Abstract Background Culicoides, also known as biting midges, carry pathogens which include Mansonella perstans. Mansonella perstans is a nematode parasite implicated in a number of disease outcomes. Even though a high prevalence of about 75% M. perstans infection has been recorded in some communities in the middle belt of Ghana, and a wide diversity of Culicoides species has been identified, the exact Culicoides species transmitting M. perstans in Ghana has not yet been deciphered. This study therefore aimed at assessing the species diversity of Culicoides and their role in the transmission of M. perstans in the middle belt of Ghana. Methods Culicoides species were sampled from 11 communities in the Asante-Akim North and Sene West districts in the middle belt of Ghana. Centre for Disease Control (CDC) UV light traps, as well as human bait (i.e. human landing catch and engorged catch) methods were used to assess the species abundance and diversity of Culicoides in the study communities in the wet and dry season. A colorimetric Loop-Mediated Isothermal Amplification (LAMP) assay was performed to assess the vector competence of the various Culicoides species. Results A total of 4810 Culicoides from 6 species were sampled. These included Culicoides inornatipennis, C. milnei, C. schultzei, C. grahamii, C. neavei, and C. imicola. Culicoides imicola was the most abundant species (56%) followed by C. grahamii (16%). Light traps sampled the most diverse species (6 species). Human landing catch and engorged catch methods identified three anthropophilic species, C. grahamii, C. milnei, and C. inornatipennis, with C. grahamii being the most anthropophilic with a peak biting time between the hours of 5 p.m. to 6 p.m. Generally, there was relatively higher species abundance in the wet than dry season. LAMP assay identified C. grahamii as the potential vector for M. perstans transmission in the middle belt of Ghana. Conclusions For the first time, we have demonstrated that C. grahamii is the potential competent vector for M. perstans transmission in the middle belt of Ghana. Little is known about the Culicoides fauna in the rain forest belt of the Littoral Region of Cameroon. This study was designed to investigate the diversity, abundance and distribution of Culicoides spp. and their role as the purported vector(s) of M. perstans. Overnight light trap collections and human landing catches (HLCs) revealed eight species of Culicoides with C. grahamii being the most abundant species followed by C. milnei. Four anthropophilic species (C. inornatipennis, C. grahamii, C. fulvithorax and C. milnei) were determined by the HLCs with a higher abundance in the 4–6 p.m. collections. The drop trap technique and Mp419 LAMP assay confirmed C. milnei to be the most efficient vector in enabling the development of the microfilarial stage to the infective larval form of M. perstans. The LAMP assay also revealed that natural transmission of this nematode is fostered by C. milnei and C. grahamii in the wild. Abstract Background Biting midges in the genus Culicoides (Diptera: Ceratopogonidae) transmit bluetongue virus (BTV) and epizootic hemorrhagic disease virus (EHDV) to ruminants, thus exerting a significant economic impact on animal agriculture worldwide. However, very little is known about the larval habitat characteristics of Culicoides species associated with BTV/EHDV transmission, particularly in southeastern USA, limiting the establishment of effective midge control strategies. In this study, we examined the habitat associations of Culicoides species abundant on a commercial cervid farm in Florida, USA and quantified several environmental variables of their habitat to identify the key variables associated with midge abundance. Methods Mud/substrate samples from three potential larval habitats on the farm (edges of streams, puddles and seepages) were brought to the laboratory and incubated for adult emergence, and the percentage organic matter, macronutrients, micronutrients, pH, electrical conductivity, moisture and microbial concentrations of the substrate were quantified. Results Strong habitat associations were observed for Culicoides haematopotus (Malloch) (stream edge), Culicoides stellifer (Coquillett) (puddles) and Culicoides loisae (Jamnback) (stream edge), the most commonly emerging midge species from the samples. Suspected vector species of BTV/EHDV on the property, C. stellifer and Culicoides venustus (Hoffman), emerged mainly from habitats with moderate-high levels of pollution (edges of puddles and seepages) as indicated by the relatively higher concentrations/levels of organic matter, nutrients and other environmental variables in these samples. The emergence of C. insignis was too low to form any meaningful conclusions. For each Culicoides species, only weak positive or negative associations were detected between midge abundance and the various environmental variables quantified. Conclusions Habitat associations of Culicoides species abundant on a local cervid/animal farm vary, most likely as a function of certain biotic/abiotic characteristics of the habitat. Further studies across a larger spatial and temporal scale will be needed to experimentally evaluate/identify the key factors more strongly associated with the abundance of target Culicoides species. This information, in the long term, can be potentially exploited to render local habitats unsuitable for midge oviposition/larval development.

When present, they are often considered a biting nuisance but in addition, they are involved in the transmission of pathogens to humans, domestic and wild animals. Data on Culicoides species in the South-West region of Cameroon dates back to the 1950s. Over the decades, ecological transformation due to agriculture and deforestation may have affected the population dynamics of Culicoides and therefore our study provides an update of their bio-ecology in the region. Furthermore, the role of various Culicoides species in the transmission of parasitic filariae of the genus Mansonella remains inconclusive in this region. This study was designed to address these unknown issues and expand on current scientific knowledge. Results Eight species of Culicoides (C. bedfordi, C. inornatipennis, C. fulvithorax, C. grahamii, C. imicola, C. milnei, C. neavei and C. kumbaensis) were collected using light traps and human baits. The preferential attraction of adult Culicoides (Diptera: Ceratopogonidae) to specific wavelengths was studied in West Bengal, India. A total of 280 collections were made with suction light traps fitted with various colours of light emitting diodes (LEDs) placed at cattle sheds during June, August and November in 2017. In addition, the numbers that have been collected in the evening and morning were compared. Locally manufactured suction light traps fitted with ultra violet (UV), blue, green, yellow, pink, red and white LEDs were compared. UV light attracted the highest number of midges followed by blue and then green LED. Culicoides peregrinus Kieffer and Culicoides oxystoma Kieffer were the most abundant followed by Culicoides fulvus Sen and Das Gupta, Culicoides innoxius Sen and Das Gupta, Culicoides anophelis Edwards and Culicoides huffi Causey. The species composition remained similar across the wavelengths. Although significant variations in midge population have been observed across the months, no significant difference in dusk and dawn abundance was noticed. To better understand the biology of local Culicoides species, a study divided into three parts was performed in northeast Germany to elucidate the feeding activity patterns (study A), preferential landing and feeding sites (study B) and host feeding preferences (study C) of Culicoides spp. using cattle and sheep as baits. Methods In study A, the activity of Culicoides spp. was monitored over a 72 h period by collecting insects at regular intervals from the interior of drop traps with cattle or sheep standing inside. In study B, Culicoides spp. were directly aspirated from the coat and fleece of cattle and sheep during the peak activity period of Culicoides. In study C, Culicoides spp. were collected using drop traps with either cattle or sheep standing inside and located 10 m apart. Results In study A, 3,545 Culicoides midges belonging to 13 species were collected, peak activity was observed at sunset. In study B, 2,024 Culicoides midges were collected. A significantly higher number of midges was collected from the belly and flank of cattle in comparison to their head region. In study C, 3,710 Culicoides midges were collected; 3,077 (83%) originated from cattle and 633 (17%) from sheep. Nearly half (46.7%) of the midges collected from cattle were engorged, significantly more than the number of engorged midges collected from sheep (7.5%). Culicoides from the Obsoletus complex (C. obsoletus and C. scoticus) were the most common Culicoides species encountered, followed by C. punctatus. Other species identified were C. dewulfi, C. chiopterus, C. pulicaris, C. lupicaris, C. pallidicornis, C. subfascipennis, C. achrayi, C. stigma, C. griseidorsum and C. subfagineus, the last two species are reported for the first time in Germany. Engorged C. chiopterus were collected in relatively high numbers from sheep, suggesting that this species may have a preference for sheep. Insect bite hypersensitivity (IBH) is a seasonal allergic skin disease in horses caused by bites of certain Culicoides spp. The aim of our study was to investigate the maternal effect on IBH and to estimate the heritability and repeatability of IBH in the Dutch Friesian horse population. Data consisted of 3,453 Dutch Friesian broodmares with 3,763 visual observations on IBH clinical symptoms scored by 12 inspectors during organized foal inspections in 2004 and 2008. Nine percent of the mares (n = 310) were scored in both years. Mares descended from 144 sires and 2,554 dams and 26.2% of the dams (n = 669) had more than 1 offspring in the data set (range: 2 to 6). Insect bite hypersensitivity was analyzed as a binary trait with a threshold animal model with and without a maternal effect, using a Bayesian approach. Observed IBH prevalence in Dutch Friesian broodmare population was 18.2%. Heritability on the liability scale was 0.16 (SD = 0.06); heritability on the observed scale was 0.07; and repeatability was 0.89 (SD = 0.03). Maternal effect was 0.17 (SD = 0.06) and significantly differed from zero, although the animal model without a maternal effect fitted the data better. These results show that genetic and permanent environmental factors affect IBH in Dutch Friesian horses. This paper reviews the species composition and distribution of Culicoides biting midges in Turkey for the first time. The Culicoides fauna of Turkey is diverse, with 57 reported species collected in 54 localities across the Marmara, Aegean, Mediterranean, and Central, Eastern and Southeastern Anatolian Regions of Turkey. No records are available for the Black Sea Region. The most widespread species include Culicoides cataneii Clastrier, C. circumscriptus Kieffer, C. festivipennis Kieffer, C. gejgelensis Dzhafarov, C. longipennis Khalaf, C. maritimus Kieffer, C. nubeculosus (Meigen), C. obsoletus (Meigen), C. odiatus Austen, C. pulicaris (Linnaeus), C. puncticollis (Becker), C. saevus Kieffer, and C. shaklawensis Khalaf, whereas C. denisoni Boorman, C. dewulfi Goetghebuer, C. fagineus Edwards, C. minutissmus (Zetterstedt), C. montanus Schakirzjanova, C. semimaculatus Clastrier, C. slovacus Orszagh, and C. turanicus Gutsevich and Smatov are represented from only one locality. Only single specimens represent the country records of C. dewulfi and C. seifadinei. The potential of Turkish Culicoides in zoonotic arbovirus, protozoa, and nematode transmission is discussed. The clinical, epidemiological and histopathological findings of two pruritic dermatites in sheep in Israel are described. The first type of dermatitis affected mainly young animals with lesions predominantly on the legs. It occurred from March to November, with a peak in June. The second type affected animals of all ages and was mainly on the ventrum. It was sporadic but occurred throughout the year with a peak in October. The morbidity rate of this syndrome reached 4.3% in one flock. The histopathology of both conditions was consistent with an allergic dermatitis. Fleas and midges were collected and identified asCtenocephalides felis felis andvarious species ofCulicoides. The population density, seasonal activity, geographical distribution and feeding behaviour preferences of the insects and the incidence of the two types of dermatitis suggest that fleas and midges were the causal agents. Flea and midge bite pruritic dermatoses should be considered in the differential diagnosis of sarcoptic and psoroptic mange. [ABSTRACT FROM AUTHOR]

After overnight fly collections using a drop trap with a human microfilaremic donor, C. milnei emerged as the potential host for transmitting Mansonella perstans. Substantial heterogeneity was observed between the trap visiting cycles of the various species (P < 0.001). The biting cycle of the main vector, C. milnei, showed two peaks (10–11 pm and 4–5 am), the highest being 10–11 pm. Conclusions: The Culicoides fauna of the South-West region of Cameroon has not changed significantly since the 1950s. Culicoides milnei was demonstrated to be the major vector of M. perstans in this part of Cameroon. It is essentially a nocturnal species which peaks in abundance between 10 and 11 pm.
