Domestic turkey production generally utilises only a few genetically improved lines, and local breeds are severely endangered as a result. Furthermore, the genetic resources of domestic turkeys have not been properly investigated, which could, ultimately, lead to the extinction of local breeds and negatively affect their corresponding genetic diversity and environmental adaptation. Although, several microsatellite markers have been designed for mapping and quantitative trait locus analysis, there is no standard panel of markers for genetic characterisation or genetic diversity assessment. Accordingly, the present study aimed to develop a set of polymorphic microsatellite markers that could be used for international turkey population studies. Thirty-nine microsatellites were selected based on polymorphism, DNA sequence and chromosome position, as well as on amplification efficiency, success rate and the absence of nonspecific amplification. The markers were screened using 105 DNA samples from local turkey breeds from Mexico, the United States, Italy, Brazil, Egypt and Spain. A total of 401 alleles were identified, with a mean number of alleles per marker of 10.28 ± 4.25. All microsatellites were polymorphic, with at least four alleles and no more than 19 alleles. Furthermore, allelic richness ranged from 3.810 to 17.985, mean heterozygosity ranged from 0.452 ± 0.229 to 0.667 ± 0.265, polymorphic information content values ranged from 0.213 (MNT264) to 0.850 (RHT0024) and the mean Fis value was 0.322. Overall, the panel was highly polymorphic and exhibited moderate Hardy–Weinberg disequilibrium, thereby indicating its value as a tool for biodiversity and population structure studies that could play an important role in promoting the conservation of local turkey breeds.Highlights Important genetic resources reside within indigenous turkey populations. These are linked to historic heritage production values and breeds. It is important to preserve this heritage and genetic diversity, which threatens to be lost as production systems focus on production characteristics. Microsatellite markers, even though, they are now replaced by single nucleotide polymorphism automatic genotyping platforms in many fields of genetics, remain a viable alternative thanks to their cheapness and simplicity of study which makes them particularly useful when the population to be studied lacks information of the prior genetic structure.

Purpose: This paper is a proposal for improving ESL text comprehension by merging a multimodal perspective on language (Kress, 2005) with an integrated psychological (or ‘metacognitive’) approach to language learning and awareness (Fairclough, 1992, p. 1). Design: A multichannel approach is presented, based on two strategies: a main ‘multichannel’ strategy for reading the text, which focuses on the assessment of the visual, auditory, cognitive and kinaesthetic/emotional channels and relative functions, and a corollary one, specifically dedicated to implementing the cognitive function. Methodology/ Approach: The approach has been extensively applied in university-level English classes (Reggio Emilia, Parma, Bologna University) using a ‘multichannel test’ based on students’ feedback to explore the effectiveness of the method. In this paper, a case study is offered: data have been collected with specific reference to the University of Bologna, A. Y. 2017/18 and 2018/2019. Originality/Value: Evidence suggests that further and diversified applications promise an empowerment of the approach and of its impact on ELT as well as on students’ awareness and satisfaction.

Through thousands of years of breeding and strong human selection, the dog (Canis lupus familiaris) exists today within hundreds of closed populations throughout the world, each with defined phenotypes. A singular geographic region with broad diversity in dog breeds presents an interesting opportunity to observe potential mechanisms of breed formation. Italy claims 14 internationally recognized dog breeds, with numerous additional local varieties. To determine the relationship among Italian dog populations, we integrated genetic data from 263 dogs representing 23 closed dog populations from Italy, seven Apennine gray wolves, and an established dataset of 161 globally recognized dog breeds, applying multiple genetic methods to characterize the modes by which breeds are formed within a single geographic region. Our consideration of each of five genetic analyses reveals a series of development events that mirror historical modes of breed formation, but with variations unique to the codevelopment of early dog and human populations. Using 142,840 genome‐wide SNPs and a dataset of 1,609 canines, representing 182 breeds and 16 wild canids, we identified breed development routes for the Italian breeds that included divergence from common populations for a specific purpose, admixture of regional stock with that from other regions, and isolated selection of local stock with specific attributes.

Livestock guarding dogs are a valuable adjunct to the pastoral community. Having been traditionally selected for their working ability, they fulfil their function with minimal interaction or command from their human owners. In this study, the population structure and the genetic differentiation of three Italian livestock guardian breeds (Sila's Dog, Maremma and Abruzzese Sheepdog and Mannara's Dog) and three functionally and physically similar breeds (Cane Corso, Central Asian Shepherd Dog and Caucasian Shepherd Dog), totalling 179 dogs unrelated at the second generation, were investigated with 18 autosomal microsatellite markers. Values for the number of alleles per locus, observed and expected heterozygosity, Hardy-Weinberg Equilibrium, F stats, Nei's and Reynold's genetic distances, clustering and sub-population formation abilities and individual genetic structures were calculated. Our results show clear breed differentiation, whereby all the considered breeds show reasonable genetic variability despite small population sizes and variable selection schemes. These results provide meaningful data to stakeholders in specific breed and environmental conservation programmes.

Columba liviais a wild bird whose domestication has led to a large number of pigeon breeds. The occasional loss or straying of domestic birds determined the origin of feral pigeons, which are now widespread all around the world. In this study, we assumed that the main contribution to feral populations is provided by domestic breeds reared in the same areas. We tested this hypothesis by analysing the variability of 12 microsatellite loci in nine Italian feral populations sampled in areas with different intensities of breeding and selecting domestic breeds. We included in the analysis samples belonging to domestic lineages commonly bred in Italy The pattern of geographic differentiation of feral populations turned out to be rather complex and only partially explained by the geographic distance between populations. This pattern can be understood only when the domestic breeds were included in the analysis. In particular, feral populations located in regions with a long-lasting tradition of pigeon breeding showed a high level of admixture with domestic breeds, in particular with Racing Homer and Piacentino. Ferals from Bolzano, Venice and Sassari were characterized by unique genetic components, almost all of which are not shared by other feral populations and by the considered domestic breeds. Our results further emphasize the complex origin of feral populations which can be properly investigated only by considering the pool of domestic pigeons bred in the considered area and their past and present distribution.

In this study, we aimed to characterise the genetic diversity of Italian pigeon breeds and describe the relationships among them and other European lineages, conjecturing which processes have led to actual breeds. We analysed the eight most diffused Italian pigeon breeds in addition to 11 foreign lineages that could have shared common ancestors or have been used in the creation of the Italian breeds. We analysed 12 autosomal microsatellite loci in 427 samples collected from 19 breeds belonging to six main groups of domestic pigeons (tumblers and highflyers, structure, owls, wattle, utility and hen). Genetic variability did not differ considerably among breeds, with an average observed heterozygosity (HO) of 0.550±0.072 (max=0.661 in breed Sottobanca; min=0.411 in breed Frillback). The 21.34% of total genetic variation found was partitioned among breeds. Italian pigeon breeds were assigned coherently to their respective groups of origin. The analysis supports the origin of homing pigeons from English Carrier and the existence of a close relationship between Old Dutch Capuchine and Italian Owl. Despite the differences in body size, pigeons of the breeds belonging to the hen group are genetically very similar. The sub-populations recognised by breeders inside Italian Owl and Triganino are not genetically supported and their identification as new breeds ought not to be yet proposed, although they are morphologically distinguishable. Consequently, it could also be discussed how the genetic characterisation of domestic lineages could give useful information in breeding and selection processes.

The chapter concerns Roman housing of the AD II in its noteworthy and rapid evolution which has its source in the political and economic conditions most favorable in the history of Rome. On the basis of some specific structures to be found in Rome and Ostia, the author makes an attempt at demonstrating to what extent this phenomenon was linked to the unifying housing policy in localities of high demographic density in the Empire, one that can already be glimpsed from the end or Nero's epoch. Subsequently, the composition solution common in the designs of new urban quarters are identified and discussed. In particular, the linear portico of the pillared type came to be introduced into the practice of urban planning, imposed by pre-constituted models. Taking into consideration principally the examples from Ostia, the author describes these particular typologies of porticos, comparing them with other structures and realities of the western part of the Mediterranean world.

According to recent archeological evidence, turkey (Meleagris gallopavo gallopavo) domestication may have occurred in Mexico around 2000 years ago. However, little is known about the phylogenetic and genealogical background underlying domestic turkey populations. This study aimed to further understand the domestication process and identify inter- or intraspecific connections between turkey populations to determine their origins, trace their global expansion, and define the species&rsquo, genetic value. Ninety-three domestic turkeys (local breeds) were sampled from populations in Brazil, Mexico, USA, Spain, Italy, Iran, and Egypt. Publicly available sequences from previous studies were also included. Standard mitochondrial DNA, genetic diversity, and haplotype network analyses were performed. Seventy-six polymorphic sites were identified. Turkeys from Mexico showed the greatest number of polymorphic sites (40), while turkeys from Italy and Brazil reported only one site each. Nucleotide diversity was also highest in Mexico and the USA (&pi, = 0.0175 and 0.0102, respectively) and lowest in Brazil and Italy. Of the six major haplogroups defined, the Mexican and USA populations appeared to have remained more stable and diverse than the other populations. This may be due to conservative husbandry policies in the rural areas of other populations, which have prevented the introduction of commercial turkey lines.

Genetic traceability has a key role in the product certification, but it is rarely implemented in sheep so far, especially in the fresh meat sector. In this study, the case of the Sambucana sheep is analysed with the aim of developing a genetic system able to certify the origin of its traditional product, the Sambucano lamb, protected by a registered trademark. A set of 14 microsatellite markers was identified as an efficient tool to genetically discriminate the Sambucana sheep from other breeds potentially involved in mislabelling and to allow for an effective allocation test of meat cuts labelled as ‘Guaranteed Sambucano lamb’. The paternity test proved to be an additional means to improve the reliability of the control. The traceability system here described is easy to implement in local minor sheep breeds and is recommended in the framework of meat certification.

Since its domestication, about 5000 years ago, the donkey (Equus asinus) has been extensively used as a work or draft animal in agricultural activities and for the transportation of people and goods. In the last century, technology improvement and growing mechanization strongly affected agriculture and the management and use of this livestock species in the industrialized countries. Nowadays, the use of donkeys for work or transport has almost disappeared, together with the need for mules or hinny breeding. During the last five decades, Italian autochthonous donkey populations suffered from a severe reduction in population size, which led to the extinction of several breeds. At present, eight breeds remain, all classified by FAO as critically endangered or endangered: Asinara, Pantesco, Grigio Siciliano, Romagnolo, Amiatino, Sardo Grigio, Martina Franca, and Ragusano. To evaluate the extant genetic variability of Italian donkeys, we typed 16 microsatellite loci in 258 individuals from these breeds. The results highlighted moderate levels of inbreeding ( F (IS) = 0.127) and a significant partition of genetic variation into breeds, as suggested by fixation index ( F (ST) = 0.109) and analysis of molecular variance (10.86% of total variation assigned to the between-breeds level) analyses. This was confirmed by a Bayesian clustering procedure that also highlighted a further partitioning at lower hierarchical levels corresponding to the farms of origin. This evidence suggests that an effective management strategy for Italian donkey populations should focus on breeds as conservation units. However, this requires a synergic management strategy at the farm level to maintain diversity and avoid inbreeding.

To understand the origin and genetic diversity of Italian horses, mitochondrial DNA D-loop sequences were generated for 163 horses from seven breeds. Sequence analysis of a 480-bp segment revealed a total of 84 haplotypes with 57 polymorphic sites, indicating multiple maternal origins and high genetic diversity. Comparison of the haplotypes with the equine mtDNA haplotype/haplogroup nomenclature showed a haplogroup distribution in the Italian breeds more similar to that found in the Middle East breeds than in the European breeds, probably due to the economic and cultural relationship with the Middle East in the past centuries.

Very little research into genetic diversity of Italian native dog breeds has been carried out so far. In this study we aimed to estimate and compare the genetic diversity of four native Italian shepherd dog breeds: the Maremma, Bergamasco, Lupino del Gigante and Oropa shepherds. Therefore, some cosmopolitan dog breeds, which have been widely raised in Italy for a long time past, have also been considered to check possible influence of these dog populations on the Italian autochthonous breeds considered here. A total of 212 individuals, belonging to 10 different dog breeds, were sampled and genotyped using 18 autosomal microsatellite loci. We analyzed the genetic diversity of these breeds, within breed diversity, breed relationship and population structure. The 10 breeds considered in this study were clearly genetically differentiated from each other, regardless of current population sizes and the onset of separate breeding history. The level of genetic diversity explained 20% of the total genetic variation. The level of H E found here is in agreement with that found by other studies. The native Italian breeds showed generally higher genetic diversity compared with the long established, well-defined cosmopolitan dog breeds. As the Border Collie seems closer to the Italian breeds than the other cosmopolitan shepherd dogs considered here, a possible utilization of this breed to improve working performance in Italian traditional working shepherd dogs cannot be ignored. The data and information found here can be utilized in the organization of conservation programs planned to reduce inbreeding and to minimize loss of genetic variability.

To identify genes with effects on meat quality and production traits we developed an adult porcine skeletal muscle cDNA library. After pre-screening this library with seven genes highly expressed in skeletal muscle, 385 non-hybridizing clones were sequenced from both ends to yield 510 expressed sequence tags (ESTs). Together with those ESTs previously generated from this library, we have produced 701 porcine skeletal muscle ESTs. These ESTs were grouped into 306 different cDNA species and compared with the human skeletal muscle transcriptional profiles obtained from different databases. Furthermore we mapped 107 of these cDNAs using a somatic cell hybrid panel with genes mapping over all the autosomes (except on chromosome 11) and on chromosome X. The mapping of these cDNAs contributed to the construction of a first genomic transcript map of the skeletal muscle tissue in pig.

Con la presente ricerca abbiamo studiato il livello di espressione di geni nel tessuto muscolare di suino con la tecnica fluoro differential display, per identificare geni espressi in modo variabile in funzione del parametro calo di prima salagione. Sono stati identificati dieci geni, dei quali sei a funzione nota e quattro non ancora caratterizzati, che mostravano un’espressione maggiore negli animali con il calo di prima salagione più alto. - Con la presente ricerca abbiamo studiato il livello di espressione di geni nel tessuto muscolare di suino con la tecnica fluoro differential display, per identificare geni espressi in modo variabile in funzione del parametro calo di prima salagione. Sono stati identificati dieci geni, dei quali sei a funzione nota e quattro non ancora caratterizzati, che mostravano un’espressione maggiore negli animali con il calo di prima salagione più alto.

Chromosomal assignments are reported for fourteen porcine expressed sequence tags (ESTs)--CALM1, CRYAB, MYH7, MYL1, PDK4, PGAM2, PYGM, REV3L, RFC1, SLN, SPTBN1, SRM160, TPM1 and YWHAG. The ESTs were derived from our porcine skeletal muscle cDNA library. The ESTs sequences selected for mapping included the presence of the 3'-untranslated region. The assignments were performed using two independent somatic cell hybrid panels providing the possibility of confirmation of the results obtained. The observed localizations are compared with the locations predicted from heterologous (human-pig, pig-human) chromosome painting data and knowledge of the map locations of the human homologues. These results add new information to the porcine genome transcript map.

The aim of this study was to provide an overview on breed's characteristics of a small dairy goat population recently identified in the Piedmont region (NW Italy): the Grey Goat of Lanzo Valleys. This goat, locally named Fiurina, is composed of about 150 heads. Increasing knowledge on threatened breeds is an effective tool to develop conservation programs aiming at preserving loss of genetic resources, economically valorizing animal-derived food products, maintaining traditions and cultural values, as well as appropriately managing native habitats. Data and samples were collected in 15 representative farms to obtain information on geographical distribution, breeding systems, morphometric measures, and genetic diversity. Milk traits, including fatty acids profile and caseins polymorphism, were also investigated. The prevalent breeding system is extensive or semi-extensive, mostly with vertical transhumance from lowland to alpine pastures during the grazing season. The breed is characterized by a peculiar color of the fleece (mixture of white, grey and black course outer hairs with brown under-down) from which the local name “Fiurina” (=speckled) derives. The breed is of medium size, with a quite high frequency of well-developed and turned backward horns. More than 80% of does have pear-shaped udder, typical of goats, with cylindrical teats directed downward. The microsatellite analysis pointed out significant distances between Fiurina and other goat breeds reared in the same area. Consequently, Fiurina has to be considered as a unique breed. Although highly variable, quite appreciable milk yields were observed. Considering milk gross composition and fatty acids profile, Fiurina goat showed comparable results to other local breeds reared in alpine environment and mainly fed with fresh and conserved forages. A remarkable amount of α-linolenic acid (0.82 g 100 g−1 fat) was detected. The breed is also characterized by an interesting and wide variability in the casein cluster, with some haplotypes (i.e., A-C-F-C′, E-A-C-B, F-C1-F-C′) detected only in Fiurina, confirming the genetic uniqueness of the breed. The balanced frequency of medium-strong and weak-null CSN1S1 alleles could be exploited for different breeding strategies.

Hybridization and introgression can impact the evolution of natural populations. Several wild canid species hybridize in nature, sometimes originating new taxa. However, hybridization with free-ranging dogs is threatening the genetic integrity of grey wolf populations (Canis lupus), or even the survival of endangered species (e.g., the Ethiopian wolf C. simensis). Efficient molecular tools to assess hybridization rates are essential in wolf conservation strategies. We evaluated the power of biparental and uniparental markers (39 autosomal and 4 Y-linked microsatellites, a melanistic deletion at the β-defensin CBD103 gene, the hypervariable domain of the mtDNA control-region) to identify the multilocus admixture patterns in wolf x dog hybrids. We used empirical data from 2 hybrid groups with different histories: 30 presumptive natural hybrids from Italy and 73 Czechoslovakian wolfdogs of known hybrid origin, as well as simulated data. We assessed the efficiency of various marker combinations and reference samples in admixture analyses using 69 dogs of different breeds and 99 wolves from Italy, Balkans and Carpathian Mountains. Results confirmed the occurrence of hybrids in Italy, some of them showing anomalous phenotypic traits and exogenous mtDNA or Y-chromosome introgression. Hybridization was mostly attributable to village dogs and not strictly patrilineal. The melanistic β-defensin deletion was found only in Italian dogs and in putative hybrids. The 24 most divergent microsatellites (largest wolf-dog FST values) were equally or more informative than the entire panel of 39 loci. A smaller panel of 12 microsatellites increased risks to identify false admixed individuals. The frequency of F1 and F2 was lower than backcrosses or introgressed individuals, suggesting hybridization already occurred some generations in the past, during early phases of wolf expansion from their historical core areas. Empirical and simulated data indicated the identification of the past generation backcrosses is always uncertain, and a larger number of ancestry-informative markers is needed.

Summary We report on a complete genome scan for quantitative trait loci (QTL) affecting milk protein percentage (PP) in the Italian Holstein-Friesian cattle population, applying a selective DNA pooling strategy in a daughter design. Ten Holstein-Friesian sires were chosen, and for each sire, about 200 daughters, each from the high and low tails of estimated breeding value for PP, were used to construct milk DNA pools. Sires and pools were genotyped for 181 dinucleotide microsatellites covering all cattle autosomes. Sire marker allele frequencies in the pools were obtained by shadow correction of peak height in the electropherograms. After quality control, pool data from eight sires were used for all subsequent analyses. The QTL heterozygosity estimate was lower than that of similar studies in other cattle populations. Multiple marker mapping identified 19 QTL located on 14 chromosomes (BTA1, 2, 3, 4, 5, 6, 8, 9, 12, 14, 17, 20, 23 and 27). The sires were also genotyped for seven polymorphic sites in six candidate genes (ABCG2, SPP1, casein kappa, DGAT1, GHR and PRLR) located within QTL regions of BTA6, 14 and 20 found in this study. The results confirmed or excluded the involvement of some of the analysed markers as the causative polymorphic sites of the identified QTL. The QTL identified, combined with genotype data of these candidate genes, will help to identify other quantitative trait genes and clarify the complex QTL patterns observed for a few chromosomes. Overall, the results are consistent with the Italian Holstein population having been under long-term selection for high PP.

Catria is 1 of the 22 native Italian horse breeds that now survive from a larger number. Thirty individuals, representative of the Catria horse, were analyzed for 11 microsatellites and compared with data of 10 breeds reared in Italy. Three different approaches, genetic distances, correspondence analysis, and clustering methods, were considered to study genetic relationships among Catria and the other horse populations. Genetic differentiation among breeds was highly significant (P < 0.01) for all loci. Average F(ST) values indicate that around 10% of the total genetic variation was explained by the between-breed differences and the 3 approaches utilized gave similar results. Italian native breeds are clearly separated from the other examined breeds. However, by the correspondence analysis, the Catria appears closer to Maremmano and Murgese. The results of Bayesian approaches give further information showing for Catria a common origin with Maremmano and Italian Heavy Draught. Genetic relationships among Catria and the other breeds are consistent with the breed's documented history. The data and information found here can be utilized in the organization of conservation programmes planned to reduce inbreeding and to minimize loss of genetic variability.

Porcine skeletal muscle genes play a major role in determining muscle growth and meat quality. Therefore, to progress towards a better understanding of the genetic factors influencing these traits, the first step is to characterize the genes expressed in skeletal muscle tissue in pig. To this aim, we constructed a porcine biceps femoris muscle cDNA library and sequenced 111 randomly isolated clones. By FASTA analysis, we identified 72 unique clones: 47 showed homology to previously identified genes in human or other mammals, 20 matched uncharacterized expressed sequence tags (ESTs), two showed no significant matches to sequences already present in DNA databases, and three other clones containing only repetitive elements were excluded from further analysis. Mitochondrial genes (16.2%), myosin heavy chain genes (9%) and the actin α skeletal muscle gene (9%) were the most abundant transcripts. Among the 47 identified genes several muscle-specific or predominant sequences expressed in skeletal muscle were found. The sequences of the clones matching uncharacterized human, mouse or porcine ESTs were tested by GRAIL in order to identify putative coding regions. The results of our analysis allowed the establishment of a first list of genes expressed in porcine skeletal muscle.

The present paper describes the isolation and linkage mapping of two isoforms of skeletal muscle myosin heavy chain in pig. Two partial cDNAs (pAZMY4 and pAZMY7), coding for the porcine myosin heavy chain-2B and -beta respectively, have been isolated from a pig skeletal muscle cDNA library. Four RFLPs were detected with the putative porcine skeletal myosin heavy chain-2B probe (pAZMY4) and one RFLP was identified with the putative myosin heavy chain-beta probe (pAZMY7). Two myosin heavy chain loci were mapped by linkage analysis performed with the five RFLPs against the PiGMaP linkage consortium ResPig database: the MYH1 locus, which identifies the fast skeletal muscle myosin heavy chain gene cluster, was located at the end of the map of porcine chromosome 12, while the MYH7 locus, which identifies the myosin heavy chain-alpha/-beta gene cluster, was assigned to the long arm of porcine chromosome 7.

Sources/description: Expressed sequence tags, European Molecular Biology Laboratory (EMBL) accession numbers AJ301262, AJ301107, and AJ301170, corresponding respectively to portions of the porcine muscle creatine kinase (CKM), peroxisomal enoyl coenzyme A hydratase 1 (ECH1), and skeletal slow troponin T1 (TNNT1) genes were isolated from an adult porcine skeletal muscle cDNA library. Polymerase chain reaction primers for CKM and ECH1 were designed in the 3¢-untranslated region to amplify fragments of 207 bp and 104 bp, respectively. For TNNT1, primers were designed in the coding sequence, each on a different exon, according to the organization of the human and mouse TNNT1 gene. Partial sequencing confirmed that the obtained amplicon of about 490 bp contained portions of exons 8 and 9 and the intervening intron of the porcine TNNT1 gene (EMBL accession number AJ495809).

Evidence was provided for a new erythrocyte ADA allele (Aw) in the Swedish Landrace breed of pigs. The enzyme activity of Aw variant was considerably less than that of the previously reported erythrocyte ADA variants A and B. The mobility of the Aw variant was identical to that of the A variant, both by electrophoresis and by isoelectric focusing. The in vitro heat stability (at 56 degrees C) of the Aw variant was similar to that of A and B variants. The leucocyte ADA activity was normal in pigs with the erythrocyte ADA Aw variant. The frequencies of the erythrocyte ADA alleles, A, B, Aw, and O (silent allele) in four breeds of pigs in Sweden were: Landrace--0.51, 0.19, 0.11, 0.19; Large White--0.91, 0.03, 0.0, 0.06; Duroc--0.95, 0.05, 0.0, 0.0; Hampshire--0.12, 0.14, 0.0, 0.74.

To investigate the mitochondrial DNA diversity of five Italian donkey breeds (Amiata, Martinafranca, Romagnolo, Asinara, and Ragusano), we sequenced the HVR I region (D-loop, 288 bp) and cytochrome b gene (274 bp) in 121 individuals. In the D-loop we found nineteen mutations corresponding to fourteen different haplotypes, while in cyt b coding gene only six mutations were found, originating five different haplotypes. In particular, three mutations out of six were non-synonymous, causing an aminoacidic substitution. About the D-loop region, the value of nucleotide diversity (π) observed within breeds was relatively low, but not far from values detected in other European breeds. Phylogenetic and network analyses disclosed the presence of two divergent maternal lineages within Italian donkeys. These haplogroups correspond to the well known lineages of ancestors (Equus asinus somaliensis and E. a. africanus), as donkeys were domesticated from distinct wild subspecies living in Eastern Africa regions. ...

Many studies have demonstrated that quantitative trait loci (QTL) can be identified and mapped in commercial dairy cattle populations using genetic markers in daughter and granddaughter designs.The final objective of these studies is to identify genes or markers that can be used in breeding schemes via marker assisted selection (MAS).

The genetic structure of Ventasso Horse (VH) was investigated using 12 microsatellites. The analyses were carried out on 117 VH individuals and the results were compared with those obtained analysing 11 other breeds reared in Italy. All microsatellites were polymorphic in VH and in the other breeds. A total of 124 alleles (from 6 to 19 alleles per microsatellite) were detected. Average heterozygosity was 0.743 in VH and ranged from 0.613 to 0.759 in the other breeds. The mean FST value had an average value of 0.0932. Genetic distances were calculated using Nei’s standard genetic distance (Ds). The smallest Ds values were found between VH and Anglo-Arab, Thoroughbred, Maremmano and Lipizzan horse breeds. Phylogenetic trees constructed using neighbour-joining method showed two clear separate clusters: the first includes Bardigiano, Haflinger and Italian Heavy Draught Horse, the second contains the other 9 breeds.

Bovine chromosome 20 (BTA20) was studied to identify QTL for milk yield and protein percentage in the Italian Holstein-Friesian breed using a selective milk DNA pooling strategy in a daughter design with sire haplotype analysis. Several QTL were identified. The effect of the known GHR F279Y and PRLR S18N mutations were in for the most part confirmed. However, it was also shown that these markers cannot explain all significant effects observed on BTA20 for the investigated traits.

Starting from 3D surveys carried out with laser scanners and photogrammetry of two of the most famous examples of Hadrian’s domes (octagonal hall of the Piccole Terme and vestibule of the Piazza d’Oro) studies on their design have been conducted. In particular, through the comparison of ichnographia and orthographia of these emblematic examples of architectural experimentation, common elements and features have emerged which, thanks to the integrated use of new measurement technologies, have allowed us to hypothesize structural dimensioning systems of the piers together to possible criteria for defining the intrados-extrados relationship. The use, by Hadrian’s architects, of precise grid-based designs to facilitate the achievement of quantities (required areas and volumes) emerges clearly, as well as modules aimed at defining alignments of horizontal and vertical structures: useful elements for an integrated reading of the individual buildings to support the archaeological investigation.
