2. Optimizing Drug Design by Merging Generative AI With Active Learning Frameworks

Abstract

Traditional drug discovery programs are being transformed by the advent of machine learning methods. Among these, Generative AI methods (GM) have gained attention due to their ability to design new molecules and enhance specific properties of existing ones. However, current GM methods have limitations, such as low affinity towards the target, unknown ADME/PK properties, or the lack of synthetic tractability. To improve the applicability domain of GM methods, we have developed a workflow based on a variational autoencoder coupled with active learning steps. The designed GM workflow iteratively learns from molecular metrics, including drug likeliness, synthesizability, similarity, and docking scores. In addition, we also included a hierarchical set of criteria based on advanced molecular modeling simulations during a final selection step. We tested our GM workflow on two model systems, CDK2 and KRAS. In both cases, our model generated chemically viable molecules with a high predicted affinity toward the targets. Particularly, the proportion of high-affinity molecules inferred by our GM workflow was significantly greater than that in the training data. Notably, we also uncovered novel scaffolds significantly dissimilar to those known for each target. These results highlight the potential of our GM workflow to explore novel chemical space for specific targets, thereby opening up new possibilities for drug discovery endeavors.

4. A Framework for Meta-Learning in Science Education for a Time of Crisis and Beyond

Abstract

Science education has an important role in educating the public on learning strategies that will generate a scientific literate population. The challenges encountered in this time of crisis calls for individuals to make well-informed decisions, based on reliable information. Understanding scientific basic concepts can inform the population on making informed decisions that will protect and prosper their communities. This study applied a grounded theory approach to propose a framework for meta-learning as a strategy that enhances science understanding and cultivates trust toward science. Meta-learning in science education is contextualized during a time of crisis and four stages are suggested for the meta-learning process. In the first stage, the learner becomes aware of a situation and activates prior knowledge. In the second stage, the learner searches and evaluates reliable information. In the third stage, the learner adjusts their behavior based on the new knowledge. Finally, in the fourth stage, the learner embraces learning as an everlasting process and re-adjusts behavior accordingly. Through meta-learning practices in science education, learners can claim agency of their learning processes and embrace a lifelong learning endeavor that will benefit themselves and those around them.

8. A Textbook Analysis to Uncover the Hidden Contributors of Science and Mathematics

Abstract

Science and mathematics are universal endeavors. However, the historical perspective of science and mathematics has been westernized across the curriculum in the USA. There is a limited number of textbook analyses centered on exploring the representation of science and mathematics under a historical standpoint. This research seeks to discuss the dominated historical viewpoints presented in the textbooks and uncover the untold historical perspectives of science and mathematics contributors from non-western regions. A discourse analysis of eleven science and mathematics textbooks published over the course of 10 years was examined under an ethnocentric and cultural imperialistic lens with a focus on the hidden curriculum. The findings reflect that science and math textbooks continue to exhibit male Eurocentric representations. This research offers counter-hegemonic discourses in the history of science and mathematics not present in the analyzed textbooks with the purpose of diversifying the scientific and mathematic historical content knowledge. By offering teachers and students with diverse perspectives of the history of science and mathematics, the curriculum becomes enriched with accurate knowledge that serves and represents all learners.

Abstract

Although active learning is well-known to positively impact student academic achievement, there is limited research on classroom behavior, interaction, and engagement in active learning settings. This research applied an explanatory case study method in order to describe students' behaviors, interactions, and engagement in an active learning classroom (ALC). Students' behaviors and interactions were categorized according to active learning teaching strategies. Findings reveal students exhibited positive engagement with the course content through the exposure of multiple active learning opportunities; however, engagement with the active learning teaching strategy varied. The findings suggest factors that contribute to the effectiveness of ALCs.

Abstract

Due to the COVID-19 pandemic, Biology Teaching Assistants (TAs) were tasked with transitioning and adapting their instruction to an online environment by quickly implementing Emergency Remote Teaching (ERT) practices. Effective online and in-person teaching requires student-centered approaches to support undergraduate student learning. Using interviews and the Approaches to Teaching Inventory (ATI), a case study was conducted to explore the impact on TAs approaches to teaching during the transition to an online emergency remote environment, and to identify areas where TAs need further support through Teaching Professional Development (TPD). The findings revealed themes regarding challenges in the ERT context, such as decreased active learning opportunities, decreased office hours attendance, decreased student engagement, and more time spent on teaching tasks. Our work provides educational researchers and practitioners with key aspects that can improve TPD for online teaching and learning. [ABSTRACT FROM AUTHOR]

Abstract

Lattice structures have become an innovative solution for the improvement of part design, as they are able to substitute solid regions, maintain mechanical capabilities, and reduce material usage; however, dimensional quality control of these geometries is challenging. X-ray computed tomography (XCT) is the most suitable non-destructive metrological technique as it is capable of characterizing internal features and hidden elements. Uncertainty estimation of XCT is still in development, and studies typically use high-resolution calibrated devices such as focal variation microscopes (FVMs) as a reference, focusing on certain parts of the lattice but not the whole structure. In this paper, an estimation of the accuracy of XCT evaluation of a complete lattice structure in comparison to a higher-resolution reference device (FVM) is presented. Experimental measurements are taken on ad hoc designed test objects manufactured in polyamide 12 (PA12) using selective laser sintering (SLS), optimized for the evaluation on both instruments using different cubic-based lattice typologies. The results confirm higher precision on XCT evaluation in both qualitative and quantitative analysis. Even with a lower resolution, XCT is able to characterize details of the surface such as re-entrant features; as well, standard deviations and uncertainties in strut diameter evaluation remain more stable in all cells in XCT, identifying on the other hand reconstruction problems on FVM measurements. Moreover, it is shown that, using XCT, no additional evaluation errors were found in inner cells, suggesting that the measurement of external elements could be representative of the whole structure for metrological purposes. Abstract

The use of metaproteomics for studying the human gut microbiota can shed light on the taxonomic profile and the functional role of the microbial community. Nevertheless, methods for extracting proteins from stool samples continue to evolve, in the pursuit of optimal protocols for moistening and dispersing the stool sample and for disrupting microbial cells, which are two critical steps for ensuring good protein recovery. Here, we evaluated different stool sample processing (SSP) and microbial cell disruption methods (CDMs). The combination of a longer disintegration period of the stool sample in a tube rotator with sonication increased the overall number of identified peptides and proteins. Proteobacteria, Bacteroidetes, Planctomycetes, and Euryarchaeota identification was favored by mechanical cell disruption with glass beads. In contrast, the relative abundance of Firmicutes, Actinobacteria, and Fusobacteria was improved when sonication was performed before bead beating. Tenericutes and Apicomplexa identification was enhanced by moistening the stool samples during processing and by disrupting cells with medium-sized glass beads combined with or without sonication. Human protein identifications were affected by sonication. To test the reproducibility of these gut metaproteomic analyses, we examined samples from six healthy individuals using a protocol that had shown a good taxonomic diversity and identification of proteins from Proteobacteria and humans. We also detected proteins involved in microbial functions relevant to the host and related mostly to specific taxa, such as B12 biosynthesis and short chain fatty acid (SCFA) production carried out mainly by members in the Prevotella genus and the Firmicutes phylum, respectively. Abstract

Macrophages are involved in the primary human response to Candida albicans. After pathogen recognition, signaling pathways are activated, leading to the production of cytokines, chemokines, and antimicrobial peptides. ATP binding proteins are crucial for this regulation. Here, a quantitative proteomic and phosphoproteomic approach was carried out for the study of human macrophage ATP-binding proteins after interaction with C. albicans. From a total of 547 nonredundant quantified proteins, 137 were ATP binding proteins and 59 were detected as differentially abundant. From the differentially abundant ATP-binding proteins, 6 were kinases (MAP2K2, SYK, STK3, MAP3K2, NDKA, and SRPK1), most of them involved in signaling pathways. Furthermore, 85 phosphopeptides were quantified. Macrophage proteomic alterations including an increase of protein synthesis with a consistent decrease in proteolysis were observed. Besides, macrophages showed changes in proteins of endosomal trafficking together with mitochondrial proteins, including some involved in the response to oxidative stress. Regarding cell death mechanisms, an increase of antiapoptotic over pro-apoptotic signals is suggested. Furthermore, a high pro-inflammatory response was detected, together with no upregulation of key mi-RNAs involved in the negative feedback of this response. Abstract

Introducción: El coronavirus SARS-CoV-2 fue propagándose por todo el mundo, declarando la OMS la pandemia en marzo de 2020. Para los profesionales de enfermería, así como el resto de los sanitarios, supuso afrontar situaciones de sobrecarga laboral, de riesgo de contagio o de cuestiones éticas, todo ello basado en un virus del cual había poca información. Dichas circunstancias causaron repercusiones sobre la vida de los profesionales de la salud, tanto en el ámbito de su salud como en el ámbito laboral. Por ello es importante examinar las consecuencias de la pandemia sobre la salud de los trabajadores de enfermería y los factores asociados. Objetivos: Analizar el impacto que tuvo la pandemia sobre los profesionales de enfermería, así como los factores de riesgo y de protección asociados Material y Métodos: Se elaboró una revisión bibliográfica mediante la búsqueda de artículos en diferentes bases de datos científicas. Después de realizar una lectura crítica de los artículos, se seleccionaron 17 publicaciones. Resultados y discusión: Los resultados obtenidos a partir de los artículos seleccionados describían los principales signos y síntomas, los factores de riesgo asociados a las alteraciones de la salud mental y la capacidad de resiliencia sobre los profesionales de enfermería durante la pandemia. Conclusión: Los profesionales de enfermería sufrieron un aumento de la prevalencia de alteraciones de la salud mental, como la ansiedad, la depresión, el trastorno de estrés postraumático o el Síndrome de Burnout. En caso de que en un futuro se pudiera sufrir la misma situación, se ve necesario el desarrollo de medidas preventivas adecuadas para el personal de enfermería con el objetivo de que estén formados adecuadamente., Grado en Enfermería

Abstract

L'acte de caminar, una de les accions més ancestrals i més invariables de l'ésser humà, ha experimentat, en canvi, una transformació notable en el seu significat, rellevància social i impacte a les ciutats. En èpoques passades, caminar era l'única opció de transport disponible, però actualment també s'ha convertit en una elecció conscient. Al llarg de la història, la presència del vianant a la ciutat ha patit canvis significatius. En temps antics, caminar era la forma dominant de desplaçar-se, però l'auge del transport motoritzat ha limitat la seva presència a les urbs. No obstant això, actualment hi ha un renovat interès per recuperar i promoure la participació activa del vianant, fomentant el caminar com una opció que és sostenible i beneficia la salut. Aquells que trien caminar tenen més control sobre el seu temps i gaudeixen de la llibertat d'aturar-se i apreciar-ne l'entorn. Sin embargo, en la actualidad, existe un renovado interés por recuperar y promover la participación activa del peatón, fomentando el caminar como una opción que es sostenible y beneficia la salud. Aquellos que eligen caminar tienen un mayor control sobre su tiempo y disfrutan de la libertad de detenerse y apreciar su entorno. 42. Comparative proteomic study of Edwardsiella tarda strains with different degrees of virulence.

Abstract

Edwardsiella tarda is an enteric opportunistic pathogen that causes a great loss in aquaculture. This species has been described as a phenotypical homogeneous group; in contrast, serological studies and molecular typing revealed a wide heterogeneity. In this work, a proteomic study of differential expression of a virulent isolate from turbot cultured in the Norwest of Spain in comparison with an avirulent collection strain was performed in order to recognize proteins involved in virulence. One hundred and three proteins that presented different abundance were successfully identified and classified into 11 functional categories according to their biological processes: amino acid, carbohydrate and lipid metabolism, tricarboxylic cycle, stress response and protein fate, protein synthesis, biogenesis of cellular components, cell rescue defence and virulence, cell membrane and transport, signal transduction and purine and pyrimidine metabolism. Twenty three protein spots detected only in turbot isolate were identified. It was shown that the same proteins appeared in different spots in the two isolates. Mass spectra obtained by MALDITOF/TOF of some of these proteins and DNA sequencing explained the changes as a result of different amino acid sequences. Source data are provided with this paper, p38α is a versatile protein kinase that can control numerous processes and plays important roles in the cellular responses to stress. Dysregulation of p38α signaling has been linked to several diseases including inflammation, immune disorders and cancer, suggesting that targeting p38α could be therapeutically beneficial. Over the last two decades, numerous p38α inhibitors have been developed, which showed promising effects in pre-clinical studies but results from clinical trials have been disappointing, fueling the interest in the generation of alternative mechanisms of p38α modulation. Here, we report the in silico identification of compounds that we refer to as non-canonical p38α inhibitors (NC-p38i). By combining biochemical and structural analyses, we show that NC-p38i efficiently inhibit p38α autophosphorylation but weakly affect the activity of the canonical pathway.
