In the rapidly emerging field of biomedical applications, multifunctional nanoparticles, especially those containing magnetic and plasmonic components, have gained significant attention due to their combined properties. These hybrid systems, often composed of iron oxide and gold, provide both magnetic and optical functionalities and offer promising avenues for applications in multimodal bioimaging, hyperthermal therapies, and magnetically driven selective delivery. This paper focuses on the implementation of advanced characterization methods, comparing statistical analyses of individual multifunctional particle properties with macroscopic properties as a way of fine-tuning synthetic methodologies for their fabrication methods. Special emphasis is placed on the size-dependent properties, biocompatibility, and challenges that can arise from this versatile nanometric system. In order to ensure the quality and applicability of these particles, various novel methods for characterizing the magnetic gold particles, including the analysis of their morphology, optical response, and magnetic response, are also discussed, with the overall goal of optimizing the fabrication of this complex system and thus enhancing its potential as a preferred diagnostic agent. The properties of fiber reinforced polymers are strongly related to the length and orientation of the fibers within the polymer matrix, the latter of which can be studied using X-ray computed tomography (XCT). Unfortunately, resolving individual fibers is challenging because they are small compared to the XCT voxel resolution and because of the low attenuation contrast between the fibers and the surrounding resin. To alleviate both problems, anisotropic dark field tomography via grating based interferometry (GBI) has been proposed. Here, the fiber orientations are extracted by applying a Funk-Radon transform (FRT) to the local scatter function. However, the FRT suffers from a low angular resolution, which complicates estimating fiber orientations for small fiber crossing angles. We propose constrained spherical deconvolution (CSD) as an alternative to the FRT to resolve fiber orientations. Instead of GBI, edge illumination phase contrast imaging is used because estimating fiber orientations with this technique has not yet been explored. Dark field images are generated by a Monte Carlo simulation framework. It is shown that the FRT cannot estimate the fiber orientation accurately for crossing angles smaller than 70 ∘ , while CSD performs well down to a crossing angle of 50 ∘ . A dark-field scanning transmission ion microscopy detector was designed for the helium ion microscope. The detection principle is based on a secondary electron conversion holder with an exchangeable aperture strip allowing its acceptance angle to be tuned from 3 to 98 mrad. The contrast mechanism and performance were investigated using freestanding nanometer-thin carbon membranes. The results demonstrate that the detector can be optimized either for most efficient signal collection or for maximum image contrast. The designed setup allows for the imaging of thin low-density materials that otherwise provide little signal or contrast and for a clear end-point detection in the fabrication of nanopores. In addition, the detector is able to determine the thickness of membranes with sub-nanometer precision by quantitatively evaluating the image signal and comparing the results with Monte Carlo simulations. The thickness determined by the dark-field transmission detector is compared to X-ray photoelectron spectroscopy and energy-filtered transmission electron microscopy measurements.

This proof‐of‐concept study shows that dark‐field microscopy provides sufficient contrast for cell visualization in fingerprints with high sebum content. Although the application is limited to smooth surfaces that do not scatter light, such as polyethylene terephthalate (PET), it was able to measure the number of cells deposited within a fingerprint residue and the reduction in cell transfer with repeated skin contact. On a PET surface, at roughly 5 N of contact force, a typical finger transfers several hundred cells onto the surface. Over subsequent finger contacts onto a clean PET surface, this number decreased exponentially until a steady state was reached, which is characterized by the transfer of (78 ± 36) cells or (0.46 ± 0.21) cells/mm2 when normalized for fingerprint area. High uncertainty in cell transfer was due to: the highly variable nature of a human finger (where the number of loose cells varies from person to person and from day to day depending on what they touch) and difficulties in controlling the contact force and finger movement such as twisting during deposition (where twisting of the finger can expose a new patch of skin to the substrate, increasing the number of cell transfer). Plasma etching was also explored as an effective way to validate dark‐field microscopy for cell counting. Although limited to inorganic substrates due to etching effects, exposing the fingerprint for less than 10 min can remove a majority of the sebum while keeping the cells intact for a before‐and‐after comparison using light microscopy. Ever since the discovery of X-rays, tremendous efforts have been made to develop new imaging techniques for unlocking the hidden secrets of our world and enriching our understanding of it. X-ray differential phase contrast imaging, which measures the gradient of a sample's phase shift, can reveal more detail in a weakly absorbing sample than conventional absorption contrast. However, normally only the gradient's component in two mutually orthogonal directions is measurable. In this article, omnidirectional differential phase images, which record the gradient of phase shifts in all directions of the imaging plane, are efficiently generated by scanning an easily obtainable, randomly structured modulator along a spiral path. The retrieved amplitude and main orientation images for differential phase yield more information than the existing imaging methods. Importantly, the omnidirectional dark-field images can be simultaneously extracted to study strongly ordered scattering structures. The proposed method can open up new possibilities for studying a wide range of complicated samples composed of both heavy, strongly scattering atoms and light, weakly scattering atoms. A simple dark field microscopy technique was used for visualization of blood vessels in normal human renal tissues and carcinoma. Phase contrast condenser ring apt for high power objectives was combined with a 10x objective in order to create a dark field illumination of the specimens examined. The endothelial lining of the vessels had been stained by using CD31 monoclonal antibodies combined with conventional peroxidase immunohistochemistry. The final DAB addition used for this technique induced an intense light scatter in the dark field microscope. This scattered light originating from the endothelial lining made the walls of the bright vessels easily detectable from the dark background.

Abstract Background In recent years, our group has developed electro-optical remote sensing methods for the monitoring and classification of aerofauna. These methods include active lidar methods and passive, so-called dark-field methods that measure scattered sunlight. In comparison with satellite- and airborne remote sensing, our methods offer a spatiotemporal resolution several orders of magnitude higher, and unlike radar, they can be employed close to ground. Whereas passive methods are desirable due to lower power consumption and ease of use, they have until now lacked ranging capabilities. Results In this work, we demonstrate how passive ranging of sparse insects transiting the probe volume can be achieved with quadrant sensors. Insects are simulated in a raytracing model of the probe volume, and a ranging equation is devised based on the simulations. The ranging equation is implemented and validated with field data, and system parameters that vary with range are investigated. A model for estimating insect flight headings with modulation spectroscopy is implemented and tested with inconclusive results. Insect fluxes are retrieved through time-lag correlation of quadrant detector segments, showing that insects flew more with than against the wind during the study period. Conclusions The presented method demonstrates how ranging can be achieved with quadrant sensors, and how it can be implemented with or without active illumination. A number of insect flight parameters can be extracted from the data produced by the sensor and correlated with complementary information about weather and topography. The approach has the potential to become a widespread and simple tool for monitoring abundances and fluxes of pests and disease vectors in the atmosphere.

Background: X‐ray dark‐field radiography could enhance mammography by providing more information on imaged tissue and microcalcifications. The dark field signal is a measure of small angle scattering and can thus provide additional information on the imaged materials. This information can be useful for material distinction of calcifications and the diagnosis of breast cancer by classifying benign and malign association of these calcifications. Methods: For this study, institutional review board approval was obtained. We present the evaluation of images acquired with interferometric grating‐based x‐ray imaging of 323 microcalcifications (166 malign and 157 benign associated) in freshly dissected breast tissue and compare the results to the information extracted in follow‐up pathological evaluation. The number of imaged calcifications is sufficiently higher than in similar previous studies. Fourteen calcification properties were extracted from the digital images and used as predictors in three different models common in discrimination analysis namely a simple threshold model, a naive Bayes model and a linear regression model, which classify the calcifications as associated with a benign or suspicious finding. Three of these fourteen predictors have been newly defined in this work and are independent from the tissue background surrounding the microcalcifications. Using these predictors no background correction is needed, as in previous works in this field. The new predictors are the length of the first and second principle component of the absorption and dark‐field data, as well as the angle between the first principle component and the dark‐field axis. We called these predictors data length, data width, and data orientation. Results: In fourfold cross‐validation malignancy of the imaged tissue was predicted. Models that take only classical absorption predictors into account reached a sensitivity of 53.3% at a specificity of 81.1%. For a combination of predictors that also include dark field information, a sensitivity of 63.2% and specificity of 80.8% were obtained. The included dark field information consisted of the newly introduced parameters, data orientation and data width. Conclusions: While remaining at a similar specificity, the sensitivity, with which a trained model was able to distinguish malign from benign associated calcifications, was increased by 10% on including dark‐field information. The roundworm Caenorhabditis elegans is widely used as a model for studying conserved pathways for fat storage, aging, and metabolism. The most broadly used methods for imaging fat in C. elegans require fixing and staining the animal. Here, we show that dark field images acquired through an ordinary light microscope can be used to estimate fat levels in worms. We define a metric based on the amount of light scattered per area, and show that this light scattering metric is strongly correlated with worm fat levels as measured by Oil Red O (ORO) staining across a wide variety of genetic backgrounds and feeding conditions. Dark field imaging requires no exogenous agents or chemical fixation, making it compatible with live worm imaging. Using our method, we track fat storage with high temporal resolution in developing larvae, and show that fat storage in the intestine increases in at least one burst during development.

The development of sensitive methods for the determination of potential bacterial contamination is of upmost importance for environmental monitoring and food safety. In this study, we present a new method combining a fast pre-enrichment step using a microporous cryogel and a detection and identification step using antimicrobial peptides (AMPs) and labelled antibodies, respectively. The experimental method consists of: (i) the capture of large amounts of bacteria from liquid samples by using a highly porous and functionalized cryogel; (ii) the detection and categorisation of Gram-positive and Gram-negative bacteria by determining their affinities toward a small set of AMPs; and (iii) the identification of the bacterial strain by using labelled detection antibodies. As proof of concept, the assessment of the three steps of the analysis was performed by using Escherichia coli and Bacillus sp. as models for Gram-negative and Gram-positive bacteria, respectively. The use of AMPs with broad specificity combined with labelled antibodies enabled the detection and potential categorization of a large spectrum of unknown or unexpected bacteria.

The speckle‐based X‐ray imaging technique (SBT), which includes the three imaging modalities of absorption, phase contrast and dark field, is widely used in many fields. However, the influence of the grain size of the diffuser, the coherence of the X‐ray source and the pixel size of the detector on the multi‐mode imaging quality of SBT is still woefully unclear. In this paper, the whole SBT process is simulated and the influence of these three factors on image quality is discussed. Based on this discussion, the grain size of the diffuser for SBT applications should be limited by the pixel size of the detector and the coherence length of the X‐ray source. According to analysis of the noise signal and correlation map, a suitable grain size is an indispensable condition for high‐quality SBT images, because an excessively small or large grain size degrades the resolution of the imaging results and generates false signals. In addition, the power spectral density of the measured raw speckle patterns demonstrates that a smaller grain can better retain high‐frequency information from an imaged sample. The simulated and experimental results verify these conclusions. The conclusions of this work will be helpful in designing suitable experimental setups for SBT applications and have the potential to promote the performance of SBT in other applications, such as X‐ray optics metrology and coherence measurement. Dark-field illumination is a simple yet elegant imaging technique that can be used to detect the presence of particles on a specular surface. However, the sensitivity of dark-field illumination to initial conditions affects its repeatability. This is problematic in cases where automation is desired. We present an improvement to the current method of using a modulation field that relies on phase calculations rather than intensity. As a result, we obtain a computational method that is insensitive to noise and provides clearly defined particle information, allowing a global threshold to be set for autonomous measurement purposes. As an emerging technology, hyperspectral imaging (HSI), which combines both advanced spectroscopy and imaging techniques, provides sufficient information for spectral and spatial analysis and is thus suitable for distribution and property investigation of nanoscale materials. Considering the applications of HSI have spread from remote sensing to quality control of macro products such as food and milk, this article reviews recent research of HSI in a new field of nanoscale materials. On the basis of fundamental parts of a HSI system, new techniques fitting specifically for nanoscale materials imaging such as dark field and Raman spectroscopy are introduced. Nanoscale materials, including metal nanoparticles, carbon nanotubes and graphene, biological components in cells and tissues, as well as multi-layer nanoscale materials, are the research hotspots utilizing HSI technology. Related research reports of the above materials are reviewed based on the physical distinction of these nanoscale materials. It is believed that HSI technology is a strongly potential technique for property investigation and manipulation of nanomaterial for various applications. [ABSTRACT FROM AUTHOR]

Purpose: X-ray dark-field imaging promises information on the small angle scattering properties even of large samples. However, the dark-field image is correlated with the object's attenuation and phase-shift if a polychromatic x-ray spectrum is used. A method to remove part of these correlations is proposed. Methods: The experimental setup for image acquisition was modeled in a wave-field simulation to quantify the dark-field signals originating solely from a material's attenuation and phase-shift. A calibration matrix was simulated for ICRU46 breast tissue. Using the simulated data, a dark-field image of a human mastectomy sample was corrected for the finger print of attenuation- and phase-image. Results: Comparing the simulated, attenuation-based dark-field values to a phantom measurement, a good agreement was found. Applying the proposed method to mammographic dark-field data, a reduction of the dark-field background and anatomical noise was achieved. The contrast between microcalcifications and their surrounding background was increased. Conclusions: The authors show that the influence of and dispersion can be quantified by simulation and, thus, measured image data can be corrected. The simulation allows to determine the corresponding dark-field artifacts for a wide range of setup parameters, like tube-voltage and filtration. The application of the proposed method to mammographic dark-field data shows an increase in contrast compared to the original image, which might simplify a further image-based diagnosis. The background level (BGL) in an extreme-UV (EUV) dark-field image for EUV mask blank inspection was analyzed. The background refers to the light scattered by the multilayer roughness of the mask blank, and the variation in the BGL is attributed to random and fixed factors. We found that the BGL variation due to the fixed factor could be successfully reproduced by simulation using surface information obtained by atomic force microscope measurement with the assumption of a multilayer roughness structure. This means that the fixed factor contributing to the BGL variation was found to be speckle noise due to the multilayer roughness. Moreover, this reproduction procedure was found to be valuable for estimating the multilayer roughness structure up to several layers below the surface. [ABSTRACT FROM AUTHOR]

A transmission environmental scanning electron microscope with use of a scintillation gaseous detection device has been implemented. This corresponds to a transmission scanning electron microscope but with addition of a gaseous environment acting both as environmental and detection medium. A commercial type of low vacuum machine has been employed together with appropriate modifications to the detection configuration. This involves controlled screening of various emitted signals in conjunction with a scintillation gaseous detection device already provided with the machine for regular surface imaging. Dark field and bright field imaging has been obtained along with other detection conditions. With a progressive series of modifications and tests, the theory and practice of a novel type of microscopy is briefly shown now ushering further significant improvements and developments in electron microscopy as a whole. Plasmonic resonances of metal-based nanoparticles are increasingly used for ultrasensitive imaging assays. In this context, the CytovivaTM microscopy platform has greatly gained in popularity. In essence, Cytoviva is an optimized dark field microscope that permits detection of particles down to a few nanometers in size. A significant limitation of Cytoviva up to now has been that it only provided for single plane imaging. The datasets produced by this technique therefore only show a partial view of the sample - not ideally suited to analysis. Here we explain how to overcome this limitation by mounting the Cytoviva condenser on an automated microscope with Z-scanning capability. Our method allows three-dimensional mapping of nanoparticles in their full three-dimensional cellular context. We apply this technique to study the interaction of silver and cerium dioxide nanoparticles with cells of the green alga, Pseudokirchneriella subcapitata, a system of significant environmental relevance because algae underlie much of the aquatic food chain. Our objective was to develop a technique to visualize in detail the interaction of nanoparticles with cells in three dimensions, such that one may, for example, determine whether a particular nanoparticle is inside a cell, at its very surface, or at a distance from it. Using the optical methods described, specimens can be observed with modified multimodal light microscopes based on interference contrast combined with phase contrast, dark- or bright-field illumination. Thus, the particular visual information associated with interference and phase contrast, dark- and bright-field illumination is joined in real-time composite images appearing in enhanced clarity and purified from typical artefacts, which are apparent in standard phase contrast and dark-field illumination. In particular, haloing and shade-off are absent or significantly reduced as well as marginal blooming and scattering. The background brightness and thus the range of contrast can be continuously modulated and variable transitions can be achieved between interference contrast and complementary illumination techniques. The methods reported should be of general interest for all disciplines using phase and interference contrast microscopy, especially in biology and medicine, and also in material sciences when implemented in vertical illuminators. Abstract: We present real and reciprocal space photoelectron emission microscopy (PEEM) results on few layer graphene using laboratory based He I and II radiation. The combination of a focused high-intensity source and high transmission PEEM electron optics provides good signal to noise ratios for the different modes of acquisition. We demonstrate work function mapping and secondary electron analysis, related to the graphene layer thickness, band structure imaging from micron scale regions by wave vector resolved PEEM (k-PEEM) and local secondary electron spectroscopy, giving information on the valence and conduction band states and the dispersion relations of the bands. Dark field PEEM is done by selecting the Dirac cone corresponding to the specific rotation of each graphene layer and allows spatial mapping of the commensurate rotation angles. The use of He II radiation increases the volume of reciprocal space accessible to k-PEEM and improves signal to background. The preferential linear polarization of the light source is used to investigate aspects of the electronic chirality near the Dirac cone. Imaging soft matter by transmission electron microscopy (TEM) is anything but straightforward. Recently, interest has grown in developing alternative imaging modes that generate contrast without additional staining. Here, we present a dark-field TEM technique based on the use of an annular objective aperture. Our experiments demonstrate an increase in both contrast and signal-to-noise ratio in comparison to conventional bright-field TEM. This study presents the synthesis of gold-Pluronic core-shell nanoparticles by a two-step method and investigates their biological impact on cancer cells, specifically nanoparticle internalization and cytotoxicity. Uniform, 9-10-nm-sized, hydrophobic gold nanoparticles were synthesized in organic phase by reducing gold salt with oleylamine, after which oleylamine-protected gold nanoparticles were phase-transferred into aqueous medium using Pluronic F127 block copolymer, resulting in gold-Pluronic core-shell nanoparticles with a mean hydrodynamic diameter of ~35 nm. The formation and phase-transfer of gold nanoparticles were analyzed by UV-Vis absorption spectroscopy, transmission electron microscopy, and dynamic light scattering. The obtained gold-Pluronic core-shell nanoparticles proved to be highly stable in salted solution. Cytotoxicity tests showed no modification of cellular viability in the presence of properly purified particles. Furthermore, dark-field cellular imaging demonstrated that gold-Pluronic nanoparticles were able to be efficiently uptaken by cells, being internalized through nonspecific endocytosis. The high stability, proven biocompatibility, and imaging properties of gold-Pluronic core-shell nanoparticles hold promise for relevant intracellular applications, with such a design providing the feasibility to combine all multiple functionalities in one nanoparticle for simultaneous detection and imaging. A distributed fast-acquisition system for synchronized multi-technique experiments is presented, in which the collection of metadata and the asynchronous merging of large data volumes from multiple detectors are managed as part of the data collection process. This fast continuous scanning scheme, named FLYSCAN, enables measurement of microscopy data on a timescale of milliseconds per pixel. Proof-of-principle multi-technique experiments, namely scanning X-ray fluorescence spectrometry combined with absorption, differential phase contrast and dark-field imaging, have been performed on biological and geological samples.
