Given the importance of anadromous Northern Dolly Varden as a consumption staple for northern residents and the climate- and development-related impacts on total mercury (THg) concentrations, temporal changes in northern Dolly Varden THg concentrations were assessed between historical (1986–1988) and contemporary (2011–2013) periods from two rivers in the north-western Canadian Arctic. In the Rat River, mean THg changed from 79 ± 42 ng/g ww in 1986–1988 to 109 ± 44 ng/g ww in 2011–2013, while in the Firth River, THg changed from 126 ± 45 ng/g ww in 1986–1988 to 178 ± 47 ng/g ww in 2011–2012. Length adjusted values indicated increases in the Firth River were driven by the increased size of fish, but increases in the Rat River were not. After factoring in size, δ13C and δ15N, [THg] was found to be most influenced over time by fish size, but also significantly modified by temporal period and foodweb position. Relationships between log[THg] versus fork-length and log[THg] versus δ13C have remained constant over time in the Rat River, but not in the Firth River, while relationships between log[THg] versus δ15N have remained constant in the Firth River, but not in the Rat River. Changes in the significance and the slope of the relationships relating δ13C and δ15N to log[THg] suggests underlying bioaccumulative processes are temporally variant and will be sensitive to climate-driven changes in the aquatic environments within which fish live and feed. Abstract Unlike the normal anadromous lifestyle, Chinese native Dolly Varden char (Salvelinus malma) is locked in land and lives in fresh water lifetime. To explore the effect of freshwater adaption on its immune system, we constructed a pooled cDNA library of hepatopancreas and spleen of Chinese freshwater Dolly Varden char (S. malma). A total of 27,829 unigenes were generated from 31,233 high-quality transcripts and 17,670 complete open reading frames (ORF) were identified. Totally 25,809 unigenes were successfully annotated and it classified more native than adaptive immunity-associated genes, and more genes involved in toll-like receptor signal pathway than those in complement and coagulation cascades (51 vs 3), implying the relative more important role of toll-like receptors than the complement system under bacterial injection for the freshwater Dolly Varden char. These huge different numbers of TLR and complement system identified in freshwater Dolly Varden char probably caused by distinct evolution pressure patterns between fish TLR and complement system, representative by TLR3 and TLR5 as well as C4 and C6 , respectively, which were under purifying and positively selecting pressure, respectively. Further seawater adaptation experiment and the comparison study with our library will no doubt be helpful to elucidate the effect of freshwater adaption of Chinese native Dolly Varden char on its immune system. Perennial groundwater springs along the Alaska and Yukon North Slope provide overwintering habitat for various organisms, including birds and fishes. We observed an American Dipper, Cinclus mexicanus, in the open water of a perennial spring situated in Fish Creek, Yukon, in Ivvavik National Park on 8 March 2018. The observation at Fish Creek was among the most northern documented sightings of an American Dipper during the winter in North America. Moreover, the observation was approximately 650 km farther north than where American Dippers have been documented overwintering in Yukon, making this the most northern Canadian observation documented for this species in any season. Additionally, the American Dipper was photographed feeding on a juvenile Dolly Varden, Salvelinus malma. Although American Dippers are known to feed on small fish, our observation was a novel documentation of a trophic interaction between both species during winter. The open-water habitat in Fish Creek, which is important for both species and has not been previously described, was short (~730 m long), shallow (mean = 20 cm deep), narrow (mean = 2.8 m wide), and cold (mean water temperature = 0.34ºC). While there is little information regarding the ecological interactions of American Dipper overwintering in the Arctic, we note that all observations in the North Slope area during winter occurred in river systems also used by Dolly Varden, which indicates that juvenile Dolly Varden could be an important source of food for American Dipper in winter. Dolly Varden (Salvelinus malma) and Whitespotted Char (Salvelinus leucomaenis) are representative native fish of the family Salmonidae that inhabit the upper reaches of rivers on Hokkaido Island, Japan. They are threatened by the invasive Rainbow Trout (Oncorhynchus mykiss). In this study, environmental DNA (eDNA) real‐time polymerase chain reaction (PCR) assays to detect these three salmonids were developed and used to clarify the distribution pattern of these fish. A specificity test for each assay was conducted using DNA extracted from both target and closely related fish, and the specificity of each assay was confirmed. Then, we carried out eDNA surveys in two mountainous rivers around Mt. Daisetsu in winter, when snow depth was maximized. In the winter surveys, eDNA of all three species were successfully detected from river water samples, including under‐ice water samples. The results of eDNA detection corresponded with the results of an earlier distribution survey performed with Japanese‐style fly‐fishing and lure‐fishing. These results suggested that the eDNA assays developed in this study are applicable for inter‐seasonal surveys for these species. Otolith strontium and multi-year mark-recapture information were used to characterize associations between migration patterns and spawning frequencies in an anadromous Dolly Varden (Salvelinus malma) population (Rat River, Northwest Territories, Canada) that undertakes a long migration between freshwater spawning/overwintering (Fish Creek; a tributary to Rat River) and marine feeding habitats (Beaufort Sea) (~800 km round trip). Reconstructions of lifetime annual migration histories among otolith annuli was matched to information on reproductive status (current-year ‘spawner’ or ‘non-spawner’) that was known in two different, sometimes successive, years for each fish. Two migratory life histories were observed: fish either migrated annually after smoltification or periodically skipped an annual ocean migration to remain in freshwater and spawn. Different spawning frequencies were detected where fish not migrating annually tended to spawn in alternate years (84.6%) more often than those migrating annually (50%). Additionally, annually migrating fish had lower longevity (≤9 years vs. ≤13 years). The evaluation of differences in spawning frequency between sexes, independent of migration tactic, revealed males (84.6%) skipped spawning more often than females (51.2%) suggesting fitness trade-offs between life histories differ between sexes. Further, some fish returned from the sea considerably earlier than the majority of other current-year migrants. The streams draining Kamchatkan volcanic areas are perfect natural models producing the long-lasting effects similar to inorganic anthropogenic pollution. The most abundant local fish species Salvelinus malma is able to reproduce under highly unfavorable conditions: contamination with heavy metals and mineral suspensions. The aim of this study was to assess the adaptability of the landlocked and anadromous populations in adverse conditions. Here, we examined the liver toxicant accumulation in the overwintered S. malma juveniles, as well as aberrations in gills, internal organs, and peripheral blood. Compared to the anadromous populations the landlocked populations demonstrate poor ability to develop the mechanisms of decreasing toxicants’ bioaccumulation. The sets of histological and hematological alterations demonstrated a striking similarity for both ecotypes, and the difference in occurrence of tissue alterations was mostly insignificant. Extreme negative effects of the heavily polluted environment were: the secondary lamellas’ fusion, necrosis of liver and spleen tissues, erythrocyte morphological abnormalities, and elimination of germ cells. We suppose that the resistance to organs’ malfunctions is not the main adaptive vector in S. malma populations in the polluted streams, and expect the same long-term pattern of injury in the resident fish populations under anthropogenic pollution. The migratory history of Dolly Varden Salvelinus malma was investigated in 30 streams in the Shiretoko Peninsula, Hokkaido, Japan, which is in the southern part of their distribution. Captured Dolly Varden were classified into two categories by their body coloration: silvery fish were thought to be an anadromous form, and non-silvery fish a fluvial form that remains in freshwater throughout its life. Forty-seven of 1,739 Dolly Varden collected in the spring and ten of 2,126 Dolly Varden collected in the autumn were silvery fish. Twenty silvery fish and 120 non-silvery fish were examined by analysis of otolith Sr:Ca ratio. Three silvery fish from autumn and 15 non-silvery fish had migrated to salt water or brackish water. The other specimens of silvery fish were regarded as smolt or pre-smolt before seaward migration. The present study suggests that Dolly Varden in the Shiretoko Peninsula have a flexible migratory history, where some of the non-silvery fish utilize marine environments while some individuals follow an anadromous life history. This study aimed to evaluate the effects of feeding frequency on growth, antioxidant status, immune response and resistance to hypoxia stress challenge on juvenile dolly varden char Salvelinus malma (average weight: 9.40 ± 0.30 g). Fish were randomly assigned to one of six feeding frequencies (1, 2, 3, 4, 5 and 6 times/day) following the same ration size for 8 weeks. Weight gain increased significantly as feeding frequency increased from 1 to 5 times/day, but decreased with further increasing feeding frequency. Feed intake, whole body crude lipid content, serum aspartate transaminase, alanine transaminase and malondialdehyde content increased significantly as feeding frequency increased. Serum total antioxidant capacity increased significantly as feeding frequency increased from 1 to 5 times/day. Hepatosomatic index, serum lysozyme activity and 50% haemolytic complement content of fish fed 1–2 times/day were lower than those of fish fed 3–6 times/day. During the hypoxia challenge, cumulative mortality increased significantly as feeding frequency or hypoxia time increased. This study indicates that lower and higher feeding frequencies had a negative influence on growth of dolly varden char; the optimal feeding frequency to enhance growth and boost immunity of this species at juvenile stage is 4 times/day. Partial migration, in which a portion of the population migrates while the rest of the population remains as residents, is a common form of migration. Alternative migratory tactics (AMTs) of partial migration are often determined by polygenic threshold traits. However, the ultimate mechanisms that drive inter‐population variations in threshold traits are not well understood. We present a simple schematic model to explain how the threshold trait changes with fitness consequences under opposing natural and artificial selection forces. We conducted a field test to evaluate the effects of migration difficulty (as a natural selective force) and selective captive breeding (as an artificial selective force) on threshold traits of a partially migratory fish. Male masu salmon Oncorhynchus masou in the Shari River system have AMTs divided into three population categories of hatchery, wild/above the waterfall, and wild/below the waterfall (control). The wild/above the waterfall salmon live in a high‐migration‐cost situation, and the threshold trait changed in a direction that promoted residency. In hatchery salmon, which are produced by migrant‐selective captive breeding, the threshold trait changed in a direction that promoted migration. In contrast, Dolly Varden charr Salvelinus malma displayed only resident tactics, and the threshold trait did not differ between the populations above and below the waterfall, indicating that environment did not explain the variation in the threshold trait. Our results support the model and suggest that opposing natural and artificial selection forces drive variations in the threshold traits and migratory patterns in the studied species. In Salmonidae, subordinate males are exposed to higher risks of sperm competition than dominant males and thus are expected to improve the sperm characteristics (sperm concentrations, sperm velocity and sperm longevity). In this study, we investigated the relationships between body size and secondary sexual characters (breeding colour, hump height and snout length), and sperm characteristics of one-year-old (newly matured) Dolly Varden char. Small males displayed higher sperm concentrations than large males. Moreover, males with dull breeding colours, but not with lesser snout length and hump height, displayed an increased sperm velocity compared to males with bright colours, suggesting a trade-off between sperm quantity and the investment in breeding colour. In addition, sperm longevity decreased as sperm swimming velocity increased. These findings indicate that small males with dull breeding colours improve the quantity and quality of their sperm to a great extent to enhance their chances of reproductive success. Recreational angling is a popular leisure activity globally and can be a threat or useful conservation tool depending on the management strategy. Miyabe charr Salvelinus malma miyabei is an endemic fish that inhabits Lake Shikaribetsu, and a management program was established to harmonise recreational fishery and conservation. To examine the suitability of this program, population assessment was conducted with the cooperation of anglers. The population size in 2014 as assessed by tag-and-release angling was estimated to be 105,300 [95% confidence interval (CI) 37,300-178,600], much higher than the reported estimate in 1995. Further, angling mortality was estimated to be quite low. Moreover, no decreasing population trend was detected on analysing 8 years of anglers' catch data. Consequently, angling has facilitated stock assessments of Miyabe charr under the current program. Assimilation of mercury (Hg) into food webs is directly influenced by ecological factors such as local habitat characteristics, species feeding behavior, and movement patterns. Total Hg concentrations ([THg]) in biota from Subarctic latitudes are driven both by broad spatial processes such as long-range atmospheric transport and more local influences such as biovectors and geology. Thus, even relatively pristine protected lands such as national parks are experiencing Hg accumulation. We analyzed [THg] and stable isotopes of carbon (δ 13 C) and nitrogen (δ 15 N) in 104 Dolly Varden ( Salvelinus malma ) collected from two rivers in southeastern Alaska, upstream and downstream of apparent anadromous migration barriers in watersheds with and without glacial coverage. To assess the potential magnitude of marine-derived THg returning to freshwater, we analyzed [THg] in ten adult pink salmon from each study system. There were no differences in Dolly Varden mean [THg] between sites after the data were standardized for fork length, but unadjusted [THg] varied relative to fish size and δ 15 N values. While previous studies generally show that [THg] increases with higher δ 15 N values, we found that Dolly Varden below migration barriers and foraging on salmon eggs had the highest δ 15 N values among all sampled individuals, but the lowest [THg]. Dolly Varden residing below anadromous barriers had δ 13 C values consistent with marine influence. Since salmon eggs typically have low [Hg], our results suggest that abundant salmon populations and the dietary subsidy they provide may reduce the annual exposure to [Hg] in egg-eating stream fishes such as Dolly Varden. In addition to identifying a suitable species for freshwater Hg monitoring in southeastern Alaska, our study more broadly implies that river characteristics, location within a river, fish size, and feeding ecology are important factors influencing Hg accumulation. Total mercury (THg) concentrations were measured for different life-history types of Northern Dolly Varden ( Salvelinus malma malma) from the Babbage River, Yukon Territory, Canada to test hypotheses regarding the influence of life-history type, trophic position, and growth rate on THg tissue concentrations. In contrast to other northern anadromous fish species, higher measured THg concentrations were found in the anadromous life-history types. Standardization for common age, however, indicated that anadromous fish had lower THg concentrations than freshwater resident life-history types. δN was the most important factor in explaining observed differences among the individuals regardless of life-history types, with growth rate also contributing to explain among-individual variation. The contrast of higher absolute, but lower age- and size-adjusted THg levels in anadromous fish was explained by a combination of two counteracting mechanisms, including: (1) a switch to feeding at higher trophic levels and the use of more THg contaminated marine prey, and (2) somatic growth dilution that, with increasing growth efficiency, decreases THg as fish grow and age. Results underscore the importance of considering life-history variation in addition to trophic patterns, and fish sizes and age when interpreting mercury concentrations in fish with varying life histories. Salmon rivers that flow from volcanoes on the Kamchatka Peninsula are subjected to chronic pollution by mineral suspensions and dissolved metals. We analyzed the impact of chronic pollution on the body morphology of Dolly Varden parr using linear measurements and geometric morphometrics. Populations from different rivers demonstrated differences in morphology, although our results suggest that contamination is not responsible for the formation of any specific morphological features. The anadromous juveniles from polluted streams are characterized by high variability in external morphology, expressed in the increased standard deviation and coefficient of variation in distributions of morphometric traits. Mean standard deviation of the trait distributions in the samples from polluted rivers is significantly higher than in samples from streams not subjected to volcanic pollution. The increase of morphometric variability occurs gradually with fish age and affects a complex of characters, including the position of fins and their length. Isolation of Dolly Varden in polluted streams has other consequences for external morphology. The landlocked populations are characterized by channelization of the development and acquire novel morphologic features, which are reflected in peculiar values of some morphometric traits (especially upper jaw length and position of dorsal fin) together with reduced dispersion in these traits. Nucleotide sequences of the mitochondrial DNA cytochrome b ( CytB) gene fragment and the control region ( D-loop) of Dolly Varden ( Salvelinus malma) from the Commander Islands and the Kol River of the Kamchatka Peninsula were examined. A high level of genetic variability of island populations comparable to that of the mainland population of western Kamchatka was demonstrated. The trophic ecology (diet and head morphology) and migration patterns of two closely related salmonid fishes, Arctic char ( Salvelinus alpinus (L., 1758)) and Dolly Varden ( Salvelinus malma (Walbaum, 1792)), were examined in tributaries of Lake Aleknagik, southwestern Alaska, to test for differentiation between species. Schoener's index of proportional overlap and multivariate analyses of diets suggested that these species had significantly different trophic niches. Arctic char and the largest individuals of both species had the most diverse diets, and sockeye salmon ( Oncorhynchus nerka (Walbaum, 1792)) eggs dominated the diet of both species, especially Arctic char. Arctic char had larger jaws and wider heads than Dolly Varden of similar body length, which may contribute to interspecific diet difference. The species also differed in migration patterns; otolith microchemistry indicated that juvenile Arctic char were produced by nonanadromous mothers, whereas the mothers of the Dolly Varden had been to sea in the season prior to spawning. The species also segregate in spawning habitat (Arctic char in the lakes and Dolly Varden in streams), as well as in juvenile rearing habitat. Our study provides the first evidence of divergent feeding and migratory ecology between sympatric juvenile Arctic char and Dolly Varden, differences that may constrain hybridization and introgression between them. Assessment of anadromous northern-form Dolly Varden Salvelinus malma malma in the Western Canadian Arctic requires reliable methods for estimating ages. Additionally, conservation efforts warrant determining whether fin rays provide a non-lethal alternative to otoliths. Precision and bias of whole and sectioned otoliths, and sectioned pectoral and pelvic fin rays were examined. Two age readers with different levels of experience ageing this species read each structure three times. Coefficient of variation (CV) was calculated to measure precision, and age bias plots were created for each method of preparation/structure within and between readers. The experienced reader demonstrated the highest precision with sectioned otoliths (CV = 1.6 %) followed by whole otoliths (CV = 4.2 %) while pectoral and pelvic fins were the lowest, CV = 7.7 % and 7.5 %, respectively. The age bias plot showed little difference between whole and sectioned otoliths, although greater imprecision/bias was evident for whole otoliths at age ≥9. Compared to otoliths, fin rays produced younger age estimates starting at 5 years; however, pelvic fins were more biased towards younger estimates than pectoral fins. The less experienced reader had greater inconsistencies, tending to overage younger and underage older samples for all methods compared to the more experienced reader, underscoring the importance of experience when estimating age for this species. We conclude that both types of fin rays are a poor non-lethal alternative to otoliths for fish ≥5 years and recommend an experienced ager could use whole otoliths up to age 8 and sectioned otoliths for fish ≥9 years (>500 mm fork length). In Arctic Alaska, Dolly Varden Salvelinus malma is highly valued as a subsistence fish; however, little is known about its marine ecology. New advances in electronic tagging, such as pop-up satellite archival tags (PSATs), provide scientists with a fishery-independent means of studying several aspects of this species' movement and ecology. To evaluate the usefulness of this technology, we attached 52 PSATs to Dolly Varden in the Wulik River, which flows from northwestern Alaska into the Chukchi Sea, to study several characteristics of the marine habits of this species. Overall, PSATs provided unprecedented information about summer dispersal of Dolly Varden, including the first evidence of offshore dispersal in the Chukchi Sea, as well as previously documented dispersal types such as movement to other rivers and southerly nearshore movements in northwestern Alaska. On the basis of minimal observable evidence of tag-induced behavioral effects, as well as movements of more than 450 km by fish at liberty (i.e., between tag deployment and release or recapture), we conclude that PSATs offer an effective alternative method for studying several aspects of Dolly Varden dispersal and ecology in areas where it is not practical or feasible to capture these fish, such as coastal and offshore regions of Arctic Alaska. The closely related chars Salvelinus malma and Salvelinus albus, which sympatrically inhabit the Kamchatka River basin and Kronotsky Lake (Kamchatka), attract the attention of the researchers because of their debated origin and taxonomic status. Previous studies of sympatric populations of these chars revealed small but statistically significant genetic differences between these species at a number of molecular markers, suggesting the presence of the genetic exchange and hybridization. In this study, based on genotypic characterization of nine microsatellite loci, a considerable level of historical and contemporary genetic migration between sympatric populations of these chars was demonstrated. At the individual level, a high degree of hybridization was observed, mainly among the Dolly Varden individuals from the studied populations. Numbers of masu salmon and Dolly Varden in streams in the Shiretoko Peninsula, location of the Shiretoko World Heritage Site in north eastern Hokkaido, appear to be declining. One concern is whether masu salmon smolts continue to migrate during July, after the existing May to June angling closure established to protect migrating smolts in eastern Hokkaido. Smolt timing and relative abundances were monitored in the Uebetsu River near the World Heritage Site from May to July during 2005-2007. Each year some masu salmon smolts emigrated during July, especially in 2005 when 82% of smolts captured were caught that month. In contrast, most anadromous Dolly Varden smolts emigrated prior to early June when stream temperatures <8°C. Mean fork lengths of smolts in both species decreased during the runs. Determination of unit-stock structure and the assignment of individuals from mixed groups to stock-of-origin is important to conservation and management of Arctic riverine fishes. Northern form Dolly Varden Char ( Salvelinus malma malma ) are closely linked to perennial groundwater sources during the freshwater stages of their life cycle. This close connectivity provides an ideal circumstance for stock discrimination using archived sagittal otoliths (1986–2007) collected from the river drainages of the study area (Firth River, Yukon Territory (YT), Babbage River, YT, Big Fish River, Northwest Territories (NT), Rat River, NT, and Vittrekwa River, NT). Laser ablation-ICP–MS techniques were employed to measure 87 Sr/ 86 Sr isotopes and trace elements (Sr, Ba, S, Zn, Mg, P) from dorso-ventral otolith thin sections. Random forest model, learning algorithm and tree classification provided 94.4% classification success for individual fish to the river systems (putative stocks). Misclassifications within the model occurred among the Vittrekwa, Babbage, and Rat river drainages. Strontium, 87 Sr/ 86 Sr ratio and Ba were the most significant discriminators within the model, although removal of the remaining elements reduced overall discriminatory power. Microchemical analyses of archived otoliths provided high classification and discrimination success thus can be used as a tool for fisheries managers in developing effective management and conservation plans regarding mixed-stock fisheries for this species. The northern Dolly Varden ( Salvelinus malma malma) displays variable life-history types and occupies freshwater habitats with varying levels of connectivity. Here, we assayed microsatellite DNA variation in northern Dolly Varden from the western Canadian Arctic to resolve landscape and life-history variables driving variation in genetic diversity and population structure. Overall, genetic variation was highest in anadromous populations and lowest in those isolated above waterfalls, with stream-resident forms intermediate between the two. Anadromous and isolated populations were genetically divergent from each other, while no genetic differentiation was detectable between sympatric anadromous and stream-resident forms. Population structure was stable over 25 years, hierarchically organized, and conformed to an isolation-by-distance pattern, but stream-isolated forms often deviated from these patterns. Gene flow occurred primarily among Yukon North Slope populations and between sympatric anadromous and resident forms. These results were sex-dependent to some extent, but were influenced more by reproductive status and life history. Our study provides novel insights into the life history, population demographic, and habitat variables that shape the distribution of genetic variation and population structure in Arctic fluvial habitats while providing a spatial context for management and conservation. Understanding the extent of interspecific hybridization and how ecological segregation may influence hybridization requires comprehensively sampling different habitats over a range of life history stages. Arctic char ( Salvelinus alpinus) and Dolly Varden ( S. malma) are recently diverged salmonid fishes that come into contact in several areas of the North Pacific where they occasionally hybridize. To better quantify the degree of hybridization and ecological segregation between these taxa, we sampled over 700 fish from multiple lake (littoral and profundal) and stream sites in two large, interconnected southwestern Alaskan lakes. Individuals were genotyped at 12 microsatellite markers, and genetic admixture ( Q) values generated through Bayesian-based clustering revealed hybridization levels generally lower than reported in a previous study (<0.6% to 5% of samples classified as late-generation hybrids). Dolly Varden and Arctic char tended to make different use of stream habitats with the latter apparently abandoning streams for lake habitats after 2-3 years of age. Our results support the distinct biological species status of Dolly Varden and Arctic char and suggest that ecological segregation may be an important factor limiting opportunities for hybridization and/or the ecological performance of hybrid char. Contact zones between divergent lineages of aquatic taxa have been described from the northeastern Pacific Ocean. We surveyed samples of Dolly Varden ( Salvelinus malma) from their North American range for variation at 14 microsatellite DNA loci. After accounting for hybridization between Dolly Varden and co-occurring bull trout ( Salvelinus confluentus) and Arctic char ( Salvelinus alpinus), we found evidence for two genetic lineages of Dolly Varden consistent with the previously recognized subspecies, northern Dolly Varden ( S. m. malma) and southern Dolly Varden ( S. m. lordii). We documented a contact zone between the two subspecies from the eastern Alaska Peninsula to Cook Inlet, Alaska, where admixture values (i.e., the proportion of the genome estimated to be composed of northern Dolly Varden, QNDV) ranged between QNDV = 0.245 and 0.754 across about 700 ocean kilometres. Populations of Dolly Varden showing low admixture (i.e., less than 5%) were located a minimum of 346 km to the west to 1200 km to the southeast, respectively, from the contact zone. The two lineages of Dolly Varden probably stem from isolation and subsequent divergence in, and dispersal from, distinct northern and southern Pleistocene glacial refugia and substantiate the treatment of S. malma as two subspecies and as at least two designatable units under Canada's Species at Risk Act. Animal migrations often evolve to exploit seasonal variation in foraging opportunities. Many migrants move among discrete habitats using environmental cues to time their arrivals with seasonal peaks in food abundance. This type of migratory behaviour has been hypothesized to be particularly vulnerable to climate change, which can generate asynchrony between the seasonal timing (phenology) of animal movements and periods of resource abundance in destination habitats., While many studies have documented climate-induced mismatches between migrants and the lower trophic levels they target, virtually none have explored the consequences of altered migration phenology on the higher trophic levels that migrants often subsidise. We explored the roles of climatic variation and prey migrations in driving the seasonal migrations of predators., We show that in a warming catchment where prey migrations have changed substantially over the last 4 decades, migrations of Dolly Varden ( Salvelinus malma) (predator) remain synchronised with annually variable Pacific salmon ( Oncorhynchus spp.) (prey) migrations and appear to be cued directly by salmon migration rather than environmental conditions., In contrast to many previous studies, our results suggest that some wide-ranging predators may be resilient to altered prey phenology. Previous research has documented the significance of total mercury (THg) as a northern contaminant in general and of fish in particular. While much research has been devoted to documenting both spatial and temporal changes in THg in consumed fish, little effort has been directed at understanding patterns of THg in Dolly Varden ( Salvelinus malma ), a prized subsistence species throughout the western North American Arctic. Here we report historical THg concentrations for anadromous Dolly Varden from 10 populations in the Yukon and Northwest Territories sampled across a range of latitudes (67–69°N) and longitudes (136–141°W) between the years 1988–91. Unadjusted mean THg concentrations ranged from 15 to 254 ng/g wet weight. Length-adjusted THg concentrations were significantly different among sites, but were not related to latitude or longitude. Within and among populations, THg was significantly related to fork-length, age, δ 15 N, and δ 13 C, with the variation in THg found among populations being best explained by size. The data serve as an important baseline against which future changes in THg levels in this important subsistence fishery may be compared to determine the significance of any observed trends. The northern Dolly Varden, Salvelinus malma malma, is a typical representative of arctic fauna distributed in northeastern Asia and northwestern North America. Because its spawning habitats were affected by Pleistocene glacial advances over most of its natural range, S. m. malma is among the most interesting objects of phylogeographic and microevolutionary studies. We reconstructed the genealogy of mt DNA haplotypes from 27 Alaskan and Asian populations to study the influence of glacial and geological vicariance events on the contemporary population genetic structure, phylogeographic subdivision and distribution of the northern Dolly Varden. Analysis of restriction site states in three PCR-amplified mt DNA regions ( ND1/ ND2, ND5/ ND6, Cytb/ D-loop; 47% of the mitochondrial genome) resolved 75 haplotypes in 436 fish. Similar patterns of subspecific variation apparent from hierarchical diversity and nested clade analyses of mt DNA haplotypes identify weak spatial differentiation and low levels of divergence. Our results suggest that (1) demographic history has been influenced by historical range expansions and recent isolation by distance, (2) present populations from Asia and North America were colonized from one main Beringian Refugium, and (3) that this taxon's ancestral population probably experienced a bottleneck in the Beringian Refugium during the late Pleistocene ( Wisconsin) glacial period. Numerous studies of population genetics in salmonids and other anadromous fishes have revealed that population structure is generally organized into geographic hierarchies (isolation by distance), but significant structure can exist in proximate populations due to varying selective pressures (isolation by adaptation). In Chignik Lakes, Alaska, anadromous Dolly Varden char ( Salvelinus malma) spawn in nearly all accessible streams throughout the watershed, including those draining directly to an estuary, Chignik Lagoon, into larger rivers, and into lakes. Collections of Dolly Varden fry from 13 streams throughout the system revealed low levels of population structure among streams emptying into freshwater. However, much stronger genetic differentiation was detected between streams emptying into freshwater and streams flowing directly into estuarine environments. This fine-scale reproductive isolation without any physical barriers to migration is likely driven by differences in selection pressures across freshwater and estuarine environments. Estuary tributaries had fewer larger, older juveniles, suggesting an alternative life history of smolting and migration to the marine environment at a much smaller size than occurs in the other populations. Therefore, genetic data were consistent with a scenario where isolation by adaptation occurs between populations of Dolly Varden in the study system, and ecological data suggest that this isolation may partially be a result of a novel Dolly Varden life history of seawater tolerance at a smaller size than previously recognized. Essential overwintering habitats for anadromous Dolly Varden Salvelinus malma on Alaska's North Slope appear to be limited to a small number of perennial springs, primarily in eastern Brooks Range drainages. Because future petrochemical development in the region continues to be a possibility, and development would require large quantities of freshwater, we sought to identify and document the overwintering areas used by Dolly Varden in the Hulahula River, eastern Brooks Range. In August 2007, we implanted 52 Dolly Varden with multi-year radio transmitters at a known overwintering area in the lower Hulahula River. Other wintering areas were located during 11 aerial surveys conducted over the next 2.5 years. A stationary receiver located in the lower Hulahula River provided migration timing information. Radio-tagged Dolly Varden used four discrete areas with perennial springs for overwintering in the Hulahula River drainage. The springs, totaling approximately 12 km in stream length, were located between river km 40 and 105. Radio-tagged Dolly Varden migrated downstream on their way to the Beaufort Sea in early June. Most tagged fish known to have survived the summer at sea returned to the Hulahula River during late July and August, but seven fish overwintered in other North Slope drainages. Within the Hulahula River drainage, 15 fish overwintered in more than one area during the three winters of the project, but only the four identified perennial spring areas were used. Large digestive organs increase rates of energy gain when food is plentiful but are costly to maintain and increase rates of energy loss when food is scarce. The physiological adaptations to this trade-off differ depending on the scale and predictability of variation in food abundance., Currently, there is little understanding of how animals balance trade-offs between the cost and capacity of the digestive system in response to resource pulses: rare, ephemeral periods of resource superabundance. We investigated the physiological and behavioural tactics of the fish Dolly Varden ( Salvelinus malma) that rear in watersheds with low in situ productivity, but experience annual resource pulses from the spawning migrations of Pacific salmon. The eggs of Pacific salmon provide high-energy food for Dolly Varden., Dolly Varden sampled 6 weeks prior to the resource pulse exhibited atrophy of the stomach, pyloric caeca, intestine and liver. Throughout the portion of the growing season prior to the resource pulse, fish exhibited empty stomachs, low indices of energy condition and muscle isotope signatures reflecting the previous resource pulse., During the resource pulse, Dolly Varden exhibited large digestive machinery, gorged on salmon eggs and rapidly stored energy in fat reserves, somatic growth and gonad development. Dolly Varden appeared to achieve nearly their entire annual energy surplus during the ∼5-week period when sockeye salmon spawn., Digestive flexibility provides Dolly Varden the energy efficiency required to survive and reproduce when resource abundance is concentrated into an annual pulse that is predictable, yet highly ephemeral. Although fish are known to incur extremely variable energy budgets, our study is one of the first to document digestive flexibility in wild fish. Our study emphasizes that fish can rely heavily on rare, high-magnitude foraging opportunities. Dolly Varden ( Salvelinus malma) are a facultatively anadromous salmonid common around much of the North Pacific Rim, but little is known about the environmental factors affecting the timing and diversity of their migration. We combined telemetry of anadromous fish with long-term monitoring of Dolly Varden upstream migration timing and environmental data in the Chignik Lakes watershed in Alaska and then compared the timing data with that of other streams where only count data were available. Telemetry revealed two upstream migration modes: midsummer and late fall at the Chignik Lakes. Weir counts indicated that timing fluctuated markedly over the monitoring period (1996-2011) and was negatively correlated with June sea surface temperature. The relationship between sea surface temperature and migration timing in other watersheds with long-term records was as follows: negative (Buskin River), positive (Auke Creek), or nonexistent (Goodnews and Kanektok rivers). Among 18 streams and rivers throughout the eastern Pacific range of Dolly Varden, median upstream migration date increased with latitude. Overall, Dolly Varden migration timing is more variable, protracted, and more strongly influenced by local sea surface temperatures than is typical of semelparous salmonids. Streams were surveyed along the Mackenzie Valley, Northwest Territories, to investigate the distribution and biology of riverine char. Bull trout (Salvelinus confluentus) were captured in 24 of 43 streams sampled from three watersheds: the Kotaneelee River, the South Nahanni River, and the Central Mackenzie Mountains. The first confirmed sympatric bull trout and Dolly Varden (Salvelinus malma) populations in the Northwest Territories are reported from the Gayna River (~65°17' N, 129°21' W). This location represents a geographical range extension for bull trout approximately 200 km northwest of the previous northernmost published location in the central Northwest Territories (~64°30' N and 125°00' W, Great Bear River). It also extends the geographical range of Dolly Varden south approximately 300 km from the closest known populations located in the Peel River basin. The identification of this geographic range expansion is a result of extending both our sampling range and our ability to identify riverine chars accurately. In bull trout, we observed two different growth patterns, which correspond to non-migratory and migratory life histories. Northern bull trout populations are relatively small and widespread, and they are separated from one another by large distances and in some cases by natural barriers. Although these populations share similar biological traits with their southern counterparts, the northern fish mature later and do not spawn as often, which increases their sensitivity to exploitation and other anthropogenic perturbations. The level of genetic differentiation of northern Dolly Varden char Salvelinus malma malma from Asia and North America was evaluated using the data on mtDNA variation (regions ND1/ND2, ND5/ND6, and Cytb/D-loop) obtained by means of PCR-RFLP analysis. For S. m. malma, the mean values of haplo-type and nucleotide diversity were 0.5261 ± 0.00388 and 0.001558, respectively. The mean estimate of the population nucleotide divergence constituted 0.055%. It was demonstrated that S. m. malma on the most part of the species range examined (drainages of the Beaufort Sea, Chukotka Sea, Bering Sea, and the Sea of Okhotsk) was characterized by the population genetic structure with the low level of genetic differentiation and divergence. At the same time, populations from the Pacific Ocean Gulf of Alaska demonstrated marked genetic differentiation, supported by the high pairwise G4ST values (from 0.4198 to 0.5211) and nucleotide divergence estimates (mean divergence, 0.129%), from Asian and North American populations. Analysis of molecular variance (AMOVA) showed that most of the mtDNA variation in S. m. malma fell in the intrapopulation component (72.5%). - Spatial structures can drive population dynamics and persistence. The roles of space and dispersal on dynamics, however, remain largely unknown in wild populations, mainly because of the difficulty in extensive/intensive field surveys at multiple locations. Here, I integrate results of ecological, demographic and genetic studies to elucidate detailed population structure and to identify the mechanisms of population persistence in a stream fish. Spatial structures, such as habitat size and connectivity, are particularly important in river and freshwater networks because such ecosystems are highly vulnerable to human activities. These issues are explored by examining results from a decadal research on the stream-dwelling Dolly Varden charr in the Sorachi River basin, Japan. More than 100 local habitats (i.e. tributaries) were examined for spatial structure of populations by, such as, species presence, population census, redd counts, analysis of population synchrony and microsatellite DNA analysis. The results indicate that (i) population demography is largely independent in each tributary, (ii) some large tributaries support self-sustaining populations, (iii) despite small population sizes in most tributaries (<30 spawning females), local extinction is rare, except in human-influenced areas and (iv) probabilities of extinction are likely reduced by immigration of fish from neighbouring tributaries. The evidence suggests that Dolly Varden in this river system function as a source-sink or mainland-island metapopulation structure and that processes influencing these structures vary within the stream network. Gametes were collected from Dolly Varden char ( Salvelinus malma) from waterbodies in a region exposed to mining-related selenium (Se) releases in British Columbia, Canada. Fertilized eggs were incubated in a laboratory and deformities were assessed on newly-hatched alevins using a graduated severity index. No effects were observed on egg or alevin survival or larval weight across the studied exposure range of 5.4 to 66 mg/kg dry weight in egg. Length of some larvae was reduced at the highest egg Se concentrations and a clear residue-response relationship was observed for larval deformity. The egg concentration corresponding to a 10% increase in the frequency of deformity (EC10) was 54 mg/kg dry weight, which is substantially higher than reported for other cold-water fish species. Hatchery-reared adult brown trout, Salmo trutta v. fario L., [215-335 mm standard length ( L), n = 82] were individually tagged and released into three sections of the Blanice River in May 2007. Wild populations of brown trout and grayling, Thymallus thymallus, L., in these sections and three non-stocked control sections were also tagged. The recapture rate of hatchery-reared adult brown trout after 6 months (18%, n = 15) was comparable to that of wild adult brown trout in stocked (15%, n = 14) and control (14%, n = 11) sections. The recapture rates of wild brown trout and grayling after 6 months were higher in control sections than in stocked sections, but the differences were not significant. The movement of recaptured large juvenile wild brown trout from stocked sections was significantly higher (36%) than from control sections (9%). Genetic variation at eight microsatellite loci was examined in 21 populations of Dolly Varden charrs, Salvelinus malma, representing five geographical regions (Kamchatka Peninsula, Sea of Okhotsk coast, Sea of Japan coast, Sakhalin Island, and Kuril islands) . Hierarchical analysis of molecular variance showed that 11% (58% in terms of R-statistics) of the variation was distributed among of northern subspecies of Dolly Varden, Salvelinus malma malma and southern Asian Dolly Varden, S. m. krascheninnikovi while similar values were attributed to the among-regional level within northern Dolly Varden 9% (7%) and southern Asian Dolly Varden 11% (14%). Permutation-based tests indicated a mutational component to genetic differentiation based on allelic size variance and suggested that divergence of the two subspecies had occurred at least 3,000 generations ago. On large spatial scales (within the Asian range of the species), populations clustered according to their geographical location. On smaller scales (within regions and subregions) correlation between genetic and geographic distances was not significant. Abstract: Previous studies have indicated that salmonid fertilization success may be very sensitive to elevated concentrations of total dissolved solids (TDS) with effects at concentrations as low as 250mgl−1 being reported. However, interpretation of these studies is complicated by poor control performance and variable concentration response relationships. To address this, a series of experiments were performed to evaluate TDS effects on Arctic Grayling (Thymallus arcticus) and Dolly Varden (Salvelinus malma) fertilization success and identify possible mechanisms for previously observed test variability and any observed effects of TDS. Results indicate that some of the experiments reported here were likely confounded by extended milt holding times prior to experiment initiation. Milt holding times >6h were shown to significantly reduce control fertilization and corresponding concentration response relationships were variable. When milt holding time was minimized during fertilization experiments, consistent control performance with >90% control fertilization was achieved and consistent concentration response relationships were observed for both species examined. Experiments performed under these conditions indicate that Arctic Grayling and Dolly Varden fertilization success is not sensitive to elevated TDS with EC20s (concentration causing 20% effect) of >2782 and >1817mgl−1 (the highest concentrations tested), respectively. However, TDS was shown to significantly affect embryo water absorption during the water hardening phase immediately following fertilization. The lowest observable effect concentrations (LOECs) for this endpoint were 1402 and 964mgl−1 for Arctic Grayling and Dolly Varden, respectively. The effect of reduced embryo turgidity, due to impaired water absorption, on resistance to mechanical damage under real world conditions needs further investigation in order to understand the implications of this observed effect. Comparative analysis of mitochondrial DNA variation was performed in white char Salvelinus albus Glubokovsky and in its putative ancestor species, northern Dolly Varden char Salvelinus malma malma Walbaum. Highly statistically significant differentiation of S. albus and S. m. malma in the areas of sympatric (Kamchatka River basin) and allopatric (Kronotskoe Lake and Kronotskaya River) residence was demonstrated. The mtDNA divergence between S. albus and S. m. malma did not exceed the range of intraspecific variation in the populations of northern Dolly Varden char. At the same time, clusterization pattern of the Salvelinus chars provides hypothesis on the common origin of two allopatric populations of white char. Genealogical analysis of haplotypes indicates that S. albus and S. m. malma currently demonstrate incomplete divergence of mitochondrial lineages. The low nucleotide divergence estimates between S. albus and S. m. malma reflect the short time period since the beginning of the divergence of ancestral lineages. Phylogeography of southern Asian Dolly Varden char was studied using the data on mtDNA variation (regions ND1/ND2, ND5/ND6, and Cytb/D loop) obtained using PCR-RFLP analysis. Analysis of contemporary population genetic structure showed that Salvelinus malma krascheninnikovi throughout the whole species range was characterized by high population differentiation in combination with rather small differences between the populations from remote regions. The genealogy of mtDNA haplotypes was reconstructed and nested clade analysis of geographical distances was performed. Geographical distribution of mtDNA haplotypes of S. m. krascheninnikovi was explained by population genetic processes (restricted gene flow), as well as by historical demographic events (range expansion and fragmentation). It was demonstrated that the main demographic events were associated with cyclic processes of the geological formation of the Sea of Japan and adjacent territories. Numerous low-head dams have been constructed for erosion control and installed in almost all mountainous streams in Japan. Shiretoko Peninsula, Hokkaido, is considered as one of the most intact areas in Japan, however, more than 331 low-head dams have been installed in 39 streams of the peninsula since the 1960s. In this study, the density of stream-dwelling Dolly Varden ( Salvelinus malma) and the habitat characteristics in dam-installed sections were investigated in Shiretoko streams and compared with unaltered sections in which no dams have been installed as a reference. Fish density in dam-installed sections was significantly lower than reference sections. Canopy cover, maximum temperature, wetted width, frequencies of pebble, bedrock and boulder, and areas of rapid, glide and riffle was significantly differed between dam-installed and reference sections. High stream temperature, occurrence of glides, and loss of boulder in dam-installed sections may reduce the suitability of habitats for fish and minimize the population size. The article presents a study about the high risk of bottlenecks and inbreeding depression in the deformations of Dolly Varden charr or Salvelinus malma. It examines the impact of high frequency to the fin deformities in the fish due to inbreeding. It also notes that deformity occurs as a result of a loss of genetic diversity that correlated with the population of the fishes.

We review the ecology and conservation of three lesser known chars (genus Salvelinus): Dolly Varden (S. malma), white-spotted char (S. leucomaenis), and bull trout (S. confluentus). Dolly Varden is distributed across the northern Pacific Rim and co-occurs with bull trout and white-spotted char at the southern extremes of its range. In contrast, bull trout and white-spotted char are naturally isolated, with the former restricted to North America and the latter distributed in northeastern Asia. Though the range of Dolly Varden overlaps with the two other chars, it is most closely related to Arctic char (S. alpinus), whereas bull trout and white-spotted char are sister taxa. Each species exhibits diverse life histories with respect to demographic characteristics, trophic ecology, and movement. This diversity appears to be tied to environmental variability (e.g., temperature, habitat connectivity), resource availability (e.g., food), and species interactions. Increasingly, these interactions involve nonnative species including nonnative salmonines and changes in food webs related to establishment of species such as Mysis shrimp in large lakes. As humans expand into the remote and pristine habitats that support these three chars, we encourage proactive consideration of the lessons learned where chars have already declined and internationally-based research and conservation. To what degree are population differences in resource use caused by competition and the occupation of adjacent positions along environmental gradients evidence of competition? Habitat use may be the result of a competitive lottery, or restricted by competition. We tested to what extent population differences in habitat use of two salmonids, cutthroat trout ( Oncorhynchus clarki) and Dolly Varden charr ( Salvelinus malma) were influenced by interspecific competition. We hypothesized that the depth distribution of Dolly Varden charr would be affected by competition from the more littoral and surface-oriented cutthroat trout, and that the depth distribution of cutthroat trout would be little affected by competition from Dolly Varden charr. Sympatric populations of cutthroat trout and Dolly Varden charr were created by reciprocal transfers of previously allopatric populations in two experimental lakes. We found evidence of asymmetric competition, as Dolly Varden charr were displaced from littoral habitats when sympatric with cutthroat trout, whereas cutthroat trout remained unaffected by the presence of Dolly Varden charr. Evolved differences between the species, and differences between experimental lakes, also contributed to population differences in habitat use, but asymmetric competition remained as the main driver of different depth distributions in sympatry. Genetic resemblance of chars Salvelinus alpinus krasheninnikovi ( Salvelinus malma krasheninnikovi) of 35 samples collected in five Kuril Islands—Shumshu, Paramushir, Onekotan, Iturup, and Kunashir—has been studied by the PCR-RAPD method. In the limits of each island, both resident isolates and anadromous forms give strictly supported clusters distinct from samples from the other islands. The samples from five islands form three superclusters: the first from Kunashir and Iturup Islands, the second from Paramushir and Onekotan Islands, and the third from Shumshu Island. The possible reasons for genetic similarity of resident and anadromous forms of Dolly Varden chars inhabiting reservoirs of a definite island are considered (the founder effect, homing, limited migration). To study the effect of isolation on the genetic variation of mDNA, Salvelinus malma krascheninnikovi and S. malma malma nonanadromous populations from different model ecosystems were examined by RFLI-PCR. The haplotype and nucleotide diversities of mtDNA were significantly decreased in three isolated populations as a result of random gene drift. The genealogy of mtDNA haplotypes was reconstructed. Phylogenetic analysis of charrs of the genus Salvelinus was performed using PCR-RFLP analysis of mitochondrial DNA (mtDNA). Three fragments of mtDNA (ND1/ND2, ND5/ND6 and Cyt b/D-loop) were amplified by PCR and examined for restriction site variation using 13 restriction endonucleases. Analysis of 313 sites of over 7672 bp of charr mtDNA was conducted. Six phylogenetic lineages of the mtDNA were revealed, corresponding to six separate charr taxa: Salvelinus leucomaenis Pallas; Salvelinus levanidovi Chereshnev, Skopetz & Gudkov; Salvelinus taranetzi Kaganovsky; Salvelinus malma krascheninnikovi Taranetz; Salvelinus malma malma Walbaum and Salvelinus alpinus alpinus. The Levanidov ( S. levanidovi) and white-spotted ( S. leucomaenis) charrs represented the most divergent lineages. These are probably closest to a common ancestor of the genus. Salvelinus taranetzi was the next divergent lineage. Salvelinus malma krascheninnikovi was placed between S. leucomaenis and S. taranetzi. Salvelinus malma malma and S. a. alpinus were sister taxa, and were on the last branch to diverge. The divergence between S. m. malma and S. a. alpinus inferred from mtDNA nucleotide sequences was 1.1%; between S. m. malma and S. taranetzi 2.8%; between S. m. malma and S. m. krascheninnikovi 4%; between S. taranetzi, S. m. krascheninnikovi and S. a. alpinus 2.8%. Salvelinus levanidovi and S. leucomaenis were almost equally remote from the other charrs by some 9% and 7%, respectively. The extent of genetic differences between the northern S. m. malma populations and the European S. a. alpinus populations indicates that they diverged recently. The existence of many putative charr species with different divergence times in northeastern Asia suggests that this region may be one of the centres of speciation of the genus Salvelinus. In 41 individuals of South Asian Dolly Varden char Salvelinus curilus, nucleotide sequences of tRNA-Pro gene fragment (27 bp) and mtDNA control region (483–484 bp) were analyzed. The fish were collected in 20 localities covering virtually the whole range of the species: Kuril Islands, Sakhalin Island, and Primorye. In addition, six individuals of three other char species ( S. albus, S. malma, and S. leucomaenis), which are closely related to S. curilus and inhabit the Russian Far East, were examined. In all, we detected 12 different variants of mtDNA haplotypes that formed three distinct groups differing in 14–20 nucleotide positions. The first group consisted of six haplotypes found in S. curilus in Kuril Islands, Sakhalin, and Primorye (mtDNA phylogroup OKHOTSKIA). The second group comprised four haplotypes representing the mtDNA phylogroup BERING, which had been described earlier (Brunner et al, 2001); they were found in S. curilus in Kuril Islands and Sakhalin, as well as in S. albus and S. malma in Kamchatka and northern Kurils. The third group included two haplotypes detected in S. leucomaenis. The existence of two mtDNA lineages (OKHOTSKIA and BERING) in S. curilus from Kurils and Sakhalin was explained by hybridization and mtDNA transfer from S. malma to S. curilus. The absence of the BERING haplotypes in S. curilus from Primorye water reservoirs is related to the physical isolation of the Sea of Okhotsk and Sea of Japan basins in past epochs. On the basis of comparing phylogenetic trees, constructed from the data on allozyme and mtDNA variation, we suggest that in this case, a mediated transfer of mtDNA in Alpinoid chars → S. malma → S. curilus chain could occur. Isolation by distance is usually tested by the correlation of genetic and geographic distances separating all pairwise populations’ combinations. However, this method can be significantly biased by only a few highly diverged populations and lose the information of individual population. To detect outlier populations and investigate the relative strengths of gene flow and genetic drift for each population, we propose a decomposed pairwise regression analysis. This analysis was applied to the well-described one-dimensional stepping-stone system of stream-dwelling Dolly Varden charr ( Salvelinus malma). When genetic and geographic distances were plotted for all pairs of 17 tributary populations, the correlation was significant but weak ( r2 = 0.184). Seven outlier populations were determined based on the systematic bias of the regression residuals, followed by Akaike's information criteria. The best model, 10 populations included, showed a strong pattern of isolation by distance ( r2 = 0.758), suggesting equilibrium between gene flow and genetic drift in these populations. Each outlier population was also analysed by plotting pairwise genetic and geographic distances against the 10 nonoutlier populations, and categorized into one of the three patterns: strong genetic drift, genetic drift with a limited gene flow and a high level of gene flow. These classifications were generally consistent with a priori predictions for each population (physical barrier, population size, anthropogenic impacts). Combined the genetic analysis with field observations, Dolly Varden in this river appeared to form a mainland-island or source-sink metapopulation structure. Intra- and interspecific phylogenetic analysis of Dolly Varden Salvelinus malma and white-spotted charr Salvelinus leucomaenis throughout Hokkaido Island was conducted using nucleotide sequences of the mitochondrial DNA control region and three nuclear DNA markers [ribosomal DNA (ITS-1), microsatellite ( u-85), SINE ( Fok-223)]. A total of 19 mtDNA haplotypes from 271 individuals of Dolly Varden, nine haplotypes from 134 white-spotted charr were defined, and identified two well-supported monophyletic clades for each species. Two haplotypes of Dolly Varden, however, were clustered together in the white-spotted charr clade. Those haplotypes were found only from several rivers in the Shiretoko Peninsula, and a neighbouring river of that region. Analyses of ribosomal DNA and Fok-223 loci revealed that both species are characterized by having completely species-specific diagnostic sequence and fragment patterns. Dolly Varden of the Shiretoko populations are typically allopatric in distribution where white-spotted charr do not currently occur. In addition, incongruence in genetic relationships between mtDNA and nuclear DNA markers give strong evidence of historical mtDNA introgression between Dolly Varden and white-spotted charr. Some white-spotted charr diagnostic alleles in a nuclear microsatellite locus ( u-85) were found in some Shiretoko Dolly Varden populations, suggesting that introgressive hybridization might have also occurred in relatively recent contact and potentially ongoing evolutionary event.
