Moringa oleifera Lam.(Moringaceae) et Sclerocarya birrea [(A. Rich.) Hochst.] (Asclepiadaceae) sont deux plantes utilisées en médecine traditionnelle au Burkina Faso dans le traitement du diabète. L’objectif de notre travail a été d’étudier les effets protecteurs des extraits hydro-éthanoliques (80 %) de fruits de Moringa oleifera (MO) et d’écorces de tronc de Sclerocarya birrea (SB), sur la viabilité et la fonctionnalité de la cellule bêta pancréatique, lors d’un stress oxydant induit par H2O2. La viabilité a été évaluée par la technique du [3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazoliumbromide] (MTT) et la fonctionnalité par la mesure de la sécrétion d’insuline induite par le glucose (8,3 mM). Nous avons montré que MO et SB à 10 μg/mL et au bout de deux heures prévenaient partiellement l’altération de la viabilité induite par H2O2. L’altération de la fonctionnalité était prévenue totalement par 10 μg/mL de MO au bout d’une heure et 1 μg/mL de SB au bout de deux heures. The aim of our study was to investigate the protective effects of ethanol (80 %) extract of Moringa oleifera (MO) fruits and stem barks of Sclerocarya birrea (SB) on the viability and functionality of the pancreatic β-cells, during oxidative stress induced by H2O2. Viability was evaluated by the [3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide] (MTT) colorimetric assay and functionality by the measurement of insulin secretion induced by glucose (8.3 mM). We showed that MO and SB(10 μg/mL) after two hours partially prevented the alteration of viability induced by H2O2 (50 μM). The alteration of the functionality is completely prevented by 10 μg/mL of MO after one hour, and 1 μg/mL of SB after two hours. Moringa oleifera and Sclerocarya birrea protected β cell viability and functionality against oxidative stress induced exogenously. During the last decade, gene therapy via ex vivo gene transfer into autologous hematopoietic stem cells has emerged as a convincing therapy for severe combined immunodeficiency caused by ILR2G mutation (SCID-X1) despite the occurrence of genotoxicity caused by the integration of first-generation retroviral vectors. However, the place of gene therapy among the therapeutic armamentarium remains to be defined. We retrospectively analyze and compare clinical outcomes and immune reconstitution in 13 consecutive SCID-X1 patients having undergone haploidentical hematopoietic stem cell transplantation (HSCT) and 14 SCID-X1 patients treated with gene therapy over the same period at a single center level: the Necker Children's Hospital (Paris, France). Our results show a clear advantage in terms of T-cell development of gene therapy over HSCT with a mismatched donor. Patients treated with gene therapy display a faster T-cell reconstitution and a better long-term thymic output. Interestingly, this advantage of gene therapy vs haploidentical HSCT seems to be independent of the existence of clinical graft-versus-host disease in the latter condition. If data of safety are confirmed over the long term, gene therapy for SCID-X1 appears to be an equal, if not superior, alternative to haploidentical HSCT.

Le Languedoc-Roussillon présente des caractéristiques particulières ou un niveau particulier de risques: le vieillissement de la population, particulièrement évident dans la région, ainsi que l'augmentation de la population en situation précaire posent des problèmes de nutrition et de prise en charge. L'alimentation de type méditerranéen, dont les bienfaits pour la santé sont reconnus, est progressivement remplacée par une alimentation mondialisée, standardisée et peu variée, ce qui entraîne, avec le développement de la sédentarité, une épidémie d'obésité/diabète avec, entre autres, des conséquences cardio-vasculaires. Parmi les maladies émergentes, il faut noter certaines formes de cancer, des maladies neuro-dégénératives (Alzheimer), et nombre de maladies infectieuses et parasitaires qui, du fait des échanges constants avec la région, risquent de se développer, les vecteurs ou réservoirs étant présents sur notre territoire. Une réflexion des responsables locaux et de la population est nécessaire.

Terminalia superba is highly regarded in some parts of Cameroon in traditional medical practice. We have studied the vasorelaxant effects of the stem bark methanol extract of T. superba on rat vascular smooth muscle. The results demonstrated that T. superba extract provoked a time-dependent relaxation of aortic rings precontracted with norepinephrine (10(-6) M). The vasorelaxant effect of the plant extract was not affected by endothelium removal or by pretreatment with indomethacin or N(W)-nitro-Larginine methyl ester (L-NAME). T. superba extract did not significantly, affect the contraction induced by 30 mM or 60 mM KCl as compared to those induced by NE. Relaxations elicited by T. superba extract were markedly reduced by glibenclamide, a putative blocker for K(ATP) channels and by tetraethylammonium, the non-specific K+ channel inhibitor. T. superba caused a time- and concentration-dependent relaxation of the rat aortic rings that were inhibited by charybdotoxin and iberotoxin but not by apamin. These finding indicate that T. superba extract at least partially relaxes the rat aorta by activating K+ channels, mainly KATP channels and large-conductance Ca2+ -activated K+ channels in rat aorta.
