1. Membrane Heating in Living Tissues Exposed to Nonthermal Pulsed EM Fields

Abstract: A bio tissue model consisting of multilayer spherical cells including four nested radial domains (nucleus, nuclear membrane, cytoplasm and plasma membrane) is worked out to derive the cell heating dynamics in presence of membrane capacitance dispersion under pulsed electromagnetic exposure. Two possible cases of frequency-dependent membrana models are discussed: plasma and nuclear membranes are dispersive, only the nuclear memebrane is dispersive . In both models an high localized heating of the membranes occurs, without significant temperature rise in the cytoplasm and nucleoplasm., Comment: 9 pages, 7 figures (preprint in piubblication on IEEE Transacton on Plasma Science)

Abstract: We report the results of a multimessenger search for coincident signals from the LIGO and Virgo gravitational-wave observatories and the partially completed IceCube high-energy neutrino detector, including periods of joint operation between 2007-2010. These include parts of the 2005-2007 run and the 2009-2010 run for LIGO-Virgo, and IceCube's observation periods with 22, 59 and 79 strings. We find no significant coincident events, and use the search results to derive upper limits on the rate of joint sources for a range of source emission parameters. For the optimistic assumption of gravitational-wave emission energy of $10^{-2}$\,M$_\odot$c$^2$ at $\sim 150$\,Hz with $\sim 60$\,ms duration, and high-energy neutrino emission of $10^{51}$\,erg comparable to the isotropic gamma-ray energy of gamma-ray bursts, we limit the source rate below $1.6 \times 10^{-2}$\,Mpc$^{-3}$yr$^{-1}$. We also examine how combining information from gravitational waves and neutrinos will aid discovery in the advanced gravitational-wave detector era.

Abstract: In this paper we report on a search for short-duration gravitational wave bursts in the frequency range 64 Hz-1792 Hz associated with gamma-ray bursts (GRBs), using data from GEO600 and one of the LIGO or Virgo detectors. We introduce the method of a linear search grid to analyse GRB events with large sky localisation uncertainties such as the localisations provided by the Fermi Gamma-ray Burst Monitor (GBM). Coherent searches for gravitational waves (GWs) can be computationally intensive when the GRB sky position is not well-localised, due to the corrections required for the difference in arrival time between detectors. Using a linear search grid we are able to reduce the computational cost of the analysis by a factor of O(10) for GBM events. Furthermore, we demonstrate that our analysis pipeline can improve upon the sky localisation of GRBs detected by the GBM, if a high-frequency GW signal is observed in coincidence. We use the linear search grid method in a search for GWs associated with 129 GRBs observed satellite-based gamma-ray experiments between 2006 and 2011. The GRBs in our sample had not been previously analysed for GW counterparts. A fraction of our GRB events are analysed using data from GEO600 while the detector was using squeezed-light states to improve its sensitivity; this is the first search for GWs using data from a squeezed-light interferometric observatory. We find no evidence for GW signals, either with any individual GRB in this sample or with the population as a whole. For each GRB we place lower bounds on the distance to the progenitor, assuming a fixed GW emission energy of $10^{-2} M_{\odot}c^{2}$, with a median exclusion distance of 0.8 Mpc for emission at 500 Hz and 0.3 Mpc at 1 kHz. Abstract: We present the results of a search for gravitational waves associated with 223 gamma-ray bursts (GRBs) detected by the InterPlanetary Network (IPN) in 2005-2010 during LIGO's fifth and sixth science runs and Virgo's first, second and third science runs. The IPN satellites provide accurate times of the bursts and sky localizations that vary significantly from degree scale to hundreds of square degrees. We search for both a well-modeled binary coalescence signal, the favored progenitor model for short GRBs, and for generic, unmodeled gravitational wave bursts. Both searches use the event time and sky localization to improve the gravitational-wave search sensitivity as compared to corresponding all-time, all-sky searches. We find no evidence of a gravitational-wave signal associated with any of the IPN GRBs in the sample, nor do we find evidence for a population of weak gravitational-wave signals associated with the GRBs. For all IPN-detected GRBs, for which a sufficient duration of quality gravitational-wave data is available, we place lower bounds on the distance to the source in accordance with an optimistic assumption of gravitational-wave emission energy of $10^{-2}M_{\odot}c^2$ at 150 Hz, and find a median of 13 Mpc. For the 27 short-hard GRBs we place 90% confidence exclusion distances to two source models: a binary neutron star coalescence, with a median distance of 12Mpc, or the coalescence of a neutron star and black hole, with a median distance of 22 Mpc. Abstract: The general problem of computing the false-alarm rate vs. detection-threshold relationship for a bank of correlators is addressed, in the context of maximum-likelihood detection of gravitational waves, with specific reference to chirps from coalescing binary systems. Accurate (lower-bound) approximants for the cumulative distribution of the whole-bank supremum are deduced from a class of Bonferroni-type inequalities. The asymptotic properties of the cumulative distribution are obtained, in the limit where the number of correlators goes to infinity. The validity of numerical simulations made on small-size banks is extended to banks of any size, via a gaussian-correlation inequality. E57, 6470, 1998) a thorough characterization in terms of receiver operating characteristics (ROCs) of stochastic-resonance (SR) detectors of weak harmonic signals of known frequency in additive gaussian noise was given. It was shown that strobed sign-counting based strategies can be used to achieve a nice trade-off between performance and cost, by comparison with non-coherent correlators. Here we discuss the more realistic case where besides the sought signal (whose frequency is assumed known) further unwanted spectrally nearby signals with comparable amplitude are present. Abstract: The final inspiral phase in the evolution of a compact binary consisting of black holes and/or neutron stars is among the most probable events that a network of ground-based interferometric gravitational wave detectors is likely to observe. Gravitational radiation emitted during this phase will have to be dug out of noise by matched-filtering (correlating) the detector output with a bank of several $10^5$ templates, making the computational resources required quite demanding, though not formidable. We propose an interpolation method for evaluating the correlation between template waveforms and the detector output and show that the method is effective in substantially reducing the number of templates required. Indeed, the number of templates needed could be a factor $\sim 4$ smaller than required by the usual approach, when the minimal overlap between the template bank and an arbitrary signal (the so-called {\it minimal match}) is 0.97. Abstract: Achieving net zero emissions will require an unprecedented transformation of our infrastructure system. Different economies will face varying challenges, but the transformation of the global economy to be environmentally sustainable and resilient includes decarbonising industry and transport, building smart energy systems, and increasing access to affordable, clean energy. Understanding the role of the private sector and financial markets will be crucial in achieving a decisive transition. At the same time, institutional investors such as pension funds are facing increasingly complex and challenging macro-conditions and financial markets. Investors are searching for organizational innovation and reconfiguration of their investment models to meet their long-term objectives. Following the Covid crisis with an increasing interest in sustainability among investors, there is an opportunity to scale up institutional investment in low carbon infrastructure and to protect and grow the retirement savings of fund members. Reframing finance will help to respond to the urgent need to act on energy transition while at the same time supporting the economy. Objective of this thesis is to examine the of role institutional investors and in particular pension funds in sustainable and low-carbon infrastructure, diversifying sources of finance and fostering the development of infrastructure as an asset class. To illustrate how long term investors can promote a more inclusive and sustainable financial system while contributing to the real economy I have adopted an economic geography and bottom up approach benefitting from the unique access to micro data for infrastructure analysis. Following this approach, this thesis builds on literature on financial intermediation and the theory of finance, design and governance of infrastructure, the geography of finance, pension fund research and emerging debates on “relational geometries.” Beyond the lack of data, there is a lack of analysis in academia and policy institutions on the connection between policies affecting investors’ portfolios strategies and political decisions for low carbon infrastructure.The institutional investor’s investment strategies and the decision to invest in infrastructure are based on strategic asset allocation and asset-liability management considerations. This thesis has analyzed how the changing nature of long-term savings is shaping the current and future demand for long-term assets and the impact of demographic and financial pressure on infrastructure investment. Asset allocation analysis has shown the overall trend to diversify investment portfolios in private markets, impact of ESG investment integration and increasing interest in low carbon infrastructure investment. At the same time government choices about different types of private funding models and forms of financing of infrastructure are based on short-term political priorities and national constraints. Renewable energy investment is growing, being part of infrastructure asset allocations. However the limited overall investment in low carbon infrastructure is showing major disconnections between investors and governments expectations. As seen in the thesis, partnership models are emerging as a way to leverage traditional bank and utility finance in primary markets and share risks as well as organizational capacity and expertise in support of the financing of infrastructure projects. Well-functioning and deep capital markets, supported by pooling assets, securitization, yieldcos and new listed vehicles could widen the access to and improve transparency of infrastructure investment while diversifying risk and lowering the cost of capital.

Abstract: Photosynthetic reactions in plants, algae, and cyanobacteria are driven by photosystem I and photosystem II complexes, which specifically reduce or oxidize partner redox biomolecules. Photosynthetic complexes can also bind synthetic organic molecules, which inhibit their photoactivity and can be used both to study the electron transport chain and as herbicides and algicides. Thus, their development, characterization, and sensing bears fundamental and applied interest. Substantial efforts have been devoted to developing photosensors based on photosystem II to detect compounds that bind to the plastoquinone sites of this complex. In comparison, photosystem I based sensors have received less attention and could be used to identify novel substances displaying phytotoxic effects, including those obtained from natural product extracts. We have developed a robust procedure to functionalize gold electrodes with photo- and redox-active photosystem I complexes based on transparent gold and a thiolate self-assembled monolayer, and we have obtained reproducible electrochemical photoresponses. Chronoamperometric recordings have allowed us to measure photocurrents in the presence of the viologen derivative paraquat at concentrations below 100 nM under lock-in operation and a sensor dynamic range spanning six orders of magnitude up to 100 mM. We have modeled their time course to identify the main electrochemical processes and limiting steps in the electron transport chain. Our results allow us to isolate the contributions from photosystem I and the redox mediator, and evaluate photocurrent features (spectral and power dependence, fast transient kinetics) that could be used as a sensing signal to detect other inhibitors and modulators of photosystem I activity.

Abstract: Introduction: In mental health prevention, person-centered, and rights-based approaches, the role of recovery is highlighted (WHO, 2021). Various evaluation tools are used in rehabilitation objectives and programs, including the Specific Levels of Functioning Scale (SLOF) (Mucci et al. Schizophr Res 2014;159 144-50) and the Recovery Assessment Scale – Domains and Stages (RAS-DS), a self-measure of mental health recovery. It includes 38 items clustered into four recovery domains and meets two functions. In addition to measuring self-reported outcomes, it increases service-user control towards objectives and recovery action plans (Honey et al. BMC Psychiatry 2023;23 500). Objectives: To evalue the efficacy of RAD-DS in a psychiatric rehabilitation facility to be used as a routine tool in daily rehabilitation activity. Methods: In our observational study, we recruited 103 inpatients (total: 103 patients, females: 38 patients, males: 65 patients) in a psychiatric rehabilitation facility. The patient presented with psychiatric disorders that met the diagnostic criteria of DMS-5 (schizophrenia, bipolar disorder, MDD, personality borderline disorder). Epidemiological data are shown in Table 1. All patients were undergoing a psychiatric rehabilitation program and were observed during a one-year evaluation. In all patients, the following rating scales were administered at baseline (T0) and after a year (T1): For the evaluation of social measures, life outcomes, and functioning and recovery: - Recovery Assessment Scale – Domains and Stages RAS-DS - Specific Levels of Functioning Scale (SLOF) - Global Assessment of Functioning (GAF) For psychopathological evaluation: - Brief Psychiatric Rating Scale (BPRS) The data were statistically analyzed with the EZAnalyze 3.0 software for the Excel platform. Results: The RAS-DS total score results (Table 2) show a not significant difference between T0 vs. T1 (mean: 101.80 vs. 104.37, p. 0.193). An improvement in the score was observed after one year of rehabilitation treatment in the subgroup " Doing things I value " (T0 vs. T1: mean 16.15 vs. 18.77, p 0.001). Statistically significant differences were observed in the subgroups " Mastering my illness " (T0 vs. T1: mean 18.3 vs. 20.85, p. 0.021). Abstract: The C9ORF72 gene repeat expansion is the most frequent cause of ALS. Long repeats alleles in ATXN-1, ATXN-2, and NIPA1 genes are associated to ALS susceptibility. Tandem Repeat Polymorphisms (TRPs) are good candidates for missing hereditability in ALS (40%), although they were never systematically analyzed as they represent a remarkable challenge to NGS. The aim of this study is to perform a systematic analysis of TRPs in ALS by combining NGS and novel bioinformatics tools. We performed our analysis from whole genome sequencing data (WGS) of 70 ALS cases. TRPs were evaluated by means of a software developed within our consortium to detect tandem repeat expansion. Validation of expanded loci was conducted by Repeat primer PCR We identified an ALS patient with a CGG expansion in 5'UTR of FRA10AC1 gene. To explore the possible role of this CGG expansion in ALS we screened a cohort of 337 ALS and 285 controls and we found 3 expanded patients (0.9%) and no control. We failed to replicated this result in a second cohort of 426 ALS patients and 733 controls (1 ALS (0.2%) and 5 controls (0.7%)) Overall, we observed the CGG expansion in 4/763 (0.5%) ALS patients and 5/1018 controls (0.5%). FRA10AC1 gene expression was not silenced by the expansion. The software we developed can detect epeat expansions from WGS data, although FRA10AC1 CGG expansion seems not to be involved in ALS pathogenesis. Abstract: The Eurofit Special Test is a battery of motor fitness tests resulting from a 10-year project of the Committee of Experts for Sports Research and is comprised of strength, speed, flexibility, and balance. The purpose of this study was to investigate whether the Eurofit Special was able to distinguish variations in functioning among individuals with intellectual disabilities. Significant differences were found in long jump flexibility 25-m dash, medicine ball throw, balance walk, sit ups in 30-s. Abstract: The purpose of the investigation was to use hand-held dynamometry to compare a single trial versus maximal or mean values of muscular strength in individuals with mental retardation. Twenty-three subjects classified as moderately mentally retarded (12 male, 11 female) performed three trials on six muscle groups. The magnitudes of the first, maximal, and mean isometric force across muscle groups were compared using a repeated measures analysis of variance and the Greenhouse-Geiser adjustment for the critical level of significance. Data analysis revealed a significant main muscle group effect and measurement score effect, but no significant differences between a single trial and mean of three trials. However, there was a significant difference between the first or mean scores and maximal scores. Intraclass correlation coefficients demonstrated consistency across muscle groups. Abstract: The paper presents a cooperative, narrative and systemic approach to life/career design and guidance, here addressed to professionals of education, and namely to teachers and school managers. This approach is here illustrated through the presentation of a cooperative inquiry, that involved a network of secondary schools (first and second degree) in the geographic area North East of Milano and Province from April 2015 to September 2016. The project’s title was OSA (an acronym for Orientating oneself and each other within the Learning System), a word that also means in Italian “to dare”, since the whole proposal wanted to be an invitation for participants to innovate their practices through a deep and dialogic transformation of their commonsense representations and implicit/explicit perspectives about choice, uncertainty, adaptation, and identity building. The Italian school, in fact, tends to offer a lot of information to students about their future career, and asks them to be ready to adapt to the job market, and to social demands, but very few occasions are offered to develop self-reflexivity, critical thinking and awareness of the larger context, that are nonetheless needed in order to freely make a deliberate choice. The project’s methodology was based on sharing stories of (dis)orientation, to foster critical analysis of life/career design and guidance practices brought forth by schools and to develop awareness of the influence of the wider context. Education to life/career design and guidance is education to the art of improvising a life, a necessary competence in times of rapid change, and besides in a world that is living a major crisis. Furthermore, the emphasis on individuals, as separated from their systems of interactions, and the dominance of linear functional thinking, do not help to see the complexity of life. Abstract: LHCII, the most abundant membrane protein on earth, is the major light-harvesting complex of plants. It is generally accepted that LHCII is associated with Photosystem II and only as a short-term response to overexcitation of PSII a subset moves to Photosystem I, triggered by its phosphorylation (state1 to state2 transition). However, here we show that in most natural light conditions LHCII serves as an antenna of both Photosystem I and Photosystem II and it is quantitatively demonstrated that this is required to achieve excitation balance between the two photosystems. This allows for acclimation to different light intensities simply by regulating the expression of LHCII genes only. It is demonstrated that indeed the amount of LHCII that is bound to both photosystems decreases when growth light intensity increases and vice versa. Abstract: Photosystem I (PSI) is an essential component of photosynthetic membranes. Despite the high sequence and structural homologies, its absorption properties differ substantially in algae, plants and cyanobacteria. In particular it is characterized by the presence of low-energy chlorophylls (red forms), the number and the energy of which vary in different organisms. The PSI–LHCI (PSI–light harvesting complex I) complex of the green alga Chlamydomonas reinhardtii (C.r.) is significantly larger than that of plants, containing five additional light-harvesting complexes (together binding ≈ 65 chlorophylls), and contains red forms with higher energy than plants. To understand how these differences influence excitation energy transfer and trapping in the system, we studied two PSI–LHCI C.r. particles, differing in antenna size and red-form content, using time-resolved fluorescence and compared them to plant PSI–LHCI. The excited state kinetics in C.r. shows the same average lifetime (50 ps) as in plants suggesting that the effect of antenna enlargement is compensated by higher energy red forms. The system equilibrates very fast, indicating that all Lhcas are well-connected, despite their long distance to the core. The differences between C.r. PSI–LHCI with and without Lhca2 and Lhca9 show that these Lhcas bind red forms, although not the red-most. The red-most forms are in (or functionally close to) other Lhcas and slow down the trapping, but hardly affect the quantum efficiency, which remains as high as 97% even in a complex that contains 235 chlorophylls.

Abstract: CP24 is a minor antenna complex of Photosystem II, which is specific for land plants. It has been proposed that this complex is involved in the process of excess energy dissipation, which protects plants from photodamage in high light conditions. Here, we have investigated the functional architecture of the complex, integrating mutation analysis with time-resolved spectroscopy. A comprehensive picture is obtained about the nature, the spectroscopic properties, and the role in the quenching in solution of the pigments in the individual binding sites. The lowest energy absorption band in the chlorophyll a region corresponds to chlorophylls 611/612, and it is not the site of quenching in CP24. Chlorophylls 613 and 614, which are present in the major light-harvesting complex of Photosystem appear to be absent in CP24. In contrast to all other light-harvesting complexes, CP24 is stable when the L1 carotenoid binding site is empty and upon mutations in the third helix, whereas mutations in the first helix strongly affect the folding/stability of the pigment-protein complex. The absence of lutein in L1 site does not have any effect on the quenching, whereas substitution of violaxanthin in the L2 site with lutein or zeaxanthin results in a complex with enhanced quenched fluorescence. Triplet-minus-singlet measurements indicate that zeaxanthin and lutein in site L2 are located closer to chlorophylls than violaxanthin, thus suggesting that they can act as direct quenchers via a strong interaction with a neighboring chlorophyll. The results provide the molecular basis for the zeaxanthin-dependent quenching in isolated CP24.

50. Pigment binding sites occupancy and functional architecture of the Photosystem II antenna complex Lhcb5

Abstract: Lhcb5 is an antenna protein that is highly conserved in plants and green algae. It is part of the inner layer of photosystem II antenna system retained in high light acclimated plants. To study the structure-function relation and the role of individual pigments in this complex, we (i) "knocked out" each of the chromophores bound to multiple (nine total) chlorophyll sites and (ii) exchanged the xanthophylls bound to the three xanthophyll sites. The occupancy and associated energy of the pigment binding sites were determined. The role of the individual pigments in protein folding, stability, energy transfer, and dissipation was studied in vitro. The results indicate that lutein has a primary role in the folding and stability of the complex, whereas violaxanthin and zeaxanthin have a negative effect on folding yield and stability, respectively. The data showed a distinct function for the L1 and L2 carotenoid binding sites, the former preferentially involved in gathering the excitation energy to chlorophyll a (Chl a), whereas the latter modulates the concentration of chlorophyll singlet excited states dependent on the xanthophylls bound to it, likely via an interaction with Chl-603. Our results also underscored the role of zeaxanthin and lutein in quenching the excitation energy, whereas violaxanthin was shown to be very effective in energy transfer. The characteristics of the isolated proteins were consistent with the observed role of Lhcb5 in vivo in catalyzing fluorescence quenching upon zeaxanthin binding. © 2009 by The American Society for Biochemistry and Molecular Biology, Inc.
