Trapped electron mode (TEM) is the main source of turbulence predicted for the unique operation regime of a flat temperature profile under low-recycling conditions in the LTX-β tokamak, while ion temperature gradient driven turbulence may also occur with gas fueling from the edge. To investigate mainly TEM scale density fluctuations, a high spatial and time resolution 2D beam emission spectroscopy (BES) diagnostic is being developed. Apart from spatially localized density turbulence measurement, BES can provide turbulence flow and flow shear dynamics. This BES system will be realized using an avalanche photodiode-based camera and narrow band interference filter. The system can acquire data at 2 MHz. Simulations with the Simulation of Spectra (SOS) code indicate that a high signal to noise ratio can be achieved with the proposed system. This will enable sampling the density fluctuations at this high time resolution. The design considerations and system optimization using the SOS code are presented.

The objective of this study was to identify the relative contribution of tenderness factors for three beef muscles with similar tenderness ratings. Longissimus lumborum (LL), tensor fascia latae (TF) and gastrocnemius (GC) were collected from 10 USDA low Choice beef carcasses and assigned to a 5 or 21 days aging period (n = 60). Sarcomere length, troponin-T degradation, collagen content, mature collagen crosslink density, intramuscular lipid content and trained panel analysis were measured. Correlation and multivariate regression analysis indicated each muscle has a specific tenderness factor that contributed to the overall tenderness evaluated by trained panelists. The equations indicated LL tenderness was driven by lipid content (P .05); TF tenderness was driven by collagen content (P .05). GC tenderness was driven by proteolysis (P .01), and only collagen content can be casually used as an overall tenderness predictor for all three cuts.

We apply a cascaded linear model analysis to a micro-channel plate x-ray framing camera. We establish a theoretical expression of the Noise Power Spectrum (NPS) at the detector's output and assess its accuracy by comparing it to the NPS of Monte Carlo simulations of the detector's response to a uniform illumination. We also demonstrate that fitting the NPS of experimental data against a parametric model based on this expression can yield valuable information on the imaging ability of framing cameras, offering an alternative approach to the usual method employed to measure their modulation transfer functions.

High temperature is a major environmental factor that limits wheat (Triticum aestivum L.) productivity. Climate models predict greater increases in night-time temperature than in daytime temperature. The objective of this research was to compare the effects of high daytime and high night-time temperatures during anthesis on physiological (chlorophyll fluorescence, chlorophyll concentration, leaf level photosynthesis, and membrane damage), biochemical (reactive oxygen species (ROS) concentration and antioxidant capacity in leaves), growth and yield traits of wheat genotypes. Winter wheat genotypes (Ventnor and Karl 92) were grown at optimum temperatures (25/15 °C, maximum/minimum) until the onset of anthesis. Thereafter, plants were exposed to high night-time (HN, 25/ 24 °C), high daytime (HD, 35/15 °C), high daytime and night-time (HDN, 35/ 24 °C) or optimum temperatures for 7 days. Compared with optimum temperature, HN, HD and HDN increased ROS concentration and membrane damage and decreased antioxidant capacity, photochemical efficiency, leaf level photosynthesis, seed set, grain number and grain yield per spike. Impact of HN and HD was similar on all traits. Greater impact on seed set, grain number and grain yield per spike was observed at HDN compared with HN and HD. These results suggest that HN and HD during anthesis cause damage of a similar magnitude to winter wheat.

The use of energy harvesting for powering wireless sensors is made more challenging in most applications by the requirement for customization to each specific application environment because of specificities of the available energy form, such as precise location, direction and motion frequency, as well as the temporal variation and unpredictability of the energy source. Wireless power transfer from dedicated sources can overcome these difficulties, and in this work, the use of targeted ultrasonic power transfer as a possible method for remote powering of sensor nodes is investigated. A powering system for pipeline monitoring sensors is described and studied experimentally, with a pair of identical, non-inertial piezoelectric transducers used at the transmitter and receiver. Power transmission of 18mW (Root-Mean-Square) through 1m of a118mm diameter cast iron pipe, with 8mm wall thickness is demonstrated. By analysis of the delay between transmission and reception, including reflections from the pipeline edges, a transmission speed of 1000m/s is observed, corresponding to the phase velocity of the L(0,1) axial and F(1,1) radial modes of the pipe structure. A reduction of power delivery with water-filling is observed, yet over 4mW of delivered power through a fully-filled pipe is demonstrated. The transmitted power and voltage levels exceed the requirements for efficient power management, including rectification at cold-starting conditions, and for the operation of low-power sensor nodes. The proposed powering technique may allow the implementation of energy autonomous wireless sensor systems for monitoring industrial and network pipeline infrastructure.

High temperature (HT) stress is one of the major environmental factors influencing yield of soybean (Glycine max L. Merr.) in the semi-arid regions. Experiments were conducted in controlled environments to study the effects of HT stress on anatomical changes of pollen and their relationship to pollen function in soybean genotype K 03-2897. Objectives of this study were to (a) quantify the effect of HT stress during flowering on pollen function and pod set and (b) observe the anatomical changes in pollen grains of soybean plants grown under HT stress. Plants were exposed to HT (38/28 °C) or optimum temperature (OT, 28/18 °C) for 14 days at flowering stage. HT stress significantly decreased in vitro pollen germination by 22.7 % compared to OT. Pollen from HT stress was deformed; it had a thicker exine wall and a disintegrated tapetum layer. HT stress decreased pod set percentage (35.2 %) compared to OT. This study showed that decreases in pollen in vitro germination by HT stress were caused by anatomical changes in pollen, leading to decreased pod set percentage under HT stress.

Aim Percutaneous tibial nerve stimulation (PTNS) is increasingly being used as a treatment for faecal incontinence (FI). The evidence for its efficacy is limited to a few studies involving small numbers of patients. The aim of the study was to assess the efficacy of PTNS in patients with urge, passive and mixed FI. Method A prospective cohort of 100 patients with FI was studied. Continence scores were determined before treatment and following 12 sessions of PTNS using a validated questionnaire [Cleveland Clinic Florida (CCF)-FI score]. The deferment time and average number of weekly incontinence episodes before and after 12 sessions of treatment were estimated from a bowel dairy kept by the patient. Quality of life was assessed prior to and on completion of 12 sessions of PTNS using a validated questionnaire [Rockwood Faecal Incontinence Quality of Life (QoL)]. Results One hundred patients (88 women) of median age of 57 years were included. Patients with urge FI (n = 25) and mixed FI (n = 60) demonstrated a statistically significant improvement in the mean CCF-FI score (11.0 ± 4.1 to 8.3 ± 4.8 and 12.8 ± 3.7 to 9.1 ± 4.4) with an associated improvement in the QoL score. This effect was not observed in patients with purely passive FI (n = 15). Conclusion The study demonstrates that PTNS benefits patients with urge and mixed FI, at least in the short term.

Boyle Molecular diagnosis on tissues and cells: how it affects training and will affect practice in the future On 25th November 2011, a symposium organized by the Royal College of Pathologists, entitled ‘Molecular diagnosis on tissues and cells’, took place in London. As trainees in histopathology and cytopathology, we were stimulated to consider the role that molecular biology is likely to play in future practice and how this is addressed by our own training. The symposium provided a basis for this article. Routine samples requiring molecular analysis are equally relevant to histopathologists and cytopathologists, and molecular biology laboratories are now using cytological as well as histological material for diagnostic testing, allowing different specimen types to be used as and when they are most appropriate. The most widely used types of molecular analysis in routine cellular pathology are EGFR testing in lung cancer, molecular testing of thyroid nodules, fluorescence in situ hybridization testing of urine samples, clonality analysis in lymphoma testing, HER2 testing in breast and gastric cancer, KRAS testing in colorectal cancer, intraoperative assessment of breast cancer sentinel nodes, molecular testing of gastrointestinal stromal tumours and mismatch repair protein analysis. Of these, the majority in the UK are carried out on histopathology samples, although many are applicable to cytological samples if adequate material is obtained. We are particularly encouraged by the potential of molecular diagnostic cytology in traditionally difficult areas, such as intraoperative assessment. We believe that increasing reliance on molecular diagnostic techniques will also herald changes in training.

Soybean (Glycine max L. Merr.) genotype K 03-2897 was grown in a controlled environment to study the effects of high-temperature (HT) stress on ultrastructural changes in leaves and its relationship with photosynthetic rate. The objectives of this study were to (i) quantify the effects of HT stress during fl owering stage on photosynthetic rate and (ii) observe the anatomical and ultrastructural changes in leaves of soybean grown under HT. Plants were exposed to HT (38/28°C) or optimum temperature (OT; 28/18°C) for 14 d at fl owering stage R2. High-temperature stress signifi cantly decreased the leaf photosynthetic rate and stomatal conductance by 20.2 and 12.8%, respectively, compared with those at OT. However, HT stress signifi cantly increased the thicknesses of the palisade and spongy layers and the lower epidermis above those at OT. In addition, HT stress damaged the plasma membrane, chloroplast membrane, and thylakoid membranes. The mitochondrial membranes, cristae, and matrix were distorted under HT stress. High-temperature stress increased the content of leaf reducing sugars by 82.6% above that at OT. Leaves under HT stress had a higher carbon isotope ratio compared with that at OT. Decreases in photosynthesis at HT stress were mediated through anatomical and structural changes in the cell and cell organelles, particularly the chloroplast and mitochondria.

ObjectivesWoody breast is a myopathy observed in chicken breast meat (Pectoralis major) characterized by its tough and rubbery texture. However, the exact causation of woody breast texture is still unknown. We hypothesize that sarcoplasmic reticulum (SR) dysfunctionality early postmortem results in rapid leakage of intracellular calcium may partially contribute to the abnormal meat texture observed in woody breast meat. The objective of this preliminary study was to investigate this hypothesis.Materials and MethodsFourteen Ross line broiler breast fillets (7 severe woody breast and 7 normal) were collected at 3 h postmortem from a commercial processing plant located in the southeast United States. The 7 woody breast samples also exhibited moderate to severe white striping. The 7 normal samples did not exhibit any signs of white striping or woody breast. Each sample was trimmed, weighed, vacuum packaged and frozen at –20°C at approximately 8 h postmortem. One 1.9 cm strip across the cranial end of each fillet was fabricated and pulverized in liquid nitrogen to measure sarcomere length (Laser Scan Confocal Microscope with a 100x/NA 1.4 objective), calpain activity (immunoblotting for µ-calpain autolysis), proteolysis (immunoblotting for troponin-T degradation) and collagen content (hydroxyproline content). Purge was also collected from each sample to evaluate protein (bicinchoninic acid assay) and free calcium concentration (atomic absorption).ResultsWoody breast fillets were heavier than normal chicken breast fillets (522.9 vs. 446.9g; P < 0.05). Woody breast samples tended to have shorter sarcomeres (1.70 vs. 2.02 µm; P = 0.0543) and less intact troponin-T compared to normal breast samples (relative intact troponin-T band density: 49.98 vs. 56.97%; P = 0.0515) at 8 h postmortem. It was interesting to note that no µ-calpain band was detected through immunoblotting for both the woody breast and normal samples at 8 h postmortem. Other studies have found similar results as poultry µ-calpain autolyzed at a much rapid rate than µ-calpain in mammalian species. In addition, the purge from woody breast samples also had higher levels of free calcium compared to normal samples (6.2 vs. 4.2 nmol calcium/mg protein; P < 0.05). Lastly, there was more collagen present in the woody breast samples compared to normal chicken breast samples (3.89 vs. 2.08 mg collagen/g muscle tissue; P < 0.05).ConclusionThe results indicated that the cause of texture abnormality of woody breast may be the combined effects of more calcium being released from the SR early postmortem resulting in shorter sarcomere length and more collagen being deposited in the chicken breast meat. Additional research with the focus on SR integrity and functionality as well as collagen crosslinks are needed to further elucidate the basic mechanism of woody breast texture formation.

To transform undergraduate biology education, faculty need to provide opportunities for students to engage in the process of science. The rise of research approaches using next-generation (NextGen) sequencing has been impressive, but incorporation of such approaches into the undergraduate curriculum remains a major challenge. In this paper, we report proceedings of a National Science Foundation–funded workshop held July 11–14, 2011, at Juniata College. The purpose of the workshop was to develop a regional research coordination network for undergraduate biology education (RCN/UBE). The network is collaborating with a genome-sequencing core facility located at Pennsylvania State University (University Park) to enable undergraduate students and faculty at small colleges to access state-of-the-art sequencing technology. We aim to create a database of references, protocols, and raw data related to NextGen sequencing, and to find innovative ways to reduce costs related to sequencing and bioinformatics analysis. It was agreed that our regional network for NextGen sequencing could operate more effectively if it were partnered with the Genome Consortium for Active Teaching (GCAT) as a new arm of that consortium, entitled GCAT-SEEK(quence). This step would also permit the approach to be replicated elsewhere.

Background: Both actinomycotic granules and pseudoactinomycotic radiate granules (PAMRAGs) occur in the female genital tract, most commonly in the endometrium. It is important to distinguish between these since the former may result in pelvic inflammatory disease and require antibiotic treatment while the latter is non-infectious and does not require specific treatment. Aims: To investigate the coexistence of actinomyces-like organisms and PAMRAGs in the same granules, and describe the presence of PAMRAGs in the cervix and the vulva. Methods: Six cases with actinomyces-like organisms and PAMRAGs in the same granules (four in the endometrium, one in a tubo-ovarian abscess, and one in both the endometrium and a tubo-ovarian abscess) are reported as well as seven examples of PAMRAGs in the cervix and one in a vulval abscess. Results: The combined granules consisted of central basophilic Gram and silver positive filamentous organisms consistent with actinomyces surrounded by radiating eosinophilic club-like formations which were Gram and silver negative, the latter consistent with PAMRAGs. The PAMRAGs in the cervix and vulva consisted entirely of Gram and silver negative radiating eosinophilic club-like formations. Conclusions: Although actinomycotic granules and PAMRAGs are distinct lesions which should be distinguished for patient management, they may coexist in the same granules. It is likely in such cases that the PAMRAGs form around the bacterial colonies which act as a nidus. The presence of radiating eosinophilic club-like formations characteristic of PAMRAGs does not preclude the presence of actinomyces. Careful morphological examination plus supportive Gram and silver stains, if necessary, allows the diagnosis of these combined granules. PAMRAGs also occur in the cervix, where it is likely that they form secondary to encrustation of inspissated mucus, and in the vulva.

A double blind trial of practolol in coronary heart disease has been conducted for 2 years. In 298 patients with acute myocardial infarction there was no reduction in overall mortality. In a group with initial heart rate over 100 per minute mortality was significantly lowered up to 1 year. Of 484 patients with coronary heart disease treatment for 2 years did not produce a significant reduction in infarction or sudden death. Beta-adrenergic blocking drugs have been shown to reduce left ventricular work and to have an antiarrhythmic action. On these grounds they would seem theoretically to have a place in the management of acute myocardial infarction. Practolol is a cardio-selective beta-blocking agent with an intrinsic sympathomimetic action, but devoid of local anaesthetic effect. It has been found effective in post infarction arrhythmias (1). In early infarction it reduces the area of necrosis as measured by surface ST segment mapping (2).

Groundwater was sampled with a straddle-packer system at the Stratmat Main Zone deposit, New Brunswick, Canada, to better understand the origin of self-potential (SP) anomalies associated with massive sulphide mineralization and to investigate processes of sulphide oxidation below the water table. Groundwater displays a marked increase in total dissolved solids (TDS; 200 to 1600 mg/l) through the sulphide zone (84–89 m in depth). Groundwater ranges from low TDS CaHCO 3 -type water at shallow depth to higher TDS CaSO 4 -type water within the massive sulphides. The massive sulphides are coincident with a large SP anomaly (−600 mV) compared to the host rocks with an associated positive temperature anomaly of 0.05°C. With increasing depth through the massive sulphide zone, base metal (Cu, Zn, Mo, Pb, Cd) and Fe concentrations increase (e.g. Fe increases from 40 to 2300 mg/l). Stable isotopic compositions in groundwater also vary with depth in the sulphide zone with δ 18 O VSMOW increasing from −12.32 to −12.00‰ and δ 13 C VPDB from −13.55 to −10.31‰. The stable isotopes, TDS and straddle-packer recovery time data indicate that groundwater flow is greatest in the upper parts of the massive sulphides. The large SP anomaly is interpreted to result from decoupled sulphide oxidation occurring between oxygenated waters impacted by the borehole and non-pyritic sulphide phases within the sulphide mass. Electrons pass from sulphide phases (e.g. pyrrhotite) through intervening pyrite to dissolved oxygen in the borehole, which renders a cathodic character to the wall of the borehole and an anodic character to the interior of the sulphide. The higher hydraulic conductivity of the upper massive sulphide has allowed penetration of dissolved oxygen, which produces a similarly decoupled oxidation reaction between the upper and lower sulphides, at least proximal to the borehole. The increasingly anodic nature of the sulphide mass with increasing depth is demonstrated by the chemical evolution of groundwater with depth. Increasing SO 4 2− , H + , Fe 2+ and Zn 2+ concentrations are evidence of the oxidation reactions occurring in deeper sulphides, whereas the higher pH in the upper massive sulphide is evidence of O 2 reduction. Increased δ 13 C VPDB with depth probably results from enhanced carbonate mineral dissolution as acid is produced. The unusual positive temperature anomaly associated with the borehole may be partly a result of exothermic oxidation of the sulphide but is more likely due to the higher thermal conductivity of massive sulphide relative to country rock. The sulphide appears to be acting as a conduit for heat from deeper areas, which suggests that a large sulphide mass extends beneath the intersected section.

Aims: It is generally considered that an unequivocal histological diagnosis of endometriosis requires the presence of endometrioid-type glands and endometrioid-type stroma. However, small nodules or plaques of endometrioid-type stroma without glands have been noticed by the authors in repeated peritoneal biopsies performed for suspected endometriosis. These are often, but not always, accompanied by typical endometriosis with glands. This form of endometriosis has been previously referred to as stromal or micronodular stromal endometriosis. However, there has been little reference to this condition in the literature. Methods: In this study, there was a review of a large series (n = 274) of peritoneal biopsies with a diagnosis of endometriosis with a view to ascertaining the frequency of stromal endometriosis. Results: Stromal endometriosis, characterised histologically by small microscopic nodules or plaques of endometrioid-type stroma, sometimes with a whorled pattern and prominent vascularity and erythrocyte extravasation, was identified in 44.9% of the biopsies. In 6.6% of the biopsies, stromal endometriosis occurred without typical endometriosis. The foci of stromal endometriosis usually had a superficial location just beneath the mesothelial surface or protruding above this. Associated histological features present in some cases included reactive mesothelial proliferation, inflammation, giant cell or granuloma formation, haemosiderin pigment deposition, microcalcification and decidualisation and myxoid change. Conclusions: Stromal endometriosis, usually in the form of superficial nodules or plaques, is a relatively common form of endometriosis which typically occurs in association with typical endometriosis but occasionally on its own. Pathologists should be aware of the existence of this form of endometriosis, the morphological features of which may be subtle. The typical location, intimately associated with surface mesothelium, may suggest that stromal endometriosis derives from mesothelial or submesothelial cells via a metaplastic process.

This review paper presents and places in context findings from 23 manuscripts based on the data sets of the National Institute of Child Health and Human Development (NICHD) Study of Early Child Care and Youth Development (SECCYD). The NICHD study tracked the development of more than 1000 children from birth through age 15. The children were born across the USA to families that were diverse in terms of their economic, educational, and ethnic background. The children also varied in terms of the timing, extent, quality, and type of their child care experiences. The findings reported in this review paper pertain to (1) predictors of child-mother attachment; (2) links between child-mother attachment and children's developmental outcomes; and (3) methods for assessing attachment. The review paper also includes suggestions for future attachment research afforded by the data sets of the NICHD SECCYD. Not all the hypotheses tested in the reviewed papers were confirmed, and those that were confirmed were associated with modest effect sizes. Some findings emerged under specific conditions, even though they were expected to be more universal. These findings need to be discussed and aligned with theory. Also, the extent to which the findings generalize beyond the historical period and the country in which the data were collected merits discussion and exploration.

The immunoglobulin degrading enzyme of Streptococcus pyogenes, IdeS, is an unusual cysteine protease produced by group A streptococci for which the only known substrate is immunoglobulin G (IgG). To date, IdeS has not been found to cleave any of the known synthetic substrates that other cysteine proteases hydrolyse, thus making the development of an IdeS detection assay difficult. Furthermore, at high doses of substrate, product generation is inhibited potentially due to the need for a dimeric enzyme complex with IgG. In this study we have developed a mass spectral assay for IdeS activity based on the detection of an Mr approximately 25,300 Fc fragment that retains the ability to bind streptococcal protein G. Using this assay procedure, evidence for a multimeric enzyme-substrate complex was obtained as well as identifying isolated heavy chains as a non-substrate inhibitor of IdeS activity. Under appropriate experimental conditions the assay could be used to detect IdeS activity in bacterial culture media or in human plasma without a requirement for purified reactants. The availability of a rapid and sensitive assay for IdeS should facilitate the detailed biochemical characterization of this unusual bacterial cysteine protease.

Surface-enhanced laser desorption/ionization time-of-flight analysis was used to monitor both the kinetics and heterogeneity of product formation during the biotinylation of a number of model proteins and peptide targets. The selected molecules were the IgG-binding protein, protein A, human serum albumin, and a synthetic peptide corresponding to the N terminus of a streptococcal M1 protein. The extent of biotinylation was determined by kinetic analysis of the shift in molecular mass from the native material. Each residue modified by reaction with N-hydroxysuccinimide biotin resulted in an addition of approximately 341 amu to the native protein or polypeptide. The novelty of the method was in the ability to determine the molecular mass shift, without first separating the targeted molecule from the biotinylating reagent. The analysis was rapid, simple, and provided information on the average number of biotin molecules added and the homogeneity of the resulting product.

Binding proteins that have high affinities for mammalian plasma proteins that are expressed on the surface of bacteria have proven valuable for the purification and detection of several biologically important molecules from human and animal plasma or serum. In this study, we have isolated a high affinity albumin-binding molecule from a group G streptococcal isolate of bovine origin and have demonstrated that the isolated protein can be biotinylated without loss of binding activity and can be used as a tracer for quantification of human serum albumin (HSA). The binding protein can be immobilized and used as a selective capture reagent in a competitive ELISA format using a biotinylated HSA tracer. In this assay format, the sensitivity of detection for 50% inhibition of binding of HSA was less than 1 microg/ml. When attached to the bacterial surface, this binding protein can be used to deplete albumin from human plasma, as analyzed by surface-enhanced laser desorption ionization time of flight mass spectrometry.

In this study, the importance of different domains of the fibrinogen molecule in the binding and assembly of a surface plasminogen (plgn) activator has been analyzed. This was achieved using SELDI technology that enabled dissociation of bound fragments from intact bacteria and accurate distinction between fibrinogen fragments based on their molecular mass. These studies indicate that Streptococcus pyogenes binds directly to human fibrinogen fragment D but not fragment E. The predominant surface proteins binding to fragment D were associated with the mrp gene product. Surface-associated fibrinogen fragment D was capable of anchoring a functional surface plgn activator complex. Taken together, these data indicated that fragment D of fibrinogen is necessary and sufficient to anchor a plgn activator complex on the surface of Streptococcus pyogenes.

A novel immunoproteomic assay, combining specificity of antibody with precision of mass spectral analysis is described, and a number of practical applications are presented. The assay is carried out in three steps. The first step of the assay involves antibody immobilization, using a bacterial Fc binding support. The second step is antigen capture and washing to remove non-specific binding. The third step involves analysis of the captured antigens by SELDI-TOF. The assay has many advantages in sensitivity, speed, and economy of reagents in detection of specific antigens or antibodies. In addition, under appropriate experimental conditions, semi-quantitative data may be obtained. By combining the increasing range of selective specific antibody reagents available, in part due to advances in antibody engineering technology, and the resolving power available, using mass spectrometry, immunoproteomics is a valuable technique in proteomic analysis. A number of examples of the application of this technique to analysis of biological systems are presented.

Very few investigations have dealt with the environmental impact of the highly toxic metal thallium (Tl), and its subsequent dispersion through natural processes and human activities such as mining and farming. This study is focused on high concentrations of Tl in rocks in SW Guizhou, China, that are related to several widely scattered disseminated gold–mercury–arsenic and coal deposits, and a primary Tl deposit within an Au–As–Hg–Tl metallogenic belt of the Huijiabao anticline. The Tl, Hg and As in the Lanmuchang Hg–Tl deposit area are associated with the abundant occurrence of sulfide minerals such as lorandite, realgar, orpiment and cinnabar. Concentrations of Tl range from 100 to 35 000 ppm in sulfide ores, and 39–490 ppm in host rocks. The enrichment of Au, Tl, Hg, As, and Sb in the Yanshang gold mineralized area reflects the occurrence of Au mineralization and its mineral assemblage of Tl–Hg–As–Sb sulfides. Thallium ranges from 0.22 to 16 ppm in Au ores and host rocks. Thallium in coals is enriched up to 46 ppm within the Au–As–Hg–Tl metallogenic belt, and is derived from the regional Au–As–Hg–Tl mineralization. Mercury and As show a similar distribution to Tl with high concentrations in sulfide ores, coals and host rocks. Human populations living near and downstream of Tl deposits and Tl-bearing ore deposits are susceptible to Tl contamination because of its high toxicity and high uptake rate by crops. The dispersion of Tl, Hg and As associated with the primary mineralization of Au–As–Hg–Tl can be traced through physical erosion and chemical weathering, producing secondary dispersion into soils, groundwater and surface water and crops. Mining activities compound the natural processes, readily dispersing Tl into the surface environment. The Lanmuchang area illustrates Tl contamination related to a Tl-rich deposit due to both natural processes and the impact of mining. The Yanshang area demonstrates Tl contamination related toa Tl-bearing gold deposit, caused by natural processes in the absence of mining activity.

Batrachochytrium dendrobatidis is a major pathogen of frogs worldwide, associated with declines in amphibian populations. Diagnosis of chytridiomycosis to date has largely relied upon histological and immunohistochemical examination of toe clips. This technique is invasive and insensitive particularly at early stages of infection when treatment may be possible. We have developed a real-time PCR Taqman assay that can accurately detect and quantify one zoospore in a diagnostic sample. This assay will assist the early detection of B. dendrobatidis in both captive and wild populations, with a high degree of sensitivity and specificity, thus facilitating treatment and protection of endangered populations, monitoring of pristine environments and preventing further global spread via amphibian trade.

Streptococcus pyogenes protein H ( sph ) is an immunoglobulin-binding protein present in the Mga regulon of certain M1 serotype isolates. Although sph is present in many strains, it is frequently not expressed. In this paper we show that protein H was highly expressed after bacteria were injected into the skin of mice and were recovered from the blood, kidney, or spleen at various times postinfection. The percentage of protein H-positive colonies increased with time, reaching 100% in the spleen and kidney within 24 to 72 h postinfection. The up-regulation of sph expression was also observed in a mga mutant.

This study explores the complicated social interactions related to gender that take place at school recess. Using grounded theory methodology and a social constructionist perspective, the researchers observed 67 recesses in New England and then examined the data to determine which themes emerged. Results demonstrate that there is great complexity in preadolescent gender relations and that their incidence and significance is in flux during recess time. Among the results noted were ways in which children reinforce their gender identities; the wide intragender variability in choice of activities especially for girls; and clear evidence of borderwork, that is, those interactions where boys and girls play together but there is something about the play that actually reinforces the gender divisions rather than diminishes them. Future research should examine ways adults can support children in developing relationships based on mutual collegiality and liking, thereby building their sense of competence and self-esteem.

The small karstic watershed of Lanmuchang, in a Hg–Tl mineralized area in SW Guizhou Province, China, exhibits an enrichment of toxic Tl in groundwater and related stream water. This affords an excellent demonstration of the natural processes of Tl dispersion, and the resultant impact on the local ecosystem. The distribution of Tl in the water system follows a decreasing concentration pattern from deep groundwater to stream water to shallow groundwater. Tl shows high levels (13–1100 μg/L) in deep groundwater within the Tl-mineralized area, decreasing with distance away from the mineralized area to background levels (0.005 μg/L). The distribution of Tl in the water system is constrained by Tl mineralization, water–rock interactions and hydrogeological conditions. Tl concentrations in waters generally correlate with concentrations of total dissolved solids, sulphate, Ca and pH values, suggesting the contribution of water-rock interactions to water geochemistry. Water–rock interactions are driven by weathering of Tl-bearing sulfides which decreases pH values in groundwater, and by dissolution of limestone enhanced by acid fluids. Tl in stream water in both the base-flow and flood-flow regimes shows higher concentrations than it does in shallow groundwater that serves as the stream's source (mainly springs, dug-well flows and karstic cave waters). Concentrations of Tl in stream water in the flood-flow regime are generally lower than in the base-flow regime due to dilution effects, but those in the waters of mid-stream are almost the same as in the base-flow regime, probably due to contribution from Tl-rich soil water seepage or from acid mine drainage (AMD). Unexpectedly, Tl concentrations in stream water in both regimes are remarkably higher (2–30 fold) downstream than up- and mid-stream. These pronounced increases of Tl concentration are likely caused by unidentified discharges of deep groundwater through fractured zones to the downstream trace. The groundwater-related Tl transfer processes affect the ecosystem through contamination of water supply and arable soil and ultimately the food chain with undoubted risks to human health. Therefore, the results of this study are important for environmental planning and regulations, and will also serve as baseline data for future research on Tl natural dispersion processes.

Protein H is an immunoglobulin-binding protein expressed by certain M1 serotypes of Streptococcus pyogenes. In a recent study of invasive group A isolates, it was found that none of the 16 M1 serotype isolates analyzed expressed protein H on their surface despite the presence of the protein H gene (sph) in approximately one-third of the isolates. Selection of stable protein H-expressing variants could be achieved by infection of prtH(+) non-expressing strains into a mouse skin and recovering bacteria from the spleen. This effect was independent of the transcription regulator Mga, since a similar effect was noted in an mga(-) mutant. Thus, host passage of S. pyogenes can lead to stable high level expression of Protein H.

Post-translational modification of the antiphagocytic M1 protein of Streptococcus pyogenes can influence its binding properties for human immunoglobulin G subclasses and its invasive potential. Current methods of monitoring this modification event involve N-terminal sequencing and are cumbersome, slow and not amenable to routine analysis. In this study we demonstrate that surface enhanced laser desorption/ionization-time of flight mass spectrometry can be used to monitor modification of the M1 protein by the secreted bacterial cysteine protease, SpeB. This method, when combined with a specific antibody capture step provides a specific, rapid and sensitive assay for key virulence factors of the important human pathogen Streptococcus pyogenes.

A detailed hydrogeochemical study at the undisturbed Halfmile Lake and Restigouche Zn–Pb deposits, Bathurst Mining Camp, New Brunswick, was initiated to elucidate processes controlling the oxidation of sulphide minerals, factors influencing base-metal dispersion in ground and surface waters, and to improve hydrogeochemical exploration methods. Groundwaters were collected using flow-through bailer and straddle-packer technologies; the latter of which proved to be the most effective under the fracture-flow dominated conditions. Groundwaters from the two deposits are compositionally distinct. At the Halfmile Lake deposit, groundwaters are dominated by low total dissolved solids (TDS −1 ) and Ca–HCO 3 -type compositions. Groundwaters at the Restigouche deposit vary in composition from shallow Ca–HCO 3 - and Ca–SO 4 -type waters to high TDS Na–Cl-type waters. Elevated sulphate and base metal contents (up to 4200 μg l −1 Zn and 1400 μg l −1 Pb) of shallow Ca–SO 4 -type groundwaters proximal to massive sulphides indicate that the sulphides are undergoing natural oxidation. The degree to which the massive sulphides are oxidized depends on the depth of penetration of dissolved oxygen and flux rates, which are controlled by the permeability of the rocks and local hydrological gradients. Transport of metals away from massive sulphides depends on physical factors that include hydrology, groundwater flow, and the degree of connectivity between groundwater and surface water environments, as well as groundwater pH, metal adsorption and colloid mobility.

Rice flour-based batter (RFBB) formulations for chicken drumstick coating were developed as an alternative for traditional wheat flour-based batter (WFBB). Physicochemical properties and storage stability of selected RFBB were evaluated and compared to WFBB. Batter pickup of RFBB formulated in combination with oxidized corn starch and methylcellulose (MC) was not significantly different from that of WFBB. In contrast, batters with only rice and corn flour (60:40% flour weight) exhibited significantly higher pickup. Rice flour batter with 15% oxidized corn starch had the lowest batter pickup. All RFBB exhibited (P0.05) lower oil absorption than WFBB. The TBA values of RFBB and WFBB increased (P0.05) with increased frozen storage time at -40 C for 90 d. The RFBB with MC exhibited the lowest TBA values, whereas WFBB had the highest values. Microstructural analysis revealed that freezing caused structural deterioration of all batters, but the RFBB with MC exhibited less freezing tolerance than other samples. The total plate counts of immediately fried or frozen fried chicken stored for 90 d were less than 1 log cfu/g sample. The RFBB with 5% oxidized corn starch and MC can replace WFBB on fried drumsticks. Additionally, RFBB results in a healthier product due to lower fat absorption.

The convenience and appeal of battered or breaded products have resulted in a sales increase of 100% since 1980. Because of the rapid growth of the Asian-American population and increasing consumption of rice and rice products, rice flour is a logical alternative for wheat flour in traditional batter formulation. The effects of ingredients used in rice flour-based batters on adhesion characteristic for deep-fat fried chicken drumsticks were studied by laser scanning confocal microscopy (LSCM) and texture analysis. Raw chicken drumsticks were predusted with egg albumin powder before dipping into batters prepared from combinations of rice flour, yellow corn flour, oxidized cornstarch, methylcellulose, or xanthan gum. The drumsticks were fried at 175 +/- 5 C until the internal temperature reached at least 71 C. For LSCM, samples were fixed overnight and were sectioned by vibratome (200 micrometer) before viewing. Batter adhesion was determined using an attachment specifically designed for chicken drumsticks. Microstructural analysis showed that batter formulated with a 50:50 mixture of rice and corn flours adhered better to drumsticks than batter with other rice flour ratios. Xanthan gum (0.2%) or methylcellulose (0.3%) alone had poor adhesion to chicken skin. However, when combined with other ingredients, xanthan gum increased the amount of batter pick-up before frying by increasing viscosity. Egg albumin significantly facilitated batter adhesion. The results from texture analysis supported the microstructural studies. As rice flour ratio increased from 50 to 70%, the binding force decreased. Rice flour showed potential as an alternative to wheat flour for batter formulas when the appropriate levels of oxidized starch, xanthan gum, and methylcellulose were included in the formulation.

Ground and surface waters collected from two undisturbed Zn–Pb massive sulphide deposits (the Halfmile Lake and Restigouche deposits) and active mines in the Bathurst Mining Camp (BMC), NB, Canada were analysed for the rare earth elements (REE). REE contents are highly variable in waters of the BMC, with higher contents typical of waters with higher Fe and lower pH. There are significant differences between ground- and surface waters and between groundwaters from different deposits. The REE contents of surface waters are broadly similar within and between deposit areas, although there are spatial variations reflecting differences in pH and redox conditions. Surface waters are characterised by strong negative Ce anomalies ([Ce/Ce*]NASC as low as 0.08), produced by oxidation of Ce3+ to Ce4+ and preferential removal of Ce4+ from solution upon leaving the shallow groundwater environment. Groundwaters and seeps typically lack significant Ce anomalies reflecting generally more reducing conditions in the subsurface environment and indicating that Ce oxidation is a rapid process in the surface waters. Deeper groundwaters at the Halfmile Lake deposit are characterised by REE patterns that are similar to the host lithologies, whereas most groundwaters at the Restigouche deposit have LREE-depleted patterns compared to NASC. Halfmile Lake deposit groundwaters have generally lower pH values, whereas Restigouche deposit groundwaters show greater heavy REE-complexation by carbonate ions. Shallow waters at the Halfmile Lake and Stratmat Main Zone deposits have unusual patterns which reflect either the adsorption of light REE onto colloids and fracture-zone minerals and/or precipitation of REE–phosphate minerals. Middle REE-enrichment is typical for ground- and surface waters and is highest for neutral pH waters. The labile portion of stream sediments are generally more middle REE-enriched than total sediment and surface waters indicating that the REE are removed from solution by adsorption to Fe- and Mn-oxyhydroxides in the order middle REE≥light REE>heavy REE.

Tamoxifen therapy may result in a variety of endometrial proliferative lesions, including adenocarcinoma, and as recently suggested, proliferative changes within endometriosis. This report describes an endometrioid adenocarcinoma arising in ovarian endometriosis in a patient taking tamoxifen. There were also foci of benign and borderline endometrioid adenofibroma in the same ovary and a synchronous endometrioid endometrial adenocarcinoma in the uterus. The spectrum of benign, borderline, and malignant endometrioid neoplasia arising within endometriosis suggests that tamoxifen, as a result of its estrogenic effects, may cause proliferative and, in rare instances, malignant changes in endometriosis.

Stream waters and sediments draining a gossan tailings pile at the Murray Brook massive sulphide deposit were collected to investigate Au mobility. Weathering of the massive sulphides at Murray Brook during the Late Tertiary period resulted in the concentration of Au in the gossan cap overlying the supergene Cu and unoxidized massive sulphide zones of the deposit.
