1. Analysis of water quality at hydrographic basin scale using satellite images, co-occurrence matrices and Bayes classifier

Abstract: Given the increased risks of water scarcity and the presence of polluting agents in water resources, this paper aims at the development and presentation of a computational tool capable of assessing water quality based on digital processing techniques applied to satellite images. Initially, a database was created for Brazilian regions, consisting of hydrographic basins' satellite images associated with the Water Quality Index (WQI), according to the criteria established by the National Water Agency (ANA). Hitherto, the database consisted of 85 images, 61 were used in the training stage and 24 in the testing stage. In both stages, the images were subjected to thresholding using Otsu's Method, binarization, linear expansion on saturation, application of a Laplacian filter, extraction of characteristics by using co-occurrence matrices and classification by the Bayes Discriminant. Such techniques were also implemented on a computational platform in the MATLAB® environment, responsible for the interface between the system and users. The proposed system presented an approximate 70% success rate regarding the classification of WQIs, which can be improved as more information is made available to improve the databases. 2. 3D{\pi}: Three-Dimensional Positron Imaging, A Novel Total-Body PET Scanner Using Xenon-Doped Liquid Argon Scintillator

Abstract: Objective: This paper introduces a novel PET imaging methodology called 3-dimensional positron imaging (3D{\pi}), which integrates total-body (TB) coverage, time-of-flight (TOF) technology, ultra-low dose imaging capabilities, and ultra-fast readout electronics inspired by emerging technology from the DarkSide collaboration. Approach: The study evaluates the performance of 3D{\pi} using Monte Carlo simulations based on NEMA NU 2-2018 protocols. The methodology employs a homogenous, monolithic scintillator composed of liquid argon (LAr) doped with xenon (Xe) with silicon photomultipliers (SiPM) operating at cryogenic temperatures. Main results: Significant enhancements in system performance are observed, with the 3D{\pi} system achieving a noise equivalent count rate (NECR) of 3.2 Mcps which is approximately two times higher than uEXPLORER's peak NECR (1.5 Mcps) at 17.3 (kBq/mL). Spatial resolution measurements show an average FWHM of 2.7 mm across both axial positions. The system exhibits superior sensitivity, with values reaching 373 kcps/MBq with a line source at the center of the field of view. Additionally, 3D{\pi} achieves a TOF resolution of 151 ps at 5.3 kBq/mL, highlighting its potential to produce high-quality images with reduced noise levels. Significance: The study underscores the potential of 3D{\pi} in improving PET imaging performance, offering the potential for shorter scan times and reduced radiation exposure for patients. The Xe-doped LAr offers advantages such as fast scintillation, enhanced light yield, and cost-effectiveness. Abstract: DarkSide-20k (DS-20k) is a dark matter detection experiment under construction at the Laboratori Nazionali del Gran Sasso (LNGS) in Italy. It utilises ~100 t of low radioactivity argon from an underground source (UAr) in its inner detector, with half serving as target in a dual-phase time projection chamber (TPC). The UAr cryogenics system must maintain stable thermodynamic conditions throughout the experiment's lifetime of >10 years. Continuous removal of impurities and radon from the UAr is essential for maximising signal yield and mitigating background. We are developing an efficient and powerful cryogenics system with a gas purification loop with a target circulation rate of 1000 slpm. Central to its design is a condenser operated with liquid nitrogen which is paired with a gas heat exchanger cascade, delivering a combined cooling power of >8 kW. Here we present the design choices in view of the DS-20k requirements, in particular the condenser's working principle and the cooling control, and we show test results obtained with a dedicated benchmarking platform at CERN and LNGS. We find that the thermal efficiency of the recirculation loop, defined in terms of nitrogen consumption per argon flow rate, is 95 % and the pressure in the test cryostat can be maintained within $\pm$(0.1-0.2) mbar. We further detail a 5-day cool-down procedure of the test cryostat, maintaining a cooling rate typically within -2 K/h, as required for the DS-20k inner detector. Additionally, we assess the circuit's flow resistance, and the heat transfer capabilities of two heat exchanger geometries for argon phase change, used to provide gas for recirculation. We conclude by discussing how our findings influence the finalisation of the system design, including necessary modifications to meet requirements and ongoing testing activities., Comment: 45 pages, 24 figures

Abstract: The dual-phase liquid argon time projection chamber is presently one of the leading technologies to search for dark matter particles with masses below 10 GeV/c$^2$. This was demonstrated by the DarkSide-50 experiment with approximately 50 kg of low-radioactivity liquid argon as target material. The next generation experiment DarkSide-20k, currently under construction, will use 1,000 times more argon and is expected to start operation in 2027. Based on the DarkSide-50 experience, here we assess the DarkSide-20k sensitivity to models predicting light dark matter particles, including Weakly Interacting Massive Particles (WIMPs) and sub-GeV/c$^2$ particles interacting with electrons in argon atoms. With one year of data, a sensitivity improvement to dark matter interaction cross-sections by at least one order of magnitude with respect to DarkSide-50 is expected for all these models. A sensitivity to WIMP--nucleon interaction cross-sections below $1\times10^{-42}$ cm$^2$ is achievable for WIMP masses above 800 MeV/c$^2$. With 10 years exposure, the neutrino fog can be reached for WIMP masses around 5 GeV/c$^2$., Comment: submitted to Nature Communications

Abstract: Experiments aimed at direct searches for WIMP dark matter require highly effective reduction of backgrounds and control of any residual radioactive contamination. In particular, neutrons interacting with atomic nuclei represent an important class of backgrounds due to the expected similarity of a WIMP-nucleon interaction, so that such experiments often feature a dedicated neutron detector surrounding the active target volume. In the context of the development of DarkSide-20k detector at INFN Gran Sasso National Laboratory (LNGS), several R&D projects were conceived and developed for the creation of a new hybrid material rich in both hydrogen and gadolinium nuclei to be employed as an essential element of the neutron detector. Thanks to its very high cross-section for neutron capture, gadolinium is one of the most widely used elements in neutron detectors, while the hydrogen-rich material is instrumental in efficiently moderating the neutrons. In this paper results from one of the R&Ds are presented. In this effort the new hybrid material was obtained as a poly(methyl methacrylate) (PMMA) matrix, loaded with gadolinium oxide in the form of nanoparticles. We describe its realization, including all phases of design, purification, construction, characterization, and determination of mechanical properties of the new material.

6. MIL-101(Fe)@ceramic-monolith for arsenic removal in aqueous solutions

Abstract: Abstract In this work, we present the preparation of a hybrid material constituted by a Metal-Organic Framework (type MIL-101(Fe)) and a natural clay ceramic monolith (NCCM), the materials were successfully assembled through an in-situ hydrothermal method. The composites were characterized by powder X-ray diffraction (PXRD), thermogravimetric analysis (TGA), Fourier-transform infrared spectroscopy (FTIR), X-ray photoelectron spectroscopy (XPS), scanning electron microscopy (SEM), N2 adsorption-desorption isotherms at 77 K and CO2 adsorption at 273 K; these techniques confirmed the obtaining of the hybrid material and, in addition, through studies before and after adsorption mechanisms involved were identified. Then, the As(V) adsorption capacity of the materials was evaluated using an aqueous arsenic solution; from which high adsorption capacities of up to 268 mg g− 1 and 61.5 mg g− 1, respectively. In addition, kinetic and equilibrium studies of As(V) adsorption were carried out. Finally, the potential capacities of the hybrid material consisting of Al2O3, AlO6-SiO4 and Fe3O4 for the adsorption of H3AsO4, were studied through density functional theory calculations.

7. Assessment of Urban Heat Islands in an Eastern Amazonian city

Abstract: The increase in heat islands in large cities, especially in tropical areas, has been a significant problem, as the rise in temperature can cause health issues for people. Therefore, this research aimed to analyze changes in land surface temperature (LST) and urban heat islands (UHI) in 1986 and 2023. The methodological procedures were carried out in four steps: I) Calculation of LST and UHI using the Landsat collection; II) Zonal statistics of the mean, the median, the minimum, maximum, and standard deviation of LST and UHI in each neighbourhood; III) Kruskal-Wallis Test applied to the average LST between 1986 and 2023; and IV) Assessment of the spatial autocorrelation of neighbourhoods using Bivariate Global and Local Moran’s I Index between LST and UHI. The results showed that in 2023, there was an increase in heat islands in central city neighbourhoods compared to 1986, as demonstrated by the Kruskal-Wallis test, which showed significant differences in LST. The global Moran’s I Index presented a value of 0.642, indicating a robust spatial autocorrelation in local studies. The neighbourhoods that showed high correlations were Campina, Cidade Velha, Cremação, Condor, Fátima, Jurunas, Nazaré, Pedreira, Reduto, São Braz, and Umarizal. The neighbourhoods that showed low correlations were Águas Lindas, Aurá, Curió-Utinga, Guanabara, and Parque Guajará. These results are directly linked to the intensification of urbanization and low vegetation cover, especially in central areas of the city, which showed a high correlation. These findings can aid decision-makers and support urban planning, focusing on neighbourhoods with higher average temperatures.

Abstract: The direct search for dark matter in the form of weakly interacting massive particles (WIMP) is performed by detecting nuclear recoils (NR) produced in a target material from the WIMP elastic scattering. A promising experimental strategy for direct dark matter search employs argon dual-phase time projection chambers (TPC). One of the advantages of the TPC is the capability to detect both the scintillation and charge signals produced by NRs. Furthermore, the existence of a drift electric field in the TPC breaks the rotational symmetry: the angle between the drift field and the momentum of the recoiling nucleus can potentially affect the charge recombination probability in liquid argon and then the relative balance between the two signal channels. This fact could make the detector sensitive to the directionality of the WIMP-induced signal, enabling unmistakable annual and daily modulation signatures for future searches aiming for discovery. The Recoil Directionality (ReD) experiment was designed to probe for such directional sensitivity. The TPC of ReD was irradiated with neutrons at the INFN Laboratori Nazionali del Sud, and data were taken with 72 keV NRs of known recoil directions. The direction-dependent liquid argon charge recombination model by Cataudella et al. was adopted and a likelihood statistical analysis was performed, which gave no indications of significant dependence of the detector response to the recoil direction. The aspect ratio R of the initial ionization cloud is estimated to be 1.037 +/- 0.027 and the upper limit is R < 1.072 with 90% confidence level, Comment: 20 pages, 10 figures, submitted to Eur. Abstract: Dark matter lighter than 10 GeV/c$^2$ encompasses a promising range of candidates. A conceptual design for a new detector, DarkSide-LowMass, is presented, based on the DarkSide-50 detector and progress toward DarkSide-20k, optimized for a low-threshold electron-counting measurement. Sensitivity to light dark matter is explored for various potential energy thresholds and background rates. These studies show that DarkSide-LowMass can achieve sensitivity to light dark matter down to the solar neutrino floor for GeV-scale masses and significant sensitivity down to 10 MeV/c$^2$ considering the Migdal effect or interactions with electrons. Requirements for optimizing the detector's sensitivity are explored, as are potential sensitivity gains from modeling and mitigating spurious electron backgrounds that may dominate the signal at the lowest energies.

Abstract: BackgroundCabo Verde, an island country in West Africa, has been affected since human colonization by epidemics of vector-borne diseases with major epidemics of dengue and zika in recent years. Although there is a national program for integrated vector control, innovative strategies that reinforce routine activities and strengthen vector control are necessary to prevent the emergence or reemergence of arboviruses and new epidemics of dengue and zika. Insecticide paints are evidenced as new technologies for the formulation of insecticides in a more residual and safe way. The TINTAEDES project aimed to assess the efficacy, acceptability, and operational deployment of an insecticide paint for Aedes control.Methodology/Principal findingsLaboratory and small-scale field trials were conducted, assessing mortality through World Health Organization cone bioassays. A community-based intervention study in the neighborhoods of Várzea and Tira Chapéu in the city of Praia, Cabo Verde, was developed. The intervention is a paint self-application model by homeowners and neighborhood volunteers. The intervention was evaluated based on entomological indicators and the responses given by the residents of the painted houses to a questionnaire on the knowledge, satisfaction, and safety of insecticidal paints. A transfluthrin-based insecticide paint was effective against wild Ae. aegypti for one year in the laboratory and semi-field conditions. Residents largely perceived a reduction in mosquito presence in the treated houses (98%).ConclusionInsecticide paints are presented as an effective innovation strategy for mosquito control, which could be implemented as a reinforcement of the measures carried out by the vector control program in the city of Praia and throughout the country.

Abstract: The Aria project consists of a plant, hosting a 350 m cryogenic isotopic distillation column, the tallest ever built, which is currently in the installation phase in a mine shaft at Carbosulcis S.p.A., Nuraxi-Figus (SU), Italy. Aria is one of the pillars of the argon dark-matter search experimental program, lead by the Global Argon Dark Matter Collaboration. Aria was designed to reduce the isotopic abundance of $^{39}$Ar, a $\beta$-emitter of cosmogenic origin, whose activity poses background and pile-up concerns in the detectors, in the argon used for the dark-matter searches, the so-called Underground Argon (UAr). In this paper, we discuss the requirements, design, construction, tests, and projected performance of the plant for the isotopic cryogenic distillation of argon. We also present the successful results of isotopic cryogenic distillation of nitrogen with a prototype plant, operating the column at total reflux.

Abstract: Abstract Pollination is vital for ecosystem functioning, especially in biodiversity‐rich regions like the Brazilian Cerrado. Our research establishes a comprehensive meta network of pollinator–plant interactions within this biome. We quantified the importance of different pollinator groups, identifying keystone species. We examined potential biases in sampling effort and the spatial behavior of interactions within the heterogeneous Cerrado plant physiognomies. Our investigation uncovered 1499 interactions among 293 plant species and 386 visitor species, with legitimate pollination accounting for 42.4% of the interactions. The network exhibited modularity, driven by bees and insects, with vertebrates bridging diurnal and nocturnal modules. While a generalized pattern emerged, high specialization existed within modules due to habitat diversity. Bees, particularly Apis mellifera (exotic) and Trigona spinipes (native), played central roles as network hubs. Hummingbirds and bats, engaged in specialized interactions showing strong connectivity within and between modules. Interestingly, invertebrate–vertebrate modules were more prevalent than expected in the meta network. However, a bias was evident, primarily within specific biogeographical districts with fragmented landscapes and intrusion from other biomes. Variations in plant species and endemism rates influenced pollinator occurrence and the Cerrado network topology. Our study offers valuable insights into pollinator–plant interactions within the Cerrado, encompassing both invertebrates and vertebrates. The modeled network represents a significant step in understanding the structural complexity of pollination networks, integrating partial networks from diverse pollination systems within heterogeneous habitats. Nevertheless, a biogeographical bias could limit a comprehensive understanding of network functionality across the Cerrado.

Abstract: Future liquid-argon DarkSide-20k and ARGO detectors, designed for direct dark matter search, will be sensitive also to core-collapse supernova neutrinos, via coherent elastic neutrino-nucleus scattering. This interaction channel is flavor-insensitive with a high-cross section, enabling for a high-statistics neutrino detection with target masses of $\sim$50~t and $\sim$360~t for DarkSide-20k and ARGO, respectively. Thanks to the low-energy threshold of $\sim$0.5~keV$_{nr}$ achievable by exploiting the ionization channel, DarkSide-20k and ARGO have the potential to discover supernova bursts throughout our galaxy and up to the Small Magellanic Cloud, respectively, assuming a 11-M$_{\odot}$ progenitor star. We report also on the sensitivity to the neutronization burst, whose electron neutrino flux is suppressed by oscillations when detected via charged current and elastic scattering. Finally, the accuracies in the reconstruction of the average and total neutrino energy in the different phases of the supernova burst, as well as its time profile, are also discussed, taking into account the expected background and the detector response., Comment: 21 pages, 8 figures

Abstract: Proportional electroluminescence (EL) in noble gases is used in two-phase detectors for dark matter searches to record (in the gas phase) the ionization signal induced by particle scattering in the liquid phase. The "standard" EL mechanism is considered to be due to noble gas excimer emission in the vacuum ultraviolet (VUV). In addition, there are two alternative mechanisms, producing light in the visible and near infrared (NIR) ranges. The first is due to bremsstrahlung of electrons scattered on neutral atoms ("neutral bremsstrahlung", NBrS). The second, responsible for electron avalanche scintillation in the NIR at higher electric fields, is due to transitions between excited atomic states. In this work, we have for the first time demonstrated two alternative techniques of the optical readout of two-phase argon detectors, in the visible and NIR range, using a silicon photomultiplier matrix and electroluminescence due to either neutral bremsstrahlung or avalanche scintillation. The amplitude yield and position resolution were measured for these readout techniques, which allowed to assess the detection threshold for electron and nuclear recoils in two-phase argon detectors for dark matter searches. Abstract: Large liquid argon detectors offer one of the best avenues for the detection of galactic weakly interacting massive particles (WIMPs) via their scattering on atomic nuclei. The liquid argon target allows exquisite discrimination between nuclear and electron recoil signals via pulse-shape discrimination of the scintillation signals. Atmospheric argon (AAr), however, has a naturally occurring radioactive isotope, $^{39}$Ar, a $\beta$ emitter of cosmogenic origin. For large detectors, the atmospheric $^{39}$Ar activity poses pile-up concerns. The use of argon extracted from underground wells, deprived of $^{39}$Ar, is key to the physics potential of these experiments. The DarkSide-20k dark matter search experiment will operate a dual-phase time projection chamber with 50 tonnes of radio-pure underground argon (UAr), that was shown to be depleted of $^{39}$Ar with respect to AAr by a factor larger than 1400. Assessing the $^{39}$Ar content of the UAr during extraction is crucial for the success of DarkSide-20k, as well as for future experiments of the Global Argon Dark Matter Collaboration (GADMC). This will be carried out by the DArT in ArDM experiment, a small chamber made with extremely radio-pure materials that will be placed at the centre of the ArDM detector, in the Canfranc Underground Laboratory (LSC) in Spain. The ArDM LAr volume acts as an active veto for background radioactivity, mostly $\gamma$-rays from the ArDM detector materials and the surrounding rock. Abstract: Abstract The Aria cryogenic distillation plant, located in Sardinia, Italy, is a key component of the DarkSide-20k experimental program for WIMP dark matter searches at the INFN Laboratori Nazionali del Gran Sasso, Italy. Aria is designed to purify the argon, extracted from underground wells in Colorado, USA, and used as the DarkSide-20k target material, to detector-grade quality. In this paper, we report the first measurement of argon isotopic separation by distillation with the 26 m tall Aria prototype. We discuss the measurement of the operating parameters of the column and the observation of the simultaneous separation of the three stable argon isotopes: $${}^{36}\hbox {Ar}$$ 36 Ar , $${}^{38}\textrm{Ar}$$ 38 Ar , and $${}^{40}\textrm{Ar}$$ 40 Ar . We also provide a detailed comparison of the experimental results with commercial process simulation software. This measurement of isotopic separation of argon is a significant achievement for the project, building on the success of the initial demonstration of isotopic separation of nitrogen using the same equipment in 2019.

Abstract: Abstract Two different reactive concentrated magnesium hydroxide (HM) pulps, produced in a lab reactor mill, and a synthetic and non-reactive HM-pulp, for comparative analyses, have been prepared and studied. Sodium polyacrylate was selected as a dispersant for improving the HM pulp fluidity, with concentrations varying from 0.42% to 2.0% on a dry basis. The two reactive pulps differed from each other mainly by the impurity level and for both, a residual MgO amount was detected, implying that the hydration reaction could proceed. Results (with and without dispersant) confirmed the formation of particle clusters as HM concentration rose until a critical value, above which the pulp behaved as a solid. Reactive HM-pulps with high impurities may represent a limiting factor in the drying operation. Among all HM-pulps studied, the purest reactive pulp with 1% dispersant presented the best flow behavior making this reactive pulp the most suitable to be tested in a pilot spray dryer.

Abstract: Abstract Background Yeasts are gaining attention as alternative ingredients in aquafeeds. However, the impact of yeast inclusion on modulation of intestinal microbiota of fish fed plant-based ingredients is limited. Thus, the present study investigates the effects of yeast and processing on composition, diversity and predicted metabolic capacity of gut microbiota of Atlantic salmon smolt fed soybean meal (SBM)-based diet. Two yeasts, Cyberlindnera jadinii (CJ) and Wickerhamomyces anomalus (WA), were produced in-house and processed by direct heat-inactivation with spray-drying (ICJ and IWA) or autolyzed at 50 °C for 16 h, followed by spray-drying (ACJ and AWA). In a 42-day feeding experiment, fish were fed one of six diets: a fishmeal (FM)-based diet, a challenging diet with 30% SBM and four other diets containing 30% SBM and 10% of each of the four yeast products (i.e., ICJ, ACJ, IWA and AWA). Microbial profiling of digesta samples was conducted using 16S rRNA gene sequencing, and the predicted metabolic capacities of gut microbiota were determined using genome-scale metabolic models. Results The microbial composition and predicted metabolic capacity of gut microbiota differed between fish fed FM diet and those fed SBM diet. The digesta of fish fed SBM diet was dominated by members of lactic acid bacteria, which was similar to microbial composition in the digesta of fish fed the inactivated yeasts (ICJ and IWA diets). Inclusion of autolyzed yeasts (ACJ and AWA diets) reduced the richness and diversity of gut microbiota in fish. The gut microbiota of fish fed ACJ diet was dominated by the genus Pediococcus and showed a predicted increase in mucin O-glycan degradation compared with the other diets. The gut microbiota of fish fed AWA diet was highly dominated by the family Bacillaceae. Conclusions The present study showed that dietary inclusion of FM and SBM differentially modulate the composition and predicted metabolic capacity of gut microbiota of fish. The inclusion of inactivated yeasts did not alter the modulation caused by SBM-based diet. Fish fed ACJ diet increased relative abundance of Pediococcus, and mucin O-glycan degradation pathway compared with the other diets.

Abstract: We present the design and characterization of TIGER (Turin Integrated Gem Electronics for Readout), a 64-channel ASIC developed for the readout of the CGEM (Cylindrical Gas Electron Multiplier) detector, the proposed inner tracker for the 2018 upgrade of the BESIII experiment, carried out at BEPCII in Beijing. Each ASIC channel features a charge sensitive amplifier coupled to a dual-branch shaper stage, optimized for timing and charge measurement, followed by a mixed-mode back-end that extracts and digitizes the timestamp and charge of the input signals. The time-of-arrival is provided by a set of low-power TDCs, based on analogue interpolation techniques, while the charge measurement is obtained either from the Time-over-Threshold information or with a sample-and-hold circuit. The ASIC has been fabricated in a 110 nm CMOS technology and designed to operate with a 1.2 V power supply, an input capacitance of about 100 pF, an input dynamic range between 3 and 50 fC, a power consumption of about 12 mW/channel and a sustained event rate of 60 kHz/channel. Abstract: Abstract Plant-induced resistance can be an important component of soybean mites biological control programs. This work evaluates the preference of predatory mite Neoseiulus californicus (Acari: Phytoseiidae) to soybean plants under single and multiple herbivory conditions by two-spotted spider mite Tetranychus urticae (Acari: Tetranychidae), and velvetbean caterpillar Anticarsia gemmatalis (Lepidoptera: Noctuidae). Using a Y olfactometer, the following scenarios were evaluated: soybean with no infestation and soybean infested with A. gemmatalis; soybean infested with T. urticae and A. gemmatalis, and soybean infested with T. urticae and with both T. urticae and A. gemmatalis. Volatile compounds released by plants were analyzed and identified by a Trace GC Ultra gas chromatograph coupled to a mass spectrometer with a solid phase micro-extraction ion-trap. The predatory mite N. californicus preferred soybean plants infested with T. urticae compared to those infested with A. gemmatalis. Multiple infestation did not interfere with its preference to T. urticae. Multiple herbivory of T. urticae and A. gemmatalis modified the chemical profile of volatile compounds emitted by soybean plants. However, it did not interfere with the search behavior of N. californicus. Out of the 29 identified compounds only five promoted predatory mite response. Thus, regardless of single or multiple herbivory by T. urticae with or without A. gemmatalis, the indirect induced resistance mechanisms operate similarly. As such, this mechanism contributes to an increase in the encounter rate between predator and prey for N. Californicus and T. urticae, and the efficacy of biological control of mites on soybean.

Abstract: Building on the successful experience in operating the DarkSide-50 detector, the DarkSide Collaboration is going to construct DarkSide-20k, a direct WIMP search detector using a two-phase Liquid Argon Time Projection Chamber (LArTPC) with an active (fiducial) mass of 23 t (20 t). The DarkSide-20k LArTPC will be deployed within a shield/veto with a spherical Liquid Scintillator Veto (LSV) inside a cylindrical Water Cherenkov Veto (WCV). Operation of DarkSide-50 demonstrated a major reduction in the dominant $^{39}$Ar background when using argon extracted from an underground source, before applying pulse shape analysis. Data from DarkSide-50, in combination with MC simulation and analytical modeling, shows that a rejection factor for discrimination between electron and nuclear recoils of $\gt3\times10^9$ is achievable. This, along with the use of the veto system, is the key to unlocking the path to large LArTPC detector masses, while maintaining an "instrumental background-free" experiment, an experiment in which less than 0.1 events (other than $\nu$-induced nuclear recoils) is expected to occur within the WIMP search region during the planned exposure. DarkSide-20k will have ultra-low backgrounds than can be measured in situ. This will give sensitivity to WIMP-nucleon cross sections of $1.2\times10^{-47}$ cm$^2$ ($1.1\times10^{-46}$ cm$^2$) for WIMPs of $1$ TeV$/c^2$ ($10$ TeV$/c^2$) mass, to be achieved during a 5 yr run producing an exposure of 100 t yr free from any instrumental background. DarkSide-20k could then extend its operation to a decade, increasing the exposure to 200 t yr, reaching a sensitivity of $7.4\times10^{-48}$ cm$^2$ ($6.9\times10^{-47}$ cm$^2$) for WIMPs of $1$ TeV$/c^2$ ($10$ TeV$/c^2$) mass.

Abstract: We report on the cryogenic characterization of Red Green Blue - High Density (RGB-HD) SiPMs developed at Fondazione Bruno Kessler (FBK) as part of the DarkSide program of dark matter searches with liquid argon time projection chambers. A dedicated setup was used to measure the primary dark noise, the correlated noise, and the gain of the SiPMs at varying temperatures. A custom-made data acquisition system and analysis software were used to precisely characterize these parameters. We demonstrate that FBK RGB-HD SiPMs with low quenching resistance (RGB-HD-LR$_q$) can be operated from 40 K to 300 K with gains in the range $10^5$ to $10^6$ and noise rates on the order of a few Hz/mm$^2$.

Abstract: The goal of this study was to evaluate the effect of a human observer on Rabbit Grimace Scale (RbtGS) scores. The study scored video footage taken of 28 rabbits before and after orthopedic surgery, as follows: 24 h before surgery ( baseline), 1 h after surgery ( pain), 3 h after analgesia administration ( analgesia), and 24 h after surgery ( 24h) in the presence and absence of an observer. Videos were assessed twice in random order by 3 evaluators who were blind to the collection time and the presence or absence of an observer. Responses to pain and analgesia were evaluated by comparing the 4 time points using the Friedman test, followed by the Dunn test. The influence of the presence or absence of the observer at each time point was evaluated using the Wilcoxon test. Intra- and interrater reliabilities were estimated using the intraclass correlation coefficient. The scale was responsive to pain, as the scores increased after surgery and had decreased by 24 h after surgery. The presence of the observer reduced significantly the RbtGS scores (median and range) at pain (present, 0.75, 0 to 1.75; absent, 1, 0 to 2) and increased the scores at baseline (present, 0.2, 0 to 2; absent, 0, 0 to 2) and 24h after surgery (present, 0.33, 0 to 1.75; absent, 0.2, 0 to 1.5). The intrarater reliability was good (0.69) to very good (0.82) and interrater reliability was moderate (0.49) to good (0.67). Thus, the RbtGS appeared to detect pain when scored from video footage of rabbits before and after orthopedic surgery. In the presence of the observer, the pain scores were underestimated at the time considered to be associated with the greatest pain and overestimated at the times of little or no pain.

Abstract: Lactic acid bacteria (LAB) are traditionally used in fermentation and food preservation processes and are recognized as safe for consumption. Recently, they have attracted attention due to their health-promoting properties; many species are already widely used as probiotics for treatment or prevention of various medical conditions, including inflammatory bowel diseases, infections, and autoimmune disorders. Some LAB, especially Lactococcus lactis, have been engineered as live vehicles for delivery of DNA vaccines and for production of therapeutic biomolecules. Here, we summarize work on engineering of LAB, with emphasis on the model LAB, L. lactis. We review the various expression systems for the production of heterologous proteins in Lactococcus spp. and its use as a live delivery system of DNA vaccines and for expression of biotherapeutics using the eukaryotic cell machinery. We have included examples of molecules produced by these expression platforms and their application in clinical disorders. We also present the CRISPR-Cas approach as a novel methodology for the development and optimization of food-grade expression of useful substances, and detail methods to improve DNA delivery by LAB to the gastrointestinal tract. Finally, we discuss perspectives for the development of medical applications of recombinant LABs involving animal model studies and human clinical trials, and we touch on the main safety issues that need to be taken into account so that bioengineered versions of these generally recognized as safe organisms will be considered acceptable for medical use.

Esse fato se deve ao progresso das técnicas médicas, o qual viabilizou a transferência dos enfermos e da morte para o contexto hospitalar. Objetivos: Conhecer as percepções de médicos residentes em relação à morte e ao preparo acadêmico e profissional para lidar com situações de óbito hospitalar. Métodos: Trata-se de estudo de abordagem quantitativa, descritiva e transversal a partir de um questionário estruturado aplicado em 40 residentes do Hospital Escola da Faculdade de Medicina de Itajubá (HE/FMIt). Resultados: Do total de residentes entrevistados, 62,5% afirmam terem tido capacitação acadêmica para lidar com a morte de pacientes; a preparação teórica e prática para comunicação de óbito aos familiares foi apontada como regular por 45,0% dos entrevistados; 57,5% sentem-se seguros no ato de transmitir a má noticia. Os recursos pessoais apontados pelos residentes para o enfrentamento das situações de óbito foram o controle emocional (60,0%) e o amor pela profissão (47,5%); os significados atribuídos à morte foram de Passagem (47,5%) e Processo (40,0%). Conclusão: Conclui-se que as concepções dos residentes revelam uma preparação para situações de morte e morrer no contexto hospitalar suficiente para a prática clínica, embora ainda com lacunas na formação, o que reforça a necessidade de estudos sobre o tema e o planejamento de ações de humanização no cuidado aos pacientes, melhorando a qualidade da assistência à saúde. This fact is due to the progress of medical techniques which, without qualification, made the transfer of patients and death to the hospital context possible. Aims: To know the perceptions of resident physicians in relation to death and to know their academic and professional preparation for dealing with deaths in hospitals. Methods: This is a quantitative, descriptive and cross-sectional study based on a structured questionnaire applied to 40 residents of the School Hospital of the Medical School of Itajubá (HE / FMIt). Results: From the total number of residents interviewed, 62.5% said they had had the academic preparation to deal with the death of patients; 45.0% said they had theoretical and practical preparation for the communication of death to family members and to the community; and 57.5% feel confident about giving the bad news. Concerning the human resources that were required for dealing with death-related situations pointed out by residents, they were emotional control (60.0%) and love for the profession (47.5%). As for the meanings attributed to death, they were Passage (47.5%) and Process (40.0%). Conclusion: It is concluded that although residents' conceptions reveal a preparation for death and dying in the hospital context that is sufficient for clinical practice, there are still gaps in their training, which reinforces the need for studies on the theme and the planning of humanization actions in the care of patients, improving the quality of health care. Abstract: BackgroundBlack soldier fly (Hermetia illucens) is a promising insect species to use as a novel ingredient in fish feeds. Black soldier fly larvae consists of three major fractions, namely protein, lipid, and exoskeleton. These fractions contain bioactive compounds that can modulate the gut microbiota in fish such as antimicrobial peptides, lauric acid, and chitin. However, it is not certain how, or which fractions of black solider fly would affect gut microbiota in fish. In the present study, black soldier fly larvae were processed into three different meals (full-fat, defatted and de-chitinized) and two fractions (oil and exoskeleton), and included in diets for Atlantic salmon (Salmo salar). Atlantic salmon pre-smolts were fed with these diets in comparison with a commercial-like control diet for eight weeks to investigate the effects of insect meals and fractions on the composition and predicted metabolic capacity of gut microbiota. The gut microbiota was profiled by 16S rRNA gene sequencing, and the predicted metabolic capacities of gut microbiota were determined using genome-scale metabolic models.ResultsThe inclusion of insect meals and fractions decreased abundance ofProteobacteriaand increased abundance ofFirmicutesin salmon gut. The diets that contained insect chitin, i.e., insect meals or exoskeleton diets, increased abundance of chitinolytic bacteria including lactic acid bacteria andActinomycesin salmon gut, with fish fed full-fat meal diet showing the highest abundances. The diets that contained insect lipids, i.e., insect meals and oil diets enrichedBacillaceaein fish gut. The fish fed diets containing full-fat insect meal had a unique gut microbiota composition dominated by beneficial lactic acid bacteria andActinomyces,and showed a predicted increase in mucin degradation compared to the other diets.ConclusionsThe present results showed that the dietary inclusion of insect meals and fractions can differently modulate the composition and predicted metabolic capacity of gut microbiota in Atlantic salmon pre-smolts. The use of full-fat black soldier fly larvae meal in diets for salmon is more favorable for beneficial modulation of gut microbiota than larvae processed by separation of lipid or exoskeleton fractions.

50. Microencapsulation of Lactic Acid Bacteria Improves the Gastrointestinal Delivery and in situ Expression of Recombinant Fluorescent Protein

Abstract: The microencapsulation process of bacteria has been used for many years, mainly in the food industry and, among the different matrixes used, sodium alginate stands out. This matrix forms a protective wall around the encapsulated bacterial culture, increasing its viability and protecting against environmental adversities, such as low pH, for example. The aim of the present study was to evaluate both in vitro and in vivo, the capacity of the encapsulation process to maintain viable lactic acid bacteria (LAB) strains for a longer period of time and to verify if they are able to reach further regions of mouse intestine. For this purpose, a recombinant strain of LAB (L. lactis ssp. cremoris MG1363) carrying the pExu vector encoding the fluorescence protein mCherry [L. lactis MG1363 (pExu:mCherry)] was constructed. The pExu was designed by our group and acts as a vector for DNA vaccines, enabling the host cell to produce the protein of interest. The functionality of the pExu:mCherry vector, was demonstrated in vitro by fluorescence microscopy and flow cytometry after transfection of eukaryotic cells. After this confirmation, the recombinant strain was submitted to encapsulation protocol with sodium alginate (1%). Non-encapsulated, as well as encapsulated strains were orally administered to C57BL/6 mice and the expression of mCherry protein was evaluated at different times (0–168 h) in different bowel portions. Confocal microscopy showed that the expression of mCherry was higher in animals who received the encapsulated strain in all portions of intestine analyzed. These results were confirmed by qRT-PCR assay. Therefore, this is the first study comparing encapsulated and non-encapsulated L. lactis bacteria for mucosal DNA delivery applications. Our results showed that the microencapsulation process is an effective method to improve DNA delivery, ensuring a greater number of viable bacteria are able to reach different sections of the bowel.
