Due to a shortage of resources, exploring new biomass fillers has become critical for paper making. In this study, we reported an eco-friendly strategy for fabricating low-cost and efficient recycled paper with high mechanical properties using corrugated cardboard waste paper and harmless municipal sludge (HMS). First, the characteristics of HMS, including the particle size, pH, specific surface area, organic functional groups and organic component content, were analyzed. Then, the influence of the amount of HMS on the properties of the recycled paper was studied. Finally, the strengthening mechanism of biomass filling recycled paper was discussed. The results showed that HMS, a biomass filler for recycled paper preparation, exhibited excellent physical and chemical properties, with a median particle size of 6.395 μm, a surface area of 39.974 m2 g−1 and organic functional groups. The tensile index of the recycled paper with 30 % HMS was 13.10 Nm/g, which was 16.4 % greater than that of the unfilled paper and showed better thermal stability. This excellent performance could be attributed to the uniform distribution of HMS on the fiber surface, which improved the accessibility of hydrogen bond formation between fibers. An initial carbon emission calculation formula for the pulp molding industry was first proposed in accordance with the calculation guidelines provided by the Intergovernmental Panel on Climate Change (IPCC). The formula was primarily developed by utilizing the greenhouse gas accounting methods used in the pulp and paper industry and the life cycle assessment (LCA) theory. Carbon dioxide emissions coming into being during the production processes of dry-press and wet-press pulp molding were calculated individually, alongside the carbon emission factors throughout the lifecycle of the products. This study also delivers a comparative analysis of pulp molding products and similar cushion packaging materials. Moreover, the findings reveal that during the production phase, particularly in the formative stages of pulp molding, substantial carbon dioxide emissions would be produced, with dry-press pulp molding products exhibiting markedly higher carbon emission factors than their wet-press counterparts. The primary objective of achieving low-carbon transformation for the pulp molding industry was to reduce the carbon emissions caused by the forming process, primarily by improving the forming process and enhancing energy efficiency or utilizing clean energy. During the gene editing process mediated by CRISPR/Cas9, precise genome editing and gene knock-in can be achieved by the homologous recombination of double-stranded DNA (dsDNA) donor template. However, the low-efficiency of homologous recombination in eukaryotic cells hampers the development and application of this gene editing strategy. Here, we developed a novel CRISPR/Cas9-hLacI donor adapting system (DAS) to enhance the dsDNA-templated gene editing, taking the advantage of the specific binding of the LacI repressor protein and the LacO operator sequence derived for the Escherichia coli lactose operon. The codon-humanized LacI gene was fused as an adaptor to the Streptococcus pyogenes Cas9 ( SpCas9 ) and Staphylococcus lugdunensis Cas9 ( SlugCas9-HF ) genes, and the LacO operator sequence was used as the aptamer and linked to the dsDNA donor template by PCR. The Cas9 nuclease activity after the fusion and the homology-directed repair (HDR) efficiency of the LacO-linked dsDNA template were firstly examined using surrogate reporter assays with the corresponding reporter vectors. The CRISPR/Cas9-hLacI DASs mediated genome precise editing were further checked, and we achieved a high efficiency up to 30.5% of precise editing at the VEGFA locus in HEK293T cells by using the CRISPR/SlugCas9-hLacI DAS. In summary, we developed a novel CRISPR/Cas9-hLacI DAS for dsDNA-templated gene editing, which enriches the CRISPR/Cas9-derived gene editing techniques and provides a novel tool for animal molecular design breeding researches.
