Perfluoroalkyl substances (PFAS) are highly persistent organic pollutants that have been detected in a wide array of environmental matrices and, in turn, biota including humans and wildlife wherein they have been associated with a multitude of toxic, and otherwise adverse effects, including ecosystem impacts. In the present study, we developed a toxicity assay for embryonic stages of mahi-mahi (Coryphaena hippurus), as an environmentally relevant pelagic fish species, and applied this assay to the evaluation of the toxicity of “legacy” and “next-generation” PFAS including, respectively, perfluorooctanoic acid (PFOA) and several perfluoroethercarboxylic acids (PFECA). Acute embryotoxicity, in the form of lethality, was measured for all five PFAS toward mahi-mahi embryos with median lethal concentrations (LC50) in the micromolar range. Consistent with studies in other similar model systems, and specifically the zebrafish, embryotoxicity in mahi-mahi generally (1) correlated with fluoroalkyl chain length and hydrophobicity, i.e., log P, of PFAS, and thus, aligned with a role of uptake in the relative toxicity; and (2) increased with continuous exposure, suggesting a possible role of developmental stage specifically including a contribution of hatching (and loss of protective chorion) and/or differentiation of target systems (e.g., liver). Compared to prior studies in the zebrafish embryo model, mahi-mahi was significantly more sensitive to PFAS which may be related to differences in either exposure conditions (e.g., salinity) and uptake, or possibly differential susceptibility of relevant targets, for the two species. Moreover, when considered in the context of the previously reported concentration of PFAS within upper sea surface layers, and co-localization of buoyant eggs (i.e., embryos) and other early developmental stages (i.e., larvae, juveniles) of pelagic fish species to the sea surface, the observed toxicity potentially aligns with environmentally relevant concentrations in these marine systems. Thus, impacts on ecosystems including, in particular, population recruitment are a possibility. The present study is the first to demonstrate embryotoxicity of PFAS in a pelagic marine fish species, and suggests that mahi-mahi represents a potentially informative, and moreover, environmentally relevant, ecotoxicological model for PFAS in marine systems.

Zearalenone (ZEA) is a mycotoxin, commonly found in agricultural products, linked to adverse health impacts in humans and livestock. However, less is known regarding effects on fish as both ecological receptors and economically relevant “receptors” through contamination of aquaculture feeds. In the present study, a metabolomics approach utilizing high-resolution magic angle spinning nuclear magnetic resonance (HRMAS NMR) was applied to intact embryos of zebrafish (Danio rerio), and two marine fish species, olive flounder (Paralichthys olivaceus) and yellowtail snapper (Ocyurus chrysurus), to investigate the biochemical pathways altered by ZEA exposure. Following the assessment of embryotoxicity, metabolic profiling of embryos exposed to sub-lethal concentrations showed significant overlap between the three species and, specifically, identified metabolites linked to hepatocytes, oxidative stress, membrane disruption, mitochondrial dysfunction, and impaired energy metabolism. These findings were further supported by analyses of tissue-specific production of reactive oxygen species (ROS) and lipidomics profiling and enabled an integrated model of ZEA toxicity in the early life stages of marine and freshwater fish species. The metabolic pathways and targets identified may, furthermore, serve as potential biomarkers for monitoring ZEA exposure and effects in fish in relation to ecotoxicology and aquaculture.

Pop-up satellite archival tags (PSATs) have been used to demonstrate habitat utilization and large-scale migrations of aquatic species and are a critical tool to manage highly migratory fish populations. Use of PSATs has increased in recent years; however, few studies have investigated the physiological and behavioral effects of carrying a PSAT. To address this gap, young-adult mahi-mahi (Coryphaena hippurus; 25–35 cm fork length) were tagged with miniature PSATs and assessed in a two-part experiment utilizing swim tunnel respirometry and behavioral analysis of free-swimming individuals. Swim tunnel respirometry revealed significant reductions in the critical and optimal swimming speeds of tagged fish (10.2% and 20.9%, respectively), as well as significant reductions in maximum metabolic rate and aerobic scope (16.1% and 21.4%, respectively). In contrast, mean and maximum velocity, acceleration, total distance traveled, survival, and feeding success of free-swimming tagged fish showed no impacts of tagging compared with untagged conspecifics held in the same tank. The results of this study highlight the importance of considering multiple methodologies to assess the impacts of tagging fish and provide insight into the data collected by PSATs deployed on wild fish.

Publicly available toxicological studies on wastewaters associated with unconventional oil and gas (UOG) activities in offshore regions are nonexistent. The current study investigated the impact of hydraulic fracturing-generated flowback water (HF-FW) on whole organism swimming performance/respiration and cardiomyocyte contractility dynamics in mahi-mahi (Coryphaena hippurus-hereafter referred to as "mahi"), an organism which inhabits marine ecosystems where offshore hydraulic fracturing activity is intensifying. Following exposure to 2.75% HF-FW for 24 h, mahi displayed significantly reduced critical swimming speeds (Ucrit) and aerobic scopes (reductions of ∼40 and 61%, respectively) compared to control fish. Additionally, cardiomyocyte exposures to the same HF-FW sample at 2% dilutions reduced a multitude of mahi sarcomere contraction properties at various stimulation frequencies compared to all other treatment groups, including an approximate 40% decrease in sarcomere contraction size and a nearly 50% reduction in sarcomere relaxation velocity compared to controls. An approximate 8-fold change in expression of the cardiac contractile regulatory gene cmlc2 was also seen in ventricles from 2.75% HF-FW-exposed mahi. These results collectively identify cardiac function as a target for HF-FW toxicity and provide some of the first published data on UOG toxicity in a marine species.

The understanding of the detection threshold and behavioral response of fishes in response to crude oil is critical to predicting the effects of oil spills on wild fish populations. The Deepwater Horizon oil spill released approximately 4.9 million barrels of crude oil into the northern Gulf of Mexico in 2010, overlapping spatially and temporally with the habitat of many pelagic fish species. Yet, it is unknown whether highly migratory species, such as mahi-mahi (Coryphaena hippurus), might detect and avoid oil contaminated waters. We tested the ability of control and oil-exposed juvenile mahi-mahi (15–45 mm) to avoid two dilutions of crude oil in a two-channel flume. Crude oil-derived polycyclic aromatic hydrocarbons (PAHs) are pervasive environmental pollutants with well-established detrimental effects on the health of marine organisms. Following large-scale oil spills in the marine environment, there is a critical need for noninvasive sampling methods to assess environmental exposure to PAHs without further perturbations to the population and for long-term monitoring following a spill. To test the efficacy of epidermal mucus mRNA as a source for noninvasive sampling, juvenile mahi-mahi (Coryphaena hippurus, ∼28 days of age) were exposed to control seawater or two concentrations of high-energy water accommodated fractions (HEWAFs; 5% or 10%) of Deepwater Horizon surface oil for 48 h. Deepwater Horizon crude oil is comprised of polycyclic aromatic hydrocarbons that cause a number of cardiotoxic effects in marine fishes across all levels of biological organization and at different life stages. Although cardiotoxic impacts have been widely reported, the mechanisms underlying these impairments in adult fish remain understudied. In this study, we examined the impacts of crude oil on cardiomyocyte contractility and electrophysiological parameters in freshly isolated ventricular cardiomyocytes from adult mahi-mahi (Coryphaena hippurus). Cardiomyocytes directly exposed to oil exhibited reduced contractility over a range of environmentally relevant concentrations (2.8-12.9 μg l-1∑PAH). This reduction in contractility was most pronounced at higher stimulation frequencies, corresponding to the upper limits of previously measured in situ mahi heart rates. To better understand the mechanisms underlying impaired contractile function, electrophysiological studies were performed, which revealed oil exposure prolonged cardiomyocyte action potentials and disrupted potassium cycling (9.9-30.4 μg l-1∑PAH). This study is the first to measure cellular contractility in oil-exposed cardiomyocytes from a pelagic fish. Results from this study contribute to previously observed impairments to heart function and whole-animal exercise performance in mahi, underscoring the advantages of using an integrative approach in examining mechanisms of oil-induced cardiotoxicity in marine fish.

Teleost fishes are diverse and successful, comprising almost half of all extant vertebrate species. It has been suggested that their success as a group is related, in part, to their unique O2 transport system, which includes pH-sensitive hemoglobin, a red blood cell β-adrenergic Na+/H+ exchanger (RBC β-NHE) that protects red blood cell pH, and plasma accessible carbonic anhydrase which is absent at the gills but present in some tissues, that short-circuits the β-NHE to enhance O2 unloading during periods of stress. However, direct support for this has only been examined in a few species of salmonids. Here, we expand the knowledge of this system to two warm-water, highly active marine percomorph fish, cobia (Rachycentron canadum) and mahi-mahi (Coryphaena hippurus). We show evidence for RBC β-NHE activity in both species, and characterize the Hb-O2 transport system in one of those species, cobia. We found significant RBC swelling following β-adrenergic stimulation in both species, providing evidence for the presence of a rapid, active RBC β-NHE in both cobia and mahi-mahi, with a time-course similar to that of salmonids. We generated oxygen equilibrium curves (OECs) for cobia blood and determined the P50, Hill, and Bohr coefficients, and used these data to model the potential for enhanced O2 unloading. We determined that there was potential for up to a 61% increase in O2 unloading associated with RBC β-NHE short-circuiting, assuming a - 0.2 ∆pHa-v in the blood. Thus, despite phylogenetic and life history differences between cobia and the salmonids, we found few differences between their Hb-O2 transport systems, suggesting conservation of this physiological trait across diverse teleost taxa.

Brevetoxins (PbTx) are a well-recognized group of neurotoxins associated with harmful algal blooms, and specifically recurrent "Florida Red Tides," in marine waters that are linked to impacts on both human and ecosystem health including well-documented "fish kills" and marine mammal mortalities in affected coastal waters. Understanding mechanisms and pathways of PbTx toxicity enables identification of relevant biomarkers to better understand these environmental impacts, and improve monitoring efforts, in relation to this toxin. Toward a systems-level understanding of toxicity, and identification of potential biomarkers, high-resolution magic angle spinning nuclear magnetic resonance (HRMAS NMR) was utilized for metabolic profiling of zebrafish (Danio rerio) embryos, as an established toxicological model, exposed to PbTx-2 (the most common congener in marine waters). Metabolomics studies were, furthermore, complemented by an assessment of the toxicity of PbTx-2 in embryonic stages of zebrafish and mahi-mahi (Coryphaena hippurus), the latter representing an ecologically and geographically relevant marine species of fish, which identified acute embryotoxicity at environmentally relevant (i.e., parts-per-billion) concentrations in both species. HRMAS NMR analysis of intact zebrafish embryos exposed to sub-lethal concentrations of PbTx-2 afforded well-resolved spectra, and in turn, identification of 38 metabolites of which 28 were found to be significantly altered, relative to controls. Metabolites altered by PbTx-2 exposure specifically included those associated with (1) neuronal excitotoxicity, as well as associated neural homeostasis, and (2) interrelated pathways of carbohydrate and energy metabolism. Metabolomics studies, thereby, enabled a systems-level model of PbTx toxicity which integrated multiple metabolic, molecular and cellular pathways, in relation to environmentally relevant concentrations of the toxin, providing insight to not only targets and mechanisms, but potential biomarkers pertinent to environmental risk assessment and monitoring strategies.

Progress towards the development of reliable hatchery technology for red snapper, Lutjanus campechanus, has been limited by inconsistent spawning and difficulties in larval rearing. We report on recent progress to improve this technology. A larval rearing trial assessing the effects of two different photoperiods was conducted at the University of Miami Experimental Hatchery (UMEH). Four 400-L tanks were subjected to a photoperiod of 16 h of full spectrum light, and four 400-L tanks were subjected to 16 h of full spectrum light and 8 h of dim light through 11 days post hatch (dph). S-strain rotifers, Brachionus rotundiformis, were used as the exclusive first feeding source and were maintained at a density of 25 ml−1 throughout the experimental period. Standard length, swim bladder inflation, and feed consumption were measured for each tank at 3, 6, 9 and 12 dph. Survival was quantified at 12 dph and at the completion of weaning onto dry feeds at 40 dph. No significant differences in growth, feed consumption, or survival rates were observed between treatments at the conclusion of the trial. However, tanks exposed to a treatment involving 8 h of dim light showed completion of swim bladder inflation in 92.5 ± 10.8 % of larvae sampled by 9 dph, while tanks maintained with 16 h of light and 8 h of darkness showed a significant reduction with only 60 ± 2.5 % swim bladder inflation at this sample date (p = 0.0131). By 12 dph, all larvae in all tanks showed >95 % swim bladder inflation success. Total survival across treatment groups through 13 dph was 66 ± 8.0 % and 4.6 ± 1.0 % at 40 dph when the larvae were fully weaned onto dry diets.

Pelagic fish embryos are thought to float in or near surface waters for the majority of their development and are presumed to have little to no control over their mobility, rendering these embryos at high risk for damages associated with surface stressors such as ultraviolet radiation (UVR). We recently challenged these long-standing paradigms by characterizing a potential mechanism of stressor avoidance in early-life stage mahi-mahi (Coryphaena hippurus) in which embryos sense external cues, such as UVR, and modify their buoyancy to reduce further exposure. It is unknown whether embryos of other marine fish with pelagic spawning strategies have similar capabilities. To fill this knowledge gap, we investigated buoyancy change in response to UVR in three additional species of marine fish that utilize a pelagic spawning strategy: yellowfin tuna (Thunnus albacares), red snapper (Lutjanus campechanus), and cobia (Rachycentron canadum). Embryos of all three species displayed increased specific gravity and loss of buoyancy after exposures to environmentally relevant doses of UVR, a response that may be ubiquitous to fish with pelagic embryos. To gain further insight into this response, we investigated recovery of buoyancy, oxygen consumption, energy depletion, and photolyase induction in response to UVR exposures in at least one of the three species listed above.

Mahi-mahi (Coryphaena hippurus) are a highly migratory pelagic fish, but little is known about what environmental factors drive their broad distribution. This study examined how temperature influences aerobic scope and swimming performance in mahi. Mahi were acclimated to four temperatures spanning their natural range (20, 24, 28, and 32{\deg}C; 5-27 days) and critical swimming speed (Ucrit), metabolic rates, aerobic scope, and optimal swim speed were measured. Aerobic scope and Ucrit were highest in 28{\deg}C-acclimated fish. 20{\deg}C-acclimated mahi experienced significantly decreased aerobic scope and Ucrit relative to 28{\deg}C-acclimated fish (57 and 28% declines, respectively). 32{\deg}C-acclimated mahi experienced increased mortality and a significant 23% decline in Ucrit, and a trend for a 26% decline in factorial aerobic scope relative to 28{\deg}C-acclimated fish. Absolute aerobic scope showed a similar pattern to factorial aerobic scope. Our results are generally in agreement with previously observed distribution patterns for wild fish. Understanding the orientation behavior and capabilities in early life history (ELH) of fishes is critical for studying their dispersal but has, surprisingly, never been tested in any pelagic species. We here investigate the ontogeny of orientation and swimming abilities of the pelagic Coryphaena hippurus Linnaeus, 1758 larvae, hereafter mahi-mahi, through their ELH stages using the Drifting In Situ Chamber (DISC) in a laboratory setup. The DISC was deployed in a large (3 m3) circular aquarium in order to control the stimulus perceived by the fish and to identify behavioral response at the individual, developmental stage, and population levels. A total of 79 individual ranging from 7 to 23 days post hatch and from preflexion to early juvenile stages were exposed to a directional light mimicking the sun&rsquo, s position. Orientation towards the light direction was tested by switching the light by 180&deg, among trials. To compare the orientation among development stages, we scaled the directionality by the swimming ability, therein defined as &ldquo, directionality competence&rdquo, The results show that while mahi-mahi directionality competence increases through ontogeny, they acquire a positive and directional phototaxis behavior at the flexion stage. This potential solar orientation is kept through the early juvenile stage. Mahi-mahi may thus be able to use the sun as a compass in the open ocean as observed in the larval stages of demersal species seeking for settlement habitats. This ability notably develops before the improvement of their swimming capabilities, suggesting that early orientation&mdash, even at micro-scale&mdash, and swimming capabilities may be equally important for the survival of the ELH stages of pelagic species.

Maintaining energy balance over a wide range of temperatures is critical for an active pelagic fish species such as the mahi-mahi (Coryphaena hippurus), which can experience rapid changes in temperature during vertical migrations. Due to the profound effect of temperature on mitochondrial function, this study was designed to investigate the effects of temperature on mitochondrial respiration in permeabilized heart and red skeletal muscle (RM) fibres isolated from mahi-mahi. As RM is thought to be more anatomically isolated from rapid ambient temperature changes compared to the myocardium, it was hypothesized that heart mitochondria would be more tolerant of temperature changes through a greater ability to match respiratory capacity to an increase in temperature and to maintain coupling, when compared to RM mitochondria. Results show that heart fibres were more temperature sensitive and increased respiration rate with temperature increases to a greater degree than RM. Respiratory coupling ratios at the three assay temperatures (20, 26, and 30 °C), revealed that heart mitochondria were less coupled at a lower temperature (26 °C) compared to RM mitochondria (30 °C). In response to an in vitro acute temperature challenge, both tissues showed irreversible effects, where both heart and RM increased uncoupling whether the assay temperature was acutely changed from 20 to 30 °C or 30 to 20 °C. The findings from this study indicate that mahi-mahi heart mitochondria were more temperature sensitive compared to those from RM.

This study examined potential interactive effects of co-exposure to Deepwater Horizon (DWH) crude oil (∼30 μg L−1 ΣPAHs) for 24 h and either hypoxia (2.5 mg O2 L−1; 40% O2 saturation) or elevated temperature (30 °C) on the swimming performance of juvenile mahi-mahi (Coryphaena hippurus). Additionally, effects of shorter duration exposures to equal or higher doses of oil alone either prior to swimming or during the actual swim trial itself were examined. Only exposure to hypoxia alone or combined with crude oil elicited significant decreases in critical swimming speed (Ucrit) and to a similar extent (∼20%). In contrast, results indicate that elevated temperature might ameliorate some effects of oil exposure on swimming performance and that effects of shorter duration exposures are either reduced or delayed.

BackgroundPsychiatric inpatient treatment is increasingly performed in settings with locked doors. However, locked wards have well-known disadvantages and are ethically problematic. In addition, recent data challenges the hypothesis that locked wards provide improved safety over open-door settings regarding suicide, absconding and aggression. Furthermore, there is evidence that the introduction of an open-door policy may lead to short-term reductions in involuntary measures. The aim of this study was to assess if the introduction of an open-door policy is associated with a long-term reduction of the frequency of seclusion and forced medication.MethodIn this 6-year, hospital-wide, longitudinal, observational study, we examined the frequency of seclusion and forced medication in 17,359 inpatient cases admitted to the Department of Adult Psychiatry, Universitäre Psychiatrische Kliniken (UPK) Basel, University of Basel, Switzerland. In an approach to enable a less restrictive policy, six previously closed psychiatric wards were permanently opened beginning from August 2011. During this process, a systematic change towards a more patient-centered and recovery-oriented care was applied. Statistical analysis consisted of generalized estimating equations (GEE) models.ResultsIn multivariate analyses controlling for potential confounders, the implementation of an open-door policy was associated with a continuous reduction of seclusion (from 8.2 to 3.5%; ηp2 = 0.82; odds ratio: 0.88) and forced medication (from 2.4 to 1.2%; ηp2 = 0.70; odds ratio: 0.90).ConclusionThis underlines the potential of the introduction of an open-door policy to attain a long-term reduction in involuntary measures.

With the expansion of marine aquaculture throughout the world comes a need for solutions to improve the economic viability and environmental sustainability of farming operations. Integrated multi-trophic aquaculture (IMTA) has been identified as a promising method of farming that typically combines multiple cultured species in connected systems in an effort to decrease the concentration of nutrients expelled as waste products from intensive finfish culture while also increasing commercial profits via subsequent extractive biomass production. This study examined the effectiveness of South Florida native red macroalgal species Agardhiella subulata for use as a productive, nutrient-extracting species for IMTA in the Atlantic, Gulf of Mexico, and Caribbean regions. When cultured in a pilot-scale, land-based IMTA system for replicated 15 day trials with American red snapper, Lutjanus campechanus, A. subulata grew up to 14.4% d−1 and significantly decreased the concentration of all measured dissolved inorganic nutrients in the effluent water. The average percent (% ± SE) removal of nutrients were: 68.8 ± 4.8 ammonia, 94.5 ± 1.3 nitrate, 96.6 ± 2.0 nitrite, and 93.2 ± 3.1 phosphate from primary culture effluents. Further evidence of A. subulata fitness for commercial IMTA practices is shown through significantly high bulk nitrogen tissue concentration of experimental (nutrient-enriched) compared to control (un-enriched) groups. Results of this study demonstrate the efficient growth and nutrient removal capabilities of A. subulata when used in IMTA applications with marine finfish production in warm water environments, as may be encountered in marine aquaculture projects in the Atlantic, Gulf of Mexico and Caribbean regions.

Aerobic exercise capacity is dependent on the cardiorespiratory system's ability to supply oxygen at a rate that meets energetic demands. In teleost fish crude oil exposure, with the associated polycyclic aromatic hydrocarbons (PAH's), reduces exercise performance and this has been hypothesized to be due to compromised cardiovascular function. In this study, we test this hypothesis by simultaneously measuring cardiovascular performance, oxygen consumption, and swim performance in a pelagic teleost, the cobia (Rachycentron canadum). Metabolic rate increased over 300% in both groups during the swim trial but as the fish approached the critical swim speed (Ucrit) MO2 was 12% lower in the oil exposed fish. Further, stroke volume was initially 35% lower while heart rate was 15% higher in the oil exposed compared to control fish. Our findings suggested, while aspects of cardiovascular and metabolic function are altered by oil exposure, additional studies are needed to further understand the homeostatic mechanisms that may sustain cardiovascular function at higher exercise intensities in cobia.

Successful culture of marine fish relies upon availability of high quality fertilized eggs obtained from broodstock. However, some of the most critical aspects of obtaining such eggs are often overlooked. These aspects include the capture, transport, acclimation, and spawning of sexually mature wild-caught fish. Mahi-mahi ( Coryphaena hippurus ), also known as dolphinfish, have been identified as one of the most promising candidate species for development of warm-water marine finfish aquaculture due to their high growth rate, market presence, and global distribution. In addition, mahi-mahi have proven to be a useful model species for physiology and environmental toxicology research, specifically in studies examining tropical and subtropical pelagic teleosts. One of the keys to aquaculture development of this species is the ability to obtain year-round production of fertilized embryos. This study documents the technical methods utilized to reach a point of consistent mahi-mahi egg production year-round, while also detailing the live transport tank and land-based spawning tank design, implementation, and operation. Following three different groups of wild-caught mahi-mahi broodstock from the point of capture throughout their lifespan, this study provides novel information on growth, survival, and spawning of this species in captivity. Results from this research have allowed for significant new insights into the effects of a variety of environmental stressors on the early life stages of this species. Furthermore, the ability to maintain consistent spawning populations of mahi-mahi in captivity has allowed for reliable and consistent production of fully-weaned fingerlings of this species, thereby resolving one of the key industry bottlenecks that has been limiting expansion of mahi-mahi commercial-scale aquaculture.

Mahi-mahi Coryphaena hippurus is a promising species for aquaculture development and has been used as a model species for oil toxicology and physiology studies. This species has one of the fastest growth rates of any marine teleost and a unique reproductive biology due to its high spawning frequency and reproductive energy allocation. These characteristics lend the species to being an excellent model for understanding broodstock nutrition for other high energetic pelagic species. Transport of coral reef fish larvae is driven by advection in ocean currents and larval swimming. However, for swimming to be advantageous, larvae must use external stimuli as guides. One potential stimulus is “odor” emanating from settlement sites (e.g., coral reefs), signaling the upstream location of desirable settlement habitat. However, specific chemicals used by fish larvae have not been identified. Dimethyl sulfide (DMS) is produced in large quantities at coral reefs and may be important in larval orientation. In this study, a choice-chamber (shuttle box) was used to assess preference of 28 pre-settlement stage larvae from reef fish species for seawater with DMS. Swimming behavior was examined by video-tracking of larval swimming patterns in control and DMS seawater. We found common responses to DMS across reef fish taxa - a preference for water with DMS and change in swimming behavior - reflecting a switch to “exploratory behavior”. An open water species displayed no response to DMS. Affinity for and swimming response to DMS would allow a fish larva to locate its source and enhance its ability to find settlement habitat. Moreover, it may help them locate prey accumulating in fronts, eddies, and thin layers, where DMS is also produced.

The understanding of the detection threshold and behavioral response of fishes in response to crude oil is critical to predicting the effects of oil spills on wild fish populations. The

Crude oil and polycyclic aromatic hydrocarbon (PAH) exposure in early life stage fish has been well-characterized to induce phenotypic malformations such as altered heart development and other morphological impacts. The effects of chemical oil dispersants on toxicity are more controversial. To better understand how chemical dispersion of oil can impact toxicity in pelagic fish, embryos of mahi-mahi (Coryphaena hippurus) were exposed to three concentrations of the chemical dispersant Corexit 9500A, or Corexit 9500A-oil mixtures (chemically enhanced water accommodated fractions: CEWAF) of Deepwater Horizon crude oil for 48 h. RNA sequencing, gene ontology enrichment, and phenotypic measurements were conducted to assess toxicity. Exposure to Corexit 9500A altered expression of less than 50 genes at all concentrations (2.5, 5, and 10 mg/L nominal concentration) and did not induce acute mortality or phenotypic malformations, corroborating other studies showing minimal effects of Corexit 9500A on developing mahi-mahi embryos. CEWAF preparations contained environmentally relevant ∑PAH concentrations ranging from 1.4 to 3.1 μg/L and similarly did not alter larval morphology. Differentially expressed genes and significantly altered pathways related to cardiotoxicity, visual impairments, and Ca2+ homeostasis reinforced previous work that expression of genes associated with the heart and eye are highly sensitive molecular endpoints in oil-exposed early life stage fish. Differential expression and gene ontology pathways were similar across the three CEWAF treatments, indicating that increased chemical dispersion did not alter molecular outcomes within the range tested here. In addition, significant sublethal molecular responses occurred in the absence of observable phenotypic changes to the heart, indicating that effects of oil on early life stage fish may not be completely dependent on cardiac function.

Many ecologically important fishes, including mahi-mahi (Coryphaena hippurus), and their offspring were directly exposed to crude oil following the Deepwater Horizon (DWH) oil spill. Early life stage fish are especially vulnerable to the toxicity of crude oil-derived polycyclic aromatic hydrocarbons (PAHs). In teleosts, yolk sac proteins are the main energy source during development and are usually catabolized into ammonia or urea among other byproducts. Although excretion of these waste products is sensitive to oil exposure, we know little about the underlying mechanisms of this process. In this study, we examined the effects of crude oil on ammonia and urea handling in the early life stages of mahi. Mahi embryos exposed to 30–32 μg L−1 ∑PAH exhibited increased urea excretion rates and greater accumulation of urea in the tissues before hatch suggesting that ammonia, which is highly toxic, was converted into less-toxic urea. Oil-exposed embryos (6.3–32 μg L−1 ∑PAH) displayed significantly increased tissue ammonia levels at 42 hpf and upregulated mRNA levels of ammonia transporters (Rhag, Rhbg and Rhcg1) from 30 to 54 hpf. However, despite increased accumulation and higher expression of ammonia transporters, the larvae exposed to higher ∑PAH (30 μg L−1 ∑PAH) showed reduced ammonia excretion rates after hatch. Together, the increased production of nitrogenous waste reinforces previous work that increased energy demand in oil-exposed embryos is fueled, at least in part, by protein metabolism and that urea synthesis plays a role in ammonia detoxification in oil-exposed mahi embryos. To our knowledge, this study is the first to combine physiological and molecular approaches to assess the impact of crude-oil on both nitrogenous waste excretion and accumulation in the early life stages of any teleosts.

Background Mahi‐mahi (Coryphaena hippurus) is a commercially and ecologically important fish species that is widely distributed in tropical and subtropical waters. Biological attributes and reproductive capacities of mahi‐mahi make it a tractable model for experimental studies. In this study, life development of cultured mahi‐mahi from the zygote stage to adult has been described. Results A comprehensive developmental table has been created reporting development as primarily detailed observations of morphology. Additionally, physiological, behavioral, and molecular landmarks have been described to significantly contribute in the understanding of mahi life development. Conclusion Remarkably, despite the vast difference in adult size, many developmental landmarks of mahi map quite closely onto the development and growth of Zebrafish and other warm‐water, active Teleost fishes., Key Findings Mahi‐mahi is a tractable model for experimental studies high‐performance pelagic predatory fish species.Biological attributes of mahi are reported in a comprehensive developmental table.Physiological, behavioral and molecular landmarks are described through the life development.Mahi has a rapid growth rate, but the developmental marks are similar to other teleost fishes.

In this study, we investigated the effect of acute increases in temperature on cardiovascular function of mahi-mahi (Coryphaena hippurus). We also describe, for the first time, an artery that supplies the gastrointestinal tract that originates from the fourth branchial artery. We used vascular casting to verify the anatomical location of this unique celiaco-mesenteric artery. We predicted that blood flow in this vessel would be correlated with the digestive state of the animal. Increasing water temperature from 25.0 to 30.5 °C resulted in a linear increase in heart rate (fH) from 165 ± 4 beats∙min−1to 232 ± 7 beats∙min−1. Over this temperature range, fH strongly correlated with water temperature (R2 = 0.79). At 31 °C fH no longer correlated with water temperature, and at 34 °C fH had dropped to 114 ± 19 beats∙min−1. Furthermore, we found that mahi are capable of maintaining constant cardiac output over a temperature range from 25 to 31 °C. Cardiac function appeared to be compromised at temperatures >31 °C. In fed anesthetized fish, blood flow was pulsatile in the celiaco-mesenteric artery and was not in fasted fish. In fed fish, blood flow in the left celiaco-mesenteric artery was 1.99 ± 0.78 ml·min−1·kg−1 compared to the total cardiac output of 168.6 ± 12.7 ml·min−1·kg−1. The data suggest that mahi can differentially regulate gastric blood flow based on feeding state, which may explain the high digestive efficiency and very high growth rates of these pelagic predators.

Windows of exposure to a weathered Deepwater Horizon oil sample (slick A) were examined for early life stage mahi-mahi (Coryphaena hippurus) to determine whether there are developmental periods of enhanced sensitivity during the course of a standard 96-h bioassay. Survival was assessed at 96 h following oil exposures ranging from 2 h to 96 h and targeting 3 general periods of development, namely the prehatch phase, the period surrounding hatch, and the posthatch phase. In addition, 3 different oil preparations were used: high- and low-energy water accommodated fractions of oil and very thin surface slicks of oil (∼1 μm). The latter 2 were used to distinguish between effects due to direct contact with the slick itself and the water underlying the slick. Considering the data from all 3 exposure regimes, it was determined that the period near or including hatch was likely the most sensitive. Furthermore, toxicity was not enhanced by direct contact with slick oil. These findings are environmentally relevant given that the concentrations of polycyclic aromatic hydrocarbons eliciting mortality from exposures during the sensitive periods of development were below or near concentrations measured during the active spill phase. The Deepwater Horizon oil spill in 2010 overlapped with the spawning of many pelagic fish species in the Gulf of Mexico, including mahi-mahi (Coryphaena hippurus). Polycyclic aromatic hydrocarbons (PAHs) released during the spill have been shown to cause photo-induced toxicity under ultraviolet (UV) radiation. In the present study, mahi-mahi embryos were exposed to high-energy water accommodated fractions of source and naturally weathered oils for up to 48 h. The timing of co-exposure with UV radiation varied between an early development exposure for 8 h or a late development exposure for 8 h. The UV co-exposure had a photo-induced toxic effect on hatching success for all oil types and exposure scenarios. A more sensitive developmental window to photo-induced toxicity was observed when UV exposure occurred late in development. Source Oil B (SOB) was over six fold more toxic and Massachusetts source oil (MASS) oil was 1.6 fold more toxic when the embryos were co-exposed to UV light late in development. Further, weathered oil from the surface (OFS) co-exposure with UV late in development resulted in bradycardia in the mahi-mahi. The present study provides evidence that the developmental window when UV co-exposure occurs has a significant effect on the degree of photo-induced toxicity of oil, as well as provides evidence that UV co-exposure may exacerbate long-term cardiac effects in developing fish. Exposure to polycyclic aromatic hydrocarbons (PAH) negatively impacts exercise performance in fish species but the physiological modifications that result in this phenotype are poorly understood. Prior studies have shown that embryonic and juvenile mahi–mahi (Coryphaeus hippurus) exposed to PAH exhibit morphological abnormalities, altered cardiac development and reduced swimming performance. It has been suggested that cardiovascular function inhibited by PAH exposure accounts for the compromised exercise performance in fish species. In this study we used in-situ techniques to measure hemodynamic responses of young adult mahi–mahi exposed to PAH for 24 h. The data indicate that stroke volume was reduced 44% in mahi–mahi exposed to 9.6 ± 2.7 μg l−1 geometric mean PAH (∑PAH) and resulted in a 39% reduction in cardiac output and a 52% reduction in stroke work. Maximal change in pressure over change in time was 28% lower in mahi–mahi exposed to this level of ∑PAH. Mean intraventricular pressures and heart rate were not significantly changed. This study suggests exposure to environmentally relevant PAH concentrations impairs aspects of cardiovascular function in mahi–mahi.

Key differences in the developmental process of pelagic fish embryos, in comparison to embryos of standard test fish species, present challenges to obtaining sufficient control survival needed to successfully perform traditional toxicity testing bioassays. Many of these challenges relate to the change in buoyancy, from positive to negative, of pelagic fish embryos that occurs just prior to hatch. A novel exposure system, the pelagic embryo-larval exposure chamber (PELEC), has been developed to conduct successful bioassays on the early life stages (ELSs; embryos/larvae) of pelagic fish. Using this unique recirculating upwelling system, it was possible to significantly improve control survival in pelagic fish ELS bioassays compared to commonly used static exposure methods. Results demonstrate that control performance of mahi-mahi (Coryphaena hippurus) embryos in the PELEC system, measured as percent survival after 96-hrs, significantly outperformed agitated static exposure and static exposure systems. Similar significant improvements in 72-hr control survival were obtained with yellowfin tuna (Thunnus albacares). The PELEC system was subsequently used to test the effects of photo-induced toxicity of crude oil to mahi-mahi ELSs over the course of 96-hrs. Results indicate a greater than 9-fold increase in toxicity of Deepwater Horizon (DWH) crude oil during co-exposure to ambient sunlight compared to filtered ambient sunlight, revealing the importance of including natural sunlight in 96-hr DWH crude oil bioassays as well as the PELEC system's potential application in ecotoxicological assessments.
