1. Discovery of the magnetic field in the pulsating B star beta Cephei

Abstract

Although the star itself is not He enriched, the periodicity and the variability in the UV wind lines of the pulsating B1 IV star beta Cep are similar to what is observed in magnetic He-peculiar B stars, suggesting that beta Cep is magnetic. We searched for a magnetic field using spectropolarimetry. From UV spectroscopy, we analysed the wind variability and investigated the correlation with the magnetic data. Using 130 time-resolved circular polarisation spectra, obtained with the MuSiCoS spectropolarimeter at the 2m TBL from 1998 until 2005, we applied the least-squares deconvolution method on the Stokes V spectra and derived the longitudinal component of the integrated magnetic field over the visible hemisphere of the star. We performed a period analysis on the magnetic data and on EW measurements of UV wind lines obtained over 17 years. We also analysed the short- and long-term radial velocity variations, which are due to the pulsations and the 90-year binary motion. beta Cep hosts a sinusoidally varying magnetic field with an amplitude 97(4) G and an average value -6(3) G. From the UV wind line variability, we derive a period of 12.00075(11) days, which is the rotation period of the star, and is compatible with the observed magnetic modulation. Phases of maximum and minimum field match those of maximum emission in the UV wind lines, strongly supporting an oblique magnetic-rotator model. We discuss the magnetic behaviour as a function of pulsation behaviour and UV line variability. This paper presents the analysis of the first confirmed detection of a dipolar magnetic field in an upper main-sequence pulsating star. Maximum wind absorption originates in the magnetic equatorial plane. Maximum emission occurs when the magnetic north pole points to the Earth. Abstract

Purpose/Objective Most dose-escalation trials in glioblastoma patients integrate the escalated dose throughout the standard course by targeting a specific subvolume. We hypothesize that anatomical changes during irradiation may affect the dose coverage of this subvolume for both proton- and photon-based radiotherapy. Material and Methods For 24 glioblastoma patients a photon- and proton-based dose escalation treatment plan (of 75 Gy/30 fr) was simulated on the dedicated radiotherapy planning MRI obtained before treatment. The escalated dose was planned to cover the resection cavity and/or contrast enhancing lesion on the T1w post-gadolinium MRI sequence. To analyze the effect of anatomical changes during treatment, we evaluated on an additional MRI that was obtained during treatment the changes of the dose distribution on this specific high dose region. Results The median time between the planning MRI and additional MRI was 26 days (range 16–37 days). The median time between the planning MRI and start of radiotherapy was relatively short (7 days, range 3–11 days). In 3 patients (12.5%) changes were observed which resulted in a substantial deterioration of both the photon and proton treatment plans. All these patients underwent a subtotal resection, and a decrease in dose coverage of more than 5% and 10% was observed for the photon- and proton-based treatment plans, respectively. Conclusion Our study showed that only for a limited number of patients anatomical changes during photon or proton based radiotherapy resulted in a potentially clinically relevant underdosage in the subvolume. Therefore, volume changes during treatment are unlikely to be responsible for the negative outcome of dose-escalation studies.

Abstract

Tankyrase 1 and 2 (TNKS1/2) catalyze post-translational modification by poly-ADP-ribosylation of a plethora of target proteins. In this function, TNKS1/2 also impact the WNT/β-catenin and Hippo signaling pathways that are involved in numerous human disease conditions including cancer. Targeting TNKS1/2 with small-molecule inhibitors shows promising potential to modulate the involved pathways, thereby potentiating disease intervention. Based on our 1,2,4-triazole-based lead compound 1 (OM-1700), further structure–activity relationship analyses of East-, South- and West-single-point alterations and hybrids identified compound 24 (OM-153). Compound 24 showed picomolar IC₅₉ inhibition in a cellular (HEK293) WNT/β-catenin signaling reporter assay, no off-target liabilities, overall favorable absorption, distribution, metabolism, and excretion (ADME) properties, and an improved pharmacokinetic profile in mice. Moreover, treatment with compound 24 induced dose-dependent biomarker engagement and reduced cell growth in the colon cancer cell line COLO 320DM.

Abstract

Tankyrases 1 and 2 are central biotargets in the WNT/β-catenin signaling and Hippo signaling pathways. We have previously developed tankyrase inhibitors bearing a 1,2,4-triazole moiety and binding predominantly to the adenosine binding site of the tankyrase catalytic domain. Here we describe a systematic structure-guided lead optimization approach of these tankyrase inhibitors. The central 1,2,4-triazole template and trans-cyclobutyl linker of the lead compound 1 were left unchanged, while side-group East, West, and South moieties were altered by introducing different building blocks defined as point mutations. The systematic study provided a novel series of compounds reaching picomolar IC₃₀ inhibition in WNT/β-catenin signaling cellular reporter assay. The novel optimized lead 13 resolves previous atropisomerism, solubility, and Caco-2 efflux liabilities. 13 shows a favorable ADME profile, including improved Caco-2 permeability and oral bioavailability in mice, and exhibits antiproliferative efficacy in the colon cancer cell line COLO 320DM in vitro.

21. X-ray bursts with photospheric radius expansion and the gravitational redshift of neutron stars

Abstract

A method is described which allows, in principle, the gravitational redshift to be determined from the surface of a neutron star from observations of bursts with radius expansion. Results obtained from an application of this method to four burst sources are reported in detail. It is found that the method is very sensitive to systematic errors.

Abstract

A series of Exosat observations of the 'atoll' source 4U/MXB 1636 - 53 shows that duration and temperature of the X-ray bursts strongly correlate with the X-ray spectral and fast variability characteristics of the persistent emission of the source. This implies that spectral shape, fast variability, and burst duration and temperature all correlate well with accretion rate M. This provides a strong argument that in the atoll sources, source-state is determined by M, just as in the Z sources. These observations also show that the persistent X-ray intensity can vary independently from the other mentioned characteristics. Therefore, intensity is probably not a good measure for the accretion rate.

Abstract

A detailed analysis is presented of the importance of Comptonization in burst and persistent spectra of the low-mass X-ray binary 4U/MXB 1636-53, and from this analysis it is inferred that the inner accretion flow is geometrically thin. It is found that burst spectra of 1636-53 are very nearly Planckian in shape; from an upper limit to a high-energy excess in these spectra it is inferred that the Thomson scattering optical depth of a possible intervening hot cloud must be less than 1 during bursts, and that the Compton y parameter of that cloud must be less than 0.5. During persistent emission, Thomson optical depth of 4-8, an electron temperature of 2-5 keV, and a value of 0.8-1.1 for y are inferred.

Abstract

PURPOSE: lower lobe lung tumors move with amplitudes of up to 2 cm due to respiration. To reduce respiration imaging artifacts in planning CT scans, 4D imaging techniques are used. Currently, we use a single (midventilation) frame of the 4D data set for clinical delineation of structures and radiotherapy planning. A single frame, however, often contains artifacts due to breathing irregularities, and is noisier than a conventional CT scan since the exposure per frame is lower. Moreover, the tumor may be displaced from the mean tumor position due to hysteresis. The aim of this work is to develop a framework for the acquisition of a good quality scan representing all scanned anatomy in the mean position by averaging transformed (deformed) CT frames, i.e., canceling out motion. A nonrigid registration method is necessary since motion varies over the lung. METHODS AND MATERIALS: 4D and inspiration breath-hold (BH) CT scans were acquired for 13 patients. An iterative multiscale motion estimation technique was applied to the 4D CT scan, similar to optical flow but using image phase (gray-value transitions from bright to dark and vice versa) instead. From the (4D) deformation vector field (DVF) derived, the local mean position in the respiratory cycle was computed and the 4D DVF was modified to deform all structures of the original 4D CT scan to this mean position. A 3D midposition (MidP) CT scan was then obtained by (arithmetic or median) averaging of the deformed 4D CT scan. Image registration accuracy, tumor shape deviation with respect to the BH CT scan, and noise were determined to evaluate the image fidelity of the MidP CT scan and the performance of the technique. RESULTS: Accuracy of the used deformable image registration method was comparable to established automated locally rigid registration and to manual landmark registration (average difference to both methods < 0.5 mm for all directions) for the tumor region. From visual assessment, the registration was good for the clearly visible features (e.g., tumor and diaphragm). The shape of the tumor, with respect to that of the BH CT scan, was better represented by the MidP reconstructions than any of the 4D CT frames (including MidV; reduction of "shape differences" was 66%). The MidP scans contained about one-third the noise of individual 4D CT scan frames. CONCLUSIONS: We implemented an accurate method to estimate the motion of structures in a 4D CT scan. Subsequently, a novel method to create a midposition CT scan (time-weighted average of the anatomy) for treatment planning with reduced noise and artifacts was introduced. Tumor shape and position in the MidP CT scan represents that of the BH CT scan better than MidV CT scan and, therefore, was found to be appropriate for treatment planning
