Objective Exploring the influence of rapamycin on the progressing of hepatocellular carcinoma(HCC) in H-ras 12V transgenic mice. Methods The expression level of miRNA in liver tissue and peritumor tissue of transgenic mice was detected by high-throughput sequencing of miRNAs. Treatment of transgenic mice with rapamycin and the changes of tumor morphology were detected. The miR-183 expression levels were detected by RT-qPCR. The protein expression levels of mTOR, ERK and PDCD4, the downstream target gene of miR-183, were detected by Western blot. Results Compared with the peritumor tissue, the miR-183 expression in hepatocellular carcinoma tissue was increased significantly (P＜0.01). The protein expression levels of PDCD4 were decreased significantly(P＜0.05). Compared with the control group, after intervention with rapamycin, the counting of liver tumors decreased significantly(P＜0.05). The liver co-efficient decreased significantly(P＜0.05). The ERK phosphorylation increased significantly (P＜0.05). The miR-183 expression was decreased significantly (P＜0.05). The protein level of PDCD4 increased significantly (P＜0.05). Conclusions Rapamycin can alleviate hepatocellular carcinoma progression in H-ras 12V transgenic mice.

The protective effects of L-aspartic acid, L-valine, and L-proline on the stability of mulberry anthocyanins were investigated. Results showed that L-aspartic acid, L-valine, and L-proline significantly enhanced (p < 0.05) the stability of mulberry anthocyanins under constant light or ascorbic acid (AA). L-Proline had the best protective effect against anthocyanin degradation. The interaction between L-proline and cyanidin-3-O-glucoside (C3G) through hydrogen bonding and van der Waals forces, which improved the stability of C3G, was confirmed using FT-IR, 1H NMR, XRD, and molecular docking analyses, as well as molecular dynamics modes. In vitro digestion experiments yielded that both 2,2-Diphenyl-1-picrylhydrazyl (DPPH) and 2,2′-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) free radical scavenging capacities of the C3G/Pro group were increased in the intestinal fluid (p < 0.05). The above findings suggest that L-proline effectively slowed down the degradation of mulberry anthocyanins, and that it could be used as an auxiliary pigment and food additive to extend the optimal flavor period of products containing mulberry anthocyanins, and can improve the bioavailability of mulberry anthocyanins. Designing versatile rubber as a multifunctional elastomer is of great importance, incorporating it with biomass-derived nanoblocks will mitigate environmental challenges. Here biosynthesized natural rubber (NR) composites with CoFe2O4-immobilized biomass carbon (BC) derived from macadamia nutshells were fabricated by facile mechanical mixing. Morphological analysis indicates that CoFe2O4 nanoparticles are uniformly anchored on the surface of BC, forming intact electromagnetic loss networks in NR matrix. As a consequence, the as-fabricated NR/CoFe2O4@BC composites demonstrate enhanced mechanical, thermal, and electromagnetic performance. Particularly, NR/CoFe2O4@BC composite shows the best microwave attenuation capacity when CoFe2O4@BC loading is 40 phr, with the minimum reflection loss (RL) of −35.00 dB and effective absorption bandwidth (RL < −10 dB) of 1.60 GHz. All results indicate that this work open new paradigm for multiple applications based on biosynthetic elastomer with the sustainable biomass derived nanoblocks.

Anthocyanins are important secondary metabolites in many plant fruits and are excellent natural pigments with strong biological activities.However, anthocyanins are easily influenced by environmental and processing factors, leading to poor storage and processing stability.Therefore, in-depth research on the processing stability of anthocyanins and expanding their application areas has become a hot topic of concern both domestically and internationally.This article elaborated on the composition and structure of anthocyanins, investigated their stability in food, and provided a comprehensive review of domestic and international research progress on improving their stability.The aim is to provide a theoretical basis for the development and application of anthocyanins. This study explores the interaction between cyanidin‐3‐O‐glucoside (C3G), a water‐soluble pigment known for its diverse functional activities, and walnut protein isolate (WPI) as a potential stabilizing agent. Given the inherent instability of C3G, particularly its limited application in the food industry due to sensitivity to thermal and oxidative conditions, this research study aims to enhance its stability. According to the results of the fluorescence quenching experiment, C3G can efficiently quench WPI's intrinsic fluorescence through static quenching. Structural exploration revealed that C3G bound WPI via hydrophobic interaction force, with the number of bound C3G molecules (n) almost equivalent to 1. The ΔG value denoting change in Gibbs free energy for C3G binding with WPI was −8.05 kJ/mol, which indicated that the interaction between C3G and WPI is spontaneous. Moreover, the conformational structures of WPI were altered by C3G binding with a decrease in α‐helix contents and an increase in β‐turn, β‐sheet, and random coil contents. The thermal degradation kinetics indicate that after interacting with WPI, the half‐life of C3G increased by 1.62 times and 1.05 times at 80 and 95°C, respectively. The results of the oxidation stability test showed that the presence of WPI effectively reduced the discoloration and degradation of C3G caused by oxidation, and improved the oxidation stability of C3G. This study's findings will help to clarify the interaction mechanism between C3G and WPI, and broaden C3G's application scope in the food processing field. [ABSTRACT FROM AUTHOR]

We compared the expression of m6A methylation modulators and PD-L1 between HCC and normal in TCGA database. HCC samples were divided into two subtypes by consensus clustering of data from m6A RNA methylation regulators. The differences in PD-L1, immune infiltration, and prognosis between the two subtypes were further compared. The LASSO regression was used to build a risk score for m6A modulators. In addition, we identified miRNAs that regulate m6A regulators. Results. We found that fourteen m6A regulatory genes were significantly differentially expressed between HCC and normal. HCC samples were divided into two clusters. Of these, there are higher PD-L1 expression and poorer overall survival (OS) in cluster 1. There was a significant difference in immune cell infiltration between cluster 1 and cluster 2. Through the LASSO model, we obtained 12 m6A methylation regulators to construct a prognostic risk score. Compared with patients with a high-risk score, patients with a low-risk score had upregulated PD-L1 expression and worse prognosis. There was a significant correlation between risk score and tumor-infiltrating immune cells. Finally, we found that miR-142 may be the important regulator for m6A RNA methylation in HCC. Conclusion. Our results suggest that m6A RNA methylation modulators may affect the prognosis through PD-L1 and immune cell infiltration in HCC patients.
