Andersen-Tawil syndrome (ATS) is a rare inheritable disease associated with loss-of-function mutations in KCNJ2, the gene coding the strong inward rectifier potassium channel Kir2.1, which forms an essential membrane protein controlling cardiac excitability. ATS is usually marked by a triad of periodic paralysis, life-threatening cardiac arrhythmias and dysmorphic features, but its expression is variable and not all patients with a phenotype linked to ATS have a known genetic alteration. The mechanisms underlying this arrhythmogenic syndrome are poorly understood. Knowing such mechanisms would be essential to distinguish ATS from other channelopathies with overlapping phenotypes and to develop individualized therapies. For example, the recently suggested role of Kir2.1 as a countercurrent to sarcoplasmic calcium reuptake might explain the arrhythmogenic mechanisms of ATS and its overlap with catecholaminergic polymorphic ventricular tachycardia. Here we summarize current knowledge on the mechanisms of arrhythmias leading to sudden cardiac death in ATS. We first provide an overview of the syndrome and its pathophysiology, from the patient's bedside to the protein and discuss the role of essential regulators and interactors that could play a role in cases of ATS. The review highlights novel ideas related to some post-translational channel interactions with partner proteins that might help define the molecular bases of the arrhythmia phenotype. We then propose a new all-embracing classification of the currently known ATS loss-of-function mutations according to their position in the Kir2.1 channel structure and their functional implications. We also discuss specific ATS pathogenic variants, their clinical manifestations, and treatment stratification. The goal is to provide a deeper mechanistic understanding of the syndrome toward the development of novel targets and personalized treatment strategies. [ABSTRACT FROM AUTHOR]

- Abstract
Adult cardiac progenitor/stem cells (CPC/CSC) are multipotent resident populations involved in cardiac homeostasis and heart repair. Assisted by complementary RNAseq analysis, we defined the fraction of the CPC proteome associable with specific functions by comparison with human bone marrow mesenchymal stem cells (MSC), the reference population for cell therapy, and human dermal fibroblasts (HDF), as a distant reference. Label-free proteomic analysis identified 526 proteins expressed differentially in CPC. iTRAQ analysis confirmed differential expression of a substantial proportion of those proteins in CPC relative to MSC, and systems biology analysis defined a clear overrepresentation of several categories related to enhanced angiogenic potential. The CPC plasma membrane compartment comprised 1,595 proteins, including a minimal signature of 167 proteins preferentially or exclusively expressed by CPC. CDH5 (VE-cadherin), OX2G (OX-2 membrane glycoprotein; CD200), GPR4 (G protein-coupled receptor 4), CACNG7 (calcium voltage-gated channel auxiliary subunit gamma 7) and F11R (F11 receptor; junctional adhesion molecule A; JAM-A; CD321) were selected for validation. Their differential expression was confirmed both in expanded CPC batches and in early stages of isolation, particularly when compared against cardiac fibroblasts. Among them, GPR4 demonstrated the highest discrimination capacity between all cell lineages analyzed.

The MAP-i Doctoral Programme in Informatics, of the Universities of Minho, Aveiro and Porto, In order to attain the promises of the Cloud Computing paradigm, systems need to be able to transparently adapt to environment changes. NoSQL databases, which are becoming pivotal systems in nowadays cloud infrastructures, exhibit the highly desirable scalability and availability properties. Scalability achieved by these databases is anchored on data independence; there is no clear relationship between data, and atomic inter-node operations are not a concern. Such assumption over data allows a paradigm shift on how to achieve the best performance. Unfortunately, current solutions put the burden on the application's developer to handle and master the speci cities of each system that is hindering a broader adoption. In this dissertation, we tackle the several shortcomings in current implementations of cloud-based NoSQL databases at three di erent levels. First, we present a cloud-enabled framework for the automatic and heterogeneous recon guration of NoSQL databases. This framework enables NoSQL databases to become autonomously elastic while providing a new load balancing component that takes into account data access patterns. Secondly, we propose a novel mechanism to partition data that takes into account the system workload. It estimates, in an autonomous way, a splitting point that leads to optimal load balancing in terms of requests. Finally, we develop a mechanism to accurately predict the resource usage of NoSQL databases resorting to an o ine trained model. Then, it can accurately estimate in real time the database resource usage for any request distribution only by knowing two parameters: i) cache hit ratio; and ii) incoming throughput. Both input values can be observed in real time or synthesized for request allocation decisions, furthermore it is su ciently simple and generic so it can be used with several databases., De modo a alcançar as promessas do paradigma da computação na nuvem, os sistemas têm de ser capazes de adaptar-se às mudanças de uma forma transparente. Os bancos de dados NoSQL que estão-se a tornar sistemas cruciais nas infraestruturas da nuvem, possuem as propriedades de escalabilidade e elevada disponibilidade. A escalabilidade está assente na independência de dados; pois não existe uma relação clara entre os mesmos, e operações atómicas que envolvam mais que um nó não são uma preocupação. Tal pressuposto permite uma mudança de paradigma na forma de alcançar o melhor desempenho. Infelizmente, as soluções atuais requerem responsabilidades adicionais a quem desenvolve as aplicações, nomeadamente a necessidade de manipular e dominar as especificidades de cada sistema. Esta situação esta a dificultar a adoção do paradigma. Nesta dissertação, abordamos v arias lacunas das atuais implementações de bases de dados NoSQL a três diferentes níveis. Primeiramente, apresentamos um sistema que permite a reconfiguração automática e heterogénea de bases de dados NoSQL, que permite ainda que essas mesmas bases de dados se tornem autonomamente elásticas e simultaneamente balancear a carga tendo em conta os padrões de acesso. Em segundo lugar, propomos um novo mecanismo de particionamento de dados que tem em conta o estado atual do sistema, ao estimar de forma autónoma qual o ponto ideal de divisão baseado nos pedidos. Finalmente, desenvolvemos um mecanismo para prever com precisão o uso de recursos pelas bases de dados NoSQL com base num modelo construído em modo online. Esse modelo permite estimar com elevada precisão e em tempo real o uso de recursos da base de dados para qualquer distribuição somente conhecendo dois parâmetros: i) a taxa de acessos com sucesso da cache e ii) o desempenho. Introduction: Intraocular refractive procedures with the implantation of a Phakic Intraocular lens have become a safe efficient and predictable alternative for treating high ametropias when the use of corneal photoablative procedures is not possible. The implantation of Phakic intraocular lens preservs the accomodative function,is a reversable refractive procedure, with minimal induction of higher order aberrations compaed with corneal photoablative procedures. Methods: An analytical review of phakic intraocular lens was performed. Results: Critical issues of phakic intraocular lens indications, ancillary tests, options regading models designs, ranging, sizing , safety matters and surgicak care was discussed. En este articulo investigue la relación que puede existir entre el conocimiento adquirido en la especialización en alta gerencia, y la actual forma de gestión seguridad en el escenario empresarial, el cual ha evolucionado desde el empirismo hasta la actual profesionalización liderada por la Universidad Militar Nueva Granada. Hoy en día, la profesionalización de la seguridad tanto en Colombia, como en Latinoamérica, es un arte que va en franco desarrollo a convertirse en una verdadera ciencia. Para nuestro caso, este desarrollo del conocimiento va desde un sinnúmero de diplomados, a un único pregrado y una única especialización. Por esta razón, desde la óptica académica se ha podido identificar que los contenidos curriculares están direccionados hacia el desarrollo de una fuerte labor técnica. Pero, ¿en la actualidad los profesionales de la seguridad necesitan un mayor nivel de formación que les genere herramientas para enfrentar los nuevos retos que demandan los cada vez mas exigentes y complejos procesos organizacionales?, la respuesta es: definitivamente, SI !!!!!!. En merito de la presente problemática, este articulo de investigación es una reflexión propositiva con visión académica que propenda por la generación de nuevas herramientas a través del desarrollo de un modelo académico de gerencia de alto nivel para la gestión de seguridad organizacional. This paper investigates the relationship that may exist between the knowledge gained specialization in senior management, and the current form of management security business scenario, which has evolved from empiricism to the current professionalization led by the Universidad Militar Nueva Granada . Today, the professionalization of security both in Colombia and in Latin America , is an art that goes into full development to become a true science. In our case , the development of knowledge ranging from countless graduates, to one undergraduate and unique expertise. For this reason, it has been from the academic perspective identify which curricula are directed towards the development of a strong technical work . But today 's security professionals need a higher level of training that will generate new tools to address the challenges demanded by increasingly demanding and complex organizational processes ?, the answer is definitely YES !!!!!! . In the last years it has been increasingly clear that KV-channel activity modulates neurotransmitter release. The subcellular localization and composition of potassium channels are crucial to understanding its influence on neurotransmitter release. To investigate the role of KV in corticostriatal synapses modulation, we combined extracellular recording of population-spike and pharmacological blockage with specific and nonspecific blockers to identify several families of KV channels. We induced paired-pulse facilitation (PPF) and studied the changes in paired-pulse ratio (PPR) before and after the addition of specific KV blockers to determine whether particular KV subtypes were located pre- or postsynaptically. Initially, the presence of KV channels was tested by exposing brain slices to tetraethylammonium or 4-aminopyridine; in both cases we observed a decrease in PPR that was dose dependent. Further experiments with tityustoxin, margatoxin, hongotoxin, agitoxin, dendrotoxin, and BDS-I toxins all rendered a reduction in PPR. In contrast heteropodatoxin and phrixotoxin had no effect. Our results reveal that corticostriatal presynaptic KV channels have a complex stoichiometry, including heterologous combinations KV1.1, KV1.2, KV1.3, and KV1.6 isoforms, as well as KV3.4, but not KV4 channels. The variety of KV channels offers a wide spectrum of possibilities to regulate neurotransmitter release, providing fine-tuning mechanisms to modulate synaptic strength. [ABSTRACT FROM AUTHOR]

La amplia magnitud de la migración venezolana en Colombia no solo se dio por la solidez de la crisis en Venezuela, sino también porque el país receptor en su historia no había afrontado un fenómeno como este, lo cual esforzó al Gobierno a establecer planes de acción instantáneos debido a que no existía una planificación específica para afrontar dicha emergencia. Los efectos del proceso migratorio son variados, puesto que se relacionan con la salud, la alimentación, el trabajo y la vivienda. Sin embargo, uno de los más importantes y de mayor trascendencia es la seguridad, ya que son las zonas fronterizas las principales áreas afectadas ante el volumen de la población y las condiciones en las que llegan a estos lugares. Adicionalmente, las medidas impuestas por el presidente Nicolás Maduro han fragmentado la tranquilidad en los departamentos limítrofes principalmente. The effects of the migration process are varied, as they relate to health, food, work and housing. However, one of the most important and important is security, since it is the border areas that are the main areas affected by the size of the population and the conditions in which they reach these places. In addition, the measures imposed by President Nicolás Maduro have fragmented tranquillity in the neighbouring departments mainly. Los negocios asociados a la prestación de servicios en el sector de la vigilancia y seguridad privada, cada día toman más realce teniendo en cuenta los altos ingresos operacionales que se pueden recibir con la prestación de los mismos. El sector de la vigilancia y seguridad privada fue regulado y normado a partir del Decreto Ley 356 del año 1994 mediante el cual se creó el Estatuto de Vigilancia y seguridad privada en el cual se especifican los requisitos para la operación en el sector. Por ser un negocio atractivo, la seguridad privada ha tenido inversión extranjera, sin embargo se encuentra un vacío jurídico para que las empresas extranjeras puedan invertir en este sector teniendo en cuenta lo contemplado en algunas normas donde se exceptúan las actividades de defensa y seguridad nacional y que los socios de estas empresas sean de nacionalidad Colombiana, sin embargo en la actualidad existe la operación de algunas de ellas. El Congreso de la República en la actualidad está estudiando un proyecto de ley para regular la vigilancia y seguridad privada y de esta manera actualizar la norma que se encuentra vigente desde el año 1994, este proyecto tiene incluido algunos artículos que probablemente afecta directamente la participación y continuidad de empresas extranjeras en el negocio de la seguridad privada. El presente ensayo analiza las diferente normatividad jurídica y el estado actual de la inversión extranjera en Colombia. The private security and surveillance sector was regulated and regulated by Decree Law 356 of 1994, which created the Statute of Surveillance and Private Security in which the requirements for the operation in the sector are specified. En el desarrollo del ensayo además se encontraran aspectos como los ámbitos de aplicación, los aportes que este genera, la importancia de su adecuada aplicación y los resultados que se pueden obtener siendo estos últimos los más influyentes para la adecuada administración del riesgo, su gestión y tratamiento. Dissertação de mestrado em Engenharia Informática, NoSQL databases opt not to offer important abstractions traditionally found in relational databases in order to achieve high levels of scalability and availability: transactional guarantees and strong data consistency. These limitations bring considerable complexity to the development of client applications and are therefore an obstacle to the broader adoption of the technology. In this work we propose a middleware layer over NoSQL databases that offers transactional guarantees with Snapshot Isolation. The proposed solution is achieved in a non-intrusive manner, providing to the clients the same interface as a NoSQL database, simply adding the transactional context. The transactional context is the focus of our contribution and is modularly based on a Non Persistent Version Store that holds several versions of elements and interacts with an external transaction certifier. In this work, we present an implementation of our system over Apache Cassandra and by using two representative benchmarks, YCSB and TPC-C, we measure the cost of adding transactional support with ACID guarantees., As bases de dados NoSQL optam por não oferecer importantes abstrações tradicionalmente encontradas nas bases de dados relacionais, de modo a atingir elevada escalabilidade e disponibilidade: garantias transacionais e critérios de coerência de dados fortes. Estas limitações resultam em maior complexidade no desenvolvimento de aplicações e são por isso um obstáculo à ampla adoção do paradigma. Neste trabalho, propomos uma camada de middleware sobre bases de dados NoSQL que oferece garantias transacionais com Snapshot Isolation. A abordagem proposta e não-intrusiva, apresentando aos clientes a mesma interface NoSQL, acrescendo o contexto transacional. Este contexto transacional e o cerne da nossa contribuição e assenta modularmente num repositório de versões não-persistente e num certificador externo de transações concorrentes. 23. Promising tools for future drug discovery and development in antiarrhythmic therapy .

- Abstract
Arrhythmia refers to irregularities in the rate and rhythm of the heart, with symptoms spanning from mild palpitations to life-threatening arrhythmias and sudden cardiac death (SCD). The complex molecular nature of arrhythmias complicates the selection of appropriate treatment. Current therapies involve the use of antiarrhythmic drugs (class I-IV) with limited efficacy and dangerous side effects and implantable pacemakers and cardioverter-defibrillators with hardware-related complications and inappropriate shocks. The number of novel antiarrhythmic drug in the development pipeline has decreased substantially during the last decade and underscores uncertainties regarding future developments in this field. Consequently, arrhythmia treatment poses significant challenges, prompting the need for alternative approaches. Remarkably, innovative drug discovery and development technologies show promise in helping advance antiarrhythmic therapies. Here, we review unique characteristics and the transformative potential of emerging technologies that offer unprecedented opportunities for transitioning from traditional antiarrhythmics to next-generation therapies. We assess stem cell technology, emphasizing the utility of innovative cell profiling using multi-omics, high-throughput screening, and advanced computational modeling in developing treatments tailored precisely to individual genetic and physiological profiles. We offer insights into gene therapy, peptide and peptibody approaches for drug delivery. We finally discuss potential strengths and weaknesses of such techniques in reducing adverse effects and enhancing overall treatment outcomes, leading to more effective, specific, and safer therapies. Altogether, this comprehensive overview introduces innovative avenues for personalized rhythm therapy, with particular emphasis on drug discovery, aiming to advance the arrhythmia treatment landscape and the prevention of SCD. Significance Statement Arrhythmias and sudden cardiac death account for 15-20% of deaths worldwide. However, current antiarrhythmic therapies are ineffective and with dangerous side effects. Here, we review the field of arrhythmia treatment underscoring the slow progress in advancing the cardiac rhythm therapy pipeline and the uncertainties regarding evolution of this field. We provide information on how emerging technological and experimental tools can help accelerate progress and address the limitations of antiarrhythmic drug discovery.
