Abstract

Background: At the time of AtTEnd trial design, standard treatment for advanced or recurrent endometrial cancer included carboplatin and paclitaxel chemotherapy. This trial assessed whether combining atezolizumab with chemotherapy might improve outcomes in this population. Methods: AtTEnd was a multicentre, double-blind, randomised, placebo-controlled, phase 3 trial done in 89 hospitals in 11 countries across Europe, Australia, New Zealand, and Asia. Enrolled patients were aged 18 years or older, and had advanced or recurrent endometrial carcinoma or carcinosarcoma, an Eastern Cooperative Oncology Group performance status of 0–2, and received no previous systemic chemotherapy for recurrence. Patients were randomly assigned (2:1) using an interactive web response system (block size of six) to either atezolizumab 1200 mg or placebo given intravenously with chemotherapy (carboplatin at area under the curve of 5 or 6 and paclitaxel 175 mg/m2 intravenously on day 1 every 21 days) for 6–8 cycles, then continued until progression. Stratification factors were country, histological subtype, advanced or recurrent status, and mismatch repair (MMR) status. Participants and treating clinicians were masked to group allocation. The hierarchically tested co-primary endpoints were progression-free survival (in patients with MMR-deficient [dMMR] tumours, and in the overall population) and overall survival (in the overall population). Primary analyses were done in the intention-to-treat population, defined as all randomly assigned patients who gave their full consent to participation in the study and data processing. Safety was assessed in all patients included in the intention-to-treat population who received at least one dose of study treatment. Here, we report the primary progression-free survival and the interim overall survival results. This study is ongoing and is registered with ClinicalTrials.gov, NCT03603184. Abstract

A new market for cocoa juice beverages is currently developing with the potential to solve the current cocoa industry challenges related to fluctuating and low cocoa prices. This study investigates the feasibility of processing cocoa pulp collected from smallholders as a side product from cocoa beans production. Producing cocoa pulp as a by-product requires re-organising the collection of pods and processing of cocoa juice in a new process. Two options are evaluated in this report: the collection of cocoa pods in a central processing unit and the collection of cocoa pods and processing of pulp in a mobile processing unit. The study identified critical success factors of a modular circular approach with different cocoa products. We calculated production and processing costs, investment costs, evaluated current market and farm-gate prices and looked into possible additional impacts from collecting cocoa pulp. In both cases cocoa pulp extraction is feasible. Further research is needed to understand the impact of selling whole pods and cocoa juice and to investigate further opportunities to make the cocoa juice production more profitable for smallholder farmers.

Abstract

La investigación profundiza el entendimiento de la trascendencia que tiene contar con un mecanismo formal de política macroprudencial enfocado en resguardar la solidez del sistema financiero. Siendo así, el estudio examinó el enfoque y facultades de los Consejos de Estabilidad Financiera para la gestión del riesgo sistémico en economías avanzadas y países miembros de la Alianza del Pacífico. Es una investigación cualitativa basada en la descripción y comprensión de la importancia de los Consejos de Estabilidad Financiera para la gestión del riesgo sistémico, para lo cual se recurrió a la teoría fundamentada de la estabilidad financiera. El principal resultado es la identificación de Consejos de Estabilidad Financiera que establecen parámetros regulatorios como Reino Unido y Estados Unidos; mientras que, en México, Chile y Colombia, funcionan como instancias de coordinación entre sus miembros que efectúan recomendaciones a fin de limitar los riesgos sistémicos. Se concluyó que, la estabilidad financiera, al ser un bien público, es un compromiso constitutivo que fomenta valores sociales, y su preservación, exige la responsabilidad de los Consejos de Estabilidad Financiera para promover el bienestar general de la población a través de la eliminación o reducción del riesgo sistémico.

Abstract

Numerous challenges – from population increase to climate change – threaten the sustainable development of cities and call for a fundamental change of urban development and green-blue resource management. Urban forests are vital in this transition, as they provide various ecosystem services and allow to re-shape and re-think cities. Based on a Europe-wide community effort with diverse experts centered around urban forests and urban greening, we propose five key research fields to generate the knowledge required to unlock fundamental changes in urban development and green-blue resource management: circular bioeconomy, climate resilience, governance, social and human environment, and biodiversity. To support the design of greener, cooler, more inclusive and resilient cities, all these research fields require inter- and transdisciplinary collaboration, engaging stakeholders in transforming urban engagement and functioning. We summarise main inter-, trans- und multi-disciplinary research paths for each field and the cross-cutting knowledge areas that can help to address the challenges many cities face (e.g., modelling and assessment of the urban microclimate). For transforming cities further knowledge is needed on e.g., urban innovation, transition, participation, and more. Finally, we address how the identified research gaps can be implemented (e.g., international coordinated research effort, interdisciplinary networks).

Abstract

Background: A better understanding of food-related behaviour and its determinants can be achieved through harmonisation and linking of the various data-sources and knowledge platforms. Scope: We describe the key decision-making in the development of a prototype of the Determinants and Intake Platform (DI Platform), a data platform that aims to harmonise and link data on consumer food behaviour. It will be part of the Food Nutrition Health Research Infrastructure (FNH-RI) that will facilitate health, social and food sciences. Approach: The decision-making was based on the evidence of user needs and data characteristics that guided the specification of the key building blocks of the DI Platform. Eight studies were carried out, including consumer online survey; interview studies of key DI Platform stakeholders; desk research and workshops. Key findings: Consumers were most willing to share data with universities, then industry and government. Trust, risk perception and altruism predicted willingness to share. For most other stakeholders non-proprietary data was most likely to be shared. Lack of data standards, and incentives for sharing were the main barriers for sharing data among the key stakeholders. The value of various data types would hugely increase if linked with other sources. Finding the right balance between optimizing data sharing and minimizing ethical and legal risks was considered a key challenge. Conclusions: The development of DI Platform is based on careful balancing of the user, technical, business, legal and ethical requirements, following the FAIR principles and the need for financial sustainability, technical flexibility, transparency and multi-layered organisational governance.

Abstract

This study describes a series of exploratory and confirmatory factor analyses that were conducted with the 44-item Social Anxiety Questionnaire for Children- 4th version (SAQ-CIV) to identify a reduced set of items that might be used to construct a new abbreviated instrument for measuring social anxiety in children and adolescents. The fourth version of the Social Anxiety Questionnaire for Children (SAQ-CIV) was administered to 12,801 non-clinical participants (ages 9 to 15 years) from 12 Latin American countries and Spain. Exploratory and confirmatory factor analysis supported a 6-factor structure of social anxiety in children, replicating a similar structure to that of adults (Caballo et al. in Behavioral Psychology/Psicología Conductual, 18(1), 5–34, 2010; Caballo et al. in Behavior Therapy, 43(2), 313–328, 2012): 1) Interactions with the opposite sex, 2) Criticism and embarrassment, 3) Speaking in public/Talking to teachers, 4) Assertive expression of annoyance and disgust, 5) Performing in public, and 6) Interactions with strangers. Each of the factors contains 4 items, yielding an abbreviated 24-item instrument, the Social Anxiety Questionnaire for Children (SAQ-C). The present results suggest this is a reliable, valid, and culturally sensitive instrument to assess social anxiety in youth.

40. Role of oxygen derived free radicals in platelet activating factor induced bowel necrosis.

Abstract

The mechanism of tissue and cell injury in ischaemic bowel necrosis is unclear. The present study investigated the role of oxygen derived free radicals in the development of bowel necrosis using injections of platelet activating factor (PAF) into the mesenteric vasculature. Animals were pretreated with allopurinol or superoxide dismutase together with catalase, before administration of PAF. Superoxide dismutase/catalase markedly improved the PAF-induced lesions, indicating that most of the intestinal damage after PAF injection is because of the release of oxygen radicals. The major source of oxygen radicals is xanthine oxidase, as allopurinol ameliorated small bowel lesions. Pretreatment with allopurinol produced a significant (p less than 0.01) preventive effect on PAF induced hypotension. In contrast, superoxide dismutase/catalase did not alter PAF induced hypotension. Superoxide dismutase/catalase pretreatment improved PAF induced haemoconcentration and leucopenia, while allopurinol showed no effect. [ABSTRACT FROM PUBLISHER]

Abstract

Novel protein feeds are needed in ecological and organic poultry production to supply natural sources of amino acids, particularly methionine. Synthetic amino acids are increasingly banned in certified organic production. Carp is an invasive fish in the United States and its harvest can help provide natural source of methionine, while reducing an undesirable population. In our previous study, we formulated a diet using extruded carp-based meal (CBM) to replace synthetic methionine in broiler diets and conducted a feeding trial to test its effect on broiler performance. For this study, birds were processed at the end of the feeding trial and evaluated for quality and sensory attributes of meat from birds fed with CBM diet. Controls were normal diet that contained methionine and the carp meal was used to replace methionine and a portion of soybean meal. Results suggest that birds fed with CBM diet did not differ from the Control in live weight, carcass weight, breast weight, ready-to-cook yield, wing yield, tenders, and thigh yields (P> 0.05). Breast meat was more light (L*) for birds from the CBM treatment compared to the Control and the pH was lower (P< 0.05). For the thigh meat, birds fed CBM diets resulted in higher protein and less fat compared to the Control (P< 0.05). In the consumer analysis, overall impression, flavor, and mouthfeel were lower for the CBM breast meat than the Control and scored lower in terms of tenderness and juiciness (P< 0.05). Interestingly, thigh meat from CBM had more omega-3 fatty acids, which has a human health benefit. The results indicate that carp-based meal hold potential as a novel protein to replace synthetic methionine in organic broiler diets.
