Background: The quest for candidate probiotics and prebiotics to develop novel synbiotics for sustainable and profitable fish farming remains a major focus for various stakeholders. In this study, we examined the effects of combining two fungal probiotics, Saccharomyces cerevisiae and Aspergillus niger with extracts of Jerusalem artichoke and white button mushroom to develop a synbiotic formulation to improve the growth and health status of zebrafish (Danio rerio). An initial in vitro study determined the most effective synbiotic combination, which was then tested in a 60-day in vivo nutritional trial using zebrafish (80 ± 1.0 mg) as a model animal. Four experimental diets were prepared: a control diet (basal diet), a prebiotic diet with 100% selected mushroom extract, a probiotic diet with 107 CFU of S. cerevisiae/g of diet, and a synbiotic diet with 107 CFU of S. cerevisiae/g of diet and 100% mushroom extract. As readouts, growth performance, survival, digestive enzyme activity and innate immune responses were evaluated. Results: In vitro results showed that the S. cerevisiae cultured in a medium containing 100% mushroom extract exhibited the maximum specific growth rate and shortest doubling time. In the in vivo test with zebrafish, feeding them with a synbiotic diet, developed with S. cerevisiae and mushroom extract, led to a significant improvement in the growth performance of zebrafish (P < 0.05). The group of zebrafish fed with the synbiotic diet showed significantly higher levels of digestive enzyme activity and immune responses compared to the control group (P < 0.05). Conclusion: Taken together, these results indicated that the combination of S. cerevisiae and mushroom extract forms an effective synbiotic, capable of enhancing growth performance and immune response in zebrafish. AbstractObjectiveMethodsResultsConclusionGastrointestinal illnesses have been reported in relation to low disaccharidase activity, yet both the prevalence of disaccharidase deficiency and its association with gastrointestinal symptoms and irritable bowel syndrome (IBS) are largely unknown. We aimed to determine the association between low activity of disaccharidase enzymes on gastrointestinal symptoms and presence of IBS.Patients referred for gastroscopic examination due to gastrointestinal complaints were consecutively included. A pinch biopsy was taken from the distal part of duodenum, and disaccharidase activity was measured using the Dahlqvist method. Gastrointestinal symptom severity was measured using IBS-Symptom Severity Score (IBS-SSS).A total of 40 patients were included. Disaccharidase deficiency was detected in 24 patients (60%). Half of the patients (n = 21) had IBS according to Rome IV criteria. A majority (75%) of all patients reported moderate to severe gastrointestinal symptoms. Moderate to severe gastrointestinal symptoms were reported by 16 patients (67%) with disaccharidase deficiency and in 14 patients (88%) with normal disaccharidase activity. Lactase deficiency was detected in 22 patients (55%), maltase deficiency in 11 patients (28%), sucrase deficiency in 9 patients (23%), isomaltase deficiency in 13 patients (33%) and glucoamylase deficiency in 12 patients (30%). The activity of all enzymes was reduced in 8 patients (20%). Degree of disaccharidase deficiency was not associated with either the severity of gastrointestinal symptoms or the diagnosis of IBS. Enzymes levels were not associated with gastrointestinal symptom scores.Our findings did not reveal any association between biochemically measured disaccharidase deficiency and gastrointestinal symptoms or the presence of IBS. Insect's resilience to adverse conditions poses a significant challenge for integrated pest control. This has resulted in huge economic losses to agriculture and forestry production as well as a range of severe ecological issues. As a physiological mechanism of insects, digestive physiology plays an important role in the process of adaptation to stress factors. However, there has been no systematic review of what stresses insects can adapt to through digestive physiology and how digestive physiology is involved in insect adaptation to stresses. In this review, the potential link between digestive physiology and adaptation of insects to biotic and abiotic stresses, including plant defense mechanisms, chemical insecticides, and entomopathogenic microorganisms, is analyzed. We point to that digestive physiology composed of digestive enzymes and gut microbial communities is an important strategy for insects to resist plant physical defense (e.g., hemicellulose, pectin, and microfibers), chemical defense (e.g., azadirachtin, diterpenoid acids, and phenolic glycosides), chemical insecticide stress, and entomopathogenic microorganism infection. In addition, the primary function of the digestive physiology in insects is to ensure energy supply during biotic and abiotic stress, assist in the metabolism of exogenous toxins (e.g., anti-insect proteins, primary metabolites, secondary metabolites, and insecticides), and improve their innate immunity against entomopathogenic microorganisms. Simple Summary: Due to the underdeveloped digestive tracts of larval fish, predigested fish protein has been included in feeds to help increase dietary protein utilization. The predigested protein has been produced from various protein sources and broken down using different methods. This has ultimately led to variable results when included in larval diets, and there is still a lack of an optimal dry diet for larval fish. This study tested the use of adult Walleye muscle as a protein source to provide the optimal dietary amino acid composition for same-species larvae. The muscle was also broken down with enzymes from the adult Walleye gut to predigest protein in a way that mimics the digestion of live Walleye. This protein source was tested against protein produced from muscle and/or enzymes from Nile Tilapia, as a non-species-specific source. The dietary inclusion of predigested protein produced from same-species muscle and digestive enzymes led to increased growth and amino acid absorption in larval Walleye. However, reduced survival in fish fed with the predigested protein suggests that further research is required to improve the pellet stability of the larval diets and reduce the loss of nutrients to the water. Fish protein hydrolysates used in larval diets have been prepared from a variety of fish species, with different enzymes used to hydrolyze the protein. This study's objectives were to determine the effect of the dietary inclusion of fish muscle hydrolysates obtained from species-specific muscle/enzymes—versus hydrolysates produced from muscle/enzymes of a different species—on the growth performance, survival, skeletal development, intestinal peptide uptake, and muscle-free amino acid (FAA) composition of larval Walleye (Sander vitreus). Eight protein products were obtained for this study, comprising an unhydrolyzed and hydrolyzed product from each combination of muscle/enzymes from Walleye and Nile tilapia (Oreochromis niloticus). Four diets were produced, and the dietary protein was provided in a 50/50 ratio of unhydrolyzed and hydrolyzed protein from the respective muscle/enzyme combination. Four groups were fed one of the corresponding formulated diets, and two groups of larvae, fed a commercial starter diet and Artemia, respectively, served as reference groups. Larval Walleye fed the diet containing protein produced with the species-specific muscle and enzymes had a significantly higher weight after the study—30% higher than any other group. A significant interaction effect between muscle and enzyme sources on the growth of Walleye larvae was observed. The species-specific combination also led to a significant increase in postprandial FAA and indispensable amino acid concentrations in muscle. No significant differences were observed between the hydrolysate-fed groups in survival, deformity occurrence, or peptide uptake. Each hydrolysate-based diet significantly reduced skeletal deformities and survival compared to the commercial diet. The results of this study suggest that species-specific muscles and enzymes produce a more optimal dietary protein source for larval fish than non-species-specific products. Further research should focus on improving the physical properties of the formulated diets to reduce possible leaching of hydrolyzed protein and improve the survival of fish larvae. The present study assessed the growth and fish performance of juvenile red sea bream fed four isonitrogenous (46–48%) and isolipidic (15%) diets containing 45% fishmeal (control) and 15%, 30%, and 45% defatted black soldier fly (BSF) Hermetia illucens meal for 67 days. Subsequent analyses showed that there were no significant differences in the growth parameters, nutrient digestibility, and body composition of fish among all treatments. Although no significant difference was observed in fish growth, low growth was observed when the fish meal was totally replaced with the BSF meal. Higher alpha diversity of intestinal microbiota was observed in fish fed the BSF meal-based diet compared to that of the control. In the treatment groups fed the BSF meal, elevations in the presence of microbial genera that produce digestive enzymes, such as chitinase (Nocardia, Vibrio, Paenibacillus, Photobacterium), lipase (Paenibacillus, Photobacterium), and trypsin-like serine protease (Bacillus, Paenibacillus) and vitamin B group [biotin (Vibrio, Paenibacillus, Bacillus), cobalamin (Pseudomonas, Bacillus, Propionibacterium), and pyridoxin (Photobacterium)], were observed. We also found an increasing trend of bacteria producing antioxidative enzymes, such as superoxide dismutase and catalase (Nocardia, Pseudomonas, Photobacterium) in the BSF groups. These results suggested that the BSF meal could replace 67% of fishmeal without causing a negative impact on growth and lead to beneficial effects on the intestinal microbiota of the red sea bream. The physiological impacts of polysaccharides and lipids in seaweed on human health are becoming clearer, but the behavior of the protein components, especially lectins, after ingestion remains poorly understood. In this study, we examined the resistance of edible red algae-derived lectins to digestive enzymes. We found that a lectin extracted from Meristotheca papulosa (MPL-1), belonging to the Jacalin-related lectin family, was relatively stable when subjected to both peptic and tryptic digestion and retained its hemagglutination activity after 24 h of digestion. The activity of MPL-1 was also maintained without a large change for 24 h following exposure to enzymes such as papain, Actinase E, and proteinase K, suggesting that MPL-1 possesses a strong resistant property against proteolytic digestion. We examined the anti-proliferation activity of the MPL fraction from the algal body against HT-29, a human colon cancer-derived cell line, and found that it showed a strong inhibitory activity with a half-maximal inhibitory concentration of 4.5 μg/mL. This activity was nullified in the presence of yeast mannan, an inhibitory sugar compound of lectin, demonstrating that MPL expressed its activity through binding to the glycan moieties on HT-29. The characterization of digestive enzymes presents in fish and their potential biotechnological uses is a welldeveloped and studied field. However, there has not been a systematic review that analyzes the state of knowledge of these enzymes at a global level. Therefore, a systematic literature search on three platforms was carried out to review and analyze existing knowledge about digestive enzymes of marine fishes from fisheries and aquaculture and their potential application in industrial processes. Using the PRISMA method for selecting journal manuscripts, we found 112 scientific articles published between 1984 and 2020 studying different digestive enzymes from 87 fish species. Most studies were carried out in Tunisia and Mexico and only 6 articles were published in South American countries. The most studied digestive enzymes were alkaline proteases, mainly trypsin, and the proposed uses for these enzymes were mainly as additives in commercial detergents. There is a vacancy in the characterization of other digestive enzymes as pepsins, lipases and amylases and the study of species that are distributed in the Southwest Atlantic Ocean. The digestive properties of starch are crucial in determining postprandial glycaemic excursions. Genistein, an active phytoestrogen, has the potential to influence starch digestion rates. We investigated the way genistein affected the digestive properties of starch in vitro. We performed enzyme kinetics, fluorescence spectroscopy, molecular docking, and molecular dynamics (MD) simulations for analysing the inhibitory properties of genistein on starch digestive enzymes as well as clarifying relevant mechanism of action. Our findings demonstrated that, following the addition of 10% genistein, the contents of slowly digestible and resistant starches increased by 30.34% and 7.18%, respectively. Genistein inhibited α-amylase and α-glucosidase, with half maximal inhibitory concentrations of 0.69 ± 0.06 and 0.11 ± 0.04 mg/mL, respectively. Genistein exhibits a reversible and non-competitive inhibiting effect on α-amylase, while its inhibition on α-glucosidase is a reversible mixed manner type. Fluorescence spectroscopy indicated that the presence of genistein caused declining fluorescence intensity of the two digestive enzymes. Molecular docking and MD simulations showed that genistein binds spontaneously to α-amylase via hydrogen bonds, hydrophobic interactions, and π-stacking, whereas it binds with α-glucosidase via hydrogen bonds and hydrophobic interactions. Titanium dioxide nanoparticles (TiO2) are released into the natural waters in different ways and cause toxicity to aquatic life. Although various studies have reported the toxicity of nanoparticles for aquatic animals, there is limited information regarding the effects of environmental factors, especially salinity, on their toxicity. Simultaneous effects of TiNPs and water salinity were studied in the Pacific oyster, Crassostrea gigas. Oysters were exposed to sub-lethal concentrations of TiNPs (low concentration: 50 µg/l, middle concentration: 500 µg/l, high concentration: 5000 µg/l) at different water salinities (Extremely low salinity: 5 g/l, low salinity: 15 g/l, middle salinity: 25 g/l, high salinity (normal salinity): 35 g/l) for 96 h. After nano-TiO2 exposure, the immune components in hemolymph (total hemocyte concentration [THC], phagocytosis), antioxidant enzyme activities in digestive gland (superoxide dismutase, catalase, glutathione peroxidase, glutathione S-transferase), oxidation stress index (malondialdehyde) in gills and the expression of immune-related genes (Vav-3 protein, cofilin, Galectin 4, pro-Cathepsin L, Rho protein, Ficolin, LPS/-1,3-glucan protein, Lysozyme, Defensin) in hemocytes were measured. The THC, phagocytosis, antioxidant enzyme activities, and immune-related gene expressions mostly decreased at salinities of 5 and 15 g/l in response to 500–5000 µg/l nano-TiO2 compared to non-exposed oysters and 50 µg/l nano-TiO2 group. An increase in water salinity up to 35 g/l mitigated the depressing effects of the nano-TiO2 on the immune and antioxidant components of the oysters. Exposure of oysters to 500–5000 µg/l nano-TiO2-induced oxidative stress at salinities of 5 and 15, since malondialdehyde levels were higher in these treatments compared to control and those in 50 µg/l nano-TiO2 group. The increase of salinity up to 35 g/l ameliorated the TiO2-induced oxidative stress. The results of the present study showed that the toxic impacts of nano-TiO2 can be ameliorated in the Pacific oyster with increase in water salinity, as the immune components, antioxidant enzyme activities recovered and oxidative stress mitigated when the salinity increased up to 35 g/l. The purpose of this study was to investigate the adaptive strategies and compensatory growth mechanisms of juvenile Epinephelus coioides under starvation stress. The fish with an initial body weight of 8.88 ± 0.06 g were starved for 0 day (F0, control), 3 days (F3), 6 days (F6), 9 days (F9), 12 days (F12), and 15 days (F15), followed by feeding periods of 30 days, 27 days, 24 days, 21 days, 18 days, and 15 days, respectively. The results are shown as follows: (1) E. coioides could achieve complete or partial compensatory growth by improving both feeding rate and feed conversion efficiency. (2) The viscerosomatic index and the hepatosomatic index of the test fish decreased linearly with the duration of starvation (P < 0.05), while the condition factor did not significantly decrease (P > 0.05). At the end of feeding, all the physical body indicators of the test fish recovered or exceeded the control levels. (3) The lipid and glycogen contents of the test fish decreased continuously throughout the entire starvation process, with noticeable protein consumption occurring in the later stages of starvation (P < 0.05). At the end of the feeding period, apart from a significant reduction in crude lipid contents in the F12 and F15 groups compared to the control group (P < 0.05), the biochemical composition of the test fish was restored or even surpassed that of the control group. (4) The activity of digestive enzymes could be stimulated by starvation, enabling the efficient utilization of energy storage substances. In conclusion, it is advisable to limit the period of starvation for juvenile E. coioides to 12 days, which means that the ratio of starvation period to feeding period should not exceed two‐thirds. A variety of therapeutic possibilities have emerged for skillfully regulating protein function or conformation through intermolecular interaction modulation to rectify abnormal biochemical reactions in diseases. Herein, a devised strategy of enzyme coordinators has been employed to alleviate postoperative pancreatic fistula (POPF), which is characterized by the leakage of digestive enzymes including trypsin, chymotrypsin, and lipase. The development of a dextrorotary (D)-peptide supramolecular gel (CP-CNDS) under this notion showcases its propensity for forming gels driven by intermolecular interaction. Upon POPF, CP-CNDS not only captures enzymes from solution into hydrogel, but also effectively entraps them within the internal gel, preventing their exchange with counterparts in the external milieu. As a result, CP-CNDS completely suppresses the activity of digestive enzymes, effectively alleviating POPF. Remarkably, rats with POPF treated with CP-CNDS not only survived but also made a recovery within a mere 3-day period, while mock-treated POPF rats had a survival rate of less than 5 days when experiencing postoperative pancreatic fistula, leak or abscess. Collectively, the reported CP-CNDS provides promising avenues for preventing and treating POPF, while exemplifying precision medicine-guided regulation of protein activity that effectively targets specific pathogenic molecules across multiple diseases. Cadmium could induce various degrees of harm to aquatic organisms. A 30-day feeding trial was conducted to investigate the effects of cadmium on growth, muscle composition, digestive enzyme activity, gene expression of antioxidants and lipid metabolism in juvenile genetic improvement of farmed tilapia (GIFT, Oreochromis niloticus, Initial weight: 21.36 ± 0.24 g). Four cadmium concentrations of aquaculture water were designed: 0, 0.2, 0.4, and 0.6 mg/L Cd2+. The main results are as follows: Compared with the control group (0 mg/L Cd2+), the weight gain rate (WGR), specific growth rate (SGR), daily growth index (DGI), and spleen index (SI) of juvenile GIFT under cadmium stress were significantly decreased (p< 0.05). The contents of crude protein and crude lipid in muscle were significantly decreased (p< 0.05), and the ash was significantly increased (p< 0.05). The activities of trypsin, lipase, and -amylase in the intestinal were significantly decreased (p< 0.05). The relative expression levels of carnitine palmityl transferase 1 (cpt-1), peroxisome proliferator-activated receptor (ppar ), ppar, hormone-sensitive lipase (hsl), lipoprotein lipase (lpl), malate dehydrogenase (mdh), leptin (lep), fatty acid synthetase (fas), cholesterol response element binding protein 1 (srebp1), squalene cyclooxygenase (sqle), and stearoyl-CoA desaturase (scd) genes in liver were significantly decreased (p< 0.05). The relative expression levels of catalase (cat), superoxide dismutase (sod), glutathione S-transferase (gst), and glutathione peroxidase (gsh-px) genes in the liver were significantly decreased (p< 0.05). Introduction: Calcein was used to develop a shell marking method for Potamocorbula laevis. Methods: The suitable conditions for markingwere investigated, includingmarking concentration, immersion time, and water temperature. The impacts and feasibility of the marking method were assessed based on the survival rate of P. laevis, the success rate of fluorescence marking, marking quality, and alterations in activities of antioxidant enzymes in the digestive gland of the experimental bivalves. Two concentrations of calcein (20 and 50 mg/L) were used and the immersion time included 1 and 2 h, respectively. The experimentwas performed in two rounds, with water temperatures of 12.84 ± 0.09 and 24.18 ± 0.04 °C, respectively. Results and discussion: The results indicated that calcein did not significantly impact the survival of P. laevis after 7 d of recovery. The catalase activity and malondialdehyde content in low temperature-marked P. laevis showed significant decreases, and the relative abundances of certain fatty acids also exhibited significant changes within 2 h post exposure to 20 mg/L of calcein. However, these indicators returned to normal levels within 7 d. The marking impact of calcein was proportional to the calcein concentration and immersion time. Higher temperature generated a negative impact on the marking effect of 20 mg/L of calcein, while no obvious impacts were observed for 50 mg/L of calcein. The marking success rates and the recapture rates of P. laevis for in situ tests in the two experimental groups were both 100% and 4.44 ± 1.29% after one month. Also, the recapture marking rates and the marking good rates of the recaptured individuals were both 100%. There were no significant differences between these parameters for 50 and 75 mg/L of calcein. Given the cost and safety of labeling, a strategy in terms of an immersion in 50 mg/L of calcein for 2 h could be considered as an effective in situ labeling scheme for P. laevis. In conclusion, calcein can be employed as a marking method for P. laevis. These findings could be potentially beneficial for development of in situ labeling technology, proliferation as well as release of shellfish in tidal flats and resource conservation. Anastomotic leakage (AL) is the leaking of non‐sterile gastrointestinal contents into a patient's abdominal cavity. AL is one of the most dreaded complications following gastrointestinal surgery, with mortality rates reaching up to 27%. The current diagnostic methods for anastomotic leaks are limited in sensitivity and specificity. Since the timing of detection directly impacts patient outcomes, developing new, fast, and simple methods for early leak detection is crucial. Here, a naked eye‐readable, electronic‐free macromolecular network drain fluid sensor is introduced for continuous monitoring and early detection of AL at the patient's bedside. The sensor array comprises three different macromolecular network sensing elements, each tailored for selectivity toward the three major digestive enzymes found in the drainage fluid during a developing AL. Upon digestion of the macromolecular network structure by the respective digestive enzymes, the sensor produces an optical shift discernible to the naked eye. The diagnostic efficacy and clinical applicability of these sensors are demonstrated using clinical samples from 32 patients, yielding a Receiver Operating Characteristic Area Under the Curve (ROC AUC) of 1.0. Large-mouth bass originally lived in rivers and lakes and now is often raised in recirculating aquaculture systems (RAS). However, the shortcoming of RAS is that there is very limited space for swimming fish in a high-density culture environment, thus leading to a lack of exercise. To investigate the effects of exercise training on growth performance, digestive enzymes, stress, and antioxidant capacity of large-mouth bass in a RAS, three training water flow velocities with three replicates were used in the present study: low water velocity: 0.5 body length per second (bl/s) (LV); medium water velocity: 1 bl/s (MV); and high water velocity: 1.5 bl/s (HV). 270 large-mouth bass (average initial body weight 47.51 ± 1.44 g, initial body length 12.71 ± 0.06 cm) were randomly divided into nine tanks (30 fish per tank). After 27 days experiment, the results showed that the growth performance of large-mouth bass, including feed intake, weight gain rate, and specific growth rate, in the HV group was significantly higher compared to the other groups. On the physiological level, the lipase and amylase activities were significantly increased in the HV group compared to the other groups. For the stress response parameters, the cortisol, glucose and lactate concentrations in the HV group were significantly increased, which may indicate higher stress in the HV group. Regarding the antioxidant enzyme activities, the activities of catalase (CAT) and glutathione peroxidase (GSH-PX) in the LV group were significantly higher than those in the HV group, which indicated that fish suffered higher oxidative stress in the LV group. Regarding the immune-related parameters, LZM and ACP activities had no significant differences among the groups, while AKP activities in the MV and HV groups were significantly higher than that in the LV group. Overall, the results demonstrated that exercise training had significant effects on the growth, digestion, stress, and immune response of large-mouth bass. It is suggested that exercise training with 1.5 bl/s can enhance fish growth, however, it also presents a potential risk of inducing stress in fish. Rhyzopertha dominica (F.) is considered as a major stored product pest. Conventional insecticides induce adverse effects on the environment. Plant derivative products constitute a promising alternative for pest control. The purpose of our research was to identify the chemical profile of essential oil (EO) extracted from Schinus molle L. (Anacardiaceae) and to evaluate the fumigant and the contact toxicity, and the repellent activity of this oil on adults of Rhyzopertha dominica F. (Coleoptera), a major pest in products that are stored around the world. The effects of this oil were examined on biochemical profile (energy reserves and Proteins), selective biomarkers like acetylcholinesterase (AChE), glutathione S-transferases (GST) and glutathione (GSH), and nucleic acids (DNA and RNA). Digestive enzymes were also investigated. The major chemical compounds identified by the GC-MS were α-phellandrene (16.613%), ß-phellandrene (12.104%), elemol (9.028%) and β-eudesmol (5.872%). This EO exhibited an insecticidal and repellent activity as a function of the exposure time and concentrations. Characteristics of the digestive enzymes of the fifth instar larvae of the beet moth were determined to achieve a superior understanding of the digestive physiology of the pest. After dissecting the larval digestive tube, optimum acidity for the activity of α-amylase and α-glucosidase was obtained 8–9, and 8 for β-glucosidase, respectively. The optimum temperatures were 35 °C and 35–40 °C for α-amylase activity and for α- and β-glucosidase, respectively. The zymogram displayed the existence of three α-amylase isoforms in the digestive tract of this insect. The maximum activity of general protease was determined in the digestive tract of this pest using azocasein substrate in the pH range of 10–11. Furthermore, the optimum temperature was 35 °C for general protease. The metal ions including iron, magnesium, manganese, zinc, and copper at 5 and 10mM concentrations had a significant influence on the activity of α-amylase, α- and β-glucosidase, trypsin, chymotrypsin, and elastase enzymes. Digestive midgut protease activity in fifth instar larvae (L5) of Helicoverpa armigera was investigated under controlled conditions, where, an artificial diet containing different concentrations (0.005 µg/g - 0.3 µg/g) of Cry1Ac (Crystalline ä-endotoxin from class A, subclass c) toxin was fed. Here, the Bacillus thuringiensis (Bt) cotton seeds were used as Cry1Ac toxin source that was determined using Envirologix ELISA (Enzyme Linked Immunosorbent Assay) kit. The results showed an increase in the general midgut proteases as well as specific proteases activity linked to enzymes such as trypsin-, chymotrypsin-, elastase-, and leucine aminopeptidase-like with the increasing toxin content of the diet. However, carboxypeptidase-A and carboxypeptidase-B protease activity remained unaltered as a consequence of the incorporation of Cry1Ac toxin in the diet, which is also reported to be absent in midgut of H. armigera as evident in the control group. Clostridium butyricum (CB) has received much attention as a probiotic; however, few studies have focused on its effects on liver health. Here, we studied the influence of CB on the liver health of spotted sea bass through transcriptomics and metabolomics studies and preliminarily explored its molecular mechanisms. This study showed that CB significantly reduced hepatic aspartate aminotransferase (AST) activity and increased alkaline phosphatase (AKP) and acid phosphatase (ACP) activity (P < 0.05). CB has demonstrated significant effects in strengthening liver immunity and can increase hepatic amylase and trypsin activities and promote hepatic catabolism of carbohydrates and amino acids. Integration of the liver transcriptomics and metabolomics showed altered transcript levels of mainly gluconeogenic, lipogenic, and amino acid metabolic pathways. It regulated the abundance of metabolic biomarkers such as arachidonate, crotonyl-CoA, and D-glucose 1-phosphate. Protein hydrolysates as a feed supplement have been reported to enhance the growth of beneficial gut bacteria and improve the host's animal intestinal balance. Thus, this study evaluates the prebiotic effects of hydrolyzed squid by-products in black tiger shrimp, Penaeus monodon, diet. A 50-day feeding trial was conducted following a 4 × 2 factorial experiment in a completely randomized design. The juvenile P. monodon were fed plant-based diet (without fishmeal) supplemented with graded levels of 0, 0.5, 1.0, and 2.0% hydrolyzed squid by-products, either with or without Bacillus subtilis BF12. A control group fed a fish meal-based diet was also included. The shrimp growth, survival, nutrient retention, digestive enzyme activities, immune-related and gut pro-inflammatory gene expression, and intestine histomorphology were the evaluated biological responses. Feeding shrimp with plant-based diet containing 0.5% hydrolyzed squid by-products and B. subtilis BF12 supplement improved growth, enhanced feed conversion, improved nutrient retentions, upregulated the immune-related genes, and downregulated the gut pro-inflammatory genes. Enhanced digestive enzyme activities were observed in treatment groups with higher hydrolyzed squid by-product supplementation. Moreover, hydrolysate supplementation and probiotic application increased the shrimp's intestinal villi height and promoted the growth B. subtilis BF12 probiotics in the gut. Collectively, the present work suggests that hydrolyzed squid by-product protein supplementation at 0.5% and B. subtilis BF12 probiotics can promote growth, enhance the immunological responses, and improve the gut health of juvenile P. monodon fed a plant-based diet. In this study, we investigated the effects of gallic tannic acid (GTA) supplementation on the growth performance, intestinal digestive enzymes, morphology and microbial composition of common carp (Cyprinus carpio). The basal diet (CK) was supplemented with 5 g/kg (TL), 10 g/kg (TM) and 20 g/kg (TH) for 6 weeks. Compared with the CK group, adding GTA (10 g/kg) significantly increased the weight gain rate (WG%), specific growth rate (SGR%) and body condition factor ratio (CF) of common carp (p < 0.05) and reduced the fish feed conversion ratio (FCR) (p < 0.05). GTA supplementation significantly increased digestive enzyme activity, including amylase (Ams), protease (Prt) and lipase (Lps) (p < 0.05). Supplementing GTA can also enhance the innate immunity of common carp by reducing the activities of serum alanine aminotransferase (Alt), aspartate aminotransferase (Ast), malondialdehyde (Mda), and increasing catalase (Cat), total superoxide dismutase (Khorshidi, Paknejad, Sodagar, Hajimoradloo, & Shekarabi) levels (p < 0.05). Furthermore, the intestinal barrier is improved. Increases in villus length, goblet cell number, and occludin a (oclna) gene expression levels were observed. RT‒PCR results showed that GTA increasedthe expression levels of insulin-like growth factor 1 (igf1) and interleukin 4 (il4) and reduce the expression levels interleukin 6 (il6) mRNA in the fish intestine (p < 0.05). Adding GTA to the diet significantly increased the abundance of beneficial bacteria Gemmobacter, Shinella and Actinobacteriota_unclassified, indirectly increased the activity of digestive enzymes and promoted the growth of fish. KEGG analysis shows that 10 g/kg of GTA can significantly enhance the lipid metabolism, carbohydrate metabolism, and amino acid metabolism pathways of the intestinal microbiota in fish (p < 0.05), thereby positively affecting the enhancement of intestinal digestive enzymes. These results indicated that adding GTA to feed can effectively improve the growth performance of common carp by promoting intestinal health. Arachidonic acid (AA) is an essential fatty acid that has vital influences on early development in marine fish larvae. In this regard, a 30-day research was run to examine the effects of live food enrichment with graded levels of AA on some physiological responses of Acanthopagrus latus larvae. Four experimental emulsions containing four levels of AA were used to enrich live foods (rotifer and Artemia), including very low (6%, N), low (12%, L), medium (24%, M), and high (37.8%, H) AA levels. Newly hatched larvae were distributed into twelve 250-L cylindrical tanks with initial density 15,000 larvae in each tank supplied with seawater (23 ± 1 °C; 40.0 ± 1.0 g L−1). Nannochloropsis oculata was used in a green water system and live foods were offered to larvae two times daily. Larvae in the H group had lower total length (7.24 ± 0.6 mm) than those in other treatments. The highest survival rate after air exposure and osmotic stress challenges was in the H group. The level of n-6 polyunsaturated fatty acids, mainly AA, was enhanced in the larval whole body with increasing AA level in the emulsion. The highest and lowest alkaline phosphatase activities were in the H and N groups, respectively. The aminopeptidase N and leucine-alanine peptidase activities in the N group were lower than those in the other treatments. The trypsin activity gradually increased with the increment of AA in the emulsions. The highest and lowest chymotrypsin, amylase, and pepsin activities were in H and N groups, respectively. The highest and lowest phospholipase A2 activity was in the M and N groups, respectively. The activity of catalase in the whole body of the M group was greater than that in the rest of the treatments. The highest and lowest superoxide dismutase activities were in M and N groups, respectively. The activity of glutathione reductase in the L and M groups was higher than that in the rest. Total antioxidant capacity in the whole body of larvae in the N group was lower than that in the other treatments and the H group showed the highest value. The whole body malondialdehyde level in the L group was lower than that in N and H groups and M showed intermediate value. Based on the findings of the present study, moderate levels of AA suggested for the enrichment of live foods in A. latus by considering n-3 long-chain polyunsaturated fatty acid content in the enrichment solution. As a euryhaline fish species that inhabits estuarine and coastal regions, the spotbanded scat (Selenotoca multifasciata) experiences growth influences during its larval stage due to variations in salinity. Here, we evaluated salinity required by early-stage spotbanded scat juveniles to achieve the highest growth performance, digestive enzyme activity, survival, and antioxidant capacity. We reared spotbanded scat juveniles (all 0.50 ± 0.05 g) in 0–35‰ salinity gradients for 50 days and recorded their survival rate every 10 days. After 50 experimental days, we measured morphological data, stomach and intestinal digestive enzyme activities, and liver antioxidant enzyme activities and malondialdehyde contents. In general, 5–15‰ salinity led to 100% survival. The 5‰ salinity group demonstrated the highest values for the following measures: final wet body weight; weight gain rate; specific growth rate; growth percentage; average daily gain; stomach amylase and lipase specific activities; and intestinal amylase, lipase, trypsin, and pepsin specific activities. However, stomach trypsin and pepsin activities did not demonstrate significant between-group differences (all p > 0.05). The 25‰ salinity group demonstrated the highest liver superoxide dismutase and glutathione peroxidase activities and malondialdehyde content. Finally, the 0‰ salinity group demonstrated the highest liver catalase activity. Thus, spotbanded scat juveniles demonstrate the highest survival rates, growth performance, and digestive enzyme activity at 5‰ salinity and the strongest oxidative stress responses at 25‰ salinity. The present study investigated the effects of a feed additive (FA) containing a probiotic consortium, fructooligosaccharide and yeast extract on growth performance, humoral immune responses, hepatic antioxidant parameters and intestine digestive enzymes, morphology and transcripts in rainbow trout, Oncorhynchus mykiss. The fish were reared for 8 weeks, feeding on diets containing 0 (CNT), 0.3 (0.3FA), 1 (1FA) and 2 (2FA) g/kg FA. The results showed that fish growth parameters were significantly and quadratically related to FA levels and FA treatments had better growth performance than CNT treatment. Intestinal amylase activity significantly increased in 2FA, whereas, intestinal protease activity increased in all FA treatments. Intestinal villus length and muscular layer thickness significantly increased in 0.3FA treatment. Blood leucocyte and lymphocyte counts, plasma lysozyme activity and hepatic glutathione content significantly increased in 0.3FA and 1FA treatments; whereas hepatic malondialdehyde significantly decreased in these treatments. Blood neutrophil and monocyte counts significantly increased in 0.3FA treatment, while plasma alternative complement activity significantly increased in 1FA treatments. Plasma bactericidal activities against Aeromonas hydrophila, Yersinia ruckeri and Streptococcus iniae, and intestinal expression of heat shock protein 70 and beta‐defensin significantly increased in all FA treatments. The abundance of A. hydrophila, Y. ruckeri and S. iniae in fish gut significantly decreased in 0.3FA treatment; these bacteria were absent in the intestines of 1FA and 2FA treatments. The present results suggest that dietary 0.3–1 g/kg of FA can significantly improve growth performance, immune response, intestinal health and hepatic antioxidant capacity in rainbow trout. Simple Summary: Fishmeal is a vital protein in aquafeed; however, its high cost and limited availability pose significant challenges. This study investigates the efficacy of chicken meal, an animal protein with a higher yield and lower cost, as a substitute for fishmeal. After feeding bullfrogs with different feed formulations for eight weeks, it was found that replacing 50% of fishmeal with chicken meal increased the survival rate of bullfrogs, thus enhancing their overall weight. Additionally, this partial substitution improved the antioxidant capacity and intestinal digestive enzyme activity of the bullfrogs. Conversely, a 100% replacement of fishmeal showed no significant effect on survival rate and resulted in reduced weight gain and lower amino acid content in muscle tissues, indicating that complete replacement is unsuitable. In pursuit of sustainable aquaculture, this study was performed to evaluate chicken meal as a substitute for fishmeal in bullfrog diets. Three experimental groups were established: a control group (FM) with 20% fishmeal, a CM50 group with 50% replacement (10% fishmeal), and a CM100 group with 100% replacement (0 fishmeal). Bullfrogs were fed for 56 days. The CM50 group exhibited significant increases in total weight gain and survival rate and a notable decrease in feed coefficient (p < 0.05). However, the CM100 group showed contrary effects. Increasing chicken meal substitution correlated with decreased amino acid content in muscle. Notably, the CM50 group demonstrated enhanced activities of antioxidant enzymes (CAT, T-AOC) and elevated gene expression levels (cat, sod, gst, etc.) in muscle and the intestine (p < 0.05), improved intestinal morphology, enhanced digestive enzyme activities (amylase, lipase), and reduced expression of inflammatory factors (il-1β, il-8, il-17, etc.). Conversely, the CM100 group's indicators regressed to levels similar to or worse than those of the FM group. Therefore, a 50% substitution of fishmeal with chicken meal effectively promoted bullfrog survival, protected the intestines, and enhanced antioxidant capacity, supporting its potential as a fishmeal alternative. The present study's aim was to investigate the effects of different protein level diets on ruminal morphology, fermentation parameters, digestive enzyme activity, microflora and metabolites of Tibetan sheep. Ninety weaned lambs (initial weight of 15.40 ± 0.81 kg, 2 months old) were selected and randomly divided into three groups (six pens/treatment, five rams/pen). Dietary treatments were formulated with 13.03% (high protein, HP), 11.58% (moderate protein, MP) and 10.20% (low protein, LP), respectively. Compared with LP, both papillae length and papillae width were significantly promoted in HP and MP (p < 0.05). The concentrations of ammonia nitrogen, total VFAs, propionic acids and butyric acids in HP were significantly increased compared to those in MP and LP (p < 0.05). The activities of protease and α-amylase in HP were significantly greater than those of LP (p < 0.05). For the ruminal microbial community, higher proportions of phylum Prevotella 1 and Succiniclasticum and genus Rikenellaceae RC9 gut group and Ruminococcus 1 were observed in HP (p < 0.05). A total of 60 differential metabolites (DMs) (28 up, 32 down) between HP and MP; 73 DMs (55 up, 18 down) between HP and LP; and 65 DMs (49 up, 16 down) between MP and LP were identified. Furthermore, four pathways of the biosynthesis of unsaturated fatty acids, tryptophan metabolism, bile secretion and ABC transporters were significantly different (p < 0.05). The abundance of phylum Prevotella 1 was negatively associated with stearic acid and palmitic acid but positively associated with the taurine. The abundance of genus Ruminococcus 1 was negatively associated with stearic acid, oleic acid, erucic acid, Indole-3-acetamide and palmitic acid but positively associated with 6-hydroxymelatonin. Glycoside hydrolases (GHs, also called glycosidases) catalyze the hydrolysis of glycosidic bonds in polysaccharides. Numerous GH genes have been identified from various organisms and are classified into 188 families, abbreviated GH1 to GH188. Enzymes in the GH32 family hydrolyze fructans, which are present in approximately 15% of flowering plants and are widespread across microorganisms. GH32 genes are rarely found in animals, as fructans are not a typical carbohydrate source utilized in animals. Here, we report the discovery of 242 GH32 genes identified in 84 animal species, ranging from nematodes to crabs. Genetic analyses of these genes indicated that the GH32 genes in various animals were derived from different bacteria via multiple, independent horizontal gene transfer events. The GH32 genes in animals appear functional based on the highly conserved catalytic blades and triads in the active center despite the overall low (35–60%) sequence similarities among the predicted proteins. The acquisition of GH32 genes by animals may have a profound impact on sugar metabolism for the recipient organisms. Our results together with previous reports suggest that the acquired GH32 enzymes may not only serve as digestive enzymes, but also may serve as effectors for manipulating host plants, and as metabolic enzymes in the non-digestive tissues of certain animals. Our results provide a foundation for future studies on the significance of horizontally transferred GH32 genes in animals. The information reported here enriches our knowledge of horizontal gene transfer, GH32 functions, and animal–plant interactions, which may result in practical applications. Trypsin inhibitors (TI) in raw soybean grain, mainly represented by the Kunitz trypsin inhibitor protein (KTI), prevent the normal activity of the digestive enzymes trypsin and chymotrypsin in humans and monogastric livestock. The inactivation of TI is achieved through costly and time-consuming heat treatment. Thermal processing also impairs the solubility and availability of the soybean grain protein. Therefore, the genetic elimination of KTI has been proposed as a suitable alternative to heat treatment. The aim of this study was to screen the collection of European soybean cultivars with six genetic markers (one SSR marker and five SNP markers) previously proposed as tightly linked to the KTI3 gene encoding the major Kunitz trypsin inhibitor seed protein of soybean and validate their usability for marker-assisted selection (MAS). The six markers were validated on a subset of 38 cultivars with wide variability in KTI content and in the F2 and F3:5 progenies of two crosses between the known high- and low-KTI cultivars. Three genetic markers (SSR Satt228 and two SNP markers, Gm08_45317135_T/G and Gm08_45541906_A/C) were significantly associated with KTI content in a subset of 38 cultivars. Low-KTI alleles were detected in both low- and high-KTI genotypes and vice versa, high-KTI alleles were found in both high- and low-KTI genotypes, indicating a tight but not perfect association of these markers with the KTI3 gene. The genetic marker SSR Satt228 showed a significant association with KTI content in the F2 progeny, while the SNP markers Gm08_45317135_T/G and Gm08_45541906_A/C allowed significant discrimination between progeny with high- vs. low-KTI progenies in the F3:5 generation. Purpose: The Pediococcus strains belong to the group of lactic acid bacteria (LAB). This study aims to isolate and identify the Pediococcus spp. from mother's milk, and investigate their tolerance to low pH and bile salts, antibacterial activity, attachment to HT-29 cells and effect on cholesterol-lowering and digestive enzymes in mice. Design/methodology/approach: Pediococcus species were isolated from mother's milk, then tested for tolerance to acid and bile salts, their antibacterial effect and attachment to HT-29 cell line. In mice experiment, the levels of triglyceride (TG), total cholesterol (TC), HDL cholesterol (HDL-C), LDL cholesterol (LDL-C) and digestive enzymes such as amylase and lipase in serum of mice were measured after administration of 108 CFU/mL bacterial suspension. Findings: A total of 24 Pediococcus spp. including 16 isolates of Pediococcus pentosaceus and 8 isolates of Pediococcus acidilactici were isolated. Fifteen isolates (62.5%) were able to survive in low pH and bile salt concentration. Ten isolates (41.6%) exhibited the most powerful inhibitory effects against both Gram-positive and Gram-negative bacterial pathogens. A total of four Pediococcus isolates (16.67%) showed strong attachment to HT-29 cells. In contrast to HDL-C and amylase, the serum levels of TC, TG and LDL-C were reduced and lipase was elevated significantly following administration of mother milk-derived Pediococcus isolates. Originality/value: The Pediococcus isolates demonstrated good probiotic properties in particular cholesterol-lowering ability in mice. The increasing frequency and duration of marine heatwaves (MHWs) due to climate change pose severe threats to aquaculture, causing drastic physiological and growth impairments in farmed fish, undermining their resilience against additional environmental pressures. To ensure sustainable production that meets the global seafood demand and animal welfare standards, cost-effective and eco-friendly strategies are urgently needed. This study explored the efficacy of the red macroalga Asparagopsis taxiformis on juvenile white seabream Diplodus sargus reared under optimal conditions and upon exposure to a MHW. Fish were fed with four experimental diets (0%, 1.5%, 3% or 6% of dried powdered A. taxiformis) for a prophylactic period of 30 days (T30) and subsequently exposed to a Mediterranean category II MHW for 15 days (T53). Biometric data and samples were collected at T30, T53 and T61 (8 days post-MHW recovery), to assess performance indicators, biomarker responses and histopathological alterations. Results showed that A. taxiformis supplementation improved catalase and glutathione S-transferase activities and reduced lipid peroxidation promoted by the MHW, particularly in fish biofortified with 1.5% inclusion level. No histopathological alterations were observed after 30 days. Additionally, fish biofortified with 1.5% A. taxiformis exhibited increased citrate synthase activity and fish supplemented with 1.5% and 3% showed improved digestive enzyme activities (e.g., pepsin and trypsin activities). Overall, the present findings pointed to 1.5% inclusion as the optimal dosage for aquafeeds biofortification with A. taxiformis, and confirmed that this seaweed species is a promising cost-effective ingredient with functional properties and great potential for usage in a climate-smart context. Previous research studies confirmed that both resveratrol (RES) and β-hydroxy-β-methyl butyric acid (HMB) improved growth performance by altering intestinal microbiota. However, the mechanism underlying of RES and HMB on intestinal function remains unclear in ruminant. In this study, supplements of RES and HMB alone or in combination were evaluated as promoters of antioxidant capacity, immune response and barrier function, and modulators of the microbiota and metabolite profiles in the jejunum of Tibetan sheep. A total of 120 two-month-old Tibetan rams were randomly divided into four treatments (n = 30 per treatment), which were supplemented with a basal diet with 1.5 g RES/d (RES group), 1.25 g HMB/d (HMB group), 1.5 g RES/d plus 1.25 g HMB/d (RES-HMB group), and without additions (Control group). The results showed that RES and HMB improved the antioxidant capacity (CAT, GSH-Px, SOD, and T-AOC), immunity (IgA, IgG, and IgM), and digestive enzyme activity (α-amylase, lipase, and chymotrypsin) of the experimental lambs (p < 0.05). Additionally, jejunal morphology including villus width, villus height, and muscle layer thickness exhibited a significant difference when rams were fed diets supplemented with RES and HMB (p < 0.05). Furthermore, the determination of fermentation parameters showed that the butyrate concentration in the RES-HMB group was greater than those in the C and RES groups (p < 0.05). When compared to the C group, barrier-related gene expression (MUC-2, ZO-1, and IL-10) was significantly increased in the RES-HMB group (p < 0.05). Dietary RES and (or) HMB supplementation significantly increased the abundance of Methanobrevibacter, Actinobacteriota and Bacillus (p < 0.05). The abundance of differential bacteria was positively associated with butyrate concentration (p < 0.05). Metabolome analysis revealed that alpha ketoglutarate, succinic semialdehyde, and diacetyl as well as butanoate metabolism pathways connected to the improvements in butyrate concentration by RES and (or) HMB supplementation. Collectively, our results suggested that RES and (or) HMB supplementation improved butyrate concentration via regulating the microbial community (Methanobrevibacter, Actinobacteriota and Bacillus) and metabolism (alpha ketoglutarate, succinic semialdehyde, and diacetyl), thus contributing to jejunal morphology, antioxidant capacity, immune response, digestive enzyme activity, and barrier function. The increasing need for new treatments for obesity and diabetes has led to the development of new drugs and food supplements that could reduce carbohydrate absorption. Many starch blockers, based on common bean proteinaceous inhibitors against α-amylase (α-AI), are already present on the market. The extraction and purification of α-amylase inhibitor from a promising common bean cultivar from Sardinia (Nieddone) is described, highlighting the unique value of the Nieddone cultivar, particularly for its inhibitory activity on digestive enzymes and its complete lack of a hemagglutination effect on human red blood cells. The purification of α-AI involved two chromatographic steps (IEC and SEC) and was essential for revealing certain properties of the inhibitor. The purified inhibitor has a tetrameric structure (α2β2) and a molecular weight of approximately 42 kDa, as determined by SEC and SDS-PAGE, confirming it as a lectin-like inhibitor. The identification of the α-AI sequence was obtained by bottom-up high-resolution mass spectrometry, which allowed us to identify a unique peptide from the α chain and six unique peptides from the β chains. α-AI exhibited an optimum temperature of around 40 °C and two pH optima at 5 and 6.5, respectively. Its remarkable stability at high temperatures was measured (approximately 25% of activity retained even after 5 h at 100 °C), whereas the raw extract lost its activity entirely after just 10 min at 90 °C. Thus, the purification process significantly enhances the thermal stability of α-AI. The demonstrated effectiveness of the purified α-AI against the α-amylase enzyme in pigs, humans and insects underscores the protein's potential for treating obesity and diabetes, as well as for managing insect pests. AbstractTheaflavins, a major kind of component in black tea, have been reported to show a variety of biological activities and health effects. However, the unstable chemical properties, low bioavailability and unclear metabolism pathways of theaflavins have left much to be desired in terms of its specific efficacy and applications. This paper provides a comprehensive knowledge on the digestion, absorption, metabolism, distribution and excretion of theaflavins. We find that pH-dependent stability, efflux transport proteins are closely related to the low absorption rate and low bioavailability of theaflavins. When pass through the gastrointestinal tract, TFDG, TF2A and TF2B are gradually degraded to TF1, and release gallic acid. Then, the theaflavins skeleton are degraded into small molecular phenolic substances under the action of enzymes and microorganisms. In addition, theaflavins are widely distributed in the human body including brain, lung, heart, kidney, liver, blood tissue in a low content and can be excreted through feces. However, the influence of digestive enzymes barrier and gut microbial barrier on theaflavins are still unclear. Importantly, most findings are reported by in vitro methods and animal experiments, the metabolites and metabolic pathways of theaflavins in human body are not fully understood and need to be further investigated. Coral polyps are composed of four tissues; however, their characteristics are largely unexplored. Here we report biological characteristics of tentacles (Te), mesenterial filaments (Me), body wall (Bo), and mouth with pharynx (MP), using comparative genomic, morpho-histological, and transcriptomic analyses of the large-polyp coral, Fimbriaphyllia ancora. A draft F. ancora genome assembly of 434 Mbp was created. Morpho-histological and transcriptomic characterization of the four tissues showed that they have distinct differences in structure, primary cellular composition, and transcriptional profiles. Tissue-specific, highly expressed genes (HEGs) of Te are related to biological defense, predation, and coral-algal symbiosis. Me expresses multiple digestive enzymes, whereas Bo expresses innate immunity and biomineralization-related molecules. Many receptors for neuropeptides and neurotransmitters are expressed in MP. This dataset and new insights into tissue functions will facilitate a deeper understanding of symbiotic biology, immunology, biomineralization, digestive biology, and neurobiology in corals. The trail aimed to explore the effect of dietary supplementation of curcumin loaded olive oil nanoemulsion (CUR‐OLNE) on growth performance, feed utilization, blood biochemical, redox status, and immune response of Litopenaeus vannamei shrimp, considering the economic efficiency of supplementation. A total of 280 healthy shrimps (3.42 ± 0.02 g) were randomly distributed into five equal groups and were fed diets containing 0 (CUR‐OLNE0), 5(CUR‐OLNE5), 10(CUR‐OLNE10), 15(CUR‐OLNE15) and 20 (CUR‐OLNE20) mg CUR‐OLNE/kg diet, respectively for 16 weeks. Among CUR‐OLNE treated groups, CUR‐OLNE20 showed the highest growth performance and feed utilization traits, including final body weight, specific growth rate, feed conversion ratio, and protein efficiency ratio. Notably, the photomicrographs provided further compelling evidence regarding the potential effect of CUR‐OLNE supplementation on muscle structure and integrity. Compared to the control, the levels of blood protein significantly induced in CUR‐OLNE15 and CUR‐OLNE20 treated groups (p < 0.05). All CUR‐OLNE ‐supplemented groups possessed lower activities of liver enzymes as well as the levels of urea and creatinine compared to the control (p < 0.05). The addition of 20 mg CUR‐OLNE/kg diet decreased the concentrations of cortisol, glucose and triglycerides. The dietary treatment significantly improved the secretion of digestive enzymes, including amylase, lipase, and protease. The lowest levels of Malondialdehyde and the highest levels of total antioxidant capacity, super oxide dismutase, catalase, lysozyme and immunoglobulin M were detected in both of CUR‐OLNE15, and CUR‐OLNE20 treated groups compared to the control (p < 0.05). There were considerable significant effects of dietary supplementation of CUR‐OLNE on economic efficiency. In conclusion, the application of nanocarriers for the delivery of dietary immune stimulants such as CUR‐OLNE to Litopenaeus vannamei shrimp is a promising strategy for improving shrimp nutrition. Gut microbial communities are highly dynamic and respond to factors such as diet and host metabolism. However, there is limited information about how these responses can generate community dynamics at different time scales, such as circadian rhythms. The aim of the present study was to characterize variations along a daily cycle in the gut microbiota of fish on different feeding regimes. In addition, an estimation of the potential contribution of the gut bacterial community to the digestive process of the fish was assessed. The greater amberjack (Seriola dumerili) was used as experimental species, a promising species for Mediterranean aquaculture. Fish were randomly distributed in two different feeding regimes, in triplicate rearing tanks. Feeding regimes were continuous feeding and time-restricted feeding (three meals per day). Fish feces for RNA-based amplicon sequencing and bacterial proteolytic capacity were sampled along a 24h cycle. Time-restricted fed fish exhibited changes in the composition of the active bacterial community promoted by feeding, thus revealing two types of bacterial community profiles: "pre-feeding" and "active-feeder" profiles. Continuous-fed fish maintained an "active-feeder" community profile throughout the whole day. Bray-Curtis and Sørensen similarity indicated that differences in the composition between the two community profiles were mainly driven by the absence/presence of infrequent ASVs (amplicon variant sequences). The potential proteolytic bacterial activity tended to be higher before feeding. Thus, gut microbiota could be a key factor to understanding fish digestion, playing a role in interactions with host metabolism. This study aimed to evaluate the impact of substituting a portion of feed with Tenebrio molitor (TM) and Elodea nuttallii (EN) on crayfish culture. A total of 270 crayfish (5.1 ± 0.4 g) were fed three different diet combinations (A: 100% feed; B: 80% feed + 10% TM + 10% EN; C: 75% feed + 15% TM + 10% EN) for 12 weeks. The findings demonstrated that group C had an important beneficial impact on the growth performance of crayfish. This was evidenced by a rise in digestive enzyme activity (trypsin, lipase, and cellulase) in the intestinal and hepatopancreas, as well as an upregulation in the expression of growth-related genes (ghsr, igfbp7, mhc, mlc1, mef2, and pax7) in the muscle. Furthermore, the assessment of the flesh quality of crayfish muscle in group C was conducted. The findings indicated a significant increase (p < 0.05) in the energy value (moisture, crude protein, and crude lipid) within the muscle. The levels of delicious amino acids (Glu, Ala, Ser, Gly, and Tyr) and polyunsaturated fatty acids (ARA, DHA) were enhanced, resulting in an improved nutritional profile and flavor of the muscle while maintaining the Σn-3/Σn-6 ratio. The remodeling of the intestinal microbiota (abundance of Proteobacteria and ratio of Firmicutes/Bacteroidota bacteria) also revealed improved growth performance. Over the years, the world has continued to be plagued by type 2 diabetes (T2D). As a lifestyle disease, obese individuals are at higher risk of developing the disease. Medicinal plants have increasingly been utilized as remedial agents for managing metabolic syndrome. The aim of the present study was to investigate the in vitro anti-hyperglycemic and anti-lipidemic potential of Croton gratissimus herbal tea infusion. The inhibitory activities of C. gratissimus on carbohydrate (α-glucosidase and α-amylase) and lipid (pancreatic lipase) hydrolyzing enzymes were determined, and the mode of inhibition of the carbohydrate digestive enzymes was analyzed and calculated via Lineweaver–Burk plots and Michaelis Menten's equation. Its effect on Advanced Glycation End Product (AGE) formation, glucose adsorption, and yeast glucose utilization were also determined. High-performance liquid chromatography (HPLC) was used to quantify the possible phenolic compounds present in the herbal tea infusion, and the compounds were docked with the digestive enzymes. C. gratissimus significantly (p < 0.05) inhibited α-glucosidase (IC50 = 60.56 ± 2.78 μg/mL), α-amylase (IC50 = 35.67 ± 0.07 μg/mL), as well as pancreatic lipase (IC50 = 50.27 ± 1.51 μg/mL) in a dose-dependent (15–240 µg/mL) trend. The infusion also inhibited the non-enzymatic glycation process, adsorbed glucose effectively, and enhanced glucose uptake in yeast cell solutions at increasing concentrations. Molecular docking analysis showed strong binding affinity between HPLC-quantified compounds (quercetin, caffeic acid, gallic acid, and catechin) of C. gratissimus herbal tea and the studied digestive enzymes. Moreover, the herbal tea product did not present cytotoxicity on 3T3-L1 cell lines. Results from this study suggest that C. gratissimus herbal tea could improve glucose homeostasis and support its local usage as a potential anti-hyperglycemic and anti-obesogenic agent. This study presents the phytochemical composition, antioxidant (hydrogen atom and single-atom transfer mechanisms), and digestive enzyme inhibitory (alpha-amylase, alpha-glucosidase, and pancreatic lipase) activities of ethanol-based extractions and traditional preparations (infusion) of the leaves of wild-edible Allium species (A. kharputense, A. affine, A. shirnakiense, and A. akaka) from the highlands of Eastern Anatolia. Among the eight extracts analyzed, ethanol extractions of the A. kharputense and A. akaka leaves exhibited better biotherapeutic activities and had the highest bioactive content. The dominant bioactive profile was composed of mainly allicin and phenolic compounds (chlorogenic acid, hesperidin, rutin, isoquercitrin, and quercetin) with small amounts of fatty acids. These data were similar to the biological activities and chemical composition of common Allium species and suggest the utilization of the extracts of wild-edible Allium species in the development of Allium-based biotherapeutics or nutraceuticals. The bovine rumen contains a large consortium of residential microbes that release a variety of digestive enzymes for feed degradation. However, the utilization of these microbial enzymes is still limited because these rumen microorganisms are mostly anaerobes and are thus unculturable. Therefore, we applied a sequence-based metagenomic approach to identify a novel 2,445-bp glycoside hydrolase family 3 β-glucosidase gene known as BrGH3A from the metagenome of bovine ruminal fluid. BrGH3A β-glucosidase is a 92-kDa polypeptide composed of 814 amino acid residues. Unlike most glycoside hydrolases in the same family, BrGH3A exhibited a permuted domain arrangement consisting of an (α/β)6 sandwich domain, a fibronectin type III domain and a (β/α)8 barrel domain. BrGH3A exhibited greater catalytic efficiency toward laminaribiose than cellobiose. We proposed that BrGH3A is an exo-acting β-glucosidase from Spirochaetales bacteria that is possibly involved in the intracellular degradation of β-1,3-/1,4-mixed linkage glucans that are present in grass cell walls. Microbial communities have been documented as playing many pivotal roles, and contributing to the growth or health performance of animal hosts. Thus, many studies are currently looking for potential beneficial bacteria "probiotics" from diverse environments, including wild species. The present study aimed to investigate the diversity and potential metabolic functions of bacterial communities in the gastrointestinal tract of wild spiny lobsters. The gastrointestinal (GI) tracts of two wild lobster species (Panulirus ornatus and Panulirus homarus) were dissected aseptically and analyzed through high-throughput sequencing, followed by PICRUSt analysis. The results exposed that the most dominant phyla inhabiting both lobster species at the post-puerulus and juvenile stages were Proteobacteria, Firmicutes, Bacteriodota, Patescibacteria, and Verrucomicrobiota, while at the genus level, the GI tracts were mostly dominated by Photobacterium, Candidatus Bacillopora, Vibrio, and Catenococcus at the post-peurulus stage, and Vibrio, Catenococcus, Acanthopleuribacter, Acinetobacter, Pseudoalteromonas, Grimontia, and Photobacterium at the juvenile stage. Further metagenomic prediction analysis discovers many potential probiont properties indicated by the detection of marker genes corresponding to many important metabolic activities, such as antimicrobial compounds (streptomycin, vancomycin, carbapenem, tetracycline, novobiocin, penicillin, cephalosporin, ansamycines, butirosin, and neomycin), antioxidants (e.g., flavonoids and carotenoids), and several important digestive enzymes (e.g., lipase, protease, and amylase). These results suggest that GI tracts of wild spiny lobsters are potential sources to discover novel probionts for aquaculture purposes. Further studies, such as the isolation of the natural product-producing bacteria, or cloning of the beneficial compound-identified genes, are highly recommended to develop novel probiotic strains for aquaculture purposes. In this study, we conducted a 16-week feeding trial to investigate the effects of a high-cassava starch diet on growth performance, liver function, and metabolism in largemouth bass (Micropterus salmoides). We formulated five diets containing varying levels of cassava starch: 12%, 9%, 6%, 3%, and 0% (termed M12, M9, M6, M3, and M0, respectively). We distributed these diets among largemouth bass with the initial body weight of 83.33 ± 0.55 g via an in-pond "raceway" aquaculture system. Our findings suggest that high level (12%) of cassava starch dietary inclusion adversely affected growth performance metrics such as weight gain rate and specific growth rate, along with feed utilization efficiency indicators, including protein efficiency, protein deposition rate, and the apparent digestibility of dry matter and protein. This negative impact was accompanied by a decrease in intestinal amylase activity. Through further transcriptomic analysis, we identified several key genes associated with carbohydrate metabolism, which underwent changes influencing liver function. Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis revealed the involvement of these differentially expressed genes (DEGs) in the tricarboxylic acid cycle (TCA cycle). Comparative metabolomics analysis further indicated that the M9 group showed significant enrichment in pathways related to amino acid metabolism and alterations in the levels of metabolites involved in carbohydrate metabolism. In conclusion, our study demonstrates that incorporating up to 9% cassava starch in the diet can enhance growth performance in largemouth bass by stimulating digestive enzyme activities and promoting glucose utilization. This study was designed to evaluate the effects of yacon (Smallanthus sonchifolius) juice byproduct (YJB) on the growth performance, digestive and antioxidant enzyme activities, and disease resistance against Streptococcus iniae of juvenile black rockfish (Sebastes schlegelii) based on different feeding schedules. Four different YJB feeding strategies were evaluated: feeding the fish a basal diet continuously (control, T0), feeding them YJB (2.5 g/kg) continuously (T1), feeding them YJB for 1 day and the basal diet the next day (T2), and feeding them YJB for 1 day and the basal diet for the following 2 days (T3). No difference in survival among the treatments was found after the 8-week feeding trial (p > 0.05). However, the T1 and T2 groups exhibited significant enhancements in final body weight, weight gain, and specific growth rate compared with the T0 and T3 groups. Furthermore, the T1 and T2 groups showed a significant improvement in feed consumption, feed efficiency, and protein efficiency ratio compared with the T0 and T3 groups. No significant differences in the condition factor or viscerosomatic and hepatosomatic indices were observed among all the groups. Intestinal amylase, trypsin, and lipase activity was significantly (p < 0.05) higher in the T1 and T2 groups than in the T0 and T3 groups. Lysozyme, superoxide dismutase, and catalase activity along with glutathione peroxidase content were significantly (p < 0.05) higher under all YJB feeding regimens than those under the control treatment. The survival rates in all the YJB treatment groups after the S. iniae challenge were significantly (p < 0.05) enhanced. In conclusion, we recommend offering YJB at day-to-day intervals to improve growth performance, digestive enzyme activity, antioxidant status, and disease resistance against S. iniae. Simple Summary: This study evaluated the effect of dietary supplementation with organic silicon on the growth performance, blood biochemistry, digestive enzymes, morphohistology, intestinal microbiota and stress resistance in juvenile hybrid Tilapia (Oreochromis mossambicus × Oreochromis niloticus). It is concluded that organic silicon had no negative effects on the physiology and metabolism of the fish. The results showed that up to 50 mg·kg−1 of organic silicon can be applied to the diet of red Tilapia, improving digestive, metabolic and growth performance. In recent decades, interest has been aroused worldwide in the use of silicon in nutrition; however, information on its effect on nutrition and metabolism of fish is limited. The objective of the research was to evaluate the effect of dietary supplementation with organic silicon on the growth performance, blood biochemistry, digestive enzymes, morphohistology and intestinal microbiota and stress resistance in hybrid Tilapia (Oreochromis mossambicus × Oreochromis niloticus). Methodologically, six levels of organic silicon (DOS) [control (0), 10, 20, 30, 40 and 50 mg·kg−1] were used to feed juvenile fish (initial weight 7.51 ± 0.25 g) grown for eight weeks in 18 aquariums (15 fish/aquarium). The results indicated that growth performance showed differences (p < 0.05) for specific growth rate, feed conversion and survival. Triglycerides, cholesterol and glucose, transaminases and digestive enzymes were significantly influenced by DOS levels. The histological study confirmed that the administered diets did not cause damage and induced significant morphological changes in the proximal intestine. The 16S rRNA gene sequencing analysis of the gut microbiota showed a high diversity and richness of OTU/Chao-1, with Fusobacteria, Proteobacteria, Bacteroidetes and Acidobacteria predominating in the DOS treatments compared to the control (p < 0.05). Induction of hypoxia stress after the feeding period showed a significant relative survival rate of 83.33% in fish fed 50 mg·kg−1. It is concluded that the DOS treatments performed better than the control treatment in most of the variables analysed. DOS had no negative effects on the fish. A 90 days of feeding trial was employed for evaluating the effects of dietary supplementation with curcumin nanoparticles (CURNPs) on growth performance, hematological and biochemical parameters, immune response, antioxidant status, and intestinal and hepatic architecture in European seabass (Dicentrarchus labrax) fingerlings. Fish (n = 300, initial body weight = 10.02 g ± 0.11 g) received supplementary CURNPs at rates of 0 (control; CURNPs0), 50 (CURNPs50), 100 (CURNPs100) and 150 (CURNPs150) mg/kg diet for 90 days. Growth performance (final body weight (FBW), daily weight gain (DWG), weight gain (WG), relative growth rate (RGR), and protein efficiency ratio (PER)) and feed uptake (feed conversion rate (FCR)) were improved significantly in CURNPs (100 and 150 mg/ kg)-supplemented groups. The hematological parameters red blood corpuscles (RBCs), hemoglobin (Hb), and packed cell volume (PCV) were improved significantly (P<0.001) in CURNPs150 and CURNPs100 groups, while CURNPs150 had the lowest values of monocytes (P = 0.019) and heterophils (P<0.005) when compared with other groups. Glucose was higher and lipid profile (total cholesterol and triglycerides) were lower in all CURNPs-supplemented groups as compared to the control group. Fish fed with CURNPS at various levels exhibited remarkable increase in digestive enzymes (amylase and lipase), immune markers (immunoglobulin G (IgG), lysozyme and phagocytic indices), and antioxidant enzymes (superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx)). A noticeable decrease (P<0.05) in malondialdehyde (MDA) and considerable upregulation of hepatic insulin-like growth factor 1 (IGF1) and interleukin-1beta (IL-1β) mRNA levels were found in CURNPs-supplemented fish in a dose-dependent manner. The down regulation of hepatic heat shock protein 70 (HSP70) was observed in CURNPs150. Intestinal and hepatic architectures were improved significantly by CURNPs. An evaluation of six dietary protein-to-energy (P/E) ratios was conducted to the early juvenile lobsters (Panulirus ornatus) by investigating the ratio effects on physiology, digestive enzyme activities, body composition, and amino acid profiles. Juvenile lobsters (0.25±0.01 g) were individually kept in aquaria and treated with six isoenergetic formulated diets of P/E ratios of 21.8, 22.5, 23.5, 24.6, 25.3 and 26.6 mg KJ–1. After 8 weeks of feeding experiment, the differentiation on dietary P/E ratios was found to have affected the specific growth rates, but not the survival, moult increment, moulting rate, moult interval and average daily moulting rate of juvenile lobsters. The dietary P/E ratio also influenced the protease, lipase, superoxidase dismutase activities, and body composition of juvenile lobster. The increase in dietary protein levels significantly affected the increase of the specific growth rate, while increased dietary lipid levels only affected the length increments. Both protein and lipid content were found in the whole body of the juveniles which reflected the treated dietary composition. Results indicated that a P/E ratio of 26.6 mg KJ–1 (55% dietary protein and 9% dietary lipids) can yield the maximum growth and digestive enzyme activities from among the studied diets. Integrative studies are important for a better understanding of the functional development of fish digestive systems and the consequent design of suitable feeding regimes and ad-hoc diets. To characterize the ontogeny of digestive function in larvae and juveniles of two atherinopsids from South America, Odontesthes bonariensis and O. hatcheri, morphologic, histologic, and intestinal enzyme biochemical analyses were performed. Both species showed a functional digestive system from one week after hatching (wah) with a sustained high-level activity of the cytosolic enzyme leucine alanine peptidase (leu-ala) until 9 and 13 wah in O. hatcheri and O. bonariensis, respectively. Maximum specific leu-ala activities in both species were higher compared to other agastric and most gastric fish. These results and the abundant pinocytic vacuoles in the posterior intestine suggest that cytosolic digestion is crucial for these species. It seems that their apparent anatomically simple digestive system is compensated by an uncommonly early development and function of the digestive tract and accessory organs accompanied by high intestinal cytosolic activity. The above findings contribute to the understanding of the Atherinopsidae digestive model and, for the first time, suggest that under farming conditions, these species could be weaned at an early developmental stage with an appropriate balanced diet. The Cattle hair meal (CHM) has the potential of usage as a dietary protein source in fish nutrition due to the qualitative nutrients present in it and the enormous quantity generated at abattoirs worldwide. This study investigated the inclusion of CHM in diet of Clarias gariepinus as fishmeal replacement. Catfish juveniles with average weight of 5.86±0.14g were stocked in 15 plastic tanks at 21fish/tank in completely randomized design. The experimental fish were fed with 40% crude protein diet at 0%, 25%, 50%, 75% and 100% inclusion of CHM at 3% body weight for duration of 8 weeks. Growth performance, carcass quality, haematology, digestive enzymes, gut morphology and histology of C. gariepinus were assessed. Growth parameters differed significantly (P<0.05) among treatments with similar responses in carcass composition. The blood parameters, digestive enzymes and intestinal morphology also showed variations among treatments. Histological observation revealed that only diet with 25% CHM inclusion showed no alteration in tissues of kidney, liver and intestine. This study recommends usage of CHM up to 25% as replacement for fishmeal in C. gariepinus diet. [ABSTRACT FROM AUTHOR]

The tiger shrimp (Penaeus monodon Fabicius, 1798) is a popular Indonesian fishery product with high market value both domestically and internationally. Its high nutritional value makes it a valuable product that is in high demand. Along with increased production, various challenges appeared in management and feed quality aspects. Multi-enzymes are one method for increasing the tiger shrimp productivity. This study aimed to identify the dose of multi-enzymes in the feed that leads to an optimal growth performance, and optimum activity of digestive enzymes (amylase, protease, lipase), as well as improving the essential amino acids index in the shrimp body. This study used a completely randomized design (CRD) with 4 treatments and 3 replications each, thus 12 experimental units were applied. The treatments used were A at 0%/kg feed, B at 40%, C at 60%, and D at 80%/kg feed. The experimental animal was the tiger shrimp with a mean weight of 0.07g/ ind. The shrimps were reared in the hatchery laboratorium of the Marine Science and Fishery Faculty, Hasanuddin University, Makassar. The parameters examined were essential amino acid index (EAAI), essential amino acid score (EAAS), enzyme type activity, specific growth rate (SGR), relative weight gain (RWG), survival rate (SR), and protein efficiency ratio (PER). The results showed that feed with 80% multienzyme/kg provided the highest EAAI compared to other doses, namely 2.64. In addition, the most EAAS deficiency occurred in methionine EAA, namely 9.29 in multi-enzyme feed with a dose of 40%/kg. SGR, RWG, and PER were not significantly different (P> 0.05), but SR showed a significant difference (P< 0.05). The activity of amylase, protease, and lipase enzymes in the tiger shrimp also increased with the addition of multienzyme doses to the feed. [ABSTRACT FROM AUTHOR]
