Sediments play an important role in organic matter mineralisation and nutrient recycling, especially in shallow marine systems. Marine ecosystem models, however, often only include a coarse representation of processes beneath the sea floor. While these parameterisations may give a reasonable description of the present ecosystem state, they lack predictive capacity for possible future changes, which can only be obtained from mechanistic modelling. This paper describes an integrated benthic–pelagic ecosystem model developed for the German Exclusive Economic Zone (EEZ) in the western Baltic Sea. The model is a hybrid of two existing models: the pelagic part of the marine ecosystem model ERGOM and an early diagenetic model by Reed et al. (2011). The latter one was extended to include the carbon cycle, a determination of precipitation and dissolution reactions which accounts for salinity differences, an explicit description of the adsorption of clay minerals, and an alternative pyrite formation pathway. We present a one-dimensional application of the model to seven sites with different sediment types. The model was calibrated with observed pore water profiles and validated with results of sediment composition, bioturbation rates and bentho-pelagic fluxes gathered by in situ incubations of sediments (benthic chambers). The model results generally give a reasonable fit to the observations, even if some deviations are observed, e.g. an overestimation of sulfide concentrations in the sandy sediments. We therefore consider it a good first step towards a three-dimensional representation of sedimentary processes in coupled pelagic–benthic ecosystem models of the Baltic Sea.

An effective way to reduce complexity in ecological modelling is by grouping species that share similar characteristics into functional groups or types. Often, the creation of plant functional groups (PFGs) is carried out for each case study in an ad-hoc way using a small set of traits. This limits the transferability of these PFGs to other geographical areas or study systems. We propose a novel generic framework to generate PFGs that considers the most important ecological dimensions, is applicable to case studies globally, and that emerges from patterns of functional redundancy across species. Based on most relevant and measured plant characteristics, we designed a multi-step process that includes: i) data harmonisation and missing values imputation; ii) species clustering based on multiple characteristics encompassing the main ecological dimensions featured in plant community ecological models (i.e., dispersal, competition, and demography) and iii) the combination of ecological dimension-specific groups into comprehensive PFGs. We demonstrate this framework by applying it to a global dataset of plant characteristics including a functional traits dataset and a plant-soil co-occurrence dataset for 19,102 species. Lastly, to test the ability of generated PFGs to summarise species’ functional variation within plant communities, we correlate taxonomical and functional diversity indices calculated at the species and at the PFGs level across a global dataset of plant communities (sPlotOpen). Our framework generated 465 global, robust data-driven PFGs with non-overlapping combinations of traits for each ecological dimension divided by growth form. The validation returned positive correlation values between PFGs and species-level diversity metrics, supporting the ability of the obtained PFGs to capture functional and taxonomic diversity patterns across a variety of plant communities worldwide. The framework allows for the easy integration of newly available species characteristics data. The obtained global PFGs, covering all main known ecological processes and environmental conditions at small resolution, can increase the predictive power and accuracy of process-based models and help furthering varying-scale ecological studies.

We report on Baltic Sea polycyclic aromatic hydrocarbon (PAH) pressure based on the U.S. EPA PAHs in view of millennial and decadal developments utilizing data from sediment deposits and seawater. Pre-industrial PAH contents ranged from 500 to 4500 ng/g TOC. Maximum PAH contents were up to 100,000 ng/g TOC and date back to the 1960s to 1970s with maximum pollutant inputs. Contemporary sediment PAH contents with 10,000 to 35,000 ng/g TOC and seawater concentrations with about 1 to 16 ng/l show spatial variability due to different local inputs and sediment characteristics. Pre-industrial compositional change from low molecular weight to high molecular weight (HMW) PAH indicates changing sources from mostly petroleum to combustion. Application of diagnostic ratios reveals petroleum and combustion as contemporary PAH sources and that traffic emissions continuously contribute to the Baltic PAH profile. REACH aims at promoting the safe use of chemicals in Europe, inter alia by identification and regulation of substances of very high concern (SVHCs). Once identified, SVHCs need to be substituted by safer alternatives. However, substitutes are frequently not safer than the substances that they replace but rather show similar hazard profiles, resulting in regrettable substitution. This paper investigates the impact of chemicals regulation on substitution of chemicals by analyzing time trends in the industrial use of chemicals from 2000 to 2014 in Scandinavia. It is shown that the use of ten water-relevant SVHCs decreased by about 90% in the considered period in Sweden as compared to a control group of unregulated substances which decreased by only 20%. A closer inspection of the use of 23 highly used plasticizers revealed that the use of regulated phthalate plasticizers decreased while the use of non-phthalate plasticizers increased. A first comparison of hazardous properties showed that during the 15-years period chemical substitution drastically reduced the chemical hazard burden of plasticizers in Scandinavia for both, the environment and human health. This study shows that regulation and the related discussion on chemicals safety have significantly reduced the chemical hazard burden from plasticizers in Scandinavia since the year 2000. It is assumed that similar trends can be found for the whole European Union. To combat regrettable substitution, mitigation options are suggested, including information-based tools for the identification of safer alternatives and an improved accessibility of information on production volumes and uses of chemicals to allow for an improved assessment of chemical's risk., (Copyright © 2018. Since the 1950s the organotin compound tributyltin (TBT) was intensively used in antifouling paints for marine vessels and it became of concern for the marine environment. Herein, we report on a study from 2015 on TBT and its metabolites monobutyltin (MBT) and dibutyltin (DBT) in sediments from the central Baltic Sea and a Baltic Sea coastal site with strong harbor activities (Warnemünde). Sublayers from a sediment core from the Arkona Basin were analyzed to investigate the long term organotin pressure for the Baltic Sea. For the central Baltic Sea total organotin (MBT+DBT+TBT) ranged from 100 to 500ng/g TOC with distinct areas of high organotin content probably due to historical inputs. For the coastal site total organotin ranged from 10,000 to 60,000ng/g TOC. MBT and DBT were the predominant organotin species detected. Water is a vital resource for natural ecosystems and human life, and assuring a high quality of water and protecting it from chemical contamination is a major societal goal in the European Union. The Water Framework Directive (WFD) and its daughter directives are the major body of legislation for the protection and sustainable use of European freshwater resources. The practical implementation of the WFD with regard to chemical pollution has faced some challenges. In support of the upcoming WFD review in 2019 the research project SOLUTIONS and the European monitoring network NORMAN has analyzed these challenges, evaluated the state-of-the-art of the science and suggested possible solutions. We give 10 recommendations to improve monitoring and to strengthen comprehensive prioritization, to foster consistent assessment and to support solution-oriented management of surface waters. The integration of effect-based tools, the application of passive sampling for bioaccumulative chemicals and an integrated strategy for prioritization of contaminants, accounting for knowledge gaps, are seen as important approaches to advance monitoring. Including all relevant chemical contaminants in more holistic "chemical status" assessment, using effect-based trigger values to address priority mixtures of chemicals, to better consider historical burdens accumulated in sediments and to use models to fill data gaps are recommended for a consistent assessment of contamination. Solution-oriented management should apply a tiered approach in investigative monitoring to identify toxicity drivers, strengthen consistent legislative frameworks and apply solutions-oriented approaches that explore risk reduction scenarios before and along with risk assessment., (Copyright © 2016. Background: This paper describes a conceptual framework for solutions-focused management of chemical contaminants built on novel and systematic approaches for identifying, quantifying and reducing risks of these substances., Methods: The conceptual framework was developed in interaction with stakeholders representing relevant authorities and organisations responsible for managing environmental quality of water bodies. Stakeholder needs were compiled via a survey and dialogue. The content of the conceptual framework was thereafter developed with inputs from relevant scientific disciplines., Results: The conceptual framework consists of four access points: Chemicals, Environment, Abatement and Society, representing different aspects and approaches to engaging in the issue of chemical contamination of surface waters. It widens the scope for assessment and management of chemicals in comparison to a traditional (mostly) perchemical risk assessment approaches by including abatement- and societal approaches as optional solutions. The solution-focused approach implies an identification of abatement- and policy options upfront in the risk assessment process. The conceptual framework was designed for use in current and future chemical pollution assessments for the aquatic environment, including the specific challenges encountered in prioritising individual chemicals and mixtures, and is applicable for the development of approaches for safe chemical management in a broader sense. The four access points of the conceptual framework are interlinked by four key topics representing the main scientific challenges that need to be addressed, i.e.: identifying and prioritising hazardous chemicals at different scales; selecting relevant and efficient abatement options; providing regulatory support for chemicals management; predicting and prioritising future chemical risks. The conceptual framework aligns current challenges in the safe production and use of chemicals. The current state of knowledge and implementation of these challenges is described., Conclusions: The use of the conceptual framework, and addressing the challenges, is intended to support: (1) forwarding sustainable use of chemicals, (2) identification of pollutants of priority concern for cost-effective management, (3) the selection of optimal abatement options and (4) the development and use of optimised legal and policy instruments.

SOLUTIONS (2013 to 2018) is a European Union Seventh Framework Programme Project (EU-FP7). The project aims to deliver a conceptual framework to support the evidence-based development of environmental policies with regard to water quality. SOLUTIONS will develop the tools for the identification, prioritisation and assessment of those water contaminants that may pose a risk to ecosystems and human health. To this end, a new generation of chemical and effect-based monitoring tools is developed and integrated with a full set of exposure, effect and risk assessment models. SOLUTIONS attempts to address legacy, present and future contamination by integrating monitoring and modelling based approaches with scenarios on future developments in society, economy and technology and thus in contamination. The project follows a solutions-oriented approach by addressing major problems of water and chemicals management and by assessing abatement options. SOLUTIONS takes advantage of the access to the infrastructure necessary to investigate the large basins of the Danube and Rhine as well as relevant Mediterranean basins as case studies, and puts major efforts on stakeholder dialogue and support. Particularly, the EU Water Framework Directive (WFD) Common Implementation Strategy (CIS) working groups, International River Commissions, and water works associations are directly supported with consistent guidance for the early detection, identification, prioritisation, and abatement of chemicals in the water cycle. SOLUTIONS will give a specific emphasis on concepts and tools for the impact and risk assessment of complex mixtures of emerging pollutants, their metabolites and transformation products. Analytical and effect-based screening tools will be applied together with ecological assessment tools for the identification of toxicants and their impacts. The SOLUTIONS approach is expected to provide transparent and evidence-based candidates or River Basin Specific Pollutants in the case study basins and to assist future review of priority pollutants under the WFD as well as potential abatement options., (Copyright © 2014 Elsevier B.V. Background: Different types of indicators have been developed to describe the impact of chemicals on society and environment. Due to the high number of substances and their different types of use, most of these indicators are directed to specific areas of interest--regarding workplace safety, environmental health or consumer health. They address a specific subset of chemicals and can be used for monitoring enterprise-specific, national or international management measures., Main Features: A survey of existing indicators for chemicals has shown that indicators already exist for a remarkable number of problem fields. As soon as the release and the environmental fate of chemicals are taken into account, the complexity of the approaches increases considerably. The distinction between indicators for drivers, pressures, state, impacts and responses, as proposed by the European Environmental Agency, supports the identification of proper indicators for a specific type of problem., Discussion and Conclusions: No single indicator exists which is able to cover the whole range of chemicals and their applications. Several indicator approaches cover at least a subset of the most relevant substances. If they are intended to be used for European monitoring, robust data must be provided by EU Member States. Chemicals in enterprises (ancillary inputs as well as process chemicals) are an important element of in-plant material flow management--in terms of occupational safety and health as well as environmental protection. Existing indicators for hazardous chemicals can be a valuable tool for process and product refinement regarding hazardous chemicals, especially for enterprises., Outlook: Indicators for production and impact of chemicals, as well as policy performance indicators, are essential elements in order to monitor the management of chemicals. They have to be established for the national and for the EU level.

Any explanation of the causes of Alzheimer's disease and of its unique cerebral pathologic features must take into account the distribution and ultrastructural localization of the pre-A4 amyloid proteins in tissues and organs. The authors have analyzed the expression of the pre-A4 amyloid proteins in several tissues by immunogold electron microscopy and by immunofluorescence. For this purpose, they have used a mouse monoclonal antibody and a guinea pig antiserum raised against two synthetic peptides corresponding to two different sequences common to all the full-length forms of the A4 amyloid precursors. They observed a tissue-specific distribution of the secreted and the transmembrane form of the precursors. The authors could determine that the secreted form is generated in vivo within the cytoplasm. In the salivary glands and in the adenohypophysis, all the immunoreactivity is associated with the process of secretion, whereas in the muscle, a staining pattern compatible with the presence of the pre-A4 amyloid proteins in the sarcoplasmic reticulum has been observed. This difference in the localization may reflect tissue-specific processing pathways and suggests that posttranslational modifications such as proteolytic removal of the transmembrane and cytoplasmic domains contribute to the structural and thus functional diversity of the A4 amyloid precursors.

The ultrastructure of spermatozoa and its genesis (spermiogenesis) have been investigated in the interstitial annelid Potamodrilus fluviatilis. The mature spermatozoa are threadlike cells which are bent at the base of the flagellum, giving the cell a hairpinlike appearance. The acrosome consists of an unusual, long, flasklike vesicle with a granum in its basal part. The cylindrical nuclear region is characterized by a monolayer of vesicles enwrapping the posterior half of the nucleus. This region is endowed with a number of altered rodlike mitochondria. No middlepiece is present. The basal body of the flagellum is obliquely arranged with respect to the long axis, giving rise to a curved flagellum, which, along most of its length, exhibits a thick layer of vacuolized cytoplasm around the axoneme. During spermiogenesis, which occurs in the body fluid, spermatids develop at the surface of syncytial masses which have been formed during meiotic divisions. The acrosome protrudes in the distal part of the cell, while the basal body of the flagellum is shifted toward the proximal region, which connects the cell with the cytophore. These are unusual features in annelid spermiogenesis. As indicated in Discussion, the phylogenetic implications of these findings include the assumption that Potamodrilus is not related to any oligochaete or even any other clitellate group or species and, hence, has to be excluded from these taxa., (Copyright © 1985 Wiley-Liss, Inc.)

To study the putative precursor proteins (PreA4(695), PreA4(751), and PreA4(770] of Alzheimer's disease A4 amyloid protein, polyclonal and monoclonal antibodies were raised against a recombinant bacterial PreA4(695) fusion protein. These antibodies were used to identify the precursors in different cell lines as well as in human brain homogenates and cerebrospinal fluid (CSF). The precursors are tyrosine-sulfated, O- and N-glycosylated membrane proteins and have half-lives of 20-30 min in cells. Cells express the polypeptides at their surface but also secrete C-terminal truncated proteins into the medium. These proteins are also found in CSF of both Alzheimer's disease patients and normal individuals. The proteins are derived from their cognate membrane-associated forms by proteolysis and have apparently lost the cytoplasmic and the transmembrane domains. Since the latter contributes to the A4 amyloid sequence, it seems possible that this proteolytic cleavage represents the first step in the formation of A4 amyloid deposits.
