rhipicephalid ticks for B. bovis in Africa, with the ultimate goal of strategic control.

Highlights • Conditions for the optimal generation interval for the “bush tick”, Haemaphysalis longicornis, in Australia have been determined. • Stabilates containing infective sporozoites of Theileria orientalis ikeda were produced from ground-up ticks (GUTS) and salivary glands of H.longicornis. • Sporozoites of Theileria orientalis required around 3–5 days of tick feeding to mature in salivary glands before transmission occurred (like T.parva). • GUTS and salivary gland stabilates produced detectable infections with T.orientalis in naive calves around 3–4 weeks after inoculation. • This study confirms the final host status of H.longicornis for T.orientalis in Australia, facilitating research for prophylaxis and vector control., The intracellular protozoal parasite Theileria orientalis ikeda has rapidly spread across South-eastern Australia since 2006, causing deaths and production losses in cattle. The 3-host “bush tick” Haemaphysalis longicornis (Neumann) appears the principal biological vector in the endemic regions. To generate sufficient numbers of ticks to produce stabilate for infection to confirm vector competency and for acaricide trials, the optimal conditions and stage-specific intervals for the generational life-cycle of H.longicornis was defined on two dogs and two steers. To determine whether H.longicornis was a definitive host for Theileria orientalis, nymphal stages were fed on a steer infected with T.orientalis and moulted adults were permitted to feed for 3 days on an uninfected calf prior to harvest. Subsequent detection of infection after inoculation of four naïve calves with stabilate produced from ground-up adult ticks or dissected salivary glands confirmed H.longicornis as one final (definitive) host for T.orientalis in Australia.

At present diagnosis of true resistance and determination of drug efficacy in Fasciola hepatica infection rely solely on terminal experiments. The coproantigen ELISA (cELISA) has been reported previously as a sensitive and specific tool appropriate to detect treatment failure, and potentially drug resistance. Two studies were conducted to determine whether the cELISA was appropriate for on-farm efficacy and resistance testing in Australian Merino sheep. In Study 1 sheep were infected orally with 50 F. hepatica metacercariae on three occasions, twelve, six and two weeks prior to a single flukicide treatment with triclabendazole, closantel or albendazole. Sheep were sampled weekly for a further seven weeks prior to necropsy. Following effective treatment, no faecal antigen was detected from 1 week. When immature stages (≤6 weeks) survived treatment, coproantigen reappeared from 6 weeks post-treatment. Therefore, cELISA conducted 1-4 weeks after treatment will demonstrate obvious treatment failure against adult F. hepatica, but is not sufficiently sensitive to detect survival of immature fluke until these reach maturity. In study 2, fluke burdens of sheep necropsied 13 weeks post single infection were compared to fecal worm egg counts (FWEC) and cELISA at necropsy. Regression analysis demonstrated that cELISA correlated strongly with fluke burden, whilst FWEC correlated weakly with cELISA. The correlation between FWEC and fluke burden was also weak, although stronger than that of FWEC with cELISA. The cELISA is an appropriate tool for monitoring effectiveness of treatments against Fasciola hepatica if an adult infection is present, however when immature stages of the parasite are present it is not as reliable. Where immature parasites are present it is recommended that initial cELISA be followed with a secondary cELISA at least 6 weeks after treatment to ensure resistance to immature stages is detected. Further testing is justified for monitoring the effectiveness of control programs by detecting adult populations that have survived a treatment regime.

The results confirm the previous studies on immunity to T. sergenti in Japan, and field experience with theileriosis in endemic zones where the carrier state appears to prevent clinical disease despite repeated, seasonal tick infestations with virulent genotypes of the parasite. This method offers a means to reduce the severity of the first wave of theilerial parasitaemia after tick challenge and possibly recover associated production losses.

Integrated parasite management (IPM) for pests, pathogens and parasites involves reducing or breaking transmission to reduce the impact of infection or infestation. For Theileria orientalis, the critical impact of infection is the first wave of parasitaemia from the virulent genotypes, Ikeda and Chitose, associated with the sequelae from the development of anaemia. Therefore, current control measures for T. orientalis advocate excluding the movement of naïve stock from non-endemic regions into infected areas and controlling the tick Haemaphysalislongicornis, the final host. In Australia, treatment of established infection is limited to supportive therapy. To update and expand these options, this review examines progress towards prevention and therapy for T. orientalis, which are key elements for inclusion in IPM measures to control this parasite.

Background In regions with endemic Theileria orientalis, recovered cattle are carriers, leaving newborn calves and introduced stock as the most susceptible groups to develop clinical disease after tick infestation with parasite transmission. Methods To gain information on the kinetics of infection and development and effects of theileriosis caused by virulent ikeda and chitose genotypes and the “benign” buffeli genotype of T.orientalis, this study sampled a total of 134 calves from 3 farms in Dorrigo, Australia, a region with multiple genotypes of the parasite. In addition, 30 introduced beef weaners were bled and weighed for 6 months after arrival. Results In both cohorts, parasitaemia with the ikeda and chitose (genotypes 1 & 2) of T.orientalis was detectable by PCR within 3–4 weeks in >95 % of the groups, with maximal gene copies generated around 5–7 weeks after birth or introduction, before declining. In contrast, parasitaemias of T.orientalis buffeli (genotype 3), increased slowly, with around 80 % of the population testing positive by PCR after 4 months. The parasitaemias of the buffeli genotype did not exceed 40,000 gene copies /ul and were similar to those exhibited by ikeda and chitose genotypes following the “first wave of parasitaemia” as cattle entered the carrier state. In the 30 introduced weaners, the early infection with T.orientalis ikeda and chitose also caused significant reductions in packed cell volume (PCV) and incurred an estimated 20Kg loss in weight gain; the latter had not been recovered by 6 months after introduction. Conclusions The results support previous findings in other endemic regions of theileriosis, and imply that amelioration of the early high levels of parasitosis by the virulent genotypes could reduce the initial impact of theileriosis on production, which appears much less affected once animals enter the carrier state.

This paper provides descriptions of four new species of the genus Semia Matsumura, 1917 from Vietnam: Semia magna sp. nov., Semia spiritus sp. nov., Semia pallida sp. nov., and Semia albusequi sp. nov. A key to the 13 species of Semia is provided.

The cat flea (Ctenocephalides felis) is the most common flea species parasitising both domestic cats and dogs globally. Fleas are known vectors of zoonotic pathogens such as vector borne Rickettsia and Bartonella. This study compared cat fleas from domestic cats and dogs in New Zealand’s North and South Islands to Australian cat fleas, using the mitochondrial DNA (mtDNA) marker, cytochrome c oxidase subunit I and II (cox1, cox2). We assessed the prevalence of Rickettsia and Bartonella using genus specific multiplexed real-time PCR assays. Morphological identification confirmed that the cat flea (C. felis) is the most common flea in New Zealand. The examined fleas (n = 43) at cox1 locus revealed six closely related C. felis haplotypes (inter-haplotype distance 1.1%) across New Zealand. The New Zealand C. felis haplotypes were identical or near identical with haplotypes from southern Australia demonstrating common dispersal of haplotype lineage across both the geographical (Tasman Sea) and climate scale. New Zealand cat fleas carried Rickettsia felis (5.3%) and Bartonella clarridgeiae (18.4%). To understand the capability of C. felis to vector zoonotic pathogens, we determined flea cox1 and cox2 haplotype diversity with the tandem multiplexed real-time PCR and sequencing for Bartonella and Rickettsia. This enabled us to demonstrate highly similar cat fleas on cat and dog populations across Australia and New Zealand.

Despite the global distribution of the brown dog tick, Rhipicephalus sanguineus (Latreille, 1806) sensu lato (s.l.), limited information exists about their identity from the Arabian Peninsula. Ticks from free roaming urban dogs and dromedary camels in Riyadh, Saudi Arabia were morphologically identified, confirmed with scanning electron microscopy and characterised at mitochondrial DNA (cox1, 12S rDNA and 16S rDNA). A total of 186 ticks were collected from 65 free roaming dogs (n = 73) and 84 dromedary camels (n = 113). Morphologically, 5.9% (11/186) were R. sanguineus s.l. and Hyalomma spp. (93.5%, 174/186). From within R. sanguineus s.l., the presence of Rhipicephalus cf. camicasi Morel, Mouchet et Rodhain, 1976 (1 dog, 2 camels) and Rhipicephalus turanicus Pomerantsev, 1936 (1 camel) is reported. The examined R. cf. camicasi form a sister group to R. sanguineus s.l. tropical lineage at all DNA markers. Dogs were parasitised by Hyalomma dromedarii Koch, 1844 (n = 59), Hyalomma impeltatum Schulze et Schlottke, 1930 (n = 1), Hyalomma excavatum Koch, 1844 (n = 2), Hyalomma turanicum Pomerantsev, 1946 (n = 1) and Hyalomma rufipes Koch,1844 (n = 1). DNA from dog blood (n = 53) from Riyadh confirmed a low prevalence of canine vector-borne pathogens that does not exceed 5.7% for Babesia spp., Mycoplasma spp., Anaplasma platys, Hepatozoon canis and Ehrlichia canis using multiplexed tandem PCR (MT-PCR) and diagnostic PCR. Low prevalence of R. sanguineus s.l. on dogs likely contributed to the low level of canine vector-borne pathogens in Saudi Arabia. We demonstrate that dogs in the central Arabian Peninsula are more commonly parasitised by Hyalomma spp. than R. sanguineus s.l.

The brown dog tick Rhipicephalus sanguineus (Latreille, 1806) is the most widely distributed tick species globally. Throughout the world there are at least two divergent lineages on dogs that are traditionally grouped into what was known as R. sanguineus. The species R. sanguineus was recently redescribed using a neotype reported from countries with a temperate climate. The second lineage distributed in countries with primarily tropical climates is currently designated R. sanguineus s.l. tropical lineage. Here, we present a comprehensive genetic evaluation of Australian brown dog ticks from across the continent that complements the morphological study of R. sanguineus sensu Roberts (1965). A total of 294 ticks were collected from dogs around Australia ― including New South Wales, Queensland, the Northern Territory and Western Australia ― for morphological identification. All ticks were morphologically identified as R. sanguineus sensu Roberts (1965). DNA was isolated from a single leg from morphologically characterised individuals from New South Wales (n = 14), Queensland (n = 18), Northern Territory (n = 7) and Western Australia (n = 13), together with ticks from Fiji (n = 1) and the Seychelles (n = 1) for comparison with Australian ticks. The study revealed three cox1 haplotypes clustered only with R. sanguineus s.l. tropical lineage’. An updated distribution of R. sanguineus sensu Roberts (1965) is compared to the 1965 distribution. In the Australian context, R. sanguineus s.l. has appeared in north-western New South Wales but remains absent from coastal New South Wales. Despite both temperate and tropical climates being present in Australia, only R. sanguineus s.l. tropical lineage was found. The evidence does not support the presence of the strictly defined brown dog tick, R. sanguineus by Nava et al. (2018) in Australia, because the examined ticks are genetically and morphologically distinct. We recommend using the term brown dog tick, R. sanguineus sensu Roberts (1965) for specimens from Australia.

Aim. The effects of the water extract of the leaves of Euphorbia Poissonii (Euphorbiacea) on frog heart contraction and its possible mechanism action were investigated. Method. By using the Langerdhorff model of the isolated frog heart preparation, the strength and frequency of heart contractions as well as cardiac output were measured under perfusion with test drugs. Results and conclusion. When assessed with digoxin and adrenaline as positive controls, the water extract (3 mg/mL) exhibited significant positive inotropic (70.00% improvement compared to baseline) and chronotropic (17.79% improvement) effects. Additionally, heart contractility was inhibited in the presence of propranolol and amlodipine, suggesting that the testing extract is having adrenergic properties. Reversible heart arrest was noticed at the dose of 6 mg/mL.

The diagnosis, monitoring and flukicide efficacy testing of fasciolosis on-farm is reliant on non-terminal methods. The coproantigen ELISA (cELISA) has been recommended for diagnosis of fasciolosis and associated flukicide efficacy testing as an alternative to fluke egg counts for monitoring parasitism. Recently experimental multi-age infections have suggested that the reliability of efficacy results can be improved by a second cELISA testing at 6 weeks post-treatment (wpt) in addition to the generally accepted 1 wpt. A field study was conducted to determine the suitability of faecal fluke egg counts (FFEC) and cELISA as diagnostic, drug efficacy testing and epidemiological tools on Australian sheep and cattle farms. Faecal samples from sheep and/or cattle on three endemic farms were taken at monthly intervals for 12 months and examined by both methods. Normal farm management was maintained during the study period and opportunistic efficacy testing, in line with each farm’s normal flukicide management was undertaken. Additionally, the suitability of the Ollerenshaw Index as a predictive model for fasciolosis under Australian conditions was examined. While both diagnostics demonstrated their value in the farm environment, the current data demonstrate a distinct and significant increase in diagnostic sensitivity for epidemiological studies by using the two tests in parallel. The agreement between the two diagnostics was found to be higher in cattle, despite the poor sensitivity of FFEC in this species. Similar levels of agreement between the two tests were demonstrated at both sheep properties, regardless of the marked difference in the intensity of F. hepatica challenge. Linear regression models demonstrated the results of the two diagnostics utilized in parallel were explained substantially (R2 = 0.91) as were series data (R2 = 0.88) when the respective models were fitted. In contrast, the fitted models for FFEC (R2 = 0.54) and cELISA (R2 = 0.58) were poor explanations for test outcomes. The outcomes of these models support previous findings that suggest that the two diagnostic tests are best utilized together, particularly in parallel. Background Despite their abundance and ecological importance, bats are under significant threat worldwide. There is little information about their distribution, roosting, and habitat requirement for most species, making assessing which species is threatened or in need of special conservation measures difficult. The knowledge gap may partly be due to limitations of the old methods of studying bats which mainly involved capture/or observational techniques. Material and methods In order to evaluate the potential of identifying insectivorous bats by their echolocation calls in the Sahelian zone of northern Cameroon, 65 bats belonging to five species were captured using standard mist netting: Mops condylurus, Chaerephon major, Mops niveiventer, Scotophilus dinganii, and Scotophilus leucogaster. The bats were identified by using morphometric measurements. An Anabat SD1 detector was later used to record echolocation calls of each individual bat in flight after it was hand-released. The sonogram of each individual bat was analyzed using Analook and categorized into two call types (frequency modulation and frequency modulation/quasi constant frequency) in order to develop a library of bat reference calls that could be used for a qualitative acoustic survey and species identification. Discriminant function analysis (DFA) was applied to search phase calls of the 65 individual bats in order to evaluate the potential for classifying calls into five species groups. Seven parameters calculated from each search phase call were used to classify calls. Results Bats where place into two groups according to the structure of calls: FM bats (Mops condylurus, Chaerephon major, Mops niveiventer) and FM/QCF bats (Scotophilus dinganii and Scotophilus leucogaster). The DFA resulted in a correct overall classification of 69.7%. Conclusion This preliminary study showed that DFA of call parameters is a feasible method that can be used to identify insectivorous bats in the region by their echolocation calls.

Aim: The objective of the present study was to evaluate the wound healing effect of a methanol extract of Allium cepa Linn. in vivo. Materials and Methods: The healing activity was studied using incision wound on diabetes suppressed wound healing rodent model. Results: Data recorded exhibited a significant improvement of the blood glucose level and scare breaking strength in diabetic mice at 300 and 400 mg/kg onion extract. Histological examination of the general morphology of wounds of extract treated group has shown a normal histological pattern of healthy skin at the same doses. Background: Vitellaria paradoxa is a traditional medicinal plant of Cameroon. Several studies on this plant have focused on the cosmetic profile of its fruits. The present study focuses on the anti-inflammatory potency of stem barks extract of this plant. Objective: The objective was to evaluate the effect of methanolic extract of V. paradoxa (VPME) stem barks on inflammatory response in rats. Materials and Methods: Anti-inflammatory effects of VPME were evaluated in acute and chronic (28 days) inflammation induced in Wistar albino rats. The effects on hyperalgesia and locomotors activity were also quantified. The relative weight of lymphoid organs was obtained as well as some hematological parameters. Results: In the carrageenan-induced inflammation, VPME (75 mg/kg) exhibited a significant (66.67%) inhibition after 1 h. On the complete Freund's adjuvant-induced rheumatoid arthritis, VPME showed a significant protective effect with 8.12% inflammation against 25.00% for the control group after 2 days of the treatment. The extract (75 and 150 mg/kg) significantly reduced the score of arthritis with a maximum obtained on day 19 th of the experimentation. There was a significant increase in the reaction time of rats on the hot plate as well as the exploratory activities of the animals in the open field. This extract significantly prevented weight, hemoglobin and red blood cells losses, and spleen hypertrophy. A protective action against skin destruction and cartilage erosion was evident. Liquid chromatography-mass spectrometry analysis of the extract revealed the presence of catechins. Conclusions: These findings suggested that V. paradoxa may contribute to the reduction of the inflammatory response.

Information on the susceptibility status of Fasciola hepatica isolates is lacking in the literature, even for those isolates considered to be laboratory reference strains. Four controlled efficacy studies were conducted on two Fasciola hepatica isolates from Australia, viz. 'Oberon' and 'Sunny Corner' with treatment at either 2, 6 or 10 weeks post-infection (wpi) as defined in each study. Fluke burdens and examination of livers occurred at necropsy in weeks 12 (Study 1) or 13 (Studies 2, 3 and 4) post-infection. The triclabendazole (TCBZ) resistance status of the Oberon isolate was confirmed in 6 and 10-week old F. hepatica, utilizing the drug alone (Fasinex; 71.5% and 31.1%, respectively) and in combination with oxfendazole (Flukazole C; 79.9% and 0%, respectively). The susceptibility of this isolate to albendazole, as well as salicylanilide and sulphonamide drugs was confirmed. The Sunny Corner isolate was confirmed as susceptible to TCBZ (>99% all stages) and closantel (>90% at ≥6 wpi).

A new cicada genus, Neoterpnosia n. gen., is described for Terpnosia oberthuri Distant, 1912 as the type species, which is placed in the subtribe Euterpnosiina Lee, 2013 of the tribe Leptopsaltriini Moulton, 1923 in the subfamily Cicadinae Latreille, 1802. Terpnosia oberthuri Distant, 1912 becomes Neoterpnosia oberthuri (Distant, 1912) n. comb. Terpnosia versicolor Distant, 1912 is transferred from Terpnosia Distant, 1892 to Neoterpnosia n. gen. to become Neoterpnosia versicolor (Distant, 1912) n. comb. Two new species, Neoterpnosia personalis n. gen., n. sp. and Neoterpnosia donghai n. gen., n. sp., are described from Vietnam and India, respectively.

Over 300 pedestrians died in the UK after being struck by plant or vehicles over the last decade, with the construction industry being the worst offender. On ‘smart motorway' projects such as the M4/M5 scheme near Bristol, the narrow working area – in most locations only 3·6 m wide – adds more potential for conflict between people and plant. Following a number of near-miss incidents, the M4/M5 site team developed an innovative and highly effective ‘life-sized' training programme to change individual behaviours. The programme has proved a great success, both in the UK and internationally.
