The article is a publication of the brick discovered in the first half of the 20th century on the territory currently identified as a part of the Golden Horde known as Hajji Tarkhan, which is deposited in the funds of the Astrakhan Museum-Reserve. The fact that the finds entered the museum funds is connected with deliberate destruction of the settlement which had begun at the end of the 16th century. The latter is confirmed by historical sources. A well-preserved inscription in Arabic is a distinctive feature of the brick; the inscription is legible and possible to translate. Another characteristic of the inscription is that the letters were drawn without observing the norms of calligraphy on slightly dried clay or after the firing in the oven. The inscription includes three words, one of which is breaking on to the next line and forming a phrase that currently does not have exact analogies for translation. Nevertheless, a proposed translation option characterizes the inscription as one of the variations of the Quranic text formulas traditionally applied to various household items (vessels, candlesticks, pencil cases, etc.) since the 10th century. The nature of the unexpected finding in the lifting ground does not allow attributing it to any other construction on the Hillfort territory. Nevertheless, it was suggested that the brick was connected with the remains of a brick wall marked on the plan of the Shareniy Bugor hillfort drafted in 1924. The described and translated inscription is a new epigraphic source for the Golden Horde archeology. In addition, this is one of rare examples of the period when the text of the inscription was completely restored and translated.

The paper introduces into scientific discourse the details of early medieval ceramic containers discovered in the course of studies by the Astrakhan Museum-Reserve and the Institute of Oceanology of the Russian Academy of Sciences conducted at the Semibugorinsky archaeological complex in 2021, and deposited at the Astrakhan Museum-Reserve. The studied vessels belong to the so-called pithos-jugs of the Saltovo-Mayaki circle of the 8th–10th centuries. Such ceramic products are widely known from the Khazar monuments of the North Caucasus, in the lower reaches of the Don, the Don area and in the Crimea, found both in the form of whole items and numerous fragments. The paper examines in detail the morphology of the published vessels and their dimensional parameters, and outlines the circle of the closest analogies for each of the items. The issues of the chronology of container ware are discussed. Taking into account the evolution of pottery traditions, mainly related to the process of firing ceramics, which led to a change in its color from gray-clay to red-clay, the authors date the presented red-clay unpolished pithos-jugs the late 9th – early beginning 10th centuries. The assumptions about their manufacture by local artisans-potters are substantiated, as evidenced by numerous discovered fragments of red-clay thick-walled ware, defective ceramic products and ceramic slag. The published vessels expand the source base on the early medieval ceramics of the Volga delta.

The article is dedicated to the publication of the materials of burials from the Early Iron Age, discovered during excavations of a crumbling kurgan which is a part of the cemetery “Bogomolnye Peski-I” close to the village of Nikolskoye, Enotaevsky district, Astrakhan region. In total, 14 burials were investigated during the rescue excavations, two of them relate to the Middle Ages, other seven belong to the Bronze Age. The article describes in detail 5 burials of the Early Iron Age. The dating of the burials is defined based on the materials and details of the burial rite. The earliest burial (No. 2) dates back to the Savromat era. It contained a decapitated rams carcase and a molded pot which is typical of assumed era. The rest of the burials belong to the Middle Sarmatian period (1st – 2nd centuries AD). One of the Middle Sarmatian burials was completely destroyed by robbers in ancient times. It was possible to find a lot of gold stripes of clothes among its containment, as well as a bronze cauldron with a tamga. Plaques and stripes are not typical of either the Savromat or the early Sarmatian cultures. Indeed, they are widely used in prestigious burials of the Middle Sarmatian culture. The burials No. 5 and No. 13, accompanied by a rich inventory, are of the greatest interest. A set of gold decorations for a funeral veil and a belt set of gold with turquoise inserts were found in burial No.5, which belong to the products of the Sarmatian polychrome style, typical of the Middle Sarmatian period. In addition, a gilded bronze dish related to Roman provincial dishes and an alabaster vessel were discovered there – a typical find of the Middle Sarmatian era. In burial No. 12, the most interesting findings, in addition to a large number of various beads, are intaglio gemstones made of red glass with plots of ancient mythology depicted on them, which are unique in their own way. Moreover, an interesting vessel shaped as a bird (duck) was found there as well. The close analogs to the latter are known in cemeteries of the 1st – 2nd centuries AD in the Kuban region. A feature of this kurgan is a large number of inlet burials of the Middle Sarmatian culture, whereas in general, the burials under individual mounds predominate in the Middle Sarmatian epoch.

Abstract A comprehensive study, including geological mapping, structural and thermochronological analysis, has been carried out on Ivanščica Mountain (NW Croatia), with the aim to reconstruct the tectonic history of the Dinarides, Southern/Eastern Alps and Pannonian Basin transitional zone. Implementation of structural and thermochronological methods enabled a subdivision of Ivanščica Mt. into two structural domains (from bottom to top): Ivanščica Parautochthon and Ivanščica Imbricate Fan and Cenozoic sedimentary cover. In addition, a sequence of deformational events in tectonic history of this transitional zone is proposed, comprising three extensional and four contractional events starting from Middle Triassic until present times. The two oldest deformational events indicate Middle Triassic (D1) and Early Jurassic (D2) extensional pulses and only occur in volcano-sedimentary successions of the Ivanščica Mt. The oldest contractional event (D3) is related to the obduction of a Neotethyan ophiolitic mélange over an Upper Triassic to Lower Cretaceous succession of the eastern margin of the Adriatic microplate, which resulted in thermal overprint of the Ivanščica Imbricate Fan structural domain in Berriasian—Valanginian times (~ 140 Ma). This event was soon followed by a second contractional event (D4), which resulted in thrusting and imbrication of the Adriatic passive margin successions together with previously emplaced ophiolitic mélange, thermal overprint of the footwall successions, fast exhumation and erosion. Apatite fission track data together with syn-tectonic deposits indicate an Hauterivian to Albian age of this D4 event (~ 133–100 Ma). These Mesozoic structures were dextrally rotated in post-Oligocene times and brought from the initially typically Dinaridic SE striking and SW verging structures to the recent SW striking and NW verging structures. The following extensional event (D5) is associated with the formation of SE striking and mostly NE dipping normal listric faults, and ENE striking dextral faults accommodating top-NE extension in the Pannonian Basin. Deformations were coupled with hanging wall sedimentation of Ottnangian to middle Badenian (middle Burdigalian to upper Langhian; ~ 18–14 Ma) syn-rift deposit as observed from the reflection seismic and well data. A short-lasting contraction (D6) was registered in the late Sarmatian (late Serravallian; ~ 12 Ma). The youngest documented deformational event (D7) resulted in reactivation of ENE striking dextral faults, formation of SE striking dextral faults as well as the formation of E to ENE trending folds and reverse faults. This event corresponds to late Pannonian (late Messinian; ~ 6 Ma) to Present NNW-SSE contraction driven by the indentation and counterclockwise rotation of Adriatic microplate. Recognized tectonic events and their timings indicate that Ivanščica was mainly affected by deformational phases related to the Mesozoic evolution of the Neotethys Ocean as well as Cenozoic opening and inversion of the Pannonian Basin. Therefore, the Mesozoic tectono-sedimentary evolution of Ivanščica Mountain proves the paleogeographic affiliation of its non-ophiolitic Mesozoic structural-stratigraphic entities to the Pre-Karst unit of the Dinarides.

Testate amoebae are a polyphyletic group of unicellular eukaryotic organisms that are characterised by a rigid shell and inhabit mostly freshwater and terrestrial ecosystems. They are particularly abundant in peatlands, especially in Sphagnum-dominated biotopes. Peatland hydrology is the most important influence on testate amoebae communities. The good preservation of the shells in peat deposits and their response to hydrological regime changes are the principles for palaeohydrological reconstructions. Any changes in the water balance of mires should be expected to have far-reaching effects on biogeochemical cycles, productivity, carbon dioxide and methane exchange.This paper presents a dataset (Darwin Core Archive – DwC-A) on the distribution of Sphagnum-dwelling testate amoebae in nine mires located in the forest-steppe subzone of the East European Plane. The dataset includes information about 86 taxa belonging to 29 genera and contains 3,123 occurrences of 49,874 individuals. The following environmental variables are provided: microtopography, oxidising and reducing potential, total mineralisation, substrate temperature, acidity, substrate wetness and water table depth. These data might be used for biogeographical and palaeoecological studies, including quantitative reconstructions.

Plastic pollution poses a significant threat to ecosystem health worldwide. This study examines the determinants of environmental changes in human-modified ecosystems through a quantitative-qualitative system dynamics modeling approach: field experiments conducted on a 310 m2 unsaturated clay-rich bed and a 2.5 m2 clay-rich shore of a plastic-impacted pond in Shenzhen, China, and a 1.17 ha plastic-impacted clay pit in Musanze, Rwanda; laboratory experiments involving Modified Proctor (MP) and California Bearing Ratio (CBR) tests on natural clay reinforced with polyethylene terephthalate (PET) microplastics, with diameters ranging from 0.25 to 5 mm and at concentrations of 1.25 %, 2.5 %, 3.75 %, 5 %, and 10 % by weight of clay; and plastic dynamic flows analyzed by modeling the life cycle of PET. Field experiments showed that mulch type and thickness were critical factors influencing crack distribution in a plastic-impacted pond bed. Specifically, cracks were dominant in areas with pronounced desiccation and lacking filamentous green algae and PET-dominated plastic waste. Along the 2.5 m moisture gradient in a plastic-impacted pond bed, temperature and moisture significantly influenced nutrients, particularly in pronounced desiccation zones. Laboratory experiments showed that microplastics altered the structural properties of natural clay, decreasing moisture content while increasing dry density and load-bearing capacity. The plastic life cycle underscored the roles of industrial and consumer practices, environmental conditions, and waste management and recycling inefficiencies in driving environmental changes in human-modified ecosystems. The findings underscore the need for effective plastic waste management and recycling to mitigate the ecological impacts of plastic pollution in ecosystems.

Abstract During the Middle Triassic, intensive volcanic activity took place along the eastern margin of Pangea, including the Greater Adria promontory, due to the Neotethyan oceanization. This resulted in the formation of various volcanic and volcaniclastic rock types. The region of NW Croatia, acting as a transition zone between the Southern Alps and the Dinarides, showcases the outcrops of these rocks. The present study investigates the facies of volcaniclastic rocks, the distribution of those facies, formation processes, as well as the genesis of the primary magma to gain a better understanding of the complex geodynamics of this region during the Middle Triassic. Six profiles across the Vudelja quarry front were surveyed using drone imaging and samples were collected for detailed petrographic and geochemical analyses. Two groups of volcaniclastic rocks were identified—mafic and intermediate/felsic. The former is represented by (I) autoclastic effusive facies and (II) resedimented autoclastic facies, while the latter is represented by (III) secondary pyroclastic facies. Mafic volcaniclastics were generated through basaltic effusions in marine environments, fragmentation in contact with seawater, mixing with shallow marine carbonate clasts, and subsequent redeposition in deeper marine areas. The secondary pyroclastic facies (III) consists of a regionally distributed felsic Pietra Verde tuff whose deposits may be related to pyroclastic density currents and syn-eruptive resedimentation by turbidite-like currents. Geochemical data indicate that parental magmas responsible for generating the mafic volcaniclastics had a calc-alkaline composition and originated in ensialic and mature arc settings of an active continental margin. The observed chemical composition is likely inherited from older, arc-related lithologies, associated with the subduction of the Paleotethys Ocean. Parental magmas are thought to have formed during continental rifting of the passive Middle Triassic margins of the Greater Adria through (i) partial melting of the heterogeneous lithospheric mantle, which had been metasomatized during an earlier Hercynian subduction, and (ii) subordinate processes related to the melting of the upper continental crust and subsequent fractionation. Ar/Ar dating on plagioclase separates yielded an age of 244.5 ± 2.8 Ma for mafic volcaniclastics. This aligns well with biostratigraphic ages of felsic tuffs which crop out on a broader regional scale of the Dinarides, the Southern Alps, and the Transdanubian Range. The overlapping ages obtained from radiometric dating of mafic volcaniclastics and biostratigraphic ages of the felsic Pietra Verde tuffs strongly suggest that the Greater Adria region experienced concurrent bimodal volcanism during the Middle Triassic.

Remotely sensed data play a crucial role in monitoring the El Niño/La Niña Southern Oscillation (ENSO), which is an oceanic-atmospheric phenomenon occurring quasi-periodically with several impacts worldwide, such as specific biological and global climate responses. Since 1980, Earth has witnessed three strong ENSO events (1982–1983, 1997–1998, 2015–2016). In September 2022, La Niña entered its third year and was unlikely to continue through 2024. Instead, since 2022, forecasts have pointed to a transition from La Niña to a Neutral phase in the summer or late 2023. The onset of El Niño occurred around April 2023, and it is anticipated by sophisticated models to be a strong event through the Northern Hemisphere winter (December 2023–February 2024). The aim of this study is to demonstrate the ability of the combination of two new methods to improve the accuracy of the above claim because El Niño apart from climate anomalies, significantly impacts Earth’s ecosystems and human societies, regulating the spread of diseases by insects (e.g., malaria and dengue fever), and influencing nutrients, phytoplankton biomass, and primary productivity. This is done by exploring first the previous major El Niño events in the period January 1876–July 2023. Our calculations show that the ongoing 2023–2024 El Niño will not be the strongest.

The presence of microplastics (MPs) is increasing at a dramatic rate globally, posing risks for exposure and subsequent potential adverse effects on human health. Apart from being physical objects, MP particles contain thousands of plastic-associated chemicals (i.e., monomers, chemical additives, and non-intentionally added substances) captured within the polymer matrix. These chemicals are often migrating from MPs and can be found in various environmental matrices and human food chains; increasing the risks for exposure and health effects. In addition to the physical and chemical attributes of MPs, plastic surfaces effectively bind exogenous chemicals, including environmental pollutants (e.g., heavy metals, persistent organic pollutants). Therefore, MPs can act as vectors of environmental pollution across air, drinking water, and food, further amplifying health risks posed by MP exposure. Critically, fragmentation of plastics in the environment increases the risk for interactions with cells, increases the presence of available surfaces to leach plastic-associated chemicals, and adsorb and transfer environmental pollutants. Hence, this review proposes the so-called triple exposure nexus approach to comprehensively map existing knowledge on interconnected health effects of MP particles, plastic-associated chemicals, and environmental pollutants. Based on the available data, there is a large knowledge gap in regard to the interactions and cumulative health effects of the triple exposure nexus. Each component of the triple nexus is known to induce genotoxicity, inflammation, and endocrine disruption, but knowledge about long-term and inter-individual health effects is lacking. Furthermore, MPs are not readily excreted from organisms after ingestion and they have been found accumulated in human blood, cardiac tissue, placenta, etc. Even though the number of studies on MPs-associated health impacts is increasing rapidly, this review underscores that there is a pressing necessity to achieve an integrated assessment of MPs’ effects on human health in order to address existing and future knowledge gaps.

Time-resolved fluorescence spectroscopy plays a crucial role when studying dynamic properties of complex photochemical systems. Nevertheless, the analysis of measured time decays and the extraction of exponential lifetimes often requires either the experimental assessment or the modeling of the instrument response function (IRF). However, the intrinsic nature of the IRF in the measurement process, which may vary across measurements due to chemical and instrumental factors, jeopardizes the results obtained by reconvolution approaches. In this paper, we introduce a novel methodology, called blind instrument response function identification (BIRFI), which enables the direct estimation of the IRF from the collected data. It capitalizes on the properties of single exponential signals to transform a deconvolution problem into a well-posed system identification problem. To delve into the specifics, we provide a step-by-step description of the BIRFI method and a protocol for its application to fluorescence decays. The performance of BIRFI is evaluated using simulated and time-correlated single-photon counting data. Our results demonstrate that the BIRFI methodology allows an accurate recovery of the IRF, yielding comparable or even superior results compared with those obtained with experimental IRFs when they are used for reconvolution by parametric model fitting.

In order to demonstrate the viability of wave energy converters usage in the Mediterranean islands, this paper explores the possibilities of optimized technical integration of wave energy converters into an island's electrical grid, with a particular focus on the island of Vis in the Adriatic Sea. After introduction, a comprehensive definition of wave energy was provided and classifications and technologies associated with wave energy converters were discussed. Additionally, different interface types between wave energy converters and the grid were discussed, as well as the potential impact of these converters on the island's grid and its electrical power system. Power matrices of two different wave energy converters (WECs), namely Wavestar and Wavebob, were adjusted to the specific sea conditions around Vis using the Froude scaling method. Also, Power Factory software tool was used to model the island's power grid, conduct simulations, and present electrical parameter values derived from real-world data collected on Vis. Various scenarios involving the integration of photovoltaic power plants and wave energy converters with the resulting data are both visually and numerically presented. Simulation results indicate a successful possibility of integrating wave energy converters to the microgrid of the island of Vis with the aim to archive near net zero electricity exchange with the mainland grid with improved power (voltage) quality in the island microgrid.

The development of a quantitative in-line UV spectroscopic method for monitoring of solute concentration during the crystallization process of the active pharmaceutical ingredient (API), ceritinib is described. The method is based on artificial neural networks (ANN). A seeded cooling crystallization process of ceritinib from tetrahydrofuran was studied as a model system. The model was constructed from collected ATR-UV spectra and temperature records within the metastable zone. The collected spectra were preprocessed with the first derivative using the Savitzky-Golay filter. ANN models with different architectures were created and the optimal architecture was chosen based on the root mean square error of prediction (RMSEP) criterion. In addition, ANN models were compared with the models obtained by the linear partial least squares regression (PLSR). Due to the nonlinear relationship in the data set, ANN models predict the solution concentration with higher accuracy compared to linear models. The developed models were successfully used in real-time solution concentration measurement during ceritinib crystallization along with a supersaturation control module developed in-house.

Abstract Background In terms of the potential influence of rapid weight loss (RWL) on the metabolic health of former combat sports athletes (CSA), the scientific literature is quite scarce. Therefore, the objective of the presented research was to determine the differences in metabolic syndrome (MetS) parameters and the prevalence of MetS between former athletes who performed RWL and athletes who did not. Methods The sample of the presented study comprised 150 participants from Serbia, equally divided into two groups: 75 former athletes who had practiced combat sports and 75 ex-athletes of various other sports who did not practice RWL during their careers. The following parameters related to the MetS were evaluated: waist circumference, high-density lipoprotein cholesterol, systolic blood pressure, diastolic blood pressure, fasting glucose, and triglycerides. ABSTRACT: Introduction. The industrial activities result in the release of a significant number of pollutants into the atmosphere, which have a negative impact on human health and the environment. The development of sustainable principles for construction sites development requires the elaboration of algorithms, assessment methodologies, and monitoring of the environmental situation at the construction site. Analysis of research works in the field of environmental monitoring of air pollution by hazardous substances has shown that the speed and reliability of the obtained information are crucial in polluted atmospheric conditions. In this article, we examine the application of an intelligent environmental monitoring system with Salavat city Republic of Bashkortostan as the example. The article presents an example of continuous eco-monitoring using three automatic air pollution monitoring stations for controlling the atmospheric air in residential areas. Methods and Materials. An intelligent eco-monitoring system is proposed, which is capable of assessing the state of the air basin of an industrial city, identifying sources of increased air pollution, processing information using an artificial neural network in real-time, generating control actions on sources of increased air pollution, and providing recommendations to businesses for optimizing their operations to prevent exceeding the maximum allowable concentrations of harmful substances. Results. The analysis of air quality in Salavat has led to the conclusion that it is necessary to install window filter-screens with nanofiber-based filtering elements in residential buildings. Discussion. The results of the air basin monitoring can be used in an integrated intelligent environmental monitoring system for assessing environmental conditions and can be made available to interested parties. Considering the results in Salavat, the project can be replicated for use in eco-monitoring systems of towns and cities in the Russian Federation. Conclusion. As a result of the project, the program utilizing artificial intelligence has been developed, which will contribute to reducing the level of air pollution in the future.

Construction of relay protections of electrical installations without the use of bulky metal- and insulation-intensive measuring current transformers (CTs) was repeatedly discussed at Conferences of the International Council on Large Electric Systems (CIGRE) as a challenge for the electric power industry. In the article, the authors present resource-saving reed switch current protection for 6–10-kV electrical installations connected to switchgear cells, which is an alternative to traditional protections. The scientific novelty of the work lies in (a) reed switch protection design on the basis of measuring the magnetic induction in different modes and at different points inside a cell, and we prove that inductance values are sufficient to detect phase short-circuits in electrical installations fed from this cell and derive the dependence of the induction on the position of an inductance coil inside the cell; (b) proven possibility of using the simplest formula of the Biot–Savart law for the calculations if the experimentally obtained coefficients are introduced into it; (c) development of a technique for calculating the parameters of the overcurrent protection based on reed switches. Experimental results are presented in graphical form and clearly show the points of EMS maxima and minima. Settings for resource-saving current protection are selected and a feasibility study of the effectiveness of resource-saving overcurrent protection is carried out. A technical and economical assessment of the efficiency of the suggested overcurrent protection is conducted.

ABSTRACT: Introduction. Approximately 20% of accidents in buildings and structures are associated with the destruction of metal structures. Nanocoatings applied to metal structures significantly improve their operational properties, but at the same time make it difficult to use traditional non-destructive testing methods that require access to the surface of the base metal. The quality of the nanocoating is controlled during its formation. To prevent the destruction of metal structures, it is necessary to identify structural heterogeneities of the base metal in micro- and sub-micron sizes, which arise in areas of increased mechanical stress concentration and transform into macrodefects leading to structural failure. Methods and tools are required to non-destructively identify the stress-strain state and damage of the base metal structure through the layer of nanocoating. Electromagnetic-acoustical (EMA) transformation, is considered promising for solving this problem. Existing EMA tools do not fully utilize the potential of EMA conversion, have insufficient sensitivity and the lack of informativity. Methods and materials. For experimental research, we selected widely used in construction structures steels. Research was conducted on the interrelationship of structural alterations in standard samples of metals and the parameters of the electro-magnetic (EMA) transducer under static and cyclic loads. Results. Load diagrams of metal samples were obtained, with accompanying photographs of microstructure in control points, along with their frequency models derived through spectral analysis of the signal from EMA transducer. Discussion. Changes in the stress-strain state of the metal and accumulation of damage in its structure result in a complex interrelated set of changes in mechanical, acoustic, and electro-physical properties, all of which are reflected in the changes in the parameters of the frequency model. Conclusion. Based on the results of the conducted research, the use of a frequency model has been proposed as an integral parameter for identifying the stress-strain state and damage of metal in the equipment. The use of an artificial neural network for analyzing frequency model parameters simplifies the process of identifying the stress-strain state and damage of metal structures and increases its reliability.

Hydra is known for its natural occurrence, anatomical simplicity, intricate physiology, regenerative capacity, and ease of maintenance and manipulation in laboratory environments. It has proven to be a valuable model organism in various disciplines. Its applications range from developmental biology, stem cell research, animal physiology to environmental toxicology including ecotoxicology. The sensitivity of Hydra to a variety of environmental stressors and chemical agents such as metals, nanomaterials, and toxic organic compounds provides valuable insights into physiological mechanisms affected by environmental stressors and pollution, and Hydra can be of great use in environmental monitoring. Furthermore, since green Hydra lives in a symbiotic relationship with unicellular photoautotrophic algae, it is a suitable model organism for symbiosis research. Recently, it has become a popular model in holobiont research. The adaptability and importance of Hydra also extends to aquatic science and aquatic ecology, particularly in the context of monitoring and water pollution. Since the 1980s, Hydra has been increasingly used in various fields of research and has established itself as an important versatile model organism in numerous scientific studies. Hydra also represents an outstanding model in the fields of education and STEM. Hydra continues to be an important model in the 21st century, contributing significantly to our understanding of the biology of water and advancing freshwater research, and possibly finding its way to regenerative medicine and tumor pathobiology research.

The ocean energy resources hold the promise of a sustainable solution within global efforts to diversify energy sources and mitigate climate change. Wave energy conversion (WEC) systems, as emerging technologies, offer adaptability and the potential to harness predictable wave energy. However, integration of WEC systems into a power grid brings challenges for system operators due to their nature of operation. Addressing these demands is a multilayered process that involves highly efficient power electronic devices, control systems, and efficient energy storage solutions. This paper specifically focuses on the methodologies of the grid integration of a specific wave energy conversion system—a point absorber developed by the company Sigma Energy. Proposed methodologies are experimentally tested using power hardware-in-loop (PHIL) emulation of a fully monitored and controlled small-scale microgrid equipped with a battery energy storage system (BESS), different emulators of loads, and distributed generators (DG).

Biska, a traditional Istrian herbal spirit, is produced by macerating mistletoe leaves and twigs in a water–ethanol base, usually grape marc spirit. The aim of this study was to determine the optimal mistletoe maceration parameters (ethanol content, concentration of plant material, and duration) for the production of biska, which have not yet been the subject of scientific research. The prepared mistletoe macerates and the collected biska samples were subjected to phenolic compound characterization, including total phenolic content (TPC), total flavonoid content (TFC), and polyphenolic profile, by ultra-high-performance liquid chromatography coupled with mass spectrometry (UPLC-ESI-MS2). In addition, the macerates and the biska samples were analyzed for their biological properties: antioxidant and antiproliferative activity. The results showed that both the macerates and the biska samples contain phenolic compounds of mistletoe such as phenolic acids, flavonoids, and coumarins, which contribute to the observed biological activities—significant antioxidant activity and antiproliferative effect. By applying the maceration parameters, which yielded the highest content of phenolic compounds as well as volatile aromatic compounds (40–80 g/L of mistletoe, 40–55% v/v ethanol, and 21 days’ maceration time), the production process can be improved to produce biska with better aromatic and biological properties.

To develop specific preventive strategies, it is necessary to investigate in detail the occurrence of injuries in young football players. This study aimed to provide details about the type, location, severity, and contact nature of muscle injuries in elite U15 Croatian football players. A total number of 130 muscle injuries occurred during the six competitive seasons, of which 88 were DOMS injuries and 42 were rupture injuries. Specifically, 87% of all muscle injuries belonged to the four major groups, with only 15 injuries to the other muscle groups: hamstrings (43), quadriceps (41), adductors (25), calves (6). The muscle injury incidence was stable during the observed period as the injury incidence rate ratio (IRR) was 1.01 (95%CI: 0.99–1.03), with 3.13 (95%CI: 3.09–3.17) muscle injuries per 1000 h of exposure in the first season and 3.06 (95%CI: 3.02–3.1) in the sixth season. A significantly higher risk for muscle injuries was discovered in the competition as the incidence for the whole observed period was 7.38 (95%CI: 7.29–7.47) compared to 2.25 (95%CI: 2.24–2.26) in the training period. Despite the relatively stable number of sustained muscle injuries, the relative proportion of muscular injuries increased over the years due to a severe decrease in the total number of injuries. Regarding the proportion of players who had sustained a muscle injury, our results suggest that approximately two out of five players can expect a muscular injury over the one-season period. Relatively high injury expectancy rates, as well as reinjury rates, require specific preventive interventions that will reduce the number and severity of new and recurred injuries. Future studies should include a wide set of predictor variables in order to establish the most important injury risk factors.

Mucopolysaccharidosis II (MPS II) is a rare lysosomal storage disease characterized by deficient activity of iduronate-2-sulfatase (I2S), leading to pathological accumulation of glycosaminoglycans (GAGs) in tissues. We used iduronate-2-sulfatase knockout (Ids KO) mice to investigate if liver-directed recombinant adeno-associated virus vectors (rAAV8-LSP-hIDSco) encoding human I2S (hI2S) could cross-correct I2S deficiency in Ids KO mouse tissues, and we then assessed the translation of mouse data to non-human primates (NHPs). Treated mice showed sustained hepatic hI2S production, accompanied by normalized GAG levels in somatic tissues (including critical tissues such as heart and lung), indicating systemic cross-correction from liver-secreted hI2S. Brain GAG levels in Ids KO mice were lowered but not normalized; higher doses were required to see improvements in brain histology and neurobehavioral testing. rAAV8-LSP-hIDSco administration in NHPs resulted in sustained hepatic hI2S production and therapeutic hI2S levels in cross-corrected somatic tissues but no hI2S exposure in the central nervous system, perhaps owing to lower levels of liver transduction in NHPs than in mice. Overall, we demonstrate the ability of rAAV8-LSP-hIDSco to cross-correct I2S deficiency in mouse somatic tissues and highlight the importance of showing translatability of gene therapy data from rodents to NHPs, which is critical for supporting translation to clinical development.

Abstract The purpose of this research was to investigate the running performance of professional soccer players in relation to seven phases which resulted in the changing or maintaining the match status in the UEFA Champion League games during season 2020/2021. Moreover, we aimed to define which match status phases occur at the earliest stage of regular game time. This study involved professional soccer players from 24 teams participating in the group stage of UEFA Champions League in season 2020/21. The match status was divided into seven phases that result in changing or maintaining the match outcome: DW (Drawing to Winning); LD (Losing to Drawing); WW (Winning to Winning); DD (Drawing to Drawing); LL (Losing to Losing); DL (Drawing to Losing); WD (Winning to Drawing). Such running performance variables as: total distance covered (TDC) and distance covered in high-intensity running (HIR) were analyzed. Players participating in the UEFA Champions League matches cover the longest TDC in DW, DL and DD phases. TDC in these stages was between 111 and 123 m min−1. The highest HIR was recorded during phases: DW, DL and LL (range between 9.91 and 10.82 m min−1). In contrast, the lowest total distance and distance in HIR is covered during WD phase (only 105.57 ± 1.89 m min−1 and 7.34 m min−1 respectively). On average, phases resulting in the change of the match status occur during the first half, while all phases maintaining the result in the second half. Coaching staffs should consider registering and analysing the physical match performance in relation to described seven match status phases. Such information allows to prepare team-specific training drills, that players should perform more often in order to change or maintain the status of the game.

Abstract Endoribonucleases govern the maturation and degradation of RNA and are indispensable in the posttranscriptional regulation of gene expression. A key endoribonuclease in Gram‐negative bacteria is RNase E. To ensure an appropriate supply of RNase E, some bacteria, such as Escherichia coli, feedback‐regulate RNase E expression via the rne 5′‐untranslated region (5′ UTR) in cis. However, the mechanisms involved in the control of RNase E in other bacteria largely remain unknown. Cyanobacteria rely on solar light as an energy source for photosynthesis, despite the inherent ultraviolet (UV) irradiation. In this study, we first investigated globally the changes in gene expression in the cyanobacterium Synechocystis sp. PCC 6803 after a brief exposure to UV. Among the 407 responding genes 2 h after UV exposure was a prominent upregulation of rne mRNA level. Moreover, the enzymatic activity of RNase E rapidly increased as well, although the protein stability decreased. This unique response was underpinned by the increased accumulation of full‐length rne mRNA caused by the stabilization of its 5′ UTR and suppression of premature transcriptional termination, but not by an increased transcription rate. Mapping of RNA 3′ ends and in vitro cleavage assays revealed that RNase E cleaves within a stretch of six consecutive uridine residues within the rne 5′ UTR, indicating autoregulation. These observations suggest that RNase E in cyanobacteria contributes to reshaping the transcriptome during the UV stress response and that its required activity level is secured at the RNA level despite the enhanced turnover of the protein.

In order to identify match running performance (MRP) characterizing the most elite soccer matchplay, this study aimed to examine position-specific differences in the MRP of players competing in “big five” (BFLTs) and “non-big five” league teams (N-BFLTs). The data were obtained from 24 teams (BFLTs; n = 14, N-BFLTs; n = 10) during the UEFA Champions League (UCL) matches (n = 20) in the 2020/21 season using a semiautomatic video system. The differences in MRP between BFLTs and N-BFLTs, while controlling for contextual factors, were examined using linear mixed model. No differences in overall MRP between fullbacks, central midfielders, wide midfielders and forwards from BFLTs and their peers from N-BFLTs were found, while only central defenders from BFLTs covered more high-intensity running than central defenders from BFLTs (moderate effects size). For players on all playing positions from BFLTs, total- and low-intensity distance covered were lower in offensive phase of game and greater in defensive phase of game compared to their peers from N-BFLTs (all large effect sizes). This study demonstrated that the most elite match-play in soccer is characterized by increased efforts in defensive phase of game, and decreased efforts in offensive phase of game. Soccer training programmes should be adapted accordingly.

Physical literacy (PL) is theorized to be an important determinant of physical fitness (PF) and physical activity levels (PALs), but studies have rarely examined possible correlations between PL, PF, and PAL in preadolescent children. This study aimed to evaluate age-specific and gender-specific correlations between the affective and cognitive domains of PL (PLAC), PF, and PAL in preadolescents. Additionally, the test–retest reliability of the applied PF tests was examined. The participants were 107 children (9 to 11 years of age; 53 girls). Apart from gender and school age (third graders, fourth graders), the variables included PLAC, PAL, and PF. PF was evaluated by anthropometrics and body composition indices, sit-ups, push-ups, PACER tests, torso lifts, and broad jump. PLAC was evaluated using the PLAYself questionnaire, and the PAQ-C was used to evaluate PAL. The test–retest reliability of the PF tests was appropriate to high (ICC: 0.65–0.91). Analyses showed nonsignificant associations between PLAC and PAL in both genders and age groups. PAL was significantly correlated with PL in girls (low correlation) and fourth graders (moderate correlation). The nonsignificant associations between PLAC and PF could be a consequence of the influence of biological maturity on PF in this age group. The correlation between PAL and PLAC can be explained by the differences in physical activity habits between genders and age groups, with participation in structured physical activities (organized sports and physical education) being the most important source of PAL in girls and older children.

In positron emission tomography (PET), the original points of emission are unknown, and the scanners record pairs of photons emitting from those origins and creating lines of response (LORs) in random directions. This presents a latent variable problem, since at least one dimension of relevant information is lost. This can be solved by a statistical approach to image reconstruction—modeling the image as a Gaussian mixture model (GMM). This allows us to obtain a high-quality continuous model that is not computationally demanding and does not require postprocessing. In this paper, we propose a novel method of GMM estimation in the PET setting, directly from lines of response. This approach utilizes some well-known and convenient properties of the Gaussian distribution and the fact that the random slopes of the lines are independent from the points of origin. The expectation–maximization (EM) algorithm that is most commonly used to estimate GMMs in the traditional setting here is adapted to lower-dimensional data. The proposed estimation method is unbiased, and simulations and experiments show that accurate reconstruction on synthetic data is possible from relatively small samples.

In this study, we aimed to document the freshwater symbiotic interactions along with the impact of the abiotic environment and anthropogenic effects on the functionality of freshwater organisms. Symbiotic green hydra (Z) and free-living brown hydra (S), either separately or both species together, were treated with the herbicide norflurazon in concentrations of 2 × 10−6 mol/L (N6) and 2 × 10−7 mol/L (N7) for 72 h. Also, hydras were treated with both norflurazon and UV radiation at a wavelength of 254 nm for 2 min or were irradiated only. The next part of the experiment was performed in the same way but with added suspensions of isolated endosymbiotic alga, free-living alga, or both algae together. Mortality, migration, tentacle and tissue damage, changes in the thickness of the mesoglea of hydras, and clustering of algae were monitored. Green hydra generally showed lower rates of migration, and mortality was observed only in green hydra exposed to UV radiation. Tentacle damage was more pronounced in green hydra and included a specific fork-like structure. The use of cryofixation and TEM enabled us to partly elucidate the effect of clustering of algae. In summary, our study provides new insights into the influence of different environmental stressors and their combination on symbiotic and free-living freshwater hydras and algae and a better understanding of interactions in freshwater ecosystems.
