This paper offers reflections about the use of a longitudinal qualitative research design in a project which explored the long term impacts of flooding in two communities in North‐East Scotland. A temporal turn in the social sciences has raised the profile of longitudinal qualitative approaches, research whose diachronic framing allows trajectories and nuanced understandings of change to emerge. With reference to research which utilised a planned prospective longitudinal design, we offer reflections on methodological and project management 'lessons learnt' from undertaking a longitudinal qualitative study. Our experiences highlighted the importance of: (i) participant recruitment processes, including a need to 'oversample' to accommodate anticipated attrition rates; (ii) developing and sustaining a relationship between participants and researchers; (iii) reporting interim findings to participants, the funder and flood risk management stakeholders via a project Steering Group in particular; and (iv) agreeing a regular reporting schedule which allowed the funder and stakeholders access to findings during the lifetime of the project which, in turn, allowed impact to be generated before the final report was presented. In sharing our experiences our intention is twofold: to open a debate in human geography about how longitudinal qualitative research could be used more widely, in natural hazards research, rural community change and other research areas; and to illustrate that longitudinal qualitative research generates insights that can contribute to evidence‐based policy development, implementation and evaluation. Incidence of endometrial cancer (EC) is rising in the developed world. The current standard of care, hysterectomy, is often infeasible for younger patients and those with high body mass index. There are limited non-surgical treatment options and a lack of biologically relevant research models to investigate novel alternatives to surgery for EC. The aim of the present study was to develop a long-term, patient-derived explant (PDE) model of early-stage EC and demonstrate its use for investigating predictive biomarkers for a current non-surgical treatment option, the levonorgestrel intra-uterine system (LNG-IUS). Fresh tumour specimens were obtained from patients with early-stage endometrioid EC. Tumours were cut into explants, cultured on media-soaked gelatin sponges for up to 21 days and treated with LNG. Formalin-fixed, paraffin embedded (FFPE) blocks were generated for each explant after 21 days in culture. Tumour architecture and integrity were assessed by haematoxylin and eosin (H&E) and immunohistochemistry (IHC). IHC was additionally performed for the expression of five candidate biomarkers of LNG resistance. The developed ex vivo PDE model is capable of culturing explants from early-stage EC tumours long-term (21 Days). This model can complement existing models and may serve as a tool to validate results obtained in higher-throughput in vitro studies. This theme section editorial introduction presents a collection of papers brought together under the heading Rural transformations, rural futures. The papers are elaborations of work presented at a European Society for Rural Sociology 'satellite event' entitled Transitioning Rural Futures. Contributions to the theme section add to the emerging corpus of work that examines potentially transformative immediate and medium-term impacts of the COVID-19 pandemic on rural areas, and also speak to other 'grand challenges' likely to transform rural places and shape rural futures: specifically, resilience, digitalisation, wellbeing, socio-economic inequalities, shifting rural policy landscapes and impacts of transnational economic events. A transnational migration lens has shaped diaspora research in recent decades. Yet exploring diasporas in the context of moves across international borders has obscured sub-national diasporas, and this has led to a gap in our understanding of how such groups can and do contribute to their communities of origin. This article aims to develop a better understanding of the Scottish islands diaspora and to explore its potential to contribute to island communities, including via internal return migration. We present findings from a survey conducted with the Scottish islands diaspora to illustrate how internal diasporas can exhibit continued connection to a 'homeland'. This includes taking an active interest in a 'home' island's future and participating in shared cultural practices that bind individuals together within the diaspora over a sustained period. We find that the Scottish islands diaspora carries many of the hallmarks of its transnational counterparts and highlight how this group has the potential to contribute to the future of Scotland's islands both at a distance and, potentially, via return migration. EVs released by adipose derived stem cells (ADSCs) have shown promise as a therapeutic for tissue repair because of their purported immune-regulatory properties. Extracellular vesicles (EVs) fromADSCs could be beneficial in improving graft retention rates for autologous fat grafting (AFG) post-mastectomy as, currently, grafted tissue rates are variable. Enriching grafted tissue with ADSC-EVs may improve retention rates by modulating macrophages resident within both the breast and lipoaspirate. We aimed to identify keymacrophage phenotypes that are modulated by ADSC-EVs in vitro. ADSCs were isolated from lipoaspirates of women undergoing AFG and characterised by flow cytometry and differentiation potential. ADSC-EVs were isolated from culture media and characterised by tuneable resistive pulse sensing, transmission electron microscopy andWestern blot. Primary monocyte-derived macrophages were polarized to an M1-like (GM-CSF, IFNγ), M2-like phenotype (M-CSF, IL-4) or maintained (M0-like; M-CSF) and ADSC-EVs were co-cultured with macrophages for 48 h. Flow cytometry and high-dimensional analysis clustered macrophages post co-culture. A manual gating strategy was generated to recapitulate these clusters and was applied to a repeat experimental run. Both runs were analysed to examine the prevalence of each cluster, representing a unique macrophage phenotype, with and without ADSC-EVs. Following the addition of ADSC-EVs, M0-like macrophages demonstrated a reciprocal shift of cell distribution froma cluster with a 'high inflammatory profile' (CD36+++CD206+++CD86+++; 16.5±7.0%; p<0.0001) to a cluster with a 'lower inflammatory profile' (CD36+CD206+CD86+; 35 ± 21.5%; p < 0.05). M1-like macrophages shifted from a cluster with a 'high inflammatory profile' (CD206++CD11b++CD36++CD163++; 26.1 ± 9.4%; p = 0.0024) to a 'lower inflammatory profile' (CD206+CD11b+CD36+CD163+; 72.8 ± 8.7%; p = 0.0007). There was no shift in M2-like clusters following ADSC-EV treatment. ADSC-EVs are complex regulators of macrophage phenotype that can shift macrophages away from a heightened pro-inflammatory state. This paper seeks to examine understandings and applications of rural community resilience. Scottish policy has shifted toward neoliberalism and community empowerment, with communities encouraged to play a proactive role in enhancing their own resilience. We argue that it is important to understand the perspectives of multiple stakeholders to identify what practical factors they believe enhance community resilience and to provide a greater understanding of the mechanisms through which community resilience can be delivered. Drawing on qualitative data collection, we question what resilience means and what factors can facilitate it in practice. We find that dual discourses of resilience emerge: the emergency which reflects the policy focus on short-term damage reduction, and the everyday which reflects the desire for more long-term adaptive capacities developing in response to gradual change in rural communities. We conclude that the discourse which stakeholders align with will affect how they understand, adopt, and practice the concept. [ABSTRACT FROM AUTHOR]

This paper presents a methodology to assess the internal and external coherence, and the robustness to future uncertainty, of place-based interventions addressing spatial (in)justice. The methodology merges elements of Theory of Change (ToC) and mechanism mapping with scenario planning. It was designed based on analysis of a range of European interventions (public policies and bottom-up initiatives), selected to highlight the ways spatial injustices have been tackled across different scales. The first phase of the methodology uses a ToC mechanism map to illustrate the logic of the intervention, including its baseline assumptions (internal) and contextual conditions (external). In the second phase, scenarios for the locality are developed based on potential states of global and local macro-trends, revealing how the contextual conditions are expected to change, and whether the intervention's baseline assumptions are likely to hold. This then allows the elaboration of spatial justice trajectories, and for the ToC mechanism map to be updated. To effectively illustrate the methodology, we show how it has been applied using a case study intervention implemented in a territorially disadvantaged area. We present the methodology as a flexible tool allowing detection of more general stylised facts, and spatial comparisons between a broad range of interventions. [ABSTRACT FROM AUTHOR]
