Photodynamic therapy (PDT) is an emerging alternative cancer treatment modality that utilizes photo-sensitivity to cause cell death upon photo-irradiation. However, PDT efficiency has been hampered by tumor hypoxia, blue-shifted excitation wavelengths, and the high dark toxicity of photo-sensitizers. We designed and synthesized two novel porphycene-based photosensitizers (TBPoS-OH and TBPoS-2OH) with potent photo-cytotoxicity and a LD50 in the nM range under both normoxic and hypoxic conditions in a variety of cell types after photo-irradiation (λ = 640 ± 15 nm). Further studies showed fast-cellular uptake for TBPoS-OH that localized lysosomes and subsequently induced cell apoptosis via the lysosomal-mitochondrial pathway. Moreover, TBPoS-OH significantly reduced tumor growth in two xenografted mouse models bearing melanoma A375 and B16 cells. Finally, TBPoS-OH exhibited no obvious immunogenicity and toxicity to blood cells and major organs in mice. These data demonstrated that these two porphycene-based photosensitizers, especially TBPoS-OH, could be developed as a potential PDT modality.

Porphyrins are good photodynamic therapy (PDT) agents due to its flexibility for modifications to achieve tumor localization and photo-cytotoxicity against cancer. Yet they are not perfect. In a Ru(polypyridyl)-porphyrin system, the Ru(polypyridyl) moiety improves the water solubility and cell permeability. Consider the similar excited state energies between Ru(polypyridyl) and porphyrin moieties; a small perturbation (e.g. Zn(II) metalation) would lead to a marked change in the energy migration process. In this work, we have synthesized a series of porphyrins conjugated with Ru(polypyridyl) complexes using different linkers and investigated their photophysical properties, which included singlet oxygen quantum yield and their in vitro biological properties, resulting from linker variation and porphyrin modification by Zn(II) metalation.

We have synthesized a bifunctional erbium–porphyrin tumor imaging and PDT agent (Er–R3) that is capable of killing bladder cancer cellsviaits selective binding to the integrin αvβ3isoform overexpressed on the cell membrane.

The new amphiphilic BODPY-porphyrin conjugate BZnPP and its precursor BZnPH were synthesised, and their linear and two-photon photophysical properties, together with their cellular uptake and photo-cytotoxicity, were studied. This amphiphilic conjugate consists of a hydrophobic BODIPY moiety and a hydrophilic tetra(ethylene glycol) chain bridging a cationic triphenylphosphonium group to an amphiphilic porphyrin ZnP through acetylide linkers at its meso positions. A large two-photon absorption cross-section (σ=1725 GM) and a high singlet oxygen quantum yield (0.52) were recorded. Intense linear- and two-photon-induced red emissions were also observed for both BZnPP and BZnPH. Further in vitro studies showed that BZnPP exhibited very efficient cellular uptake and strong photocytotoxic but weak dark cytotoxic properties towards human breast carcinoma MCF-7 cells. In summary, the two-photon-induced emission and the potent photo-cytotoxicity of BZnPP make it an efficacious dual-purpose tumour-imaging and photodynamic therapeutic agent in the tissue-transparent spectral windows.

Biological systems have developed an intact network and strategies in response to various environmental pressures such as irradiation, viral invasion and oxidative stress. Therefore, elucidation of the cellular response mechanism toward oxidative stress can contribute to the knowledge of redox regulation. By using a newly developed gadolinium based photodynamic therapy (PDT) agent Gd-N and SILAC quantified proteomic analysis, we observed 485 proteins dysregulated in expression, 106 in phosphorylation and 1050 in oxidation. Interestingly, lysosome was discovered as the main organelle affected by Gd-N induced singlet oxygen, along with the down regulation of a majority of lysosomal acid hydrolases and proton pump complex ATP6V/TCIRG1. Besides, phosphorylation sites with sequence patterns "TP" or "SP" were enriched in dysregulated phosphoproteins. Protein oxidation also shows sequence patterns in target proteins with "M.D" or "KM" taking methionine as the central residue. Oxidized proteins were most enriched in the pathways of Parkinson's disease, an oxidative stress closely related neurodegenerative disease. In conclusion, our study reveals new insights into the cellular mechanism to oxidative stress and may contribute to the discovery of new targets and development of novel PDT agents.

A water-soluble light-responsive antitumor agent, PtEuL, based on a cisplatin-linked europium-cyclen complex has been synthesized and evaluated for controlled cisplatin release by linear/two-photon excitation in vitro with concomitant turn-on and long-lived europium emission as a responsive traceable signal.

We report herein a novel iridium(III) complex 1 as an antitumour necrosis factor agent and the first metal-based inhibitor of TACE enzymatic activity. Complex 1 inhibited TNF-α secretion and p38 phosphorylation in human monocytic THP-1 cells.

The hypoxia inducible factor (HIF) pathway has been considered to be an attractive anti-cancer target. One strategy to inhibit HIF activity is through the disruption of the HIF-1α–p300 protein-protein interaction. We report herein the identification of an osmium(II) complex as the first metal-based inhibitor of the HIF-1α–p300 interaction. We evaluated the effect of complex 1 on HIF-1α signaling pathway in vitro and in cellulo by using the dual luciferase reporter assay, co-immunoprecipitation assay, and immunoblot assay. Complex 1 exhibited a dose-dependent inhibition of HRE-driven luciferase activity, with an IC50 value of 1.22 μM. Complex 1 interfered with the HIF-1α–p300 interaction as revealed by a dose-dependent reduction of p300 co-precipitated with HIF-1α as the concentration of complex 1 was increased. Complex 1 repressed the phosphorylation of SRC, AKT and STAT3, and had no discernible effect on the activity of NF-κB. We anticipate that complex 1 could be utilized as a promising scaffold for the further development of more potent HIF-1α inhibitors for anti-cancer treatment.

A novel porphyrin–polypyridyl ruthenium(II) conjugate (TPP–Ru), in which the ruthenium(II) polypyridyl moiety is linked to the β-position of the tetraphenylporphyrin via a Schiff base linkage, has been synthesized and characterized by 1H NMR, HRMS and UV–visible spectroscopy. The relative singlet oxygen quantum yield and two-photon absorption cross-section of this conjugate, together with its photo-induced DNA cleavage and cytotoxic activities were measured. The results show that the amphiphilic ruthenium(II) polypyridyl–porphyrin conjugate is an effective DNA photocleavage agent, with potential application in one- and two-photon absorption anti-cancer photodynamic therapy.

A series of luminescent Ir(III) complexes were synthesised and evaluated for their ability to act as G-quadruplex-selective probes. A novel Ir(III) complex was found to be highly selective for G-quadruplex DNA, and was employed in a label-free G-quadruplex-based detection assay for nicking endonuclease activity in aqueous solution. A proof-of-concept of this probe has been demonstrated by using Nb·BbvCI as a model enzyme. In this assay, a DNA substrate comprised of oligonucleotides ON1 (5′-GTG3TAG3CG3T2G2CTGAG2TGA-3′) and ON2 (5′-TCAC2TCAGC2A2C2-3′) initially exists in a duplex conformation, resulting in a low luminescence signal due to the weak interaction between the Ir(III) complex and duplex DNA. Upon cleavage by Nb·BbvCI, the guanine-rich sequence is released and folds into a G-quadruplex, which greatly enhances the luminescence of the Ir(III) probe. This method was highly sensitive for Nb·BbvCI over other DNA-modifying enzymes.

In this paper, aiming at the trans-membrane transport properties of the different DLCs related with the cytotoxicities, we have studied photophysics of DLCs in RMs. MTT assays indicated that DLC 1 were more cytotoxic than DLC 2 and 3. Steady-state absorption and fluorescence method have been used to characterize the binding model of three DLCs (1, 2, and 3) in RMs and bovine serum albumin (BSA). The progressive red-shift of compound 1 indicated that it can experience the hydrophilic and hydrophobic environment owing to the rigid structure, thus it can more easily cross the double membrane of cell than compounds 2 and 3. In conclusion, we simulated a model of compound 1 in RMs. The present study of F16 derivatives in microenvironment would provide some useful information to the cell membrane simulation and design of DLCs.

Targeting STAT5 is an appealing therapeutic strategy for the treatment of hematologic malignancies and inflammation. Here, we present the novel osmium(II) complex 1 as the first metal-based inhibitor of STAT5B dimerization. Complex 1 exhibited superior inhibitory activity against STAT5B DNA binding compared to STAT5A DNA binding. Moreover, 1 repressed STAT5B transcription and blocked STAT5B dimerization via binding to the STAT5B protein, thereby inhibiting STAT5B translocation to the nucleus. Furthermore, 1 was able to selectively inhibit STAT5B phosphorylation without affecting the expression level of STAT5B.

5,10,15,20-tetrakis(4-amidinophenyl)porphyrin (Por 1), its Zn complex (Por 2) and its conjugate with a tetraphenylporphyrin to form a bisporphyrin (Por 3) were prepared. The monomeric Por 1 and Por 2 showed both intercalative and external binding with DNA whereas only external DNA binding was seen in the bisporphyrin, Por 3 by circular dichroism and UV-vis. The DNA photocleavage activities of these porphyrins followed the order: Por 1 ~ Por 2 > Por 3, which did not correlate with their measured 1O2 production rates. It suggests 4-amidinophenylporphyrins are promising new photodynamic therapeutic agents.

We report an in-depth study on meso-aryl nitration of tetraphenylporphyrin. In contrast to previous studies, new evidence reveals that tetrakis(p-nitrophenyl) derivative can be obtained as a major product. Successful isolation of the barely soluble product toward a remarkable yield of nearly 90% has been reached by means of a solid phase extraction technique. Distribution of different nitro-porphyrin components is reassessed with respect to varying acid content in the reactions. An ortho-effect model is proposed to describe the formation mechanism.

A series of cyanine fluorophores based on fused aromatics as an electron donor for DNA sensing and two-photon bioimaging were synthesized, among which the carbazole-based biscyanine exhibits high sensitivity and efficiency as a fluorescent light-up probe for dsDNA, which shows selective binding toward the AT-rich regions. The synergetic effect of the bischromophoric skeleton gives a several-fold enhancement in a two-photon absorption cross-section as well as a 25- to 100-fold enhancement in two-photon excited fluorescence upon dsDNA binding.

Dyads of a N-confused porphyrin (NCP) moiety covalently linked to a porphyrin free-base (H 2 P) or a zinc(II) porphyrinate (ZnP) moiety via a flexible alkyl chain of variable length have been synthesized. Photoluminescence study demonstrated an efficient excitation energy transfer from H 2 P/ZnP moiety to the NCP moiety. Measurement of the near-IR emission of singlet oxygen produced by these dyads via photosensitization showed that the NCP–ZnP dyads ( Ф Δ = (0.61–0.65) ± 0.13) were better 1 O 2 generators than the NCP–P dyads ( Ф Δ = (0.36–0.41) ± 0.08).

An amphiphilic porphyrin appended with a Ru(II)–polypyridyl complex (Ru–P) showing a moderate two-photon absorption cross-section (178.0 ± 26.8 GM), high singlet oxygen quantum yield and rapid cellular uptake was synthesized. In vitro study using human nasopharyngeal carcinoma cells showed that Ru–P exhibited a strong two-photon induced fluorescence upon uptake, lysosomal localization and potent two-photon induced cytotoxicity. These results show that Ru–P, which was designed to enhance its cellular uptake, can potentially be used as an efficacious bifunctional two-photon tumor-imaging and photodynamic therapeutic agent despite its moderate two-photon absorption cross-section.

A series of (4-cyanophenyl)porphyrins have been prepared and structurally characterized by 1H NMR, IR and UV/Vis spectroscopy, mass spectrometry, and elemental analysis. The relative singlet-oxygen quantum yields, two-photon absorption cross-sections and DNA photocleavage activities of these porphyrins were measured. The results show that (4-cyanophenyl)porphyrins with a properly designed hydrophobic/hydrophilic balance for better cellular uptake have potential application in two-photon-absorption photodynamic therapy. A novel iridium(III) complex was synthesized and evaluated for its ability to target JMJD2 enzymatic activity. The iridium(III) complex 1 can inhibit JMJD2 activity and was selective for JMJD2 activity over JARID, JMJD3, and HDAC activities. Moreover, 1 suppressed the trimethylation of the p21 promoter on H3K9me3 and interrupted the JMJD2D–H3K9me3 interactions in human cells, suggesting that it could act as an epigenetic modulator. To our knowledge, 1 represents the first metal-based JMJD2 inhibitor reported in the literature.

[structure: see text] The fluorescent photoinduced electron transfer (PET) chemosensors 1-3 were synthesized from cholic acid. 1 and 2 containing amidothiourea groups as anion receptive sites demonstrated much higher affinity toward anions than 3 containing traditional thiourea H-bond donating group. Comparative studies on their binding affinity toward carboxylates, dihydrogen phosphate, and halides revealed that the amidothiourea moiety on the C17 side chain could work cooperatively with H-bond donating groups on C7 and C12 to bind spherical halogen anions. An unexpected specific fluorescence enhancement of 1 by coordinating bromide ion was observed.

A water-soluble porphyrinato ytterbium complex linked with rhodamine B (Yb-2) showed mitochondria-specific subcellular localization and strong two-photon-induced NIR emissions (λ(em) = 650 nm, porphyrinate ligand π → π* transition; λ(em) = 1060 nm, Yb(III) (5)F(5/2) → (5)F(7/2) transitions; σ(2) = 375 GM in DMSO) with an impressive Yb(III) NIR emission quantum yield (1% at λ(ex) = 340 nm; 2.5% at λ(ex) = 430 nm) in aqueous solution.

We report herein the synthesis and application of a series of novel cyclometalated iridium(III) complexes 1-3 bearing a rhodamine-linked NˆN ligand for the detection of Cu(2+) ions. Under the optimised conditions, the complexes exhibited high sensitivity and selectivity for Cu(2+) ions over a panel of other metal ions, and showed consistent performance in a pH value range of 6 to 8. Furthermore, the potential application of this system for the monitoring of Cu(2+) ions in tap water or natural river water samples was demonstrated.

Two new 1,2,3,5-dithiadiazoles, [4-(4′-C5H4N)CN2S2] (L1) and [4-(3′-C5H4N)CN2S2] (L2), with different pyridyl groups at the 4-position were prepared. As Lewis bases, the dithiadiazoles reacted with Lewis acids via their pyridyl groups to form acid-base adducts with retention of the five-membered dithiadiazole ring. Ligands L1 and L2 reacted with excess triethylborane to give L1BEt3 and L2BEt3 (1a and 1b) and with Mn(CO)5Br to give the fac-Mn(CO)3Br(L1)2 (2a) and fac-Mn(CO)3Br(L2)2 (2b) complexes. When reacted with Pd0 complexes, namely, Pd(PPh3)4 and a mixture of [Pd2(dba)3] and dppf, the dithiadiazoles underwent ring-opening reaction by reductive cleavage of the S–S bond of the dithiadiazole ring. The resulting formation of Pd–S bonds gave the corresponding trinuclear complexes [Pd3{μ-SNC(4′-C5H4N)NS-S,S′}2(PPh3)4] (3a), [Pd3{μ-SNC(3′-C5H4N)NS-S,S′}2(PPh3)4] (3b), and [Pd3{μ-SNC(4′-C5H4N)NS-S,S′}2(dppf)2] (4) [dba = dibenzylideneacetone, dppf = 1,1′-bis(diphenylphosphanyl)ferrocene]. Complexes 3a and 3b reacted with excess triethylborane to give the corresponding acid-base adducts [Pd3{μ-SNC(4′-C5H4NBEt3)NS-S,S′}2(PPh3)4] (5a) and [Pd3{μ-SNC(3′-C5H4NBEt3)NS-S,S′}2(PPh3)4] (5b.) Complex 3a reacted with Mn(CO)5Br to give {[Pd3{μ-SNC(4′-C5H4N)NS-S,S′}2(PPh3)4][MnBr(CO)3]}n (6). The structures of 3a, 5a, and 5b were established by X-ray crystallography.

The chemiluminescence of 1DLgO2 expelled from the UV photolysis of vanadium(V)-peroxo (VP) complexes in aqueous solution was observed for the first time. Using phenalenone photosensitization as a standard reference, the 1O2 quantum yields from the photolysis of 12 VP complexes were measured. No good correlation was found between the relative DNA-photocleavage activities and the 1O2 quantum yields of these complexes. This lack of correlation underscores the significant contribution of the complex-DNA binding interaction in determining the observed DNA-photocleavage activity of the complex. The small 1O2 quantum yields observed in these complexes were interpreted in terms of the minor 1O2 pho-toelimination channel relative to other more efficient relaxation processes.

An optode device that contains two optical ion-selective membranes for potassium and sodium was used to obtain optode spectra of mixtures of these two ions. Such spectra are a non-linear function of the concentrations of the two ions. A back-propagation artificial neural network (BP-ANN) model was used to analyse these mixture optode spectra. The results showed that the BP-ANN technique is satisfactory for treating such non-linearity embedded in the data. In addition, an index for addressing the quality of the model approximation was defined and shown to be highly effective in controlling over-fitting in BP-ANN training.

Six water-soluble free-base porphyrin-Ru(II) conjugates, 1-3, and Zn(II) porphyrin-Ru(II) conjugates, 4-6, with different linkers between the hydrophobic porphyrin moiety and the hydrophilic Ru(II)-polypyridyl complex, have been synthesized. The linear and two-photon-induced photophysical properties of these conjugates were measured and evaluated for their potential application as dual in vitro imaging and photodynamic therapeutic (PDT) agents. Conjugates 1-3, with their high luminescence and singlet oxygen quantum yields, were selected for further study of their cellular uptake, subcellular localization, and cytotoxic and photocytotoxic (under linear and two-photon excitation) properties using HeLa cells. Conjugate 2, with its hydrophobic phenylethynyl linker, was shown to be highly promising for further development as a bifunctional probe for two-photon (NIR) induced PDT and in vitro imaging. Cellular uptake and subcellular localization properties were shown to be crucial to its PDT efficacy.

Two axially ligated rhodamine-Si(IV)-phthalocyanine (Rh-SiPc) conjugates, bearing one and two rhodamine B, were synthesized and their linear and two-photon photophysical, subcellular localization and photocytotoxic properties were studied. These Rh-SiPc conjugates exhibited an almost exclusive mitochondrial localizing property in human nasopharyngeal carcinoma (HK-1) cells and human cervical carcinoma (HeLa) cells. Strong photocytotoxic but low dark cytotoxic properties were also observed for the two Rh-SiPc conjugates toward the HK-1 cells. Using nuclei staining method and flow cytometric DNA content analysis, apoptotic cell death was induced by these conjugates upon photoactivation. This observation is consistent with their mitochondrial localization property. The observed properties of these conjugates qualify them as promising PDT agents.

Two Ru(II) polypyridyl-porphyrin and Zn(II) porphyrin conjugates (Ru-L and Ru-Zn-L) have been synthesized and their photophysical properties studied. The two conjugates, which contained a hydrophobic tetraphenylporphyrin L conjugated via an acetylide linker at its β-position with a hydrophilic Ru(II) polypyridyl complex, showed high singlet oxygen quantum yields (>70%) and substantial two-photon absorption cross-sections (∼500 GM). Ru-L gave strong emissions at ∼660 and ∼733 nm through linear or two-photon excitation. Solvatochromism was observed in the fluorescence spectra of Ru-L and Ru-Zn-L, where in less polar solvents (i.e., toluene and dichloromethane) their fluorescence emissions became slightly blue-shifted with a 3-fold reduction in intensity relative to those observed in polar solvents (i.e., acetonitrile and methanol). Cell-based studies of these complex conjugates were conducted using human nasopharyngeal carcinoma HK-1 and cervical carcinoma HeLa cells on which Ru-L showed rapid cellular uptake, low dark-cytotoxicity, and high photo-cytotoxicity. Furthermore, Ru-L can be excited and emits in the “biological window” in vitro, making it a potential potent new generation photodynamic therapeutic agent capable of singlet oxygen generation and in vitro near-infrared emission.

A series of cationic lanthanide porphyrinate complexes of the general formula [(Por)Ln(H(2)O)(3)](+) (Ln(3+)=Yb(3+) and Er(3+)) were synthesized in moderate yields through the interaction of meso-pyridyl-substituted porphyrin free bases (H(2)Por) with [Ln{N(SiMe(3))(2)}(3)]·x[LiCl(thf)(3)], and the corresponding neutral derivatives [(Por)Ln(L(OMe))] (L(OMe)(-)=[(η(5)-C(5)H(5))Co{P(=O)(OMe)(2)}(3)](-)) were also prepared from [(Por)Ln(H(2)O)(3)](+) by the addition of the tripodal anion, L(OMe)(-), an effective encapsulating agent for lanthanide ions. Furthermore, the water-soluble lanthanide(III) porphyrinate complexes--including [(cis-DMPyDPP)Yb(H(2)O)(3)]Cl(3) (cis-DMPyDPP=5,10-bis(N-methylpyridinium-4'-y1)-15,20-di(phenyl)porphyrin), [(trans-DMPyDPP)Yb(H(2)O)(3)]Cl(3) (trans-DMPyDPP=5,15-bis(N-methylpyridinium-4'-y1)-10,20-di(phenyl)porphyrin), [(TMPyP)Yb(L(OMe))]I(4), and [(TMPyP)Er(L(OMe))]I(4) (TMPyP=tetrakis(N-methylpyridinium-4-y1)porphyrin)--were obtained by methylation of the corresponding complexes with methyl iodide and unambiguously characterized. The binding interactions and photocleavage activities of the water-soluble lanthanide(III) porphyrinate complexes towards DNA were investigated by UV-visible, fluorescence, and near-infrared luminescence spectroscopy, as well as circular dichroism and gel electrophoresis.

A mitochondria-permeable ruthenium(II) complex has been designed as a responsive probe which may be used to sensitize the formation of singlet oxygen (Phi(Delta) = 0.93) and cause local damage in cellulo when exposed to UV and near-infrared laser excitation.

Porphyrin derivatives werefound to accumulate readily in tumors, presumably because oftheir high vascular permeability, as well as their affinity for pro-liferating endothelium, and the lack of lymphatic drainage intumors.

Aristolochic acid (AA), derived from the herbal genus Aristolochia and Asarum, has recently been shown to be associated with the development of nephropathy. Upon enzyme activation, AA is metabolized to the aristolactam-nitrenium ion intermediate, which reacts with the exocyclic amino group of the DNA bases via an electrophilic attack at its C7 position, leading to the formation of the corresponding DNA adducts. The AA-DNA adducts are believed to be associated with the nephrotoxic and carcinogenic effects of AA. In this study, liquid chromatography coupled with electrospray ionization mass spectrometry (LC-MS) was used to identify and quantify the AA-DNA adducts isolated from the kidney and liver tissues of the AA-dosed rats. The deoxycytidine adduct of AA (dC-AA) and the deoxyadenosine-AA adduct (dA-AA) were detected and quantified in the tissues of rats with one single oral dose (5mg or 30mg AA/kg body weight). The deoxyguanosine adduct (dG-AA), however, was detected only in the kidney of rats that were dosed at 30mg AA/kg body weight for three consecutive days. The amount of AA-DNA adducts found in the rats correlated well with the dosage.

Sequence-specific photo-modification of DNA has been demonstrated, for the first time, in a vanadium(V)-peroxo complex, NH4[VO(O2)2(5,6-Me2phen)] (where 5,6-Me2phen = 5,6-dimethyl-1,10-phenanthroline). Using molecular cloning technique, a consensus sequence motif of 5'-G(A/G)TA(T/C)C was identified associated with the two specific photo-modification sites, 5'-ATC and 5'-TACC found on a plasmid DNA, pBluescript, by a modified Sanger sequencing technique. DNA supercoiling was shown to be a critical prerequisite for this observed sequence-specific photo-modification activity.

The DNA-photocleavage activities (at 365 nm) of 15 vanadium(V)−peroxo complexes were examined. Singlet oxygen was implicated as the species responsible for the DNA scission. The singlet oxygen was shown to be produced from the photolysis of the vanadium(V)−peroxo complex using a singlet oxygen EPR spin trap, 2,2,6,6-tetramethyl-4-piperidone. A mechanism for the photolysis of the [VO(O_2)_2(bpy)]- anion at neutral pH has been proposed.

The insulin-mimetic effects exhibited by vanadate, hydrogen peroxide, and some peroxovanadates have recently been shown to occur, at least in part, through an activation of the insulin receptor tyrosine kinase activity. In this study, we examine the effects of these compounds on insulin receptor binding using receptor preparations from human placental membranes. Among the 16 vanadium(V)-peroxo complexes studied, the [VO(O2)2(bipy)]- ion, where bipy = 2,2′-bipyridine, was found to increase insulin receptor binding by 24%, whereas the [VO(O2)2(en)]- ion, where en = ethylenediamine, was found to reduce insulin receptor binding by about the same amount under steady-state conditions. Scatchard analysis of the binding data indicates that the observed effect of the [VO(O2)2(bipy)]- ion on insulin receptor binding is exerted mainly at the high-capacity low-affinity sites. Furthermore, this modulatory effect is reversible and requires a continuous presence of the compound. By perturbing the membrane environment of the insulin receptor, we have shown that an intact membrane structure is essential for an observable effect. The observed modulation of insulin receptor binding by peroxovanadates is interpreted in terms of a ternary complex model in which the peroxovanadate acts as an allosteric effector modulating the binding equilibrium between insulin and its receptor.

Plasticized PVC membranes incorporating the hexaester of calix[6]arene, potassium tetrakis(4-chlorophenyl) borate and a neutral H+-selective chromo-ionophore (ETH 5294) have been used as reversible sensing devices for optical determination of potassium ion in pH-buffer solutions. At pH 9.0, the absorbance response of the optode shows a good correlation with the theoretically derived formula in the range from 1 x 10(-6) to 1 x 10(-2) mol l(-1) potassium ion. Typical response times (t95) in the samples are 3 min, the relative standard deviation for repeated measurements being within 0.5%. The selectivity of this potassium-selective membrane allows its application for the reversible optical detection of potassium concentration in human serum samples.

Several acetylene bridged porphyrin-monophthalocyaninato ytterbium(III) hybrids, PZn-PcYb, PH(2)-PcYb and PPd-PcYb, have been prepared and characterized by (1)H and (31)P NMR, mass spectrometry, and UV-vis spectroscopy. Their photophysical and photochemical properties, especially the relative singlet oxygen ((1)O(2)) quantum yields and the two-photon absorption cross-section (σ(2)), were investigated. These three newly synthesized compounds exhibited very large σ(2) values and substantial (1)O(2) quantum yields upon photo-excitation, making them potential candidates as one- and two-photon photodynamic therapeutic agents.
