OBJECTIVES: Prone positioning (PP) during invasive mechanical ventilation (MV) improves outcomes of patients with severe ARDS. Recent studies suggest that PP in spontaneously breathing non-intubated patients with acute respiratory failure is well tolerated and improves oxygenation. However, little is known regarding assisted ventilation in intubated patients with ARDS undergoing PP. We conducted a retrospective review of our experience proning two clinical series of patients with moderate and severe ARDS (related to COVID-19 in one cohort, unrelated in the other), many of whom were underpressure support ventilation (PSV). METHODS: Retrospective analysis, in one 22-bed mixed ICU. The patients included in the analysis were aged ≥18 years, met the Berlin definition for moderate or severe ARDS (related or not to COVID-19) and underwent prone positioning under invasive mechanical ventilation. RESULTS: Thirty nine patients were included in the analysis. 20 patients had COVID-19 related ARDS while 19 had ARDS related to other etiologies. A total of 113 prone positioning episodes were analyzed, 84 in PSV and 29 in volume assist-control ventilation (VAC). Direct imaging instruments have the spatial resolution to resolve exoplanets from their host star. This enables direct characterization of the exoplanets atmosphere, but most direct imaging instruments do not have spectrographs with high enough resolving power for detailed atmospheric characterization. We investigate the use of a single-mode diffraction-limited integral-field unit that is compact and easy to integrate into current and future direct imaging instruments for exoplanet characterization. This achieved by making use of recent progress in photonic manufacturing to create a single-mode fiber-fed image reformatter. The fiber-link is created with 3D printed lenses on top of a single-mode multi-core fiber that feeds an ultrafast laser inscribed photonic chip that reformats the fiber into a pseudo-slit. We then couple it to a first-order spectrograph with a triple stacked volume phase holographic grating for a high efficiency over a large bandwidth. The prototype system has had a successful first-light observing run at the 4.2 meter William Herschel Telescope. The measured on-sky resolving power is between 2500 and 3000, depending on the wavelength. Recently we have demonstrated the potential of a hybrid astrophotonic device, consisting of a multi-core fiber photonic lantern and a 3D waveguide reformatting component, to efficiently reformat the multimode point spread function of a telescope to a diffracted limited pseudo-slit. Here, we report on an investigation into the potential of this device to mitigate modal noise-one of the main hurdles of multi-mode fiber-fed spectrographs. The modal noise performance of the photonic reformatter and other fiber feeds was assessed using a bench-Top spectrograph based on an echelle grating. In a first method of modal noise quantification, we used broadband light as the input, and assessed the modal noise performance based on the variations in the normalized spectrum as the input coupling to the fiber feed is varied. In a second method, we passed the broadband light through an etalon to generate a source with spectrally narrow peaks. We then used the spectral stability of these peaks as the input coupling to the fiber feed was varied as a proxy for the modal noise. Using both of these approaches we found that the photonic reformatter could significantly reduce modal noise compared to the multi-mode fiber feed, demonstrating the potential of photonic reformatters to mitigate modal noise for applications such as near-IR radial velocity measurements of M-dwarf stars.

We report on the conception and the fabrication of a 3D photonic reformatter of 73 waveguides and its associated opaque mask in a wide collaboration to develop a multi-core fiber-fed integral field spectrograph (MCIFU) centered on the Jband. The reformatter is a 3D structure that light from the input quasi-hexagonal multicore fiber is spread out by rearrangement to avoid individual core spectra overlapping when the light is dispersed. The reformatter is fabricated using ultrafast laser inscription (ULI) in a borosilicate glass of 20 mm length. Using a similar ULI process, a 73-hole mask was fabricated in silica glass that precisely matched the waveguides at the output of the reformatter. The output surface of the mask was coated with a 120 nm layer of chromium to block scattered light generated in the bulk material and enhance the signal-to-noise. All inscribed waveguides, characterized using a stable laser centered at 1310 nm from the multicore fiber to the output mask, present consistent single-mode output behavior with a maximum throughput exceeding 60%. Over the 73 cores, the average throughput was measured at 40%. First observations of the full MCIFU device during on-sky measurements have shown promising results to the potential of this novel fiber integral field unit.

We report the ultrafast laser inscription (ULI) and characterization of 3 dB directional achromatic couplers for K-band between 2 and 2.4 μm. The couplers were fabricated in commercial Infrasil glass using 1030 nm femtosecond laser pulses. Straight waveguides inscribed using optimal fabrication parameters exhibit an average propagation loss of ∼1.21 dB over full range of K-band with a single-mode behavior for a length of 17 mm. Directional couplers with different interaction lengths and waveguide widths were fabricated and characterized. We demonstrate that 3 dB achromatic directional couplers for K-band can be fabricated using ULI. These results show that ULI can fabricate highquality couplers for future applications in astronomical interferometry. Our eventual aim is to develop a two-telescope K-band integrated optical beam combiner to replace JouFLU at CHARA.

The Multi-Core Integral-Field Unit (MCIFU) is a diffraction-limited near-infrared integral-field spectrograph designed to detect and characterise exoplanets and disks in combination with extreme adaptive optics (xAO) instruments. It has been developed by an extended consortium as an experimental path finder for medium resolution spectroscopic upgrades for xAO systems. To allow it to achieve its goals we manufactured a fibre link system composed of a custom integrated fiber, with 3D printed microlenses and an ultrafast laser inscribed reformatter. Here we detail the specific requirements of the fibre link, from its design parameters, through its manufacture the laboratory performance and discuss upgrades for the future.

Cold temperatures are a threat to temperate plants, and Arabidopsis thaliana has acquired an adaptive gene expression network controlled by CBF transcription factors. The CBFs are sufficient to enable plants to survive otherwise lethal subzero temperatures. Constitutive CBF expression causes delayed flowering and stunted growth, and plants have evolved the ability to restrict CBF expression to occur only in the cold. This allows plants to anticipate likely freezing events and selectively deploy cold tolerance. The mechanism by which cold stress is sensed is however unknown. Here we show that protein translation rates in plants are proportional to temperature, and reduced translation rates trigger a rise in intracellular free calcium that activates the CAMTA transcription factors, and these directly activate cold-induced gene expression.

Funder: CIFRE, Chromatin immunoprecipitation-sequencing (ChIP-seq) is a robust technique to study interactions between proteins, such as histones or transcription factors and DNA. This technique in combination with RNA-sequencing (RNA-seq) is a powerful tool to better understand biological processes in eukaryotes. We developed a combined ChIP-seq and RNA-seq protocol for tree buds (Prunus avium L., Prunus persica L Batch, Malus x domestica Borkh.) that has also been successfully tested on Arabidopsis thaliana and Saccharomyces cerevisiae. Tree buds contain phenolic compounds that negatively interfere with ChIP and RNA extraction. In addition to solving this problem, our protocol is optimised to work on small amounts of material. Furthermore, one of the advantages of this protocol is that samples for ChIP-seq are cross-linked after flash freezing, making it possible to work on trees growing in the field and to perform ChIP-seq and RNA-seq on the same starting material. Focusing on dormant buds in sweet cherry, we explored the link between expression level and H3K4me3 enrichment for all genes, including a strong correlation between H3K4me3 enrichment at the DORMANCY-ASSOCIATED MADS-BOX 5 (PavDAM5) loci and its expression pattern. This protocol will allow analysis of chromatin and transcriptomic dynamics in tree buds, notably during its development and response to the environment.

Optical biopsy describes a range of medical procedures in which light is used to investigate disease in the body, often in hard-to-reach regions via optical fibres. Optical biopsies can reveal a multitude of diagnostic information to aid therapeutic diagnosis and treatment with higher specificity and shorter delay than traditional surgical techniques. One specific type of optical biopsy relies on Raman spectroscopy to differentiate tissue types at the molecular level and has been used successfully to stage cancer. However, complex micro-optical systems are usually needed at the distal end to optimise the signal-to-noise properties of the Raman signal collected. Manufacturing these devices, particularly in a way suitable for large scale adoption, remains a critical challenge. In this paper, we describe a novel fibre-fed micro-optic system designed for efficient signal delivery and collection during a Raman spectroscopy-based optical biopsy. Crucially, we fabricate the device using a direct-laser-writing technique known as ultrafast laser-assisted etching which is scalable and allows components to be aligned passively. The Raman probe has a sub-millimetre diameter and offers confocal signal collection with 71.3% &plusmn, 1.5% collection efficiency over a 0.8 numerical aperture. Proof of concept spectral measurements were performed on mouse intestinal tissue and compared with results obtained using a commercial Raman microscope.

Recently, we demonstrated how an astrophotonic light reformatting device, based on a multicore fibre photonic lantern and a three-dimensional waveguide component, can be used to efficiently reformat the point spread function of a telescope to a diffraction-limited psuedo-slit [arXiv:1512.07309]. Here, we demonstrate how such a device can also efficiently mitigate modal noise -- a potential source of instability in high resolution multi-mode fibre-fed spectrographs). To investigate the modal noise performance of the photonic reformatter, we have used it to feed light into a bench-top near-infrared spectrograph (R {\approx} 9,500, {\lambda} {\approx} 1550 nm). One approach to quantifying the modal noise involved the use of broadband excitation light and a statistical analysis of how the overall measured spectrum was affected by variations in the input coupling conditions. This approach indicated that the photonic reformatter could reduce modal noise by a factor of six when compared to a multi-mode fibre with a similar number of guided modes. Another approach to quantifying the modal noise involved the use of multiple spectrally narrow lines, and an analysis of how the measured barycentres of these lines were affected by variations in the input coupling. Using this approach, the photonic reformatter was observed to suppress modal noise to the level necessary to obtain spectra with stability close to that observed when using a single mode fibre feed. These results demonstrate the potential of using photonic reformatters to enable efficient multi-mode spectrographs that operate at the diffraction limit and are free of modal noise, with potential applications including radial velocity measurements of M-dwarfs., Comment: 13 pages, 9 figures (including appendix)

We present the fabrication and characterization of 3 dB asymmetric directional couplers for the astronomical K-band at wavelengths between 2.0 and 2.4 µm. The couplers were fabricated in commercial Infrasil silica glass using an ultrafast laser operating at 1030 nm. After optimizing the fabrication parameters, the insertion losses of straight single-mode waveguides were measured to be ∼ 1.2 ± 0.5 d B across the full K-band. We investigate the development of asymmetric 3 dB directional couplers by varying the coupler interaction lengths and by varying the width of one of the waveguide cores to detune the propagation constants of the coupled modes. In this manner, we demonstrate that ultrafast laser inscription is capable of fabricating asymmetric 3 dB directional couplers for future applications in K-band stellar interferometry. Finally, we demonstrate that our couplers exhibit an interferometric fringe contrast of > 90 % . This technology paves the path for the development of a two-telescope K-band integrated optic beam combiner for interferometry to replace the existing beam combiner (MONA) in Jouvence of the Fiber Linked Unit for Recombination (JouFLU) at the Center for High Angular Resolution Astronomy (CHARA) telescope array.
