Anthracycline antibiotics play an important role in cancer chemotherapy. The need to improve their therapeutic index has stimulated an ongoing search for anthracycline analogs with enhanced properties. This review aims to summarize the common synthetic approaches to benzo[g]quinoxaline-5,10-diones and their uses in heterocyclic chemistry. Because of the valuable biological activities of the 1,4-diazaanthraquinone compounds, a summary of the most promising heterocyclic quinones is provided together with their antitumor properties.

International audience; Cross-coupling reactions between 2-methyl-2-propen-1-ol and various boronic acids are used to obtain aromatic-(2-methylallyl) derivatives. However, deboronation or isomerization side reactions may occur for several boronic acids. We describe herein the synthesis of original alkenes with good yields under mild reaction conditions that decrease these side reactions. The scope of this environmentally benign reaction is thereby extended to a wide variety of boronic acids. A mechanistic study was conducted and suggested a plausible catalytic cycle mechanism, pointing to the importance of the Lewis acidity of the boronic acid used.

International audience; Object: Environmental monitoring is usually conducted by surface sampling to detect and quantify the presence of cytotoxic drugs after their reconstitution and administration. This technique reveals the origins of residual contamination and is an important component in order to protect healthcare workers from the potential risk of occupational exposure. The aim of this work is to compare various techniques and results of surface sampling for cytotoxics.Materials and methods: For each technique, sample processing methods and their analysis were compared from literature data. Sampling is often performed by the wiping technique. After treatment, various single or multi-compound technical analyses are used, in particular liquid or gas chromatography involving different detection methods: ultraviolet, mass spectrometry, plasma torch, voltammetry. Some methods are validated to ensure reliability.Results: Despite published guidelines and the use of isolator technology for the preparation of cytotoxic drugs, workplace contamination persists, leading to chemotherapeutic agents’ exposure of healthcare workers. Efforts need to be maintained with particular emphasis on harmonization and on determining alert level for cytotoxic contamination.

International audience; a Manganese(III) acetate-mediated peroxycyclization between 2-hydroxy-3-methylnaphthoquinone and various alkenes was performed to obtain dihydronaphtho[2,3-c][1,2]dioxine-5,10(3H,10aH)-diones. The reactivity of symmetrical or unsymmetrical 1,1-disubstituted alkenes and monosubstituted alkenes allowed the synthesis of more than 50 original molecules. Focusing on the excellent reactivity of 2-hydroxy-3-methylnaphthoquinone, we describe the first example of Mn(OAc)3 reactivity with nitro-substituted alkenes. The scope, limitations and stereochemistry of the products synthesized are discussed. Starting from monosubstituted alkenes, the instability of a pair of diastereoisomers was observed, leading to ring opening.

Hemolytic uremic syndrome is a rare disease, frequently responsible for renal insufficiency in children. Recent findings have led to renewed interest in this pathology. The discovery of new gene mutations in the atypical form of HUS and the experimental data suggesting the involvement of the complement pathway in the typical form, open new perspectives for treatment. This review summarizes the current state of knowledge on both typical and atypical hemolytic uremic syndrome pathophysiology and examines new perspectives for treatment., Le syndrome hémolytique urémique est une maladie rare, souvent responsables de l'apparition d une insuffisance rénale chez les enfants. Des découvertes récentes ont conduit à un regain d'intérêt dans cette pathologie. La découverte de nouvelles mutations génétiques dans la forme atypique et les données expérimentales suggérant l'implication de la voie du complément dans la forme typique ouvrent de nouvelles perspectives pour le traitement. Cette revue résume l'état actuel des connaissances sur la physiopathologie du syndrome hémolytique urémique typique et atypique et présente les nouvelles perspectives de traitement.

Neisseria meningitidis infections are a major public health problem worldwide. Although conventional approaches have not led to development of a serogroup B meningococcal vaccine, a new technique based on genome sequencing has created new perspectives. Recently, a universal serogroup B meningococcal vaccine, Bexsero®, was licensed in Europe, Australia and United States, following several clinical studies demonstrating its immunogenicity and safety. Availability of this vaccine could contribute positively to human health, by significantly reducing the incidence of meningococcal infections. However, unfavorable cost-effectiveness analysis means that routine vaccination is not currently recommended. Another serogroup meningococcal vaccine, Trumemba®, was also recently licensed in United States. Like any drug, Bexsero® and Trumemba® will require close observation to assess their impact on meningococcal epidemiology., Les infections à Neisseria meningitidis sévissent dans le monde entier et restent un véritable problème de santé publique. Toutefois, la vaccinologie conventionnelle n’a pas permis le développement d’un vaccin contre le méningocoque de sérogroupe B. Une nouvelle technique, la vaccinologie inverse, a alors ouvert de nouvelles perspectives. Récemment, un vaccin universel antiméningococcique B, Bexsero®, a été autorisé en Europe, en Australie et aux Etats-Unis, suite à des études cliniques d’immunogénicité et d’innocuité concluantes. La mise à disposition d’un tel vaccin peut apporter une réelle contribution à l’amélioration de la santé humaine, pouvant réduire significativement l’incidence des infections méningococciques. Cependant, la vaccination systématique n’est pas recommandée actuellement, en raison d’une analyse coût-efficacité défavorable. Un second vaccin antiméningococcique B, Trumemba®, a également été récemment autorisé aux Etats-Unis. Comme tout médicament, ces deux vaccins nécessiteront une surveillance étroite, permettant d’étudier leur impact sur l'épidémiologie méningococcique.

A convenient microwave irradiation protocol was utilized for the synthesis of b-ketosulfones 1-5 in good yields. These sulfones reacted with alkenes through a radical oxidative cyclization mediated by Mn(OAc)3. Dihydrofurans 6-10 were obtained in moderate to good yields starting from 1,1-disubstituted alkenes. Dihydrofurans 11-15 were synthesized in moderate yields and unexpected cyclopropanes 16-19 were obtained in low yields starting from 1,2-disubstituted alkenes. This protocol offers access to various dihydrofurans which could be tested for their antiparasitic potential.

International audience; Purposes. A new methodology to evaluate the medication-use system based on a risk cartography tool, has been developed. This work has been promoted by the Observatoire du medicament et des dispositifs m dicaux steriles et de l'innovation therapeutique (OMEDIT) from Provence- Alpes-Cote d'Azur (PACA)-Corse regions. Methods. This new methodology has been developed with Excel (Microsoft (R)) and has led to the mediEVAL tool. It consists in two categories of Excel files: evaluating Excel files (1 for each job of the medication-use system) and synthesis Excel files which allow to compile a group of evaluating files for a defined area (department, hospital.). Results and conclusion. mediEVAL is a new tool to evaluate quality and risk management of the entire medication-use system which has to be used by private or public hospitals of PACA and Corsica areas in their appropriate medication-use contract. Then, the OMEDIT can get data to provide an inventory of fixtures of the PACA-Corse area medication-use system situation.
