Search

A needs assessment was conducted by the Utah State University Dairy Extension Team to identify the most urgent issues faced by Utah dairy farmers. The assessment found that the top five priority needs were implementing new technology, improving public perception of water use, managing risk, succession planning, and reducing feed costs. The results will guide dairy Extension programming in Utah. Several workshops and events have already been planned to address these needs. Overall, the assessment will help USU Extension provide research-based educational programs to meet the needs of the dairy industry in Utah.

Japanese encephalitis virus (JEV), a zoonotic flavivirus, is principally transmitted by hematophagous mosquitoes, continually between susceptible animals and incidentally from those animals to humans. For almost a century since its discovery, JEV was geographically confined to the Asia-Pacific region with recurrent sizable outbreaks involving wildlife, livestock, and people. However, over the past decade, it has been detected for the first time in Europe (Italy) and Africa (Angola) but has yet to cause any recognizable outbreaks in humans. JEV infection leads to a broad spectrum of clinical outcomes, ranging from asymptomatic conditions to self-limiting febrile illnesses to life-threatening neurological complications, particularly Japanese encephalitis (JE). No clinically proven antiviral drugs are available to treat the development and progression of JE. There are, however, several live and killed vaccines that have been commercialized to prevent the infection and transmission of JEV, yet this virus remains the main cause of acute encephalitis syndrome with high morbidity and mortality among children in the endemic regions. Therefore, significant research efforts have been directed toward understanding the neuropathogenesis of JE to facilitate the development of effective treatments for the disease. Thus far, multiple laboratory animal models have been established for the study of JEV infection. In this review, we focus on mice, the most extensively used animal model for JEV research, and summarize the major findings on mouse susceptibility, infection route, and viral pathogenesis reported in the past and present, and discuss some unanswered key questions for future studies.

In this study we determined the impact of increasing dietary whole cottonseed (WCS) on rumen fermentation, nutrient digestibility, and microbial community composition. This study contributes novel information to the dairy community deepening the understanding of how including different levels of WCS can affect the rumen environment. This research was conducted in continuous culture fermenters. Treatments included a control diet without WCS, or the control diet plus 5, 10, or 15% (dry matter) WCS. The control diet was a 50:50 orchardgrass hay:concentrate mixture fed twice daily. In the second experiment, soybean meal and cottonseed meal (CSM) were included, and rations were balanced for fiber and protein. For the first experiment, no treatment effect was observed for neutral detergent fiber (NDF) or starch digestibility, or for acetate concentration. Butyrate and ammoniacal nitrogen concentrations increased, while propionate concentration tended to decrease as WCS was added. Increasing WCS in the diet decreased protozoa count and relative abundance of archaea. Firmicutes abundance increased whereas Bacteriodetes decreased with the addition of WCS. Lachnospiraceae and Prevotellaceae were the most abundant bacteria families in all the samples. The two most abundant genera across samples were Prevotella and Pseudobutyribrio, and the addition of WCS decreased the abundance of both. For the second experiment, with diets adjusted for fiber and protein, inclusion of WCS did not affect NDF digestibility or ammonia production. Acetate and propionate decreased with the addition of WCS while no treatment effect was observed for butyrate production. Bacteria population increased with inclusion of WCS while the opposite was observed for archaea. Our results indicate that increasing levels of WCS up to 15% (dry matter) diet do not negatively impact fiber and starch digestion in the rumen. Furthermore, inclusion of WCS at affected the microbial community composition at different levels. In conclusion, supplementation of WCS had an overall positive impact on the rumen as it did not affect rumen fermentation or nutrient digestibility, and it increased bacteria population and decreased archaea concentrations.

Although traditional ryegrass pastures are still commonly used, they require intense management and input and do not perform well during the summer months. Multispecies systems have been recognised as more sustainable, needing less fertiliser and being tolerant to drought and heat. While data on monoculture system fat composition exist, information for multispecies is scarce. The present study compared the fatty acid (FA), carotenoid and fat-soluble composition of a multispecies system (consisting of perennial ryegrass, timothy, white clover, red clover, chicory and plantain) (MULTI) with two other conventional grazing systems (monoculture perennial ryegrass (PRG) and a binary mixture of perennial ryegrass and white clover (PRG+WC)) over the Irish late-summer grazing season of dairy cattle (July–September). The results showed that the three systems had similar levels of FAs (p > 0.05), with mean α-linolenic acid values of 20.00 mg/g in PRG, 18.51 mg/g in PRG+WC, and 17.90 mg/g in MULTI and mean linoleic acid values of 3.84 mg/g in PRG, 4.16 mg/g in PRG+WC, and 4.39 mg/g in MULTI. Fourfold and twofold increases in the concentrations of α-linolenic acid and linoleic acid, respectively, were observed throughout July–September. Variations in stearic acid concentrations were not significant (p > 0.05), and palmitic acid concentrations increased over time (p < 0.05). The average values of lutein (36.68 mg/kg in PRG, 31.26 mg/kg in PRG+WC, and 35.75 mg/kg in MULTI) and α-tocopherol (2.28 mg/kg in PRG, 1.49 mg/kg in PRG+WC, and 1.83 mg/kg in MULTI) were similar among the grazing systems (p > 0.05), and the average β-carotene value was 25% higher in monoculture ryegrass (140.74 mg/kg) than in the multispecies (102.51 mg/kg) (p < 0.05). Nonetheless, the levels of β-carotene found in the multispecies were still above the recommended intake required for ruminants. In conclusion, multispecies pastures could be included in dairy cattle production systems as a sustainable alternative to traditional pastures while also providing typically adequate sources of fats for animal diets.

Atrial Fibrillation (AF) is the most common cardiac arrythmia in adults with likely sex-specific risk factors. Female sex hormones may be important in modulating risk for atrial fibrillation. We hypothesize that pregnancy and progesterone (P4), a hormone found in high levels during pregnancy with abrupt withdrawal immediately after parturition, modulates AF susceptibility in female goats. Cardiac specific TGF-?1 transgenic female goats and age-matched wild-type (WT) female goats were utilized. Pacemakers were implanted in all animals for continuous arrhythmia monitoring and AF inducibility. AF inducibility was evaluated using 5 separate 10 s bursts of atrial pacing (160 - 200 ms). In the first study, all animals were bred (n=12) via natural service and circulating progesterone levels and AF inducibility were monitored throughout gestation. In the second study, seven female goats were treated with high-dose exogenous oral P4 supplementation over a two-week period, and AF inducibility was assessed at baseline, during P4 supplementation and 48-hours following P4 supplementation. Serum P4 was measured using a chemiluminescent assay. Cardiac tissue was collected from both atria from goats in a high P4 state and a P4 withdrawal. RNA was extracted and gene expression analysis was ran on several genes related to AF. Atrial tissue was also During pregnancy, P4 levels increased compared to baseline with a sharp withdrawal during the post-partum period. This decrease in P4 levels post-parturition was associated with an increase in AF inducibility in both TGF-?1 and WT goats. Exogenous P4 supplementation increased circulating P4 compared to baseline with a decrease in P4 levels below baseline at 48-hours post supplementation. Consistent with our hypothesis and observations from the pregnancy studies, abrupt withdrawal of P4 was associated with increased AF inducibility when compared to baseline and P4 supplementation. Several genes were differentially expressed in the P4 withdrawal group compared to the high P4 group. CACN2D, CACNB2, and CAMK2D, all calcium related genes were significantly downregulated in the progesterone withdrawal (P4 WD) group when compared to the high progesterone (HIGH P4) group. These data support the hypothesis that progesterone is an important modulator of AF susceptibility in females. More specifically, abrupt reduction of progesterone significantly increases AF risk. Our gene expression data shows that P4 modulates calcium signaling in the atrium.

Consumption of the total Western diet (TWD) in mice has been shown to increase gut inflammation, promote colon tumorigenesis, and alter fecal microbiome composition when compared to mice fed a healthy diet, i.e., AIN93G (AIN). However, it is unclear whether the gut microbiome contributes directly to colitis-associated CRC in this model. The objective of this study was to determine whether dynamic fecal microbiota transfer (FMT) from donor mice fed either the AIN basal diet or the TWD would alter colitis symptoms or colitis-associated CRC in recipient mice, which were fed either the AIN diet or the TWD, using a 2 × 2 factorial experiment design. Time-matched FMT from the donor mice fed the TWD did not significantly enhance symptoms of colitis, colon epithelial inflammation, mucosal injury, or colon tumor burden in the recipient mice fed the AIN diet. Conversely, FMT from the AIN-fed donors did not impart a protective effect on the recipient mice fed the TWD. Likewise, the composition of fecal microbiomes of the recipient mice was also affected to a much greater extent by the diet they consumed than by the source of FMT. In summary, FMT from the donor mice fed either basal diet with differing colitis or tumor outcomes did not shift colitis symptoms or colon tumorigenesis in the recipient mice, regardless of the basal diet they consumed. These observations suggest that the gut microbiome may not contribute directly to the development of disease in this animal model.

Ruminant production systems are major contributors to greenhouse gases emissions, with animal feeding practices being the main cause for methane and nitrous oxide’s release. Although feeding animals forages has been proven to be more sustainable, traditional ryegrass monocultures still require a lot of input (e.g., fertilisers and pesticides). Multi-species swards, consisting of different swards, such as grasses, forage legumes and herbs, need less management and fertiliser, produce more dry matter, and also add a variety of phytochemicals into the animal diet. In particular, polyphenols have been associated with a positive impact on animal health and productivity. However, data on the phenolic composition of multi-species sward components is still scarce, and little is known about the change in concentration over the grazing season. The present study investigated the antioxidant activity of six forage species (perennial ryegrass, timothy, white clover, red clover, chicory and plantain) over the Irish grazing season, using FRAP, DPPH•• and ORAC assays. The forages were screened for individual phenolic compounds using Liquid-Chromatography-Triple-Quadruple-Mass-Spectrometry. Plantain exhibited the highest antioxidant capacity, being almost one and a half times higher than timothy and double that of chicory. Chlorogenic acid was the most abundant polyphenol in perennial ryegrass, timothy and plantain. Overall, formononetin and biochanin A levels were higher in red clover, white clover and in chicory, in comparison to other forages (p < 0.05). Variations in antioxidant capacity and polyphenol composition were more significant between species (p < 0.01) than between season within species (p > 0.05). This study suggests that multi-species swards, regardless of the grazing month, offer a potential sustainable alternative to monoculture swards with significant antioxidant activity and nutraceutical compounds.

In-vitro evidence suggests that the lipid component of foods alters vitamin D absorption. This single-blinded, cross-over postprandial study examined the effect of changing the lipid component of a 20 µg vitamin D3 fortified dairy drink on postprandial 25(OH)D concentrations. Participants consumed one dairy drink per visit: a non-lipid, a pre-formed oleic acid micelle, an olive oil and a fish oil dairy drink. There was a significant time*drink*baseline status effect on 25(OH)D concentrations (p = 0.039). There were no time*drink, time or drink effects on 25(OH)D in vitamin D sufficient participants (>50nmol/L). However, there was an effect of time on changes in 25(OH)D concentrations after the olive oil dairy drink (p = 0.034) in vitamin D insufficient participants (

The objective of the current scoping review is to gather the current literature on the use of validated precision monitoring technologies for dairy cattle. This review will list the current precision monitoring technologies that have been validated for dairy cattle, describe the statistical method(s) utilized to validate them, and suggest potential practical applications of the technologies listed.

Embryos resulting from assisted reproductive technologies, such as in vitro fertilization (IVF) or intracytoplasmic sperm injection (ICSI), develop with lower efficiencies than embryos resulting from their in vivo counterparts. The reasons behind the developmental discrepancies remain largely unknown. Because the egg is the primary determinant of embryo developmental success, it is reasonable to consider inherent egg quality as a possible cause. The hypothesis for this project is that there are distinct mRNA transcript patterns, or molecular “fingerprints,” that distinguish high- versus low-quality eggs developing within the same environment. In this study, a small cytoplasmic biopsy was removed from 40 eggs and stored for later use. The eggs were chemically activated to stimulate development in the absence of sperm and cultured in vitro. Following an eight-day development period, embryos that reached the blastocyst stage (success) and embryos that failed to develop were identified. This was repeated five times, for a total of six experimental replicates. The relative transcript levels of 48 genes were evaluated in the egg biopsies corresponding to successful and failed embryos using quantitative PCR. Analysis of the qPCR data revealed no significant differences in levels of transcripts for any of the 48 genes between the successful and failed development groups. Therefore, the evidence does not support differential expression between the cytoplasmic biopsies from oocytes of divergent developmental success. However, this study provides technical innovation for future research of oocyte quality.

Three-dimensional (3D) food printing is a type of additive manufacture in which foods with certain rheological characteristics are mechanically layered to create 3D structures from a digitally model. 3D food printing can potentially offer consumers a range of benefits, such as personalised nutrition, customisable textures and unique structures and shapes. This multidisciplinary thesis outlines research completed in three key areas: engineering, food science and consumer science, reflecting the importance of evaluating 3D food printing in this holistic manner. The primary objectives of this work was three-fold: (1) to develop a practical 3D food printer design suitable for printing dairy-based ingredients; (2) to identify and develop suitable dairy-based recipes and investigate factors affecting their printability; and (3) to explore determinants of Irish consumer willingness to try 3D food printing applications. The rationale behind two selected 3D food printer designs (Cartesian and Delta), as well as a brief comparison between the functionality of both designs, are reported. The effects of printing parameters (i.e., nozzle diameter, distance to print bed, print speed etc.) on print quality are also discussed. Certain physicochemical factors such as pH and structural protein content were found to significantly affect the printability and texture of a basic processed cheese recipe. Samples with a higher pH (pH 5.8) tended to print less accurate grids and were significantly softer and less gummy, chewy and resilient than those with lower pH (≤ pH 5.6). Printed processed cheese recipes formulated with fresh curd (high structural protein content) resulted in significantly harder prints, yet printed grids inaccurately due to the material dragging during printing. Viscosity profiles were created for each processed cheese recipe using rheological methods and correlated with absolute printing precision values to identify a range of suitable recipes for accurate printing. The effect of various factors on the printability and functionality of Micellar Casein Concentrate (MCC) suspensions was also investigated. Increasing calcium chloride concentration (5 mM) of suspensions lead to significantly harder printed samples, which had lower dissolution and solubility rates than control or printed samples with lower calcium chloride concentration (1 mM). MCC suspensions printed in porous lattice structures were found to dissolve at a quicker rate than those printed in a hemisphere structure due to a higher surface area to volume ratio. An example of possible 3D-printed product concepts demonstrating the potential of MCC as a printing material is presented. In order to achieve a balanced and comprehensive understanding of 3D food printing technology from a number of perspectives, consumer acceptance research was incorporated into this study to compliment the food science and engineering narrative. Using qualitative methods, a series of themes were identified as forming Irish consumers’ perceptions of 3D food printing applications. Consumers’ affinity for naturalness and a strong association for unprocessed, homemade meals were considered barriers to acceptance of 3D-printed foods. As an extension of this study, data from quantitative research further revealed perceived personal relevance as a significant determinant affecting the dependent variable (i.e., willingness to try 3D food printing applications in the food service sector). Trust in science was found to diminish the negative effects of novel food technology neophobia on willingness to try. Potential solutions for negating factors affecting consumer acceptance are also discussed, which may be of benefit to those looking to market 3D food printing applications in the Irish marketplace. The findings from these studies present an opportunity for food sector stakeholders to utilise this knowledge as part of their foundation to build upon and create novel 3D food printer designs and printable formulations which are suitable for acceptance in the consumer market.

Research suggests that androgens increase skeletal muscle growth by modulating polyamine biosynthesis. As such, the objective of this study was to investigate effects of anabolic hormones, polyamine precursors, and polyamines relative to proliferation, protein synthesis, and the abundance of mRNA involved in polyamine biosynthesis, proliferation, and protein synthesis in C2C12 and Sol8 cells. Cultures were treated with anabolic hormones (trenbolone acetate and/or estradiol), polyamine precursors (methionine or ornithine), or polyamines (putrescine, spermidine, or spermine). Messenger RNA was isolated 0.5 or 1, 12, or 24 h post-treatment. The cell type had no effect (p > 0.10) on proliferation, protein synthesis, or mRNA abundance at any time point. Each treatment increased (p < 0.01) proliferation, and anabolic hormones increased (p = 0.04) protein synthesis. Polyamines increased (p < 0.05) the abundance of mRNA involved in polyamine biosynthesis, proliferation, and protein synthesis. Treatment with polyamine precursors decreased (p < 0.05) the abundance of mRNA involved in proliferation and protein synthesis. Overall, C2C12 and Sol8 myoblasts do not differ (p > 0.10) in proliferation, protein synthesis, or mRNA abundance at the time points assessed. Furthermore, anabolic hormones, polyamines, and polyamine precursors increase proliferation and protein synthesis, and polyamines and their precursors alter the abundance of mRNA involved in growth.

At calving, nutrient requirements of dairy cows increase to support milk synthesis. Energy and protein requirements are increased at the initiation of lactation (Moore et al., 2000). Additionally, calcium requirements increase tremendously to meet the demands of lactation (Moore et al., 2000). Calving and subsequent milk synthesis can cause calcium concentrations in the blood to drop. When the demand for calcium exceeds the cow’s ability to mobilize calcium, hypocalcemia (low blood calcium) occurs, which can negatively impact production. This fact sheet reviews hypocalcemia in dairy cows and how to implement hypocalcemia prevention strategies.

These data are part of the dissertation research of Michael D. Briscoe. This research analyzed the association between modernization and farmer alienation from their work and their cows, as well as their overall life satisfaction. The results show that modernization has mixed effects on farmer alienation. This statistical finding was supported by interviews with farmers, who expressed that large farms can make it hard to maintain a relationship with their cows, but that technology actually replaces negative aspects of this relationship and allows for new forms of interaction.

Significance There is a pressing need for new antiinfluenza therapeutic agents. We show that a molecularly engineered banana lectin (carbohydrate-binding protein) has broad-spectrum activity against all influenza strains tested, including drug-resistant and currently circulating strains; is safe upon repeated administration in mice; and, moreover, is efficacious at treating lethal influenza infection via clinically pertinent routes of administration. We demonstrate that the lectin binds to the viral hemagglutinin glycoprotein and exerts its primary antiviral effect via inhibition of an early stage of the viral life cycle, viral membrane fusion to the host endosomal membrane. Our findings indicate that this engineered lectin, which has a mechanism of action quite distinct from the presently available agents, has potential as an antiinfluenza agent., There is a strong need for a new broad-spectrum antiinfluenza therapeutic, as vaccination and existing treatments are only moderately effective. We previously engineered a lectin, H84T banana lectin (H84T), to retain broad-spectrum activity against multiple influenza strains, including pandemic and avian, while largely eliminating the potentially harmful mitogenicity of the parent compound. The amino acid mutation at position 84 from histidine to threonine minimizes the mitogenicity of the wild-type lectin while maintaining antiinfluenza activity in vitro. We now report that in a lethal mouse model H84T is indeed nonmitogenic, and both early and delayed therapeutic administration of H84T intraperitoneally are highly protective, as is H84T administered subcutaneously. Mechanistically, attachment, which we anticipated to be inhibited by H84T, was only somewhat decreased by the lectin. Instead, H84T is internalized into the late endosomal/lysosomal compartment and inhibits virus–endosome fusion. These studies reveal that H84T is efficacious against influenza virus in vivo, and that the loss of mitogenicity seen previously in tissue culture is also seen in vivo, underscoring the potential utility of H84T as a broad-spectrum antiinfluenza agent.

Characterization of the bovine leukocyte antigen (BoLA) DRB3 gene has shown that specific alleles associate with susceptibility or resilience to the progression of bovine leukemia virus (BLV), measured by proviral load (PVL). Through surveillance of multi-farm BLV eradication field trials, we observed differential phenotypes within seropositive cows that persist from months to years. We sought to develop a multiplex next-generation sequencing workflow (NGS-SBT) capable of genotyping 384 samples per run to assess the relationship between BLV phenotype and two BoLA genes. We utilized longitudinal results from milk ELISA screening and subsequent blood collections on seropositive cows for PVL determination using a novel BLV proviral load multiplex qPCR assay to phenotype the cows. Repeated diagnostic observations defined two distinct phenotypes in our study population, ELISA-positive cows that do not harbor detectable levels of provirus and those who do have persistent proviral loads. In total, 565 cows from nine Midwest dairy farms were selected for NGS-SBT, with 558 cows: 168 BLV susceptible (ELISA-positive/PVL-positive) and 390 BLV resilient (ELISA-positive/PVL-negative) successfully genotyped. Three BoLA-DRB3 alleles, including one novel allele, were shown to associate with disease resilience, *009:02, *044:01, and *048:02 were found at rates of 97.5%, 86.5%, and 90.3%, respectively, within the phenotypically resilient population. Alternatively, DRB3*015:01 and *027:03, both known to associate with disease progression, were found at rates of 81.1% and 92.3%, respectively, within the susceptible population. This study helps solidify the immunogenetic relationship between BoLA-DRB3 alleles and BLV infection status of these two phenotypic groupings of US dairy cattle.

Simple Summary Alternatives to antibiotic therapy for mastitis in dairy cattle are of interest to the dairy industry and society. Reduced use of antibiotics while maintaining or improving animal welfare is desirable. We studied the intramammary infusion of casein hydrolysate, alone or combined with standard dry cow treatment, at the beginning of the dry period before cows have their next calf. The effects on mammary involution and milk quality suggested that infusion of casein hydrolysate alone or combined with internal teat sealant may be an alternative to antibiotic dry cow therapy. Abstract Alternatives to routine antibiotic treatment of dairy cattle during the dry period before their next calving are of interest. This was a preliminary study of whether intramammary infusion of casein hydrolysate, administered alone or combined with standard dry treatment, accelerated the rate of mammary involution early in the dry period. Four treatments were studied in a split udder design. One udder half was assigned a treatment, and the contralateral half was administered dry cow treatment + internal teat sealant as a control. Treatments were casein hydrolysate, casein hydrolysate + dry cow treatment, casein hydrolysate + teat sealant and casein hydrolysate + dry cow treatment + teat sealant. Cows (n = 16) were blocked by a number of intramammary infections per udder half (0 or 1+) and randomized to treatments. Milk production was not different between control or treated udder halves post-calving. A generalized linear mixed model tested for differences between the treatment groups in the concentration of mammary involution indicators in milk: somatic cell count, bovine lactoferrin and bovine serum albumin. At 7 days, dry udder halves treated with casein hydrolysate had higher milk concentrations of lactoferrin than those treated with casein hydrolysate + dry cow treatment, casein hydrolysate + teat sealant and control. At 10 days dry, bovine serum albumin was higher in udder halves treated with casein hydrolysate than in those treated with casein hydrolysate + dry cow treatment, casein hydrolysate + dry cow treatment + teat sealant and control. Post-calving, casein hydrolysate-treated udder halves produced 51% of total milk, unchanged from before dry-off. There were seven total intramammary infections entering the dry period, all caused by coagulase-negative staphylococci. Cure rates (3/7, 43%) were not different among all treatments and control, partly because of the small sample size. Intramammary infusion of casein hydrolysate at the end of lactation may be an alternative or possible adjunct to antibiotic dry cow therapy.
