The average bone-to-implant contact of the Mg-xGd implants was comparable to that of Ti and higher than for PEEK. The high-resolution crystal structure of the trimeric major light-harvesting complex of photosystem II (LHCII) is often perceived as the basis for understanding its light-harvesting and photoprotective functions. However, the LHCII solution structure and its oligomerization or aggregation state may generally differ from the crystal structure and, moreover, also depend on its functional state. In this regard, small-angle scattering experiments provide the missing link by offering structural information in aqueous solution at physiological temperatures. Herein, we use small-angle scattering to investigate the solution structures of two different preparations of solubilized LHCII employing the nonionic detergents n-octyl-β-d-glucoside (OG) and n-dodecyl-β-D-maltoside (β-DM). The data reveal that the LHCII-OG complex is equivalent to the trimeric crystal structure. Remarkably, however, we observe─for the first time─a stable oligomer composed of three LHCII trimers in the case of the LHCII-β-DM preparation, implying additional pigment-pigment interactions. The latter complex is assumed to mimic trimer-trimer interactions which play an important role in the context of photoprotective nonphotochemical quenching.

The development of PEO surface treatments for Nb (niobium) provides a possibility to produce functionalized coatings with a combination of new compositions and phases. In the frame of this work, PEO coatings on pure Nb are produced in three electrolytes containing aluminate, phosphate and silicate. Furthermore, the influence of the electrolyte composition on the PEO process and the microstructure, composition and properties of the coatings were studied. It was observed that most of the coating forming species did not participate in the plasma reactions directly, while the chemical-, electrochemical- and thermal stimulated reactions are dominating the coating formation. The results show that all the coatings are mainly composed of the Nb$_2$O$_5$ phase. An additional mixture of oxides (Al$_2$O$_3$ and AlNbO$_4$) is found in the coating formed in aluminate-containing electrolyte, and the coating formed in phosphate-containing electrolyte contains a small amount of Nb$_2$(PO$_4$)$_3$ and amorphous phosphate. No additional crystalline phase other than the amorphous silica phase is detected in the coating formed in silicate-containing electrolyte. Additionally, different morphology of the coatings is revealed and the most uniform coating is produced in silicate-containing electrolyte, which further improves its corrosion resistance. This coating also shows the highest photocatalytic activity due to the combination of Nb2O5 and silica. Since all PEO coatings on pure Nb show superior corrosion resistance and photocatalytic activity, they might be interesting for a wide range of applications ranging from transportation, biomedical implants to environmental protection.

The exceptional properties of metal organic frameworks (MOFs) put them also in focus as perspective materials for corrosion protection. In this work, the formation of a novel ZIF-8@PEO coating is reported for the first time. It was synthesized by controllable recrystallization of a PEO layer formed on zinc alloy Z1 into ZIF-8 in the presence of 2-methylimidazole organic linkers. The multi-stage mechanism of PEO to ZIF-8 rearrangement is proposed. Cross section, glow discharge optical emission spectroscopy and nano-focused synchrotron X-ray diffraction demonstrated that varying of synthesis parameters, the ZIF-8@PEO coating with different distribution of ZIF-8 through PEO layer can be prepared. Based on the results of laser scanning microscopy, the surface smoothing was observed with increasing the degree of the PEO-to-ZIF-8 rearrangement. Containing two components, the novel ZIF-8@PEO coating is expected to combine admirable physical-chemical properties of both PEO and ZIF-8. The molecular mechanisms responsible for outstanding lubrication of natural systems, like articular joints, have been the focus of scientific research for several decades. One essential aspect is the lubrication under pressure, where it is important to understand how the lubricating entities adapt under dynamic working conditions in order to fulfill their function. We made a structural investigation of a model system consisting of two of the molecules present at the cartilage interface, 1,2-dipalmitoyl-sn-glycero-3-phosphocholine and hyaluronan, at high hydrostatic pressure. Phospholipid layers are found at the cartilage surfaces and are able to considerably reduce friction. Their behavior under load and varied solution conditions is important as pressures of 180 bar are encountered during daily life activities. We focus on how divalent ions, like Ca2+, affect the interaction between DPPC and hyaluronan, as other investigations have indicated that calcium ions influence their interaction. It could be shown that already low amounts of Ca2+ strongly influence the interaction of hyaluronan with DPPC. Our results suggest that the calcium ions increase the amount of adsorbed hyaluronan indicating an increased electrostatic interaction. Most importantly, we observe a modification of the DPPC phase diagram as hyaluronan absorbs to the bilayer which results in an Lα-like structure at low temperatures and a decoupling of the leaflets forming an asymmetric bilayer structure.

During the implant preparation process, the material's surface and microstructure are being changed by stresses induced by machining. Hence degradable metal implants need to be fully characterized in terms of the influence of machining on the resulting microstructure and corrosion performance.In this study, micro-computed tomography (��CT) is used for the quantification of the degradation rate of biodegradable implants. To our best knowledge, for the first time quantitative measures are introduced to describe the degradation homogeneity in 3D. This information enables a prediction in terms of implant stability during the degradation in the body.Two magnesium gadolinium alloys, Mg-5Gd and Mg-10Gd (all alloy compositions are given in weight% unless otherwise stated), in the shape of M2 headless screws have been investigated for their microstructure and their degradation performance up to 56 days. During the microstructure investigations particular attention was paid to the localized deformation of the alloys, due to the machining process. In vitro immersion testing was performed to assess the degradation performance quantified by subsequent weight loss and volume loss (using ��CT) measurements.Although differences were observed in the degree of screw's near surface microstructure being influenced from machining, the degradation rates of both materials appeared to be suitable for application in orthopedic implants. From the degradation homogeneity point of view no obvious contrast was detected between both alloys. However, the higher degradation depth ratios between the crests and roots of Mg-5Gd ratios may indicated a less homogeneous degradation of the screws of these alloys on contract to the ones made of Mg-10Gd alloys. Due to its lower degradation rates, its more homogeneous microstructure, its weaker texture and better degradation performance extruded Mg-10Gd emerged more suitable as implant material than Mg-5Gd., Published by Elsevier, Amsterdam [u.a.]

Such manipulations can induce artifacts or result in a tremendous workload by, e.g., the preparation of multiple 2D images to retrieve the volume information. A non-invasive technique allowing to image the soft tissue in a 3D fashion is propagation-based phase contrast computed tomography. We explore the methods' unique properties to assess the 3D distribution and size of human chondrocytes within collagen scaffolds in a liquid environment without embedding. To seek if the identification of differences in cell distribution is possible, we have seeded cartilage cells on collagen scaffolds that were unstimulated or stimulated by alternating electric fields for 7 days. Analysis of the 3D cell distributions reveals that the migration depth of the chondrocytes into the scaffold is nearly doubled along with the total number of cells due to the applied electric field. Further analysis shows no specific size distribution of the chondrogenic cells. Today, substantial attention is given to biomaterial strategies for bone regeneration, and among them, there is a growing interest in using immunomodulatory biomaterials. The ability of a biomaterial to induce neo vascularization and macrophage polarization is a major factor in defining its success. Magnesium (Mg)-based degradable alloys have attracted significant attention for bone regeneration owing to their biodegradability and potential for avoiding secondary removal surgeries. However, there is insufficient evidence in the literature regarding the early inflammatory responses to these alloys in vivo. In this study, we investigated the early body responses to Mg-0.45wt%Zn-0.45wt%Ca pin-shaped alloy (known as ZX00 alloy) in rat femora 2, 5, and 10 days after implantation. We used 3D micro computed tomography (µCT), histological, immunohistochemical, histomorphometrical, and small angle X-ray scattering (SAXS) analyses to study new bone formation, early macrophage polarization, neo vascularization, and bone quality at the implant bone interface. The expression of macrophage type 2 biological markers increased significantly after 10 days of Mg alloy implantation, indicating its potential in stimulating macrophage polarization. Our biomineralization results using µCT as well as histological stained sections did not indicate any statistically significant differences between different time points for both groups. The activity of alkaline phosphatase (ALP) and Runt-related transcription factor 2 (Runx 2) biological markers decreased significantly for Mg group, indicating less osteoblast activity. Generally, our results supported the potential of ZX00 alloy to enhance the expression of macrophage polarization in vivo; however, we could not observe any statistically significant changes regarding biomineralization.

Plasma electrolytic oxidation (PEO) with in‐situ anatase particle addition was applied to functionalize the surface of AA2024 alloy. A base potassium titanium‐oxide oxalate dihydrate aqueous electrolyte was used with up to 30 g/L$^{-1}$ anatase particle addition. The coatings’ morphology and phase composition as a function of the anatase concentration in the electrolyte were characterized by scanning electron microscopy (SEM), X‐ray diffraction (XRD) and glow discharge optical emission spectroscopy (GDOES). Photocatalytic activity, stability in chloride solution and tribological properties were also determined. The main coating forming phases are anatase and rutile on top of a mixed interface region consisting of TiAl$_2$O$_5$ as reaction product between the TiO$_2$ and an Al$_2$O$_3$ barrier layer on top of the Al substrate. The mixed layer is extending with increasing amount of particles added, due to intensified discharges. In addition, anatase to rutile phase ratio increases due to the additional anatase particles in the electrolyte. Thus, the photocatalytic activity is improving with the particle addition. The coatings’ mechanical resistance is dropping first before increasing again with more particles added. Chemical and restored mechanical stability seems to be related to the extended mixed interface formation, which strengthen the bond to the substrate when more particles are added.

However, the degradation processes in vivo and in particular the interaction of the bone with the degrading material need to be further investigated. In this study we are presenting the first quantitative comparison of the bone ultrastructure formed at the interface of biodegradable Mg–5Gd and Mg–10Gd implants and titanium and PEEK implants after 4, 8 and 12 weeks healing time using two-dimensional small angle X-ray scattering and X-ray diffraction. Differences in mineralization, orientation and thickness of the hydroxyapatite are assessed. We find statistically significant (p < 0.05) differences for the lattice spacing of the (310)-reflex of hydroxyapatite between titanium and Mg–xGd materials, as well as for the (310) crystal size between titanium and Mg–5Gd, indicating a possible deposition of Mg within the bone matrix. The (310) lattice spacing and crystallite size further differ significantly between implant degradation layer and surrounding bone (p < 0.001 for Mg–10Gd), suggesting apatite formation with significant amounts of Gd and Mg within the degradation layer., Published by Elsevier, [Amsterdam]

One prominent area is the synovial fluid which exhibits remarkable lubrication properties. However, the synovial fluid is a multi-component system where different macromolecules interact in a synergetic fashion. Within this study we focus on the interaction of hyaluronan and phospholipids, which are thought to play a key role for lubrication. We investigate how the interactions and the association structures formed by hyaluronan (HA) and 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC) are influenced by the molecular weight of the bio polymer and the ionic composition of the solution. We combine techniques allowing us to investigate the phase behavior of lipids (differential scanning calorimetry, zeta potential and electrophoretic mobility) with structural investigation (dynamic light scattering, small angle scattering) and theoretical simulations (molecular dynamics). The interaction of hyaluronan and phospholipids depends on the molecular weight, where hyaluronan with lower molecular weight has the strongest interaction. Furthermore, the interaction is increased by the presence of calcium ions. Our simulations show that calcium ions are located close to the carboxylate groups of HA and, by this, reduce the number of formed hydrogen bonds between HA and DPPC. The observed change in the DPPC phase behavior can be attributed to a local charge inversion by calcium ions binding to the carboxylate groups as the binding distribution of hyaluronan and 1,2-dipalmitoyl-sn-glycero-3-phosphocholine is not changed., Published by MDPI7969, Basel

The manner in which nature has solved lubrication issues has fascinated scientists for centuries, in particular when considering that lubrication is achieved in aqueous media. The most outstanding system in this respect is likely the synovial joint, where close to frictionless motion is realized under different loads and shear rates. This review article focuses on two components present in the synovial area, hyaluronan and phospholipids. We recapitulate what has been learned about their interactions at interfaces from recent experiments, with focus on results obtained using reflectivity techniques at large scale facilities. In parallel, modelling experiments have been carried out and from these efforts new detailed knowledge about how hyaluronan and phospholipids interact has been gained. In this review we combine findings from modelling and experiments to gain deeper insight. Finally, we summarize what has been learned of the lubrication performance of mixtures of phospholipids and hyaluronan.

The adsorption of hydrophilic or amphiphilic multiblock copolymers provides a powerful means to produce well-defined "smart" surfaces, especially if one or several blocks are sensitive to external stimuli. The load frame is fully integrated into the beamline control system and can be controlled via a feedback loop. All relevant parameters (load, displacement, temperature, etc.) are continuously logged. It can be operated in compression or tensile mode applying forces of up to 1 kN and is compatible with all contrast modalities available at IBL and HEMS i.e. conventional attenuation contrast, propagation based phase contrast and differential phase contrast using a grating interferometer. The modularity and flexibility of the load frame allows conducting a wide range of experiments. E.g. compression tests to understand the failure mechanisms in biodegradable implants in rat bone or to investigate the mechanics and kinematics of the tessellated cartilage skeleton of sharks and rays, or tensile tests to illuminate the structure-property relationship in poplar tension wood or to visualize the 3D deformation of the tendonbone insertion. Nanoparticles (NPs) that form by self-assembly of amphiphilic poly(N-(2-hydroxypropyl)-methacrylamide) (pHPMA) copolymers bearing cholesterol side groups are potential drug carriers for solid tumor treatment. Here, we investigate their behavior in solutions of human serum albumin (HSA) in phosphate buffered saline. Mixed solutions of NPs, from polymer conjugates with or without the anticancer drug doxorubicin (Dox) bound to them, and HSA at concentrations up to the physiological value are characterized by synchrotron small-angle X-ray scattering and isothermal titration calorimetry. When Dox is absent, a small amount of HSA molecules bind to the cholesterol groups that form the core of the NPs by diffusing through the loose pHPMA shell or get caught in meshes formed by the pHPMA chains. These interactions are strongly hindered by the presence of Dox, which is distributed in the pHPMA shell, meaning that the delivery of Dox by the NPs in the human body is not affected by the presence of HSA.

Magnesium (Mg) and its alloys degrade under physiological conditions. But how strong is the connection between the implant, the corrosion layer and the surrounding tissue, namely bone? Biomechanical tests like push-out tests have shown that a degraded Mg-pin is surprisingly well integrated with the bone “as reported by Castellani et al. (Acta Biomater 7(1):432–440, 2011) [1]”. High-resolution synchrotron tomography offers a deep look into the microstructure of the material as well as of the bone during deformation until fracture happens. Here we present first data from an in situ tomography experiment of a biodegradable Mg-based implant under compressive load showing how Mg implants are incorporated into bone.

The structure of monomeric and trimeric photosystem I (PS I) of Thermosynechococcus elongatus BP1 (T. elongatus) was investigated by small-angle X-ray scattering (SAXS). The scattering data reveal that the protein-detergent complexes possess radii of gyration of 58 and 78 Å in the cases of monomeric and trimeric PS I, respectively. The results also show that the samples are monodisperse, virtually free of aggregation, and contain empty detergent micelles. The shape of the protein-detergent complexes can be well approximated by elliptical cylinders with a height of 78 Å. Monomeric PS I in buffer solution exhibits minor and major radii of the elliptical cylinder of about 50 and 85 Å, respectively. In the case of trimeric PS I, both radii are equal to about 110 Å. The latter model can be shown to accommodate three elliptical cylinders equal to those describing monomeric PS I. A structure reconstitution also reveals that the protein-detergent complexes are larger than their respective crystal structures. The reconstituted structures are larger by about 20 Å mainly in the region of the hydrophobic surfaces of the monomeric and trimeric PS I complexes. This seeming contradiction can be resolved by the addition of a detergent belt constituted by a monolayer of dodecyl-β-D-maltoside molecules. Assuming a closest possible packing, a number of roughly 1024 and 1472 detergent molecules can be determined for monomeric and trimeric PS I, respectively. Taking the monolayer of detergent molecules into account, the solution structure can be almost perfectly modeled by the crystal structures of monomeric and trimeric PS I.

Single-phase strontium ferromolybdate (Sr2FeMoO6–δ) samples with different degrees of the superstructural ordering of the Fe/Mo cations were obtained from partially reduced SrFeO3–х, SrMoO4 precursors by the solid-state technology. The study of the temperature dependences of the magnetization measured in the field-cooling and zero-field-cooling regimes indicated an inhomogeneous magnetic state of the samples. The presence of magnetic regions of different nature has also been revealed by the small-angle neutron scattering. For the Sr2FeMoO6–δ samples with different superstructural ordering of the Fe/Mo cations and for all values of the magnetic field induction in the range up to 1.5 T and of the scattering vector in the interval 0.1 >q >0.005 Å–1, the analytical dependence I ~ q–α obeys the Porod law (α ≈ 4), which corresponds to an object with a smooth and well-marked surface and polydisperse grain size. Deviations from the Porod law in the q > 0.1 Å–1 region and a weakening of the neutron scattering in applied magnetic fields may be ascribed to magnetic inhomogeneities with diameters D

We present the first measurements of the iron L2/3-edge of the compounds FeO, Fe2O3, and Fe3O4 at ambient pressure and of FeCO3 at high pressures of 2.4 and 40 GPa using a diamond anvil cell by X-ray Raman scattering spectroscopy, a bulk sensitive probe of soft X-ray absorption edges making use of hard X-rays. We show that the spectral shape of the Fe L2/3-edge can be analyzed quantitatively to reveal the oxidation state of iron in matter. Consequently, in situ X-ray Raman scattering spectroscopy at the iron L-edge at high pressure and temperature opens exciting perspectives to characterize the local coordination, oxidation, and spin state of iron at high pressure and temperature, conditions that are of relevance for e.g. geological sciences or chemical processing.

This study focuses on the preparation and characterization of magnetic switchable thin iron oxide–polymer films. In a series of experiments, the formation and growth of iron oxide under ultrathin polysiloxane layers was controlled by changing the concentration of iron ions in the aqueous subphase or by varying the residence time of ammonia in the gas phase above the liquid sample. The growth of the combined film structures is studied in situ by interfacial rheology, optical microscopy, and x-ray scattering experiments and ex situ by scanning electron microscopy. Different stages of iron oxide aggregation, from a very thin layer of amorphous iron oxide with thickness of a few nanometers up to micrometer thick coatings of crystalline maghemite (γ-Fe2O3) were investigated. The specific interactions between the inorganic iron oxide and the polymer membranes cause the creation of new composite materials which are sensitive to magnetic forces.

Nonionic dendritic amphiphiles that self-assemble into defined supramolecular aggregates are useful for the efficient solubilization of active agents, for example, in drug delivery. We investigated a new class of dendritic amphiphiles based on a hydrophilic polyol dendron head connected to a two-chain hydrophobic block. In analogy to phospholipids, these molecules form well-organized layers in bulk (vesicles) or at the water surface (Langmuir monolayer). The actual study focuses on the phase behavior and microscopic structure of mixed Langmuir layers of theses dendritic amphiphiles with the well-known phospholipid DPPC. The combination of surface pressure area isotherms with X-ray grazing incident diffraction and Brewster angle microscopy gives us information on the phase behavior of the mixed monolayers and the orientation of the amphiphiles inside the condensed domains with molecular resolution. We could prove that the dendritic generation and, by this, the headgroup size of the amphiphilic molecules have a significant influence on their interaction with DPPC at the air-water interface. Thus, our findings are important for the understanding of mixed lipid membranes in general as well as for the preparation of artificial membranes and vesicles with adjustable properties, e.g., their drug delivery potential.

Phospholipids fulfill an important role in joint lubrication. They, together with hyaluronan and glycoproteins, are the biolubricants that sustain low friction between cartilage surfaces bathed in synovial fluid. In this work we have investigated how the friction force and load bearing capacity of 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC) bilayers on silica surfaces are affected by temperature, covering the temperature range 25-52°C. Friction forces have been determined utilizing the AFM colloidal probe technique, which showed that DPPC bilayers are able to provide low friction forces over the whole temperature interval. However, the load bearing capacity is improved at higher temperatures. We interpret this finding as being a consequence of lower rigidity and higher self-healing capacity of the DPPC bilayer in the liquid disordered state compared to the gel state. The corresponding structure of solid supported DPPC bilayers at the silica-liquid interface has been followed using X-ray reflectivity measurements, which suggests that the DPPC bilayer is in the gel phase at 25°C and 39°C and in the liquid disordered state at 55°C. Well-defined bilayer structures were observed for both phases. The deposited DPPC bilayers were also imaged using AFM PeakForce Tapping mode, and these measurements indicated a less homogeneous layer at temperatures below 37°C.

Proteins are only marginally stable and are hence very sensitive to environmental conditions, such as high and low temperatures or high hydrostatic pressures. In nature, living organisms are able to compensate for extreme environmental conditions and hence rescue proteins from denaturation by using osmolytes. Organic osmolytes are accumulated under anhydrobiotic, thermal, and pressure stresses. Among those osmolytes are amino acids, polyols and sugars (e.g., glycerol and trehalose), methylamines such as trimethylamine-Noxide (TMAO), and urea. TMAO has been found to enhance protein folding and ligand binding most efficiently. On the other hand, urea, a highly concentrated waste product in mammalian kidneys, is a perturbant. It is also a major organic osmolyte in marine elasmobranch fishes. Interestingly, TMAO has been found to counteract perturbations imposed by urea and hydrostatic pressure in deep-sea animals, most effectively at a 2:1 urea:TMAO ratio. In the deep sea, hydrostatic pressures up to the 1 kbar (100 MPa) range prevail, and living organisms have to cope with such extreme environmental conditions. High hydrostatic pressure generally destabilizes the protein structure, inhibits polymerization of proteins and ligand binding. Interestingly, TMAO has been shown to largely offset these pressure effects. In fact, it was found that the amount of TMAO in the cells of a series of marine organisms increases linearly with the depth of the ocean. For that reason, TMAO is thought to serve as pressure counteractant. The term “piezolyte” has been coined for such kind of cosolute. About the underlying mechanism of stabilization by TMAO at ambient pressure conditions several experimental and theoretical (molecular dynamics simulations) articles have been published in recent years. TMAO is largely excluded from the protein surface and enhances the water structure causing greater organization through more and stronger hydrogen bonding among water molecules. However, the mechanism of this “chemical chaperon” at high hydrostatic pressure (HHP) conditions is still unclear. To yield a deeper understanding of this phenomenon, we determined the intermolecular interaction of dense protein solutions in the absence and presence of cosolvent mixtures of TMAO and urea also under HHP conditions. Small-angle Xray scattering (SAXS) experiments on dense lysozyme solutions have been carried out in the pressure range from 1 bar up to 4 kbar. The SAXS technique accurately monitors structural alterations of the protein solution and yields quantitative information on the state-dependent protein– protein interaction potential. As lysozyme is a highly stable protein, pressure-induced effects will only be attributed to changes in the protein–protein interaction of the native protein and how this is influenced by osmolytes. No pressureinduced unfolding of the protein occurs in the pressure range covered. Complementary thermodynamic data, that is, the temperature of unfolding and the volume change upon unfolding of the protein, were obtained by differential scanning (DSC) and pressure perturbation calorimetry (PPC), respectively. To verify that the protein is folded at all solution conditions studied, SAXS measurements on diluted lysozyme solutions (cP= 10 mgmL ) were carried out in the whole pressure range covered. For diluted protein solutions, the scattering intensity I(q) is proportional to the form factor P(q) (q= (4p/l)sin(V/2) is the wave vector transfer, l the wavelength of the X-rays, and V the scattering angle), which depends on the structure and size of the protein. For dilute lysozyme solutions, the radius of gyration of the particle, Rg, could be determined. We found a constant Rg value of (15.1 0.4) up to 4 kbar, indicating the absence of unfolding even at the highest pressure applied. In the case of concentrated protein solutions, the interaction between the particles gives rise to an additional scattering contribution. This SAXS signal can be described as the product of the form factor and an effective structure factor, which is related to the intermolecular structure factor S(q). To relate the structure factor to the protein–protein interaction potential, statistical mechanical model approaches have to be employed. Here, the mean-spherical approximation (MSA) in combination with the DLVO (Derjaguin– Landau–Verwey–Overbeek) potential V(r) has been used. The pair potential V(r) is given as the sum of a hard sphere potential VHS(r), a repulsive screened Coulomb-like potential VSC(r) and an attractive Yukawian potential VY(r), which is frequently used to describe protein–protein interactions (for details, see the Supporting Information). The influence of pressure on the structure and protein-protein interaction potential of dense protein solutions was studied and analyzed using small-angle x-ray scattering in combination with a liquid state theoretical approach. The structural as well as the interaction parameters of dense lysozyme solutions are affected by pressure in a nonlinear way. The structural properties of water lead to a modification of the protein-protein interactions below 4 kbar, which might have significant consequences for the stability of proteins in extreme natural environments.

A structural interpretation of the thermodynamic stability of proteins requires an understanding of the structural properties of the unfolded state. High-pressure small-angle x-ray scattering was used to measure the effects of temperature, pressure, denaturants, and stabilizing osmolytes on the radii of gyration of folded and unfolded state ensembles of staphylococcal nuclease. A set of variants with the internal Val-66 replaced with Ala, Tyr, or Arg was used to examine how changes in the volume and polarity of an internal microcavity affect the dimensions of the native state and the pressure sensitivity of the ensemble. The unfolded state ensembles achieved for these proteins with high pressure were more compact than those achieved at high temperature, and were all very sensitive to the presence of urea and glycerol. Substitutions at the hydrophobic core detectably altered the conformation of the protein, even in the folded state. The introduction of a charged residue, such as Arg, inside the hydrophobic interior of a protein could dramatically alter the structural properties, even those of the unfolded state. The data suggest that a charge at an internal position can interfere with the formation of transient hydrophobic clusters in the unfolded state, and ensure that the pressure-unfolded form of a protein occupies the maximum volume possible. Only at high temperatures does the radius of gyration of the unfolded state ensemble approach the value for a statistical random coil.

The structural change of Langmuir layers composed of alkyltrichlorosilanes under the influence of ammonia (NH3) was investigated. X-ray reflectivity and grazing incidence diffraction measurements along with surface pressure and surface potential measurements were performed in order to characterize the network structure. The data show an increase of the scattered intensity after addition of ammonia while the domain and unit cell size of the film did not change. These results show a higher surface coverage, which is not caused by a simple compression of the lipid tails. The effect can be attributed to a closing of voids in the polymer film caused by temporary breaking and annealing of the chemical bonds in the network by an increase of pH.

The stability of fluid interfaces is important in many technical fields, e.g. suspensions, emulsions and foams. In this publication we investigated the influence of maghemite nanoparticles (γ-Fe2O3) on the surface stability of different surfactant films (SDS, CTAB, Brij 35). We investigated the interactions between nanoparticles and surfactant films by means of surface dilatation and surface shear rheological experiments. For further characterizations we used X-ray reflectivity (XRR) measurements, dynamic light scattering (DLS) and zeta (ζ)-potential measurements. For CTAB and more obvious for SDS it was found that at low to moderate surfactant concentrations, the viscoelasticity of the interface was increased drastically in the presence of the iron oxide nanoparticles. For films of Brij 35, however, the nanoparticles did not have any influence on the surface rheology.

The flexible alkyl ether functionalized metal–organic framework [Zn2(BME-bdc)2(dabco)]n (BME-bdc = 2,5-bis(2-methoxyethoxy)-1,4-benzenedicarboxylate, dabco = 1,4-diazabicyclo[2.2.2]octane) shows remarkable structural changes upon selective adsorption of CO2 as determined by in situ X-ray diffraction at 195 K. Upon accommodation of carbon dioxide [Zn2(BME-bdc)2(dabco)]n transfers from a narrow pore form to an open pore form, which exhibits a much higher unit cell volume. Due to the slow adsorption kinetics an unexpected metastable intermediate form could be identified.
