Abstract

Designing synthetic microenvironments for cellular investigations is a very active area of research at the crossroads of cell biology and materials science. The present work describes the design and functionalization of a three-dimensional (3D) culture support dedicated to the study of neurite outgrowth from neural cells. It is based on a dense self-assembled collagen matrix stabilized by 100-nm-wide interconnected native fibrils without chemical crosslinking. The matrices were made suitable for cell manipulation and direct observation in confocal microscopy by anchoring them to traditional glass supports with a calibrated thickness of similar to 50 mu m. The matrix composition can be readily adapted to specific neural cell types, notably by incorporating appropriate neurotrophic growth factors. Both PC-12 and SH-SY5Y lines respond to growth factors (nerve growth factor and brain-derived neurotrophic factor, respectively) impregnated and slowly released from the support. Significant neurite outgrowth is reported for a large proportion of cells, up to 66% for PC12 and 49% for SH-SY5Y. It is also shown that both growth factors can be chemically conjugated (EDC/NHS) throughout the matrix and yield similar proportions of cells with longer neurites (61% and 52%, respectively). Finally, neurite outgrowth was observed over several tens of microns within the 3D matrix, with both diffusing and immobilized growth factors. (C) 2012 Acta Materialia Inc. Published by Elsevier Ltd. Abstract

The synthesis of core–shell magnetic mesoporous nanoparticles (MMSNs) through a phase transfer process is usually performed at the 100–250 mg scale. At the gram scale, nanoparticles without cores or with multicore systems are observed. Iron oxide core nanoparticles (IO) were synthesized through a thermal decomposition procedure of α-FeO(OH) in oleic acid. A phase transfer from chloroform to water was then performed in order to wrap the IO nanoparticles with a mesoporous silica shell through the sol–gel procedure. MMSNs were then functionalized with DTPA (diethylenetriaminepentacetic acid) and used for the separation of metal ions. Their toxicity was evaluated. The phase transfer procedure was crucial to obtaining MMSNs on a large scale. Three synthesis parameters were rigorously controlled: temperature, time and glassware. The homogeneous dispersion of MMSNs on the gram scale was successfully obtained. After functionalization with DTPA, the MMSN-DTPAs were shown to have a strong affinity for Ni ions. Furthermore, toxicity was evaluated in cells, zebrafish and seahorse cell metabolic assays, and the nanoparticles were found to be nontoxic. We developed a method of preparing MMSNs at the gram scale. After functionalization with DTPA, the nanoparticles were efficient in metal ion removal and separation; furthermore, no toxicity was noticed up to 125 µg mL−1 in zebrafish. [ABSTRACT FROM AUTHOR]

Abstract

Porous silicon nanoparticles as a novel platform in gene therapy, have shown to be an efficient vehicle for the delivery of nucleic acids in cells. For the first time, a family of porous silicon nanoparticles has been produced featuring an amino-acid functionalized cationic external surface aiming at pDNA complexation. The amino acid-based pDNA nanocarriers, displaying an average diameter of 295 nm, succeeded in transfection of HEK293 cells with an efficiency 300 times superior to “bare” porous silicon nanoparticles.

Abstract

International audience; In this work, a combination of optical and electrochemical studies are performed in order to elaborate a suitable porous silicon (PSi) substrate for biosensing and biomaterial applications. We report on the electrochemical behavior of PSi multilayer stabilized by thermal oxidation, applying electrochemical impedance spectroscopy and cyclic voltammetry measurements. The strength of the method is evidenced when adsorption of small (12-mer) peptides is monitored. Our aim is to combine the advantages offered by the large sensing area of the PSi multilayers and the high sensitivity of the electrochemical technique for improved biosensing.

Abstract

International audience; Mineralization of calcium phosphate crystals was examined on porous silicon (PSi) scaffolds with respect to the dependence of crystal formation on the average pore size of PSi. PSi multilayer wafers fabricated by electrochemical etching were moni- tored via their reflectivity spectra over time in a biologically relevant osteogenic solution. The evolution of the mean refractive index of the PSi scaffold is the signature of the in situ crystals nucleation within the pores. Crystals were then characterized using atomic force microscopy (AFM), scanning electron microscopy (SEM) and Raman micro-spectroscopy. The results in- dicate that the average pore size of PSi highly influences nucleation and crystal formation.

Abstract

A challenge regarding the design of nanocarriers for drug delivery is to prevent their recognition by the immune system. To improve the blood residence time and prevent their capture by organs, nanoparticles can be designed with stealth properties using polymeric coating. In this study, we focused on the influence of surface modification with polyethylene glycol and/or mannose on the stealth behavior of porous silicon nanoparticles (pSiNP, ~200 nm). In vivo biodistribution of pSiNPs formulations were evaluated in mice 5 h after intravenous injection. Results indicated that the distribution in the organs was surface functionalization-dependent. Pristine pSiNPs and PEGylated pSiNPs were distributed mainly in the liver and spleen, while mannose-functionalized pSiNPs escaped capture by the spleen, and had higher blood retention. The most efficient stealth behavior was observed with PEGylated pSiNPs anchored with mannose that were the most excreted in urine at 5 h. The biodegradation kinetics evaluated in vitro were in agreement with these in vivo observations. The biocompatibility of the pristine and functionalized pSiNPs was confirmed in vitro on human cell lines and in vivo by cytotoxic and systemic inflammation investigations, respectively. With their biocompatibility, biodegradability, and stealth properties, the pSiNPs functionalized with mannose and PEG show promising potential for biomedical applications. [ABSTRACT FROM AUTHOR]

48. Porphyrin-Based Mesoporous Organosilica Nanoparticles for Two-Photon Rhabdomyosarcoma Targeting.

Abstract

Recently, we described porphyrin-based periodic mesoporous organosilica (PMO) nanoparticles synthesized from a large functional octatriethoxysilylated porphyrin precursor. The framework of the nanoparticles was formed by J-aggregates of porphyrins allowing two-photon excitation photodynamic therapy (TPE-PDT) and NIR imaging. In this study, we functionalized these PMO with polyethylene glycol (PEG) moieties and an analogue of mannose 6-phosphate functionalized on anomeric position (AMFA). These AMFA are known to efficiently target mannose 6-phosphate receptors (M6PR) which are over-expressed in various cancer cell lines (breast, prostate). Here we show that M6PR is also over-expressed in rhabdomyosarcoma (RMS) cell lines. We targeted this receptor with PMO-AMFA and efficiently performed TPE imaging and TPE-PDT of RMS cells. Furthermore the same treatment did not affect healthy myoblasts, which have a low expression of M6PR. [ABSTRACT FROM AUTHOR]
