A complete and comprehensive chemical and biological study of Drimys granadensis, a native Ecuadorian aromatic plant, was conducted. By conventional steam distillation from dried leaves, a yellowish, translucent essential oil (EO) with a density of 0.95 and a refractive index of 1.5090 was obtained. The EO was analyzed by gas chromatography coupled to a mass spectrometer (GC/MS) and an FID detector (GC/FID), respectively. Enantiomeric distribution was also carried out by GC/MS using a chiral selective column (diethyl tert-butylsilyl-BETA-cyclodextrin). The microdilution broth method was employed to assess the antibacterial and antifungal activity of the EO against a panel of opportunistic microorganisms. Antioxidant capacity was measured using diphenyl picryl hydrazyl (DPPH) and azino-bis 3-ethylbenzothiazoline-6-sulfonic acid (ABTS) radicals. Finally, the inhibitory potential of the EO against acetylcholinesterase was also valued. Sixty-four chemical compounds, constituting 93.27% of the total composition, were identified, with major components including γ-muurolene (10.63%), spathulenol (10.13%), sabinene (5.52%), and δ-cadinene (4.22%). The characteristic taxonomic marker of the Drimys genus, Drimenol, was detected at very low percentages (<2%). Two pairs of enantiomers ((1S,5R)-(+)-α-pinene/(1S,5S)-(–)-α-pinene; (1S,5R)-(+)-β-pinene/(1S,5S)-(–)-β-pinene) and one pure enantiomer (1R,4S)-(–)-camphene were identified. Regarding antimicrobial potency, the EO exhibited a significant moderate effect on Listeria monocytogenes with a minimal inhibitory concentration (MIC) value of 250 µg/mL, while with the remaining microorganisms, it exerted less potency, ranging from 500 to 2000 µg/mL. The EO displayed moderate effects against the ABTS radical with a half scavenging capacity of 210.48 µg/mL and no effect against the DPPH radical. The most notable effect was noticed for acetylcholinesterase, with a half inhibition concentration (IC50) of 63.88 ± 1.03 µg/mL. These antiradical and anticholinesterase effects hint at potential pharmacological applications in Alzheimer's disease treatment, although the presence of safrole, albeit in low content (ca. 2%), could limit this opportunity. The first chemical and enantioselective analyses of the essential oil from the leaves of Mauria heterophylla Kunth were carried out. The dry plant materialproduced an essential oil with a 0.09 ± 0.05% yield by weight. The volatile fraction was submitted to GC-MS (qualitative) and GC-FID (quantitative) analyses, on two different stationary phases. A total of forty-four constituents were detected and quantified with at least one column, corresponding to about 85.5% of the whole oil mass. Limonene (15.2%), (E)-β-caryophyllene (11.1%), α-phellandrene (10.9%), germacrene D (6.6%), α-pinene (6.4%), myrcene (3.9%), β-phellandrene (2.8%), bicyclogermacrene (2.8%), (E)-β-farnesene (2.2%) and δ-cadinene (1.2%) were major components. Furthermore, the study was complemented with the enantioselective analysis of eleven common chiral terpenes, carried out on two β-cyclodextrin-based chiral selectors. Among the chiral metabolites, (1R,4S)-(+)-camphene, (1R,5R)-(+)-β-pinene, (S)-(+)-α-phellandrene and (S)-(+)-β-phellandrene were enantiomerically pure, whereas α-pinene, sabinene, limonene, linalool, terpinen-4-ol, α-terpineol and germacrene D were present as scalemic mixtures. In Ecuador, Hedyosmum cumbalense and Hedyosmum spectabile are valued for their well-known aromatic characteristics and therapeutic benefits. In this study, fresh and dried leaves of these species were steam-distilled to obtain their essential oils (EOs) for chemical characterization and assessment of their antimicrobial, antioxidant, and anticholinesterase properties. Gas chromatography coupled to mass spectrometry (GC-MS) and a flame-ionized detector (GC-FID) with a nonpolar column was employed to determine the chemical composition, along with the enantioselective analysis. The antimicrobial activity was evaluated against three Gram-positive, two Gram-negative, and two sporulated fungi. The radical scavenging properties were evaluated by DPPH and ABTS assays. A total of 66 and 57 compounds were identified for H. cumbalense and H. spectabile, respectively. Two pairs of enantiomers for each species were also detected, with (1R,5R)-(+)-sabinene and (1S,5S)-(–)-sabinene found in both specimens. A poor effect against Gram-positive cocci was observed on H. cumbalense (MIC of 4000 µg/mL). Both oils displayed weak antifungal activities, exhibiting a MIC of 1000 µg/mL. H. cumbalense had a good scavenging effect assessed by the ABTS radical (SC50 = 96.02 ± 0.33 µg/mL). Both EOs showed a strong anticholinesterase potential with an IC50 value of 61.94 ± 1.04 µg/mL for H. cumbalense and 21.15 ± 1.03 µg/mL for H. spectabile. [ABSTRACT FROM AUTHOR]

The tropical montane forests of the E Andean cordillera in Ecuador receive episodic Sahara-dust inputs particularly increasing Ca deposition. We added CaCl2 to isolate the effect of Ca deposition by Sahara dust to tropical montane forest from the simultaneously occurring pH effect. We examined components of the Ca cycle at four control plots and four plots with added Ca (2 × 5 kg ha-1 Ca annually as CaCl2) in a random arrangement. Between August 2007 and December 2009 (four applications of Ca), we determined Ca concentrations and fluxes in litter leachate, mineral soil solution (0.15 and 0.30 m depths), throughfall, and fine litterfall and Al concentrations and speciation in soil solutions. After 1 y of Ca addition, we assessed fine-root biomass, leaf area, and tree growth. Only < 3% of the applied Ca leached below the acid organic layer (pH 3.5-4.8). The added CaCl2 did not change electrical conductivity in the root zone after 2 y. In the second year of fertilization, Ca retention in the canopy of the Ca treatment tended to decrease relative to the control. After 2 y, 21% of the applied Ca was recycled to soil with throughfall and litterfall. One year after the first Ca addition, fine-root biomass had decreased significantly. Decreasing fine-root biomass might be attributed to a direct or an indirect beneficial effect of Ca on the soil decomposer community. Because of almost complete association of Al with dissolved organic matter and high free Ca2+ : Al3+ activity ratios in solution of all plots, Al toxicity was unlikely. A novel chemical profile essential oil, distilled from the aerial parts of Clinopodium taxifolium (Kunth) Govaerts (Lamiaceae), was analysed by Gas Chromatography-Mass Spectrometry (GC-MS, qualitative analysis) and Gas Chromatography with Flame Ionization Detector (GC-FID, quantitative analysis), with both polar and non-polar stationary phase columns. The chemical composition mostly consisted of sesquiterpenes and sesquiterpenoids (>70%), the main ones being (E)-β-caryophyllene (17.8%), α-copaene (10.5%), β-bourbonene (9.9%), δ-cadinene (6.6%), cis-cadina-1(6),4-diene (6.4%) and germacrene D (4.9%), with the non-polar column. The essential oil was then submitted to enantioselective GC analysis, with a diethyl-tert-butyldimethylsilyl-β-cyclodextrin diluted in PS-086 chiral selector, resulting in the following enantiomeric excesses for the chiral components: (1R,5S)-(−)-α-thujene (67.8%), (1R,5R)-(+)-α-pinene (85.5%), (1S,5S)-(−)-β-pinene (90.0%), (1S,5S)-(−)-sabinene (12.3%), (S)-(−)-limonene (88.1%), (S)-(+)-linalool (32.7%), (R)-(−)-terpinen-4-ol (9.3%), (S)-(−)-α-terpineol (71.2%) and (S)-(−)-germacrene D (89.0%). The inhibition activity against acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) of C. taxifolium essential oil was then tested, resulting in selective activity against BChE with an IC50 value of 31.3 ± 3.0 μg/mL (positive control: donepezil, IC50 = 3.6 μg/mL). The essential oils of Myrcianthes myrsinoides and Myrcia mollis, belonging to the Myrtaceae family, were obtained by steam distillation. They were analyzed by gas chromatography-mass spectrometry (GC-MS), gas chromatography-flame ionization detector (GC-FID), enantioselective gas chromatography, and gas chromatography-olfactometry (GC-O). A total of 58 compounds for Myrcianthes myrsinoides essential oil (EO) and 22 compounds for Myrcia mollis EO were identified and quantified by GC-MS with apolar and polar columns (including undetermined components). Major compounds (>5.0%) were limonene (5.3%–5.2%), 1,8-cineole (10.4%–11.6%), (Z)-caryophyllene (16.6%–16.8%), trans-calamenene (15.9%–14.6%), and spathulenol (6.2%–6.5%). The enantiomeric excess of eight chiral constituents was determined, being (+)-limonene and (+)-germacrene D enantiomerically pure. Eight components were identified as determinant in the aromatic profile: α-pinene, β-pinene, (+)-limonene, γ-terpinene, terpinolene, linalool, β-elemene and spathulenol. For M. mollis, the major compounds (>5.0%) were α-pinene (29.2%–27.7%), β-pinene (31.3%–30.0%), myrcene (5.0%–5.2%), 1,8-cineole (8.5%–8.7%), and linalool (7.7%–8.2%). The enantiomeric excess of five chiral constituents was determined, with (S)-α-pinene and (+)-germacrene D enantiomerically pure. The metabolites β-pinene, 1,8-cineole, γ-terpinene, terpinolene, linalool, and (E)-β-caryophyllene were mainly responsible for the aroma of the EO. Finally, the M. myrsinoides essential oil has an inhibitory activity for cholinesterase enzymes (IC50 of 78.6 μg/mL and 18.4 μg/mL vs. acethylcholinesterase (AChE) and butyrylcholinesterase (BChE) respectively). In this research, the leaves of Lepechinia heteromorpha (Briq.) Epling, Lepechinia radula (Benth.) Epling and Lepechinia paniculata (Kunth) Epling have been collected in order to perform a phytochemical study. The first species was distilled to obtain a novel essential oil (EO), while the others were submitted to ethyl acetate extraction and secondary metabolite isolation. The chemical composition of the EO from L. heteromorpha has been investigated by Gas Chromatography-Mass Spectrometry (GC-MS) and Gas Chromatography with Retention Indices (GC(RI)), identifying 25 constituents. A major compound, (−)-ledol (21.2%), and a minor compound, (−)-caryophyllene oxide (1.0%), were isolated from the EO and their structures confirmed by Nuclear Magnetic Resonance (NMR) spectroscopy. Other major constituents of the EO were viridiflorene (27.3%), (E,E)-α-farnesene (1.4%), spirolepechinene and (E)-β-caryophyllene (7.1% each), allo-aromadendrene (6.1%), camphor (1.7%), limonene (1.3%) and β-phellandrene (4.6%). The enantiomeric composition of the EO monoterpene fraction was also studied, determining the enantiomeric excess and distribution of α-pinene, limonene, β-phellandrene and camphor. The ethyl acetate extract of L. radula and L. paniculata were fractionated by column chromatography. Spathulenol, angustanoic acid E and 5-hydroxy-4′,7-dimethoxy flavone were isolated from L. radula extract; ledol, guaiol and (−)-carnosol were found in L. paniculata. [ABSTRACT FROM AUTHOR]

Phytochemical investigation of Croton thurifer, collected in Loja-Ecuador, led to the isolation of seven known compounds identified as: (3R, 20S) -3-palmitate-20-hydroxydammar-24-ene (1); (3R, 20S)-3- acetoxy-20-hydroxydammar-24-ene (2); trans-phytol (3); vomifoliol (4); ß-sitosterol (5); trans-tiliroside (6) and sparsifol (7). The structures of the isolated compounds were determined by NMR, MS, as well as by comparison with literature data. The hypoglycemic activity of the crude extracts and isolated compounds was assessed by their ability to inhibit a-glucosidase activity. The hexane and methanol extracts did not show any inhibitory activity on a-glucosidase, while the ethyl acetate extract (EtOAc) showed very low inhibitory activity with IC50= 1.77 mg/mL. Two of the seven isolated compounds exhibited good inhibitory activity with IC50 values much higher than acarbose (377 µg/mL). trans-tiliroside was the compound with the highest inhibitory activity (IC50 = 114.85 µg/mL), while (3R, 20S)-3-Acetoxy-20-hydroxydammar-24-ene had moderate inhibitory activity (IC50=292.87). Trans-tiliroside exherted a non-competitive inhibition with Km values between 368 and 358 µM and Vmax value from 695 and 325 nM/min for unhibited and inhibited reaction, respectively and a Ki of 163,6 µM.
