Seedling establishment is a most critical stage that determined the crop production at later stages. An in vitro investigation was carried out at the Laboratory of the Department of Plant Breeding and Genetics, Sri Karan Narendra College of Agriculture, Jobner (Rajasthan) in Rabi season 2016-17 to study the response of fenugreek genotypes for their germination and early seedling parameters under five salt stress levels viz. 0.0 mM, 40 mM, 80 mM, 120 mM and 160 mM. Fifteen seeds of each genotype were sown in sterilized Petridish and seedling observations were recorded. Results revealed that the average value was found maximum in the control and minimum at higher salinity level for most of traits. Higher reduction was found at higher salinity levels for most of the characters viz., plumule length, radicle length, seedling length, plumule to radicle length ratio, plumule fresh weight, radicle fresh weight, plumule dry weight, radicle dry weight and seedling vigour index but germination percentage showed less reduction. The contrast between mean of various genotypes in control conditions (S0) versus Sm (mean of S1, S2, S3 and S4) for each character also showed that overall mean was highest in control as compared to the overall mean of salinity levels for all the traits. This investigation concluded that higher level of salinity (above 60 mM) adversely most affected to seed germination and other early seeding traits rather than (below 40 mM) lower salinity levels. Based upon the rank totals of genotype over different salinity levels S1, S2, S3 and S4 (Sm) and characters, the RMt-303 was found to be most desirable genotype followed by UM-385, RMt-143 and RMt-305. These lines would be used as a viable option under saline conditions and better exploit in hybridization programme.

Clustered regularly interspaced short palindromic repeats/CRISPR associated nuclease 9 (CRISPR-Cas9) system was instigated first into eukaryotes in the last ten years are becoming productive and worldwide application for genome modification. Genome engineering through insertion of foreign DNA insert have numerous disadvantages which could be overcome by use of DNA-free genome editing. Various ways of DNA-free genome editing mediated by CRISPR/Cas systems are CRISPR/Cas delivery as ribonucleoprotein, delivery of CRISPR/Cas as virus-like particles and agrobacterium-based delivery of CRISPR/Cas. Crop improvement through DNA-free genome editing via CRISPR/Cas have been applied in rice, wheat, maize, tomato, soybean and rare species like Nicotiana benthamiana etc. It is method of choice for precise genome editing without genome shuffle in an organism.

Background: Drought and moisture stress is the most limiting factor affecting growth and productivity of crop plants. The genotypes may differ in their ability to withstand drought or moisture stress. Therefore, the research was carried out to identify suitable genotypes in moisture stress environments. Methods: The forty eight genotypes of fenugreek were planted in randomized block design with three replications under two environmental conditions namely, (i) normal irrigation (E1) and (ii) staggered irrigation (E2) during rabi 2016-2017. Eight stress tolerance indices viz., stress tolerance (TOL), stress susceptibility index (SSI), stress tolerance index (STI), mean production (MP), geometric mean production (GMP), yield index (YI), stress susceptibility percentage index (SSPI) and modified stress tolerance index (MSTI) were calculated based on seed yield under moisture stress (Ys) and non-stress condition (Yp). The correlation coefficient and mean rank of stress indices were calculated to identify the moisture stress tolerant genotypes. Result: The seed yield under non-stress condition (Yp) had significant positive association with TOL, SSI, STI, MP, GMP, SSPI and K1STI. The seed yield under stress condition (Ys) had significant positive association with STI, MP, GMP, YI and K2STI, while significant negative association with TOL, SSI and SSPI. The results showed that MP, GMP and STI indices were more effective in identifying high yielding genotypes in both stress and non-stress conditions while TOL, SSI, YI and SSPI under moisture stress condition. Based upon the mean seed yield and stress tolerance indices, the genotypes UM-124, UM-60, UM-55, UM-28 and UM-4 were found tolerant to staggered irrigation conditions. Hence, these genotypes may be used further in breeding programmes for moisture stress tolerance.

Background: Groundnut is an important oilseed crop in the India. The groundnut kernels serve as a rich source of edible oil (48-50%) and quality protein (25-28%). In crop improvement programme, availability of sufficient genetic variability is of immense importance. The knowledge of nature and magnitude of genetic variance controlling yield and yield components is a prerequisite for improvement of yield in any crop. Methods: The present investigation was undertaken to assess the genetic variability and character associations for pod yield and component characters in 45 genotypes of groundnut. The genotypes were raised in randomized complete block design with three replications during kharif 2019 at SKNAU, Jobner (Rajasthan). Result: The analysis of variance revealed significant differences among the genotypes for all the characters studied indicating presence of wide genetic variation for different characters. In the present investigation, high genetic advance coupled with high heritability and GCV was observed for biological yield per plant, 100-kernel weight, kernel yield per plant, dry pod yield per plant and pods per plant which indicated prevalence of additive gene action in their expression and these traits possessed high selective value. Kernel yield per plant, pods per plant and biological yield per plant exhibited significantly positive correlation with dry pod yield per plant both at genotypic and phenotypic levels, while shelling percentage at genotypic level only. Three characters viz., kernel yield per plant, pods per plant and biological yield per plant could be considered as direct selection parameters for yield improvement in groundnut because they exerted positive direct effect on dry pod yield per plant.
