2. Differential effects of isoproterenol on the activity of angiotensin-converting enzyme in the rat heart and aorta

Abstract

The excessive stimulation of beta-adrenergic receptors in the heart induces myocardial hypertrophy. There are several experimental data suggesting that this hypertrophy may also depend, at least partially, on the increase of local production of angiotensin II secondary to the activation of the cardiac renin-angiotensin system. In this study we investigated the effects of isoproterenol on the activity of angiotensin-converting enzyme (ACE) in the heart and also in the aorta and plasma. Male Wistar rats weighing 250 to 305 g were treated with a dose of (±)-isoproterenol (0.3 mg kg-1 day-1, N = 8) sufficient to produce cardiac hypertrophy without deleterious effects on the pumping capacity of the heart. Control rats (N = 7) were treated with vehicle (corn oil). The animals were killed one week later. ACE activity was determined in vitro in the four cardiac chambers, aorta and plasma by a fluorimetric assay. A significant hypertrophy was observed in both ventricular chambers. Abstract

Elevated blood pressure remains the single biggest risk factor contributing to the global burden of disease and mortality. May Measurement Month is an annual global screening campaign aiming to improve awareness of blood pressure at the individual and population level. Adults (≥18 years) recruited through opportunistic sampling were screened at sites in 92 countries during May 2019. Ideally, 3 blood pressure readings were measured for each participant, and data on lifestyle factors and comorbidities were collected. Hypertension was defined as a systolic blood pressure ≥140 mm Hg, or a diastolic blood pressure ≥90 mm Hg (mean of the second and third readings) or taking antihypertensive medication. When necessary, multiple imputation was used to estimate participants' mean blood pressure. Mixed-effects models were used to evaluate associations between blood pressure and participant characteristics. Of 1 508 130 screenees 482 273 (32.0%) had never had a blood pressure measurement before and 513 337 (34.0%) had hypertension, of whom 58.7% were aware, and 54.7% were on antihypertensive medication. Of those on medication, 57.8% were controlled to <140/90 mm Hg, and 28.9% to <130/80 mm Hg. Of all those with hypertension, 31.7% were controlled to <140/90 mm Hg, and 350 825 (23.3%) participants had untreated or inadequately treated hypertension. Of those taking antihypertensive medication, half were taking only a single drug, and 25% reported using aspirin inappropriately. This survey is the largest ever synchronized and standardized contemporary compilation of global blood pressure data. This campaign is needed as a temporary substitute for systematic blood pressure screening in many countries worldwide.

Abstract

Aims: To investigate the anti-inflammatory activity of an invasive and Hp65- producing strain Lactococcus lactis NCDO2118 FnBPA+ (pXYCYT:Hsp65) in acute 2,4,6-trinitrobenzene sulphonic acid (TNBS)-induced colitis in mice as an innovative therapeutic strategy against Crohn's disease (CD). Methods and Results: The pXYCYT:Hsp65 plasmid was transformed into the L. lactis NCDO2118 FnBPA+ strain, resulting in the L. lactis NCDO2118 FnBPA+ (pXYCYT:Hsp65) strain. Then, the functionality of the strain was evaluated in vitro for Hsp65 production by Western blotting and for invasion into Caco-2 cells. The results demonstrated that the strain was able to produce Hsp65 and efficiently invade eukaryotic cells. Subsequently, in vivo, the anti-inflammatory capacity of the recombinant strain was evaluated in colitis induced with TNBS in BALB/c mice. Oral administration of the recombinant strain was able to attenuated the severity of colitis by mainly reducing IL-12 and IL-17 levels and increasing IL-10 and secretory immunoglobulin A levels. Conclusions: The L. lactis NCDO2118 FnBPA+ (pXYCYT:Hsp65) strain contributed to a reduction in inflammatory damage in experimental CD. Significance and Impact of the Study: This study, which used L. lactis for the production and delivery of Hsp65, has scientific relevance because it shows the efficacy of this new strategy based on therapeutic protein delivery into mammalian enterocytes. Abstract

CfAvr4, a chitin-binding effector protein produced by the Dothideomycete tomato pathogen Cladosporium fulvum, protects the cell wall of this fungus against hydrolysis by secreted host chitinases during infection. However, in the presence of the Cf-4 immune receptor of tomato, CfAvr4 triggers a hypersensitive response (HR), which renders the pathogen avirulent. Recently, several orthologues of CfAvr4 have been identified from phylogenetically closely related species of Dothideomycete fungi. Of these, DsAvr4 from Dothistroma septosporum also triggers a Cf-4-dependent HR, but CaAvr4 and CbAvr4 from Cercospora apii and Cercospora beticola, respectively, do not. All, however, bind chitin. To identify the region(s) and specific amino acid residue(s) of CfAvr4 and DsAvr4 required to trigger a Cf-4-dependent HR, chimeric and mutant proteins, in which specific protein regions or single amino acid residues, respectively, were exchanged between CfAvr4 and CaAvr4 or DsAvr4 and CbAvr4, were tested for their ability to trigger an HR in Nicotiana benthamiana plants transgenic for the Cf-4 immune receptor gene. Based on this approach, a single region common to CfAvr4 and DsAvr4 was determined to carry a conserved proline residue necessary for the elicitation of this HR. In support of this result, a Cf-4-dependent HR was triggered by mutant CaAvr4 and CbAvr4 proteins carrying an arginine-to-proline substitution at this position. This study provides the first step in deciphering how Avr4 orthologues from different Dothideomycete fungi trigger a Cf-4-dependent HR.

Face ?s dificuldades que o setor da olivicultura enfrenta para escoar os produtos, ? necess?rio encontrar alternativas para reduzir as perdas e ao mesmo tempo valorizar o produto em causa. Assim, a ind?stria precisa de estar constantemente a inovar de forma a dar resposta ?s necessidades dos produtores e satisfazer os desejos de um maior n?mero de consumidores. Neste trabalho estudou-se o tempo de vida de quatro tipos de pasta de azeitona pasteurizada, embaladas em bisnagas, durante 120 dias de armazenamento. Inicialmente foi realizado um estudo preliminar onde foram elaboradas v?rias pastas de azeitona, com diferentes ingredientes, de forma a obter a formula??o otimizada, com as caracter?sticas desejadas, nomeadamente de textura, facilidade em barrar (f?cil espalhabilidade), homog?nea, baixa granulometria e sem separa??o de fases. A qualidade das pastas de azeitona foi monitorizada atrav?s da contagem de microrganismos a 30?C, par?metros f?sico-qu?micos (pH, atividade da ?gua, cor, teor de s?lidos sol?veis e textura) e avalia??o sensorial (an?lise quantitativa descritiva e teste de consumidores). A an?lise sensorial mostrou que os provadores detetaram que todas as pastas de azeitona apresentaram alguns defeitos nos atributos cheiro e sabor, principalmente a partir do 30? dia de armazenamento. Observou-se que existem diferen?as significativas entre as pastas de azeitona (preta e verde) em alguns par?metros sensoriais, com signific?ncia estat?stica (p0,92), tendo-se observado a partir desse dia um decr?scimo nestes valores, at? aw=0,84. N?o se registaram grandes altera??es da firmeza e da adesividade da textura das pastas de azeitona, ao longo do tempo de armazenamento. Conclui-se que, at? ao 62? dia de armazenamento, as diferentes pastas de azeitona revelaram qualidade microbiol?gica adequada durante o tempo de armazenamento, sendo consideradas seguras para o consumidor. No entanto, a an?lise sensorial atua como fator limitante, sendo que o tempo de vida das pastas de azeitona em bisnagas ? 30 dias, uma vez que a partir do 30? dia de armazenamento j? n?o apresentavam qualidade suficiente para os provadores. Ainda se pode concluir que, tendo em conta a an?lise sensorial, as pastas de azeitona que evidenciaram melhores resultados nas caracter?sticas positivas e com menor perda das caracter?sticas organol?ticas foram as pastas de azeitona preta com alho e com alecrim. Table olives are a typical Portuguese cuisine ingredient and an important component of Mediterranean diets. Several olive varieties are cultivated in Portugal. Olives are fermented and processed in order to remove bitterness and improve its preservation. Given the difficulties that the olive growing sector faces to sell the products, it is necessary to find alternatives to reduce losses and at the same time add value to this important agricultural product. Thus, the industry needs to be constantly innovating in order to meet the needs of producers and satisfy the wishes of a greater number of consumers. This work studied the lifespan of four types of pasteurized olive pastes, packaged in tubes, during 120 days of storage. A preliminary study was conducted using several olive pastes with different ingredients in order to obtain the optimized formulation, with the desired characteristics, such as texture, with high spreadability, homogeneous, with low particle size and without phase separations. Olive paste quality was monitored through total counts of microorganisms to 30?C, physico-chemical parameters (pH, water activity, color, soluble solids and texture) and sensory evaluation (quantitative descriptive analysis and acceptability (consumer) tests. Sensory analysis showed that judges perceived that olive pastes presented some defects in smell and taste attributes, mainly from the 30th day of storage onwards. Significant differences (p < 0.05) between black and green olive pastes were detected in some sensory parameters, along the storage time. In what concerns consumer opinions, green olive pastes were those that have obtained higher acceptance, which was expected due to the fact that green olives confer a more pleasant acidic taste. Nevertheless, both black and green olive pastes were faced moderately pleasant. Microbiological results were below the regulated limit for microorganism counts at 30? C, in all samples, along storage time, and could therefore be considered as safe for the consumer, relatively to this parameter In assessing physicochemical parameters, some changes over storage time were registered, namely an increase in Lightness (L *) and pH was evolved in a similar manner. It was seen that the pH of green olive pastes was always below 4, except on the 90th day, when pH exceeded 5. All samples had high water activity (aw) until the 6th day of storage (aw > 0.92), and showed a decrease afterwards until stabilization at aw = 0.84. There were no major changes in firmness and adhesiveness of the texture of olive folders, along storage time. It is concluded that until the 62nd day of storage, the different olive-folders presented appropriate microbiological quality during storage time, being considered safe for the consumer. However, the sensory analysis acts as a limiting factor, so the lifespan of olive in tubes is 30 days, cause from the 30th day of storage no longer had sufficient quality to the tasters. It also can be concluded that, given the sensory analysis, olive pastes that showed best results in positive characteristics and with less loss of organoleptic characteristics were black olive pastes with garlic and Rosemary.

41. Oclusão Venosa Central Retiniana e Trombofilias - A Propósito de Um Caso Clínico

Abstract

Introdução: A incidência da oclusão venosa central de retina (OVCR) varia de 2 a 8 por 1,000 e aumenta com a idade. Os factores de risco cardiovasculares, algumas trombofilias e síndromes de hiperviscosidade podem facilitar o aparecimento da OVCR. A mutação do gene da protrom- bina (G20210A) aumenta os níveis de protrombina plasmática em 30%, elevando assim o risco de trombose venosa. Não existe, no entanto, nenhum estudo científico que mostre a associação entre OVCR e esta mutação. Caso clinico: Homem de 21 anos com diminuição súbita da acuidade visual do OD, sem ante- cedentes patológicos conhecidos. Ao exame objectivo apresentava uma AV corrigida OD 0,12 e AV OE 1,0, pressão intra-ocular OD de 30mmHg e OE de 34 mmHg. A fundoscopia demonstrou edema da papila, ingurgitamento venoso temporal e nasal e hemorragias superficiais no OD. Realizou estudo imagiológico crâneo-encefálico e das órbitas, bem como analítico e genético. Detectou-se heterozigotia para a variante da protrombina 20210A. Iniciou terapêutica dirigida para a hipertensão ocular e anti-agregação plaquetária, e fez também uma injecção intra-vítrea única de Bevacizumab 1,25mg por apresentar apresentar edema macular. A evolução foi favo- rável com AV ODE=1,0. Conclusão: Apesar da associação entre a mutação do gene da protrombina e oclusão venosa da retina não estar bem esclarecida, os portadores desta mutação têm um risco aumentado de trom- bose venosa. A OVCR tem uma origem multifactorial, neste caso a hipertensão ocular poderá ser o principal factor de risco, no entanto a descoberta desta mutação é um factor que poderá elevar ainda mais este risco num doente jovem., Revista Sociedade Portuguesa de Oftalmologia, Vol. 37 N.º 1 (2013)

Abstract

In disease-suppressive soils, plants are protected from infections by specific root pathogens due to the antagonistic activities of soil and rhizosphere microorganisms. For most disease-suppressive soils, however, the microorganisms and mechanisms involved in pathogen control are largely unknown. Our recent studies identified Actinobacteria as the most dynamic phylum in a soil suppressive to the fungal root pathogen Rhizoctonia solani. Here we isolated and characterized 300 isolates of rhizospheric Actinobacteria from the Rhizoctonia-suppressive soil. Streptomyces species were the most abundant, representing approximately 70% of the isolates. Streptomyces are renowned for the production of an exceptionally large number of secondary metabolites, including volatile organic compounds (VOCs). VOC profiling of 12 representative Streptomyces isolates by SPME-GC-MS allowed a more refined phylogenetic delineation of the Streptomyces isolates than the sequencing of 16S rRNA and the house-keeping genes atpD and recA only. VOCs of several Streptomyces isolates inhibited hyphal growth of R. solani and significantly enhanced plant shoot and root biomass. Coupling of Streptomyces VOC profiles with their effects on fungal growth, pointed to VOCs potentially involved in antifungal activity. Subsequent assays with five synthetic analogs of the identified VOCs showed that methyl 2-methylpentanoate, 1,3,5-trichloro-2-methoxy benzene and the VOCs mixture have antifungal activity. In conclusion, our results point to a potential role of VOC-producing Streptomyces in disease suppressive soils and show that VOC profiling of rhizospheric Streptomyces can be used as a complementary identification tool to construct strain-specific metabolic signatures

49. Recycling of textile waste by reinforcing earthy render with micro-fiber

Abstract

Earth construction is worldwide spread. It is a valuable heritage to preserve because of its cultural and socio-economical relevance. Furthermore, earth construction, since it needs significantly less energy than modern construction techniques, and works with local materials, it contributes for thesustainable development. In Portugal, there is an impressive earth construction heritage that requires maintenance. Among the Portuguese traditional building techniques related to earth construction, the rammed earth, the adobe and the tabique are the most relevant ones. This work is focused on attempting to apply textile waste micro-fiber as an alternative sustainable reinforcement solution of earthy render of tabique walls. Therefore, several earthy render samples reinforced with 1% textile waste micro-fiber content were prepared and mechanically tested. The main experimental results are presented and discussed, and the main conclusions are drawn. This study also contributes for recycling a specific waste, namely short fibers from needling machines used to produce nonwoven fabrics.

Abstract

Portugal is considered by the World Health Organization (WHO) a risk country for the practice of Female Genital Mutilation (FGM). Objectives: To evaluate the knowledge that health professionals from Maternity Dr. Alfredo da Costa (MAC) have regarding FGM. Population and Methods: Analysis of surveys delivered to health professionals from MAC (a hospital dedicated to reproductive health), between April and June 2008, addressing issues related to the knowledge about FGM. Results: Authors collected 112 valid surveys involving 38 doctors, 48 nurses and 26 medical auxiliaries/administrative personnel. From the respondents, 106 (95%) had heard about FMG practice before, the media being the most reported source of information; 59 (53%) replied they could be able to recognize FGM cases in their clinical practice; however, only 31 (28%) claimed to know the FGM type classiication and 32 (29%) admitted to be prepared to recognize and manage these situations in their own clinical practice; 9 had been consulted explicitly by a FGM practice complication and 1 doctor had admitted having been asked to perform/execute FGM; 13 (12%) recognized that the Portuguese legislation its this practice. Regarding the practice of FGM, 100 (89%) of respondentes stated that it should not be maintained and 97 (87%) stated that it should not be tolerated. However, 42 (38%) considered that if these practices were a reality, then they should be medical assisted. Discussion: Health professionals can play an important role in eliminating the practice of FGM, not only by the proper clinical management of this situation, but also by preventing those communities at risk to resort to FGM. Most health professionals are not prepared to deal with FGM in their clinical practice. It is important to promote a better knowledge on the subject and to create protocols for proper clinical management.
