Abstract The cylindrical drift chamber is the most innovative part of the MEG II detector, the upgraded version of the MEG experiment. The MEG II chamber differs from the MEG one because it is a single volume cylindrical structure, instead of a segmented one, chosen to improve its resolutions and efficiency in detecting low energy positrons from muon decays at rest. In this paper, we show the characteristics and performances of this fundamental part of the MEG II apparatus and we discuss the impact of its higher resolution and efficiency on the sensitivity of the MEG II experiment. Because of its innovative structure and high quality resolution and efficiency the MEG II cylindrical drift chamber will be a cornerstone in the development of an ideal tracking detector for future positron-electron collider machines.

Abstract The MEG II experiment, based at the Paul Scherrer Institut in Switzerland, reports the result of a search for the decay $$\upmu ^+ \rightarrow {\textrm{e}}^+ \upgamma $$ μ + → e + γ from data taken in the first physics run in 2021. No excess of events over the expected background is observed, yielding an upper limit on the branching ratio of $${\mathcal {B}} (\upmu ^+ \rightarrow {\textrm{e}}^+ \upgamma ) < 7.5 \times 10^{-13}$$ B ( μ + → e + γ ) < 7.5 × 10 - 13 (90% CL). The combination of this result and the limit obtained by MEG gives $${\mathcal {B}} (\upmu ^+ \rightarrow {\textrm{e}}^+ \upgamma ) < 3.1 \times 10^{-13}$$ B ( μ + → e + γ ) < 3.1 × 10 - 13 (90% CL), which is the most stringent limit to date. A ten-fold larger sample of data is being collected during the years 2022–2023, and data-taking will continue in the coming years.

Whenever oral treatment or botulinum toxin injections fail to control severe spasticity, a trial with intrathecal baclofen is recommended no earlier than 1 year after brain injury. When irreversible contractures are to be avoided, such a trial might be done earlier. Some have briefly reported cognitive modifications with this treatment.During the trial period, intrathecal baclofen is continuously infused by a portable external pump through an intrathecal catheter. The daily dose is adjusted according to the clinical response. If the expected response is obtained by reduction of spasticity, a programmable pump is then implanted. Throughout the procedure, close neuropsychological follow-up is pursued.Two persons with extremely severe brain injury and spasticity received a programmable pump less than 10 months after trauma. Unexpectedly, one emerged from the minimally conscious state and the other from post-traumatic amnesia.Intrathecal baclofen should be considered within the first year after brain injury whenever spasticity does not respond to medication. ITB lessens the degree of spasticity which in turn facilitates care and, thus, has the potential to limit contractures. After severe brain injury, this treatment might trigger recovery from altered states of consciousness, improve cognition and facilitate rehabilitation.

AIMS: To evaluate the value of our driving simulator in deciding whether or not to allow patients with physical and/or cognitive deficits to resuming driving and to analyze whether or not the medical expert's final decision is based more on the results of the driving simulator than those of the neuropsychological examination. METHODS: One hundred and twenty-three patients were evaluated with the driving simulator. Thirty-five of those with cognitive deficits also underwent a neuropsychological examination prior to the medical expert's decision on driving aptitude. In cases of uncertainty or disagreement, a driving assessment in real conditions was performed by a driving instructor. RESULTS: In cases of physical handicap, the medical expert's decision concurred with that of the occupational therapist. For brain-injured patients, there was a significant correlation between the neuropsychologist's opinion and that of the occupational therapist (kappa=0.33; P=0.01). However, the sensibility and specificity were only 55 and 80%, respectively. In this paper we present the results of a search for nuclearites in the penetrating cosmic radiation using the scintillator and track-etch subdetectors of the MACRO apparatus. The analyses cover the beta = v/c range at the detector depth (3700 hg/cm(2)) 10(-5) < beta < 1; for beta = 2 x 10(-3) the flux limit is 2.7 x 10(-16) cm(-2)s(-1)sr(-1) for an isotropic flux of nuclearites, and twice this value for a flux of downgoing nuclearites.

We present the final results of the search for stellar gravitational collapses obtained by the MACRO experiment. The detector was active for a stellar collapse search for more than 11 years and it was sensitive to collapses occurring all over in our galaxy for 8.6 years. A real time system for a prompt recognition of neutrino bursts was developed and was operating on-line for almost the whole life of the experiment. No signal compatible with a neutrino burst from a galactic supernova was observed.

The primary cosmic ray (CR) proton, helium and CNO fluxes in the energy range 80-300 TeV are studied at the National Gran Sasso Laboratories by means of EAS-TOP (Campo Imperatore, 2005 m a.s.l.) and MACRO (deep underground, 3100 m w.e., the surface energy threshold for a muon reaching the detector being Eμth~1.3 TeV). The measurement is based on: (a) the selection of primaries based on their energy/nucleon (i.e., with energy/nucleon sufficient to produce a muon with energy larger than 1.3 TeV) and the reconstruction of the shower geometry by means of the muons recorded by MACRO in the deep underground laboratories; (b) the detection of the associated atmospheric Cherenkov light (C.l.) signals by means of the C.l. detector of EAS-TOP. The C.l. density at core distance r>100 m is directly related to the total primary energy E0. Proton and helium (``p+He'') and proton, helium and CNO (``p+He+CNO'') primaries are thus selected at E0~=80 TeV, and at E0~=250 TeV, respectively. Their flux is measured: Jp+He(80TeV)=(1.8+/-0.4)×10-6 m-2s-1sr-1TeV-1, and Jp+He+CNO(250TeV)=(1.1+/-0.3)×10-7 m-2s-1sr-1TeV-1, their relative weights being: Jp+He/Jp+He+CNO(250TeV)=0.78+/-0.17. By using the measurements of the proton spectrum obtained from the direct experiments and hadron flux data in the atmosphere, we obtain for the relative weights of the three components at 250 TeV: Jp:JHe:JCNO=(0.20+/-0.08):(0.58+/-0.19):(0.22+/-0.17). This corresponds to the dominance of helium over proton primaries at 100-1000 TeV, and a possible non-negligible contribution from CNO. The lateral distribution of Cherenkov light in Extensive Air Showers (EASs), which is related to the rate of energy deposit of the primary in the atmosphere, is measured for a selected proton and helium primary beam, and good agreement is found when compared with the one calculated with the CORSIKA/QGSJET simulation model.

To evaluate the value of our driving simulator in deciding whether or not to allow patients with physical and/or cognitive deficits to resuming driving and to analyze whether or not the medical expert's final decision is based more on the results of the driving simulator than those of the neuropsychological examination.One hundred and twenty-three patients were evaluated with the driving simulator. Thirty-five of those with cognitive deficits also underwent a neuropsychological examination prior to the medical expert's decision on driving aptitude. In cases of uncertainty or disagreement, a driving assessment in real conditions was performed by a driving instructor.In cases of physical handicap, the medical expert's decision concurred with that of the occupational therapist. For brain-injured patients, there was a significant correlation between the neuropsychologist's opinion and that of the occupational therapist (kappa=0.33; P=0.01). However, the sensibility and specificity were only 55 and 80%, respectively. The correlation between an occupational therapy decision based on the driving simulator and that of the medical expert was very significant (kappa=0.81; P0.0001) and the sensibility and specificity were 84 and 100%, respectively. In contrast, these values were lower (63 and 71%, respectively) for the correlation between the neuropsychologist's opinion and that of the medical expert.Our driving simulator enables the danger-free evaluation of driving aptitude. The results mirror an in situ assessment and are more sensitive than neuropsychological examination. In fact, the neuropsychologist's opinion often is more negative or uncertain with respect to the patient's real driving aptitude. When taking a decision on a patient's driving aptitude, the medical expert is more inclined to trust the results of the driving simulator.

Abstract We present the first direct search for lepton flavour violating muon decay mediated by a new light particle X, $$\upmu ^+ \rightarrow \mathrm {e}^+\mathrm {X}, \mathrm {X} \rightarrow \upgamma \upgamma \ $$ μ + → e + X , X → γ γ . This search uses a dataset resulting from $$7.5\times 10^{14}$$ 7.5 × 10 14 stopped muons collected by the MEG experiment at the Paul Scherrer Institut in the period 2009–2013. No significant excess is found in the mass region 20–45 MeV/c $$^2$$ 2 for lifetimes below 40 ps, and we set the most stringent branching ratio upper limits in the mass region of 20–40 MeV/c $$^2$$ 2 , down to $${\mathcal {O}}(10^{-11})$$ O ( 10 - 11 ) at 90% confidence level.

High energy gamma ray astronomy is now a well established field and several sources have been discovered in the region from a few GeV up to several TeV. If sources involving hadronic processes exist, the production of photons would be accompanied by neutrinos too. Other possible neutrino sources could be related to the annihilation of WIMPs at the center of galaxies with black holes. We present the results of a search for point-like sources using 1100 upward-going muons produced by neutrino interactions in the rock below and inside the MACRO detector in the underground Gran Sasso Laboratory. These data show no evidence for a possible neutrino point-like source or for possible correlations between gamma ray bursts and neutrinos. They have been used to set flux upper limits for candidate point-like sources which are in the range 10^-14-10^-15 cm-2 s-1., Comment: 37 pages, 15 figures, replacement due to a typo in tab. 6, AASLaTex, submitted to ApJ

Abstract The MEG experiment, designed to search for the $${\mu ^+ \rightarrow \hbox {e}^+ \gamma }$$ μ+→e+γ decay, completed data-taking in 2013 reaching a sensitivity level of $${5.3\times 10^{-13}}$$ 5.3×10-13 for the branching ratio. In order to increase the sensitivity reach of the experiment by an order of magnitude to the level of $$6\times 10^{-14}$$ 6×10-14 , a total upgrade, involving substantial changes to the experiment, has been undertaken, known as MEG II. We present both the motivation for the upgrade and a detailed overview of the design of the experiment and of the expected detector performance.

Comparing the frequency of two distant optical clocks will enable sensitive tests of fundamental physics. Here, the authors compare two strontium optical-lattice clocks 690 kilometres apart to a degree of accuracy that is limited only by the uncertainty of the individual clocks themselves.

Lepton Flavour Violation in the charged lepton sector (CLFV) is forbidden in the Minimal Standard model and strongly suppressed in extensions of the model to include finite neutrino mixing. On the other hand, a wide class of Supersymmetric theories, even coupled with Grand Unification models (SUSY-GUT models), predict CLFV processes at a rate within the reach of new experimental searches operated with high resolution detectors at high intensity accelerators. As the Standard model background is negligible, the observation of one or more CLFV events would provide incontrovertible evidence for physics beyond Standard model, while a null effect would severely constrain the set of theory parameters. Therefore, a big experimental effort is currently (and will be for incoming years) accomplished to achieve unprecedented sensitivity on several CLFV processes. In this paper we review past and recent results in this research field, with focus on CLFV channels involving muons and tau's. We present currently operating experiments as well as future projects, with emphasis laid on how sensitivity enhancements are accompanied by improvements on detection techniques. Limitations due to systematic effects are also discussed in detail together with the solutions being adopted to overcome them.

The final analysis of atmospheric neutrino events collected with the MACRO detector is presented. Three different classes of events, generated by neutrinos in different energy ranges, are studied looking at rates, angular distributions and estimated energies. The results are consistent for all the subsamples and indicate a flux deficit that depends on energy and path length of neutrinos. The no -oscillation hypothesis is excluded at about 5 standard deviations, while the hypothesis of muon neutrino oscillating in tau neutrino gives a satisfactory description of all data. The parameters with highest probability in a two flavour scenario are maximum mixing and Delta m^2 = 0.0023 eV^2. This result is independent of the absolute normalization of the atmospheric neutrino fluxes. The data can also be used to put experimental constrain on this normalization.

Many galactic and extragalactic astrophysical sources are currently considered promising candidates as high energy neutrino emitters. Astrophysical neutrinos can be detected as upward-going muons produced in charged-current interactions with the medium surrounding the detector. The expected neutrino fluxes from various models start to dominate on the atmospheric neutrino background at neutrino energies above some tens of TeV. We present the results of a search for an excess of high energy upward-going muons among the sample of data collected by MACRO during ~5.8 years of effective running time. No significant evidence for this signal was found. As a consequence, an upper limit on the flux of upward-going muons from high-energy neutrinos was set at the level of 1.7 10^(-14) cm^(-2) s^(-1) sr^(-1). The corresponding upper limit for the diffuse neutrino flux was evaluated assuming a neutrino power law spectrum. The primary cosmic ray (CR) proton, helium and CNO fluxes in the energy range 80-300 TeV are studied at the National Gran Sasso Laboratories by means of EAS-TOP (Campo Imperatore, 2005 m a.s.l.) and MACRO (deep underground, 3100 m w.e., the surface energy threshold for a muon reaching the detector being E-mu(th) approximate to 1.3 TeV). The measurement is based on: (a) the selection of primaries based on their energy/nucleon (i.e., with energy/nucleon sufficient to produce a muon with energy larger than 1.3 TeV) and the reconstruction of the shower geometry by means of the muons recorded by MACRO in the deep underground laboratories; (b) the detection of the associated atmospheric Cherenkov light (C.l.) signals by means of the C.l. detector of EAS-TOP. The C.l. density at core distance r > 100 m is directly related to the total primary energy E-0. Proton and helium ("p + He") and proton, helium and CNO ("p + He + CNO") primaries are thus selected at E-0 approximate to 80 TeV, and at E-0 similar or equal to 250 TeV, respectively. Their flux is measured: J(p+He)(80 TeV) = (1.8 +/- 0.4) x 10(-6) m(-1)-s(-1) sr(-1) TeV-1, and J(p+He+CNO)(250 TeV) = (1.1 +/- 0.3) x 10(-7) m(-2)-s(-1) sr(-1) TeV-1, their relative weights being J(p+He)(J(p+He+CNO)) over bar (250 TeV) = 0.78 +/- 0.17. By using the measurements of the proton spectrum obtained from the direct experiments and hadron flux data in the atmosphere, we obtain for the relative weights of the three components at 250 TeV: J(p) : J(He) : J(CNO) = (0.20 +/- 0.08) : (0.58 +/- 0.19) : (0.22 +/- 0.17). This corresponds to the dominance of helium over proton primaries at 100-1000 TeV, and a possible non-negligible contribution from CNO. The lateral distribution of Cherenkov light in Extensive Air Showers (EASs), which is related to the rate of energy deposit of the primary in the atmosphere, is measured for a selected proton and helium primary beam, and good agreement is found when compared with the one calculated with the CORSIKA/QGSJET simulation model. We present the final results of the search for stellar gravitational collapses obtained by the MACRO experiment. The detector was active for a stellar collapse search for more than 11 years and it was sensitive to collapses occurring all over in our galaxy for 8.6 years. A real time system for a prompt recognition of neutrino bursts was developed and was operating on-line for almost the whole life of the experiment. No signal compatible with a neutrino burst from a galactic supernova was observed.

The cosmic ray primary composition in the energy range between 10**15 and 10**16 eV, i.e., around the "knee" of the primary spectrum, has been studied through the combined measurements of the EAS-TOP air shower array (2005 m a.s.l., 10**5 m**2 collecting area) and the MACRO underground detector (963 m a.s.l., 3100 m w.e. of minimum rock overburden, 920 m**2 effective area) at the National Gran Sasso Laboratories. The used observables are the air shower size (Ne) measured by EAS-TOP and the muon number (Nmu) recorded by MACRO. The two detectors are separated on average by 1200 m of rock, and located at a respective zenith angle of about 30 degrees. The energy threshold at the surface for muons reaching the MACRO depth is approximately 1.3 TeV. Such muons are produced in the early stages of the shower development and in a kinematic region quite different from the one relevant for the usual Nmu-Ne studies. The measurement leads to a primary composition becoming heavier at the knee of the primary spectrum, the knee itself resulting from the steepening of the spectrum of a primary light component (p, He). The result confirms the ones reported from the observation of the low energy muons at the surface (typically in the GeV energy range), showing that the conclusions do not depend on the production region kinematics. Thus, the hadronic interaction model used (CORSIKA/QGSJET) provides consistent composition results from data related to secondaries produced in a rapidity region exceeding the central one. Such an evolution of the composition in the knee region supports the "standard" galactic acceleration/propagation models that imply rigidity dependent breaks of the different components, and therefore breaks occurring at lower energies in the spectra of the light nuclei.

We present the final results of the search for exotic massive particles in the cosmic radiation performed with the MACRO underground experiment. Magnetic monopoles and nuclearites flux upper limits obtained with the CR39 nuclear track subdetector, the scintillation and streamer tube subdetectors are given. Searches at high altitude with the SLIM experiment are in progress.

A search for lightly ionizing particles has been performed with the MACRO detector. This search was sensitive to particles with charges between $\fractional{5}$$e$ and close to the charge of an electron, with $\beta$ between approximately 0.25 and 1.0. Unlike previous searches both single track events and tracks buried within high multiplicity muon showers were examined. In a period of approximately one year no candidates were observed. Assuming an isotropic flux, for the single track sample this corresponds to a 90% C.L. upper flux limit $\Phi \leq 9.2 \times 10^{-15} cm^{-2} sec^{-1} sr^{-1}$ ., Comment: 15 pages, 7 figures, to be submitted to Phys. In this paper, the first of a two-part work, we present the reconstruction and measurement of muon events detected underground by the MACRO experiment at Gran Sasso (E-mu greater than or equal to 1.3 TeV in atmosphere). The main aim of this work is to discuss the muon multiplicity distribution as measured in the detector. The data sample analyzed consists of 4.4 x 10(6) muon events, of which similar to 263 000 are multiple muons, corresponding to a total live time of 5850 h. In this sample, the observed multiplicities extend above N-mu = 35, with intermuon separations up to 50 m and beyond. Additional complementing measurements, such as the inclusive muon Aux, the angular distribution, and the muon separation distribution (decoherence), are also included. The physical interpretation of the results presented here is reported in the following companion paper.

Upward-going muon events collected with the lower half of the MACRO detector have been analyzed in order to search for Weakly Interacting Massive Particles possibly trapped in the core of the Sun and of the Earth. No statistically significant signal could be discriminated over the possible fluctuations of the atmospheric neutrino background. Assuming a particular neutralino model, the explorable neutralino mass region has been considered.

A study of the primary cosmic ray composition in the energy range 5ṡ1014−5ṡ1015 eV is performed through the analysis of the deep underground muons and of the e.m. component of Extensive Air Showers detected in coincidence by the MACRO and EAS-TOP experiments at the Gran Sasso Laboratory. We conclude that: a) pure proton and Iron primary beams are excluded by the experimental data; b) a fraction of nuclei heavier than Helium is necessary to explain the experimental data both below and above the knee; c) a mixed composition, obtained a priori from extrapolations of the spectra directly measured below 100 TeV, yields a reasonable account of the observations up to the knee of the primary spectrum.

The MACRO experiment has been running as a supernova neutrino detector since 1989 and is sensitive to the whole galaxy since the beginning of 1992. A galactic supernova would produce some hundreds of events in the detector, We describe our stellar gravitational collapse online monitors and alarm system, and present the results of a search for neutrino bursts from supernovae during a period of 1.5 yr. - Abstract
Background: Whenever oral treatment or botulinum toxin injections fail to control severe spasticity, a trial with intrathecal baclofen is recommended no earlier than 1 year after brain injury. When irreversible contractures are to be avoided, such a trial might be done earlier. Some have briefly reported cognitive modifications with this treatment., Methods: During the trial period, intrathecal baclofen is continuously infused by a portable external pump through an intrathecal catheter. The daily dose is adjusted according to the clinical response. If the expected response is obtained by reduction of spasticity, a programmable pump is then implanted. Throughout the procedure, close neuropsychological follow-up is pursued., Results: Two persons with extremely severe brain injury and spasticity received a programmable pump less than 10 months after trauma. Unexpectedly, one emerged from the minimally conscious state and the other from post-traumatic amnesia., Conclusions: Intrathecal baclofen should be considered within the first year after brain injury whenever spasticity does not respond to medication. ITB lessens the degree of spasticity which in turn facilitates care and, thus, has the potential to limit contractures. After severe brain injury, this treatment might trigger recovery from altered states of consciousness, improve cognition and facilitate rehabilitation.

- Abstract
Background: Cardiac computed tomographic angiography (CCTA) permits simultaneous assessment of coronary artery disease (CAD) and left ventricular mass (LVM). While increased LVM predicts mortality and is associated with obstructive CAD, the relationship of LVM with non-obstructive CAD is unknown., Methods: We evaluated 212 consecutive patients undergoing 64-detector row CCTA at 2 sites without evident cardiovascular disease or obstructive (≥70%) CAD by CCTA. LVM was measured by CCTA using Simpson's method of disks and indexed to body surface area (LVMI) and height to the allometric power of 2.7(LVM/ht2.7). CCTAs were evaluated by scoring a modified AHA 16-segment coronary artery model for none = 0 (0% stenosis), mild = 1 (1-49% stenosis) or moderate = 2 (50-69% stenosis). Overall CAD plaque burden was estimated by summing scores across all segments for a segment stenosis score (SSS, max = 32)., Results: The mean age was 53.3 ± 12.8 with 52% female, 48% hypertensive, and 7.4% diabetic. The mean LVM was 109 ± 32.5 g; 58.5% had any coronary artery plaque. In multivariable linear regression, SSS was significantly associated with increased LVM, LVMI and LVM/ht2.7. LVM increased by 2.0 g for every 1-point increase in SSS (95% CI 0.06-3.4, p = 0.006). Agatston scores provided no additional predictive value for increased LVM above and beyond SSS., Conclusion: Non-obstructive CAD visualized by CCTA is associated with increased LVM independent of effects of clinical risk factors and calcium scoring. Whether addition of LVM to stenosis assessment in patients undergoing CCTA enhances risk prediction of future CAD events warrants investigation., (Copyright © 2011 Society of Cardiovascular Computed Tomography. Aims: To evaluate the value of our driving simulator in deciding whether or not to allow patients with physical and/or cognitive deficits to resuming driving and to analyze whether or not the medical expert's final decision is based more on the results of the driving simulator than those of the neuropsychological examination., Methods: One hundred and twenty-three patients were evaluated with the driving simulator. Thirty-five of those with cognitive deficits also underwent a neuropsychological examination prior to the medical expert's decision on driving aptitude. In cases of uncertainty or disagreement, a driving assessment in real conditions was performed by a driving instructor., Results: In cases of physical handicap, the medical expert's decision concurred with that of the occupational therapist. For brain-injured patients, there was a significant correlation between the neuropsychologist's opinion and that of the occupational therapist (kappa=0.33; P=0.01). However, the sensibility and specificity were only 55 and 80%, respectively. The correlation between an occupational therapy decision based on the driving simulator and that of the medical expert was very significant (kappa=0.81; P<0.0001) and the sensibility and specificity were 84 and 100%, respectively. In contrast, these values were lower (63 and 71%, respectively) for the correlation between the neuropsychologist's opinion and that of the medical expert., Conclusion: Our driving simulator enables the danger-free evaluation of driving aptitude. The results mirror an in situ assessment and are more sensitive than neuropsychological examination. In fact, the neuropsychologist's opinion often is more negative or uncertain with respect to the patient's real driving aptitude. When taking a decision on a patient's driving aptitude, the medical expert is more inclined to trust the results of the driving simulator.

- Abstract
Specificity for double-stranded DNA can arise due to somatic mutations within one of the branches of an autoreactive B cell clone. However, it is not known whether a different autospecificity predates anti-dsDNA and whether separate offshoots of an expanding B cell clone retain or evolve alternative specificities. We compared 3H9, an anti-dsDNA IgG, to 4H8 and 1A11, antibodies produced by hybridomas representing an alternative branch of the 3H9 B cell clone. All three IgG bound chromatin in ELISA and apoptotic cells in confocal microscopy, yet only 3H9 bound dsDNA, as measured by plasmon resonance. Moreover, we demonstrate that despite the unique specificity of 3H9 for dsDNA, all three clone members exhibited indistinguishable binding to chromatin. The binding to chromatin and apoptotic cells was unaffected by N-linked glycosylation in L chain CDR1, a modification that results from a replacement of serine 26 with asparagine in 4H8 and 1A11. These data provide the first evidence that specificity for nucleosome epitopes on apoptotic cells provides the initial positive stimulus for somatic variants that comprise a B cell clone, including those that subsequently acquire specificity for dsDNA. Conversely, selection of autoreactive B cells for binding to apoptotic cells leads to clonal expansion, antibody diversification, and the development of linked sets of anti-nuclear autoantibodies.

In antiphospholipid syndrome (APS) patients, some antiphospholipid antibodies (APA) are directed against negatively-charged phospholipids, while other APA are specific for phospholipid-proteins such as beta2-glycoprotein I (beta2GPI). Increased levels of oxidized low density lipoproteins (oxLDL) are present in atherosclerosis patients and these patients develop anti-oxLDL antibodies. Several studies have reported cross-reactivities between APA and anti-oxLDL antibodies, and some authors have suggested that most APA are specific for oxidized forms of phospholipids. In contrast, other studies report that APA react with both reduced and oxidized cardiolipin. In this context, we have re-examined the literature surrounding antibodies found in atherosclerosis and the APS. We have also included results from a panel of anti-phospholipid monoclonal antibodies from W/B mice, an APS model, which indicates that these antibodies do not display any preference for oxidized epitopes on lipid molecules.

- Abstract
Antiphospholipid antibodies (APA) are present in a variety of autoimmune disorders, including systemic lupus erythematosus (SLE) and the antiphospholipid syndrome (APS). Increasing evidence suggests that a subset of APA can also be detected in patients with atherosclerosis. In this review, we discuss the specificities of the autoantibodies that are present during both APS and atherosclerosis. A critical and unresolved question is whether these APA are specific for epitopes that result from lipid oxidation. Despite the fact that APA are present in patients with systemic autoimmunity and that they may participate in the pathogenesis of APS, recent studies have paradoxically proposed a beneficial role for some APA in atherosclerosis. We review the evidence that some APA specificities may be protective against plaque formation, and we discuss the putative mechanisms by which some APA could be useful in the prevention of atherosclerosis.

- Abstract
Objective: Patients with antiphospholipid syndrome (APS) are at a high risk of developing atherosclerotic complications. Conversely, individuals with primary atherosclerosis have an increased prevalence of antiphospholipid antibodies (aPL) and antibodies to oxidized low-density lipoproteins (ox-LDL). Several studies suggest that these two antibody populations may in fact overlap, although it is unclear how aPL contribute to pathogenesis. In this study, we characterized an IgG monoclonal aPL and assessed its ability to modulate atherosclerosis in low-density lipoprotein receptor-deficient (LDLR(-/-)) mice., Methods: The cardiolipin-reactive monoclonal antibody FB1 was obtained from an (NZW x BXSB)F(1) mouse, a strain with APS features that make it prone to fatal myocardial infarctions. Using an enzyme-linked immunosorbent assay, we investigated the binding of this antibody to phospholipid and LDL antigens. We also passively administered FB1 to atherosclerosis-prone mice to determine its effect on atherogenesis., Results: In contrast to earlier studies of aPL that were specific for oxidized forms of LDL, FB1 cross-reacted with both native LDL and ox-LDL. In vivo, passive administration of FB1 significantly reduced plaque formation in atherosclerosis-prone LDLR(-/-) mice., Conclusion: These results indicate that some aPL may play a protective role in atherogenesis and suggest a novel approach to the prevention of atherosclerosis.
