Metallomics is a rapidly growing research area investigating the interaction of metals with biological mol- ecules e.g. DNA, proteins and metabolites in living systems. It aims the understanding of all metal de- pendent metabolic processes such as uptake, transport, storage and excretion at molecular level. In order to access the qualitative and quantitative information of metals, which mostly occur at trace amounts and in the form of non-covalent complexes with biological ligands in a highly complex biological matrix, and to elucidate the metal-dependent life processes, metallomics utilizes the state of the art analytical and spectroscopic tech- niques. The mostly used approach for the analysis of metal complexes by preserving native metal species is the hyphenation of a chromatographic or an electrophoretic technique for high resolution separation with an elemental e.g., ICP-MS or molecular e.g., ESI-MS or MALDI-MS spectrometry technique for detection and/ or identification. X-ray absorption and X-ray fluorescence spectrometry and in-silico approaches with bioinfor- matics are among other main techniques/methodologies contributing the research activities in metallomics. This study highlights the basic terms, primarily used analytical approaches, state-of-the art instrumental tech- niques and very representative recent applications in the field., Metallomik, canlılarda metallerin biyolojik moleküllerle örn.,DNA, proteinler ve metabolitler etkileşimini inceleyen ve hızla gelişen bir araştırma alanıdır. Metallerin biyolojik sistemde geçirdiği tüm metabolik süreçlerin alım, taşınma, depolanma ve atılım gibi moleküler düzeyde anlaşılmasını hedefler. Metaller, biyolojik sistemlerde eser düzeylerde, genellikle kovalent olmayan koordinasyon kompleksleri halinde ve oldukça karmaşık bir biyolojik çevrede bulunur. Bu nedenle metallomik, metallere ilişkin nitel ve nicel bilgi edinilmesi ve metal-bağımlı yaşamsal süreçlerin aydınlatılması amacıyla, ileri analitik ve spektroskopik tekniklerden yararlanır. Metalik türlerin fizyolojik ortamdaki doğal formunu korumak suretiyle metal komplekslerinin analizi için en yaygın yaklaşım, Hibrit Tekniklerin kullanılmasıdır; bu sistemde, bileşenlerin yüksek çözünürlükte ayrımı için kromatografik veya elektroforetik bir teknik ile tayin ve/veya tanımlama için element örn., ICPMS veya moleküle örn., ESI-MS veya MALDI-MS özgül bir spektrometrik teknik birlikte kullanılmaktadır. X-ışınları absorpsiyon ve X-ışınları floresans spektroskopisi teknikleri ile bilgisayar-destekli teknolojileri içeren biyoinformatik yaklaşımlar da metallomik alanındaki araştırmalara katkı sağlayan diğer araçlar arasındadır. Bu çalışmada alanla ilgili temel kavramlar, kullanılan başlıca analitik yaklaşımlar ve ileri aletsel teknikler ele alınmış ve yakın zamanda yapılmış çalışmalardan örnekler seçilerek derlenmiştir

Sunulan çalışmada, periton içine tek dozda hekzavalan krom (Cr(VI)) çözeltisi injekte edilen deney farelerine ait karaciğer, böbrekler, beyin, akciğerler, kalp ve erbezleri dokularında krom, bakır, demir, mangan ve çinko düzeyleri tayin edilmiştir. Sonuçlar kontrol grubu deneklerine ait örneklerle karşılaştırıldığında krom içeriklerinin karaciğer ve böbreklerde yaklaşık 40 kat, diğer dokularda ise 3-5 kat arttığı; Cu, Fe, Mn and Zn dengesinin ise etkilendiği görülmüştür.Farklı organlardan elde edilen sitosolik özütlerdeki esansiyel eser elementlerin değişik molekül ağırlıklı biyomoleküller arasında dağılımı, dedektör olarak UV-VIS ve ICP-MS kullanılan boyutça eleme kromatografisi (Superdex-75 kolonlu) hibrit sistem kullanılarak tayin edilmiştir. Cr(VI) maruziyeti Fe, Mn, Cu and Zn-protein komplekslerine ait elusyon profillerinde önemli farklılıklara neden olmuştur. Özellikle karaciğer ve böbrekler olmak üzere Mn, Fe ve Zn-bağlayıcı proteinlerin biyolojik sentezlenmelerinde tetiklenmeler veya azalmalar saptanmıştır.MALDI-TOF-MS analizi öncesi değişik molekül ağırlığındaki krom içeren biyomoleküllere ait fraksiyonlar toplanmış ve doğal ve denature edici (üre ve DTT varlığında) şartlarda tripsin enzimi yardımıyla parçalanarak Cr-peptid kompleksleri elde edilmiştir. Daha küçük aralıktaki boyutça eleme kolonu (Superdex Peptide) kullanılarak kompleksler izole edilmiş ancak ilgili proteinlerin tanımlanması yapılamamıştır. Örnek hazırlama ve analitik ölçümlere ilişkin deneysel işlemler optimize edilmiştir. In the presented study, concentrations of chromium, copper, iron, manganese and zinc were determined in liver, kidney, brain, lung, heart and testis of mice upon intraperitoneal injection of a single dose hexavalent chromium (Cr(VI)). As results, comparing to control subject, chromium concentrations increased ca. 40-fold in liver and kidney and by a factor of 3-5 in all the other tissues. The homeostasis of Cu, Fe, Mn and Zn was also affected.The distribution of essential trace elements between different molecular weight biomolecules was evaluated in the cytosols of the different mouse organs by size-exclusion chromatography (Superdex-75 column) with UV-VIS and ICP-MS detection. The administration of Cr(VI) resulted in differences in the elution profiles of Fe, Mn, Cu and Zn-protein complexes. Bioinduced Mn, Fe and Zn-binding proteins could be detected in some tissues, especially in liver and kidney.Different molecular weight fractions containing chromium were heartcut and submitted to tryptic digestion prior to MALDI-TOF-MS analysis. Cr-peptide complexes could be recovered both in non-denaturing and in denaturing (in the presence of urea and DTT) conditions. They were isolated by size-exclusion chromatography with a smaller separation range (Superdex Peptide) but could not be identified. Experimental parameters in both sample preparation and analytical measurement were optimized and discussed through the analytical procedures. 165

Sunulan bu tezde, Au(lll), Pt(II), Pd(II) ve Ag(I) iyonlarının sulu çözeltiler ve anot çamuru, atık fotoğraf geliştirme çözeltisi, deniz suyu ve çeşme suyu gibi doğal örneklerden ayrılması, deriştirilmesi, tayini ve geri kazanımında seçici bir sorbent olarak poli(N-Hidroksimetilmetakrilamid-Alliltiyoüre), [poli(NHMA-AT)] hidrojellerinin sentezi ve karakterizasyonları sunulmuştur. Poli(NHMA-AT) hidrojelleri, ışın polimerizayonu tekniği ile 60Co-gama kaynağında sentezlenip, hidrojellerin karakterizasyonları, suda şişme özellikleri, FT-IR spektrumLarı, elementel analizleri, TGA termogramLarı alınarak yapılmıştır. Hidrojellerin kıymetli metal iyonları için gösterdiği adsorpsiyon ve desorpsiyon özelliklerine, pH etkisi, başlangıç metal iyon derişimi, adsorpsiyon denge zamanı, hidrojelin fiziksel şekli, adsorpsiyon ve desorpsiyon hacimLeri gibi çeşitli faktörlerin etkisi araştırılmıştır. Au(lll), Pt(ll), Pd(ll) (doygun çözeltisi değil, 750 ppm başlangıç iyon derişiminde) ve Ag(l) iyonları için sorbentin maksimum adsorpsiyon kapasitesi sırasıyla 651,39, 476,89, 405,16 ve 689,11 mg g-1 veya 3,37, 2,45, 3,81 and 6,39 mmol g-1 olarak bulunmuştur. Kıymetli metal iyonlarının sorbentten desorpsiyonu için 3 M HCI içinde 0,8 M tiyoüre çözeltisi kullanılmıştır ve desorpsiyon yüzdeleri % 97'den büyük olarak tespit edilmiştir. Yinelenen adsorpsiyon-desorpsiyon işlemLeri sorbentin en az 5 defalık kullanım için uygunluğunu göstermiştir. Deniz suyu örneklerinde altın derişimi, İzmit Körfezinde 15.83 ± 2.64 ng L-1, Seferihisar'da ise 30.48 ± 3.81 ng L-1 olarak bulunmuştur. Anot çamuru çözeltilerinde, 1 193.9 ± 4.9 ug.L1 Au(lll), 42.4 ± 9.3 jag.L`1 Pt(ll) ve 3.2 ± 0.2 jıg.L`1 Pd(ll) tayin edilmiştir. Atık fotoğraf geliştirme çözeltisinin altın ve platin içerikleri ölçülmüş ve sonuç olarak 285,9 ± 1,1 jıg.L`1 Au(lll) ve 31.5 ± 1.3 ng.L`1 Pt(ll) bulunmuştur. Yöntemin doğruluğu, atık fotoğraf geliştirme çözeltisine eklenen bilinen miktarlarda standartlar kullanılarak test edilmiştir. In the submitted study, poly(N-Hydroxymethylmethacrylamide-Allylthiourea) hydrogels [poly(NHMA-AT)] were prepared as a specific sorbent for separation, peconcentration, recovery and also determination of uitratrace amounts of Au(IIi), Pt(li), Pd(li) and Ag(l) ions from aquatic systems and natural samples such as anode slime, residual photographic developer solutions, seawater samples and tap water, in batch system. Poly(NHMA-AT) hydrogels were produced by irradiation polymerization technique using 60Co-gamma irradiation source, for characterization of the hydrogels, water uptake properties, FT-IR spectras, elemental analyses and TGA thermograms were examined. The effect of various experimental conditions such as the effect of pH, initial precious metal ion concentration, adsorption equilibrium time, physical shape of the sorbent, adsorption and desorption volume on the adsorption and desorption properties of the hydrogels for precious metal ions were investigated. Maximum adsorpiton capacities of the hydrogel were found to be 651.39, 476.89, 405.16 and 689.1 1 mg g-1 or 3.37, 2.45, 3.81 and 6.39 mmol g-1 for Au(III), Pt(ll), Pd(II) and Ag(l) Ions, respectively. Desorption of the precious metal ions were examined using a 0.8 M thiourea in 3 M HCI solution. Desorption ratios are greater than 97% for all studied precious metal ions. Adsorption-desorption cycles showed the feasibility of repeated use at least 5 times of the hydrogel. In seawater samples, gold concentration was found as 15.83 ± 2.64 ng L-1 in İzmit Gulf and 30.48 ± 3.81 ng L-1 in Seferihisar/lzmir. In anode slime solution, 1193.9 ± 4.9 ug L-1 for Au(lll), 42.4 ± 9.3 ug L-1 for Pt(ll), and 3.2 ± 0.2 ug L-1 for Pd(ll) were detected. Gold content of the residual photographic fixer solution was found 285.9 ±1.1 ug L-1, and platinum content was found as 31.5 ± 1.3 ug L-1 for studied sample. The accuracy of the system was checked with residual photographic developer solution adding known amount of Pt(ll) spike. Sunulan tezde balık örneklerinde cıva türlemesine yönelik özgün bir analitik yöntem geliştirilmiştir. Cıva türlerinin özütlenmesi yumuşak şartlar altında (60 °C ve pH 2,0) ve 2 dakika içerisinde başarılmıştır. Türlerin ayrılması ise yüksek performanslı sıvı kromatografi (HPLC) sisteminin indüktif eşleşmiş plazma kütle spektrometresi (ICP-MS) ile hibritleştirilmesiyle gerçekleştirilmiştir. Organik bileşence zengin mobil faz kullanımı ve plazmaya oksijen ilavesi türlerin 4 dakika içerisinde ayrılabilmesine olanak tanımıştır. Metodun gözlenebilme sınırı inorganik cıva ve metilcıva için sırasıyla 0,2 and 0,1 ng/g olarak hesaplanmıştır. Metot, sertifikalı referans örnekler ve Karadeniz kaynaklı bazı balık örneklerine başarı ile uygulanmıştır. In the proposed study a novel analytical procedure was developed for extraction and speciation of mercury in fish samples. The method consists of the recovery of mercury species by microwave assisted extraction at mild conditions (60 °C and pH 2.0) in 2 min. Species separation was accomplished by hyphenation of high performance liquid chromatography (HPLC) to inductively coupled plasma mass spectrometry (ICP-MS). The use of organic-rich mobile phase and the oxygen addition into plasma allowed species separation in 4 min. Limit of detection for the method was calculated as 0.2 and 0.1 ng/g for inorganic mercury and methylmercury, respectively. The method was successfully applied to certified reference materials and some commercialized fish samples from Black Sea. 48

Tuz (NaCl) en çok tüketilen gıda katkı maddeleri arasında yer alan ve aynı zamanda tıp, boyacılık, tekstil v.b. endüstrilerde kullanılmaktadır. Marketlerdeki tuz örnekleri kaynağına (deniz, göl ya da madencilik) ve üretim süreçlerine bağlı olarak farklı miktarlarda metalleri içerebilir. Eser elementler fayda ve zararları açısından biyolojik süreçlerde hayati bir rol oynamaktadır. Nikel gibi bazı elementlerin eser düzeyde dahi insan sağlığı üzerindeki toksik etkileri iyi bilinmektedir. Diğer taraftan, çinko, bakır, demir gibi normal vücut fonksiyonları için gerekli olan metallerin yüksek derişimlerde toksik etkileri bulunmaktadır. İnsan beslenmesindeki önemi açısından tuz örnekleri gıda güvenliği ve kalite kontrolünün sağlanması için rutin olarak analiz edilmelidir. İndüktif eşleşmiş plazma-kütle spektrometrisi (ICP-MS), birçok örnek türünde eser element analizlerinde yüksek duyarlılık, nispeten bileşik bağımsız sinyal oluşturması, geniş doğrusal aralık ve çoklu element analizine uygunluğu nedeniyle tercih edilen bir yöntem olmuştur. Ancak, ICP-MS konileri, lensleri üzerinde tuz birikimi ve plazma içinde oluşmuş çok atomlu türlerden kaynaklı girişimler, tuz çözeltilerinin rutin analizi için bir sorundur. Ayrıca, argon plazma içinde Na ve Cl iyonlarının yüksek bolluğu analitlerin iyonizasyonunu baskılamaktadır. Bu tez yüksek tuz ortamında aerosol seyrelme tekniği uygulanarak eser elementlerin pratik tayinine yönelik bazı çalışmalar içermektedir. Yüksek matriksli örnek çözeltilerin doğrudan analizi sağlayan bu yaklaşımda; örnek aerosol oluşumunu azaltmak için taşıyıcı gaz akışı azaltılır, plazma girişinden önce seyrelme gazı ile örnek aerosolü seyreltilir. Salt (NaCl) is among the most widely consumed food additives and is also used in medicine, dyeing, textile industry etc. Salt samples in the market may contain diverse amounts of metals, depending on its source (sea, lake or mining) and production processes. Trace elements play vital role in biological processes in terms of their essentiality and/or toxicity. Toxic effects of some elements such as nickel on human health are well-known even at trace levels. On the other hand, metals such as zinc, copper and iron which are essential for normal body functions exert toxic effects at high concentrations. Since its importance in human nutrition, salt samples should be routinely analyzed to assure food safety and quality control. Inductively Coupled Plasma-Mass Spectrometry (ICP-MS) has been a method of choice for trace element analysis in many sample types because of its high detection power, relatively compound independent signal response, wide linear range, and also multi element detection capability. However, the interferences from polyatomic species formed in plasma and salt deposition on cones and lens of ICP-MS is a problem for routine analysis of salty solutions. Moreover, high abundance of Na and Cl ions suppresses the ionization of analytes in the argon plasma. This thesis comprises some studies for rapid determination of trace elements in high salt media applying aerosol dilution technique. In this approach, the carrier gas flow is decreased to reduce the formation of sample aerosol which is further diluted by dilution gas prior to its introduction into plasma, allowing direct analysis of high-matrix sample solutions. 50
