20. Nematicidal activity of a hydroalcoholic extract of the edible mushroom Neolentinus ponderosus on L3 larvae of Haemonchus contortus.

Abstract

Objective: In the present study, the in vitro and in vivo effect of the hydroalcoholic extract of Neolentinus ponderosus (EHNP) on L3 larvae of Haemonchus contortus was evaluated. Material and methods: The N. ponderosus fungus was cultivated in potato dextrose liquid medium for 7 days at 120 rpm and 25 °C. Subsequently, the EHNP was obtained; in vitro bioassays were performed in 96-well plates. Furthermore, an in vitro confrontation with different concentrations of EHNP was carried out at 72 h against L3 larvae of H. contortus. The controls used were distilled water and ivermectin at 5 mg/mL. Subsequently, the in vivo activity of EHNP was evaluated using the gerbil against H. contortus L3 larvae as an experimental model. The experimental design consisted of four groups with: (1) distilled water, (2) fenbendazole at 20 mg/kg of body weight, (3) EHNP at a dose of 81 µg/mL, and (4) EHNP at a dose of 40 µg/mL. Results: In vitro study showed 97% mortality of the parasite H. contortus at a concentration of 3.4 mg/mL and a lethal concentration (LC90) of 2 mg/mL EHNP. In the in vivo assessment the highest mortality was (49%) at 72 h at a concentration of 81 µg/mL bw. Conclusion: The result of the present study shows that EHNP has nematicidal activity in vitro and in vivo tests (close to 97% and 50%, respectively), the fungus N. ponderosus should be considered in future tests to elucidate the secondary metabolites through spectroscopic studies. Abstract

This study was focused on the application of an aerobic biofiltration (BF) with Ficus benjamina wood chips as support medium, inoculated with two basidiomycete fungi, Phanerochaete chrysosporium (BF 1) and Trametes versicolor (BF 2), to treat Tequila vinasses from a Tequila industry. The biofiltration system was compared with a biofilter system without basidiomycete fungi (BF W), in order to determine the influence of fungi on the treatment of vinasses. Three different vinasses/water ratios (30/70, 40/60, and 50/50) were evaluated. The maximum removals of chemical oxygen demand (COD) obtained during each operation step were 72% (BF 1), 72% (BF 2), and 8% (BF W) for 30 vinasses/70 water; 72% (BF 1), 73% (BF 2), and 66% (BF W) for 40 vinasses/60 water; and 22% (BF 1), 20% (BF 2), and 18% (BF W) for 50 vinasses/50 water. The total organic carbon (TOC) removal was significantly increased using a volumetric organic load of 5.5 kg COD m−3d−1. During the operation of the biofilters, the enzymatic activity of laccase was present, even at the step of highest concentration of vinasses.

Abstract

Background: Enzymatic activity and laccase isoenzymes number of Pleurotus ostreatus grown in different pH values of the growing medium in submer ged fermentation and incubated in buffer solutions of different initial pH values were determined. The expression profiles of five laccase genes (Lacc1, Lacc4, Lacc6, Lacc9 and Lacc10) in these cultures w ere also studied. Results: The highest laccases activity was obtained in cult ures grown at initial pH of 4.5 and the lowest in cultures grown at initial pH of 8.5. Isoe nzyme profiles were different in all the cases. Lac c1, Lacc4, Lacc6 and Lacc10 were expressed in all the c ultures. Conclusions: The initial pH of the growing medium is an importa nt factor for regulating the expression of laccase genes, having an effect on the activity and on the laccase isoenzymes number produced by Postreatus in SmF. This is the first report on the influence of different initial pH values of the growing medium on the laccases activity, laccase isoenzymes number and laccases expression profiles of Postreatus grown in submerged fermentation.

Abstract

The heterologous expression of the gene LACP83 (encoding a laccase) from Pleurotus ostreatus in Escherichia coli was characterized. The laccase enzyme activity and kinetics of bacterial growth with an inducer (IPTG) and without inducer were determined. The maximum enzymatic activity was observed at 7 h post induction with a value of 3740 ± 342 U/L, which was similar to that reported for the native strain of P. ostreatus at 144 h of culture. Furthermore, the induction of laccase with IPTG reduced the specific growth rate of recombinant E. coli BL21 by approximately 50%. These results support the use this system for the recombinant production of the enzyme on an industrial scale. [ABSTRACT FROM AUTHOR]

Abstract

False smut of rice, caused by Ustilaginoidea virens (Cooke) Takahashi (teleomorph: Villosiclava virens), is one of the most important diseases affecting rice worldwide. Agrobacterium tumefaciens-mediated transformation was used to identify functional genes in U. virens. In this study, we selected a single-copy insertion mutant T133 with deficiency in producing conidia by screening the T-DNA insertion mutant library of U. virens. The UvPRO1-deletion mutant was successfully obtained after cloning the targeted gene by analysis of the T-DNA insert site of mutant T133. Further research showed that the UvPRO1 mutant was reduced in growth rate and could not produce conidia in PSB medium, while sensitivities to sodium dodecyl sulfate, Congo red, and hyperosmotic stress increased. Moreover, the UvPRO1 deletion mutant hyphae could extend along the surface of spikelets at 1-3 dpi, but mycelia became shriveled and completely lost the ability to infect spikelets at 4 dpi. The relative expression level of UvPRO1 at 8 dpi was more than twice as high as that at 1-2 dpi. These results suggest that UvPRO1 plays a critical role in hyphal growth and conidiation, as well as in stress response and pathogenesis. These findings provide a novel mode of action for the PRO1 protein in fungi and improve the understanding of the function of UvPRO1 in the life cycle of U. virens. [ABSTRACT FROM AUTHOR]

Abstract

Biocompounds present in various edible fungi of the basidiomycetes family such as Hericium erinaceus, Lentinula edodes and Pleurotus eryngii have lead to a great interest because they have been reported to have beneficial properties for health. There are various methods for the extraction of biocompounds, in which the majority is needed the use of thermal methods which affect the action of biocompounds, the objective of this work was the application of the process of the physical method of sonication, a friendly method with the environment, the amount of total soluble solids and its relationship with the antioxidant, antimicrobiological action against Escherichia coli and Staphylococcus aureus and laccase enzymatic. For the extraction of the sample, the dehydrated fruiting fungus was used after sonication and removal of solvents, the antioxidant capacity was evaluated by colorimetric methods: DPPH and ABTS.
