Norfloxacin is an antimicrobial in treating urinary tract infections with low water solubility. This study aims to know the effect of norfloxacin-urea co-crystal formation on the solubility of norfloxacin and the potential for scale-up when prepared by ultrasound-assisted slurry co-crystallization method. Identification of the screening result of the norfloxacin-urea (1 : 1) co-crystal formation by a wet grinding method using an ethanol-acetone (1 : 1) solvent mixture was performed by powder X-ray diffractometer (PXRD). The ultrasound-assisted slurry co-crystallization method was used for co-crystal formation with five-fold the weight of norfloxacin and urea than the wet grinding method. The co-crystal product prepared by the ultrasound-assisted slurry co-crystallization method was observed for its crystal morphology and characterized by PXRD and differential scanning calorimeter (DSC). Solubility and dissolution tests in water and acetate buffer solution pH 4.0 were used to evaluate the physicochemical properties. Identification of co-crystal screening by PXRD revealed the formation of norfloxacin-urea co-crystal. The PXRD pattern of the norfloxacin-urea co-crystal product prepared by the ultrasound-assisted slurry co-crystallization method was similar to the wet grinding method. Norfloxacin-urea co-crystal has a different melting point and crystal morphology from pure norfloxacin and urea. The solubility and dissolution rate of norfloxacin-urea co-crystal was higher in water and not significantly different in acetate buffer solution pH 4.0 compared to pure norfloxacin. This study showed that the norfloxacin-urea co-crystal formation could enhance the solubility of norfloxacin in water and had the potential for scale-up when prepared using the ultrasound-assisted slurry co-crystallization method.

Cette contribution interroge la mise en tourisme du Guizhou, en Chine du Sud, à travers le cas de trois villages de la minorité ethnique shui. La promotion de ces villages au rang de sites touristiques officiels suit un modèle d’aménagement standardisé, ayant pour double objectif le développement économique et la construction d’une identité nationale. Elle implique aussi des transformations souvent importantes du paysage quotidien des populations. Pris entre les actions développementalistes de l’État et les stratégies aménagistes locales, les paysages constituent un véritable enjeu : au final, à qui appartiennent les paysages de ces villages shui ? Après la présentation de trois situations contrastées, les représentations qu’ont les habitants des transformations de leur paysage sont analysées. Celles-ci révèlent l’évolution de leur regard et leur adhésion au modèle dans ses principes d’aménagement. Mais des tensions émergent, ce qui témoigne du faible pouvoir dont disposent les populations dans le processus économique et politique de développement touristique.

Oil palm and its pollinators are native to Africa. Notwithstanding, it has been extensively cultivated outside its native range, so also has its pollinators been acquired and introduced. A concern, however is that most documentation on oil palm and its pollinators are non-indigenous, with little information of African origin commenting on their activities in Africa. Thus, a study on the insect pollinators of oil palm was carried out on two fruit forms: Dura and Tenera, (ages ten and seven, respectively), in Ghana. The aim was to understand how average monthly population density of the pollinators relates to one another, and to two bunch parameters (fruitset and fruit-to-bunch ratio). Nine species (all in the order Coleoptera) were consistently seen on the spikelets of anthesising male inflorescences, and eleven species on receptive female inflorescences. Six were of the Elaeidobius genus: E. kamerunicus, E. plagiatus, E. singularis, E. subvittatus, E. bilineatus, and Elaeidobius sp. 1 (unidentified species); two each were of the Microporum genus: M. congolenses, M. dispar, and the Prosoestus genus: P. scuptilis, P. minor; while one species was of either Atheta or Forficula. The pollinators were observed to be positively related, and to varying degree of significance. Fruitset level was acceptably high, indicating adequate pollinator population and efficient pollination. There was a positive relationship between population density of the pollinators and fruitset, and a negative relationship between the bunch parameters. A number of other insects were seldom observed around the male and female inflorescences; their role could not be defined.

We describe the 2-year follow-up of an open-label trial (CT-AMT-011-01) of AAV1-LPLS447X gene therapy for lipoprotein lipase (LPL) deficiency (LPLD), an orphan disease associated with chylomicronemia, severe hypertriglyceridemia, metabolic complications and potentially life-threatening pancreatitis. The LPLS447X gene variant, in an adeno-associated viral vector of serotype 1 (alipogene tiparvovec), was administered to 14 adult LPLD patients with a prior history of pancreatitis. Primary objectives were to assess the long-term safety of alipogene tiparvovec and achieve a 40% reduction in fasting median plasma triglyceride (TG) at 3-12 weeks compared with baseline. Cohorts 1 (n=2) and 2 (n=4) received 3 × 1011 gc kg−1, and cohort 3 (n=8) received 1 × 1012 gc kg−1. Cohorts 2 and 3 also received immunosuppressants from the time of alipogene tiparvovec administration and continued for 12 weeks. Alipogene tiparvovec was well tolerated, without emerging safety concerns for 2 years. Half of the patients demonstrated a 40% reduction in fasting TG between 3 and 12 weeks. TG subsequently returned to baseline, although sustained LPLS447X expression and long-term changes in TG-rich lipoprotein characteristics were noted independently of the effect on fasting plasma TG. The 271 000 km² Lake Athabasca drainage in Northern Canada encompasses ecologically-rich and sensitive ecosystems, intensive agricultural lands, vast forests, glacier-clad mountains, and abundant oil reserves in the form of tar-sands. In this study, streamflow variability and trends in eight rivers feeding the 7800 km² Lake Athabasca are investigated over the period 1960-2010. Hydrological regimes and trends are established using a robust regime shift detection method and the Mann-Kendall (MK) test, respectively. Results show that the Athabasca River, which provides ~57% of the total annual lake inflow of 34.06 km³ yr-1, experiences marked declines in recent decades impacting lake levels and its ecosystem. The Fond du Lac River, which contributes ~ 30% of total Lake Athabasca inflow, has an increasing trend of 0.021 km³ yr-1 over 1970-2010 according to the MK test, equating to a 0.86 km³ discharge increase from Fond du Lac River to the lake. From 1960 to 2010 there has been approximately a 21.2% reduction of average discharge equivalent to a 7.22 km³ recession in the Lake Athabasca causing lake levels to drop. The lake level has a trend of -0.008myr-1 which is equivalent to a 0.39m decline in the lake level over 1960-2010. The total lake inflow trend over 1977-2010 is -0.207 km³ yr-1 or a reduction of 25.67 km³ by 2100 by linear extrapolation. This may imply a further reduction of 2m to 3m in lake level that is in the range of a 5200-yr historical minimum inferred from proxy data in nearby sediment cores. [ABSTRACT FROM AUTHOR]

Summary In Nunavik, common practices and food habits such as consumption of raw meat and untreated water place the Inuit at risk for contracting zoonotic diseases. The aim of this study was to determine the seroprevalence of seven zoonotic infections among the permanent residents of Nunavik. The study was conducted in the fall 2004 as part of the Nunavik Health Survey. Blood samples from adults aged 18-74 years ( n = 917) were collected and analysed for the presence of antibodies against Trichinella spp., Toxocara canis, Echinococcus granulosus, Brucella spp., Coxiella burnetii, Leptospira spp. and Francisella tularensis. Information on sociodemographic characteristics, traditional activities, drinking water supply and nutrition was gathered using english/inuktitut bilingual questionnaires. The chi-squared test was used to evaluate associations between seropositivity and other measured variables. Statistically significant variables were included in a multivariate logistic regression model to control for confounding factors. Estimated seroprevalences were 8.3% for E. granulosus, 3.9% for T. canis, 5.9% for Leptospira spp. and 18.9% for F. tularensis. Seroprevalence was ≤1% for Trichinella spiralis, Brucella spp. and C. burnetii. For most infections, seropositivity tended to increase with age. In multivariate analyses, seroprevalence was positively (i.e. directly) associated with age and residence in the Ungava coast area for F. tularensis; age and residence in the Hudson coast area for T. canis; female gender, lower level of schooling and frequent cleaning of water reservoirs for E. granulosus. No risk factor for Leptospira spp. infection was identified. No associations were detected with regards to food habits or environmental exposures. A small but significant portion of the Nunavik population has serologic evidence of exposure to at least one of the pathogenic microorganisms investigated. As a result of their intimate contact with the land and their nutritional habits, the Inuit of Nunavik are considered to be at risk from zoonotic infections. To better understand the risk factors for Toxoplasma gondii infection, a serosurvey was conducted in Nunavik, Québec, in September 2004. A representative sample of the Inuit adult population of Nunavik participated in this cross-sectional study ( n = 917). Antibodies (IgG) against T. gondii were detected by immunoassay. Information on sociodemographic characteristics, traditional activities, domestic environment and nutrition was gathered by questionnaire and explored as variables explanatory of seropositive results. Associations found to be statistically significant in univariate analyses were assessed by multivariable logistic regression to control for confounding factors. Almost two thirds (59.8%) of the Inuit of Nunavik were found to be seropositive for T. gondii. In multivariate analyses, risk factors for seropositivity were: increasing age, gender (women > men), lower level of education, consumption of potentially contaminated water (determined by an index of risk from waterborne infections), frequent cleaning of water reservoirs, and consumption of seal meat and feathered game. There was some variation in seroprevalence between the Ungava Bay coast (52.3%) and the Hudson Bay coast (65.6%), the two main regions of Nunavik, but this variation was not significant in the multivariable logistic regression model. This cross-sectional study demonstrated high T. gondii seroprevalence in the Inuit population and revealed that age, gender, schooling and community of residence all influence serostatus in this population. Variables related to drinking water and food choices may also influence the risk of infection. These results raise important questions about T. gondii transmission in Nunavik including possible links between terrestrial and marine cycles. [ABSTRACT FROM AUTHOR]

A spatial filter (SF) is used to reduce cloud coverage in MODIS 8-day maximum snow cover extent products (MOD10A2) from 2000-2007 to assess the topographic control on snow cover fraction (SCF) and snow cover duration (SCD) in the Quesnel River Basin (QRB) of British Columbia, Canada. Results show that the SF reduces cloud coverage and improves by 2% the accuracy of snow mapping in the QRB. The SF shows larger SCF and longer SCD than MOD10A2, with higher altitudes experiencing longer snow cover and perennial snow above 2500 m. The gradient of SCF with elevation (d(SCF)/d(elevation)) during the snowmelt season is 8% (100 m)-1. The average melt rates of SCF are similar for different 100 m elevation bands at about 5.5% (8 days)-1 for altitudes <1500m with decreasing values with elevation to near 0% (8 days)-1 for altitudes >2500 m. Different combinations of slopes and aspects also affect the SCF with a maximum difference of 20.9% at a given time. Correlation coefficients between SCD and elevation attain 0.96 (p<0.001). Mean gradients of SCD with elevation are 3.8, 4.3, and 11.6 days (100 m)-1 for the snow onset, snowmelt, and entire year, respectively. The SF decreases the standard deviations of SCDs compared to MOD10A2 with a maximum difference near 0.63 days, 0.89 days, and 1.04 days for the snow onset, snowmelt and entire year, respectively. The Cape Saint Paul Wilt Disease (CSPWD), a lethal-yellowing type disease of coconut has been in Ghana since 1932. Aerial and/or ground surveys were undertaken to assess the current status of the disease spread. The survey showed that the spread of the disease for the past 5 years has mainly been the expansion of existing foci. However, new outbreaks were identified at Glidzi in the Volta, Bawjiase and Efutu Breman in Central regions. After the resurgence in the Volta region in 1995, the Woe-Tegbi-Dzelukope corridor has remained endemic, but less aggressive. Pockets of healthy groves remain along all the coastline and inland of known disease zones. Eradication of diseased palms at Ampain focus lying just about 60 km to the Ivorian border, and disease situations on new replanting with MYD × VTT hybrid are discussed.

The Lethal Yellowing (LY) disease is one of the main threats to coconut industry in many parts of Africa and the Caribbean. Planting resistant varieties has long been recognized as one of the most promising ways of controlling the disease. Considerable efforts have been devoted throughout the world to screening suitable varieties and have often involved international cooperation. It has proven to be a lengthy and difficult task. We present an overview of these efforts with special mention to Ghana, Jamaica and Mexico. Although no variety so far has been proven fully and permanently resistant, treating resistance level as a threshold trait makes it possible to demonstrate significant differences among varieties, which can be exploited effectively to make genetic improvement a component of an integrated control strategy. Based on past experience, we make a few suggestions to increase the diversity of resistance sources and increase the level and the sustainability of resistance to LY in coconut.

Lethal Yellowing disease locally called Cape Saint Paul wilt disease (CSPWD) is the bane of the coconut industry in Ghana and is caused by a phytoplasma. In Ghana, there are areas where the disease has re-infected re-plantings long after decimating all the palms in the area. This brings to the fore the possibility of alternate hosts in the spread of the disease because the pathogen is an obligate parasite. In this work, a number of plants were screened for their host status to the CSPWD pathogen. The presence of phytoplasmas in these plants was tested by polymerase chain reaction analysis using universal phytoplasma primers P1/P7 and CSPWD-specific primers G813/GAKSR. Although Desmodium adscendens tested positive to the CSPWD-specific primers, cloning and sequencing did not confirm it as an alternate host. The identification of alternate hosts will help us to evolve sound control strategies against the spread of the disease.

The Cape Saint Paul Wilt Disease (CSPWD) is a coconut lethal yellowing type disease (LY) and is the single most serious threat to coconut cultivation in Ghana. The recommended disease management strategy is the cultivation of disease-resistant coconut varieties. More than 38 varieties have been screened for their resistance to CSPWD since 1956 and the results are reviewed in this paper. Two varieties, Sri Lanka Green Dwarf (SGD) and Vanuatu Tall (VTT), have shown high resistance to the disease, and their hybrid (SGD × VTT) is under observation to determine its performance. A programme to rehabilitate the CSPWD-devastated areas was started in 1999. Emerging results indicate that the MYD × VTT hybrid being used for the programme, succumbs to the disease under intense disease pressure. A redirection of the rehabilitation programme and the screening of more varieties are recommended.

Epidemiological studies are of major importance in understanding the determinants of plant diseases in order to control the risks of their spreading. A research programme on the epidemiology of coconut lethal yellowing, or Cape Saint Paul Wilt Disease (CSPWD), in Ghana was launched in March 2007. The objective was to characterize the distribution and spread of the disease in space and time at various scales, and their relation with the environment. This article presents the general strategy used to evaluate the incidence of CSPWD along with the environmental, ecological and agronomical variables at regional level. A survey was undertaken on 1,166 plots of Coconut Sector Development Project (CSDP) planted with Malayan Yellow Dwarf (MYD) × Vanuatu Tall (VTT) hybrids in Western Region and Central Region. Preliminary results on the distribution of CSPWD and outside variables at regional scale, along with their relations, are given.

Abstract Background Arctic populations are exposed to mercury, lead and cadmium through their traditional diet. Studies have however shown that cadmium exposure is most often attributable to tobacco smoking. The aim of this study is to examine the trends in mercury, lead and cadmium exposure between 1992 and 2004 in the Inuit population of Nunavik (Northern Québec, Canada) using the data obtained from two broad scale health surveys, and to identify sources of exposure in 2004. Methods In 2004, 917 adults aged between 18 and 74 were recruited in the 14 communities of Nunavik to participate to a broad scale health survey. Blood samples were collected and analysed for metals by inductively coupled plasma mass spectrometry, and dietary and life-style characteristics were documented by questionnaires. Results were compared with data obtained in 1992, where 492 people were recruited for a similar survey in the same population. Results Mean blood concentration of mercury was 51.2 nmol/L, which represent a 32% decrease (p < 0.001) between 1992 and 2004. Mercury blood concentrations were mainly explained by age (partial r2 = 0.20; p < 0.0001), and the most important source of exposure to mercury was marine mammal meat consumption (partial r2 = 0.04; p < 0.0001). In 2004, mean blood concentration of lead was 0.19 μmol/L and showed a 55% decrease since 1992. No strong associations were observed with any dietary source, and lead concentrations were mainly explained by age (partial r2 = 0.20.; p < 0.001). Blood cadmium concentrations showed a 22% decrease (p < 0.001) between 1992 and 2004. Once stratified according to tobacco use, means varied between 5.3 nmol/L in never-smokers and 40.4 nmol/L in smokers. Blood cadmium concentrations were mainly associated with tobacco smoking (partial r2 = 0.56; p < 0.0001), while consumption of caribou liver and kidney remain a minor source of cadmium exposure among never-smokers. Conclusion Important decreases in mercury, lead and cadmium exposure were observed. Mercury decrease could be explained by dietary changes and the ban of lead cartridges use likely contributed to the decrease in lead exposure. Blood cadmium concentrations remain high and, underscoring the need for intensive tobacco smoking prevention campaigns in the Nunavik population.
