Background In rainbow trout (Oncorhynchus mykiss), the endocrine control of spermiation is not fully understood. Besides 11ketotestosterone (11KT) and 17alpha, 20beta-dihydroxyprogesterone (MIS), the potential physiological ligand of the mineralocorticoid receptor (MR) 11-deoxycorticosterone (DOC), is a credible candidate in O. mykiss spermiation regulation as spermiation is accompanied with changes in aqueous and ionic flows. Methods In this study, we investigated potential roles of DOC during spermiation 1) by describing changes in blood plasma DOC level, MR mRNA abundance during the reproductive cycle and MR localization in the reproductive tract 2) by investigating and comparing the effects of DOC (10 mg/kg) and MIS (5 mg/kg) supplementations on sperm parameters 3) by measuring the in vitro effect of DOC on testis MIS production. Results The plasma concentration of DOC increased rapidly at the end of the reproductive cycle to reach levels that were 10–50 fold higher in mature males than in immature fish. MR mRNA relative abundance was lower in maturing testes when compared to immature testes, but increased rapidly during the spermiation period, immediately after the plasma rise in DOC. At this stage, immunohistochemistry localized MR protein to cells situated at the periphery of the seminiferous tubules and in the efferent ducts. Neither DOC nor MIS had significant effects on the mean sperm volume, although MIS treatment significantly increased the percentage of males producing milt. However, a significant reduction in the spermatocrit was observed when DOC and MIS were administrated together. Finally, we detected an inhibitory effect of DOC on testis MIS production in vitro. Conclusion These results are in agreement with potential roles of DOC and MR during spermiation and support the hypothesis that DOC and MIS mechanisms of action are linked during this reproductive stage, maybe controlling milt fluidity. They also confirm that in O. mykiss MIS is involved in spermiation induction.

Le développement des élevages industriels de truite et de saumon a accru la nécessité d’étaler la production sur l’ensemble de l’année. Dans ce but la manipulation de la photopériode est nécessaire pour obtenir des pontes toute l’année chez ces espèces à reproduction annuelle. A titre d’exemple, l’exposition des géniteurs de truite arc-en-ciel à des jours longs pendant les 2 mois suivant la ponte, puis à des jours courts pendant 4 mois, induit une nouvelle ponte 6 mois après la précédente. Par ailleurs, il est possible de réduire l’incidence de la maturation sexuelle des femelles quand les poissons sont soumis à un éclairement continu dès le début de l’alimentation. Ce résultat permet de disposer d’une alternative à la triploïdisation pour produire les individus immatures dont le marché a besoin.

Evolution de la gametogenèse, du poids du corps et des gonades au cours du cycle reproducteur du brochet (Esox lucius)

In the rainbow trout (Salmo gairdneri) and the tench (Tinca tinca) at the time of spermatogonia divisions a rise in the plasma gonadotropin levels was observed. Just before or during the spawning period, E2 17β and plasma gonadotropin reach their maximum levels. (2) When the effects of raising the natural cycle temp of the tench were studied (Δt +3°C and +6°C), in conditions where nycthemeral and seasonal rhythmicity were maintained, a significant increase in fertility was observed with the increase in temp (the 1st spawning period was earlier and there was a rise in the number of spawnings). This effect of temp seems to act at the level of the hypothalamo-pituitary system.

Le cycle reproducteur des poissons est sous la dépendance de l'hypophyse qui élabore et secrète les hormones gonadotropes. Ces hormones interviennent lors du déroulement de la gamétogenèse et sont produites activement pendant la maturation ovocytaire puis l'ovulation. La technique d'induction de ponte dite technique d'hypophysation consiste en un apport exogène d'hormone gonadotrope par injection d'hypophyses pour provoquer le déroulement de phases du cycle sexuel qui ne se produisent pas spontanément en condition de captivité comme l'ovulation.

Des spécimens immatures ou pré-pubertaires de mâles et de femelles de truites arc-en-ciel de 2 ans, de 500 à 600 g, ont été exposés, à partir du 21 juin, à une lumière continue (groupe CL); un groupe de truites témoins (groupe C) ont été exposées à la photopériode naturelle de la région parisienne et ont subi les variations saisonnières de température. L'échantillonnage périodique des gonades, pour fin d'analyses quantitatives de la spermatogenèse et évaluation du diamètre des follicules, a permis de suivre la gamétogenèse. Des techniques radioimmunologiques ont été utilisées pour mesurer les variations des concentrations de l'hormone GTH dans l'hypophyse et dans le plasma. La quantité et la qualité des gamètes ont été évaluées au cours de la fraye. À la lumière continue, la gamétogenèse et la fraye sont retardées d'environ 2 mois par rapport aux poissons témoins gardés à la photopériode naturelle. De plus, la gamétogenèse ainsi retardée comporte certaines particularités. Chez les mâles, la proportion de cellules germinales dans le testicule est modifiée comparativement au groupe témoin; par exemple, il y a présence de spermatides dans les lobules testiculaires au moment où commence la spermiation. La production de sperme, telle que mesurée dans quatre échantillons, est plus faible chez les mâles soumis à la lumière continue que chez les poissons témoins. Chez les femelles, durant la gamétogenèse, le diamètre des follicules et l'indice gonadosomatique ont tendance à être plus faibles chez le groupe CL que chez les poissons témoins, mais le diamètre des follicules diminue éventuellement et la fécondité est la même chez les deux groupes. Les variations des concentrations de GTH ne montrent de particularités que dans le plasma, en décembre. Le contenu de GTH dans l'hypophyse est plus faible en décembre chez les deux groupes de poissons des deux sexes, quel que soit le stade de reproduction atteint et quelle que soit la photopériode. Il n'y a pas de différences entre les indices hépatosomatiques et viscérosomatiques des deux groupes de poissons.

Male and female rainbow trout in their first reproductive cycle were subjected to long days (16L : 8D; 24L : 0D and 16L : 8D shifting progressively to 24L : 0D within 6 months). All the fish were maintained at ambient temperatures of between 3 and 16°C. Mean ovulation and spermiation times were 2 months later in the long-day group than in the control group, and ovulation and onset of spermiation were spread over a long period. The quantity and quality of the sexual products obtained were comparable in all groups. In subsequent experiments with the female groups, 50% of the females kept for the first time under permanent light ovulated or were about to ovulate 1–2 months later than the controls. All females initially kept in LD during the first cycle also showed delayed ovulation but only a few ovulated. In the ovaries of ovulated and non-ovulated females, small vitellogenetic oocytes were found, indicating abnormalities in recruitment. Fish exposed for the first time to long days showed a delay in spawning but had normal ovulation.

Bilateral castration of male rainbow Trout resulted in increased levels of circulating gonadotropin, as measured by radioimmunoassay, but the response varied depending on the season. In early May the increase was 12 ng/ml after 30 days, when compared to the intact control but later in the month at the time when a peak of gonadotropin level is observed in normal fish, the increase was 25 ng/ml after 13 days. At the beginning of August, when spermatogenesis commences, the increase was 9 ng/ml after 12 days. In September, when spermatogenesis is ending, an increase of 20 ng/ml was found after 7 days, but only 50% of the animals responded. In December, during spermiation, an increase of 90 ng/ml was found after 7 days., Chez la Truite arc-en-ciel ayant subi une castration bilatérale, le niveau de la gonadotropine circulante mesurée par radioimmunologie augmente, mais le type de réponse varie selon la période de l'année à laquelle l'opération est pratiquée. En mai, avant l'apparition du pic de sécrétion observé dans les conditions naturelles, l'augmentation des teneurs en gonadotropine après castration est lente et faible (8 ng/ml de plus que les témoins intacts après 30 jours). Elle est beaucoup plus rapide lorsque la castration est pratiquée fin mai, lors de l'apparition de ce pic (différence de 12 ng/ml après 13 jours). Ensuite, les différences par rapport aux témoins sont de 9 ng/ml après 12 jours en août (démarrage de la spermatogenèse), 10 ng/ml après 7 jours (spermatogenèse achevée) et 45 ng/ml après 7 jours en décembre (spermiation).

To determine the effect of gonadal feedback on plasma GTH level, female rainbow trout were ovariectomized at three stages at the end of the reproductive cycle: at the end of vitellogenesis, during germinal vesicle migration and during the post-ovulatory period. A group of controls and one of castrates in each experiment were given an injection of physiological salt solution, and a third group of castrates was supplemented with estradiol-17 beta (E2) twice a week (200 micrograms/kg) from the day of surgery. The blood was sampled twice a week, and the GTH measured by RIA. At the end of vitellogenesis, castration induced a significant rise in the gonadotropic hormone level (P less than 0.001 from post-surgical day 5), and that response, unimpeded by E2, was homogeneous in all the fish. During germinal vesicle migration, the response to castration and to supplementary E2 varied with the individual. Ovariectomy induced a significant increase in GTH (P less than 0.005 from day 3), but that increase was immediate in 5 females and delayed in the other 4; E2 prevented GTH rise in only 6 females. At the post-ovulatory period we found no significant difference between the control fish and the castrates and E2, at least temporarily, prevented the post-ovulatory rise in GTH which is usually found in trout.

Nous disposons maintenant de quelques informations sur les mecanismes qui controlent la gametogenese et son deroulement au cours du cycle reproducteur. Les effets des facteurs du milieu les plus importants commencent aussi a etre mieux connus et l'ensemble de ces connaissances ainsi acquises permet actuellement de maitriser certaines phases de la reproduction et ouvre des perspectives d'application en pisciculture.

International audience; Gonadal development and gametogenesis have been studied in the brown trout ( Salmo trutta) beginning at the establishment of the first reproductive cycle. Monthly samples were taken from 14-month old fishes beginning in July and extending over a 20-month period. Complete gametogenesis was observed in males from the first year of sampling and from the following year in females. Spermatogenesis was analysed over the first two cycles and a quantitative study showed that the volume of spermatozoa produced in relation to body weight was slightly higher at the end of the second cycle. At the end of the second reproductive cycle, the process of spermiation ceased while there were still spermatozoa in the testes, although most of them had been evacuated at the end of the first cycle. Spermatogenesis seemed to be more efficient during the second cycle but spermiation yield was higher during the first cycle. Oogonia were always present in the female ovaries and there was mitotic activity in July of the second year of life and in October-November of the third year. Previtellogenetic stages (accumulation of RNA and lipid) were already present at 14 months of age when sampling started. During the second year of life there was limited accumulation of yolk in the oocytes but it did not modify total ovary weight. In this population, vitellogenesis was complete and most of the females began to ovulate only in the third year. Before the end of the cycle in October-November, the number of previtellogenetic oocytes (stages 1, 2 and 3) increased markedly. This was followed in December by an increase in the number of oocytes containing yolk bodies, representing no doubt the class of oocytes undertaking vitellogenesis in the next cycle. the patterns of initiation of the first and second cycles thus appeared to be very different.

Les limites de ces techniques sont discutées., This paper discusses the key role played by the hypothalamo-pituitary axis which integrates the environmental factors and produces and secretes one or more hormones implicated in gonadal development. Due to sex steroid secretion, these hormones act on other organs, such as the liver, and mobilize body stores, integrating them into the oocytes in the form of yolk. Physical (photoperiod, temperature), physico-chemical (changes in water quality), sociological and dietary factors are among those influencing sex cycle regulation. Some of the tools available at present for controlling certain phases of the cycle are hormonal supplementation (hypophysation) or variation of environmental parameters, such as temperature. The limits of these techniques are discussed.

The endocrinology of teleosts reproduction : an approach to the control of fish reproduction

Des données quantitatives recueillies pendant la période de spermiation du premier cycle reproducteur chez la Truite arc-en-ciel ont montré que : — La durée de la période de spermiation est très variable : moyenne = 5 semaines [extrêmes 1 à 10 semaines). — La quantité moyenne de spermatozoïdes récoltés mesurée sur 57 mâles est de 60 milliards (438 millions/g poids corporel) et n'est pas en relation avec le poids corporel. Cette valeur représente 48 % de la quantité totale des spermatozoïdes produits. — Les réserves gonadiques qui subsistent dans les gonades à la fin de la période de spermiation (spermatozoïdes non éjaculés qui seront résorbés dans les testicules) sont de 68 milliards de spermatozoïdes par mâle (mesures établies sur 12 mâles). — La production spermatogénétique (spermatozoïdes récoltés + réserves gonadiques en fin de spermiation est de 134 milliards de spermatozoïdes (mesures sur 12 mâles). — Au cours de la période de spermiation la concentration du sperme en spermatozoïdes décroit : variation entre 15 et 10 milliards de spermatozoïdes par ml.Abstract : Quantitatives data collected during the reproductive season in young rainbow trout male (first spermatogenetic cycle) showed the following facts : 1) The duration of the spermiation period (running stage) is variable ; average : 5 weeks — extrêmes 1 to 10 weeks. 2) Average number of spermatozoa collected by stripping and measured by a spectrophotometric technique was 60. 109 (438. 106/g body weight) n = 57 males. International audience; Population structure and annual reproduction cycle of the De B alsac's mouse (Heimyscus fumosus) were studied in a wild population of south-western Gabon. The reproductive status of the mice was investigated at autopsy, and age of each animal was estimated by tooth wear patterns and body weight. Sex-ratio was equilibrated whatever the season and the ageclass. Females tended to reach sexual maturity more slowly than males. However, the average age at puberty differed between individuals of the same sex. A wide range of tooth-wearclasses and weight-classes were present in most months of the year, and sexually active individuals of both sexes were captured in most months. However pregnant or lactating females were only captured from August to March, while very young individuals were only captured from October to March. Thus, the reproduction of H. fumosus would be partly seasonal with an interruption from April to July (end of the short wet season, and beginning of the long dry season). We discussed the potential role of food availability on this seasonality; La reproduction et la structure de population du Muridé africain Heimyscus fumosus ont été étudiées dans une population sauvage du sud-ouest du Gabon. Pour chaque animal, nous avons noté, lors de l'autopsie, s'il était sexuellement actif et, pour les femelles, si elles étaient prégnantes ou allaitantes. De plus, l'âge des animaux a été estimé en fonction de leur usure dentaire et de leur poids. Le sexe-ratio obtenu était équilibré quels que soient l'âge ou la saison. Bien que l'âge de la puberté diffère entre individus de même sexe, les femelles atteindraient leur maturité sexuelle plus tardivement que les mâles. La plupart des mois, de larges gammes de classes de poids et de classes d'âge dentaires étaient présentes. Bien que des individus sexuellement actifs aient été capturés tout au long de l'année, les femelles allaitantes et prégnantes ne l'ont été que d'août à mars, tandis que les très jeunes individus l'ont été d'octobre à mars. Ainsi, la reproduction de H. fumosus serait partiellement saisonnière, avec une interruption d'avril à juillet (fin de la petite saison des pluies et début de la grande saison sèche). Nous discutons du rôle potentiel de la disponibilité des ressources sur cette saisonnalité de la reproduction.
