The present study delved into the chemical composition, antioxidant, and anti-inflammatory properties of three dry edible beans: Black (BL), Great Northern (GN), and Pinto (PN). The beans were soaked, cooked, and subjected to in vitro gastrointestinal (GI) digestion. BL bean exhibited significantly higher gastric (42%) and intestinal (8%) digestion rates. Comparative assessment of soluble GI-digested fractions (<3 kDa) revealed that the GN bean exhibited the highest abundance of dipeptides ( P < 0.05). The BL bean fraction displayed a 4-fold increase in tripeptides ( P < 0.05). Both BL and PN bean fractions are high in essential free amino acids, flavonols, and derivatives of hydroxybenzoic acid when compared to the GN bean. All the beans exhibited the ability to mitigate TNF-α-induced pro-inflammatory signaling; however, the BL bean fraction was the most effective at lowering AAPH-induced oxidative stress in HT-29 cells, followed by the GN bean ( P < 0.05). In contrast, a low antioxidant effect was observed with PN beans.

Gymnema sylvestre is a commonly used herb in Ayurvedic medicine. The demand for its extracts in the commercial and pharmaceutical fields has been steadily increasing in recent years. Its extracts are used to treat various ailments as well as for their antimicrobial properties. This study has evaluated the antimicrobial effects of different G. sylvestre extracts and of eight triterpenes isolated from the most active extract on six bacterial poultry pathogens i.e. Bacillus subtilis, Enterococcus faecalis, Staphylococcus aureus, Pseudomonas aeruginosa, Escherichia coli and Enterobacter aerogenes. In particular, it has been evaluated the minimum inhibitory concentration (MIC) and minimal bactericidal concentration (MBC) of all extracts and isolated triterpenes. Finally, the cytotoxicity activity of triterpenes was evaluated by MTT assay and their antioxidant activity in basal and oxidant conditions by DCFH-DA assay. [ABSTRACT FROM AUTHOR]

Gymnema sylvestre has been used in Asian traditional medicine for its anti-microbial, antihypercholesterolemic, hepatoprotective and sweet suppressing properties and activities. G. sylvestre has also been used extensively in chewing gum, as a health food for preventing obesity and diabetes, and as a tea. This study has evaluated the total phenolic content and antioxidant activity of the aqueous and organic G. sylvestre extracts and their sub-fractions for the initial characterization of the biological properties of the isolated compounds. An in vivo cell model was used to calculate the concentration inhibiting cell growth by 50% and the ability to exert antioxidant activity. All compounds inhibit cell growth in a dose-dependent manner, with an IC50 value ranging between 29 and 1462 μM. The effects on intracellular ROS levels are extremely variable, but it is of interest that some of the compounds appear to display an antioxidant effect. [ABSTRACT FROM AUTHOR]

Objective The CUPRAC–BCS and antioxidant activity (AOA) assays have been developed as indirect methods to determine the total antioxidant capacity (TAC). We examined these assays as markers, in erythrocytes, under oxidative stress (induced by storage) and with antioxidants – vitamin C, L-carnitine, and curcumin. Methods Antioxidants were added to blood samples, stored and assayed for TAC and reactive oxygen species (ROS). Results Vitamin C at lower concentrations and L-carnitine and curcumin at higher concentrations proved beneficial to the erythrocytes. Discussion CUPRAC–BCS and AOA effectively determined TAC of stored erythrocytes. This was in accordance with the levels of ROS, and hence, CUPRAC–BCS and AOA can be used as reliable markers of TAC, in erythrocytes during oxidative stress. [ABSTRACT FROM AUTHOR]

Abstract: Silybin is the major flavonolignan of silymarin and it displays a plethora of biological effects, generally ascribed to its antioxidant properties. Herein we shall describe an efficient synthetic strategy to obtain a variety of new and more water-soluble silybin and 2,3-dehydrosilybin (DHS) derivatives in which the 23-hydroxyl group was converted to a sulfate, phosphodiester, or amine group, using a solution-phase approach. Furthermore a new and efficient method for the preparation of DHS from silybin was developed and optimised. The antioxidant properties of the new compounds were evaluated in a cellular model in vivo and they displayed an antioxidant activity comparable to or higher than silybin and DHS, being able to prevent H2O2-induced generation of intracellular reactive oxygen species (ROS). Most of the derivatives also displayed a better hydrophilicity while retaining the biological activities of silybin and they might broaden the in vivo applications of this class of natural compounds. • Guavira pomace is an agroindustrial waste of guavira pulp industry. • It was a source of arabinan, homogalacturonan and arabinogalactan. • These dietary fibres were able to scavenge ABTS radicals. • Dietary fibres protected murine fibroblasts against H 2 O 2 -induced toxicity. • Guavira pomace is a valuable source of antioxidant dietary fibres. Exotic fruits and their co-products may be valuable sources of antioxidant dietary fibres (DF) which are useful for food industry and human health. In this study, we aimed to characterize DF obtained from guavira fruit pomace and investigate its antioxidant potential employing TEAC assay as well as a cell model. The DF were chemically characterized as containing arabinan, highly-methoxylated homogalacturonan and arabinogalactan. The DF-containing fraction (CPW) presented ABTS free radical scavenger activity. MTT and DCFH-DA assay were performed to assess, respectively, changes in cell viability and the potential intracellular antioxidant activity against H 2 O 2 -induced oxidative stress in murine NIH 3T3 fibroblast. CPW exhibited no effects on cell viability, moreover, when administered 48 h prior the induction of H 2 O 2 toxic effects, it protected the cells, significantly increasing the cell viability compared to control. This protection may be related to the observed reduction of reactive oxygen species levels. Thus, the pre-treatment of cells with guavira DF for 48 h remarkably induced a cytoprotection against pro-oxidant conditions, and may be a valuable functional compound recovered from an unexploited agroindutrial waste.
