Bu çalışmada, farklı dozlarda bor (sodyum pentaborat) içeren boğa sperması sulandırıcılarının çözüm sonu toplam motilite (TMOT), progresif motilite (PMOT), spermatozoa hareket parametreleri (VCL, VSL, VAP, LIN, STR, WOB, ALH, BCF), plazma membranı ve akrozom bütünlüğü (PMAI) ve DNA fragmentasyon indeksi (DFI) parametreleri üzerine etkisi değerlendirildi. Dört baş boğadan toplam 8 ejakülat incelendi. Her ejakülat 5 eşit parçaya bölündü ve kontrol grubu olarak standart Tris sulandırıcısıyla sulandırıldı. Deney grubu sulandırıcıları olarak 0,5 g glikoza ek olarak 0,4 g bor eklenmiş Tris sulandırıcısı (Grup 1) ve glikoz yerine 0.5; 0.7; 0.9 g bor eklenmiş Tris sulandırıcıları (Grup 2, Grup 3, Grup 4) kullanıldı. Spermalar çözdürüldükten sonra TMOT, PMOT, spermatozoa hareket parametreleri, PMAI ve DFI değerleri belirlendi. Toplam motilite ve progresif motilite verilerine göre (%66.04, %50.79) Grup 1 sulandırıcısının daha yüksek değerlerde olmasına rağmen gruplar arası farklılıklar istatistiki açıdan önemsiz bulundu (P>0.05). En yüksek VSL, LIN, STR ve WOB değerlerinin (28.15 µm/s, %42.98, %71.69, %59.71) Grup 1’de olduğu görüldü (P0.05). En yüksek PMAI değerinin Grup 1’de olduğu, DFI değerlerinin ise tüm gruplarda benzer olduğu sonucuna ulaşıldı (P>0.05). Sonuç olarak borun çözdürme işleminden sonra spermatozoanın kinetik ve yapısal özellikleri üzerinde yararlı etkileri olduğu ortaya konmuştur. Ek olarak DFI açısından zararlı bir etkisi olmadığı gözlemlenmiştir. Bu veriler değerlendirildiğinde borun (sodyum pentaborat) alternatif bir sperma sulandırıcısı bileşeni olarak kullanılabileceği sonucuna ulaşılmıştır, In this research, post-thaw effects of bull semen extenders including different amounts of boron (sodium pentaborate) on total motility (TMOT), progressive motility (PMOT), kinetic parameters of spermatozoa (VCL, VSL, VAP, LIN, STR, WOB, ALH, BCF), plasma membrane and acrosome integrity (PMAI) and DNA fragmentation index (DFI) were evaluated. Eight ejaculates were examined from four different bulls. Each ejaculate was split in to five aliquots and extended with standard Tris extender (Control) and 0.4 g boron with 0.5 g glucose (Group 1) 0.5; 0.7; 0.9 g boron without glucose (Group 2, Group 3 and Group 4 respectively) were used as experimental groups. After thawing TMOT, PMOT, kinetic traits of spermatozoa, PMAI and DFI were determined. According to motility and progressive motility (66.04%, 50.79%), Group 1 had higher values than the others, but differences between groups were not statistically significant (P>0.05). The highest values of VSL, LIN, STR and WOB were determined (28.15 µm/s, 42.98%, 71.69%, 59.71%) in Group 1 (P0.05). The highest PMAI value was determined as 60.84% in Group 1 (P>0.05). The DFI rates were found similar in all control and experimental groups (P>0.05). In conclusion, it was observed that the boron (sodium pentaborate) has positive effects on post-thaw movement and structural traits. In addition, boron has no detrimental effect on DNA fragmentation index in semen freezing. Thus, boron could be used as an alternative semen extender compound

Bu çalışma, koç spermasının dondurulmasındabireysel farklılıkları ortaya koymak ve bu farklılık üzerine sulandırıcı etkisinibelirlemek amacıyla yapıldı. Ejakulatlar 5 Karayaka koçundan sezon içidönemde suni vajina yardımıyla haftada bir kez alındı. Sperma örnekleri bireysel olarak 4 farklı sperma sulandırıcısı (Yağsız süt tozu, Tris, Andromed, Bioxcell) ilesulandırıldı. Sulandırılmış spermaörneklerini içeren payetler +4 °C’de 2 saat süreyle ekilibre edildikten sonra sıvı azotbuharında (-120 °C) 15 dakika süreyle donduruldu ve sıvı azot (-196 °C) içinde saklandı. Tüm gruplardaki payetler 37 °C’de 30 saniye süreyle çözdürüldü. Her bir koçaait çözdürülmüş sperma örneklerinde spermatozoa motilitesi (%), canlı spermatozoa(%) ve anormal spermatozoa (%) yönünden değerlendirildi.Yağsız süt tozu sulandırıcısı ile dondurulansperma örneklerinde çözüm sonu daha yüksek motilite elde edilirken, en düşükmotilite oranı Bioxcell sulandırıcısından elde edilmiştir. Sonuç olarak, sunulan çalışmada elde edilen in vitro sperma kalitesibulgularına göre yağsız süt tozu sulandırıcısının diğer sulandırıcılardan dahaüstün olduğu ve koçlar arası bireysel farkı azalttığıbulunmuştur., Thisstudy was conducted to evaluate individual differences in freezing ram spermand to determine the extenders and individual sperm interaction. The ejaculates were collected from 5 Karayaka rams by artificialvagina twice a week during the breeding season. The each ejaculate was splitinto 4 equal aliquots and diluted with skimmed milk, Tris, Andromed andBioxcell extenders. Straws contained semen was equilibrated at +4 °C for 2 h and than frozen in vapor of (15 min at -120 °C) liquid nitrogen and stored in liquid nitrogen. Frozen semen wasthawed in a water bath at 37 °C for 30 seconds. In thawed semen, percentages of spermatozoamotility, viability, and abnormal spermatozoa, for each ejaculate weredetermined. The post-thaw sperm motility was significantly higher in skimmedmilk extender post-thaw, while lowest motility rate was obtained from Bioxcelldiluent. Consequently, it was found that skimmed milk extender was better thanthe other extenders and decreased individual differences between rams.

Teke spermasının dondurulması tür, yaş, sezon, bireysel ve bölgesel farklılıklar gibi birçok faktöre bağlı olduğu çok iyi bilinmektedir. Buradan hareket ederek Ankara Tekesi (Capra Hircus Ancryrensis) dondurulabilirliğini bölgesel (R) ve bireysel (I) olarak karşılaştırılması amaçlandı. Sperma her bir bölgeden 2 teke olacak şekilde toplamda 6 adet tekeden (Ayaş, Lalahan, Nallıhan) suni vagina ile alındı. Daha sonra taze sperma bireysel olarak Tris bazlı sulandırıcı ile sulandırıldı, alışıma bırakıldı (+5 C⁰/2 saat), 0,25 ml’lik payetlere dolduruldu, sıvı azot buharında (-120 C⁰/10 dk) donduruldu ve sıvı azotta (-196 C⁰) saklandı. Dondurulmuş payetler su banyosunda (37 C⁰/30 sn) çözdürüldü ve progresif motilite, total motilite ve kinetik parametreler (VCL, VSL, VAP, LIN, STR, WOB, ALH, BCF and hyperactivity) bilgisayar destekli sperm analiz cihazında (CASA) incelendi. Ayrıca çözüm sonu canlılık (SYBR 14/PI), akrozomal bütünlük (FITC-PNA/PI) ve mitokondrial aktivite (JC-I) floresan mikroskop ile değerlendirildi.Taze sperma parametreleri açısından R ve I’da istatistiksel olarak farklılık bulunmadı parametreler açısından istatistiksel olarak farklılık görülmemesine rağmen en yüksek progresif ve total motilite sırasıyla 9,97 ± 4,05; 58,42 ± 9,40 Nallıhan bölgesinde tespit edildi. Ayrıca, en yüksek canlılık oranı 64,29±5,28 ile Ayaş grubunda bulundu (P0,05). Sonuç olarak taze sperma verileri açısından R ve I’da istatistiki bir farklılık görülmemesine rağmen farklı bölgelerde bulunan Ankara Tekelerinde dondurma öncesi ve sonrasında spermanın kalitesi açısından önemli bilgiler vermiştir.Dondurulmuş çözdürülmüş, It is well accepted that freezability of buck semen depends on many different factors, such as breed, age, season, region and even invidual differences. Therefore, we aim to compare freezability of Angora buck semen (Capra hircus ancryrensis) from different regions (R) and inviduals (I). Semen was collected with artificial vagina from 6 bucks; 2 each from 3 different R in Ankara (Lalahan, Ayaş, Nallıhan). Then, fresh semen was extended individually with Tris based extender, equilibrated (+5 °C/2h), loaded into 0.25 straws, frozen in liquid nitrogen vapour (-120 °C/15 minutes) and stored in liquid nitrogen (-196 °C). Frozen straws were thawed in water bath (37 °C/30 seconds) and percentages of total motility, progressive motility and sperm kinetic parameters (VCL, VSL, VAP, LIN, STR, WOB, ALH, BCF and hyperactivity) were assesed with computer assisted sperm analyzer (CASA). Viability (SYBR-14/PI), acrosomal integrity (FITC-PNA/PI) and mitochondrial activity (JC-1) were evaluated by fluorescein microscope. Reproductive success is the most important component of a profitable cattle livestock enterprise. The most important aim in dairy or beef cattle is the reproductive success. The profitability of business in dairy cattle breeding is measured by the milk yield and in beef cattle breeding; it is measured by the speed of live weight gain. For both cases it is essential for cattle to give birth once in a year. This can only be achieved with a proper reproduction management. Therefore, these parameters are utilized to measure the reproductive performance of cattle. In this review, the effect on fertility of reproductive parameters, which hold and important place in the management of the cattle enterprise, is discussed. Reprodüktif başarı karlı bir sığır işletmesinin en önemli bileşenidir. Süt ya da et sığırı yetiştiriciliğinde en önemli hedef ise reprodüktif başarıdır. İşletmenin karlılığı, süt sığırı yetiştiriciliğinde süt verimi ile et sığırı yetiştiriciliğinde hızlı canlı ağırlık kazanımı ile ölçülmektedir. Her iki durum için de yetiştirilen ineklerin yılda bir kez yavru vermesi şarttır. Yılda bir yavru almak ise ancak iyi bir reprodüktif yönetim ile sağlanabilir. Bu yüzden sığırların reprodüktif performanslarının ölçülmesinde bu parametrelerden faydalanılır. Bu derlemede, sığırcılık işletmelerinin yönetiminde önemli yer tutan reprodüktif parametrelerin dölverimi üzerine olan etkisi ele alınmıştır.

In this study, the fertilizing ability of in vitro capacitated bull spermatozoa in two different capacitation media (Hypotaurine and Follicular Fluid) was investigated. Ovaries were obtained from slaughterhouse and were transported to the laboratory in saline (0.9% NaCl) at 30-35ºC. Cumulus oocyte complexes were collected from antral follicles of 2 to 8 mm in diameter. After collected oocytes were classified, they were transferred to maturation medium. Oocytes were cultured for 24 hours in a 100 ml drops of maturation media overlaid with mineral oil under 5% CO2, 95% humidity in air at 38.5ºC. At the end of this incubation, oocytes which displayed cumulus expansion were considered as matured. Ham's F-10 was used as the maturation medium containing 20% ECS, 10 IU/ml FSH, 10 IU/ml LH. Cryopreserved bull semen was used for fertilization of in vitro matured oocytes. Motile spermatozoa were separated by the swim-up technique. MDM solution containing 0.5 mM hypotaurine and 25% follicular fluid was used for the capacitation of separated motile spermatozoa. Hypotaurine treated spermatozoa were incubated for 7 hours and follicular fluid treated spermatozoa were incubated for 4 hours. At the end of these incubations, the obtained sperm suspension was added to fertilization medium. The final concentration of sperm in the medium was 1-3x106 sperm/ml. Oocytes and sperm were cocultured for 18 h. At the end of this culture period, oocytes were fixed and stained for evaluation of fertilization. Oocytes were considered to be fertilized when female and male pronuclei were visible in the cytoplasm. Finally, from 878 collected oocyes, 385 oocytes were matured (43.84%). This study was carried out to determine the effect of transcervical and intravaginal artificial insemination on the pregnancy rate in dogs. Semen was collected by the massage method from dogs brought to our department. Collected semen was examined in the laboratory and stored in liquid nitrogen after dilution with Tris extender. During the study 31 bitches brought to our department for artificial insemination were used. One of the bitches was not evaluated. Oestrus was detected by vaginal cytology and the bitches were inseminated twice during the oestrus period. Fifteen of the 30 bitches were inseminated transcervically and the other 15 intravaginally. The pregnancy rate was 60.00% with the transcervical insemination method and 26.66% with the intravaginal insemination method (p < 0.05). In conclusion, we have detected that transcervical insemination with frozen dog semen is more effective than intravaginal insemination in terms of fertility.

Araştırmada, Alageyiklerin bazı androlojik parametrelerinin ortaya konması amaçlanmıştır. Çalışma için, Ankara Sincan Hayvanat Bahçesi’nde barındırılan yaşları 2-4 arasında değişen 4 erkek Alageyik değerlendirmeye alınmıştır. Alageyiklerin testis çapı, yüksekliği ve genişlik ölçüleri kaydedilmiştir. Ejakülat miktarı, sperma pH’sı, spermatozoa yoğunluğu laboratuvarda değerlendirilmiş; toplam motilite, progresif motilite, anormal spermatozoa, akrozom bütünlüğü ve spermatozoa hareket parametreleri ise CASA (Computer Aid Sperm Analyser) ile belirlenmiştir. Testis boyutları ile ejakülat miktar ve yoğunluğu arasındaki ilişki değerlendirilmiştir. Testis yükseklik, çap ve genişlik ölçüleri sırasıyla 12.7 ± 2.73 cm, 7.8 ± 1.29 cm, 5.5 ± 0.69 cm olarak, ejakülat miktarı 2.4 ± 0.47 ml, spermatozoa konsantrasyonu 1.1 ± 0.36 x 109 , pH 7.2 ± 0.39, toplam motilite %68.6 ± 6.15, progresif motilite %10.3 ± 6.97, anormal spermatozoa %18.3 ± 3.50, akrozom bütünlüğü %99.4 ± 1.2 olarak belirlenmiştir. Testis boyutlarının (yükseklik, genişlik, çap) ejakülat miktarı ve spermatozoa yoğunluğuyla korelasyona sahip olduğu tespit edilmiştir. Four male deer ranging from 2-4 ages at Ankara Sincan Zoo were used in this study Testis diameter, width and height dimensions were measured. Ejaculate volume, semen pH, spermatozoa concentration were determined at laboratory. Total motility, progressive motility, abnormal spermatozoa, acrosome integrity and spermatozoa kinetic parameters were analyzed by CASA (Computer Aid Sperm Analyzer). The relation between testicle dimensions and semen volume and concentration have been evaluated. Therefore, testis height, diameter and width dimensions were measured as 12.7 ± 2.73 cm, 7.8 ± 1.29 cm, 5.5 ± 0.69 cm, respectively. Ejaculate volume 2.4 ± 0.47 ml, total spermatozoa 1.1 ± 0.36 x 109, pH 7.2 ± 0.39, motility %68.6 ± 6.15, progressive motility %10.3 ± 6.97, abnormal spermatozoa %18.3 ± 3.50, acrosome integrity %99.4 ± 1.2 were found. High correlation was observed between testis dimensions (height, width, and diameter) and ejaculate volume and spermatozoa concentration. In this study, the effects of post-­‐mating FGA/P and of split eCG/PMSG injections at different doses were evaluated for the induction of oestrus in Angora goats during the breeding season. In total, thirty-­‐six multiparous does were assigned into two separate synchronisation trials. In Experiment 1 (double FGA), 22 does were administered all firstly an intravaginal sponge containing 20 mg FGA for 11 days. On day 9, the animals then received i.m. injections of 125 mg cloprostenol, and of 400 IU eCG concurrently. On day 6 post-­‐mating, animals were randomly assigned into two groups, as control (n=13), single and double FGA group (n=9), receiving the second FGA for 11 days. In Experiment 2 (split eCG), 14 does were randomly assigned into eCG 750 and eCG 500 groups (n=7 each), with multiple injections (on 5 consecutive days) of 750 (250 and 100 in double and 50 IU at last) or of 500 IU (100 IU daily). The results showed that; i) double/post-­‐mating FGA had no favourable effect upon the reproductive parameters studied, ii) the eCG itself led to a considerably poorer fertility and twinning in Angora does during the breeding season. Ankara Keçilerinde Sezon-­‐içi Östrus Senkronizasyonu Amacıyla Çift Doz FGA Sünger veya Bölünmüş eCG Uygulamalarının Üreme Özellikleri Üzerine Etkileri, Sunulan çalışmada, üreme sezonundaki Ankara keçilerinde çiftleşme sonrası FGA/P4 ve farklı dozlarda bölünmüş eCG/PMSG enjeksiyonlarının östrus senkronizasyonundaki etkileri araştırıldı. Toplam 36 baş anaç (önceden doğum yapmış) keçi iki ayrı senkronizasyon grubuna ayrıldı. Çalışma 1’de (çift doz FGA), keçilere 11 gün süreyle 20 mg FGA içeren vagina-içi sünger uygulandı. Hayvanlara 9. günde, 125 mg cloprostenol ve 400 IU eCG birlikte enjekte edildi. Çiftleşme sonrası 6. günde, hayvanlar tek (kontrol, n=13) veya çift doz FGA grubu (n=9), ikinci FGA 11 gün süreyle, olmak üzere rastgele iki deneme grubuna ayrıldı. Çalışma 2’de (bölünmüş eCG), 14 baş keçi rasgele tarzda eCG 750 ve eCG 500 gruplarına ayrılarak (n=7, her biri), ilk gruba 750 IU eCG (iki kez 250, iki kez 100 ve son kez 50 IU) verilirken, diğer gruba ise 500 IU eCG (günlük 100 IU) çoklu enjeksiyon tarzında (ardışık 5 gün süreli) olarak verildi. Sonuç olarak; i) çift doz/çiftleşme sonrası FGA uygulamasının çalışılan üreme parametreleri üzerine bir katkısının olmadığı, ii) sadece eCG uygulamasının ise yavru verimi ve ikizlik yönünden belli düzeyde daha düşük oranlara yol açtığı kanısına varıldı.

The aim of this study was to assess the use of - 152°C ultra freezer for freezing and storing bull semen as an alternative to freezing over the liquid nitrogen vapor and storing in liquid nitrogen in terms of certain post-thaw spermatological parameters. Two ejaculates, one per each bull, were used without pooling. Four different freezing and storing protocols were tested. At 1 week, 2 months and 6 months after cryopreservation, the frozen semen was thawed and the seminal parameters were determined. As a conclusion, this study has confirmed that the use of -152 °C ultra-low freezer for freezing and storing bull semen for 6 months is a viable alternative to liquid nitrogen, Bu çalışmanın amacı, boğa spermasının -152 °C derin dondurucuda dondurulup saklanmasının sıvı azot buharında dondurma ve sıvı azotta saklamaya çözüm sonu kimi spermatolojik parametreler bakımından bir alternatif olup olmayacağının araştırılmasıdır. Bu çalışmada iki adet boğanın birer ejakulatı miks yapılmadan kullanıldı. Araştırmada dört farklı dondurma ve saklama protokolü denendi. Dondurma işleminden bir hafta, iki ve altı aylık periyotlar sonrasında payetler çözdürüldü ve çözüm sonu bazı spermatolojik özellikler değerlendirildi. Sonuç olarak bu çalışma -152°C derin dondurucunun boğa spermasının dondurulması ve altı ay süreyle saklanmasında sıvı azota bir alternatif olarak kullanılabilirliğini ve boğa spermasının dondurulması ve saklanmasında iyi bir ortam sağladığını doğrulamaktadır

The objective of this study was to evaluate the reproductive performance of primiparous and multiparous Saanen goats after intrauterine laparoscopic artificial insemination with frozen semen. Twenty-four Saanen goats, divided in 2 groups: group 1 consisted of 11 primiparous goats and group 2 consisted of 13 multiparous goats. Estrus was synchronized by 20 mg fluorogestone acetate (FGA)-impregnated intravaginal sponges and the IM administration of 125 mg of cloprostenol (PGF2a) and eCG (400 IU), 48 h before sponge removal. Intrauterine fixed time artificial insemination (AI) was carried out 55 h after removal of the FGA sponges. Results of the present study indicate that primiparous Saanen goats exhibited a higher pregnancy rate than multiparous goats. However, pregnancy and conception rate did not differ significantly between primiparous and multiparous goats (66% and 53%; P > 0.05). In conclusion, the technique of laparoscopic intrauterine insemination in multiparous and primiparous goats is used successfully under rural conditions.

Bu çalışmada, farklı koyun ırklarında progestagen-eCG ile östrus senkronizasyonu sonrası bazı üreme parametreleri-nin karşılaştırmalı olarak irdelenmesi amaçlandı. Araştırmada, 12 baş Akkaraman, 9 baş İvesi ve 8 baş Kıvırcık koyun kullanıldı. Kızgınlıkları toplulaştırmak amacıyla, tüm koyunlara 20 mg kronolon (flugeston asetat) emdirilmiş süngerler 14 gün süreyle vaginaya yerleştirildi. Süngerler geri alınırken, hayvan başına 400 IU eCG enjeksiyonu uygulandı. Östrusları tespit edilen koyunlar elde sıfat yöntemi ile çiftleştirildi. Elde edilen östrus oranı, son uygulama-östrus aralığı, östrus süresi, gebelik oranı, doğum oranı, ikizlik oranları ve yavru verimleri sırasıyla Akkaraman ırkında; %83.3, 34.5±3.9 saat, 27.1±2.3 saat, %75, %75, %25 ve 1.25, İvesi ırkında; %88.8, 38.3±4.3 saat, 25.4±2.0 saat, %77.7, %77.7, %22.2 ve 1.22 ve Kıvırcık ırkında ise; %100, 25.7±1.7 saat, 22.5±1.4 saat, %100, %87.5, %75 ve 1.75 olarak gözlendi. Irklar arasında östrus oranı, gebelik oranı ve doğum oranı yönlerinden önemli bir fark belirlenemedi (P>0.05). Ancak, son uygulama-östrus aralığı, östrus süresi, ikizlik oranları ve yavru verimleri yönünden istatistiksel olarak önemli farklar saptandı (P

Bu çalışma Ankara tekesi spermasının farklı oranlarda yumurta sarısı içeren yağsız süt tozu sulandırıcısı ile sulandırılıp dondurulması ve dondurma sonrası spermatolojik özelliklerin saptanması amacıyla yapılmıştır. Çalışmada, 3 baş ergin Ankara keçisi tekesinden alınan ejakülatlar kullanılmıştır. Ejakülatlar çiftleştirme mevsiminde suni vajen kullanılarak haftada iki kez, üç hafta süresince toplanmıştır. Alınan ejakülatlar birleştirildikten sonra 5 eşit hacme bölünmüş ve 37°C’deki su banyosunda % 5, 10, 15, 20 oranında yumurta sarısı içeren ve içermeyen (kontrol) yağsız süt tozu sulandırıcısıyla sulandırılmıştır. Sulandırılan spermalar payetlere (0.25 ml) çekilerek 5°C’de ekilibrasyona bırakılmıştır. Ekilibrasyonu izleyen süreçte payetler sıvı azot buharında dondurularak daha sonra değerlendirilmek üzere -196° C’deki sıvı azotta saklanmıştır. Çözdürme sonrası en yüksek spermatozoa motilitesi (% 65.0±4.53) % 10 yumurta sarısı içeren yağsız süt tozu sulandırıcısında bulunurken, en düşük spermatozoa motilitesi (% 30.0±3.56) yumurta sarısı içermeyen yağsız süt tozu sulandırıcısından elde edilmiştir. Anormal spermatozoa bakımından % 10 ve 15 yumurta sarısı içeren gruplar (% 22.6±4.01 ve 24.5±3.70) arasındaki istatistiksel farklılıklar önemsiz, yumurta sarısı içermeyen kontrol grubundaki anormal spermatozoa oranı (% 44.6±4.41) diğer sulandırıcı gruplarından istatistiksel açıdan önemli oranda yüksek bulunmuştur (P, This study was carrried out to determine the effects of different amount of egg yolk added to skimmed milk extender of Angora buck semen on spermatologic parameters after frozen-thawed semen. Ejaculate samples were obtained from three mature Angora bucks were used in this study. Ejaculates were collected using an artificial vagina twice a week, for three weeks running, during the breeding season. After pooling, each pooled ejaculate was split into five equal aliquots and diluted with based extender containing egg yolk (5, 10,15, 20 %) or no egg yolk (control). Diluted samples were aspirated into 0.25 ml straws, equilibrated at 5°C. Following equilibration, the straws were frozen in liquid nitrogen vapour and stored at -196°C until evaluation. Highest motility after post-thaw was obtained from skimmed milk extender containing 10 % egg yolk as 68.5±4.53 % and lowest motility was obtained from skimmed milk extender containing 0 % egg yolk as 30.0±3.56 %. In the present study, testes characteristics and spermatologic features of 10 Karayaka rams kept under Ankara were determined. The average scrotal circumference, volume, thickness, testis length, diameter were 29.80 ± 1.08 cm, 573.70 ± 25.66 ml, 0.57 ± 0.01 cm, 12.31 ± 0.48 cm, 5.9 0 ± 0.18 cm respectively. From each ram 10 ejeculates were collected twice a week using an artificial vagina during the nonbreeding period. The spermatological characteristics of samples were as following: Ejaculate volume average 0.98 ± 0.08 ml, spermatozoa motility 79.35 ± 0.88 %, spermatozoa concentration 3653.16 ± 179 x109/ml, percentage of abnormal spermatozoa 10.99 ± 0.58 %, percentage of dead spermatozoa 13.35 ± 0.63 %, HOS test 75.84 ± 1.16 % and pH averaged 6.92 ± 0.04 in Karayaka rams. Sunulan çalışmanın amacı, in vitro olarak çözüm sonrası teke spermasının spermatolojik özellikleri üzerine Tris ve Andromed sulandırıcılarının etkisini araştırmaktır. Çalışmanın materyalini 2 baş Saanen ırkı teke oluşturmuştur. Spermalar aşım sezonu içerisinde her tekeden 5’er ejekülat olmak üzere haftada iki kez sun’i vajen yöntemiyle alınmıştır. Alınan spermalar iki eşit hacme bölündükten sonra 37oC’ ye ayarlanmış su banyosunda Tris ve Andromed sulandırıcılarıyla sulandırılmıştır. Sulandırma 1:10 oranında yapılmıştır. Sulandırılmış sperma 0.25 ml’lik payetlere çekilmiş, 4oC’ de 2.5 saat ekilibrasyona bırakılmıştır. Payetler sıvı azot buharında (-110oC/-130oC arasında 10 dakika) dondurularak, sıvı azot içinde (-196oC) muhafaza edilmiştir. Sperma su banyosunda 37o C’ da 30 saniyede çözdürülerek spermatolojik muayeneler yapılmıştır. Bu çalışmada, çözüm sonrası; spermatozoa motilitesi (%) (34±4.1; 20±3.5), anormal spermatozoa oranı (%) (45.40±3.7; 62.40±5.7), akrozoma bağlı anormal oranı (%) (24.80±2.8; 42.80±2.7) ve hypo-osmotik şişme test oranı (HOST) (%) (36±3.5; 27.20±2.3) yönünden Andromed ve Tris sulandırıcıları arasında istatistiki olarak önemli bir fark belirlenmiş (P0.05). Sonuç olarak, Andromed sulandırıcısının Tris sulandırıcısından çözüm sonrası bazı spermatolojik parametreler üzerine daha olumlu etki yaptığı saptanmıştır., The aim of this study was to evaluate for the effects of Tris-glucose-citric acid- egg yolk and Andromed extender, free of egg yolk, on sperm characteristics of buck semen after thawing in vitro. Two Saanen buck were used in this study. Semen was collected by an artificial vagina twice a week in autumn (mating season). After being divided into 2 equal aliquots, the collected semen diluted in tris diluent or andromed at 37oC in water bath. Dilutions were carried out at a proportion of 1:10. The diluted semen was placed in 0.25 ml plastics straws and equilibrated (at 4oC 2.5 hours) and frozen in vapour of liquid nitrogen (LN) (15 minutes at -110/-130 o C). Frozen straws were stored in liquid nitrogen (-196 oC). Straws were thawed in the water bath at 37oC for 30 seconds and spermatological examinations were carried out. The results demonstrated that spermatozoa motility (%), abnormal spermatozoa rate (%), acrosome abnormal rate (%) and hypo-osmotic swelling test (HOST) rate (%) were in frozen-thawed semen processed in Andromed (34±4.1, 45.40±3.7, 24.80±2.8, 36±3.5) were significantly (P0.05). Bu araştırmada, gökkuşağı alabalığında spermatolojik parametreler ve aralarındaki korelasyonlar değerlendirilmiştir. Ayrıca, iki farklı sulandırıcıyla (Sulandırıcı I ve II) spermalar dondurularak önemli olan spermatolojik özellikler belirlenmiş ve bu sulandırıcıların spermatozoa canlılığı üzerine etkileri incelenmiştir. Spermanın dondurulmasında tesadüfi seçilmiş 24 gökkuşağı alabalığından alınan ejakulatlar kullanılmıştır. Çalışmada kullanılan alabalıklardan, masaj yöntemiyle sperma alınarak spermatolojik özellikler [sperma miktarı (ml), spermatozoa motilitesi (%), spermatozoa yoğunluğu (x 109 /ml), pH] tespit edilmiştir. Sperma iki farklı sulandırıcı kullanılarak, sıvı azot buharında dondurulmuştur. Klinik olarak normal bulunan aygırda spermatolojik özelliklerden sperma miktarı, motilitesi, yoğunluğu, anormal ve ölü-canlı spermatozoa oranı ile sperma pH’sı sırasıyla 70.0 ml, % 75.0, 93.750 x 106/ml, % 16.0, %19.0 ve 7.5 olarak saptanmış ve aygır normal aşım davranışları göstermiştir. Klinik olarak tek taraflı kriptorşidi bulunan aygırda ise ortalama spermatolojik özellikler sırasıyla 56.0 ml, % 70.0, 120 x 106/ml, % 19.0, % 22.0 ve 7.7 olarak saptanmış, alınan 3 ejakülatta da aşırı libido gözlenmiştir. Bu araştırmada, infertilite problemleri olan kısrakların klitoris ve serviks uterilerinden, aygırların ise orificium urethra externa, corpus penis ve fossa glandislerinden alınan örneklerin bakteriyolojik yoklamaları yapıldı ve izole edilen etkenierin antibiyotiklere duyarlılıkları belirlendi.lnfertilite problemi olan 86 kısrağın 65 (% 75.6)'inin kiltorisinden ve12 (% 13.9)'sinin serviksinden izolasyon yapılabildi. Kısrakların klitoris ve serviks uterilerinden E.coli, Staph.aureus, Staph.epidermidis, Streptococcus sp., Str.agalactiae, Acinetobacter sp., K.pneumoniae, Bacillus sp., Corynebacterium sp., Pseudomonas sp., Moraxella sp. izole ve identifiyeedildi. Incelenen 40 aygırın urethra, fossa glandis ve corpus penisinden ise Streptococcus sp., E.coli, Acinetobacter sp., Staph.aureus, Staph.epidermidis, K.pneumoniae, Corynebacterium sp., Moraxella sp., Bacillus sp., Al caligenes sp., Neisseria sp., Pseudomonas sp. izole ve identifiye edildi. From clitorises and certix uteries of the mares,E.eoli, Staph.aureus, Staph.epidermidis, Streptoeoeeus sp., Str. agalactiae, Aeinetobaeter sp., K.pneumoniae, Baeillus sp., Corynebaeterium sp., Pseudomonas sp., Moraxella sp. were isolated and identified. From urethras, fossa glandises and corpus penises of the examined 40 stallions, Streptoeoeeus sp., E.eoli, Aeinetobaeter sp., Staph.aureus, Staph.epidermidis,K.pneumoniae, Corynebaeterium sp., Moraxella sp., Baeillus sp.,Alealigenes sp., Neisseria sp., Pseudomonas sp. were isolated and identified. In none of the samples from the infertile mares and the stallionsT.equigenitalis was obtained.As a result of antibiogram test, Gram (+) bacteria isolated from themares were sensitive to ampicilline,chloramphenicol, kanamycine, neomycine,penicilline, tetracycline and for Gram (-) bacteria were sensitive to colistine sulfate, tetracycline, ampicilline (except for E.eoli and Pseudomonas sp.), chloramphenicol, kanamycine, neomycine, streptomycine (except for Pseudomonas sp.).Gram (+) bacteria isolated from the stallions were sensitive to ampicilline, chloramphenicol, colistine sulfate, kanamycine, neomycine, peniciline, streptomycine, tetraeycline and for gram (-) bacteria were sensitive tochloramphenicol, tetraeycline, colistine sulfate, kanamycine, neomycine, streptomycine (except for Arizona sp.).

59 - ÖZET Bu çalışma, Ankara keçisi tekesinin spermatolojik özelliklerini saptamak, teke spermasını dondurmak ve iki değişik yöntemle sinkronize edilip nativ, sulandırılmış ve donmuş sperma ile tohumlanan keçilerden elde edilen döl verimini araştırmak amacıyla yapılmıştır. Araştırma, Lalahan Hayvancılık Araştırma Enstitüsünde yapılmış ve kurumun Ankara keçisi sürüsünden rastlantısal olarak seçilen 5 teke ve 120 keçi kullanılmıştır. Araştırmada kullanılan 5 tekenin sun'i vagenle alınan 10'ar ejekülatlarında sperma miktarı(ml), spermatozoa motilitesi (%), spermatozoa yoğunluğu (xlO9), sperma pH'sı, ölü-canlı spermatozoa oranı (%), anormal spermatozoa oranı (%); dondurulan spermalarda çözüm sonu motilıte oranı (%) ve çözüm sonu anormal akrozom oram (%) saptanmıştır. Beş tekenin 10'ar ejekülatlarında saptanan spermatolojik özelliklerin ortalama değerleri yukarıda belirtilen sırasıyla 0.90 + 0.03, 81.0 ± 1.31, 2.892 ± 0.116, 6.67 ± 0.06, 11.6 ± 0.9, 3.63 ± 0.07, 40.1 ± 2.2, 23.36 ± 3.07 olarak bulunmuştur. Donmuş spermalarda saptanan çözüm sonu termo-rezistens test bulguları (%) ise, 0, 2, 4, 6 ve 8. saatlerde sırasıyla 40.1, 16.7, 2.9, 0.1 ve 0.0 olarak gerçekleşmiştir. Beş Ankara keçisi tekesinden alınan 10'ar ejekülat, spermatolojik özellikleri saptandıktan sonra Tris-yumurta sarısı- gl işerin sulandırıcısı ile 100 x 106spermatozoa/0.25 mi olacak şekilde sulandırılmış ve payetlere çekilerek 2 saat ekilibrasyon sonu sıvı azot buharında dondurularak depolanmıştır.- 60 - Çalışmada kullanılan toplam 120 Ankara keçisi 60'ar keçilik iki gruba ayrılarak östrusları senkronize edilmiştir. En az 3 ay süre ile erkeklerden ayrı tutulan 60 keçilik birinci gruba çiftleşme sezonuna girişte 12 gün süre ile penisleri bez altlık ile bağlanmış teke katılmış ve bu grupta %83.33 östrus beldekleri gözlenmiştir. Çifteşme sezonunda ise, 60 kişilik ikinci gruba 11 gün ara ile 2.5mg PGF2a i. m. injekte edilmiş ve %86.67 oranında östrus gözlenmiştir. Her iki grupta östrus gösteren keçiler nativ, sulandırılmış ve donmuş sperma ile tohumlanmışlar ve sırasıyla birinci grupta %68.4, 46.6 ile 37.5 gebelik ve %63.1, 33.3 ile 37.5 doğum oranı elde edilmiştir. Aynı şekilde tohumlanan ikinci grup keçilerde ise, %85.7, 55.0 ile 38.8 gebelik ve %64.2, 50.0 ile 27.7 doğum oranı saptanmıştır. - 61 - SUMMARY This study has been realized to detect spermatological features and freeze of Angora goat buck semen. Fertility results were also investigated of Angora goats synchronized with two different methods and inseminated with fresh, diluted or frozen semen. Studies have been carried out in Lalahan Animal Research Institute. Five Angora buck and 120 Angora goat does were randomly selected. Semen volume (cc), sperm motility {%), sperm concentration (xlO9), semen pH, live-dead ratio (%) and abnormal spermatozoa rates (%) were determined a total of 50 semen samples collected from 5 Angora goat bucks by artificial vagina. In deep frozen semen samples post-thaw motility (%), and the rate of abnormal acrosomes (%) were also detected. Average values obtained were 0.90 ± 0.03, 81.0 ± 1.31, 2.89 + 0.11, 6.67 ± 0.06, 11.6 ± 0.9, 3.63 ± 0.07, 40.1 ± 2.2 and 23.36 ± 3.07, respectively. A total of 50 ejeculates collected from 5 Angora goat bucks were extended by Tris-egg yolk-glycerine extender up to 0.25 ml of extended samples contains about 100x10 6 of spermatozoa and filled into straws. After two hours of equilibration period, diluted semen samples were frozen in liquid nitrogen vapour and transferred into the liquid nitrogen. In the early breeding season, 120 Angora does were divided into two equal groups. First group of animals, which had been kept in isolation from males for 3 months were introduced to the Angora goat bucks. After a 12 days introductory period, 83.33 % of females detected in oestrus.- 62 - Second group of animals, which injected with 2.5 mg of dinoprost thromethamine eleven days apart in breeding season, came into estrus at the rate of 86.67%. All the oestrus animals in first and second groups were inseminated with fresh, diluted or frozen Angora goat semen. Pregnancy and birth rates in first group were 68.4, 46.6, 37.5 and 63.1, 33.3, 37.5, respectively.
