1. Drone-Borne LiDAR and Photogrammetry Together with Historical Data for Studying a Paleo-Landslide Reactivated by Road-Cutting and Barrier Construction outside Jerusalem

Abstract: Assessment of landslide hazards often depends on the ability to track possible changes in natural slopes. To that end, historical air photos can be useful, particularly when slope stability is compromised by visible cracking. Undocumented landsliding rejuvenates a paleo-landslide on a busy motorway connecting Jerusalem to a small Jewish settlement. Recently, a plan for broadening the motorway was approved, and we were asked to study the hazards of the road by Israeli NGOs and Palestinian residents of the area. We captured high-resolution topography around the unstable slope using drone-borne photogrammetry and LiDAR surveys. The modern data allow us to analyze historic air photos and topo maps to assess the level of sliding prior to and during modern landscaping. Our results indicate horizontal offsets of ~0.9–1.8 m and vertical offsets of 1.54–2.95 m at selected sites. We next assess the possible role of anthropogenic versus natural factors in compromising slope stability. We analyze monthly rain records together with seismic catalogs spanning several decades. Shortly after the motorway construction in 1995, a January 1996 rainstorm triggered a massive rockfall. The rockfall blocked traffic with up to 4 m-diameter boulders. We found that while a certain level of rain is a necessary condition for mobilizing the rock mass, it is the anthropogenic intervention that caused the rockfall in this site. We conclude that the recent plan for broadening the motorway jeopardizes the lives of vehicle passengers and the lives of future residents should the development materialize.

2. An early Byzantine ecclesiastical complex at Ashdod-Yam: correlating geophysical prospection with excavated remains

Abstract: In this study we show the successful deployment of Ground Penetrating Radar (GPR) together with Electrical Resistivity Surveys (ERS) in guiding the archeological excavations at Ashdod-Yam (southern coast of Israel). This approach enabled the precise identification of excavation targets relating to an Early Byzantine ecclesiastical complex located in a residential neighborhood of the modern city of Ashdod. Applied over the course of five years, the combined use of GPR and ERS, interspersed with phases of archeological excavation, not only facilitated an efficient exploration but also ensured the preservation of valuable historical structures. The geophysical data, corroborated by drone images of the post-excavation site, revealed a striking correlation between excavation and non-intrusive survey data. This study not only charts a successful excavation journey but serves as a methodological blueprint for future archeological explorations. The techniques and strategies detailed here have broader implications for the preservation and public presentation of historical sites.

Abstract: Abstract To date, the most complete paleolake‐level reconstructions for the late Pleistocene water bodies that once occupied the Dead Sea depression have been based on the combination of dating of lake sediments and terrestrial materials. However, despite these major accomplishments, there is still limited spatial control regarding the water levels, suggesting some degree of uncertainty concerning the magnitude and rate of lake‐level changes. Here, we re‐examine the late Pleistocene lake‐level changes in the Dead Sea during the transition from paleolake Lisan to the present‐day Dead Sea. We rely on systematic dating of fossil stromatolites including 84 radiocarbon and 15 U‐series ages, stable‐isotope measurements, paleobiology, high‐resolution topography, and numerical modeling to assess lake‐level changes. Our results indicate that the highstand of paleolake Lisan was of shorter duration and the transition between Lake Lisan and the Dead Sea occurred at least 5 Kyrs earlier than previously indicated. By refining the timeline and accuracy of lake‐level positions during the transition paleolake Lisan—Dead Sea, our study offers new insights into the regional and local paleo‐climatic conditions during the last glacial period in this region.

5. Estimation of Low-Velocity Landfill Thickness with Multi-Method Seismic Surveys

Abstract: Conventional geophysical methods are suitable for estimating the thicknesses of subsoil layers. By combining several geophysical methods, the uncertainties can be assessed. Hence, the reliability of the results increases with a more accurate engineering solution. To estimate the base of an abandoned landfill, we collected data using classical approaches: high-resolution seismic reflection and refraction, with more modern methods including passive surface wave analysis and horizontal-to-vertical spectral ratio (HVSR) measurements. To evaluate the thickness of the landfill, three different datasets were acquired along each of the two seismic lines, and five different processing methods were applied for each of the two arrays. 7. GPR Mapping of buried monumental retaining walls at biblical Kiriath-Yearim near Jerusalem

Abstract: In 2019 a ground-penetrating radar (GPR) investigation was conducted at Kiriath-Yearim near Jerusalem, one of the largest Bronze and Iron Ages mounds in the highlands of the southern Levant. The main objective was to test an archaeological hypothesis regarding the existence of an Iron Age summit compound which was supported by monumental stone retaining walls.We used a wheeled GPR with simultaneous central frequencies of 200, 400, and 800 MHz. Despite attenuation in the topsoil, scattering by distributed rock fragments, and asphalt pavement, we were able to penetrate almost a meter and detect anomalies.Some of the GPR anomalies are consistent with the above-mentioned theory, meaning that their locations agree with data retrieved in nearby control trenches and with the plan of the site.The excavation findings, jointly with the GPR interpretation, indeed imply the existence of a rectangular monumental summit compound that dates to the 8th century BCE.

Abstract: Summary: Genetics have nominated many schizophrenia risk genes and identified convergent signals between schizophrenia and neurodevelopmental disorders. However, functional interpretation of the nominated genes in the relevant brain cell types is often lacking. We executed interaction proteomics for six schizophrenia risk genes that have also been implicated in neurodevelopment in human induced cortical neurons. The resulting protein network is enriched for common variant risk of schizophrenia in Europeans and East Asians, is down-regulated in layer 5/6 cortical neurons of individuals affected by schizophrenia, and can complement fine-mapping and eQTL data to prioritize additional genes in GWAS loci. A sub-network centered on HCN1 is enriched for common variant risk and contains proteins (HCN4 and AKAP11) enriched for rare protein-truncating mutations in individuals with schizophrenia and bipolar disorder. Our findings showcase brain cell-type-specific interactomes as an organizing framework to facilitate interpretation of genetic and transcriptomic data in schizophrenia and its related disorders.

10. Drone-Borne LiDAR and Photogrammetry Together with Historical Data for Studying a Paleo-Landslide Reactivated by Road-Cutting and Barrier Construction outside Jerusalem.

Abstract: Assessment of landslide hazards often depends on the ability to track possible changes in natural slopes. To that end, historical air photos can be useful, particularly when slope stability is compromised by visible cracking. Undocumented landsliding rejuvenates a paleo-landslide on a busy motorway connecting Jerusalem to a small Jewish settlement. Recently, a plan for broadening the motorway was approved, and we were asked to study the hazards of the road by Israeli NGOs and Palestinian residents of the area. We captured high-resolution topography around the unstable slope using drone-borne photogrammetry and LiDAR surveys. The modern data allow us to analyze historic air photos and topo maps to assess the level of sliding prior to and during modern landscaping. Our results indicate horizontal offsets of ~0.9–1.8 m and vertical offsets of 1.54–2.95 m at selected sites. We next assess the possible role of anthropogenic versus natural factors in compromising slope stability. We analyze monthly rain records together with seismic catalogs spanning several decades. Shortly after the motorway construction in 1995, a January 1996 rainstorm triggered a massive rockfall. The rockfall blocked traffic with up to 4 m-diameter boulders. We found that while a certain level of rain is a necessary condition for mobilizing the rock mass, it is the anthropogenic intervention that caused the rockfall in this site. We conclude that the recent plan for broadening the motorway jeopardizes the lives of vehicle passengers and the lives of future residents should the development materialize. [ABSTRACT FROM AUTHOR]

Abstract: Abstract Although pancreatic ductal adenocarcinoma (PDAC) survival is poor, there are differences in patients’ response to the treatments. Detection of predictive biomarkers explaining these differences is of the utmost importance. In a recent study two genetic markers (CD44-rs353630 and CHI3L2-rs684559) were reported to be associated with survival after PDAC resection. We attempted to replicate the associations in 1856 PDAC patients (685 resected with stage I/II) from the PANcreatic Disease ReseArch (PANDoRA) consortium. We also analysed the combined effect of the two genotypes in order to compare our results with what was previously reported. Additional stratified analyses considering TNM stage of the disease and whether the patients received surgery were also performed. We observed no statistically significant associations, except for the heterozygous carriers of CD44-rs353630, who were associated with worse OS (HR = 5.01; 95% CI 1.58–15.88; p = 0.006) among patients with stage I disease. This association is in the opposite direction of those reported previously, suggesting that data obtained in such small subgroups are hardly replicable and should be considered cautiously. The two polymorphisms combined did not show any statistically significant association. Our results suggest that the effect of CD44-rs353630 and CHI3L2-rs684559 cannot be generalized to all PDAC patients.

Abstract: To date, the most complete paleolake‐level reconstructions for the late Pleistocene water bodies that once occupied the Dead Sea depression have been based on the combination of dating of lake sediments and terrestrial materials. However, despite these major accomplishments, there is still limited spatial control regarding the water levels, suggesting some degree of uncertainty concerning the magnitude and rate of lake‐level changes. Here, we re‐examine the late Pleistocene lake‐level changes in the Dead Sea during the transition from paleolake Lisan to the present‐day Dead Sea. We rely on systematic dating of fossil stromatolites including 84 radiocarbon and 15 U‐series ages, stable‐isotope measurements, paleobiology, high‐resolution topography, and numerical modeling to assess lake‐level changes. Our results indicate that the highstand of paleolake Lisan was of shorter duration and the transition between Lake Lisan and the Dead Sea occurred at least 5 Kyrs earlier than previously indicated. By refining the timeline and accuracy of lake‐level positions during the transition paleolake Lisan—Dead Sea, our study offers new insights into the regional and local paleo‐climatic conditions during the last glacial period in this region. Plain Language Summary: Index points, reflecting the unique position of the water levels in space and time, have been broadly used in marine realms to reconstruct past sea‐level changes. Here we study lake‐level variations using index points based on geomorphic and biological markers collected along the western and eastern coasts of the Dead Sea. Previous studies indicate that during the last glacial period (20–30 ka), the Dead Sea depression was filled by paleolake Lisan, which lasted 10 Kyrs. As it dried, dozens of paleo‐shorelines remained etched in the landscape. Lake level index points estimated from 99 radiometric ages in these paleo‐shorelines suggest that Lisan's highstand was shorter and the transition to the Dead Sea occurred 5 Kyrs earlier than previously thought. These novel results suggest an initial and wet period followed by drier conditions, altering the regional paleo‐climatic narrative during the last glacial period. Abstract: Background & aimsCrohn's disease (CD) has the highest prevalence in Ashkenazi Jewish populations. We sought to identify rare, CD-associated frameshift variants of high functional and statistical effects.MethodsWe performed exome sequencing and array-based genotype analyses of 1477 Ashkenazi Jewish individuals with CD and 2614 Ashkenazi Jewish individuals without CD (controls). To validate our findings, we performed genotype analyses of an additional 1515 CD cases and 7052 controls for frameshift mutations in the colony-stimulating factor 2-receptor β common subunit gene (CSF2RB). Intestinal tissues and blood samples were collected from patients with CD; lamina propria leukocytes were isolated and expression of CSF2RB and granulocyte-macrophage colony-stimulating factor-responsive cells were defined by adenomatous polyposis coli (APC) time-of-flight mass cytometry (CyTOF analysis). Variants of CSF2RB were transfected into HEK293 cells and the expression and functions of gene products were compared.ResultsIn the discovery cohort, we associated CD with a frameshift mutation in CSF2RB (P = 8.52 × 10(-4)); the finding was validated in the replication cohort (combined P = 3.42 × 10(-6)). Incubation of intestinal lamina propria leukocytes with granulocyte-macrophage colony-stimulating factor resulted in high levels of phosphorylation of signal transducer and activator of transcription (STAT5) and lesser increases in phosphorylation of extracellular signal-regulated kinase and AK straining transforming (AKT). Cells co-transfected with full-length and mutant forms of CSF2RB had reduced pSTAT5 after stimulation with granulocyte-macrophage colony-stimulating factor, compared with cells transfected with control CSF2RB, indicating a dominant-negative effect of the mutant gene. Monocytes from patients with CD who were heterozygous for the frameshift mutation (6% of CD cases analyzed) had reduced responses to granulocyte-macrophage colony-stimulating factor and markedly decreased activity of aldehyde dehydrogenase; activity of this enzyme has been associated with immune tolerance.ConclusionsIn a genetic analysis of Ashkenazi Jewish individuals, we associated CD with a frameshift mutation in CSF2RB. Intestinal monocytes from carriers of this mutation had reduced responses to granulocyte-macrophage colony-stimulating factor, providing an additional mechanism for alterations to the innate immune response in individuals with CD.

Abstract: ImportanceA recently published study of national data by McGrath et al in 2014 showed increased risk of schizophrenia (SCZ) in offspring associated with both early and delayed parental age, consistent with a U-shaped relationship. However, it remains unclear if the risk to the child is due to psychosocial factors associated with parental age or if those at higher risk for SCZ tend to have children at an earlier or later age.ObjectiveTo determine if there is a genetic association between SCZ and age at first birth (AFB) using genetically informative but independently ascertained data sets.Design, setting, and participantsThis investigation used multiple independent genome-wide association study data sets. The SCZ sample comprised 18 957 SCZ cases and 22 673 controls in a genome-wide association study from the second phase of the Psychiatric Genomics Consortium, and the AFB sample comprised 12 247 genotyped women measured for AFB from the following 4 community cohorts: Estonia (Estonian Genome Center Biobank, University of Tartu), the Netherlands (LifeLines Cohort Study), Sweden (Swedish Twin Registry), and the United Kingdom (TwinsUK). Schizophrenia genetic risk for each woman in the AFB community sample was estimated using genetic effects inferred from the SCZ genome-wide association study.Main outcomes and measuresWe tested if SCZ genetic risk was a significant predictor of response variables based on published polynomial functions that described the relationship between maternal age and SCZ risk in offspring in Denmark. We substituted AFB for maternal age in these functions, one of which was corrected for the age of the father, and found that the fit was superior for the model without adjustment for the father's age.ResultsWe observed a U-shaped relationship between SCZ risk and AFB in the community cohorts, consistent with the previously reported relationship between SCZ risk in offspring and maternal age when not adjusted for the age of the father. We confirmed that SCZ risk profile scores significantly predicted the response variables (coefficient of determination R2 = 1.1E-03, P = 4.1E-04), reflecting the published relationship between maternal age and SCZ risk in offspring by McGrath et al in 2014.Conclusions and relevanceThis study provides evidence for a significant overlap between genetic factors associated with risk of SCZ and genetic factors associated with AFB. It has been reported that SCZ risk associated with increased maternal age is explained by the age of the father and that de novo mutations that occur more frequently in the germline of older men are the underlying causal mechanism. This explanation may need to be revised if, as suggested herein and if replicated in future studies, there is also increased genetic risk of SCZ in older mothers.

22. Quantitative PCR from human genomic DNA: The determination of gene copy numbers for congenital adrenal hyperplasia and RCCX copy number variation.

Abstract: Quantitative PCR (qPCR) is used for the determination of gene copy number (GCN). GCNs contribute to human disorders, and characterize copy number variation (CNV). The single laboratory method validations of duplex qPCR assays with hydrolysis probes on CYP21A1P and CYP21A2 genes, residing a CNV (RCCX CNV) and related to congenital adrenal hyperplasia, were performed using 46 human genomic DNA samples. We also performed the verifications on 5 qPCR assays for the genetic elements of RCCX CNV; C4A, C4B, CNV breakpoint, HERV-K(C4) CNV deletion and insertion alleles. Precision of each qPCR assay was under 1.01 CV%. Accuracy (relative error) ranged from 4.96±4.08% to 9.91±8.93%. Accuracy was not tightly linked to precision, but was significantly correlated with the efficiency of normalization using the RPPH1 internal reference gene (Spearman's ρ: 0.793-0.940, p>0.0001), ambiguity (ρ = 0.671, p = 0.029) and misclassification (ρ = 0.769, p = 0.009). A strong genomic matrix effect was observed, and target-singleplex (one target gene in one assay) qPCR was able to appropriately differentiate 2 GCN from 3 GCN at best. The analysis of all GCNs from the 7 qPCR assays using a multiplex approach increased the resolution of differentiation, and produced 98% of GCNs unambiguously, and all of which were in 100% concordance with GCNs measured by Southern blot, MLPA and aCGH. We conclude that the use of an internal (in one assay with the target gene) reference gene, the use of allele-specific primers or probes, and the multiplex approach (in one assay or different assays) are crucial for GCN determination using qPCR or other methods.

Abstract: Widespread sharing of long, identical-by-descent (IBD) genetic segments is a hallmark of populations that have experienced recent genetic drift. Detection of these IBD segments has recently become feasible, enabling a wide range of applications from phasing and imputation to demographic inference. Here, we study the distribution of IBD sharing in the Wright-Fisher model. Specifically, using coalescent theory, we calculate the variance of the total sharing between random pairs of individuals. We then investigate the cohort-averaged sharing: the average total sharing between one individual and the rest of the cohort. We find that for large cohorts, the cohort-averaged sharing is distributed approximately normally. Surprisingly, the variance of this distribution does not vanish even for large cohorts, implying the existence of "hyper-sharing" individuals. The presence of such individuals has consequences for the design of sequencing studies, since, if they are selected for whole-genome sequencing, a larger fraction of the cohort can be subsequently imputed. We calculate the expected gain in power of imputation by IBD, and subsequently, in power to detect an association, when individuals are either randomly selected or specifically chosen to be the hyper-sharing individuals. Using our framework, we also compute the variance of an estimator of the population size that is based on the mean IBD sharing and the variance in the sharing between inbred siblings. Finally, we study IBD sharing in an admixture pulse model, and show that in the Ashkenazi Jewish population the admixture fraction is correlated with the cohort-averaged sharing., Comment: Includes Supplementary Material

Abstract: Copy number variants (CNVs) are risk factors in neurodevelopmental disorders, including autism, epilepsy, intellectual disability (ID) and schizophrenia. Childhood onset schizophrenia (COS), defined as onset before the age of 13 years, is a rare and severe form of the disorder, with more striking array of prepsychotic developmental disorders and abnormalities in brain development. Abstract: Instrumental strong motion data are not common around the Dead Sea region. Therefore, calibrating a new attenuation equation is a considerable challenge. However, the Holy Land has a remarkable historical archive, attesting to numerous regional and local earthquakes. Combining the historical record with new seismic measurements will improve the regional equation. On 11 July 1927, a rupture, in the crust in proximity to the northern Dead Sea, generated a moderate 6.2 ML earthquake. Up to 500 people were killed, and extensive destruction was recorded, even as far as 150 km from the focus. We consider local near-surface properties, in particular, the shear-wave velocity, as an amplification factor. Where the shear-wave velocity is low, the seismic intensity far from the focus would likely be greater than expected from a standard attenuation curve. In this work, we used the multichannel analysis of surface waves (MASW) method to estimate seismic wave velocity at anomalous sites in Israel in order to calibrate a new attenuation equation for the Dead Sea region. Our new attenuation equation contains a term which quantifies only lithological effects, while factors such as building quality, foundation depth, topography, earthquake directivity, type of fault, etc. remain out of our scope. Nonetheless, about 60 % of the measured anomalous sites fit expectations; therefore, this new ground-motion prediction equation (GMPE) is statistically better than the old ones. From our local point of view, this is the first time that integration of the 1927 historical data and modern shear-wave velocity profile measurements improved the attenuation equation (sometimes referred to as the attenuation relation) for the Dead Sea region. In the wider context, regions of low-to-moderate seismicity should use macroseismic earthquake data, together with modern measurements, in order to better estimate the peak ground acceleration or the seismic intensities to be caused by future earthquakes. This integration will conceivably lead to a better mitigation of damage from future earthquakes and should improve maps of seismic hazard.

Abstract: The Information and Communications Technology sector may be organized into individual technologies or solutions. Theoretical literature review argues digitalization technologies make up enterprise architecture and shape business processes or business models. It also shows contemporary business models market solutions for a given value or impact, rather than sell individual technologies per se. Empirical data confirms enterprise architecture is the market for system integration. Market data shows the market leader is Software AG. The goal of this article is to build a theory with an emerging case example of more technologically and business model sophisticated solutions than the theoretical and market benchmark. The methodology is a dual case study on important market players, Siemens and PTC. The standalone cases allow for similar solutions which cannot be otherwise tested or validated. Reference is also paid to other market players within the article. The empirical data analysis is compared to acceptable benchmarks, theoretical literature review and the enterprise architecture market leader Software AG. Findings show emerging technologies add capability maturity levels to accepted system integration. Network business models with a quantifiable and provable customer value proposition: bespoke or personalized products, reasonable cost per item, and zero inventory achieve the vision of mass customization. The business models are enabled by the highest level of digitalization capability maturity, not considered in the status quo literature review and at the leading enterprise architect. They place the textile industry at the forefront of technological innovation and extended reality-based marketing.

Abstract: Objective: Primary aldosteronism is a major cause of secondary hypertension. Its two principal forms are bilateral adrenal hyperplasia (BAH) and aldosterone-producing adenoma (APA) whose differentiation is clinically pivotal. There is a major clinical need for a reliable and easily accessible diagnostic biomarker for case identification and subtyping. Circulating microRNAs were shown to be useful as minimally invasive diagnostic markers. Our aim was to determine and compare the circulating microRNA expression profiles of adenoma and hyperplasia plasma samples, and to evaluate their applicability as minimally invasive markers.Methods: One hundred and twenty-three samples from primary aldosteronism patients were included. Next-generation sequencing was performed on 30 EDTA-anticoagulated plasma samples (discovery cohort). Significantly differently expressed miRNAs were validated by real-time reverse transcription-qPCR in an independent validation cohort (93 samples).Results: We have found relative overexpression of miR-30e-5p, miR-30d-5p, miR-223-3p, and miR-7-5p in hyperplasia compared to adenoma by next-generation sequencing. Validation by qRT-PCR confirmed significant overexpression of hsa-miR-30e-5p, hsa-miR-30d-5p, and hsa-miR-7-5p in hyperplasia samples. Regarding the microRNA expressional variations, adenoma is more heterogeneous at the miRNA level compared to hyperplasia.Conclusion: Three microRNAs were significantly overexpressed in hyperplasia samples compared to adenoma samples, but their sensitivity and specificity values are not good enough for introduction to clinical practice.

Descriptive statistical tools as Kruskal–Wallis, Mann–Whitney U, Levene’s F tests, Receiver Operating Characteristic (ROC) curve analysis and AUC (Area Under the Curve) with 95% confidence interval (CI) were performed.ResultsOur literature review showed extreme variability of CRP used as an inclusion criterion or as a primary outcome or both in past and current trials on AP. In our cohort, CRP levels on admission poorly predicted mortality and severe cases of AP; AUC: 0.669 (CI:0.569–0.770); AUC:0.681 (CI: 0.601–0.761), respectively. CRP levels measured within 24 h from the onset of pain failed to predict mortality or severity; AUC: 0.741 (CI:0.627–0.854); AUC:0.690 (CI:0.586–0.793), respectively. The highest CRP during hospitalization had equally poor predictive accuracy for mortality and severity AUC:0.656 (CI:0.544–0.768); AUC:0.705 (CI:0.640–0.769) respectively. CRP within 24 h from the onset of pain used as an inclusion criterion markedly increased the combined event rate of mortality and severe AP (13% for CRP > 25 mg/l and 28% for CRP > 200 mg/l).ConclusionCRP within 24 h from the onset of pain as an inclusion criterion elevates event rates and reduces the number of patients required in trials on AP.
