Image feature detection serves as the cornerstone for numerous vision applications, and it has found extensive use in agricultural harvesting. Nevertheless, determining the optimal feature extraction technique for a specific situation proves challenging, as the Ground Truth correlation between images is exceedingly elusive in harsh agricultural harvesting environments. In this study, we assemble and make publicly available the inaugural agricultural harvesting dataset, encompassing four crops: rice, corn and soybean, wheat, and rape. We develop an innovative Ground Truth-independent feature detector assessment approach that amalgamates efficiency, repeatability, and feature distribution. We examine eight distinct feature detectors and conduct a thorough evaluation using the amassed dataset. The empirical findings indicate that the FAST detector and ASLFeat yield the most exceptional performance in agricultural harvesting contexts. • The operation of unloading grain from harvesters is laborious and inefficient. • Small grain trucks are widespread but lack research in automation. • Developed an automated grain unloading system with a complete operational process. • The system is streamlined and reliable, enhancing intelligence and efficiency. Traditional track-driven rice combine harvesters, during the grain unloading process, often depend on the operator's frequent adjustment of the grain unloader's position while closely monitoring the accumulation of grain within the truck. Because of the structural characteristics of the harvester and the narrowness of the rural terrain, visibility is often obstructed, thereby increasing the difficulty of operation. It is estimated that the time consumed for unloading grain constitutes almost half of the total harvesting time, significantly reducing the efficiency of the harvester. To address these challenges, this study initially proposes an automated grain unloading system for track-driven rice combine harvesters based on stereo vision and details its operational process. Subsequently, we designed a method for acquiring the expected unloading points based on instance segmentation and another method for acquiring the actual unloading points based on geometric location information. Furthermore, this study introduces a method for determining the depth of the grain unloading truck's frame based on the angle of elevation of the unloading tube and proposes a method for acquiring unloading times at each expected point based on a model mapping grain pile height to unloading time. The experimental results confirm the high stability and reliability of the proposed automated unloading system. The maximum relative error between the computed expected unloading points and the actual expected points is less than 4 %. The maximum relative error between the actual unloading points and the true values is also less than 4 %. The mean absolute error between the obtained depth of the grain unloading truck's frame and the actual value is 0.014 m, with a root mean square error of 0.017 m. At each expected unloading point, the mean absolute error between the actual and expected grain pile heights is 0.012 m, with a root mean square error of 0.014 m. Overall, this study effectively enhances the efficiency of agricultural harvesting, reduces labor requirements, and provides a strong impetus for the automation and intelligence of combined harvesting machinery. We explore the semantic-rich structured information derived from the knowledge graphs (KGs) associated with the user-item interactions and aim to reason out the motivations behind each successful purchase behavior. Existing works on KGs-based explainable recommendations focus purely on path reasoning based on current user-item interactions, which generally result in the incapability of conjecturing users' subsequence preferences. Considering this, we attempt to model the KGs-based explainable recommendation in sequential settings. Specifically, we propose a novel architecture called Reinforced Sequential Learning with Gated Recurrent Unit (RSL-GRU), which is composed of a Reinforced Path Reasoning Network (RPRN) component and a GRU component. RSL-GRU takes users' sequential behaviors and their associated KGs in chronological order as input and outputs potential top-N items for each user with appropriate reasoning paths from a global perspective. Our RPRN features a remarkable path reasoning capacity, which is regulated by a user-conditioned derivatively action pruning strategy, a soft reward strategy based on an improved multi-hop scoring function, and a policy-guided sequential path reasoning algorithm. An insight into electronic structure and optical feature of S-adsorbed 3C–SiC (111) surface is carried out employing first-principles calculation. It is found that the T o and B position systems with adsorption energies of 3.880 and 3.895, respectively, are relatively stable compared to the T w and C systems. Impurity energy levels are present near Fermi level in C and T w position adsorption systems and the band-gap decreases obviously in the two systems. A raindrop-like electron cloud of S atom can be observed in T w adsorption system and the order of the chemical bond strength in the adsorption system is (B , T o ) > T w > C. The B and T o adsorption systems have good light permeability in the visible and infrared regions, while the C and T w adsorption systems are relatively suitable as dielectric materials and have high service life when they as devices in the ultraviolet region. Abstract: This study was designed to investigate the phthalates exposure levels in general population in Chongqing City of China, and to determine the possible associations between phthalate exposure and male reproductive function parameters. We recruited 232 general men through Chongqing Family Planning Research Institute and Reproductive Center of Chongqing. In a single spot urine sample from each man, phthalate metabolites, including mono-butyl phthalate (MBP), mono-ethyl phthalate (MEP), mono-(2-ethylhexyl) phthalate (MEHP), mono-benzyl phthalate (MBzP), phthalic acid (PA), and total PA were analyzed using solid phase extraction and coupled with high-performance liquid chromatography and detection by tandem mass spectrometry. Semen parameters were dichotomized based on World Health Organization reference values. Sperm DNA damage were analyzed using the alkaline single-cell gel electrophoresis assay. Reproductive hormones were determined in serum by the radioimmunoassay kit. We observed a weak association between urinary MBP concentration and sperm concentration in Chongqing general population. MBP levels above the median were 1.97 times (95% confidence interval [CI] 0.97–4.04) more likely to have sperm concentration below the reference value. There were no other associations between phthalate metabolites and reproductive function parameters after adjusted for potential risk factors. Our study suggested that general population in Chongqing area of China exposure to the environmental level of phthalate have weak or without adverse effects on the reproduction. How to improve the diffusion coefficient of Lithium ions in active materials is very necessary, which is responsible for the electrochemical performance. In this present contribution, LiFe 10/12 Co 1/12 Mn 1/12 P 11/12 S 1/12 O 4 (LF(CM)P(S)O) are designed and the stability, lithium ion diffusion rate, lithium intercalation potential, discharge curve, and electronic performances are systematically explored. The LF(CM)P(S)O shows the better stability with the formation energy of 2.01 eV. The lithium ion diffusion barrier of the system is reduced from 1.02 to 0.57 eV after doping, predicting that the lithium ion diffusion rate in LF(CM)P(S)O is 10 orders of magnitude faster than that of intrinsic system. The characteristic transition from p-type to n-type semiconductor is present due to the doping of S, and the doping of Mn and Co leads to the generation of impurity bands and the reduction of band gap from 3.78 to 0.737 eV. The effective mass of electrons in the doped system is 1.59m 0 , much greater than 1m 0 , which is conducive to the improvement of its conductivity. The charge density difference and electron localization function (ELF) demonstrates that the disappearance of electron enrichment and the decrease of the localization are responsible for the rapid migration of the lithium ion. The high operating voltage, charge-discharge platform of 4.7 V and the voltage difference contribute to the promotion of electrochemical performance, which open a window for the commercialization application of Li-ion battery. The calculated results show that the stability of B doping system is higher than that of the Al doping system and site 1 and 2 positions have equal contribution on the stability of systems. The doping of B and Al promotes the generation of impurity bands, and the disappearance of band gap, which is beneficial to the conductivity improvement of T-carbon. In order to enhance crop harvesting efficiency, an automatic-driving tracked grain vehicle system was designed. Based on the harvester chassis, we designed the mechanical structure of a tracked grain vehicle with a loading capacity of 4.5 m3 and a grain unloading hydraulic system. Using the BODAS hydraulic controller, we implemented the design of an electronic control system that combines the manual and automatic operation of the chassis walking mechanism and grain unloading mechanism. We utilized a hybrid A* algorithm to plan the traveling path of the tracked grain vehicle, and the path-tracking controller of the tracked grain vehicle was designed by combining fuzzy control and pure pursuit algorithms. Leveraging binocular vision technology and semantic segmentation technology, we designed an automatic grain unloading control system with functions of grain tank recognition and grain unloading regulation control. Finally, we conducted experiments on automatic grain unloading control and automatic navigation control in the field. The results showed that both the precision of the path-tracking control and the automatic unloading system meet the requirements for practical unoccupied operations of the tracked grain vehicle. The research on electronic properties of graphene/β-Si 3 N 4 (0001) heterostructure are carried out by regulating the interlayer spacing employing DTF calculations. The calculated results show that all interfaces are bonded by van der Waals (vdW) force, the configuration of N t1 atom relative to the center of C ring is more stable with the equilibrium interlayer distance D 0 of 2.908 Å and binding energy per C atom of −0.531 eV. The energy band structure is similar to the superposition of two parts of heterojunction and retains the shape of the Dirac cone with a direct band gap of 67 meV. Combined with differential density map and population analysis, it is found that 0.03 e spontaneously flow from the graphene layer to the β-Si 3 N 4 (0001) layer, results in the presence of a built-in electric field, which is conducive to the separation of electrons and holes. It is noteworthy that the band gap decreases obviously when the layer spacing gradually increases from 2.0 Å to 3.2 Å. Bisphenol A (2,2-bis(4-hydroxyphenyl)propane, BPA) is ubiquitous in the environment, wildlife, and humans. Evidence from past studies suggests that BPA is associated with decreased semen quality. However, the molecular basis for the adverse effect of BPA on male reproductive toxicity remains unclear. We evaluated the effect of BPA on mouse spermatocytes GC-2 cells and adult mice, and we explored the potential mechanism of its action. The results showed that BPA inhibited cell proliferation and increased the apoptosis rate. The testes from BPA-treated mice showed fewer spermatogenic cells and sperm in the seminiferous tubules. In addition, BPA caused reactive oxygen species (ROS) accumulation. Previous study has verified that mitochondrion was the organelle affected by the BPA-triggered ROS accumulation. We found that BPA induced damage to the endoplasmic reticulum (ER) in addition to mitochondria, and most ER stress-related proteins were activated in cellular and animal models. Knocking down of the PERK/EIF2α/chop pathway, one of the ER stress pathways, partially recovered the BPA-induced cell apoptosis. In addition, an ROS scavenger attenuated the expression of the PERK/EIF2α/chop pathway-related proteins. Taken together, these data suggested that the ROS regulated PERK/EIF2α/chop pathway played a vital role in BPA-induced male reproductive toxicity. [ABSTRACT FROM AUTHOR]

Ego-motion estimation is a foundational capability for autonomous combine harvesters, supporting high-level functions such as navigation and harvesting. This paper presents a novel approach for estimating the motion of a combine harvester from a sequence of stereo images. The proposed method starts with tracking a set of 3D landmarks which are triangulated from stereo-matched features. Six Degree of Freedom (DoF) ego motion is obtained by minimizing the reprojection error of those landmarks on the current frame. Then, local bundle adjustment is performed to refine structure (i.e., landmark positions) and motion (i.e., keyframe poses) jointly in a sliding window. Both processes are encapsulated into a two-threaded architecture to achieve real-time performance. Our method utilizes a stereo camera, which enables estimation at true scale and easy startup of the system. Quantitative tests were performed on real agricultural scene data, comprising several different working paths, in terms of estimating accuracy and real-time performance. The experimental results demonstrated that our proposed perception system achieved favorable accuracy, outputting the pose at 10 Hz, which is sufficient for online ego-motion estimation for combine harvesters. Abstract: OECD 421 screening test protocol was used to evaluate the reproductive toxicity of organic extracts of river water. These extracts, at doses of 0, 2, 16, or 80 equivalent L/kgbody weight(bw), were administered to rats daily by oral gavage. Vacuole degeneration of seminiferous epithelium was observed in the dosed groups. The caudal sperm counts/epididymis weight and the percentages of sperm with abnormal heads were increased in the group dosed with 80L/kgbw/day compared to the control group. Exposure of rats to organic extracts at the high dose resulted in a decrease in testosterone levels. The serum level of follicle-stimulating hormone increased in the groups dosed with 16 or 80L/kgbw/day compared to the control group. The number of apoptotic spermatid increased in the dosed groups. The results suggest that the extracts cause pathological damage to testicular tissue and disruption of spermatogenesis. [Copyright &y& Elsevier]

To explore the clinicopathological features of a rare dedifferentiated liposarcoma (DDLPS) with meningothelial‐like whorls, we retrospectively analyzed 46 reported cases and 1 case that we encountered. Fluorescence in situ hybridization (FISH) analysis of the MDM2 amplification status of our case was also performed. Our case involved a 73‐year‐old male patient who had a mass in the upper part of his left arm for 10 years and was treated by surgical ablation of the tumor because of the mass' recent rapid enlargement. Microscopically, the tumor tissues showed coexistence of well‐differentiated and dedifferentiated components, the latter of which included meningothelial‐like whorls and inflammatory myofibroblastic tumor‐like structures. The dedifferentiated components diffusely expressed vimentin, MDM2, CDK4, p16, and smooth muscle actin. They were also focally positive for desmin but negative for S‐100, CD117, CD34, ALK, EMA, SOX‐10, p53, and β‐catenin. FISH detection showed MDM2 amplification. In conclusion, subcutaneous DDLPS with meningothelial‐like whorls and inflammatory myofibroblastic tumor‐like features is rare. This case broadens the histopathological lineage of DDLPS, and confirms DDLPS with myogenic differentiation. The use of the combination of MDM2, CDK4, p16, and FISH to detect MDM2 amplification is a reliable basis for the diagnosis of DDLPS with meningothelial‐like whorls. [ABSTRACT FROM AUTHOR]

Objectives. To study the associations between kisspeptin levels in seminal plasma and blood plasma and semen quality. Materials and Methods. We conducted a male reproductive health survey in June 2014. A total of 666 volunteers were recruited from colleges in Chongqing, China. All volunteers completed a questionnaire including information on domestic characteristics and some potential confounders. We tested the kisspeptin levels in both blood and seminal plasma. Total seminal kisspeptin was calculated as the concentration of kisspeptin in seminal plasma multiplied by semen volume. Semen samples were tested according to the 2010 World Health Organization's (WHO) guidelines. Spearman correlation and multivariate linear regression were used to explore the association between kisspeptin concentrations in seminal plasma and blood plasma and semen quality. Potential confounders that were adjusted for included age, abstinence time, body mass index (BMI), grade, and smoking. Results. The median of kisspeptin levels in seminal plasma was 60,000 times higher than kisspeptin in blood plasma (28.0 × 106 pg/ml versus 448.9 pg/ml). Each interquartile range (IQR) of kisspeptin in seminal plasma was associated with a 4.6% (95% confidence interval [CI]: 1.6%–7.6%) increase in sperm concentration. Each IQR of total kisspeptin was associated with a 7.7% (95% CI: 4.4%–11.0%) increase in total sperm number and a 7.8% (95% CI: 4.0%–11.7%) increase in total motile sperm count. Kisspeptin levels were further classified into quartiles and Q1 was set as the reference level. Subjects in the high total kisspeptin group had 57.5% (95% CI: 33.2%–86.2%) higher total sperm number than the reference group. Conclusion. The positive association between kisspeptin levels in seminal plasma and semen quality supported an important role for the KISS1/GPR54 system in male reproductive health. Summary: This study explores whether sleep duration is associated with sperm chromatin integrity. To do so, we conducted a three‐phase panel study of 796 male volunteers from colleges in Chongqing (China) from 2013 to 2015. Sleep duration was measured using a modified Munich Chronotype Questionnaire. Sperm DNA integrity was examined via Sperm Chromatin Structure Assay and Comet assay. Setting 7–7.5 h day−1 of sleep duration as a reference, either longer or shorter sleep duration was associated negatively with high DNA stainability (HDS) (P = 0.009), which reflected the immaturity of sperm chromatin. The volunteers with > 9.0 h day−1 sleep and those with ≤ 6.5 h day−1 sleep had 40.7 and 30.3% lower HDS than did volunteers with 7–7.5 h day−1 sleep. No association was found between sleep duration and DNA fragmentation index or Comet assay parameters. This study suggests that sleep duration is associated with sperm chromatin integrity. It has been well demonstrated that polycyclic aromatic hydrocarbons (PAHs) can cause reproductive toxicity, and shorter telomere length in sperm may be one of the factors causing male infertility. However, whether exposure to PAHs is associated with sperm telomere length (STL) has never been evaluated. The present study aimed to assess the potential association between PAHs exposure and STL, and to explore potential biomarkers that may predict the effects of low-level exposure to PAHs on human sperm. Questionnaires and biological samples were collected from 666 volunteers participating in the Male Reproductive Health in Chongqing College Students (MARHCS) cohort study in 2014. Semen parameters were measured for 656 participants, while urinary PAH metabolites, STL and sperm apoptosis were successfully measured for 492, 444 and 628 participants, respectively. The linear regression analysis revealed that increased levels of urinary 1-hydroxypyrene (1-OHPyr) and 1-hydroxynapthalene (1-OHNap) were associated with decreased STL (− 0.385; 95% CI, − 0.749, − 0.021 for 1-OHPyr; and − 0.079; 95% CI, − 0.146, − 0.011 for 1-OHNap). The significant negative associations remained after adjusting for potential confounders. However, no significant associations were observed between urinary PAH metabolites and semen quality or sperm apoptosis. We also administrated rats with benzo[ a ]pyrene (B[ a ]P; 0, 1, 5, and 10 mg/kg) for 4 weeks and found shorter STL and decreased telomerase expression in germ cells in a dose-dependent manner. In conclusion, environmental exposure to some PAHs may be associated with decreased human STL, and the in vivo animal results also demonstrate the adverse effects of B[ a ]P on telomere of male germ cells. Several methods are currently available for selection when conducting sperm cryopreservation, however, these methods might cause different degrees of damage on sperm DNA. The aim of the this study is to compare the effects of storage at −80 °C (in ultra-low temperature refrigerator) and at −196 °C (in liquid nitrogen) on sperm DNA damage, thus to provide a reference for choosing the right method according to different aims. We randomly collected 28 semen samples from college students of Chongqing city. The samples stored at −80 °C were neat semen samples and the samples stored at −196 °C were mixed with additional cryoprotectants. Each sample was subjected to two freezing-thawing cycles, and the sperm DNA damage levels of fresh and thawed samples were measured by single cell gel electrophoresis (SCGE) and sperm chromatin structure assay (SCSA). Both SCGE and SCSA assays showed cryopreservation induced significant damage to sperm DNA. However, storage at −196 °C lead to more severe damage to sperm DNA than storage at −80 °C measured by SCSA. Sperm DNA damage increased simultaneously with the higher frequency of freezing-thawing cycles. We concluded that storage of neat semen samples at −80 °C had milder damage to sperm DNA than storage at −196 °C mixed with cryoprotectants. Epidemiological and experimental evidence for detrimental effects of cell phone use on semen quality is still equivocal. And that recruiting participants from infertility clinic not from general population may raise the possibility of a selection bias. To investigate effects of cell phone use on semen parameters in a general population,We screened and documented the cell phone use information of 794 young men from the Male Reproductive Health in Chongqing College students (MARHCS) cohort study in 2013, followed by 666 and 568 in 2014 and 2015, respectively. In the univariate regression analyses, we found that the daily duration of talking on the cell phone was significantly associated with decreased semen parameters, including sperm concentration [β coefficient = − 6.32% per unit daily duration of talking on the cell phone (h); 95% confidence interval (CI), − 11.94, − 0.34] and total sperm count (− 8.23; 95% CI, − 14.38, − 1.63) in 2013; semen volume (− 8.37; 95% CI, − 15.93, − 0.13) and total sperm count (− 16.59; 95% CI, − 29.91, − 0.73) in 2015]. Internet use via cellular networks was also associated with decreased sperm concentration and total sperm counts in 2013 and decreased semen volume in 2015. Multivariate analyses were used to adjust for the effects of potential confounders, and significant negative associations between internet use and semen parameters remained. Consistent but nonsignificant negative associations between talking on the cell phone and semen parameters persisted throughout the three study years, and the negative association was statistically significant in a mixed model that considered all three years of data on talking on the cell phone and semen quality. Our results showed that certain aspects of cell phone use may negatively affect sperm quality in men by decreasing the semen volume, sperm concentration, or sperm count, thus impairing male fertility. Sertoli cells (SCs) in the testes provide physical and nutritional support to germ cells. The vimentin cytoskeleton in SCs is disrupted by dibutyl phthalate (DBP), which leads to SCs dysfunction. In a previous study, we found that peroxisome proliferator-activated receptor alpha (PPARα) influenced the distribution of vimentin by affecting its phosphorylation in DBP-exposed SCs. In the present study, we investigated the role of Smad2/3 in regulating the expression of vimentin in DBP-exposed SCs. We hypothesized that Smad2/3 affects the distribution of vimentin by regulating its expression and that there is cross talk between Smad2/3 and PPARα. The real-time PCR and ChIP-qPCR results showed that SB431542 (an inhibitor of Smad2/3) could significantly attenuate the expression of vimentin induced by DBP in SCs. Phosphorylated and soluble vimentin were both downregulated by SB431542 pretreatment. WY14643 (an agonist of PPARα) pretreatment stimulated, while GW6471 (an antagonist of PPARα) inhibited, the activity of Smad2/3; SB431542 pretreatment also inhibited the activity of PPARα, but it did not rescue the DBP-induced collapse in vimentin. Our results suggest that, in addition to promoting the phosphorylation of vimentin, DBP also stimulates the expression of vimentin by activating Smad2/3 in SCs and thereby induces irregular vimentin distribution. The aim of the study was to examine the association between polymorphisms of DNA repair genes and chromosomal damage of 1,3-butadiene- (BD-) exposed workers. The study was conducted in 45 pairs of occupationally exposed workers in a BD product workshop and matched control workers in an administrative office and a circulatory water workshop in China. Newly developed biomarkers (micronuclei, MNi; nucleoplasmic bridges, NPBs; nuclear buds, NBUDs) in the cytokinesis-blocked micronucleus (CBMN) cytome assay were adopted to detect chromosomal damage. PCR and PCR-restriction fragment length polymorphism (RFLP) are adopted to analyze polymorphisms of DNA repair genes, such as X-ray repair cross-complementing Group 1 (XRCC1), O6-methylguanine-DNA methyltransferase (MGMT), poly (adenosine diphosphate-ribose) polymerases (ADPRT), and apurinic/apyrimidinic endonucleases (APE1). The BD-exposed workers exhibited increased frequencies of MNi and NPBs when compared to subjects in the control group. The results also show that the BD-exposed workers carrying XRCC1 diplotypes TCGA-CCGG (4.25±2.06‰) (FR=2.10, 95% CI: 1.03–4.28) and TCGG-TCGA (5.80±3.56‰) (FR=2.75, 95% CI: 0.76–2.65) had statistically higher NBUD frequencies than those who carried diplotype TCGG-TCGG (1.89±1.27‰). Background: Elevated levels of reactive oxygen species (ROS) are detected in 25% to 80% of infertile men. They are involved in the pathology of male infertility. Understanding the effect of increasing levels of ROS on the differential expression of sperm proteins is important to understand the cellular processes and or/pathways that may be implicated in male infertility. The aim of this study was to examine differentially expressed proteins (DEPs) in spermatozoa from patients with low, medium and high ROS levels. Methods: A total of 42 infertile men presenting for infertility and 17 proven fertile men were enrolled in the study. ROS levels were measured by chemiluminescence assay. Infertile men were divided into Low (0- < 93 RLU/s/106 sperm) (n = 11), Medium (>93-500 RLU/s/106 sperm) (n = 17) and High ROS (>500 RLU/s/106sperm) group (n = 14). All fertile men had ROS levels between 4-50 RLU/s/106 sperm. 4 subjects from fertile group and 4 each from the Low, Medium and High ROS were pooled. Protein extraction, protein estimation, gel separation of the proteins, in-gel digestion, LTQ-orbitrap elite hybrid mass spectrometry system was conducted. The DEPs, the cellular localization and pathways of DEPs involved were examined utilizing bioinformatics tools. Results: 1035 proteins were identified in the 3 groups by global proteomic analysis. Of these, 305 were DEPs. 51 were unique to the Low ROS group, 47 Medium ROS group and 104 were unique to the High ROS group. 6 DEPs were identified by Uniprot and DAVID that had distinct reproductive functions and they were expressed only in 3 ROS groups but not in the control. Conclusions: We have for the first time demonstrated the presence of 6 DEPs with distinct reproductive functions only in men with low, medium or high ROS levels. Objective To compare the sperm protein profile between infertile men with unilateral varicocele and infertile men with bilateral varicocele. Methods This prospective study investigated 50 infertile patients with clinical varicocele (33 unilateral and 17 bilateral) seeking fertility workup between March 2012 and April 2014. Routine sperm parameters, reactive oxygen species, total antioxidant capacity, and sperm deoxyribonucleic acid fragmentation were assessed in their semen. Sperm protein profile was characterized only in pooled samples of 5 unilateral and 3 bilateral varicocele samples, respectively, using sodium dodecyl sulfate-polyacrylamide gel electrophoresis and an Linear Trap Quadrupole-Orbitrap Elite hybrid mass spectrophotometer system. Differences in protein expression were analyzed using gel analysis software, followed by protein identification using mass spectroscopy analysis. Differentially expressed proteins and their abundance were quantified by comparing spectral counts, followed by bioinformatics analysis. Results Unique expression of 64 proteins in the bilateral group and 31 proteins in the unilateral group was obtained. Core functions of the top protein interaction networks were post-translational modification (∼122 proteins associated with acetylation), protein folding, free-radical scavenging, cell death, and survival. The top molecular and cellular functions were protein degradation, free radical scavenging, and post-translational modifications, whereas the top pathways were protein ubiquitination and mitochondrial dysfunction. Major biological pathways for the 253 differentially expressed proteins were metabolism, apoptosis, and signal transduction. Conclusion Functional proteomic profiling helps identify the differential processes or pathways that are affected based on the nature of varicocele (bilateral or unilateral) and provide insights into the mechanistic implications of varicocele-associated male infertility. Dibutyl phthalate (DBP) is a peroxisome proliferator which can lead to germ cell loss from Sertoli cells. Collapse of vimentin filaments occurs in Sertoli cells after DBP exposure. Peroxisome proliferator activated receptor α (PPARα) is a key receptor which could be activated by DBP. The role of PPARα in this process was investigated. Results showed that, PPARα was activated in DBP-exposed Sertoli cells, GW6471 inhibited the activity of PPARα, phosphorylation level of vimentin and concentration of soluble vimentin was higher in DBP-treated Sertoli cells than GW6471+DBP-treated cells. These results suggest that PPARα directly or indirectly mediated phosphorylation of vimentin on Ser 83, and PPARα may play an important role in regulating the reorganization of vimentin filaments during exposure of Sertoli cells to DBP. Context: Smoke inhalation injury is the main cause of fatalities for fire victims. Understanding in the pathophysiology of the injury has not been fully explored in recent years. To further explore the pathophysiological mechanism, a dynamic and controllable animal model is necessary. Objective: To develop a rat model of smoke inhalation injury to simulate human victims in air-restricted vehicle cabin fires. Materials and methods: Smoke concentration, including CO, O2, VOCs and smoke temperature under different combustion conditions, were detected. Levels of COHb, respiratory function, lung wet-to-dry weight ratio and protein concentration in BALF and blood were measured. Pathological evaluations of lung in tissues were conducted at 1, 6, 24 and 48 h post-exposure. Results: Smoke concentration rose with the increase of combustion temperature and decrease of oxygen flow. Further, 215 kinds of VOCs in the smoke were detected, and the concentrations of benzene, methylbenzene, ethylbenzene, dimethylbenzene, phenylethylene and trimethyl-benzene was 32.93, 402.06, 764.03, 113.73, 1006.61 and 89.28 mg/m³, respectively. Significant hypoxemia and CO poisoning occurred in rats. The FCOHb after exposure for 14min immediately rose to (44.2 ± 12.3) % and then gradually decrease to a normal level at 300 min post-exposure. At 24 h post-exposure, Penh increased significantly (p<0.05), and high pulmonary vascular permeability and significant lung edema [p<0.05) were observed in the smoke inhalation group. Discussion and conclusion: In summary, the novel rat model of smoke inhalation injury system used in the study is dynamic and controllable, and appropriate for use in smoke inhalation injury studies of air-restricted cabins in vehicles. DNA methylation is involved in tissue-specific and developmentally regulated gene expression. Here, we screened a novel methylation gene Sox30, whose methylation might contribute to its regulation and testis development in mice. Sox30 is a member of Sox transcription factors, and is considered to be involved in spermatogonial differentiation and spermatogenesis. However, the precise function and regulatory expression pattern remain unclear. In the present study, we found that Sox30 is highly expressed in adult testes but not in ovaries. Sox30 expression begins in early development, and in the testes, it is specifically increased coincidentally with development until adulthood. Moreover, Sox30 is expressed not only in testis germ cells, but also in sertoli cells. Sox30 is hypo-methylated in testis, epididymis and lung of adult mice, in which Sox30 is expressed. By contrast, Sox30 is hypermethylated in ovary, heart, brain, liver, kidney, spleen, pancreas, muscle, intestine, pituitary gland, blood and hippocampus of adult mice, in which the Sox30 is absent. Importantly, decreased methylation at CpG islands of Sox30 is observed in mouse developmental testes after birth, which is associated with enhanced Sox30 expression. However, the hypermethylated status of Sox30 is maintained in ovaries that does not express Sox30 during this period. Further, following demethylation treatment using 5-aza-dC, Sox30 expression is restored in GC2, TM3 and TM4 cell lines. This observation convincingly confirms that methylation really contributes to Sox30 silencing. Abstract: The health risks of heavy metals to local residents via consumption of food crops cultivated in the reclaimed soils of the Pearl River Estuary (PRE) were investigated. Soils in the river estuary were found to be significantly contaminated with heavy metals, with mean concentrations of Cd, 0.858; Pb, 48.7; Cr, 112.4; Cu, 57.3; Zn, 210.8; and Ni, 40.0mgkg−1. Rice and root vegetables were also polluted severely. The percentages of rice samples that exceeded the maximum allowable levels were 94.3, 91.4, 88.6, and 17.1% for Pb, Cr, Cd, and Cu, respectively. The accumulation factor (AF) of Cd was the highest compared with the other five metals. The total values of the health risk index (HRI) of Cd and Cu from all the crops were 3.683 and 1.665, respectively. They exceeded considerably the allowable level 1.0. The HRI values of all the metals were mostly from rice consumption. These results suggest that more attention should be directed to the potential health risks of heavy metals to residents living in the tidal flat reclaimed areas. [Copyright &y& Elsevier]

1, 3-Butadiene (BD) is a high-efficiency carcinogen in rodents and was classified as a human carcinogen in 2008 by the International Agency for Research on Cancer. However, its ability to induce genetic damage and the influence of metabolic polymorphisms to such damage in humans are both controversial claims. This study was conducted to investigate the relationships between exposure to BD, the polymorphisms of metabolic genes and the chromosomal damage in 45 pairs of occupationally exposed workers in a BD product workshop and matched control workers in an administrative office and circulatory water workshop in China. Exposure to BD was evaluated by personal sampling and stationary sampling. Different chromosomal damage endpoints in peripheral blood lymphocytes were determined using the cytokinesis-blocked micronucleus (CBMN) cytome assay; polymorphisms of metabolic genes [cytochrome P450 2E1 (CYP2E1), glutathione S-transferases (GST) and microsomal epoxide hydrolase (mEH)] in BD-exposed group were detected by polymerase chain reaction (PCR) or PCR–restriction fragment length polymorphism analysis. The results show that the average BD measurements of the exposed group were significantly higher than those for the control group (a personal sampling and stationary sampling, respectively). The BD-exposed workers exhibited increased frequencies of micronuclei (MNi) (8.00 ± 3.78‰ versus 5.62 ± 2.41‰) and nucleoplasmic bridges (NPBs) (2.58 ± 2.79‰ versus 1.13 ± 1.34‰) and a decreased nuclear division index (2.20 ± 0.14 versus 2.35 ± 0.27) when compared subjects in the control group. Meanwhile, BD-exposed workers carrying CYP2E1 c1c2/c2c2 or mEH intermediate (I)/high (H) group had a significantly higher NPB frequency than those carrying CYP2E1 c1c1 [frequency ratio (FR) = 2.60, 95% confidence interval (CI) 1.72–3.93; P < 0.0001) or the mEH low(S) group (FR = 2.06, 95% CI% 1.17–3.62; P < 0.05), respectively. Our study suggests that MNi and NPB frequency in CBMN cytome assay could be potential genotoxic biomarkers for BD exposure in humans. Abstract: Many of the coastal tidal flats in China that were polluted with heavy metals are now being reclaimed for arable land. The safety of these soils for agriculture is of great concern. The present study investigated the sediment chemical properties, concentrations, and speciation of heavy metals at different levels of desalination during a controlled leaching experiment. After leaching with fresh water, the average reductions in the heavy metal species examined in 0–65cm depth sediment were 32.1% for Pb, 26.2% for Cd, 14.0% for Zn, 13.8% for Cu, and 11.0% for Cr, while the Ni concentration in sediment did not change significantly. The amounts of Cd, Pb, Cr, Cu, and Zn bound to the reducible fraction, the amounts of Cd, Pb, and Zn bound to the exchangeable fraction, the amounts of Pb, Cr, Cu, and Zn associated with the carbonate fraction, and the Cu associated with the oxidizable fraction all decreased significantly. Complexation with salt anions, ion exchange between the cations and the metal ions, removal of , dissolution of carbonate, and the redox potential variations all contributed to the decreases in Pb, Cd, Zn, Cu, and Cr. These results suggest that leaching with fresh water can also remove a fraction of the heavy metal contamination when it diminishes sediment salinity. [ABSTRACT FROM AUTHOR]

Abstract: Nestin, a member of the sixth class of intermediate filament proteins, has long been known not only as a specific marker for central nervous system stem cells, but also as an indicator of the degree of neural stem cell differentiation. Immunohistochemical analysis showed nestin expression in the epidermis and the upper two-third of the hair follicle in non-balding human scalp skin, but not in the lower third of the follicle. Expression was very weak in the sebaceous gland. The level of nestin mRNA transcription was much higher in the bulge region of the follicle than in the sebaceous gland. These results show that the pattern of nestin expression is related to the differentiation of epidermal stem cells. [Copyright &y& Elsevier]

Male reproductive damage in the general population comprises different disorders in various biomarkers, which could be respectively caused by a number of exposure factors. However, researchers considering the environmental/behavioral/psychological exposures together to evaluate their contribution to male reproductive damage are still lacking. The present study investigated the comprehensive association between 138 environmental/behavioral/psychological exposures and 32 male reproductive biomarkers in 796 young Chinese men using graph-guided fused lasso (GFLASSO) and hierarchical clustering methods. All biomarkers were found to be associated with various exposures. A combination of these exposures not only predicted the levels of single biomarkers in another test dataset, but also identified the comprehensive reproductive features by clustering the men into five subgroups with distinct damages representing disrupted spermatogenesis with abnormal sperm morphology, low sperm motility with DNA fragmentation, chromatin immaturity, aberrant endocrine, or DNA strand breakage. The findings can be used to suggest a novel way to identify the males with a high risk of reproductive damage and develop personalized preventive strategies. In our previous study, we had identified SOX30 as a novel tumor suppressor that acts through direct regulation of p53 transcription in human lung cancer. Here, we sought to determine the clinical relevance of SOX30 expression in a series of surgically-resected non-small cell lung cancer (NSCLC) patients. Analysis of SOX30 expression and clinico-pathologic features reveal a significant correlation of SOX30 expression with histological type (n = 220, P = 0.008) and clinical stage (n = 220, P = 0.024). Kaplan-Meier analysis indicates an association of high SOX30 expression with better prognosis in NSCLC patients (n = 220, P = 0.007). Via multivariate Cox-regression analysis, SOX30 expression is revealed to be an independent prognostic factor for overall survival (OS) of NSCLC patients (n = 220, P = 0.014, hazard ratio (HR) = 0.816). In particular, SOX30 is a favorable and independent prognostic factor in one main subtype of NSCLC, lung adenocarcinoma (ADC) patients (n = 150, P = 0.000, HR = 0.405), but not in another main subtype of NSCLC, squamous cell carcinoma patients. Furthermore, high expression of SOX30 represents a favorable and independent factor for the prognosis of ADC patients at clinical stage II (P = 0.013), with positive lymph node (P = 0.003), at histological grade 2 (P = 0.000) or grade 3 (P = 0.025).
