The aim of this research was to investigate the antioxidant, and anticholinergic properties, phenolics profile and antibacterial activities of Abelmoschus esculentus ethanol extract. The analysis of phenolic compounds was performed with LC–MS/MS. The antioxidant capacity (radical scavenging, metal-reducing power and total antioxidant activity) was assessed by DPPH, ABTS, Cu2+–Cu+ reducing (CUPRAC), Fe3+–Fe2+ reducing and ferric thiocyanate methods. The antibacterial activity was determined by disc diffusion and MIC (Minimum inhibitory concentration) methods and the anticholinergic property was predicted by inhibition of acetylcholinesterase (AChE). Acetohydroxamic acid was determined as the main phenolic compound with the highest amount in the flower extract. Also, quercetin, myricetin, fumaric acid, vanillic acid, ellagic acid, 4-hydroxybenzoic acid, salicylic acid, caffeic acid, kaempferol, butein, protocatechic acid, catechin hydrate, oleuropein and other components were detected, respectively. The ethanolic extract of the plant has 29.41% DPPH radical scavenging activity, 20.59% ABTS radical scavenging activity, and also moderate metal reduction potential. Also, the extract showed an inhibition effect on the AChE with IC50 values (0.18 mg mL−1). The ethanol extract of the plant showed antibacterial effect on Staphylococcus aureus, Escherichia coli, and Salmonella Typhimurium at different levels. These results suggested that extract of the flowers of A. esculentus extract might play a role in the treatment of some diseases with its antioxidant, anticholinergic, and antibacterial activity., Bu araştırmanın amacı, Abelmoschus esculentus’a ait çiçek kısımınlarının etanolik ekstraktının antioksidan ve antikolinerjik özelliklerini, fenolik bileşik profilini ve antibakteriyel aktivitesini araştırmaktır. Fenolik bileşiklerin analizi LC-MS/MS ile gerçekleştirilmiştir. Antioksidan kapasitesi (radikal giderme, metal indirgeme gücü ve toplam antioksidan aktivite) DPPH, ABTS, Cu2+-Cu+ indirgeme (CUPRAC), Fe3+-Fe2+ indirgeme ve ferrik tiyosiyanat yöntemleri ile değerlendirilmiştir. Antibakteriyel aktivite, disk difüzyon ve MIC (Minimum inhibitör konsantrasyonu) yöntemleri ile belirlenmiştir. Antikolinerjik özellik ise, asetilkolinesterazın (AChE) inhibisyonu ile tespit edilmiştir. Bitki özütünde miktar bakımından en fazla bulunan başlıca fenolik bileşiğin asetohidroksamik asit olduğu belirlenmiştir. Ayrıca, sırasıyla kuarsetin, myrisetin, fumarik asit, vanillik asit, ellagik asit, 4-hidroksibenzoik asit, salisilik asit, kafeik asit, kemferol, bütein, protokateşik asit, kateşin hidrat, oleuropein ve diğer bileşenler tespit edilmiştir. Bitkinin etanolik özütü, %29.41 DPPH radikal giderme aktivitesi gösterirken, %20.59 ABTS radikal giderme aktivitesine ve ayrıca orta düzeyde metal indirgeme potansiyeline sahiptir. Ayrıca ekstrakt, 0.18 mg mL-1 IC50 değeri ile AChE üzerinde bir inhibisyon etkisi göstermiştir. Bitkinin etanol özütü, Staphylococcus aureus, Escherichia coli ve Salmonella Typhimurium üzerinde farklı düzeylerde antibakteriyel etki göstermiştir. Elde edilen sonuçlar, A. esculentus çiçek özütünün, sahip olduğu antioksidan, antikolinerjik ve antibakteriyel özellikleri ile bazı hastalıkların tedavisinde rol oynayabileceğini düşündürmektedir.

Dikici, Emrah (Aksaray, Yazar), Plants generate secondary metabolites, which could be used as novel therapeutic compounds. These compounds are well known for their beneficial effects on human health. Thus, it is important to evaluate total phenolic and flavonoid contents as well as antioxidant activities of different plants. The present study was conducted to determine the total phenolic/flavonoid content and antioxidant activity of ethanol extract prepared from leaves of Phlomis pungens L. The total phenolic content of the ethanol extract of the sample were determined using the Folin-Ciocalteu reagent. Antioxidant activity of Phlomis pungens L. was determined using different in vitro experimental models, which include DPPH, FRAP, CUPRAC and potassium thiocyanate method. Total phenolic and flavonoid contents of Phlomis pungens L. were found to be 68.4 mg GAE/g and 4.9 mg QE/g, respectively. Antioxidant activity of the sample was comparable to commercial antioxidant standards (BHT, - tocopherol and trolox). The results showed that Phlomis pungens L has moderate free radical scavenging and reducing capacity. Overall, this study discovers total phenolic and flavonoid content as well as antioxidant properties of Phlomis pungens L, Bitkiler terapötik bileşikler olarak kullanılabilen ikincil metabolitleri üretirler. Bu bileşikler, insan sağlığı üzerindeki olumlu etkileri açısından iyi bilinmektedir. Bu nedenle farklı bitkilerin antioksidan aktivitelerinin yanı sıra toplam fenolik ve flavonoid içeriğinin değerlendirilmesi önemlidir. Bu çalışma Phlomis pungens L.’nin yapraklarından hazırlanan etanol özütünün toplam fenolik/flavonoid içeriğini ve antioksidan aktivitesini belirlemek amacıyla yürütülmüştür. Numunenin etanol özütünün toplam fenolik içeriği Folin-Ciocalteu reaktifi kullanılarak belirlendi. Phlomis pungens L.’nin antioksidan aktivitesi, DPPH, FRAP, CUPRAC ve potasyum tiyosiyanat yönteminin içeren farklı in vitro deneysel modeller kullanılarak belirlendi. Phlomis pungens L.’nin toplam fenolik ve flavonoid içeriği sırasıyla 68.4 mg GAE/g ve 4.9 mg QE/g olarak bulundu. Numunenin antioksidan aktivitesi, ticari antioksidan standartlarıyla (BHT, α-tokoferol ve trolox) kıyaslandı. Sonuçlar, Phlomis pungens L’nin orta düzeyde serbest radikal süpürme ve indirgeme Me kapasitesine sahip olduğunu gösterdi. Genel olarak, bu çalışma Phlomis pungens L’nin antioksidan özelliklerinin yanı sıra toplam fenolik ve flavonoid içeriğini ortaya çıkarmıştır.

Bu çalışma alıç (Crataegus monogyna) bitkisinin yapraklarının antioksidan aktivitesini belirlemek maksadı ile gerçekleştirilmiştir. Bu amaçla alıçın yaprak kısımları Erzincan ili İliç ilçesi Bağıştaş köyünden 2011 yılı mayıs ayı sonlarında toplandı, kurutuldu ve etanol ekstresi hazırlandı. Etanol ekstresi hazırlanan alıç (Crataegus monogyna) yaprakları üzerinde ferrik tiyosiyanat metoduna göre total antioksidan aktivite, kuprak metodu ile Cu2+ ve potasyum ferriksiyanat indirgeme metodu ile Fe3+ iyonları indirgeme kapasitesi, 1,1-difenil-2-pikril-hidrazil (DPPH) giderme aktivitesi tayinleri, toplam fenolik ve flavonoit bileşik miktar tayinleri yapıldı. Alıç (Crataegus monogyna) bitkisinin yapraklarından elde edilen 10 ? g mL-1 konsantrasyonluk etanol ekstresinin linoleik asit emülsiyonunun lipit peroksidasyonunu %40,9 inhibe ettiği gözlendi. Aynı konsantrasyonda BHT % 68,3, troloks'un ise % 29,9'luk bir inhibisyona sahip olduğu gözlendi. Toplam fenolik içeriği trolokstan daha fazla olduğu tespit edilen alıç (Crataegus monogyna) yüksek DPPH giderme ve metal indirgeme aktivitesi sergiledi. Etanol ekstresi elde edilen Crataegus monogyna bitkisinin antioksidan aktivitesi araştırılırken, butillenmiş hidroksi toluen (BHT), ?-tokoferol ve onun suda çözünen bir analoğu olan troloks referans antioksidan bileşikleri olarak kullanıldı. This study had been aimed to examine the antioxidant activitie of leaves of hawthorn (Crataegus monogyna). For this purpose, leaves of hawthorn (crataegus monogyna) was collected from Bağıştaş village which İliç, a town of Erzincan province in May 2011. It was dried and ethanol extraction was prepared. On prepared ethanol extraction, total antioxidant activity was determined by ferroccyanate reduction method, reduction capacity of Cu2+ was determined by Cuprac method, reduction capacity of Fe3+ ions were determined by thiocyanate reduction method, the activitiy of scavenging of 1,1-diphenyl-2-picryl-hydrazyl (DPPH) was determined and was determined the total amount of the compound of phenolic and flavonoid. It was observed that 10 ? g mL-1 concentrated of ethanol extract obtained from hawthorn (Crataegus monogyna) plant inhibited the lipid peroxidation of linoleic acid emulsion as 40.9 % respectively. In the same concentration, Butylated hydroxy toluene (BHT) and trolox caused inhibitions of 68.3% and 29.9 %, respectively. The content of total phenolic activity, DPPH scavenging and metal reduction activity of hawthorn (Crataegus monogyna) which was detected more than trolox. Butylated hydroxy toluene (BHT), ?-tocopherol and its an analoge which is solved in water, namely Trolox, were used as reference antioxidant compounds, while the antioxidant activity of hawthorn (Crataegus monogyna) plant whose ethanol extraction was taken out was examined. 73
