ObjectivesNanofluid technology is an important means to improve the thermal conductivity of energy storage materials in solar thermal power plants. However, the nanoparticles in actual molten salt based nanofluid usually aggregate and settle down, which significantly affects the thermal performance of the composites. To gain a deeper insight into the variation of thermophysical properties of molten salt based nanofluids, the silica nanoparticles with different aggregation morphologies were constructed in solar salt.MethodsThe effects of system temperature, mass fraction of nanoparticles, and micromorphology of aggregates were investigated based on the molecular dynamics simulation and lattice Boltzmann method.ResultsIt is found that the inclusion of nanoparticles can effectively enhance the thermal conductivity of the base fluid. Compared with the uniformly dispersed nanofluid, the heat conduction performance of the aggregated nanofluid is better, and it decreases with the increase of fractal dimension of agglomerates. Moreover, the thermal conductivity of nanofluid is negatively correlated with temperature and aggregate size, and positively correlated with mass fraction of nanoparticles, backbone length of each aggregate and degree of agglomeration.ConclusionsThe research results reveal the mechanism of nanoparticle aggregation on the heat transport within phase change materials and provide critical reference on the design of thermophysical properties of molten salt based nanofluids.

Abstract As one of the most common tumors in women, the pathogenesis and tumor heterogeneity of breast cancer have long been the focal point of research, with the emergence of tumor metastasis and drug resistance posing persistent clinical challenges. The emergence of single-cell sequencing (SCS) technology has introduced novel approaches for gaining comprehensive insights into the biological behavior of malignant tumors. SCS is a high-throughput technology that has rapidly developed in the past decade, providing high-throughput molecular insights at the individual cell level. Furthermore, the advent of multitemporal point sampling and spatial omics also greatly enhances our understanding of cellular dynamics at both temporal and spatial levels. The paper provides a comprehensive overview of the historical development of SCS, and highlights the most recent advancements in utilizing SCS and spatial omics for breast cancer research. The findings from these studies will serve as valuable references for future advancements in basic research, clinical diagnosis, and treatment of breast cancer.

Abstract Intercellular cell adhesion molecule-1 (ICAM-1) is frequently overexpressed in non-small cell lung cancer (NSCLC) and associated with poor prognosis. However, the mechanism underlying the negative effects of neoplastic ICAM-1 remains obscure. Herein, we demonstrate that the survival of NSCLC cells but not normal human bronchial epithelial cells requires an anti-apoptosis signal triggered by fibrinogen γ chain (FGG)–ICAM-1 interaction. ICAM-1–FGG ligation preserves the tyrosine phosphorylation of ICAM-1 cytoplasmic domain and its association with SHP-2, and subsequently promotes Akt and ERK1/2 activation but suppresses JNK and p38 activation. Abolishing ICAM-1–FGG interaction induces NSCLC cell death by activating caspase-9/3 and significantly inhibits tumor development in a mouse xenograft model. Finally, we developed a monoclonal antibody against ICAM-1–FGG binding motif, which blocks ICAM-1‒FGG interaction and effectively suppresses NSCLC cell survival in vitro and tumor growth in vivo. Thus, suppressing ICAM-1–FGG axis provides a potential strategy for NSCLC targeted therapy.

Influenza virus infection poses a continual menace to public health. Here, we developed soluble trimeric HA ectodomain vaccines by establishing interprotomer disulfide bonds in the stem region, which effectively preserve the native antigenicity of stem epitopes. The stable trimeric H1 ectodomain proteins exhibited higher thermal stabilities in comparison with unmodified HAs and showed strong binding activities towards a panel of anti-stem cross-reactive antibodies that recognize either interprotomer or intraprotomer epitopes. Negative stain transmission electron microscopy (TEM) analysis revealed the stable trimer architecture of the interprotomer disulfide-stapled WA11#5, NC99#2, and FLD#1 proteins as well as the irregular aggregation of unmodified HA molecules. Immunizations of mice with those trimeric HA ectodomain vaccines formulated with incomplete Freund’s adjuvant elicited significantly more potent cross-neutralizing antibody responses and offered broader immuno-protection against lethal infections with heterologous influenza strains compared to unmodified HA proteins. Additionally, the findings of our study indicate that elevated levels of HA stem-specific antibody responses correlate with strengthened cross-protections. Our design strategy has proven effective in trimerizing HA ectodomains derived from both influenza A and B viruses, thereby providing a valuable reference for designing future influenza HA immunogens.

Rheumatoid arthritis is a chronic autoimmune disease of undetermined etiology characterized by symmetric synovitis with predominantly destructive and multiple joint inflammation. Cytoplasmic DNA sensors that recognize protein molecules that are not themselves or abnormal dsDNA fragments play an integral role in the generation and perpetuation of autoimmune diseases by activating different signaling pathways and triggering innate immune signaling pathways and host defenses. Among them, melanoma deficiency factor 2 (AIM2) recognizes damaged DNA and double-stranded DNA and binds to them to further assemble inflammasome, initiating the innate immune response and participating in the pathophysiological process of rheumatoid arthritis. In this article, we review the research progress on the source of cytoplasmic DNA, the mechanism of assembly and activation of AIM2 inflammasome, and the related roles of other cytoplasmic DNA sensors in rheumatoid arthritis.

A bio-based shape memory composite was prepared with chitosan as filler and deproteinized epoxidized natural rubber （ENR) as matrix, and the microstructure, mechanical properties, and shape memory performance of the shape memory composite materials were investigated by Fourier transform infrared spectroscopy, atomic force microscope, electronic universal testing machine, etc. The results showed that the addition of chitosan dispersed the ordered crystalline phases of ENR into smaller and more uniformly sized particles throughout the system. Due to the strengthening and toughening effect of chitosan, the abrasion resistance of the shape memory composite material was enhanced and its mechanical properties improved gradually. Furthermore, the addition of chitosan further improved the shape memory performance of ENR.

Abstract Clinically, chronic pain and depression often coexist in multiple diseases and reciprocally reinforce each other, which greatly escalates the difficulty of treatment. The neural circuit mechanism underlying the chronic pain/depression comorbidity remains unclear. The present study reports that two distinct subregions in the paraventricular thalamus (PVT) play different roles in this pathological process. In the first subregion PVT posterior (PVP), glutamatergic neurons (PVPGlu) send signals to GABAergic neurons (VLPAGGABA) in the ventrolateral periaqueductal gray (VLPAG), which mediates painful behavior in comorbidity. Meanwhile, in another subregion PVT anterior (PVA), glutamatergic neurons (PVAGlu) send signals to the nucleus accumbens D1‐positive neurons and D2‐positive neurons (NAcD1→D2), which is involved in depression‐like behavior in comorbidity. This study demonstrates that the distinct thalamo‐subcortical circuits PVPGlu→VLPAGGABA and PVAGlu→NAcD1→D2 mediated painful behavior and depression‐like behavior following spared nerve injury (SNI), respectively, which provides the circuit‐based potential targets for preventing and treating comorbidity.

ABSTRACT: Hyperuricemia (HUA) is a metabolic disorder caused by excessive production of uric acid (UA) or impaired uric acid metabolism. Smilax China L. has a wide range of pharmacological activities such as immunomodulatory, anti-inflammatory, and antioxidant. Its roots and rhizomes have been widely used for the treatment of HUA. However, its mechanisms for treating HUA and reducing renal impairment have not been fully elucidated. In the present study, we evaluated the effect of Smilax China L. extract (SC) on UA metabolism and further explored its mechanism of action by feeding a high-calcium and high-protein diet to chickens to induce a model of HUA in chickens. SC significantly reduced serum UA levels and improved renal function in hyperuricemic chickens. Meanwhile, SC was able to inhibit the activity of xanthine oxidase (XOD) in vivo and in vitro, reducing the production of uric acid. In addition, SC was able to increase the expression of Breast Cancer Resistance Protein (BCRP) in the kidney and ileum and increase uric acid excretion. Therefore, our results suggest that SC may be a candidate for anti-hyperuricemia.

Objective: To evaluate the T-lymphocyte subset distribution and the diagnostic and prognosis value of double-negative T (DNT) cells in colorectal cancer (CRC). Methods: This retrospective study compared the T-lymphocyte subsets and DNT of 114 patients with CRC with those of 107 healthy controls (HC). The diagnostic potential of DNT and T-lymphocyte subsets was assessed using the receiver operating characteristic (ROC) curve, and prognostic values were evaluated using the Kaplan–Meier curve and the Cox regression model. Results: The percentages of CD8+ T cells and DNT cells, and value of carcinoembryonic antigen (CEA), were remarkably higher in patients with CRC than in those with HC, but the ratio of CD4+/CD8+ was decreased. Using ROC curve analysis, DNT cell percentage, CEA, and CD4+/CD8+ ratio all had good diagnostic efficacy, with areas under the curve (AUCs) of 0.865, 0.786 and 0.624, respectively. The central neuropeptide GLP-1 is synthesized by preproglucagon (PPG) neurons in the brain. GLP-1 receptors are widely distributed in central nervous system. Hippocampus is a key component of the limbic system which is involved in learning, memory, and cognition. Previous studies have shown that overexpression of GLP-1 receptors in the hippocampus could improve the process of learning and memory. However, up to now, the direct electrophysiological effects and possible molecular mechanisms of GLP-1 in hippocampal CAl neurons remain unexplored. The present study aims to evaluate the effects and mechanisms of GLP-1 on the spontaneous firing activity of hippocampal CAl neurons. Employing multibarrel single-unit extracellular recordings, the present study showed that micro-pressure administration of GLP-1 receptor agonist, exendin-4, significantly increased the spontaneous firing rate of hippocampal CA1 neurons in rats. Furthermore, application of the specific GLP-1 receptor antagonist, exendin(9−39), alone significantly decreased the firing rate of CA1 neurons, suggesting that endogenous GLP-1 modulates the firing activity of CA1 neurons. Co-application of exendin(9−39) completely blocked exendin-4-induced excitation of hippocampal CA1 neurons. Finally, the present study demonstrated for the first time that the transient receptor potential canonical 4 (TRPC4)/TRPC5 channels may be involved in exendin-4-induced excitation. The present studies may provide a rationale for further investigation of the modulation of GLP-1 on learning and memory as well as its possible involvement in Alzheimer's disease.

Reducing the blade aerodynamic loads is extremely important to extend the life of wind turbine blades. Trailing edge flaps are effective means to reduce blade aerodynamic loads, however, its regulation ability weakens greatly at large angles of wind attack. The combined regulation ability on the aerodynamic characteristics of wind turbine blades by S-slot and trailing edge flaps and by adding S-slot at 50% of the chord length of turbine blade was investigated. The shear stress transport (SST) turbulence model was used to study the effect of combined control of S-slot and trailing edge flaps on the aerodynamic characteristics of S809 airfoil at Reynolds number of 106. The synergistic mechanism between the S-slot and trailing edge flaps was further analyzed. The results show that the deflection of trailing edge flaps can significantly change the gas flow rate in the slot, and effectively adjust the pressure difference in the middle of the blade. This combined control strategy can significantly improve the stall phenomenon of airfoil at large angles of wind attack, thereby significantly enhancing the ability of trailing edge flaps to control the lift coefficient.

Abstract Background The early allopolyploid Brassica napus was a hybrid of two Brassica species, that had undergone a whole genome duplication event followed by genome restructuring, including deletions and small scale duplications. A large number of homologous genes appeared functional divergence during species domestication. Due to the high conservation of de novo glycerolipid biosynthesis, multiple homologues of glycerol-3-phosphate acyltransferases (GPATs) have been found in B. napus. Moreover, the functional variances among these homologous GPAT-encoding genes are unclear. Results In this study, four B. napus homologous genes encoding glycerol-3-phosphate acyltransferase 9 (BnaGPAT9) were characterized. Although a bioinformatics analysis indicated high protein sequence similarity, the homologues demonstrated tissue-specific expression patterns and functional divergence. Yeast genetic complementation assays revealed that BnaGPAT9-A1/C1 homologues but not BnaGPAT9-A10/C9 homologues encoded functional GPAT enzymes. Furthermore, a single nucleotide polymorphism of BnaGPAT9-C1 that occurred during the domestication process was associated with enzyme activity and contributed to the fatty acid composition. The seed-specific expression of BnGPAT9-C1 1124A increased the erucic acid content in the transformant seeds. Conclusions This study revealed that BnaGPAT9 gene homologues evolved into functionally divergent forms with important roles in erucic acid biosynthesis.

Abstract Background Lung cancer remains a major global health concern due to its high incidence and mortality rates. With advancements in medical treatments, an increasing number of early-stage lung cancer cases are being detected, making surgical treatment the primary option for such cases. However, this presents challenges to the physical and mental recovery of patients. Peplau known as the “mother of psychiatric associations” has formulated a theory of interpersonal relationships in nursing. Through effective communication between nurses and patients over four periods, she has established a good therapeutic nurse-patient relationship. Therefore, this study aimed to explore the effect of perioperative multimodal nursing based on Peplau’s interpersonal relationship theory on the rehabilitation of patients with surgical lung cancer. Methods We retrospectively analyzed 106 patients with non-small cell lung cancer who underwent thoracoscopic lobectomy at our department between June 2021 and April 2022. Patients were categorized into two groups according to the different nursing intervention techniques. The Peplau’s group comprised 53 patients who received targeted nursing interventions, and the control group comprised 53 patients who received conventional nursing care. We observed the patients’ illness uncertainty, quality of life, and clinical symptoms in both groups. Results Patients in the Peplau’s group had significantly lower illness uncertainty scores and a significantly higher quality of recovery than those in the control group. However, there were no significant differences in length of post-anesthesia care unit stay, complication rates, and visual analog scores between both groups. Conclusion The multimodal perioperative nursing based on Peplau’s interpersonal relationship theory not only reduces the illness uncertainty of patients with lung cancer surgery and improves their QoR but also expands the application of this theory in clinical practice, guiding perioperative nursing of patients with lung cancer. Implications These findings provide practical information for standardized care in a hectic anesthetic care setting. Impact The assessed anesthesia nursing model helps reduce uncertainty and promote early recovery in patients with cancer at various stages of their disease, which expands the scope of therapeutic practice and existing theories. It also serves as a guide for care in the anesthesia recovery room. Reporting method We adhered to the relevant Equator guidelines and the checklist of items in the case–control study report. Patient or public contribution Patients cooperated with medical staff to complete relevant scales.

Based on the recent work [J. Stat. Phys. 190, 193 (2023), J. Stat. Phys. 190, 181 (2023)], we formulate the first law and the second law of stochastic thermodynamics in the framework of general relativity. These laws are established for a charged Brownian particle moving in a heat reservoir and subjecting to an external electromagnetic field in generic stationary spacetime background, and in order to maintain general covariance, they are presented respectively in terms of the divergences of the energy current and the entropy density current. The stability of the equilibrium state is also analyzed.

Background The persistently high incidence of stroke in many nations is suggestive of an area for further improvement on existing strategies of primary stroke prevention. Although the era of digitalisation has led to the increasing use of mobile applications (apps) in healthcare, more studies are needed to determine the efficacy of apps in producing the desired health outcomes across different nations and cultures. Objective To describe the development and evaluate the usability of a mobile app in delivering a culturally adapted stroke prevention educational programme for middle-aged adults in the Republic of China. Methods The educational programme was developed in three phases. In Phase 1, the process involved analysing requirements and designing structured modules. Phase 2 concentrated on expert consultation and technical development to deliver the educational programme. Phase 3 included a usability trial and refinement of the educational program based on trial results. Results Educational content was derived from the Chinese Guidelines for the Prevention and Treatment of Stroke and the Dietary Guidelines for Residents. The WeChat platform was used to deliver the educational programme. Participants expressed satisfaction with the content, interface, and functions of the apps, indicating that the apps have good usability. Conclusions The development process of the Educational Programme was designed to maximise the culturally appropriate, and impact of lifestyle changes and stroke prevention. An app-based educational programme that has demonstrated good usability is a vital factor prior to deploying it in an intervention to evaluate its effects on health outcomes.

Abstract The impact of changes in global temperatures and precipitation on climate distribution remains unclear. Taking the annual global average temperatures and precipitation as the origin, this study determined the climate distribution with the distances of temperature and precipitation from their global averages as the X and Y axes. The results showed that during 1980–2019, the global temperature distribution converged toward the mean (convergence), while the precipitation distribution moved away from the mean (divergence). The combined effects of both led to a convergence in the global climate distribution. During 2025–2100, significant climate convergence is observed under two emission scenarios (SSP245 and SSP585). However, the climate convergence and the area of change in climate type remains insignificant only under SSP126, suggesting that the diversity of the global climate pattern can be maintained under a sustainable emission pathway (SSP126), whereas high emission pathways will lead to greater uniformity in global climate.

Objective Although stroke is prevalent among older people, there is a rising incidence among the younger subpopulations, particularly middle-aged adults. A healthy diet is one of the key modifiable factors to primary prevention of stroke among these subpopulations, yet there is limited understanding of the dietary habits among middle agers who have the risk factor(s) but no occurrence of stroke. This study aims to explore the views on perceptions and the self-management of middle-aged adults at risk of stroke on a healthy diet and to identify the enablers and barriers that could inform the future development of dietary interventions.Design This study used an interpretive descriptive qualitative design, employing semistructured purposive sampling for focus group discussions. Thematic analysis was conducted on the transcribed interviews and field notes, facilitated by NVivo 12.0 Plus software.Setting Community settings in Zhengzhou City, Henan Province.Participants Middle-aged adults (aged 45–59) were identified as at risk of stroke due to the presence of one or more modifiable risk factors.Result A total of seven focus group discussions were audio recorded. Four main themes emerged, which were: (1) cognitive understanding of a healthy diet; (2) dietary practices; (3) knowledge acquisition and (4) barriers to dietary adherence.Conclusions The middle-aged adults at risk of stroke were generally aware of the risk and attempted to practise healthy eating. The existing educational programmes on following a healthy diet in the prevention of disease need to be made more comprehensible, accessible and equitable, especially for those from socioeconomically disadvantaged communities.

Abstract Liquid–liquid phase separation (LLPS) is a novel principle for interpreting precise spatiotemporal coordination in living cells through biomolecular condensate (BMC) formation via dynamic aggregation. LLPS changes individual molecules into membrane-free, droplet-like BMCs with specific functions, which coordinate various cellular activities. The formation and regulation of LLPS are closely associated with oncogenesis, tumor progressions and metastasis, the specific roles and mechanisms of LLPS in tumors still need to be further investigated at present. In this review, we comprehensively summarize the conditions of LLPS and identify mechanisms involved in abnormal LLPS in cancer processes, including tumor growth, metastasis, and angiogenesis from the perspective of cancer hallmarks. We have also reviewed the clinical applications of LLPS in oncologic areas. This systematic summary of dysregulated LLPS from the different dimensions of cancer hallmarks will build a bridge for determining its specific functions to further guide basic research, finding strategies to intervene in LLPS, and developing relevant therapeutic approaches.

Abstract Achievement of national climate targets and the corresponding costs would entirely depend on regional actions within the country. However, because of substantial inequalities and heterogeneities among regions, especially in developing economies, aggressive or uniform actions may exacerbate inequity and induce huge economic losses, which in turn challenges the national climate pledges. Hence, this study extends prior research by proposing economically optimal strategies that can achieve national climate targets and ensure the greatest local and national benefits as well as regional equality. Focusing on the biggest developing country China, we find this strategy can avoid up to 1.54% of cumulative GDP losses for approaching carbon neutrality, and more than 90% of regions would obtain economic gains compared either with existing independently launched targets or with the uniform strategy that all regions achieve peak carbon emissions before 2030. We also provide optimal carbon mitigation pathways to regional peak carbon, carbon intensity and energy consumption.

Osmotic stress is a major threaten to the growth and yield stability of Brassica napus. Post-translational modification with O-linked β-N-acetylglucosamine (O-GlcNAc) is ubiquitous in plants, and participates in a variety of signal transduction and metabolic regulation. However, studies on the role of O-GlcNAc transferase (OGT) in osmotic stress tolerance of plants are limited. In previous study, a O-glycosyltransferase, named BnaC09.OGT, was identified from the B. napus variety ‘Zhongshuang 11’ by yeast one hybrid with promoter of BnaA01.GPAT9. It was found that BnaC09.OGT localized in both nucleus and cytoplasm. The spatiotemporal expression pattern of BnaC09.OGT exhibited tissue specificity in developmental seed, especially in 15 days after pollination. In view of osmotic stress inducing, the BnaC09.OGT overexpression and knockout transgenic lines were constructed for biological function study. Phenotypic analysis of BnaC09.OGT overexpression seedlings demonstrated that BnaC09.OGT could enhance osmotic stress tolerance than WT and knockout lines in euphylla stage under 15% PEG6000 treatment after 7 days. In addition, compared with WT and knockout lines, overexpression of BnaC09.OGT had significantly higher activities of antioxidant enzymes (SOD and POD), higher content of soluble saccharide, and while significantly less content of malondialdehyde, proline and anthocyanidin under 15% PEG6000 treatment after 7 days. On the other hand, the unsaturated fatty acid content of BnaC09.OGT overexpression was significantly higher than that of WT and knockout lines, so it is speculated that the BnaC09.OGT could increase unsaturated fatty acid biosynthesis for osmotic stress tolerance by promoting the expression of BnaA01.GPAT9 in glycerolipid biosynthesis. In summary, the above results revealed that the function of BnaC09.OGT provides new insight for the analysis of the pathway of O-glycosylation in regulating osmotic stress tolerance in B. napus.

Objective To investigate sex difference of dynamic cerebral autoregulation (dCA) in different body positions. Methods A total of 130 healthy adults (65 males and 65 females) who underwent physical examination in Xuanwu Hospital Capital Medical University from August 2021 to November 2022 were included. Non⁃invasive continuous blood pressure monitor and transcranial Doppler ultrasonography (TCD) were used to monitor beat⁃to⁃beat blood pressure and middle cerebral artery (MCA) cerebral blood flow velocity (CBFV), respectively. Transfer function analysis (TFA) was used to compare and analyze the dCA parameters (absolute gain value and percentage gain, phase, coherence) in the range of very low frequency (0.02-0.07 Hz), low frequency (0.07-0.20 Hz) and high frequency (0.20-0.50 Hz) of different positions and sex. Results The phase in very low frequency and low frequency were significantly lower in the standing position than in the supine position [(57.37 ± 12.45)° vs. (66.72 ± 16.64)°, t = ⁃ 6.428, P = 0.000; (36.82 ± 11.59)° vs. (43.02 ± 11.51)°, t = ⁃ 6.052, P = 0.000], while the coherence in very low frequency and low frequency were significantly higher in the standing position than in the supine position (0.73 ± 0.06 vs. 0.66 ± 0.06, t = ⁃ 8.947, P = 0.000; 0.76 ± 0.06 vs. 0.73 ± 0.07, t = ⁃ 2.693, P = 0.007). In both supine and standing positions, absolute gain value of male in the low frequency [(0.91 ± 0.23) cm/(s·mm Hg) vs. (1.04±0.23)cm/(s·mm Hg), t =3.075, P =0.003; (0.84±0.19)cm/(s·mm Hg) vs. (0.95±0.21)cm/(s·mm Hg), t = 3.102, P = 0.002] and high frequency [(0.94 ± 0.25) cm/(s·mm Hg) vs. (1.11 ± 0.27) cm/(s·mm Hg), t = 3.740, P = 0.000; (0.91 ± 0.21) cm/(s·mm Hg) vs. (1.05 ± 0.23) cm/(s·mm Hg), t = 3.747, P = 0.000] was lower than those of female, while the phase in the low frequency was higher than that of female [(45.25 ± 10.21)° vs. (40.81 ± 12.37)°, t = ⁃ 2.239, P = 0.027; (38.90 ± 12.25)° vs. (34.74 ± 10.57)°, t = ⁃ 2.072, P = 0.040]. Conclusions Postural changes can also affect the cerebral autoregulation, and dCA can be improved in the supine position; there is no significant difference in dCA between male and female in the very low frequency, but there is a certain difference between the sex in the low frequency and high frequency.

Modeling of non-carbon dioxide (non-CO2) greenhouse gas (GHG) emissions in integrated assessment models (IAMs) is critical to achieving long-term climate targets. This study selects nine IAMs to reveal the modeling mechanisms and mitigation of non-CO2 GHGs. It further explores the impact of heterogeneity in the consideration of non-CO2 GHGs in IAMs on the climate targets and carbon dioxide removal (CDR) measures. The study shows that some IAMs cover fewer mitigation measures and do not include agricultural non-CO2 GHG emissions or fluorinated gases. The achievement of long-term climate targets can be affected by the lower mitigation potential of non-CO2 GHGs and the heterogeneity in non-CO2 GHG emissions will add uncertainty to the amount of negative CDR emissions.

ABSTRACT: Heat stress (HS) in broilers can be an environmental stressor that leads to intestinal inflammation and intestinal barrier damage. In order to examine the effect of Ban Lian Zi Jin San (BLZJS) on intestinal inflammation and barrier function in heat-stressed broilers, a model of chronic cyclic HS in broilers was established. A total of 300 twenty-one-day-old broilers were divided into 5 treatments at random. Broilers in 3 BLZJS dosage groups were kept in an ecologically controlled room at 37℃ ± 2℃ for 6 wk, and fed basal diets supplemented with 0.5, 1, and 2% BLZJS. Broilers in HS group were housed in the same room, but fed the basal diets. The findings indicated that supplementation of BLZJS significantly declined serum HS indexes levels (HSP70, HSP90), and increased serum antioxidant capacity (SOD and T-AOC) in broilers (P < 0.05). Besides, supplementation of BLZJS significantly inhibited the expression of HS indexes (HSP70 and HSP90), genes related to TLR4 inflammatory signal pathway (TLR4, MyD88, TRIF, IRAK-4, and NF-κB), inflammatory factors (IL-6 and TNF-α), and upregulated anti-inflammatory cytokines (IL-10) and intestinal tight junction-related genes (Occludin, Claudin-1, and ZO-1) in broiler jejunum (P < 0.05). On the other hand, supplementation of BLZJS could significantly reduce the protein expression of NF-κB and HSP70 in chick jejunum (P < 0.05). In conclusion, BLZJS inhibited the activation of TLR4 signal pathway and reduced the production of inflammatory factors, restoring the level of intestinal tight junction protein and protecting jejunal intestinal barrier function in heat-stressed broilers.

Histones have a vital function as components of nucleosomes, which serve as the fundamental building blocks of chromatin. Histone deacetylases (HDACs), which target histones, suppress gene transcription by compacting chromatin. This implies that HDACs have a strong connection to the suppression of gene transcription. Histone deacetylase 7 (HDAC7), a member of the histone deacetylase family, may participate in multiple cellular pathophysiological processes and activate relevant signaling pathways to facilitate the progression of different tumors by exerting deacetylation. In recent years, HDAC7 has been increasingly studied in the pathogenesis of tumors. Studies that are pertinent have indicated that it has a significant impact on the growth and metastasis of tumors, the formation of the vascular microenvironment, and the emergence of resistance to drugs. Therefore, HDAC7 could potentially function as a potent predictor for tumor prognosis and a promising target for mitigating drug resistance in tumors. This review primarily concentrates on elucidating the structure and function of HDAC7, its involvement in the development of various tumors, and its interplay with relevant signaling pathways. Meanwhile, we briefly discuss the research direction and prospect of HDAC7.

Abstract CD44 plays a key role in the communication of CSCs with the microenvironment and the regulation of stem cell properties. UALCAN was used to analyze the expression of CD44 in bladder cancer (BLCA) and normal tissue. The UALCAN was utilized to analyze the prognostic value of CD44 in BLCA.
