To manage Earth in the Anthropocene, new tools, new institutions, and new forms of international cooperation will be required. Earth Virtualization Engines is proposed as an international federation of centers of excellence to empower all people to respond to the immense and urgent challenges posed by climate change.

The confrontation of complex Earth system model (ESM) codes with novel supercomputing architectures poses challenges to efficient modeling and job submission strategies. The modular setup of these models naturally fits a modular supercomputing architecture (MSA), which tightly integrates heterogeneous hardware resources into a larger and more flexible high-performance computing (HPC) system. While parts of the ESM codes can easily take advantage of the increased parallelism and communication capabilities of modern GPUs, others lag behind due to the long development cycles or are better suited to run on classical CPUs due to their communication and memory usage patterns. To better cope with these imbalances between the development of the model components, we performed benchmark campaigns on the Jülich Wizard for European Leadership Science (JUWELS) modular HPC system. We enabled the weather and climate model Icosahedral Nonhydrostatic (ICON) to run in a coupled atmosphere–ocean setup, where the ocean and the model I/O is running on the CPU Cluster, while the atmosphere is simulated simultaneously on the GPUs of JUWELS Booster (ICON-MSA). Both atmosphere and ocean are running globally with a resolution of 5 km. In our test case, an optimal configuration in terms of model performance (core hours per simulation day) was found for the combination of 84 GPU nodes on the JUWELS Booster module to simulate the atmosphere and 80 CPU nodes on the JUWELS Cluster module, of which 63 nodes were used for the ocean simulation and the remaining 17 nodes were reserved for I/O. With this configuration the waiting times of the coupler were minimized. Compared to a simulation performed on CPUs only, the MSA approach reduces energy consumption by 45 % with comparable runtimes. ICON-MSA is able to scale up to a significant portion of the JUWELS system, making best use of the available computing resources. A maximum throughput of 170 simulation days per day (SDPD) was achieved when running ICON on 335 JUWELS Booster nodes and 268 Cluster nodes.

State-of-the-art Earth system models typically employ grid spacings of O(100 km), which is too coarse to explicitly resolve main drivers of the flow of energy and matter across the Earth system. In this paper, we present the new ICON-Sapphire model configuration, which targets a representation of the components of the Earth system and their interactions with a grid spacing of 10 km and finer. Through the use of selected simulation examples, we demonstrate that ICON-Sapphire can (i) be run coupled globally on seasonal timescales with a grid spacing of 5 km, on monthly timescales with a grid spacing of 2.5 km, and on daily timescales with a grid spacing of 1.25 km; (ii) resolve large eddies in the atmosphere using hectometer grid spacings on limited-area domains in atmosphere-only simulations; (iii) resolve submesoscale ocean eddies by using a global uniform grid of 1.25 km or a telescoping grid with the finest grid spacing at 530 m, the latter coupled to a uniform atmosphere; and (iv) simulate biogeochemistry in an ocean-only simulation integrated for 4 years at 10 km. Comparison of basic features of the climate system to observations reveals no obvious pitfalls, even though some observed aspects remain difficult to capture. The throughput of the coupled 5 km global simulation is 126 simulated days per day employing 21 % of the latest machine of the German Climate Computing Center. Extrapolating from these results, multi-decadal global simulations including interactive carbon are now possible, and short global simulations resolving large eddies in the atmosphere and submesoscale eddies in the ocean are within reach.

From June to August 2020, an observational network of 103 meteorological ground-based stations covered the greater area (50 km × 35 km) of Hamburg (Germany) as part of the Field Experiment on Sub-mesoscale Spatio-Temporal variability at Hanseatic city of Hamburg (FESST@HH). The purpose of the experiment was to shed light on the sub-mesoscale (𝒪(100) m–𝒪(10) km) structure of convective cold pools that typically remain under-resolved in operational networks. During the experiment, 82 custom-built, low-cost APOLLO (Autonomous cold POoL LOgger) stations sampled air temperature and pressure with fast-response sensors at 1 s resolution to adequately capture the strong and rapid perturbations associated with propagating cold pool fronts. A secondary network of 21 weather stations with commercial sensors provided additional information on relative humidity, wind speed, and precipitation at 10 s resolution. The realization of the experiment during the COVID-19 pandemic was facilitated by a large number of volunteers who provided measurement sites on their premises and supported station maintenance. This article introduces the novel type of autonomously operating instruments, their measurement characteristics, and the FESST@HH data set (https://doi.org/10.25592/UHHFDM.10172; Kirsch et al., 2021b). A case study demonstrates that the network is capable of mapping the horizontal structure of the temperature signal inside a cold pool, and quantifying a cold pool's size and propagation velocity throughout its life cycle. Beyond its primary purpose, the data set offers new insights into the spatial and temporal characteristics of the nocturnal urban heat island and variations of turbulent temperature fluctuations associated with different urban and natural environments.

The science guiding the EUREC4A campaign and its measurements is presented. EUREC4A comprised roughly 5 weeks of measurements in the downstream winter trades of the North Atlantic – eastward and southeastward of Barbados. Through its ability to characterize processes operating across a wide range of scales, EUREC4A marked a turning point in our ability to observationally study factors influencing clouds in the trades, how they will respond to warming, and their link to other components of the earth system, such as upper-ocean processes or the life cycle of particulate matter. This characterization was made possible by thousands (2500) of sondes distributed to measure circulations on meso- (200 km) and larger (500 km) scales, roughly 400 h of flight time by four heavily instrumented research aircraft; four global-class research vessels; an advanced ground-based cloud observatory; scores of autonomous observing platforms operating in the upper ocean (nearly 10 000 profiles), lower atmosphere (continuous profiling), and along the air–sea interface; a network of water stable isotopologue measurements; targeted tasking of satellite remote sensing; and modeling with a new generation of weather and climate models. In addition to providing an outline of the novel measurements and their composition into a unified and coordinated campaign, the six distinct scientific facets that EUREC4A explored – from North Brazil Current rings to turbulence-induced clustering of cloud droplets and its influence on warm-rain formation – are presented along with an overview of EUREC4A's outreach activities, environmental impact, and guidelines for scientific practice. Subseasonal-to-seasonal (S2S) prediction, especially the prediction of extreme hydroclimate events such as droughts and floods, is not only scientifically challenging, but also has substantial societal impacts. Motivated by preliminary studies, the Global Energy and Water Exchanges (GEWEX)/Global Atmospheric System Study (GASS) has launched a new initiative called “Impact of Initialized Land Surface Temperature and Snowpack on Subseasonal to Seasonal Prediction” (LS4P) as the first international grass-roots effort to introduce spring land surface temperature (LST)/subsurface temperature (SUBT) anomalies over high mountain areas as a crucial factor that can lead to significant improvement in precipitation prediction through the remote effects of land–atmosphere interactions. LS4P focuses on process understanding and predictability, and hence it is different from, and complements, other international projects that focus on the operational S2S prediction. More than 40 groups worldwide have participated in this effort, including 21 Earth system models, 9 regional climate models, and 7 data groups. This paper provides an overview of the history and objectives of LS4P, provides the first-phase experimental protocol (LS4P-I) which focuses on the remote effect of the Tibetan Plateau, discusses the LST/SUBT initialization, and presents the preliminary results. Multi-model ensemble experiments and analyses of observational data have revealed that the hydroclimatic effect of the spring LST on the Tibetan Plateau is not limited to the Yangtze River basin but may have a significant large-scale impact on summer precipitation beyond East Asia and its S2S prediction. Preliminary studies and analysis have also shown that LS4P models are unable to preserve the initialized LST anomalies in producing the observed anomalies largely for two main reasons: (i) inadequacies in the land models arising from total soil depths which are too shallow and the use of simplified parameterizations, which both tend to limit the soil memory; (ii) reanalysis data, which are used for initial conditions, have large discrepancies from the observed mean state and anomalies of LST over the Tibetan Plateau. Innovative approaches have been developed to largely overcome these problems.

Clouds play a potentially important role in Arctic climate change but are poorly represented in current atmospheric models across scales. To improve the representation of Arctic clouds in models, it is necessary to compare models to observations to consequently reduce this uncertainty. This study compares aircraft observations from the Arctic CLoud Observations Using airborne measurements during polar Day (ACLOUD) campaign around Svalbard, Norway, in May–June 2017 and simulations using the ICON (ICOsahedral Non-hydrostatic) model in its numerical weather prediction (NWP) setup at 1.2 km horizontal resolution. By comparing measurements of solar and terrestrial irradiances during ACLOUD flights to the respective properties in ICON, we showed that the model systematically overestimates the transmissivity of the mostly liquid clouds during the campaign. This model bias is traced back to the way cloud condensation nuclei (CCN) get activated into cloud droplets in the two-moment bulk microphysical scheme used in this study. This process is parameterized as a function of grid-scale vertical velocity in the microphysical scheme used, but in-cloud turbulence cannot be sufficiently resolved at 1.2 km horizontal resolution in Arctic clouds. By parameterizing subgrid-scale vertical motion as a function of turbulent kinetic energy, we are able to achieve a more realistic CCN activation into cloud droplets. Additionally, we showed that by scaling the presently used CCN activation profile, the hydrometeor number concentration could be modified to be in better agreement with ACLOUD observations in our revised CCN activation parameterization. This consequently results in an improved representation of cloud optical properties in our ICON simulations.

The atmospheric energy budget is analysed in numerical simulations of tropical cloud systems to better understand the physical processes behind aerosol effects on the atmospheric energy budget. The simulations include both shallow convective clouds and deep convective tropical clouds over the Atlantic Ocean. Two different sets of simulations, at different dates (10–12 and 16–18 August 2016), are simulated with different dominant cloud modes (shallow or deep). For each case, the cloud droplet number concentration (CDNC) is varied as a proxy for changes in aerosol concentrations without considering the temporal evolution of the aerosol concentration (for example due to wet scavenging, which may be more important under deep convective conditions). It is shown that the total column atmospheric radiative cooling is substantially reduced with CDNC in the deep-cloud-dominated case (by ∼10.0 W m−2), while a much smaller reduction (∼1.6 W m−2) is shown in the shallow-cloud-dominated case. This trend is caused by an increase in the ice and water vapour content at the upper troposphere that leads to a reduced outgoing longwave radiation, an effect which is stronger under deep-cloud-dominated conditions. A decrease in sensible heat flux (driven by an increase in the near-surface air temperature) reduces the warming by ∼1.4 W m−2 in both cases. It is also shown that the cloud fraction response behaves in opposite ways to an increase in CDNC, showing an increase in the deep-cloud-dominated case and a decrease in the shallow-cloud-dominated case. This demonstrates that under different environmental conditions the response to aerosol perturbation could be different.

Cirrus clouds play an important role in determining the radiation budget of the earth, but many of their properties remain uncertain, particularly their response to aerosol variations and to warming. Part of the reason for this uncertainty is the dependence of cirrus cloud properties on the cloud formation mechanism, which itself is strongly dependent on the local meteorological conditions. In this work, a classification system (Identification and Classification of Cirrus or IC-CIR) is introduced to identify cirrus clouds by the cloud formation mechanism. Using reanalysis and satellite data, cirrus clouds are separated into four main types: orographic, frontal, convective and synoptic. Through a comparison to convection-permitting model simulations and back-trajectory-based analysis, it is shown that these observation-based regimes can provide extra information on the cloud-scale updraughts and the frequency of occurrence of liquid-origin ice, with the convective regime having higher updraughts and a greater occurrence of liquid-origin ice compared to the synoptic regimes. Despite having different cloud formation mechanisms, the radiative properties of the regimes are not distinct, indicating that retrieved cloud properties alone are insufficient to completely describe them. This classification is designed to be easily implemented in GCMs, helping improve future model–observation comparisons and leading to improved parametrisations of cirrus cloud processes.

The spatio-temporal variability of integrated water vapour (IWV) on small scales of less than 10 km and hours is assessed with data from the 2 months of the High Definition Clouds and Precipitation for advancing Climate Prediction (HD(CP)2) Observational Prototype Experiment (HOPE). The statistical intercomparison of the unique set of observations during HOPE (microwave radiometer (MWR), Global Positioning System (GPS), sun photometer, radiosondes, Raman lidar, infrared and near-infrared Moderate Resolution Imaging Spectroradiometer (MODIS) on the satellites Aqua and Terra) measuring close together reveals a good agreement in terms of random differences (standard deviation ≤1 kg m−2) and correlation coefficient (≥ 0.98). The exception is MODIS, which appears to suffer from insufficient cloud filtering. For a case study during HOPE featuring a typical boundary layer development, the IWV variability in time and space on scales of less than 10 km and less than 1 h is investigated in detail. For this purpose, the measurements are complemented by simulations with the novel ICOsahedral Nonhydrostatic modelling framework (ICON), which for this study has a horizontal resolution of 156 m. These runs show that differences in space of 3–4 km or time of 10–15 min induce IWV variabilities on the order of 0.4 kg m−2. This model finding is confirmed by observed time series from two MWRs approximately 3 km apart with a comparable temporal resolution of a few seconds. Standard deviations of IWV derived from MWR measurements reveal a high variability (> 1 kg m−2) even at very short time scales of a few minutes. These cannot be captured by the temporally lower-resolved instruments and by operational numerical weather prediction models such as COSMO-DE (an application of the Consortium for Small-scale Modelling covering Germany) of Deutscher Wetterdienst, which is included in the comparison. However, for time scales larger than 1 h, a sampling resolution of 15 min is sufficient to capture the mean standard deviation of IWV. The present study shows that instrument sampling plays a major role when climatological information, in particular the mean diurnal cycle of IWV, is determined.

A long-standing task in climate research has been to distinguish between anthropogenic climate change and natural climate variability. A prerequisite for fulfilling this task is the understanding of the relative roles of external drivers and internal variability of climate and the carbon cycle. Here, we present the first ensemble simulations over the last 1200 years with a comprehensive Earth system model including a fully interactive carbon cycle. Applying up-to-date reconstructions of external forcing including the recent low-amplitude estimates of solar variations, the ensemble simulations reproduce temperature evolutions consistent with the range of reconstructions. The 20th-century warming trend stands out against all pre-industrial trends within the ensemble. Volcanic eruptions are necessary to explain variations in pre-industrial climate such as the Little Ice Age; yet only the strongest, repeated eruptions lead to cooling trends that differ significantly from the internal variability across all ensemble members. The simulated atmospheric CO2 concentrations exhibit a stable carbon cycle over the pre-industrial era with multi-centennial variations somewhat smaller than in the observational records. Early land-cover changes have modulated atmospheric CO2 concentrations only slightly. We provide a model-based quantification of the sensitivity (termed γ) of the global carbon cycle to temperature for a variety of climate and forcing conditions. We diagnose a distinct dependence of γ on the forcing strength and time-scales involved, thus providing a possible explanation for the systematic difference in the observational estimates for different segments of the last millennium.

Abstract Background Uveal melanoma is the most common non-cutaneous melanoma and is an intraocular malignancy affecting nearly 7,000 individuals per year worldwide. Of these, approximately 50% will progress to metastatic disease for which there are currently no effective curative therapies. Despite advances in molecular profiling and metastatic stratification of uveal melanoma tumors, little is known regarding their underlying biology of metastasis. Our group has identified a disseminated neoplastic cell population characterized by co-expression of immune and melanoma proteins, circulating hybrid cells (hybrids), in patients with uveal melanoma. Compared to circulating tumor cells, which lack expression of immune proteins, hybrids are detected at an increased prevalence in peripheral blood and can be used as a non-invasive biomarker to predict metastatic progression. Methods To ascertain mechanisms underlying enhanced hybrid cell dissemination we identified hybrid cells within primary uveal melanoma tumors using single cell RNA sequencing (n = 8) and evaluated their gene expression and predicted ligand-receptor interactions in relation to other melanoma and immune cells within the primary tumor. We then verified expression of upregulated hybrid pathways within patient-matched tumor and peripheral blood hybrids (n = 4) using cyclic immunofluorescence and quantified their protein expression relative to other non-hybrid tumor and disseminated tumor cells. Results Among the top upregulated genes and pathways in hybrid cells were those involved in enhanced cell motility and cytoskeletal rearrangement, immune evasion, and altered cellular metabolism. In patient-matched tumor and peripheral blood, we verified gene expression by examining concordant protein expression for each pathway category: TMSB10 (cell motility), CD74 (immune evasion) and GPX1 (metabolism). Both TMSB10 and GPX1 were expressed on significantly higher numbers of disseminated hybrid cells compared to circulating tumor cells, and CD74 and GPX1 were expressed on more disseminated hybrids than tumor-resident hybrids. Lastly, we identified that hybrid cells express ligand-receptor signaling pathways implicated in promoting metastasis including GAS6-AXL, CXCL12-CXCR4, LGALS9-P4HB and IGF1-IGFR1. Conclusion These findings highlight the importance of TMSB10, GPX1 and CD74 for successful hybrid cell dissemination and survival in circulation. Our results contribute to the understanding of uveal melanoma tumor progression and interactions between tumor cells and immune cells in the tumor microenvironment that may promote metastasis.

Antimicrobials are widely used to cure intramammary infections (IMI) in dairy cows during the dry period (DP). Nevertheless, the IMI cure is influenced by many factors and not all quarters benefit from antimicrobial dry cow treatment (DCT). To evaluate the true effect of antibiotic DCT compared to self-cure and the role of causative pathogens on the IMI cure, a retrospective cross-sectional study was performed. The analysis included 2987 quarters infected at dry-off (DO). Information on DCT, causative pathogens, somatic cell count, milk yield, amount of lactation, Body Condition Score, and season and year of DO were combined into categorical variables. A generalized linear mixed model with a random cow, farm and year effect and the binary outcome of bacteriological cure of IMI during the DP was conducted. In the final model, a significant effect ( p < 0.05) on DP cure was seen for the DO season and the category of causative pathogens (categories being: Staphylococcus aureus , non-aureus staphylococci, streptococci, coliforms, 'other Gram-negative bacteria', 'other Gram positive bacteria', non-bacterial infections and mixed infections), while antibiotic DCT (vs. non-antibiotic DCT) only showed a significant effect in combination with the pathogen categories streptococci and 'other Gram-positive bacteria'.

To optimise udder health at the herd level, identifying incurable mastitis cases as well as providing an adequate therapy and culling strategy are necessary. Cows with clinical mastitis should be administered antibiotic medication if it is most likely to improve mammary cure. The somatic cell count (SCC) in milk of the monthly implemented Dairy Herd Improvement (DHI) test represents the most important tool to decide whether a cow has a promising mammary cure rate. Differential cell count (DCC) facilitates the specification of the immunological ability of defence, for example by characterising leukocyte subpopulations or cell viability. The aim of this study was to assess the DCC and cell viability in DHI milk samples regarding the cytological (CC) and bacteriological cure (BC) of the udder within a longitudinal study, thereby gaining a predictive evaluation of whether a clinical mastitis benefits from an antibiotic treatment or not. The cows enrolled in this study had an SCC above 200,000 cells/mL in the previous DHI test. Study 1 assessed the CC by reference to the SCC of two consecutive DHI tests and included 1010 milk samples: 28.4% of the mammary glands were classified as cytologically cured and 71.6% as uncured. The final mixed logistic regression model identified the total number of non-vital cells as a significant factor associated with CC. An increasing amount of non-vital cells was related to a lower individual ability for CC. Cows which were in the first or second lactation possessed a higher probability of CC than cows having a lactation number above two. If animals developed a clinical mastitis after flow cytometric investigation, the BC was examined in study 2 by analysing quarter foremilk samples microbiologically. Taking 48 milk samples, 81.3% of the mammary glands were classified as bacteriologically cured and 18.7% as uncured. The percentage of total non-vital cells tended to be lower for cows which were cured, but no significance could be observed. This study revealed that the investigation of the proportion of non-vital cells in DHI milk samples can enhance the prognosis of whether an antibiotic treatment of clinical mastitis might be promising or not. Prospectively, this tool may be integrated in the DHI tests to facilitate the decision between therapy or culling.

To reduce ineffective antimicrobial usage in the treatment of non-severe clinical mastitis (CM) in cows with long-lasting udder diseases, non-antibiotic therapy with a non-steroidal anti-inflammatory drug (NSAID) was conducted and evaluated in a non-blinded, positively controlled, non-inferiority trial. Therefore, three-time systemic ketoprofen treatment at intervals of 24 h was evaluated in comparison with the reference treatment of solely antibiotic therapy in a field study on nine free-stall dairy farms located in Northern Germany. Cows with previous CM cases in current lactation and/or with long-lasting high somatic cell counts in preceding dairy herd improvement test days were randomly allocated to one of the two treatment groups in cases of mild to moderate CM. Quarter foremilk samples of the affected quarters were taken for cyto-bacteriological investigation before treatment as well as ~14 and 21 d after termination of therapy. Both treatment groups were compared regarding the bacteriological cure (BC) as the primary outcome. Clinical cure (CC) and no CM relapse within 60 d after the end of treatment (no R60) were chosen as secondary outcomes. The study resulted in the following outcomes: Streptococcus uberis was most frequently identified in microbiological culture from pre-treatment samples, followed by Staphylococcus aureus and Escherichia coli and other coliforms. No significant differences between the NSAID treatment and the reference treatment were detected regarding CC and CM recurrence (no R60). Although the sole ketoprofen therapy resulted in a numerically lower likelihood of BC, there were no significant differences to the reference treatment. Considering the selection criteria in this study, the results indicate that in mild to moderate CM cases exclusive treatment with ketoprofen may constitute an alternative to antimicrobial intramammary therapy, providing an opportunity for reduction of antibiotic usage. However, non-inferiority evaluations were inconclusive. The aim of this study was to define the time-related period of intramammary infections and its relation to risk factors for intramammary infections and clinical mastitis at cow and quarter levels. In total, 269 German Holstein Frisian dairy cows on three farms in Northern and Eastern Germany were included in this study. Quarter milk samples were collected at dry-off, 3 ± 1 days after calving and 17 ± 3 days after calving, for cytomicrobiological examination. Risk factors at quarter- and cow-level associated with intramammary infections and clinical mastitis were recorded during the trial period. Data were analyzed using logistic regression procedures and odds ratios were calculated. Calving for the second time increased the odds of clinical mastitis during the first 100 days of lactation compared to cows calving for the third time or more. A high milk yield after calving was a risk factor for new infections, with environmental pathogens 17 ± 3 days postpartum. A body condition score after calving less than 3.5 was associated with a decreased risk of having an intra-mammary infection (IMI) with non- aureus staphylococci and coryneforms 3 ± 1 days postpartum and consistent body condition between dry-off and early lactation decreased the risk of intramammary infections after calving. The absence of a ring of hyperkeratosis at the teat apex shown at dry-off was associated with a lower risk of intramammary infections with environmental pathogens 17 ± 3 days postpartum. This study shows the important influence of the dry period and early lactation on intramammary infections and clinical mastitis postpartum in dairy cows. Udder quarters may have eliminated pathogens during the dry period in 43.6% of cases in this study. Additionally, new infections occurred during early lactation, so 5.1% more quarters were infected 17 ± 3 days compared to 3 ± 1 days postpartum. New infections can be traced to non- aureus staphylococci and Staphylococcus aureus from dry-off up until 3 ± 1 days postpartum, and to non- aureus staphylococci, Staphylococcus aureus and Streptococcus uberis, after calving. In total, 88.7% of the infected quarters showed new infections with another pathogen species 3 ± 1 days postpartum than at dry-off, and 89.2% of the quarters 17 ± 3 days postpartum than 3 ± 1 days postpartum. In conclusion, the early lactation has just as important an influence on intramammary infections postpartum in dairy cows as the dry period. There is the possibility that udder quarters eliminate pathogens during the early lactation, especially during the dry period. However, there is also the danger that new infections manifest, with a large proportion of new infections occurring after calving. Thus, additional control strategies are of great importance to prevent new infections occurring during early lactation as well as during the dry period to reduce negative effects on milk yield and culling hazards in dairy cows by minimizing the associated risk factors.

To reduce the negative effects of mastitis in dairy heifers in early lactation on the future milking performance, the aim of this study was to define the time-related period of intramammary infections and to relate this to risk factors at heifer and quarter level for intramammary infections and subclinical mastitis. In total, 279 German Holstein Frisian heifers in three farms in Northern and Eastern Germany were included in this study. Quarter milk samples for cytomicrobiological examination were collected 3 ± 1 days after calving and 17 ± 3 days after calving, and risk factors at heifer and quarter level associated with intramammary infections and clinical mastitis were recorded during the trial period. Data were analyzed using logistic regression procedures and odds ratios were calculated. Calving at older ages increased the odds of intramammary infections with non- aureus staphylococci (NAS) and coryneforms 17 ± 3 days after calving compared to heifers calving at a younger age. Detaching of milking cups during milking is a risk factor for new infections between day 3 ± 1 and 17 ± 3 postpartum. The milk yield after calving is associated with a decrease in intramammary infections with environmental pathogens and clinical mastitis. A high milk yield assists the development of udder edema with an increased risk of intramammary infections with NAS and coryneforms. An increased somatic cell count (SCC) after calving increased the odds of intramammary infections with contagious pathogens 17 ± 3 days postpartum. The early lactation has an important influence on udder health and intramammary infections postpartum in dairy heifers. Udder quarters eliminated pathogens during early lactation by 6.9% for cases in this study. New infections manifest themselves up until 17 ± 3 days postpartum, especially with Corynebacterium spp. and NAS. In total, 82.9% of the infected quarters showed new infections with another pathogen species 17 ± 3 days postpartum than 3 ± 1 days postpartum. In conclusion, the early lactation has an important influence on udder health and intramammary infections postpartum in heifers with the possibility that udder quarters eliminate pathogens, but also the danger that new infections manifest themselves. Thus, the prevention of new infections by minimizing the associated risk factors is of great importance.

Recurrent clinical mastitis contributes to around half of all infections having an economic impact in the dairy industry. It leads to milk yield reduction, increased risk of mortality, and culling, and may be caused by new infections or a persistent infection after previous treatment. Disease management is dependent on the infecting species, necessitating accurate identification of the pathogen in the range of persistent and reinfection cases among recurrent infections using culture and molecular biological analysis. Milk samples from diagnosed clinical mastitis cases were collected from three Northern German dairy farms between 2011 and 2015. Totally, 2043 diagnosed mastitis cases were examined at quarter level (1598 (78.2 %) first and 445 (21.8 %) recurrent mastitis cases in lactation). Among the recurrent cases, 145 (32.6 %) cases were confirmed to harbor the same pathogenic species as previous infections. RAPD PCR confirmed the same species strain in 49 (11 %) of the recurrent infections. The contribution of new infections as compared to persistent infections in cases of clinical mastitis is clear from the data. Bovine clinical mastitis quarter foremilk samples were collected from 15 German dairy farms for the isolation of Streptococcus uberis strains. Samples were also collected from the 8 spots where Streptococcus uberis was most expected in the dairy environment to investigate the transmission behavior of Streptococcus uberis within the farm. The selected environmental spots for sampling were the inner surface of the milking liner, drinking troughs (on pasture and in the barn), exit area of milking parlor, bedding material from the lying area in the barn, passageway to pasture, lying area of soil or vegetation on pasture, and the barn area in front of the milking parlor. We performed pulsed-field gel electrophoresis on 237 Streptococcus uberis isolates to identify environmental strains that matched those from mastitis milk. The same strains were detected on the passageway to the pasture, milking parlor waiting area, in one of the liners, and a drinking trough. Streptococcus uberis strains showed high variability within farms and because identical strains (in mastitis milk and environment) were found in different environmental localizations, its transmission appears to be farm specific. Thus, to establish a farm-specific mastitis control strategy, the main environmental sources of Streptococcus uberis must be analyzed for matching strains. We discuss scientific features and computational performance of kilometre-scale global weather and climate simulations, considering the Icosahedral Non-hydrostatic (ICON) model and the Integrated Forecast System (IFS). Scalability measurements and a performance modelling approach are used to derive performance estimates for these models on upcoming exascale supercomputers. This is complemented by preliminary analyses of the model data that illustrate the importance of high-resolution models to gain improvements in the accuracy of convective processes, a better understanding of physics dynamics interactions and poorly resolved or parametrized processes, such as gravity waves, convection and boundary layer. Machine milking-induced alterations of teat tissue may impair local defense mechanisms and increase the risk of new intramammary infections. The objective of the current study was to assess the influence of short-term and long-term alterations of teat tissue and infectious status of the udder quarter on the risk of naturally occurring new intramammary infections, inflammatory responses, and mastitis. Short-term and long-term changes in teat condition of right udder quarters of 135 cows of a commercial dairy farm in Saxony-Anhalt, Germany, were recorded monthly for 10 mo using simple classification schemes. Quarter milk samples were collected from all examined quarters at each farm visit. Bacteriological culture results and somatic cell counts of quarter milk samples were used to determine new inflammatory responses (increase from ≤100,000 cells/mL to >100,000 cells/mL between 2 samples), new infections (detection of a pathogen from a quarter that was free of the same pathogen at the preceding sampling), and new mastitis (combination of new inflammatory response and new infection). Separate Poisson mixed models for new inflammatory responses, new infections, and new mastitis caused by specific pathogens or groups of pathogens (contagious, environmental, major, minor, or any) were used to estimate risk ratios and 95% confidence intervals. Data preparation and parameter estimation were performed using the open source statistical analysis software R. We observed no effect of any variable describing teat condition on the risk of new intramammary infections, inflammatory responses, or mastitis. Historically, medicine and nursing has had a hierarchical and patriarchal relationship, with physicians holding monopoly over knowledge-based practice of medical care, thus impeding interprofessional collaboration. Power gradient prevents nurses from demanding cooperative patient rounding. We surveyed attitudes of nursing staff at our tertiary care community hospital, before and after implementation of a patient-centered interprofessional (hospitalist-nurse) rounding process for patients. There was a substantial improvement in nursing staff satisfaction related to the improved communication (7%-54%, p < 0.001) and rounding (3%-49%, p < 0.001) by hospitalist providers. Patient-centered rounding also positively impacted nursing workflow (5%-56%, p < 0.001), nurses' perceptions of value as a team member (26%-56%, p = 0.018) and their job satisfaction (43%-59%, p = 0.010). Patient-centered rounding positively contributed to transforming the hospitalist-nurse hierarchical model to a team-based collaborative model, thus enhancing interprofessional relationships.

Jewel beetles of the genus Melanophila and some pyrophilous species of the flat bugs genus Aradus show a pyrophilous behaviour and have developed so-called photomechanical infrared (IR) receptors. In a spherical photomechanical IR sensillum incoming IR radiation is converted into micromechanical action, finally stimulating the dendritic tip of a mechanosensitive sensory cell. The tip is located inside a tiny cuticular sphere with a diameter of about 12 μm. The material properties of the different cuticular components of this sphere are of great importance for stimulus generation. We measured the modulus and hardness of the outer exocuticular shell of the sphere and the mesocuticle inside the core. Measurements were made by nanoindentation at sensilla which were partly cut open under dry as well as under rewetted (i.e. quasi-natural) conditions. We found that in the rewetted sensilla the outer exocuticular shell of the sphere in the Melanophila sensillum is about 50% harder and 20% stiffer than reference exocuticle, and that in both species especially the rewetted mesocuticle of the inner core of the IR sensilla is significantly softer (about 80% in Melanophila) and more compliant (about 90% also in Melanophila) than the reference mesocuticle. The findings can be interpreted as special adaptations of the cuticular microdomains of photomechanical infrared sensilla to enhance thermomechanical performance and, thereby, sensitivity., (Copyright © 2012 Acta Materialia Inc. The mechanical properties of the sternal cuticle of the locust were investigated by nanoindentation. Modulus and hardness of the exo-, meso-, and endocuticular layers were locally measured under dry and fully wetted conditions in the normal (i.e. perpendicular to the outer surface) as well as in the transverse direction (i.e. parallel to the alignment of the respective layers). The results show that water has a major impact on the mechanical properties of all layers. After drying the endocuticle, in particular, became harder by a factor of up to 9 and stiffer by a factor of up to 7.4. Additionally the gradual decrease in hardness and Young's modulus from the outer exo- to the inner endocuticle, characteristic of native cuticle, was eliminated or even reversed in dried cuticle. A pronounced anisotropy was revealed in all layers when comparing data obtained by probing in the normal (lower values) vs. probing in the transverse direction (higher values). Cyclic drying and rewetting of the endocuticle showed that the mechanical properties can be reproducibly changed by altering the water content. Based on our results we propose a new role of the epicuticle: fine-tuning of the mechanical properties of the different cuticular layers can be accomplished by setting the local cuticular transpiration., (Copyright © 2011 Acta Materialia Inc. Beetles of the genus Melanophila and certain flat bugs of the genus Aradus actually approach forest fires. For the detection of fires and of hot surfaces the pyrophilous species of both genera have developed infrared (IR) receptors, which have developed from common hair mechanoreceptors. Thus, this type of insect IR receptor has been termed photomechanic and shows the following two special features: (i) The formation of a complex cuticular sphere consisting of an outer exocuticular shell as well as of a cavernous microfluidic core and (ii) the enclosure of the dendritic tip of the mechanosensitive neuron inside the core in a liquid-filled chamber. Most probably a photomechanic IR sensillum acts as a microfluidic converter of infrared radiation which leads to an increase in internal pressure inside the sphere, which is measured by a mechanosensitive neuron.A simple model for this biological IR sensor is a modified Golay sensor in which the gas has been replaced by a liquid. Here, the absorbed IR radiation results in a pressure increase of the liquid and the deflection of a thin membrane. For the evaluation of this model analytical formulas are presented, which permits the calculation of the pressure increase in the cavity, the deformation of the membrane and the time constant of an artificial leak to compensate ambient temperature changes. Some organic liquids with high thermal expansion coefficients may improve the deflection of the membrane compared to water.

Streptococcus uberis and Staphylococcus aureus are important mastitis pathogens in dairy cows in the entire world. Recent publications showed that an extended therapy can be more efficient in combating such intramammary infections. The objective of this study was to evaluate the effect of an extended intramammary therapy to treat moderate and severe mastitis cases in lactating dairy cows under field conditions in northern Germany. From December 2005 to August 2007, a total of 157 clinical mastitis cases on 10 farms in northern Germany were enrolled in the study and randomly assigned (blocked by parity and body temperature) to one of two treatment groups (intrammammary lincomycin/neomycin 1.5-d (ALK) or 5-d (ALL)). Clinical, microbiological and cytomicrobiological cure rates were evaluated. Treatments were initiated before culture results. Cows were observed and evaluated on d 1 to 6, 19 and 26. Six cases in 157 (3.8%) resulted in a therapy change in between 48 h after mastitis detection. Overall, treatments were not significantly different to controls regarding clinical cure rate. However, when the microbiological cure rate was evaluated, differences were observed. ALL appeared form infections. We conclude that in farms with Streptococcus uberis mastitis, the 5-d extended lincomycin/neomycin treatment regimen was significantly more efficient in microbiological cure than the standard 1.5-d treatment.

This field study focused on the possible effects of increased milking frequency (milking four times a day in comparison with milking twice a day) on clinical and bacteriological cure rates of clinical, antibiotically treated mastitis cases. Parameters tested were clinical, microbiological and full (cytomicrobiological) cure as well as the development of milk yield after the clinical mastitis episode. Cows from a large dairy herd meeting the study criteria (n=93) were assigned to two treatment groups by a systematic randomization scheme (blocked by body temperature <=or >39.5 degrees C). Both groups were randomly divided by experimental treatments: a) antibiotic intramammary treatment and milking 2-times a day; b) antibiotic intramammary treatment and milking 4-times a day. Treatments were initiated before the culture results were known. Cows were surveyed and evaluated on days 1-6, 24 and 31. No significant differences between treatment and control groups regarding clinical cure, microbiological cure, full cure and milk production could be established. Applying a 4-times a day milking regime did not lead to any significant effect, either positive or negative. Therefore, the results suggest that milking 4-times a day as a supporting therapy for mild, moderate and severe antimicrobially treated mastitis cases cannot be recommended.

The objective of this study was to evaluate the piglet as a suitable animal model for human diseases of high altitude. We studied 12 piglets, 4-10 weeks old, in a hypobaric chamber under conditions of high altitude at a pressure of 1/2 atmosphere (to approximately 320 Torr) for various periods of time (12, 24, 36, 48, and 72 hours) with continuous monitoring. The animals were decompressed every 24 hours for grooming and feeding. Two animals were studied as nonexposed controls, and one was studied as a control in the chamber without decompression. The animals were euthanized after the exposure, and a complete autopsy was performed. The tissues were then analyzed with light and electron microscopy. The animals all exhibited clinical features of ataxia, tachypnea with Cheyne-Stokes respiration, and lethargy. One animal vomited. The histologic and ultrastructural analysis showed normal organs, particularly lung and brain. The piglet may be a suitable animal model for the study of high altitude-related diseases in humans, but prolonged uninterrupted exposure and a delay in euthanasia after exposure to high altitude may be necessary for the development of reactive pathologic changes.
