Denphal-type Dendrobium is the famous cut and potted flower in the world, and most cultivated in tropical and subtropical regions. However, it often suffers from cold in winter in subtropical regions. To verify the physiological response of Denphal-type Dendrobium under low temperature in this study, the mature and young plantlets of Dendrobium Udomsri Beauty were treated under 15, 10, and 5 °C, respectively. And then the electrical conductivity (EC), soluble protein, soluble sugar, free proline, malondialdehyde (MDA), chlorophyll content, and the rate of defoliation after regrowth were measured. The results showed that both mature plant and young seedlings of Dendrobium Udomsri Beauty, the EC, soluble protein, soluble sugar, free proline, MDA content, and defoliation rate were increased with the decrease of treatment temperature and the extension of treatment times. The content of chlorophyll decreased gradually with the decrease of treatment temperature and the extension of treatment times. The correlation analysis showed that soluble sugar, free proline, MDA, chlorophyll content, and defoliation rate were significantly correlated with the semi-lethal temperature. It is indicated that the content of free proline, MDA, chlorophyll, and defoliation rate could be used as the effective indexes for the comprehensive assessment of cold tolerance of Dendrobium Udomsri Beauty. Moscatilin, a bibenzyl derivative from the Dendrobium genus, has been traditionally used in Chinese medicine. Recent studies suggest its potential as a powerful anticancer agent due to its diverse pharmacological properties.This review aims to consolidate current research on moscatilin's anticancer mechanisms, structure–activity relationships, and therapeutic potential to assess its viability for clinical use. A literature search was performed in PubMed/MedLine, Scopus, and Web of Science.The search focused on "cancer," "moscatilin," "anticancer," "bioactivity," "dendrobium," and "pharmacological properties." Relevant studies on molecular mechanisms, preclinical and clinical efficacy, and bioavailability were reviewed. Moscatilin exhibits significant anticancer effects in lung, breast, colorectal, and pancreatic cancers. It induces apoptosis via the JNK/SAPK pathway, inhibits cell proliferation, and suppresses metastasis. Structure–activity relationship studies reveal that phenolic groups and a two-carbon bridge are crucial for its efficacy. Additionally, moscatilin shows good bioavailability and a favorable safety profile, with low toxicity to healthy cells. Moscatilin demonstrates considerable potential as an anticancer agent, targeting multiple cancer progression pathways. SUMMARY Erianin, crepidatin, and chrysotobibenzyl are typical medicinal polymethoxylated bibenzyls (PMBs) that are commercially produced in Dendrobium species. PMBs' chemo‐diversity is mediated by the manifold combinations of O‐methylation and hydroxylation in a definite order, which remains unsolved. To unequivocally elucidate the methylation mechanism of PMBs, 15 possible intermediates in the biosynthetic pathway of PMBs were chemically synthesized. DcOMT1–5 were highly expressed in tissues where PMBs were biosynthesized, and their expression patterns were well‐correlated with the accumulation profiles of PMBs. Moreover, cell‐free orthogonal tests based on the synthesized intermediates further confirmed that DcOMT1‐5 exhibited distinct substrate preferences and displayed hydroxyl‐group regiospecificity during the sequential methylation process. The stepwise methylation of PMBs was discovered from SAM to dihydro‐piceatannol (P) in the following order: P → 3‐MeP → 4‐OH‐3‐MeP → 4‐OH‐3,5‐diMeP → 3,3'(4′),5‐triMeP → 3,4,4′,5‐tetraMeP (erianin) or 3,3′,4,5‐tetraMeP (crepidatin) → 3,3′,4,4′,5‐pentaMeP (chrysotobibenzyl). Furthermore, the regioselectivities of DcOMTs were investigated by ligand docking analyses which corresponded precisely with the catalytic activities. In summary, the findings shed light on the sequential catalytic mechanisms of PMB biosynthesis and provide a comprehensive PMB biosynthetic network in D. catenatum. Background: Dendrobium officinale Kimura et Migo (D. officinale) is parasitic on rocks or plants with very few mineral elements that can be absorbed directly, so its growth and development are affected by nutritional deficiencies. Previous studies found that phosphorus deficiency promotes polysaccharides accumulation in D. officinale, the expression of DoCSLA6 (glucomannan synthase gene) was positively correlated with polysaccharide synthesis. However, the molecular mechanism by which the low phosphorus environment affects polysaccharide accumulation remains unclear. Results: We found that DoSPX1 can reduce phosphate accumulation in plants and promote the expression of PSIs genes, thereby enhancing plant tolerance to low phosphorus environments.Y1H and EMSA experimental show that DoMYB37 can bind the promoter of DoCSLA6. DoSPX1 interact with DoMYB37 transiently overexpressed DoSPX1 and DoMYB37 in D. officinale protocorm-like bodies, decreased the Pi content, while increased the expression of DoCSLA6. Conclusions: The signaling pathway of DoSPX1-DoMYB37-DoCSLA6 was revealed. Dendrobium species, which are perennial herbs widely distributed in tropical and subtropical regions, are notable for their therapeutic properties attributed to various bioactive compounds, including dendrobine-type sesquiterpenoid alkaloids (DTSAs). The objective of this review article is to provide a comprehensive overview of recent advances in the biosynthesis of DTSAs, including their extraction from Dendrobium species and endophytes, elucidation of associated genes through genomic and transcriptomic sequencing in both Dendrobium spp. and endophytes, exploration of the biosynthetic pathways of DTSAs, and drawing conclusions and outlining future perspectives in this field. Alkaloids, predominantly nitrogen-containing compounds found in medicinal orchids, include over 140 types discovered across more than 50 species. DTSAs, identified in 37 picrotoxane alkaloids, have a distinctive five-membered nitrogen heterocyclic ring. This review highlights endophytic fungi as alternative sources of DTSAs, emphasizing their potential in pharmaceutical applications when plant-derived compounds are scarce or complex. Genomic and transcriptomic sequencing of Dendrobium spp. and their endophytes has identified key genes involved in DTSAs biosynthesis, elucidating pathways such as the mevalonate (MVA) and 2-C-methyl-D-erythritol 4-phosphate (MEP) pathways. Genes encoding enzymes, such as acetyl-CoA C-acetyltransferase and diphosphomevalonate decarboxylase, are positively associated with dendrobine production. Despite significant advancements, the complexity of terpenoid biosynthesis in different subcellular compartments remains a challenge. Future research should focus on leveraging high-quality genomic data and omics technologies to further understand and manipulate the biosynthetic pathways of DTSAs and enhance their medicinal use. Background: Dendrobium Sw. represents one of the most expansive genera within the Orchidaceae family, renowned for its species' high medicinal and ornamental value. In higher plants, the ankyrin (ANK) repeat protein family is characterized by a unique ANK repeat domain, integral to a plethora of biological functions and biochemical activities. The ANK gene family plays a pivotal role in various plant physiological processes, including stress responses, hormone signaling, and growth. Hence, investigating the ANK gene family and identifying disease-resistance genes in Dendrobium is of paramount importance. Results: This research identified 78 ANK genes in Dendrobium officinale Kimura et Migo, 77 in Dendrobium nobile Lindl., and 58 in Dendrobium chrysotoxum Lindl. Subsequently, we conducted comprehensive bioinformatics analyses on these ANK gene families, encompassing gene classification, chromosomal localization, phylogenetic relationships, gene structure and motif characterization, cis-acting regulatory element identification, collinearity assessment, protein-protein interaction network construction, and gene expression profiling. Concurrently, three DoANK genes (DoANK14, DoANK19, and DoANK47) in D. officinale were discerned to indirectly activate the NPR1 transcription factor in the ETI system via SA, thereby modulating the expression of the antibacterial PR gene. Hormonal treatments with GA3 and ABA revealed that 17 and 8 genes were significantly up-regulated, while 4 and 8 genes were significantly down-regulated, respectively. DoANK32 was found to localize to the ArfGAP gene in the endocytosis pathway, impacting vesicle transport and the polar movement of auxin. Conclusion: Our findings provide a robust framework for the taxonomic classification, evolutionary analysis, and functional prediction of Dendrobium ANK genes. The three highlighted ANK genes (DoANK14, DoANK19, and DoANK47) from D. officinale may prove valuable in disease resistance and stress response research. DoANK32 is implicated in the morphogenesis and development of D. officinale through its role in vesicular transport and auxin polarity, with subcellular localization studies confirming its presence in the nucleus and cell membrane. ANK genes displaying significant expression changes in response to hormonal treatments could play a crucial role in the hormonal response of D. officinale, potentially inhibiting its growth and development through the modulation of plant hormones such as GA3 and ABA. Dendrobium species, esteemed as a group of orchid plants with important ornamental and medicinal value, are currently experiencing significant developmental advancements. This research encompasses a comprehensive review of Dendrobium tissue culture literature spanning 2008 to 2023, sourced from the Web of Science Core Collection database. Employing the bibliometric analysis tool CiteSpace, we conducted a visualization and presentation of data encompassing research institutions, authorship patterns, key terminology, and emergent themes, elucidating the current status, hotspots, and frontiers of Dendrobium tissue culture research. After thorough screening, a total of 261 papers were included in this study. The publication count exhibited a consistent annual increase. Chinese Acad Sci, Univ Sains Malaysia, North Eastern Hill Univ, Univ Debrecen, and Univ Putra Malaysia emerged as primary research institutions, while a core team led by J.A. Teixeira da Silva, S. Sreeramanan, S. Kumaria, and N. Chen played a dominant role. Central research themes revolve around optimizing the tissue culture system of medicinal Dendrobium, and exploring the genetic stability and gene function. Glutathione S-transferases (GSTs) are members of a protein superfamily with diverse physiological functions, including cellular detoxification and protection against oxidative damage. However, there is limited research on GSTs responding to cadmium (Cd) stress. This study classified 46 GST genes in Dendrobium officinale (D. officinale) into nine groups using model construction and domain annotation. Evolutionary analysis revealed nine subfamilies with diverse physical and chemical properties. Prediction of subcellular localization revealed that half of the GST members were located in the cytoplasm. According to the expression analysis of GST family genes responding to Cd stress, DoGST5 responded significantly to Cd stress. Transient expression of DoGST5-GFP in tobacco leaves revealed that DoGST5 was localized in the cytoplasm. DoGST5 overexpression in Arabidopsis enhanced Cd tolerance by reducing Cd-induced H2O2 and O2− levels. These findings demonstrate that DoGST5 plays a critical role in enhancing Cd tolerance by balancing reactive oxygen species (ROS) levels, offering potential applications for improving plant adaptability to heavy metal stress. Dendrobium hybrids have a significant role in the present floral sector. The aim of this research was to evaluate how various light qualities affect the physiological and biochemical traits of Dendrobium 'Shuijing'. In order to determine the optimal light quality for in vitro cultivation of Dendrobium plantlets, we examined the correlations between growth, antioxidant capacity, and nutrient and chlorophyll levels, as well as chlorophyll fluorescence. The growth rate was compared by using different light qualities emitted by the LED light source. These included red light (R), blue light (B), and three ratios: 8R:2B, 7R:3B, and equal proportions of both colors, known as white, fluorescent light (CK). The combination of 7R:3B resulted in noticeable enhancements in leaf count, root length, root activity, fresh and dry weight measurements, antioxidant capability, as well as chlorophyll content and fluorescence. Specifically, the mixture of red and blue LED lights at a ratio of 7R:3B led to increased leaf number, root length, root activity, fresh and dry weight measurements, antioxidant ability, and chlorophyll content with improved fluorescence. In order to explore the effect of light quality on the growth and development of Dendrobium, the chlorophyll content and chlorophyll fluorescence parameters of plants under all light quality conditions were analyzed by using a linear regression model with other physiological and biochemical indexes. A significant correlation between non-photochemical quenching (NPQ) and leaf length was also observed. The content of chlorophyll b showed significant correlations with both root number and leaf number. Furthermore, chlorophyll a, along with its ratio to chlorophyll b, significantly correlated with root length. Chlorophyll b and the relative electron transport rate of PSII (ETRII) significantly correlated with root activity and the free proline content (FPC) and catalase (CAT) activity. The photochemical quenching coefficient (qP) significantly correlated with total soluble sugars content (SSC) and peroxidase (POD) activity. The correlation between the quantum yield of PSII (Fv/Fm ratio) and superoxide dismutase (SOD) activity was found to be significant. Similarly, the effective quantum yield of PSII photochemistry (ΦPSII) showed significant correlations with fresh weight, dry weight, soluble protein content (SPC), and ascorbate peroxidase (APX) activity. Through a principal component analysis (PCA), it was observed that plants cultivated under the 7R:3B light treatment achieved significantly better comprehensive scores compared to those grown under different light treatments. In conclusion, growth achieved under an LED emitting a ratio of 7R:3B light yielded the most robust Dendrobium hybrid plantlets within a controlled environment. [ABSTRACT FROM AUTHOR]

Erianin, a bibenzyl compound found in dendrobium extract, has demonstrated broad anticancer activity. However, its mechanism of action in gastric cancer (GC) remains poorly understood. LKB1 is a tumor-suppressor gene, and its mutation is an important driver of various cancers. Yet some studies have reported contradictory findings. In this study, we combined bioinformatics and in vitro and in vivo experiments to investigate the effect and potential mechanism of Erianin in the treatment of GC. The results show that LKB1 was highly expressed in patients' tumor tissues and GC cells, and it was associated with poor patient prognosis. Erianin could promote GC cell apoptosis and inhibit the scratch repair, migration, invasion, and epithelial–mesenchymal transition (EMT) characteristics. Erianin dose-dependently inhibited the expression of LKB1, SIK2, SIK3, and PARD3 but had no significant effect on SIK1. Erianin also inhibited tumor growth in CDX mice model. Unexpectedly, 5-FU also exhibited a certain inhibitory effect on LKB1. The combination of Erianin and 5-FU significantly improved the anti-tumor efficacy of 5-FU in the growth of GC cells and xenograft mouse models. In summary, Erianin is a potential anti-GC compound that can inhibit GC growth and EMT properties by targeting the LKB1-SIK2/3-PARD3-signaling axis. Dendrobium section Densiflora species hold important ornamental and medicinal value. To improve the efficiency of breeding and cultivation, in this study, we aimed to elucidate the types of photosynthetic carbon assimilation pathways in D. brymerianum, D. chrysotoxum, D. densiflorum, D. harveyanum, D. sulcatum, D. thyrsiflorum, D. jenkinsii, D. lindleyi, all of which are known native Chinese species of Dendrobium section Densiflora, by analyzing stable carbon isotope ratios(δ13C), diurnal net CO2 assimilation, and titratable acidity. The results showed that the δ13C values of D. jenkinsii and D. lindleyi were greater than-20‰, identifying as crassulacean acid metabolism（CAM） plants, while the δ13C values of other 6 species were less than-20‰, identifying as C3 plants. Following drought stress, D. jenkinsii had net CO2 uptake during the whole night, and D. lindleyi had net CO2 exchange for over two-thirds of the night. The difference in titratable day and night acid level（△H+） between these two plants was markedly higher than that of C3 plants, providing further evidence that both are CAM plants. All C3 species exhibited marked net CO2 uptake during the daytime; however, acid accumulation in them was more markedly at night, exceeding 4 mmol·kg-1. The only exception was D. brymerianum, which did not show night acid accumulation, indicating that this effect is unique to the obligate C3 plants, while the remaining five species were facultative CAM plants. Epiphytism is an example of an obligate interspecific interaction between plants, where a plant species grows on another plant species for physical support. In epiphytic orchids that rely on suitable host tree species (phorophytes) and mycorrhizal fungi for germination and growth, a distinct preference has been observed for particular host tree species. However, the factors driving this host preference remain unclear.In this study, we investigated whether mycorrhizal fungi that promote seedling development determine the preference of phorophytes in the epiphytic orchid Dendrobium wangliangii. First, we identified the preferred host tree species for this orchid. Next, in vitro and in situ germination experiments were conducted to identify potential fungi that promote seed germination. Finally, high‐throughput sequencing was used to assess the distribution of these fungi on both colonized host trees and potential host trees, including individual trees of the host species not yet colonized by the orchid and trees of another species growing in the vicinity of a colonized tree.Dendrobium wangliangii exhibited a preference for trees within the Fagaceae family. Two Fusarium species from the bark of host trees were identified as the primary fungi that promote seed germination. Successful protocorm formation and seedling development were observed exclusively on preferred phorophytes. Fungal community analyses further indicated that both Fusarium spp. occurred on all potential host trees, suggesting that the host preference of this epiphytic orchid is not constrained by the distribution of suitable mycorrhizal fungi.Synthesis: Regarding the epiphytes that depend on mycorrhizal fungi for successful seedling recruitment, we conclude that strong epiphyte–phorophyte associations do not arise from the limited distribution of mycorrhizal fungi but are more likely the result of other factors inherent in the phorophyte environment. Dendrobium stem is a valuable food with medicinal and edible properties. Due to its high medicinal value and price, closely related Dendrobium varieties are often sold as imitations on the market. Therefore, there is an urgent need to develop new methods that can quickly identify Dendrobium stem and its closely related species. The Heracles Neo ultra-fast gas phase electronic nose was used in this study to determine and analyze the composition and contents of volatile organic compounds (VOCs) in Dendrobium stem and samples of five other species closely related to it. A total of 20 VOCs were identified, and a fingerprint map of the VOCs was constructed. Principal component analysis (PCA), Euclidean distance, and other methods were used to comprehensively process and analyze the obtained VOC information. The AroChemBase database was also used for qualitative analysis of the compounds. The results showed that there are significant differences in the odor fingerprint spectra of Dendrobium stem and the five other closely related species. The main types of compounds in Dendrobium stem and its five closely related species were organic esters, aldehydes, ketones, and olefins. Among them, 3-methylbutanal and n-butanol were characteristic compounds of the Dendrobium stem sample, while the VOCs acetonitrile and trometamol were present in the five related Dendrobium species samples. The Heracles Neo ultra-fast gas phase electronic nose can quickly and accurately identify Dendrobium stem and its five closely related species. It can also be used for the quality evaluation of Dendrobium stem, providing a theoretical reference for reducing the phenomenon of medicinal confusion in the Dendrobium stem market. Dendrobium loddigesii is a precious traditional Chinese medicine with high medicinal and ornamental value. However, the characterization of its mitochondrial genome is still pending. Here, we assembled the complete mitochondrial genome of D. loddigesii and discovered that its genome possessed a complex multi-chromosome structure. The mitogenome of D. loddigesii consisted of 17 circular subgenomes, ranging in size from 16,323 bp to 56,781 bp. The total length of the mitogenome was 513,356 bp, with a GC content of 43.41%. The mitogenome contained 70 genes, comprising 36 protein-coding genes (PCGs), 31 tRNA genes, and 3 rRNA genes. Furthermore, we detected 403 repeat sequences as well as identified 482 RNA-editing sites and 8154 codons across all PCGs. Following the sequence similarity analysis, 27 fragments exhibiting homology to both the mitogenome and chloroplast genome were discovered, accounting for 9.86% mitogenome of D. loddigesii. Synteny analysis revealed numerous sequence rearrangements in D. loddigesii and the mitogenomes of related species. Phylogenetic analysis strongly supported that D. loddigesii and D. Amplum formed a single clade with 100% bootstrap support. The outcomes will significantly augment the orchid mitochondrial genome database, offering profound insights into Dendrobium's intricate mitochondrial genome architecture. [ABSTRACT FROM AUTHOR]

The purpose of this research was to study the growth and bioactive compounds of one-year-old Dendrobium friedericksianum Rchb.f. cultured under different LEDs light conditions. The pseudobulb of orchid extracts was used to study edible coatings. No significant differences of plant height and diameter were found when culturing under different LEDs light conditions for 6 months. The analysis of bioactive compounds from the pseudobulb of Dendrobium friedericksianum Rchb.f. incubated under different light conditions for 7 months found that the IC value of extract of 50 orchid grown under purple light was statistically significantly lower than that of other light colors, corresponding to the total phenolic content of orchid extracts and the total flavonoid content from the orchids extracts from the purple light cultured. The results for culturing orchids under the LEDs to control the required amount of active ingredient were generally better compared to the natural planting. Edible coatings were studied from orchid extracts with the objective of finding ways to delay changes in quality and to extend the shelf life of Kluai Khai and lime. It was found that there were no significant differences for the application of coating agent to delay the color change of Kluai Khai and lime, and delay changes in firmness, sweetness, pH value and the acid content of Kluai Khai. Dendrobium is the second largest genus of Orchidaceae, distributed in southern and southwestern China. The number of Dendrobium is large, but some taxa have not been discovered. During the investigation of orchid resources in Xizang, two Dendrobium species were discovered, which were identified as D. macraei Lindl. and D. ritaeanum King & Pantl. Both were recorded for the first time in China. The results are as follows: D. macraei is similar to D. plicatile, with the difference that the edge of the middle lobe of lip is entire, apex concave; D. ritaeanum is similar to D. angustifolium, the flowers of the former are very small, and sepals and petals do not have purple stripes, and the lip is triangular up to 1.4 cm in length. The detailed morphological characteristics and photographs of the two species are provided. The specimens are stored in the Herbarium of Xizang Agricultural and Animal Husbandry University. This discovery enriches the local data and the potential medicinal plant resource reserve of orchids in China, and is of great significance to the biodiversity study of orchids and the investigation of medicinal orchid resources.

BACKGROUND RESULTS CONCLUSION Dendrobium officinale flos (DOF), a novel food raw material, is used in Chinese folk medicine to nourish the stomach. However, there is still no available study to evaluate the effects of DOF on animal models of acute gastric injury and its mechanism by modern pharmacological research.Herein, we characterized the major components of an aqueous extract of DOF and assessed its potential ameliorative effects in a rat model of acute gastric mucosal injury. The DOF water extract showed significant protective effects on the gastric mucosa and exhibited excellent antioxidant and anti‐inflammatory activities. Acute gastric injury rat models induced by ethanol (6 mL kg−1) were pretreated with different doses of DOF water extract (50–100 mg kg−1 day−1), and the biological effects of DOF extract in gastric tissues were evaluated. DOF extract alleviated the symptoms of ethanol‐stimulated acute gastric mucosal injury, as evidenced by a significant reduction in gastric injury index and the degree of gastric pathological changes. Additionally, treatment with DOF extract upregulated mucin expression in the gastric mucosa, attenuated oxidative stress, decreased the release of inflammatory mediators (TNF‐α, IL‐6), suppressed the expression of key proinflammatory enzymes (COX‐2 and iNOS), reduced the phosphorylation of p38 MAPK and p65 NF‐κB and increased the level of PGE2 in gastric tissues.DOF exerts protective effects against ethanol‐induced acute gastric mucosal injury, mainly by inhibiting inflammation and oxidative stress. © 2024 Society of Chemical Industry. Nonalcoholic fatty liver disease (NAFLD) is marked by hepatic steatosis accompanied by an inflammatory response. At present, there are no approved therapeutic agents for NAFLD. Dendrobium Huoshanense polysaccharide (DHP), an active ingredient extracted from the stems of Dendrobium Huoshanense, and exerts a protective effect against liver injury. However, the therapeutic effects and mechanisms of action DHP against NAFLD remain unclear. DHP was extracted, characterized, and administered to mice in which NAFLD had been induced with a high-fat and high-fructose drinking (HFHF) diet. Our results showed that DHP used in this research exhibits the characteristic polysaccharide peak with a molecular weight of 179.935 kDa and is composed primarily of Man and Glc in a molar ratio of 68.97:31.03. DHP treatment greatly ameliorated NAFLD by significantly reducing lipid accumulation and the levels of liver function markers in HFHF-induced NAFLD mice, as evidenced by decreased serum levels of aspartate aminotransferase (AST), alanine aminotransferase (ALT), total cholesterol (TC) and total triglyceride (TG). Furthermore, DHP administration reduced hepatic steatosis, as shown by H&E and Oil red O staining. DHP also inhibited the Toll-like receptor 4 (TLR4)/nuclear factor- kappa B (NF-κB) signaling pathway expression, thereby reducing levels of hepatic proinflammatory cytokines. Besides, untargeted metabolomics further indicated that 49 metabolites were affected by DHP. These metabolites are strongly associated the metabolism of glycine, serine, threonine, nicotinate and nicotinamide, and arachidonic acid. In conclusion, DHP has a therapeutic effect against NAFLD, whose underlying mechanism may involve the modulation of TLR4/NF-κB, reduction of inflammation, and regulation of the metabolism of glycine, serine, threonine, nicotinate and nicotinamide metabolism, and arachidonic acid metabolism. The extract of Dendrobium huoshanense, a traditional Chinese medicinal and food homologous plant belonging to the family Orchidaceae, was previously reported to have hypoglycemic and antioxidant effects. In this study, the direct effects of polysaccharide (DHP) and non-polysaccharide (NDHP) components of D. huoshanense, as well as its water extract (DHWE) were compared with that of metformin (an antidiabetic drug) on the gut microbiota (collected from fecal flora) of rats with streptozotocin-induced type 1 diabetes (T1D) using an in vitro fermentation method. The results showed that DHWE, DHP, and NDHP reduced pH and increased bacterial proliferation and short-chain fatty acid (SCFA) content in fermentation broth. DHWE, DHP, NDHP and metformin promoted the production of acetic and propionic acid, acetic acid, propionic acid and butyric acid, and propionic acid, respectively. DHWE, DHP, and NDHP reduced the abundance of Proteobacteria (subdominant pathogenic bacteria) and increased the abundance of Firmicutes (dominant beneficial gut bacteria). NDHP also reduced the abundance of Bacteroidetes (beneficial and conditional pathogenic). Metformin increased the abundance of Proteobacteria and reduced the abundance of Firmicutes and Bacteroidetes. At the genus level, NDHP promoted the proliferation of Megamonas and Megasphaera and decreased harmful bacteria (e.g., Klebsiella), and DHP increased the abundance of Prevotellaceae (opportunistic and usually harmless). By contrast, metformin increased the abundance of harmful bacteria (e.g., Citrobacter) and reduced the abundance of beneficial bacteria (e.g., Oscillospira). Dendrobium officinale Kimura & Migo is a perennial herbaceous plant of the genus Dendrobium in the family of Orchidaceae with high medicinal value. Melatonin (MT) is an indole-like tryptamine with functions such as regulating plant growth and development. This experiment investigated the effects of different concentrations of MT on the growth and development of protocorms of D. officinale protocorm-like bodies (PLBs). The results showed that the changes in morphological indicators such as color, cluster size, and surface changes were more significant under 75 µM MT than those of 0 µM (CK), and the appearance of white on the PLB surface was expedited, which was more conducive to the proliferation of PLBs. MT treatment of 100 µM inhibited the differentiation of adventitious buds, and the contents of photosynthetic pigments, polysaccharides, and flavonoids were significantly increased. Moreover, as compared with CK, the activities of superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) in PLBs increased significantly, while the content of malondialdehyde (MDA) decreased gradually with 75 µM or less. In conclusion, a concentration of 75 µM melatonin can enhance the rapid propagation rate of D. officinale in vitro, providing insights into the effects of melatonin on the growth of tissue-cultured D. officinale seedlings. Determination of D. crumenatum mutant obtained from in vitro mutation breeding needs a long time due to its long-life cycle. SSR molecular markers can be used for early mutant detection. Specific SSR markers developed in D. crumenatum are not yet available. Alternative published SSR markers were developed from D. catenatum. The aims of this study are to screen the most informative SSR markers generated from D. catenatum tested in irradiated D. crumenatum population and to determine the gamma irradiation dose resulting the most mutants. Ten SSR markers were randomly selected and tested in 25 individuals of D. crumenatum plantlets irradiated with several doses (0, 5, 10, 15, and 20 Gy; 5 plantlets each dose). The result showed 7 of 10 primers were polymorphic and the other three were monomorphic. All seven polymorphic primers can be used to identified intraspecific variation in the D. crumenatum mutant population. Markers dnsr28 and dnsr98 were the most informative, with the highest polymorphic information content (PIC) value of 0.5. Irradiation D. crumenatum protocorms using 10-15 gray doses were detected as the highest mutant percentage obtained up to 100% in the sample tested. This resulting marker information can be used to screen wider mutant population to decrease the non-mutant individuals in the population for maintenance and cost efficiency. This study assessed the phenolic and flavonoid contents, and antioxidant, antibacterial, as well as cytotoxic properties of the protocorm extract of Dendrobium transparens and compared it to its wild equivalents. Methanol was used to extract compounds from the stems (DTSE) and protocorms (DTPE). DTSE contained 61.889 mg flavonoid and 82.00 mg phenolic content per gram extract equivalent to quercetin and gallic acid, respectively. At a concentration of 191.23 µg/mL, DTSE exhibited a 50% DPPH radical scavenging efficiency. Compared to the 3T3 cell line (2108.87 µg/mL), the DTPE's cytotoxic ability against the HeLa (229.30 µg/mL) and U251 (213.90 µg/mL) cell lines was found to be significantly stronger. However, the U251 cell line was strongly cytotoxic to DTSE (75.84 µg/mL). At a dose of 2000 mg/kg, neither DTSE nor DTPE caused any discernible harm in mice. They could inhibit the growth of Acinetobacter baumannii, Escherichia coli, Klebsiella pneumoniae, Pseudomonas aeruginosa, and Staphylococcus aureus. Dendrobium officinale Kimura & Migo in the genus Dendrobium of Orchidaceae is an important medicinal plant that produces various bibenzyl and phenanthrene derivatives. In some orchids, these derivatives have been reported to increase with fungal infection. Bibenzyl biosynthesis is regulated by bibenzyl synthase (BBS). Although six genes of the BBS family have been registered from D. officinale, their gene regulation mechanisms are unclear. The infection of Dendrobium with mycorrhizal fungi also reportedly increases the expression of genes involved in biosynthesis; however, the effect of mycorrhizal fungi on bibenzyl production is unknown. The present study examined the effects of three mycorrhizal fungi isolated from D. officinale on BBS gene expression and bibenzyl production over time. One of the Tulasnellaceae operational taxonomic units induced BBS gene expression and increased two representative bibenzyls, gigantol and dendrophenol, at specific time points. Furthermore, 19 BBS sequences were cloned from 12 Dendrobium species, and a phylogenetic analysis was performed. The results indicated that repeated BBS gene duplication occurred during the evolution of the genus, and further duplication occurred after speciation. Dendrobium officinale (D. officinale), often used as a dual-use plant with herbal medicine and food applications, has attracted considerable attention for health-benefiting components and wide economic value. The antioxidant ability of D. officinale is of great significance to ensure its health care value and safeguard consumers' interests. However, the common analytical methods for evaluating the antioxidant ability of D. officinale are time-consuming, laborious, and costly. In this study, near-infrared (NIR) spectroscopy and chemometrics were employed to establish a rapid and accurate method for the determination of 2,2′-azinobis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) scavenging capacity, 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging capacity, and ferric reducing antioxidant power (FRAP) in D. officinale. The quantitative models were developed based on the partial least squares (PLS) algorithm. Two wavelength selection methods, namely the genetic algorithm (GA) and competitive adaptive reweighted sampling (CARS) method, were used for model optimization. The CARS-PLS models exhibited superior predictive performance compared to other PLS models. The root mean square errors of cross-validation (RMSECVs) for ABTS, FRAP, and DPPH were 0.44%, 2.64 μmol/L, and 2.06%, respectively. The results demonstrated the potential application of NIR spectroscopy combined with the CARS-PLS model for the rapid prediction of antioxidant activity in D. officinale. Dendrobium, a highly effective traditional Chinese medicinal herb, exhibits significant variations in efficacy and price among different varieties. Therefore, achieving an efficient classification of Dendrobium is crucial. However, most of the existing identification methods for Dendrobium make it difficult to simultaneously achieve both non-destructiveness and high efficiency, making it challenging to truly meet the needs of industrial production. In this study, we combined Laser-Induced Breakdown Spectroscopy (LIBS) with multivariate models to classify 10 varieties of Dendrobium. LIBS spectral data for each Dendrobium variety were collected from three circular medicinal blocks. During the data analysis phase, multivariate models to classify different Dendrobium varieties first preprocess the LIBS spectral data using Gaussian filtering and stacked correlation coefficient feature selection. Subsequently, the constructed fusion model is utilized for classification. The results demonstrate that the classification accuracy of 10 Dendrobium varieties reached 100%. Compared to Support Vector Machine (SVM), Random Forest (RF), and K-Nearest Neighbors (KNN), our method improved classification accuracy by 14%, 20%, and 20%, respectively. Additionally, it outperforms three models (SVM, RF, and KNN) with added Principal Component Analysis (PCA) by 10%, 10%, and 17%. This fully validates the excellent performance of our classification method. Finally, visualization analysis of the entire research process based on t-distributed Stochastic Neighbor Embedding (t-SNE) technology further enhances the interpretability of the model. This study, by combining LIBS and machine learning technologies, achieves efficient classification of Dendrobium, providing a feasible solution for the identification of Dendrobium and even traditional Chinese medicinal herbs. Polysaccharides from Dendrobium officinale have garnered attention for their diverse and well-documented biological activities. In this study, we isolated three ethanol-fractionated polysaccharides from Dendrobium officinale (EPDO) and investigated their digestive properties and effects on gut microbiota regulation in vitro. The results indicated that after simulating digestion in saliva, gastric, and small intestinal fluids, three EPDOs, EPDO-40, EPDO-60 and EPDO-80, with molecular weights (Mw) of 442.6, 268.3 and 50.8 kDa, respectively, could reach the large intestine with a retention rate exceeding 95%. During in vitro fermentation, the EPDOs were broken down in a "melting" manner, resulting in a decrease in their Mw. EPDO-60 degraded more rapidly than EPDO-40, likely due to its moderate Mw. After 24 h, the total production of short-chain fatty acids (SCFAs) for EPDO-60 reached 51.2 ± 1.9 mmol/L, which was higher than that of EPDO-80. Additionally, there was an increase in the relative abundance of Bacteroides, which are capable of metabolizing polysaccharides. EPDO-60 also promoted the growth of specific microbiota, including Prevotella 9 and Parabacteroides, which could potentially benefit from these polysaccharides. Most notably, by comparing the gut microbiota produced by different fermentation carbon sources, we identified the eight most differential gut microbiota specialized in polysaccharide metabolism at the genus level. Functional prediction of these eight differential genera suggested roles in controlling replication and repair, regulating metabolism, and managing genetic information transmission. This provides a new reference for elucidating the specific mechanisms by which EPDOs influence the human body. These findings offer new evidence to explain how EPDOs differ in their digestive properties and contribute to the establishment of a healthy gut microbiota environment in the human body. Dendrobium officinale (D. officinale) as a well-known herbal and functional food material is often adulterated with lower-price Dendrobium species. In this study, we aimed to develop an integrated method of nuclear magnetic resonance spectroscopy and artificial neural network (NMR-ANN) to identify and quantify the adulteration of D. officinale powder with other cheaper species. Microwave-assisted water extraction was selected as a time-saving and green method for sample preparation. The results indicate that the NMR-ANN method effectively quantified the adulteration rate of D. officinale powder with the root mean squared error (RMSE) of 4.92, mean absolute error (MAE) of 3.56 and coefficient of determination (R2) of 0.98 in the model test phase and with the RMSE of 7.65, MAE of 6.30 and R2 of 0.98 in the double-blinded test phase. The whole evaluation process can be done in 6 min and 5 s. ZF-HD transcription factors, which are unique to land plants, are involved in the regulation of abiotic stress response and related signaling pathways, and play a crucial role in plant growth and development. Dendrobium is one of the largest genera of orchids, with a high ornamental and ecological value. However, the specific functions of the ZF-HDs in Dendrobium remain unknown. In this study, we identified a total of 53 ZF-HDs from D. chrysotoxum (17), D. catenatum (23), and D. huoshanense (13), and analyzed their physicochemical properties, phylogenetic relationships, chromosomal locations, protein structures, conserved motifs, and expression patterns. The phylogenetic relationships revealed that 53 ZF-HDs were classified into six subfamilies (ZHDI–V and MIF), and all ZF-HD proteins contained motif 1 and motif 4 conserved domains, while a minority of these proteins had exons. The analysis of cis-elements in the promoters of ZF-HDs from three Dendrobium species showed that growth- and development-related elements were the most prevalent, followed by hormone response and abiotic stress response elements. Through collinearity analysis, 14 DchZF-HDs were found to be collinear with DhuZF-HDs, and 12 DchZF-HDs were found to be collinear with DcaZF-HDs. Furthermore, RT-qPCR analysis revealed that DchZF-HDs play a regulatory role in the development of lateral organs during the flowering process. The results indicated that DchZHD2 plays a role in the unpigmented bud stage, while DchMIF8 and DchZHD16 play significant roles during the pigmented bud and initial bloom stages. The ARF gene family is a representative transcription factor that plays a crucial role in the regulation of various growth and development processes in plants. Although the ARF gene family has been identified in five Orchidaceae species, limited research has been conducted on the ARF gene within Dendrobium. To explore ARF family genes in different Dendrobium species, we selected chromosome-level genomic data from D. nobile, D. chrysotoxum, and D. huoshanense for genome-wide identification, and to analyze expression patterns in the D. nobile flower. In this study, 13, 18, and 23 ARF genes were identified in the genomes of D. chrysotoxum, D. huoshanense, and D. nobile, respectively. These genes were then subsequently classified into four classes (Classes I, II, III, and IV) based on our phylogenetic analysis. Additional protein sequence analysis found that 30 ARF proteins with three classically conserved structural domains (BDB, MR, and RD) were present in the three Dendrobium species. Our gene structure comparative analysis also found the same evolutionary branch with similar intron-exon structural features. Specifically, Class I and Class III display longer introns that potentially constitute a distinctive characteristic of Dendrobium. Gene expression patterns analysis showed the potential involvement of DnoARF 5, 7, 10, 11, 12, 13, 16, 17, and 23 in initial differentiation and flower bud formation. Likewise, transcriptomic analysis and RT-qPCR expression profiles indicated flower-specific expression patterns for four ARF genes (DnoARF5, DnoARF6, DnoARF16, and DnoARF22), which suggest an important impact on flower development and regulation. Ultimately, this study provides comprehensive data to elucidate the potential functions of ARF genes in three Dendrobium species and suggests new insights for further exploration concerning the function and regulatory mechanisms in Dendrobium during flower development. This study aimed to explore the inhibitory effect and mechanism of the total alkaloids of Dendrobium officinale Kimura et Migo (DENA) against cholesterol esterase (CE). DENA was characterised by SEM, 1H NMR, and X-ray diffraction (XRD). The inhibitory effect and mechanism of DENA against CE were investigated through fluorescence chromatography, circular dichroism, and molecular docking. DENA inhibited CE activity (IC50 = 1.08 ± 0.09 mg mL−1), characterised by a non-competitive inhibition mechanism. Furthermore, DENA induced a fluorescence quenching in CE, causing a blue shift in the λmax. This coincided with a transition in the secondary structure of CE from a layered to a helical structure by circular dichroism, indicating a significant reduction in its stability. Moreover, molecular docking confirmed that DENA binds to amino acid residues in the enzyme through hydrogen bonds and hydrophobic interactions, leading to structural changes and reduced enzyme activity. Orchids are crucial for the horticulture industry. Mycorrhizal fungi benefit crops by improving nutrition, plant growth, and disease resistance. However, the mycorrhizal association of horticultural hybrid orchids is poorly understood. To address this, we investigated mycorrhizal colonization in the entire root system and assessed the mycorrhizal community using a Dendrobium cultivar, D. Stardust 'Firebird', obtained from three nurseries. Additionally, we isolated and tested mycorrhizal fungi in symbiotic culture to assess their role in the seed germination and growth of Dendrobium species. All plants were colonized by mycorrhizal fungi, with a higher colonization rate in mature than in juvenile plants. Molecular identification of mycorrhizal fungi by Sanger and high-throughput sequencing revealed that the cultivar was associated with a phylogenetically diverse group of fungi, including mycorrhizal fungi from Tulasnellaceae, and several wood-decaying fungi. The Tulasnellaceae isolates significantly enhanced the seed germination of three Dendrobium species and increased the survival rate and growth of asymbiotic seedlings of D. moniliforme. Dendrobium officinale powder is rich in polysaccharies and has special rheological properties, which can be used to develop starch-based 3D printing food materials. In this study, the effect of Dendrobium officinale powder on the adaptability of starch-based 3D printing gel (SG) was investigated. The structure and characteristics of starch-based gel 3D printing materials were analyzed by rheology, texture, scanning electron microscopy (SEM), x-ray diffraction, and fourier transform infrared spectroscopy (FT-IR) combined with Computational Fluid Dynamics (CFD) simulation. The results showed that the addition of Dendrobium officinale powder had a significant effect on the microstructure, rheological and textural properties of starch-based gels. With the addition of Dendrobium officinale from 0 g (SG) to 1.00 g (SG-1.00D), the hardness of starch-based gel increased from (59.81 ±1.39) g to (105.67±2.05) g, and the adhesion increased from (43.40±2.07) g-s to (78.59±0.23) g-s. It was found that addition of 0.15 g Dendrobium officinale powder can optimize the network structure of the gel system. The addition of Dendrobium can cause the migration of FT-IR characteristic peaks, and enhance the intensity of XRD diffraction characteristic marker peaks near 17°, indicating that stronger hydrogen bonds are generated, but no new substances are formed. The printing speed of the SG-0.15D could reach 6.13x10-2 1/s with best 3D printing adaptability. This research provides a theoretical basis and direction for the development of using Dendrobium officinale powder in 3D food printing materials, and providestechnical reference and support for the actual production of Dendrobium officinale 3D printing materials. Background: Dendrobium officinale Kimura et Migo, a renowned traditional Chinese orchid herb esteemed for its significant horticultural and medicinal value, thrives in adverse habitats and contends with various abiotic or biotic stresses. Acid invertases (AINV) are widely considered enzymes involved in regulating sucrose metabolism and have been revealed to participate in plant responses to environmental stress. Although members of AINV gene family have been identified and characterized in multiple plant genomes, detailed information regarding this gene family and its expression patterns remains unknown in D. officinale, despite their significance in polysaccharide biosynthesis. Results: This study systematically analyzed the D. officinale genome and identified four DoAINV genes, which were classified into two subfamilies based on subcellular prediction and phylogenetic analysis. Comparison of gene structures and conserved motifs in DoAINV genes indicated a high-level conservation during their evolution history. The conserved amino acids and domains of DoAINV proteins were identified as pivotal for their functional roles. Additionally, cis-elements associated with responses to abiotic and biotic stress were found to be the most prevalent motif in all DoAINV genes, indicating their responsiveness to stress. Furthermore, bioinformatics analysis of transcriptome data, validated by quantitative real-time reverse transcription PCR (qRT-PCR), revealed distinct organ-specific expression patterns of DoAINV genes across various tissues and in response to abiotic stress. Examination of soluble sugar content and interaction networks provided insights into stress release and sucrose metabolism. Conclusions: DoAINV genes are implicated in various activities including growth and development, stress response, and polysaccharide biosynthesis. Dendrobium nobile is an important orchid plant that has been used as a traditional herb for many years. For the further pharmaceutical development of this resource, a combined transcriptome and metabolome analysis was performed in different parts of D. nobile. First, saccharides, organic acids, amino acids and their derivatives, and alkaloids were the main substances identified in D. nobile. Amino acids and their derivatives and flavonoids accumulated strongly in flowers; saccharides and phenols accumulated strongly in flowers and fruits; alkaloids accumulated strongly in leaves and flowers; and a nucleotide and its derivatives and organic acids accumulated strongly in leaves, flowers, and fruits. Simultaneously, genes for lipid metabolism, terpenoid biosynthesis, and alkaloid biosynthesis were highly expressed in the flowers; genes for phenylpropanoids biosynthesis and flavonoid biosynthesis were highly expressed in the roots; and genes for other metabolisms were highly expressed in the leaves. Furthermore, different members of metabolic enzyme families like cytochrome P450 and 4-coumarate-coA ligase showed differential effects on tissue-specific metabolic accumulation. Members of transcription factor families like AP2-EREBP, bHLH, NAC, MADS, and MYB participated widely in differential accumulation. ATP-binding cassette transporters and some other transporters also showed positive effects on tissue-specific metabolic accumulation. These results systematically elucidated the molecular mechanism of differential accumulation in different parts of D. nobile and enriched the library of specialized metabolic products and promising candidate genes. The WUSCHEL-related homeobox (WOX) transcription factor plays a vital role in stem cell maintenance and organ morphogenesis, which are essential processes for plant growth and development. Dendrobium chrysotoxum, D. huoshanense, and D. nobile are valued for their ornamental and medicinal properties. However, the specific functions of the WOX gene family in Dendrobium species are not well understood. In our study, a total of 30 WOX genes were present in the genomes of the three Dendrobium species (nine DchWOXs, 11 DhuWOXs, and ten DnoWOXs). These 30 WOXs were clustered into ancient clades, intermediate clades, and WUS/modern clades. All 30 WOXs contained a conserved homeodomain, and the conserved motifs and gene structures were similar among WOXs belonging to the same branch. D. chrysotoxum and D. huoshanense had one pair of fragment duplication genes and one pair of tandem duplication genes, respectively; D. nobile had two pairs of fragment duplication genes. The cis-acting regulatory elements (CREs) in the WOX promoter region were mainly enriched in the light response, stress response, and plant growth and development regulation. The expression pattern and RT-qPCR analysis revealed that the WOXs were involved in regulating the floral organ development of D. chrysotoxum. Among them, the high expression of DchWOX3 suggests that it might be involved in controlling lip development, whereas DchWOX5 might be involved in controlling ovary development. In conclusion, this work lays the groundwork for an in-depth investigation into the functions of WOX genes and their regulatory role in Dendrobium species' floral organ development. Dendrobium officinale is a valuable traditional Chinese herbal plant that is both medicinal and edible. However, the yield of wild Dendrobium officinale is limited. Adverse stress affects the growth, development, and yield of plants, among which low temperature is the primary limiting factor for introducing Dendrobium officinale to high-latitude areas and expanding the planting area. Therefore, this study aims to explore the variations in growth ability, cold resistance, and contents of bioactive compounds among different Dendrobium officinale strains. Four strains of Dendrobium officinale were selected as experimental materials and were subjected to low-temperature stress (4 °C). The agronomic traits, physiological indices, as well as the expressions of cold resistance-related genes (HSP70, DcPP2C5, DoCDPK1, and DoCDPK6) in the roots and leaves of Dendrobium officinale, were determined. The contents of bioactive compounds, including polysaccharides, flavonoids, and phenols were also measured. Compared with the other strains, Xianju had the highest seed germination and transplantation-related survival rates. Under low-temperature stress, Xianju exhibited the strongest cold resistance ability, as revealed by the changes in water contents, chlorophyll levels, electrical conductivities, enzyme activities, and expressions of the cold resistance-related genes. Additionally, the polysaccharide content of Xianju increased the most, while the stem flavonoid and leaf phenol contents were elevated in all four strains under cold treatment. Therefore, selecting excellent performing strains is expected to expand the planting area, improve the yield, and increase the economic benefits of Dendrobium officinale in high latitude areas with lower temperatures. Introduction: DNA-binding with one finger (Dof) transcription factors (TFs) are a unique family of TFs found in higher plants that regulate plant responses to light, hormones, and abiotic stresses. The specific involvement of Dof genes in the response to environmental stresses remains unknown in D. huoshanense. Methods: A total of 22 Dof family genes were identified from the D. huoshanense genome. Results: Chromosome location analysis showed that DhDof genes were distributed on 12 chromosomes, with the largest number of Dof genes located on chromosome 8. The phylogenetic tree revealed that DhDofs could be categorized into 11 distinct subgroups. In addition to the common groups, DhDof4, DhDof5, DhDof17, and the AtDof1.4 ortholog were clustered into the B3 subgroup. Group E was a newly identified branch, among which DhDof6, DhDof7, DhDof8, and DhDof9 were in an independent branch. The conserved motifs and gene structure revealed the differences in motif number and composition of DhDofs. The dof domain near the N-terminus was highly conserved and contained a C2-C2-type zinc finger structure linked with four cysteines. Microsynteny and interspecies collinearity revealed gene duplication events and phylogenetic tree among DhDofs. Large-scale gene duplication had not occurred among the DhDofs genes and only in one pair of genes on chromosome 13. Synteny blocks were found more often between D. huoshanense and its relatives and less often between Oryza sativa and Arabidopsis thaliana. Selection pressure analysis indicated that DhDof genes were subject to purifying selection. Expression profiles and correlation analyses revealed that the Dof gene under hormone treatments showed several different expression patterns. DhDof20 and DhDof21 had the highest expression levels and were co-expressed under MeJA induction. The cis-acting element analysis revealed that each DhDof had several regulatory elements involved in plant growth as well as abiotic stresses. qRT-PCR analysis demonstrated that DhDof2 was the main ABA-responsive gene and DhDof7 was the main cold stress-related gene. IAA suppressed the expression of some Dof candidates, and SA inhibited most of the candidate genes. Background: It has been reported that ferroptosis participates in the pathophysiological mechanism of spinal cord injury (SCI). Our preliminary experiments verified that dendrobium nobile polysaccharide (DNP) improved the behavioral function of SCI rats. Therefore, the purpose of this study was to examine the role of DNP on ferroptosis and its neuroprotective mechanism in SCI rats. Methods: Adult female sprague dawley (SD) rats were exposed to SCI by Allen's method, followed by an intragastric injection of 100 mg/kg DNP per day for 2 weeks. Behavioral features were verified by the Basso-Beattie-Bresnahan (BBB) scale and footprint evaluation. Iron content and glutathione (GSH) were assessed spectrophotometrically. Mitochondrial morphology was examined by transmission electron microscopy. The expression of ferroptosis-related genes, including System Xc-light chain (xCT), G-rich RNA sequence binding Factor 1 (GRSF1) and glutathione peroxidase 4 (Gpx4), was examined by real-time polymerase chain reaction (PCR) and western blot. The spinal cavity was defined using hematoxylin-eosin (HE) staining, and neuronal modifications were detected by immunofluorescence. Results: Compared with the SCI group, the BBB score of rats in the DNP group increased at 7 d, 14 d, 21 d, and 28 d. The differences between the two groups were statistically significant. At 12 h post-injury the iron content began to decrease. At 24 h post-injury the iron content decreased significantly in the DNP group. The morphological changes of the mitochondrial crest and membrane in the DNP group were ameliorated within 24 h. Compared with the sham group, the expression of xCT, GSH, Gpx4, and GRSF1 were significantly reduced after SCI. After DNP treatment, the expression of xCT, Gpx4, and GSH were higher. The tissue cavity area was significantly reduced and the amount of NeuN+ cells was increased in the DNP group at 14 d and 28 d after SCI. In the present study, Dendrobium officinale polysaccharides (DOPs) are incorporated into sodium alginate (SA) for the development of SA‐DOPs films. The physical, mechanical, and functional properties of SA‐DOPs composite films and their ability to preserve fresh grapes are investigated. The results reveal that the addition of DOPs can significantly enhance the thickness, swelling ratio, tensile strength, elongation at break, and thermal stability of the films (p < 0.05) whereas the water solubility and water vapor permeability are decreased. Fourier transform‐infrared (FT‐IR) spectroscopy results reflect interaction of chemical groups and bonds between SA and DOPs. The result of scanning of electron microscopy (SEM) indicates smooth for SA‐DOPs films while control film exhibits rough. The SA‐DOPs films exhibit potential antioxidant effects on 1,1‐diphenyl‐2‐picrylhydrazyl (DPPH) and 2,2'‐azino‐bis(3‐ethylbenzothiazoline‐6‐sulfonic acid) (ABTS) free radicals. Furthermore, the composite films exhibit remarkable inhibition of the growth of Escherichia coli and Staphylococcus aureus (p < 0.05). In addition, the SA‐DOPs coatings can effectively reduce weight loss, inhibit polyphenol oxidase activity, extend the shelf life of the grapes at room temperature (25 ± 1 °C) for 14 days. Simple Summary: This study investigated the effectiveness of soluble silicon fertilizer (SiO2) in protecting dendrobium orchids from thrips pest complexes and fungal pathogens to enhance yield and quality. Over a nine-month greenhouse trial in Hawaii, three dendrobium varieties ('Uniwai Supreme', 'Uniwai Mist', and 'Uniwai Royale') received alternating weekly applications of soluble silicon fertilizer (50 mg Si/plant/application) via either soil drench and/or foliar methods. The primary objectives were to assess the impact of soluble silicon fertilizer on marketable yield, spray length, thrips and fungal damage, and plant vigor. The results revealed significant benefits of silicon fertilization across all varieties. 'Uniwai Supreme' exhibited the most substantial increase in marketable yield (73.0%) and 'Uniwai Mist' had the least increase in marketable yield, with a 50.6% increase compared to untreated controls. These findings have practical implications for orchid cultivation and pest management practices, demonstrating the potential of soluble silicon to improve orchid health and productivity by reducing pest damage. By mitigating the impact of thrips and fungal pathogens, silicon fertilization can contribute to more sustainable orchid production methods. Ultimately this research supports the adoption of silicon supplementation as a strategy to enhance Integrated Pest Management (IPM) tactics and promote environmentally friendly agriculture. The effects of soluble silicon fertilization on monocots and dicots have been widely studied. However, little is known regarding its effects on protecting epiphytes against insect and fungal pests. The efficacy of silicon fertilizer to reduce damage by thrips pest complexes, namely: Thrips palmi Karny, Frankliniella occidentalis Pergande, Chaetanaphothrips orchidii Moulton, and Chaetanaphothrips signipennis Bagnall (Thysanoptera: Thripidae), and the fungal pathogens: Botrytis cinerea Persoon (Helotiales: Sclerotiniaceae) and Fusarium spp. Link (Hypocreales: Nectriaceae) was examined during a nine-month greenhouse trial in Hawaii. The trial assessed yield, quality, and pest damage across three common varieties of dendrobiums. All replicates received additional soluble silicon fertilizer applications alternating weekly between soil drench and foliar (50 mg Si/plant) applications. Yield, quality, and spray length, pest damage, plant vigor, SPAD, and leaf temperature were measured. Data were analyzed using a generalized linear model (glm) with repeated measures followed by post-hoc pair-wise comparisons in R, version 4.3.1. Treatment effects were significant at p < 0.001 for the majority of the explanatory variables including: marketable yield, spray length, thrips damage, and fungal damage. Overall, the lavender variety ('Uniwai Supreme') benefited the most from silicon applications with a 73.0% increase in marketable yield, compared to the white variety ('Uniwai Mist'), which had an increase of 50.6% marketable sprays in contrast to its untreated control. Si benefits conferred to the purple variety ('Uniwai Royale') were intermediate to the lavender and white varieties. Background: Dendrobium officinale flowers (DOF) have the effects of antiaging and nourishing yin, but it lacks pharmacological research on skin aging. Objective: Confirming the role of DOF in delaying skin aging based on the "in vitro animal–human" model. Methods: In this experiment, three kinds of free radical scavenging experiments in vitro, D‐galactose‐induced aging mouse model, and human antiaging efficacy test were used to test whether DOF can improve skin aging through anti‐oxidation. Results: In vitro experiment shows that DOF has certain scavenging effect on 2,2‐diphenyl‐1‐picrylhydrazyl (DPPH) free radical, hydroxyl free radical, and superoxide free radical, and its IC50 is 0.2090 μg/mL, 15.020, and 1.217 mg/mL respectively. DOF can enhance the activities of T‐AOC, SOD, CAT, and GSH Px in the serum of aging mice, increase the content of GSH, and reduce the content of MDA when administered with DOF of 1.0, 2.0, and 4.0 g/kg for 6 weeks. In addition, it can enhance the activity of SOD in the skin of aging mice, increase the content of Hyp, and decrease the content of MDA, activated Keap1/Nrf2 pathway in the skin of aging mice. Applying DOF with a concentration of 0.2 g/mL on the face for 8 weeks can significantly improve the skin water score and elasticity value, reduce facial wrinkles, pores, acne, and UV spots, and improve the facial brown spots and roughness. Conclusion: DOF can significantly improve skin aging caused by oxidative stress, and its mechanism may be related to scavenging free radicals in the body and improving skin quality. Dendrobium nobile Lindl. polysaccharide (DNP1) showed good anti-inflammatory activity in our previous study. In this study, the structural characterization of DNP1 and its mode of action on TLR4 were investigated. Structural characterization suggested that DNP1 was a linear glucomannan composed of (1 → 4)-β-Manp and (1 → 4)-β-Glcp residues, and the acetyl group was linked to the C-2 of Manp. The possible repeating structural units of DNP1 were [→4)-2-OAc-β-Manp-(1→]3 →4)-β-Glcp-(1→. Surface plasmon resonance (SPR) binding test results showed that DNP1 did not bind directly to TLR4. The TLR4 and MD2 receptor blocking tests confirmed that DNP1 needs MD2 and TLR4 to participate in its anti-inflammatory effect. The binding energy of DNP1 to TLR4-MD2 was −7.9 kcal/mol, indicating that DNP1 could bind to the TLR4-MD2 complex stably. Therefore, it is concluded that DNP1 may play an immunomodulatory role by binding to the TLR4-MD2 complex and inhibiting the TLR4-MD2-mediated signaling pathway. [ABSTRACT FROM AUTHOR]

Blue light is the higher-energy region of the visible spectrum. Excessive exposure to blue light is known to induce oxidative stress and is harmful to the eyes. The stems of Dendrobium nobile Lindl. (Orchidaceae), named Jinchaishihu, have long been used in traditional Chinese medicine (TCM) for nourishing yin, clearing heat, and brightening the eyes. The polysaccharide is one of the major components in D. nobile. However, the effect on ocular cells remains unclear. This study aimed to investigate whether the polysaccharide from D. nobile can protect the eyes from blue light-induced injury. A crude (DN-P) and a partially purified polysaccharide (DN-PP) from D. nobile were evaluated for their protective effects on blue light-induced damage in ARPE-19 and 661W cells. The in vivo study investigated the electroretinographic response and the expression of phototransduction-related genes in the retinas of a Drosophila model. The results showed that DN-P and DN-PP could improve blue light-induced damage in ARPE-19 and 661W cells, including cell viability, antioxidant activity, reactive oxygen species (ROS)/superoxide production, and reverse opsin 3 protein expression in a concentration-dependent manner. The in vivo study indicated that DN-P could alleviate eye damage and reverse the expression of phototransduction-related genes, including ninaE, norpA, Gαq, Gβ76C, Gγ30A, TRP, and TRPL, in a dose-dependent manner in blue light-exposed Drosophila. In conclusion, this is the first report demonstrating that D. nobile polysaccharide pretreatment can protect retinal cells and retinal photoreceptors from blue light-induced damage. Background: The discovery of traditional plants' medicinal and nutritional properties has opened up new avenues for developing pharmaceutical and dietary strategies to prevent atherosclerosis. However, the effect of the antioxidant Dendrobium officinale polysaccharide (DOP) on atherosclerosis is still not elucidated. Purpose: This study aims to investigate the inhibitory effect and the potential mechanism of DOP on high-fat diet-induced atherosclerosis in Apolipoprotein E knockout (ApoE−/−) mice. Study design and methods: The identification of DOP was measured by high-performance gel permeation chromatography (HPLC) and Fourier transform infrared spectroscopy (FTIR). We used high-fat diet (HFD)-induced atherosclerosis in ApoE−/− mice as an animal model. In the DOP intervention stage, the DOP group was treated by gavage with 200 μL of 200 mg/kg DOP at regular times each day and continued for eight weeks. We detected changes in serum lipid profiles, inflammatory factors, anti-inflammatory factors, and antioxidant capacity to investigate the effect of the DOP on host metabolism. We also determined microbial composition using 16S rRNA gene sequencing to investigate whether the DOP could improve the structure of the gut microbiota in atherosclerotic mice. Results: DOP effectively inhibited histopathological deterioration in atherosclerotic mice and significantly reduced serum lipid levels, inflammatory factors, and malondialdehyde (F/B) production. Additionally, the levels of anti-inflammatory factors and the activity of antioxidant enzymes, including superoxide dismutase (SOD) and glutathione peroxidase (GSH-PX), were significantly increased after DOP intervention. Furthermore, we found that DOP restructures the gut microbiota composition by decreasing the Firmicutes/Bacteroidota (F/B) ratio. The Spearman's correlation analysis indicated that serum lipid profiles, antioxidant activity, and pro-/anti-inflammatory factors were associated with Firmicutes, Bacteroidota, Allobaculum, and Coriobacteriaceae_UCG-002. Thiacloprid (THI) is a neonicotinoid insecticide, and its wide‐ranging use has contributed to severe environmental and health problems. Dendrobium officinale polysaccharide (DOP) possesses multiple biological activities such as antioxidant and antiapoptosis effect. Although present research has shown that THI causes kidney injury, the exact molecular mechanism and treatment of THI‐induced kidney injury remain unclear. The study aimed to investigate if DOP could alleviate THI‐induced kidney injury and identify the potential molecular mechanism in quails. In this study, Japanese quails received DOP (200 mg/kg) daily with or without THI (4 mg/kg) exposure for 42 days. Our results showed that DOP improved hematological changes, biochemical indexes, and nephric histopathological changes induced by THI. Meanwhile, THI exposure caused oxidative stress, apoptosis, and autophagy. Furthermore, THI and DOP cotreatment significantly activated the nuclear factor erythroid 2‐related factor 2/heme oxygenase‐1 (Nrf2/HO‐1) pathway, restored antioxidant enzyme activity, and reduced apoptosis and autophagy in quail kidneys. In summary, our study demonstrated that DOP mitigated THI‐mediated kidney injury was associated with oxidative stress, apoptosis, and autophagy via activation of the Nrf2/HO‐1 signaling pathway in quails. Natural polysaccharides are important active biomolecules. However, the analysis and structural characterization of polysaccharides are challenging tasks that often require multiple techniques and maps to reflect their structural features. This study aimed to propose a new heart-cutting two-dimensional liquid chromatography (2D–LC) method for separating and analyzing polysaccharides to explore the multidimensional information of polysaccharide structure in a single map. That is, the first-dimension liquid chromatography (1D–LC) presents molecular-weight information, and the second-dimension liquid chromatography (2D–LC) shows the fingerprints of polysaccharides. In this 2D–LC system, the size-exclusion chromatography–hydrophilic interaction chromatography (SEC–HILIC) model was established. Coupling with a charged aerosol detector (CAD) eliminated the need for the derivatization of the polysaccharide sample, allowing the whole process to be completed within 80 min. The methods were all validated in terms of precision, linearity, stability, and repeatability. The capability of the new 2D-LC method was demonstrated in determining various species of natural polysaccharides.
