Search

Helping student service members and veterans (SSM/Vs) earn a college degree is central to supporting them post-service. Yet, this generation of SSM/Vs faces challenges in higher education, including problems adapting, poor health, and administrative constraints, contributing to worsened academic outcomes and a sense of isolation on campus. This monograph synthesizes research on the challenges facing SSM/Vs. It also frames common aspects of successful programs aiding SSM/Vs as three areas for intervention: how administrators and faculty can create a welcoming campus for SSM/Vs; ways universities can create support systems for SSM/V social, health, and academic success; and engaging community partnerships to enable these efforts. Whether SSM/Vs overcome their challenges and unlock their strengths is contingent on opportunities provided within the school itself, and by its faculty, administrators, students, and community. Central to these efforts should be the goal of inclusivity enhancement, community building, and reduction of health-related dysfunction among SSM/Vs.

Abstract Cereus jamacaru DC. occurs in the central-eastern region of Brazil and is among the best-known cacti in the country. Its phytochemical properties and fruits hold great significance to the local communities. We examined published ethnobotanical studies regarding C. jamacaru (mandacaru) undertaken between 2000 to 2024, to better understand its traditional uses and relationships with human communities in Brazil. The literature review was carried out by consulting the databases Google Scholar (166 articles), Scopus (1 article) and Web of Science (2 articles), and followed the PRISMA 2020 protocol. A total of 169 articles were identified, of which 45 were deemed relevant. The most frequent uses of C. jamacaru include medicinal (cited in 32 articles), human consumption as a non-conventional food (22), and forage (18). C. jamacaru is often cited to treat health conditions such as kidney problems, flu and inflammations, but it’s also used for construction and crafts. Our results highlight the diverse and vital uses of C. jamacaru for human communities. Furthermore, we demonstrate the pattern of knowledge production regarding the ethnobotany of this species and the challenges for its conservation and the preservation of associated traditional knowledge.

Background:. In recent years, the aesthetic market has become more prominent for being not only a tool to correct unwanted features but also a social means for improving quality of life. The use of 30% polymethylmethacrylate (PMMA) as a gluteal filler has grown in popularity all over the world, being sought after by men and women for body remodeling because it provides a natural look as well as significant long-lasting results. Methods:. A retrospective, multicenter study analyzed the medical records of 2801 patients who underwent a total of 4725 gluteal filler procedures between January 2009 and December 2021 at the different locations of Clínica Leger. The study protocol was approved by the Brazilian National Research Ethics Commission. Results:. The medical records of 2801 patients (2666 women and 135 men) who underwent 4725 gluteal filler injections were retrospectively analyzed. A total of 922,776 mL of PMMA was used (average = 329 mL/patient). The occurrence of 101 adverse events (2.1%) was observed, and no statistically significant relationship between the mean injected volume and the incidence of complications could be found. Conclusions:. According to the data presented in this study, the use of PMMA injections with the appropriate volumes for aesthetic treatments and corrections is safe and effective.

Abstract Species of the genus Podocarpus L’Hér. ex Pers.present biological activities, such as analgesic, antioxidant, antifungal, acting in the fight against anemia, depurative and fortifying. Podocarpus lambertii Klotzch ex Endl. is a Brazilian native species popularly known as maritime pine and lacks information about its phytochemical profile and possible biological activities. The study was conducted to determine the phytochemical composition of soluble plant extracts of acetone (EA), ethyl acetate (EAE) and hexane (HE) from leaves of P. lambertii; evaluate the antimicrobial potential by the broth microdilution technique; antioxidant potential by the DPPH method, as well as to evaluate the biofilm inhibition capacity by the crystal violet assay and reduction of the yellow tetrazolium salt (MTT). Phytochemical screening detected the presence of flavonoids, triterpenoids, steroids, tannins, alkaloids and saponins. All extracts showed antimicrobial activity on the microorganisms tested, and the EA showed the best results. High free radical scavenging potential was observed only in EAE (96.35%). The antibiofilm potential was observed in the EAE extract. The results contribute to the knowledge of the species and indicate the potential of P. lambertii extracts as a source of plant bioactives for the development of new alternative strategies to control resistant microorganisms.

IntroductionThe inflammatory response after spinal cord injury (SCI) is an important contributor to secondary damage. Infiltrating macrophages can acquire a spectrum of activation states, however, the microenvironment at the SCI site favors macrophage polarization into a pro-inflammatory phenotype, which is one of the reasons why macrophage transplantation has failed.MethodsIn this study, we investigated the therapeutic potential of the macrophage secretome for SCI recovery. We investigated the effect of the secretome in vitro using peripheral and CNS-derived neurons and human neural stem cells. Moreover, we perform a pre-clinical trial using a SCI compression mice model and analyzed the recovery of motor, sensory and autonomic functions. Instead of transplanting the cells, we injected the paracrine factors and extracellular vesicles that they secrete, avoiding the loss of the phenotype of the transplanted cells due to local environmental cues.ResultsWe demonstrated that different macrophage phenotypes have a distinct effect on neuronal growth and survival, namely, the alternative activation with IL-10 and TGF-β1 (M(IL-10+TGF-β1)) promotes significant axonal regeneration. We also observed that systemic injection of soluble factors and extracellular vesicles derived from M(IL-10+TGF-β1) macrophages promotes significant functional recovery after compressive SCI and leads to higher survival of spinal cord neurons. Additionally, the M(IL-10+TGF-β1) secretome supported the recovery of bladder function and decreased microglial activation, astrogliosis and fibrotic scar in the spinal cord. Proteomic analysis of the M(IL-10+TGF-β1)-derived secretome identified clusters of proteins involved in axon extension, dendritic spine maintenance, cell polarity establishment, and regulation of astrocytic activation.DiscussionOverall, our results demonstrated that macrophages-derived soluble factors and extracellular vesicles might be a promising therapy for SCI with possible clinical applications.

The capuchin monkey (Sapajus apella), a New World monkey species, exhibits prominent characteristics that make it an ideal model for neuroscience research. These characteristics include its phylogenetic traits, telencephalization coefficient, anatomical structures and pathways, genetic profile, immune responses, cognitive abilities, and complex behavioral repertoires. Traditionally, methodologies for stereotactic neurosurgery in research models have relied on the use of brain atlases. However, this approach can lead to errors due to the considerable variation in brain size and shape among individual monkeys. To address this issue, we developed a protocol for deriving individual coordinates for each monkey using a straightforward and relatively inexpensive method involving MRI imaging. Our protocol utilizes a specially designed, 3D-printed stereotactic head-holder that is safe to use with an MR magnet, non-invasive placement of fiducial markers, and post-processing with open-source software. This approach enhances MRI data visualization, improves anatomical targeting, and refines the design of neurosurgical experiments. Our technique could also prove beneficial in other areas of neuroscience research that require accurate calculation of stereotaxic coordinates. Furthermore, it could be useful for other nonhuman primate species for which brain atlases are typically unavailable.

Parkinson’s disease (PD) is recognized as the second most prevalent primary chronic neurodegenerative disorder of the central nervous system. Clinically, PD is characterized as a movement disorder, exhibiting an incidence and mortality rate that is increasing faster than any other neurological condition. In recent years, there has been a growing interest concerning the role of the gut microbiota in the etiology and pathophysiology of PD. The establishment of a brain–gut microbiota axis is now real, with evidence denoting a bidirectional communication between the brain and the gut microbiota through metabolic, immune, neuronal, and endocrine mechanisms and pathways. Among these, the vagus nerve represents the most direct form of communication between the brain and the gut. Given the potential interactions between bacteria and drugs, it has been observed that the therapies for PD can have an impact on the composition of the microbiota. Therefore, in the scope of the present review, we will discuss the current understanding of gut microbiota on PD and whether this may be a new paradigm for treating this devastating disease.

Abstract Background Polyarthritis has been associated with canine visceral leishmaniasis (CanVL), and co-infection with Ehrlichia canis is common and may alter clinical manifestations. Methods A total of 89 dogs presenting CanVL were subdivided into two groups: (1) G1, consisting of 46 dogs seronegative to Ehrlichia spp., and (ii) G2, consisting of 43 dogs seropositive to Ehrlichia spp. Eight joints (carpal, tarsal, stifles and elbows) from each dog were evaluated by radiography and synovial fluid (SF) cytologic analysis. Results Overall, 74 of the 89 (83.1%) dogs presented joint abnormalities suggestive of osteoarthritis by radiography (G1: 40/46 [86.9%]; G2: 34/43 [79.0%]), with no statistically significant between-group difference. All dogs with abnormal joint X-ray images presented radiographic lesions bilaterally, independent of the characteristics of the lesion. Soft tissue swelling around the joint and joint space narrowing were more commonly observed in G1 than in G2 dogs. There was no significant between-group difference in terms of other radiographic abnormalities suggestive of osteoarthritis (evident trabecular pattern, subchondral bone sclerosis, osteolysis, osteolytic–proliferative lesions or bone proliferation). SF from 174/315 (55.2%) and 152/307 (49.5%) joints from G1 and G2 dogs, respectively, presented an inflammatory infiltrate, but there was no significant association between the presence of inflammatory infiltrate and group. There was also no statistical difference between groups in either of the evaluated joints in terms of the percentage of neutrophils or mononuclear cells. Leishmania spp. amastigotes were found in 69/315 (21.9%) joints from G1 dogs and in 100/307 (32.5%) joints from G2 dogs (Fisherʼs exact test, P = 0.002, odds ratio = 0.5, 95% confidence interval = 0.4–0.8). The neutrophilic infiltrate was significantly higher in joints with amastigote forms in both G1 (Mann–Whitney U-test, U (18) = 817, Z = -3.76, P = 0.0001) and G2 dogs (Mann–Whitney U-test, U (18) = 6543, Z = − 5.06, P

ABSTRACT Sandy-textured soils are naturally more vulnerable to the erosion process and their exploitation, although possible, is often performed inappropriately, favoring its degradation. In this context, this study aimed to classify the rainfall erosivity in a region of sandy soils to identify critical situations of soil and water loss and also correlate it with rainfall data to assess whether there is temporal dependence of this variable using geostatistical techniques. The potential for alternative and sustainable production systems to be used in regions with sandy soils was also analyzed. Historical data of precipitation in the study region were analyzed to determine the average monthly and annual erosivity indices, which were classified and its temporal dependence was assessed by applying geostatistics. NDVI data from satellite images were used to investigate the soil cover pattern in different production systems. Geostatistics was adequate for the analysis of rainfall erosivity, which showed moderate to strong temporal dependence. It was classified between strong and very strong and was highly dependent on precipitation, with events of higher erosion potential between October and March in the studied region. The vicious circle of degradation of sandy soils, such as those of the Bolsão region of Mato Grosso do Sul, Brazil, can be modified by adopting alternative and sustainable production systems that value the maximization of soil cover.

Introduction Obsessive-compulsive disorder (OCD) is a psychiatric disorder affecting 1.3% of the population worldwide where both genetic and environmental factors, such as perinatal events and neuroinflammation, are thought to contribute to the etiology of the disorder. In the past, the description of clinical entities such as Pediatric Autoimmune Neuropsychiatric Disorder Associated with Streptococcal Infections (PANDAS), in which an acute neuropsychiatric syndrome with prominent obsessive-compulsive features emerges in children infected with group-A beta-hemolytic streptococcus (GABHS), sparked the hypothesis that infections may be a risk-modifying factor for the development of OCD. Along with streptococcal infections, other pathogens such as Toxoplasma gondii have been implicated in the pathophysiological models of the disorder, although causal associations have not been established for any of beforementioned pathogens. Objectives To perform a systematic review and meta-analysis about the presence of biological evidence of infection in patients diagnosed with OCD. Methods We conducted a systematic review and a meta-analysis (PROSPERO registration CRD42021223415) by performing a standardized electronic database search in MEDLINE/PubMed, Web of Science, Embase and Scopus. Search was conducted on 17/10/2022. Eligible papers included case-control and cohort studies using a comparator group, that tested for specific biomarkers providing evidence of infection in patients diagnosed with OCD; exclusion criteria included studies without quantitative or qualitative measures of infection, case reports, systematic or scope reviews, and animal studies. Selection process was conducted according to PRISMA 2020 statement guidelines. Study quality was assessed through Newcastle-Ottawa Quality Assessment Scale. Results We identified 8911 records through the search after duplicate removal. A total of 22 studies met inclusion criteria after selection process, and 15 were eligible for meta-analysis. Most evidence concerned Toxoplasma gondii (10 studies), and patients with OCD appear to have higher odds of being infected compared to controls, with a meta-analytic odds ratio of 2.39 (95% IC 1.60-3.58), when comparing 467 patients with 5411 controls. However, most studies were methodologically heterogeneous, which compromises the interpretation of meta-analytic results. Information regarding other agents, including GABHS, Borna disease virus and Toxocara canis was gathered but due to an insufficient number of papers it was not possible to perform a meta-analysis for each of them. Conclusions Our work suggests that albeit exhaustively reported in the literature, there is no strong evidence of the over-representation of biomarkers of infection in patients with OCD compared to control volunteers. Methodologically robust studies are needed to further test this hypothesis. Disclosure of Interest None Declared

ABSTRACT CROPWAT is a model that uses water balance to study the water factor and productivity in production systems. In this sense, this study aimed to calibrate the CROPWAT for modeling rainfed and irrigated soybean production systems. Climate and soil data from a soybean-producing region in the south of Mato Grosso do Sul, Brazil, were used to implementing simulations in nine agricultural years, and crop shortfalls were calculated and compared with reference data. Statistical indices were applied to evaluate the performance of the model in its calibration. For validation, two field trials with 12 cultivars were implemented under irrigation and rainfed and the mean productivity in each water management was compared with the CROPWAT estimate by Student’s t-test. Accuracy (r) was 0.976 (“very strong”), precision (r2) was equivalent to 95.3%, and the indices of agreement (d) and performance (c) were considered excellent (0.95 and 0.93, respectively) in the calibration. Validation demonstrated that the hypothesis that CROPWAT correctly estimated soybean productivity under irrigated and rainfed systems cannot be rejected at 1 or 5% significance levels.

Objectives: The majority of available scores to assess mortality risk of coronavirus disease 2019 (COVID-19) patients in the emergency department have high risk of bias. Therefore, this cohort aimed to develop and validate a score at hospital admission for predicting in-hospital mortality in COVID-19 patients and to compare this score with other existing ones. Methods: Consecutive patients (≥ 18 years) with confirmed COVID-19 admitted to the participating hospitals were included. Logistic regression analysis was performed to develop a prediction model for in-hospital mortality, based on the 3978 patients admitted between March–July, 2020. The model was validated in the 1054 patients admitted during August–September, as well as in an external cohort of 474 Spanish patients. Results: Median (25–75th percentile) age of the model-derivation cohort was 60 (48–72) years, and in-hospital mortality was 20.3%. The validation cohorts had similar age distribution and in-hospital mortality. Seven significant variables were included in the risk score: age, blood urea nitrogen, number of comorbidities, C-reactive protein, SpO2/FiO2 ratio, platelet count, and heart rate. The model had high discriminatory value (AUROC 0.844, 95% CI 0.829–0.859), which was confirmed in the Brazilian (0.859 [95% CI 0.833–0.885]) and Spanish (0.894 [95% CI 0.870–0.919]) validation cohorts, and displayed better discrimination ability than other existing scores. It is implemented in a freely available online risk calculator (https://abc2sph.com/). Conclusions: An easy-to-use rapid scoring system based on characteristics of COVID-19 patients commonly available at hospital presentation was designed and validated for early stratification of in-hospital mortality risk of patients with COVID-19.

Category: Arthroscopy; Midfoot/Forefoot Introduction/Purpose: Hallux rigidus is a common pathological condition of the foot, responsible for a great functional limitation. Metatarsophalangeal arthrodesis is an excellent therapeutic alternative for advanced stages of the disease, usually performed using an open technique. Forefoot arthroscopy has been developed in the last decade, with progressive expansion of indications, bringing encouraging results. Our objective is present the clinical-radiographic results of patients undergoing arthroscopic metatarsophalangeal arthrodesis of the hallux, showing the technique and reporting complications. Methods: This study involves a series of 9 patients (10 feet), all with advanced hallux rigidus (Coughlin-Shurnas grades 3 and 4). All patients underwent an arthroscopic procedure through two dorsal portals with small-joint instruments. Outcomes were assessed using the AOFAS functional score and a visual analogue scale of pain. The radiographic parameters of interest were metatarsal-phalangeal angulation in the anteroposterior (pre- and postoperative) and lateral (post-operative) views, as well as ray shortening. Results: All patients evolved with consolidation in radiographic control at 6 weeks postoperatively and showed a marked improvement in functional scores. We observed a reduction in the metatarsophalangeal angle, without shortening of the radius and of the finger. No patient had transfer metatarsalgia or complaints of cosmetic problems. Conclusion: Arthroscopic metatarsophalangeal arthrodesis is a very relevant therapeutic option for managing advanced stages of hallux rigidus, with minimal aggression and providing excellent results, in addition to a low incidence of complications.

The treatment can be conservative or surgical; the latter is better indicated in high-performance athletes. The pursue of faster recovery, reductions in skin complications and re-ruptures have brought about improvements to the tendon repair technique and to early assisted rehabilitation programs. Our goal was to check whether or not the adjuvant use of 2-octyl cyanoacrylate provides additional mechanical resistance to the calcaneal tenorraphy, when compared to suture alone, using the same tenorraphy technique. Methods: We used 45 three-month-old New Zealand rabbits, distributed into three groups of 15 animals each. The rabbits underwent two surgical procedures at a 30-day interval. In the first procedure, we performed the complete calcaneal tendon section on the right side, and then we repaired it using three different techniques: suture alone in group 1, suture with 2-octyl cyanoacrylate in group 2, and 2-octyl cyanoacrylate alone in group 3. After 30 days, we slaughtered the rabbits, and resected their right-side calcaneal tendon, and sent the specimen for pathology analysis. Then, we performed a cross section of the left calcaneal tendon, and used the same three repair techniques. After 15 minutes, we subjected the repaired tendons to the linear traction mechanical strength test. Abstract Spinal cord injury (SCI) leads to dramatic impairments of motor, sensory, and autonomic functions of affected individuals. Following the primary injury, there is an increased release of glutamate that leads to excitotoxicity and further neuronal death. Therefore, modulating glutamate excitotoxicity seems to be a promising target to promote neuroprotection during the acute phase of the injury. In this study, we evaluated the therapeutic effect of a FDA approved antiepileptic drug (levetiracetam-LEV), known for binding to the synaptic vesicle protein SV2A in the brain and spinal cord. LEV therapy was tested in two models of SCI—one affecting the cervical and other the thoracic level of the spinal cord. The treatment was effective on both SCI models. Treated animals presented significant improvements on gross and fine motor functions. The histological assessment revealed a significant decrease of cavity size, as well as higher neuronal and oligodendrocyte survival on treated animals. Molecular analysis revealed that LEV acts by stabilizing the astrocytes allowing an effective uptake of the excess glutamate from the extracellular space. Overall, our results demonstrate that Levetiracetam may be a promising drug for acute management of SCI.

The failure of axons to regenerate after a spinal cord injury (SCI) remains one of the greatest challenges in neuroscience. The initial mechanical trauma is followed by a secondary injury cascade, creating a hostile microenvironment, which not only is not permissive to regeneration but also leads to further damage. One of the most promising approaches for promoting axonal regeneration is to maintain the levels of cyclic adenosine monophosphate (cAMP), specifically by a phosphodiesterase-4 (PDE4) inhibitor expressed in neural tissues. Therefore, in our study, we evaluated the therapeutic effect of an FDA-approved PDE4 inhibitor, Roflumilast (Rof), in a thoracic contusion rat model. Results indicate that the treatment was effective in promoting functional recovery. Rof-treated animals showed improvements in both gross and fine motor function. Eight weeks post-injury, the animals significantly recovered by achieving occasional weight-supported plantar steps. Histological assessment revealed a significant decrease in cavity size, less reactive microglia, as well as higher axonal regeneration in treated animals. Molecular analysis revealed that IL-10 and IL-13 levels, as well as VEGF, were increased in the serum of Rof-treated animals. Overall, Roflumilast promotes functional recovery and supports neuroregeneration in a severe thoracic contusion injury model and may be important in SCI treatment.

Background: Parkinson's disease (PD) is a neurodegenerative disease characterized by intracellular inclusions named Lewy bodies (LB), and alpha-synuclein (asyn) is the major component of these protein aggregates. The precise physiological and pathological roles of asyn are not fully understood. Nevertheless, asyn present in LB is ubiquitinated but fails to reach the 26S proteasome. The mutation A30 P is related to an aggressive and early-onset form of PD. Tumor necrosis factor receptor-associated factor 6 (TRAF6) is an E3 ubiquitin ligase, and it interacts and ubiquitinates the asyn in atypical chains (lysine K6, K27, K29, and K33). Methods: Here, we investigated the role of TRAF6 interaction with asyn and the involvement of nuclear factor κB (NF-κB), a key transcription factor in pro-inflammatory signaling pathway activation. Results and Conclusion: We demonstrated that TRAF6 binds to both WT and the mutant form A30 P asyn in an SH-SY5Y cell model. Additionally, the interaction between TRAF6 and WT asyn induced an increase in the activation of NF-κB, leading to changes in TNF, IL-1β and IL-10 levels and culminating in reduced cell viability. Interestingly, the activation of NF-κB and gene regulation were not found in A30 P asyn. These data point to a novel role of TRAF6 in the pathophysiology of PD.

Abstract Background Aedes aegypti and Culex quinquefasciatus are the main urban vectors of arthropod-borne viruses causing human disease, including dengue, Zika, or West Nile. Although key to disease prevention, urban-mosquito control has met only limited success. Alternative vector-control tactics are therefore being developed and tested, often using entomological endpoints to measure impact. Here, we test one promising alternative and assess how three such endpoints perform at measuring its effects. Methods We conducted a 16-month, two-arm, cluster-randomized controlled trial (CRCT) of mosquito-disseminated pyriproxyfen (MD-PPF) in central-western Brazil. We used three entomological endpoints: adult-mosquito density as directly measured by active aspiration of adult mosquitoes, and egg-trap-based indices of female Aedes presence (proportion of positive egg-traps) and possibly abundance (number of eggs per egg-trap). Using generalized linear mixed models, we estimated MD-PPF effects on these endpoints while accounting for the non-independence of repeated observations and for intervention-unrelated sources of spatial-temporal variation. Results On average, MD-PPF reduced adult-mosquito density by 66.3% (95% confidence interval, 95% CI: 47.3–78.4%); Cx. quinquefasciatus density fell by 55.5% (95% CI: 21.1–74.8%), and Ae. aegypti density by 60.0% (95% CI: 28.7–77.5%). In contrast, MD-PPF had no measurable effect on either Aedes egg counts or egg-trap positivity, both of which decreased somewhat in the intervention cluster but also in the control cluster. Egg-trap data, therefore, failed to reflect the 60.0% mean reduction of adult Aedes density associated with MD-PPF deployment. Conclusions Our results suggest that the widely used egg-trap-based monitoring may poorly measure the impact of Aedes control; even if more costly, direct monitoring of the adult mosquito population is likely to provide a much more realistic and informative picture of intervention effects. In our CRCT, MD-PPF reduced adult-mosquito density by 66.3% in a medium-sized, spatially non-isolated, tropical urban neighborhood. Broader-scale trials will be necessary to measure MD-PPF impact on arboviral-disease transmission.

Major depressive disorder (MDD) is a multidimensional psychiatric disorder that is estimated to affect around 350 million people worldwide. Generating valid and effective animal models of depression is critical and has been challenging for neuroscience researchers. For preclinical studies, models based on stress exposure, such as unpredictable chronic mild stress (uCMS), are amongst the most reliable and used, despite presenting concerns related to the standardization of protocols and time consumption for operators. To overcome these issues, we developed an automated system to expose rodents to a standard uCMS protocol. Here, we compared manual (uCMS) and automated (auCMS) stress-exposure protocols. The data shows that the impact of the uCMS exposure by both methods was similar in terms of behavioral (cognition, mood, and anxiety) and physiological (cell proliferation and endocrine variations) measurements. Given the advantages of time and standardization, this automated method represents a step forward in this field of preclinical research.

Parkinson’s disease (PD) is the second most prevalent neurodegenerative disorder worldwide. Clinically, it is characterized by a progressive degeneration of dopaminergic neurons (DAn), resulting in severe motor complications. Preclinical and clinical studies have indicated that neuroinflammation can play a role in PD pathophysiology, being associated with its onset and progression. Nevertheless, several key points concerning the neuroinflammatory process in PD remain to be answered. Bearing this in mind, in the present review, we cover the impact of neuroinflammation on PD by exploring the role of inflammatory cells (i.e., microglia and astrocytes) and the interconnections between the brain and the peripheral system. Furthermore, we discuss both the innate and adaptive immune responses regarding PD pathology and explore the gut–brain axis communication and its influence on the progression of the disease.

The gonadal development of the characin Astyanax aff. bimaculatus from Funil Reservoir, an impoundment in the middle reaches of the Paraiba do Sul River in southeastern Brazil was analysed through histological, histochemical and histometric techniques, and the spawning season was determined. Unbalanced sex ratio was found, with females outnumbering and reaching larger sizes than males. The following stages of oogenic cell development were identified for females: primary growth, pre-vitellogenic (cortical alveolar) and vitellogenic. In males, the following stages were determined: spermatogonia primary and secondary, spermatocyte primary and secondary, spermatid and spermatozoa. The spawning occurs most of the year, peaking in January-February, as indicated by the gonadosomatic index. In the peak of the spawning period, the condition factor and hepatosomatic indices decreased suggesting use of stored body energy reserves during the reproduction. Spawning in batches, small-sized oocytes and a long reproductive season suggest that Astyanax aff. bimaculatus fits to an opportunist strategy. Such strategy may facilitate the use of a wide range of lentic and lotic body waters in the Neotropical region.

Understanding local stakeholders' perception and their relation with the landscape and its natural resources is an important step for successfully implementing Forest Landscape Restoration (FLR). Here, we present a case study on FLR in the context of a global biodiversity hotspot—the Brazilian Atlantic Forest, using a participatory approach to include local stakeholders' knowledge and perception of the landscape into project planning. We analyzed the land use and cover, and organized a workshop with focus group methodology associated with maps and other visual representations to assess local perceptions of economic activities, production chains and their impacts on the landscape and ecosystem services. The study area encompasses seven municipalities mainly covered by native vegetation and pastures. Despite pastureland being the prominent land use in the region, they are not engaged in associations, most do not live in the region, and few participated in the workshop. Most participants were small and medium-scale landowners involved in agricultural activities who demonstrated a detailed knowledge of the territory, a disposition toward combining conservation with production practices, and a positive perception regarding ecotourism, agroecological approaches, water, and soil conservation. The participatory approach proved effective to complement the initial assessment while revealing novel aspects of the landscape and the landowners, helping test our hypotheses and adjust the engaging narratives for future FLR activities planning in the region, including environmental law compliance. More studies associating social and natural science, including participatory methods and local communities' perception, are needed to fully comprehend the drivers of stakeholders' engagement. This case study provides useful insights for other researchers and practitioners to design more effective plans for future land management.

The reproductive physiology of fish can be changed by the presence of pollutants in the water, which act as endocrine disrupting compounds (EDC). We evaluated the impacts of water contaminants in polluted reservoirs acting as possible EDC on the reproductive physiology of Astyanax fasciatus and Hoplias malabaricus males. We used biomarkers with different levels of biological organization. Hoplias malabaricus adult males were collected in the summer and winter at five different sites in the Tietê River Basin: the Ponte Nova reservoir (PN), considered a reference site due to the low anthropogenic influence; the Billings reservoir (BIL) at two different branches; and the Guarapiranga reservoir (GUA) at two different branches. Astyanax fasciatus adult males were collected at PN and BIL. BIL and GUA are subjected to great anthropogenic action. We analyzed gonadal histomorphology, testosterone (T), 11-ketotestosterone (11-KT), estradiol (E2) plasma levels, and gene expression of hepatic vitellogenin (vtgA) and pituitary follicle stimulating hormone (fshβ). In the PN reservoir (reference), the biomarkers analyzed in both species did not differ between the periods analyzed. This is an evidence that the animals keep the same reproductive activity during both seasons. The changes in the plasma concentration of gonadal steroids in both species in polluted reservoirs suggest the presence of EDC compounds in the water and/or adjusts of the physiological setpoint to allow the reproduction in such adverse conditions. The use of vtgA as biomarker suggests the presence of estrogenic compounds, mainly in BIL, but with a more evident response of H. malabaricus. However, even considering physiological changes, both species present testes during the maturation phase that allow the reproduction in an environment with a high degree of pollution.

Cervical spinal cord trauma represents more than half of the spinal cord injury (SCI) cases worldwide. Respiratory compromise, as well as severe limb motor deficits, are among the main consequences of cervical lesions. In the present work, a Gellan Gum (GG)-based hydrogel modified with GRGDS peptide, together with adipose tissue-derived stem/stromal cells (ASCs) and olfactory ensheathing cells (OECs), was used as a therapeutic strategy after a C2 hemisection SCI in rats. Hydrogel or cells alone, and a group without treatment, were also tested. Four weeks after injury, compound muscle action potentials (CMAPs) were performed to assess functional phrenic motor neuron (PhMN) innervation of the diaphragm; no differences were observed amongst groups, confirming that the PhMN pool located between C3 and C5 was not affected by the C2 injury or by the treatments. In the same line, the vast majority of diaphragmatic neuromuscular junctions remained intact. Five weeks post-injury, inspiratory bursting of the affected ipsilateral hemidiaphragm was evaluated through EMG recordings of dorsal, medial and ventral subregions of the muscle. All treatments significantly increased EMG amplitude at the ventral portion in comparison to untreated animals, but only the combinatorial group presented increased EMG amplitude at the medial portion of the hemidiaphragm. No differences were observed in forelimb motor function, neither in markers for axonal regrowth (neuronal tracers), astrogliosis (GFAP) and inflammatory cells (CD68). Moreover, using Von Frey testing of mechanical allodynia, it was possible to find a significant effect of the group combining hydrogel and cells on hypersensitivity; rats with a SCI displayed an increased response of the contralateral forelimb to a normally innocuous mechanical stimulus, but after treatment with the combinatorial therapy this behavior was reverted almost to the levels of uninjured controls. These results suggest that our therapeutic approach may have beneficial effects on both diaphragmatic recovery and sensory function.

Vascular disruption following spinal cord injury (SCI) decisively contributes to the poor functional recovery prognosis facing patients with the condition. Using a previously developed gellan gum hydrogel to which the adhesion motif GRGDS was grafted (GG-GRGDS), this work aimed to understand the ability of adipose-derived stem cells (ASCs) to impact vascular organization of human umbilical vein endothelial cells (HUVECs), and how this in turn affects neurite outgrowth of dorsal root ganglia (DRG) explants. Our data shows that culturing these cells together lead to a synergistic effect as showed by increased stimulation of neuritogenesis on DRG. Importantly, HUVECs were only able to assemble into vascular-like structures when cultured in the presence of ASCs, which shows the capacity of these cells in reorganizing the vascular milieu. Analysis of selected neuroregulatory molecules showed that the co-culture upregulated the secretion of several neurotrophic factors. On the other hand, ASCs, and ASCs + HUVECs presented a similar profile regarding the presence of angiotrophic molecules herein analyzed. Finally, the implantation of GG-GRGDS hydrogels encapsulating ASCs in the chick chorioallantoic membrane (CAM) lead to increases in vascular recruitment toward the hydrogels in comparison to GG-GRGDS alone. This indicates that the combination of ASCs with GG-GRGDS hydrogels could promote re-vascularization in trauma-related injuries in the central nervous system and thus control disease progression and induce functional recovery.

ABSTRACT The hydrogen is a fuel with a high energy density. Studies have sought to verify the potential of different residues and configurations of reactors in the production of biohydrogen. This study evaluated the influence of the organic loading rate on the production of biohydrogen from cassava starch wastewater (CSW) in anaerobic sequencing batch biofilm reactors (AnSBBR) operated in a fed-batch. In the tests, the organic loading rates of 9.0; 13.5 and 18.0 gCT L−1 d−1 were used, at cycle times (CT) of 4; 4 and 2 h (tests I, II and III, respectively). The reactor was inoculated with sludge from a pilot anaerobic reactor used in the treatment of CSW, heat treated (heated at 90°C for 15 min). With the increase of the organic load (condition I to II) with the same cycle time (CT: 4 h), there were increases in the hydrogen molar productivity (PrM) and hydrogen yield in relation to applied total carbohydrate (RMCA) and removed (RMCR). However, with the increase in the organic loading rate in which the CT was reduced from 4 to 2 h (condition II to III), there was a decrease in PrM and RMCA and RMCR. The best performance of the reactor was verified in condition II (organic loading rate: 13.5 gTC L− 1 d−1; CT: 4 h), in which the PrM was 35.8 molH2 m−3 d−1 and RMCA and RMCR were 2.24 and 3.67 molH2 kgTC−1, respectively.

ABSTRACT The hydrodynamic behavior of an anaerobic fixed bed reactor (AFBR) was evaluated in the treatment of cattle slaughterhouse wastewater. The AFBR was operated at hydraulic retention time (HRT) of 14, 11 and 8 h. Stimulus-response assays were carried out with Eosin Y and the experimental data were adjusted to the single-parameter theoretical models of dispersion and N-continuous stirred tank reactors in series (N-CSTR). The experimental results of the residence time distribution curves showed that at lower flow rate, the reactor showed plug flow behavior with correlation coefficient (r) of 0.88 and number of dispersion of 0.2 for high dispersion (HD). However, at higher and intermediate flow rates, the AFBR behave as a complete mixture flow, (r) of 0.94 and 0.96, respectively. Residence time distribution curves in the AFBR showed a good approximation of the complete mixing model at hydraulic residence time of 11 and 8 h, with 5 and 2 N-CSTR reactors in series, respectively. The volume of dead zones corresponding to 43.0, 37.4 and 11.2% of the volume of the reactor for HRT of 14, 11 and 8 h, respectively, was noted, and hydraulic short circuiting were not confirmed.

ABSTRACT The aim of this study was to evaluate the behavior of an anaerobic reactor of packed-bed and continuous flow, in relation to the biogas and methane production and the removal of organic load, from the wastewater of cassava starch extraction. For this purpose, were tested, in a reactor with a useful volume of 2.82 L and support mean of polypropylene filled with polyethylene foam, the organic loading rate (OLR) of 2.5, 5.0, 8.0 and 10.0 g L-1d-1. It was evaluated the removal of COD, the pH, the VA/TA ratio, the biogas and the methane production. It was concluded from this study that it is possible to produce biogas with methane percentages higher than 80% (maximum) and remove the organic load more than 90% under the tested conditions. In relation to stability, the reactor can be considered stable for the parameters, pH, the relationship between volatile acidity and total alkalinity and organic load removal. The best results in relation to the average volumetric biogas production (0.174 and 0.311 L g COD-1) were obtained for the higher organic loading rate (of 8 and 10 g L-1d-1, respectively) and the observed tendency is that with the increase of the organic loading rate, the specific biogas production will increase as well as the specific production of methane.

Abstract Background Glioblastoma (GBM) is a highly aggressive primary brain cancer, for which curative therapies are not available. An emerging therapeutic approach suggested to have potential to target malignant gliomas has been based on the use of multipotent mesenchymal stem cells (MSCs), either unmodified or engineered to deliver anticancer therapeutic agents, as these cells present an intrinsic capacity to migrate towards malignant tumors. Nevertheless, it is still controversial whether this innate tropism of MSCs towards the tumor area is associated with cancer promotion or suppression. Considering that one of the major mechanisms by which MSCs interact with and modulate tumor cells is via secreted factors, we studied how the secretome of MSCs modulates critical hallmark features of GBM cells. Methods The effect of conditioned media (CM) from human umbilical cord perivascular cells (HUCPVCs, a MSC population present in the Wharton’s jelly of the umbilical cord) on GBM cell viability, migration, proliferation and sensitivity to temozolomide treatment of U251 and SNB-19 GBM cells was evaluated. The in vivo chicken chorioallantoic membrane (CAM) assay was used to evaluate the effect of HUCPVCs CM on tumor growth and angiogenesis. The secretome of HUCPVCs was characterized by proteomic analyses. Results We found that both tested GBM cell lines exposed to HUCPVCs CM presented significantly higher cellular viability, proliferation and migration. In contrast, resistance of GBM cells to temozolomide chemotherapy was not significantly affected by HUCPVCs CM. In the in vivo CAM assay, CM from HUCPVCs promoted U251 and SNB-19 tumor cells growth. Proteomic analysis to characterize the secretome of HUCPVCs identified several proteins involved in promotion of cell survival, proliferation and migration, revealing novel putative molecular mediators for the effects observed in GBM cells exposed to HUCPVCs CM. Conclusions These findings provide novel insights to better understand the interplay between GBM cells and MSCs, raising awareness to potential safety issues regarding the use of MSCs as stem-cell based therapies for GBM.

ABSTRACT The anaerobic digestion (AD) operation complexity raise the importance of stability testing to verify whether the operating conditions are under control and whether the process is working as required. The current method of verifying process stability is by a range of the ratio between volatile fatty acidity and total alkalinity (VFA/ TA) within which stable conditions are brought about; these rates vary with the content of organic material. A few indicators as pH or constant biogas production are also used. Therefore, the aim of this study was to propose the use of Shewhart control chart for individual measures as a stability test for AD, as well as to demonstrate its use in poultry litter AD. The method showed to be advantageous since it standardizes a variation range for the process according to the average and verifies the stability by direct measures, such as organic material removal efficiency and biogas production.

ABSTRACT The aim of this study was to monitor the stabilization of a sequential batch reactor (SBR) regarding the partial nitritation of poultry slaughterhouse wastewater, aiming at preparing it for a reactor with bacteria that promote anaerobic ammonia oxidation (anammox). A cylindrical reactor with a useful volume of 3.5 L, mechanical agitation (50 rpm), and suspended biomass was used in the experiment. The strategies adopted for nitrite-oxidizing bacteria (NOB) inhibition were cycle time (CT: 24, 16, 6, 5, 4 h and real-time monitoring), free ammonia accumulation, alkalinity restriction and low concentrations of dissolved oxygen (DO). The ammonification process, observed at all experimental phases, negatively influenced the control of partial nitritation process. Alkalinity restriction under low cycle times (4 and 5 h) conditions, together with low dissolved oxygen concentration, was a key factor for nitrite-oxidizing bacteria inhibition. Stability of partial nitritation was only reached by real-time monitoring, being the cycles stopped when 50% N–NH4+ and 50% N–NO2− were present in the reactor.

ABSTRACT Fixed-bed reactors have been considered promise alternatives for hydrogen production due to their simple construction and increase in the biomass retention. The purpose of this study was to investigate the biological production of hydrogen in anaerobic fixed-bed reactors with cassava starch wastewater used as substrate. Different support materials and arrangements of fixed-bed were used to evaluate the biological production of hydrogen in anaerobic continuous fixed-bed reactors, with cassava starch wastewater as substrate - recycled low-density polyethylene scraps, in packed bed (R1), recycled low-density polyethylene cylinders, in ordained bed (R3) and bamboo stems, in vertical arrangement (R2 and R4). In R1 the initial pH was adjusted for 6.0, with hydraulic retention time (HRT) of 4 h and organic loading rate (OLR) of 9.5 g.L−1.d−1. In R2 the initial pH was maintained in 4.5, with HRT of 4 h and OLR of 9.5 g.L−1.d−1. R3 and R4 were operated with initial pH of 4.5, HRT of 4 h and OLR of 13.5 g.L−1.d−1. The volumetric hydrogen production (VHPR) was favored by lower OLR applied, evenin different pH ranges (6.0 and 4.5). VHPR values of 229 mL H2.L−1.d−1and 248 mL H2.L−1.d−1 were obtained in R1 and R2, respectively. Both in the bamboo stems bed as in the polyethylene cylinders bed, the increase of OLR and the reduction of the initial pH resulted in a diminishing of VHPR to 175 mL H2.d−1.L−1 (R3) and 145 m LH2 .d−1.L−1 (R4). Higher concentrations of butanol (821.32 and 1,529.22 mg.L−1) and ethanol (915.41 and 924.41 mg.L−1) were verified in the reactors with bamboo stems. In R4, the increase of OLR and the reduction of the initial pH contributed to the increase of butanol concentration in 1.8 times, diminishing the VHPR in 41.68%, yield in 63.95% and H2 in 37.6%, and indicating that the effect of pH is more pronounced with the increase of OLR, leading to the solventogenesis.

The process of cassava starch extraction is associated with the generation of a large volume of liquid wastes from root washing and starch extraction (5 to 7 L.kg−1root). In this context, the aim of this study was to evaluate the treatment of cassava starch wastewater in a horizontal anaerobic fixed bed reactor to the removal of organic matter and generation of biogas. For this purpose, two fixed bed reactors filled with different types of support materials were used: bamboo rings (R1) and flexible PVC rings (R2). The reactors were constructed in polyvinyl chloride (PVC) with 90 cm length and 15 cm diameter. To evaluate the reactors were carried out 13 assays (A1 to A13) with increases in the organic loading rate (OLR, 1.7 to 15.0 g.L−1.d−1) and a decrease in the hydraulic retention time (HRT, 4.0 to 0.8 days). Biogas production followed a similar pattern in both reactors. The highest biogas productions of 1.4 L.L−1.d−1 (R1) and 1.0 L.L−1.d−1 (R2) were verified in the assay A6 in which were applied highest influent concentration (15.1g.L−1), intermediates OLR (5.6 g.L−1.d−1) and HRT (2.7 d). The chemical oxygen demand (COD) removal increased with the OLR increase resulting in COD removal values of up to 99%. Total solids removal efficiencies of 86.2 and 85.5% were achieved in R1 and R2, respectively. In both reactors, the contact surface of bamboo (132 m2.m−3) and PVC (191 m2.m−3) supported biomass attachment even in the highest OLR (15 g.L−1.d−1) and low HRT (0.8 d). Both support material provided conditions to the reactors resist the variations in operating conditions and reestablish the equilibrium after possible hydraulic and organic shocks load, constituting a robust system for the treatment of cassava starch wastewater. In these conditions the COD and solids removal remained satisfactory. After a 450-days reactor running, no changes were observed in the composition and structure of the support material, indicating that bamboo and PVC are possible cheap and efficient alternatives for biomass immobilization.

Citalopram is a selective serotonin reuptake inhibitor, and although widely used as an antidepressant, this drug has also demonstrated interesting repairing properties leading to motor recovery and pathology amelioration in animal models of stroke and degeneration. Here, we tested the efficacy of both 7-day and 8-week citalopram treatment in a contusive spinal cord injury (SCI) rat model. A combination of behavioral tests, histological and serum cytokine analysis was used to assess overall recovery. Despite promoting a mild reduction of inflammatory cells as well as an early, but transient increase of specific serum cytokines, citalopram administration showed no overall beneficial effects on motor performance or lesion extension. Our results do not support citalopram treatment as a therapeutic strategy for SCI.

This article presents a dataset obtained from the deployment of an IEEE 802.15.4g SUN (Smart Utility Network) single-hop network (11 nodes) in a large industrial scenario (110,044 m 2 ) for a long period of time (99 days). The dataset contains ∼11 M entries with RSSI (Received Signal Strength Indicator), CCA (Clear Channel Assessment), and PDR (Packet Delivery Ratio) values. The analyzed results show a high variability in the average RSSI (i.e., between −82.1 dBm and −101.7 dBm) and CCA (i.e., between −111.2 dBm and −119.9 dBm) values, which is caused by the effects of multi-path propagation and external interference. Despite being above the sensitivity limit for each modulation, these values result in poor average PDR values (i.e., from 65.9% to 87.4%), indicating that additional schemes are needed to meet the link reliability requirements of industrial applications. Hence, the presented dataset will allow researchers and practitioners to propose novel mechanisms and evaluate their performance using realistic conditions, enabling the dependability vision of the RAW (Reliable and Available Wireless) WG (Working Group) at the IETF (Internet Engineering Task Force).

ABSTRACT This study aimed at analyzing the energetic substrate (ES) in the main storage tissues of Steindachneridion parahybae, throughout the reproductive cycle in captivity. Differently from wild, in captivity, feeding is not interrupted during the reproductive period, the females do not spawn spontaneously, and they are sedentary. Adult females were sampled monthly and based on their histology and gonadosomatic index (GSI), ovaries were classified into: previtellogenic (PRV), vitellogenic (VTG), and regression (REG) stages. Ovaries at the VTG stage showed higher protein and lipids levels than at the PRV stage with a positive correlation between these substrates and the GSI. Muscle was the main source of proteins transferred to the ovaries, according to the negative correlation between these organs. Lipids remained unchanged in the liver, which is an important supplier in vitellogenesis, a pattern that probably occurs due to the continuous feeding. Muscular glycogen levels were higher at the VTG and REG than at the PRV stages. Plasma triglycerides were also higher during REG, while glucose levels were more elevated during the VTG stage. These results suggest that with constant food supply, the pattern of deposition of ES in S. parahybae is different from that described for other wild potamodromous species.
