8. An integrated multidisciplinary approach to study the effects of copper and osmotic stress in fish

Abstract

Since many estuarine zones are impacted by copper contamination, there is an on-going effort to develop Biotic Ligand Models (BLMs) predicting copper toxicity in transitional environments. In the first stage of this project, a critical analysis of the BLM framework identified some aspects of the model that required further investigation. In particular, a BLM for estuaries needed (a) a better characterization of the dissolved organic matter (DOC) and its effect on copper availability, and (b) the inclusion in the model’s equation of a salinity-correction factor modulating the relationship between copper accumulation on the biotic ligand and toxicity. The first issue was addressed by modelling the data produced using a Chelex resin method to determine the labile fraction of copper in samples of mixed riverine and estuarine waters. A refined and simplified BLM equation was then presented, accounting for both the DOC characteristics and the relevance of the osmotic gradient in modulating the relationship between copper accumulation and toxicity. A critical analysis of the literature on copper toxicity and salinity led to the hypothesis that copper-exposed fish are more sensitive to osmotic stresses, as copper interferes with their osmoregulatory pathways. In particular, the cytosolic isoform-2 of the enzyme carbonic anhydrase (CA2) was identified as an osmotic effector protein targeted by copper and involved in osmotic stress response pathways, hence representing a mechanistic link between the combined effects of copper exposure and osmotic stress. To test this hypothesis, two in vivo studies were performed, using the euryhaline fish sheepshead minnow (Cyprinodon variegatus) and applying different rates of salinity changes as a way of dosing osmotic stress. The results showed a disturbance in plasma ion homeostasis after the salinity transitions, but notably the magnitude of the disturbance was greater in the copper-exposed individuals, suggesting a sensitizing effect of copper on the responses of fish to osmotic stress. Gene expression data demonstrated that CA2 is targeted by copper and confirmed the role of the enzyme in osmoregulatory pathways, as further supported by a promoter analysis of the gene coding for zebrafish CA2, which revealed the presence of osmotic-stress related elements. Overall, these results suggest that CA2 is an osmotic effector protein whose response can be activated by a medium level of osmotic stress through a combination of transcriptional and post-translational control circuits.

Abstract

Background In February 2021 the “Cortina2021” Alpine World Ski Championships took place in the mountain town of Cortina d'Ampezzo (Italy), thus gaining the definition of “first sporting event of global significance in the time of covid-19 pandemic”. Though the event took place behind closed doors and in a context of national public health restrictive measures, it counted almost 6,000 participants. Methods A rigorous “Covid-19 Prevention and Hygiene Protocol” was drawn up by the local and regional health authorities together with the organizing committee, in order to contain the risk of viral circulation within the venue. Results Overall about 20,000 rapid antigen nasal swabs were performed to screen-detect sars-cov2 cases: only 22 of these were found positive, whereas 25 people were quarantined as close contacts. Conclusions The experience of “Cortina2021” proves that mass gatherings can take place even in times of epidemic threats, if adequately planned and by providing public health recommendations tailored on the viral circulation foreseen by the time the event is due. If new epidemics should arise in the future, hopefully the public health management of “Cortina2021” may be a template for next mass gatherings to be held, as far as possible, despite the viral circulation.

Abstract

In March 2020 the covid-19 pandemic led to the abruption of most of the routine outpatient activities in the Italian hospitals and Prevention Departments, including those vaccinations which were not urgent and/or scheduled for the age 0-6. As soon as a milder phase of the pandemic made it possible, since June 2020, in the mountain territory of the province of Belluno (Veneto, North-East of Italy), in the Alps, 12.152 doses of vaccine against tick-borne encephalitis have been administered by means of the innovative “drive-through” modality. In this context of hazardous delays, the Prevention Department of Belluno (Italy) tested its resilience: a simple and effective method of taking charge was implemented, by initially phoning to the positive case and imposing the isolation measure on him or her and later on proceeding with the conventional contact tracing. Abstract

Background: Non‐small cell lung cancer (NSCLC) is the first cause of cancer‐related death among men and the second among women worldwide. It also poses an economic threat to the sustainability of healthcare services. This study estimated the direct costs of care for patients with NSCLC by stage at diagnosis, and management phase of pathway recommended in local and international guidelines. Methods: Based on the most up‐to‐date guidelines, we developed a very detailed "whole‐disease" model listing the probabilities of all potentially necessary diagnostic and therapeutic actions involved in the management of each stage of NSCLC. We assigned a cost to each procedure, and obtained an estimate of the total and average per‐patient costs of each stage of the disease and phase of its management. Results: The mean expected cost of a patient with NSCLC is 21,328 € (95% C.I. −20 897−22 322). This cost is 16 291 € in stage I, 19530 € in stage II, 21938 € in stage III, 22175 € in stage IV, and 28 711 € for a Pancoast tumor. In the early stages of the disease, the main cost is incurred by surgery, whereas in the more advanced stages radiotherapy, medical therapy, treatment for progressions, and supportive care become variously more important. Conclusions: An estimation of the direct costs of care for NSCLC is fundamental in order to predict the burden of new oncological therapies and treatments on healthcare services, and thus orient the decisions of policy‐makers regarding the allocation of resources. Abstract

Deregulation of the tyrosine kinase signalling is often associated with tumour progression and drug resistance, but its underlying mechanisms are only partly understood. In this study, we investigated the effects of the receptor tyrosine kinase AXL on the stability of the MDMX-MDM2 heterocomplex and the activity of p53 in melanoma cells. Our data demonstrated that AXL overexpression or activation through growth arrest-specific 6 (Gas6) ligand stimulation increases MDMX and MDM2 protein levels and decreases p53 activity. Upon activation, AXL stabilizes MDMX through a post-translational modification that involves phosphorylation of MDMX on the phosphosite Ser314, leading to increased affinity between MDMX and MDM2 and favouring MDMX nuclear translocation. Ser314 phosphorylation can also protect MDMX from MDM2-mediated degradation, leading to stabilization of the MDMX-MDM2 complex. We identified CDK4/6 and p38 MAPK as the two kinases mediating AXL-induced modulation of the MDMX-MDM2 complex, and demonstrated that suppression of AXL, either through siRNA silencing or pharmacological inhibition, increases expression levels of p53 target genes P21, MDM2, and PUMA, improves p53 pathway response to chemotherapy, and sensitizes cells to both Cisplatin and Vemurafenib. Abstract

Glyphosate is the best-selling herbicide in the world and in 2015 the International Agency for Research on Cancer listed it among the "probable carcinogens for humans", opening a scientific debate that is still going on. On these premises, in 2016, extraordinary samplings of glyphosate, its metabolite AMPA, and a similar compound, ammonium glufosinate, were carried out in 12 wells of the water network intended for domestic consumption in the territory of the Local Health Unit 2 - District of Pieve di Soligo, Province of Treviso, Veneto region, Italy. The area includes 13 municipalities at high-intensity "Prosecco d.o.c.g." wine production. Traces of glyphosate (maximum reached 0.08 µg/L) and AMPA (maximum reached 0.25 µg/L, beyond the legal limit of 0.1 µg/L for drinking water) were detected in 2 wells supplying an urban area. No samples contained traces of ammonium glufosinate. Following these findings, an inter-municipal order to suspend the use of glyphosate was introduced and then entered definitively in the rural police regulation concerning all the municipalities in the Prosecco d.o.c.g. area, which led to the elimination of glyphosate and AMPA also in the initially contaminated wells. The case shows that high-consumption herbicides can reach the drinking water network of a city surrounded by territories with high agricultural activity. Moreover, the combined intervention of the institutions was fundamental to eliminate a "probable carcinogen" from the urban drinking water and to promote the abandonment of potentially harmful agricultural practices in favor of solutions with reduced environmental and health impact.
