Abstract

Abstract The adipose tissue (AT) surrounding breast cancer (BC) plays a pivotal role in cancer progression and represents an optimal source for new biomarker discovery. The aim of this work was to investigate whether specific AT factors may have prognostic value in estrogen receptor-positive (ER+) BC. Proteoglycan Versican (VCAN), Insulin-like Growth Factor 1 (IGF1), Reticulon 4B (RTN4), chemokines CCL5 (also known as RANTES) and interleukin 8 (IL-8) are expressed in AT and may play important roles in BC progression. Peritumoral AT and tumoral biopsies were obtained from patients with ER+ BC (N = 23). AT specimens were collected also from healthy women (N = 17; CTRL-AT). The analysis of gene expression by qPCR revealed significantly higher mRNA levels of VCAN, IGF1, RTN4, and CCL5 in BC-AT compared to the CTRL-AT, and no difference in IL-8 mRNA levels. VCAN positively correlated with patient Body Mass Index (BMI) in BC-AT, while not in CTRL-AT. Moreover, VCAN and IGF1 positively correlated with RTN4 and negatively with CCL5. Interestingly, VCAN correlated with tumoral Ki67, while IGF1 with tumoral OCT4 that, in turn, correlated with tumoral Ki67 and patient BMI. Thus, peritumoral AT content of VCAN, and IGF1 are related to BC proliferation and aggressiveness.

AimsGene fusions assays are key for personalised treatments of advanced human cancers. Their implementation on cytological material requires a preliminary validation that may make use of cell line slides mimicking cytological samples. In this international multi-institutional study, gene fusion reference standards were developed and validated.MethodsCell lines harbouringEML4(13)–ALK(20) andSLC34A2(4)–ROS1(32) gene fusions were adopted to prepare reference standards. Eight laboratories (five adopting amplicon-based and three hybridisation-based platforms) received, at different dilution points two sets of slides (slide A 50.0%, slide B 25.0%, slide C 12.5% and slide D wild type) stained by Papanicolaou (Pap) and May Grunwald Giemsa (MGG). Analysis was carried out on a total of 64 slides.ResultsFour (50.0%) out of eight laboratories reported results on all slides and dilution points. While 12 (37.5%) out of 32 MGG slides were inadequate, 27 (84.4%) out of 32 Pap slides produced libraries adequate for variant calling. The laboratories using hybridisation-based platforms showed the highest rate of inadequate results (13/24 slides, 54.2%). Conversely, only 10.0% (4/40 slides) of inadequate results were reported by laboratories adopting amplicon-based platforms.ConclusionsReference standards in cytological format yield better results when Pap staining and processed by amplicon-based assays. Further investigation is required to optimise these standards for MGG stained cells and for hybridisation-based approaches.

Abstract

Objetivo: descrever o papel da família na opção pela vasectomia, o período de aconselhamento profissional e os resultados diante do procedimento na perspectiva de homens adultos. Método: investigação descritiva e exploratória de abordagem qualitativa. Os dados foram coletados em agosto de 2013, por meio de entrevista, junto a 13 homens que haviam sido submetidos à esterilização cirúrgica há, no mínimo, seis meses, em um município paranaense. Resultados: a família representou para os homens fator decisório ante a escolha pela cirurgia, pois foi descrita como suporte emocional. Apesar de o enfermeiro não ter participado do processo de aconselhamento para a realização da vasectomia, o procedimento cirúrgico e seus resultados foram descritos como satisfatórios. Conclusão: os enfermeiros necessitam participar mais ativamente do aconselhamento aos homens que buscam os serviços de saúde para a realização da vasectomia e a família deve ser incluída neste processo por constituir suporte para a decisão masculina.

Abstract

Il lavoro di ricerca ha avuto quale obiettivo l'analisi del contesto dell'insula 14 della Regio I di Pompei. Le singole unità abitative dell'isolato sono state analizzate nella loro complessità stratigrafica, per avviare una prima riflessione sull'evoluzione interna subita dalle strutture e allo stesso tempo cercare di ricostruire, nel modo più accurato possibile, la fase di occupazione immediatamente precedente l'evento eruttivo. I risultati di questa analisi sono stati integrati con i dati provenienti dallo studio dei materiali, seguendo un approccio contestuale che desse rilevanza anche al repertorio dell'instrumentum domesticum per fornire, lì dove possibile, ulteriori indicazioni utili a chiarire funzionalità e/o riutilizzo degli ambienti.

24. UN CEMENTIZIO CON MOTIVO INEDITO DA UN CUBICULUM DI POMPEI (REGIO I, 14).

Abstract

Excavations in the Regio I, insula 14 took place between the ’50s and the late ’80s of the 20th, though the area remains largely unpublished. In the first attempt to trace the history of this district of the city, two new decorative floors were discovered in the northern house of the insula (number 5) both located in two small cubicula. The decoration is not equally preserved and some details are known today only thanks to the data recorded during the excavation. Nevertheless, such “discovery” helps to reconstruct a little bit of the history of the house, as well as to enrich Pompeii’s catalog with further artefacts. [ABSTRACT FROM AUTHOR]

Abstract

Background Surgical strategies are being adapted to face the COVID‐19 pandemic. Recommendations on the management of acute appendicitis have been based on expert opinion, but very little evidence is available. This study addressed that dearth with a snapshot of worldwide approaches to appendicitis. Methods The Association of Italian Surgeons in Europe designed an online survey to assess the current attitude of surgeons globally regarding the management of patients with acute appendicitis during the pandemic. Questions were divided into baseline information, hospital organization and screening, personal protective equipment, management and surgical approach, and patient presentation before versus during the pandemic. Results Of 744 answers, 709 (from 66 countries) were complete and were included in the analysis. Most hospitals were treating both patients with and those without COVID. There was variation in screening indications and modality used, with chest X‐ray plus molecular testing (PCR) being the commonest (19·8 per cent). Conservative management of complicated and uncomplicated appendicitis was used by 6·6 and 2·4 per cent respectively before, but 23·7 and 5·3 per cent, during the pandemic (both P, The COVID‐19 pandemic required reorganization of surgical services, affecting patients with common surgical diseases including acute appendicitis. No evidence is available on the topic. This study found global variation in screening policies, use of personal protective equipment and intraoperative directives. Abstract

AimsGene fusions assays are key for personalised treatments of advanced human cancers. Their implementation on cytological material requires a preliminary validation that may make use of cell line slides mimicking cytological samples. In this international multi-institutional study, gene fusion reference standards were developed and validated.MethodsCell lines harbouring EML4(13)–ALK(20) and SLC34A2(4)–ROS1(32) gene fusions were adopted to prepare reference standards. Eight laboratories (five adopting amplicon-based and three hybridisation-based platforms) received, at different dilution points two sets of slides (slide A 50.0%, slide B 25.0%, slide C 12.5% and slide D wild type) stained by Papanicolaou (Pap) and May Grunwald Giemsa (MGG). Analysis was carried out on a total of 64 slides.ResultsFour (50.0%) out of eight laboratories reported results on all slides and dilution points. While 12 (37.5%) out of 32 MGG slides were inadequate, 27 (84.4%) out of 32 Pap slides produced libraries adequate for variant calling. The laboratories using hybridisation-based platforms showed the highest rate of inadequate results (13/24 slides, 54.2%). Conversely, only 10.0% (4/40 slides) of inadequate results were reported by laboratories adopting amplicon-based platforms.ConclusionsReference standards in cytological format yield better results when Pap staining and processed by amplicon-based assays. Further investigation is required to optimise these standards for MGG stained cells and for hybridisation-based approaches.

Abstract

Lifting tasks, among manual material handling activities, are those mainly associated with low back pain. In recent years, several instrumental-based tools were developed to quantitatively assess the biomechanical risk during lifting activities. In this study, parameters related to balance and extracted from the Centre of Pressure (CoP) data series are studied in fatiguing frequency-dependent lifting activities to: i) explore the possibility of classifying people with LBP and asymptomatic people during the execution of task; ii) examine the assessment of the risk levels associated with repetitive lifting activities, iii) enhance current understanding of postural control strategies during lifting tasks. Data were recorded from 14 asymptomatic participants and 7 participants with low back pain. The participants performed lifting tasks in three different lifting conditions (with increasing lifting frequency and risk levels) and kinetic and surface electromyography (sEMG) data were acquired. Kinetic data were used to calculated the CoP and parameters extracted from the latter show a discriminant capacity for the groups and the risk levels. Furthermore, sEMG parameters show a trend compatible with myoelectric manifestations of muscular fatigue. Correlation results between sEMG and CoP velocity parameters revealed a positive correlation between amplitude sEMG parameters and CoP velocity in both groups and a negative correlation between frequency sEMG parameters and CoP velocity. The current findings suggest that it is possible to quantitatively assess the risk level when monitoring fatiguing lifting tasks by using CoP parameters as well as identify different motor strategies between people with and without LBP. Abstract

Cholesterol granulomas are common in the mastoid antrum and air cells of the temporal bone. In the paranasal sinuses, especially in the frontal sinus, they have occasionally been mentioned in the literature. The pathogenesis is unknown, but the majority of the authors support the concept of airway obstruction in the cells well pneumatised of temporal bone and paranasal sinuses. The authors report a case of cholesterol granuloma of the frontal sinus treated with radical surgical techniques, and they also recommend an endoscopic approach to frontal sinus to restore or enlarge the nose-frontal canal and promote drainage and ventilation of the frontal sinus.

Abstract

[EN] This essay deals with the analysis of the semaphoric intersection of Avenida Pío XII, Avenida de Campanar, Calle de la Vall de la Ballestera and Calle Joaquín Ballester located in the city of Valencia. In order to carry out this, the document studies the capacity and and the level of service, since, it presents problems of congestion producing delays and discomforts to the users. Once these analyzes have been carried out, a series of alternatives are proposed to provide a feasible solution to the problem. In this light, the method of determination for the calculation of the capacity at traffic light intersections will be collected in this document and, on the other hand, the information obtained from the selected traffic light intersection will be captured. The methodology to be used throughout the study will be the one defined in the HCM 2010, Highway Capacity Manual 2010. Following the procedures explained in volume 3 "Interrupted Flow", chapter 18 of the HCM 2010, will proceed to evaluate the capacity and the quality of service provided to the users of the vehicles crossing the mentioned intersection. In addition to this, the methodology allows identifying the source of problems and provides a measure of strategies for their improvement. Once the calculations have been made using the HCM 2010 procedure, it is observed that the capacity obtained does not correspond to reality, so it is proposed to use a different analysis that studies the storage of the intersection. Thus, few alternatives are proposed to solve the problem and it is suggested proposals to solve the congestion produced., [CA] A aquest treball s'analitza la intersecció semaforitzada de l'Avinguda Pío XII, l'Avinguda de Campanar, el carrer de la Vall de la Ballestera i el carrer Joaquín Ballester situats a la Ciutat de València. Amb la finalitat de realitzar un estudi sobre la seva capacitat i nivell de servei, ja que, presenta problemes de congestió produint esperes així com incomoditats als usuaris. Una vegada realitzats aquests anàlisis, es proposen una serie d'alternatives per poder proporcionar una solució factible al problema. D'aquesta manera, a aquest document s'arreplegarà el mètode de determinació per al càlcul de la capacitat en interseccions semaforitzades i, d'altra banda, es plasmarà la informació obtinguda de la intersecció semaforitzada seleccionada. La metodologia que s'utilitzarà en tot l'estudi es regira per la definida en el HCM 2010, Highway Capacity Manual 2010. Seguint els procediments que s'expliquen al volum 3 "Interrupted Flow", capítol 18 del HCM 2010, es procedirà a avaluar la capacitat i la qualitat del servei que es proporciona als usuaris dels vehicles que creuen la intersecció esmentada. D'altra banda, la metodologia permet identificar la font de problemes i proporcionar una seria d¿estratègies per a la seva millora. Una vegada realitzats els càlculs mitjançant el procediment del HCM 2010, s¿observa que la capacitat obtinguda no es correspon amb la realitat, per la qual cosa, es proposa la utilització d'anàlisis que estudien l'emmagatzematge de la intersecció. Abstract

Objetivo: descrever o papel da família na opção pela vasectomia, o período de aconselhamento profissional e os resultados diante do procedimento na perspectiva de homens adultos. Método: investigação descritiva e exploratória de abordagem qualitativa. Os dados foram coletados em agosto de 2013, por meio de entrevista, junto a 13 homens que haviam sido submetidos à esterilização cirúrgica há, no mínimo, seis meses, em um município paranaense. Resultados: a família representou para os homens fator decisório ante a escolha pela cirurgia, pois foi descrita como suporte emocional. Apesar de o enfermeiro não ter participado do processo de aconselhamento para a realização da vasectomia, o procedimento cirúrgico e seus resultados foram descritos como satisfatórios. Conclusão: os enfermeiros necessitam participar mais ativamente do aconselhamento aos homens que buscam os serviços de saúde para a realização da vasectomia e a família deve ser incluída neste processo por constituir suporte para a decisão masculina.

Abstract

During a non-invasive procedure, participants both helped and helped by a confederate with features that create social distance (membership in an ethnic outgroup or another social group). For this purpose, we created a set of virtual scenarios in which the confederate's ethnicity (white vs. black) and appearance (business man vs. beggar, with casual dress as a control condition) were crossed. The study aimed to explore how the emotional reactions of participants changed according to the confederate's status signals as well as signals that they belong to the same or a different ethnic group. Participants' alertness, calmness, and engagement were monitored using electroencephalogram (EEG) during the original virtual reality (VR) video sessions. Participants' distress and empathy when exposed to helping interactions were self-assessed after the VR video sessions. The results pointed out that, irrespective of whether they helped the confederate or were helped by him/her, white participants showed higher levels of alertness when exposed to helping interactions involving a white beggar or a black businessman, and their emotional calmness and engagement were higher when interacting with a black beggar or a white businessman. The results for self-assessed distress and empathy followed the same tendency, indicating how physiological and self-assessed measures can both contribute to a better understanding of the emotional processes in virtual intergroup helping situations. Based on the presented results, the methodological and practical implications of VR in terms of enhancing self-reflective capacities in intergroup helping processes are discussed. Abstract

Objective: To describe the role of the family in opting for vasectomy, the period of professional counseling and outcomes before the procedure from the perspective of adult men. Method: Descriptive and exploratory qualitative research approach. Data were collected in August 2013, through interviews, along with 13 men who had undergone surgical sterilization for at least six months in a city in the Paraná. Results: family represented the deciding factor for the choice men before the surgery because it was described as emotional support. Although the nurse did not attend the counseling process for performing vasectomy, surgical procedure and its results were described as satisfactory. Conclusion: nurses need to participate more actively in counseling to men who seek health services to perform the vasectomy and the family should be included in this process to provide support for male decision.

47. Recreational boating as a vector of spread of non-indigenous species around the Mediterranean Sea

Abstract

This study seeks to identify the hotspots for the secondary spread of non-indigenous species (NIS) resulting for recreational boating travel patterns across the Mediterranean Sea, and investigate the potential for internal spread within each study region. Three Mediterranean regions will be sampled for: 1) NIS richness in the marinas and NIS richness and abundance from recreational vessels; 2) biopollution level of transient recreational boats; 3) geographic range of NIS within each region; 4) hull cleaning and travel history of recreational boats. The results will then be tested with other abiotic factors to determine which factors are significant in contributing to making some marinas more high-risk for the presence and transfer of alien species. Finally, a spatial connectivity network analysis will be presented of the 30 sampled marinas by combining a risk-level for each marina along with recreational boating travel patterns. Marina selection is based on the busiest marinas for recreational boaters in each region in combination with securing permission and feasibility. Samples for Amathia verticillatum will be collected from each region and genetically tested to determine if they share the same initial or secondary source of invasion.
