Background: Reliable Human Papillomavirus (HPV) testing and genotyping are essential for quality assurance in HPV-based primary screening, disease management and for monitoring the impact of HPV vaccination. The clinical validation of HPV molecular diagnostic assays has significantly contributed to these objectives; however, little emphasis has been placed on assuring sample quality. This study aimed to evaluate the accuracy of sample cellularity assessment using the C-C Motif Chemokine Receptor 5 (CCR5) gene target as a marker of sample adequacy in molecular diagnostics. Methods: Jurkat cell line samples were counted using both a Thoma cell-counting chamber and Fluorescence-Activated Cell Sorting (FACS). Jurkat cell line samples at three different concentrations were subsequently evaluated using the OncoPredict HPV Quality Control (QC) real-time PCR assay, employing CCR5 for molecular cellularity quantification. Results: The cellularity values obtained were comparable across the three different methods for all dilutions of the cell line tested. Conclusions: The results obtained from this study show that CCR5 represents a promising molecular marker for the accurate quantification of sample cellularity, confirming its use as a reliable sample adequacy control, thus reducing the risk of “false-negative” results.

Impairments in the function of the hypothalamic-pituitary-adrenal (HPA) axis and enhanced glucocorticoid receptor (GR) activity in the central amygdala (CeA) are critical mechanisms in the pathogenesis of alcohol use disorder (AUD). The GR antagonist mifepristone attenuates craving in AUD patients, alcohol consumption in AUD models, and decreases CeA γ-aminobutyric acid (GABA) transmission in alcohol-dependent rats. Previous studies suggest elevated GR activity in the CeA of male alcohol-preferring Marchigian-Sardinian (msP) rats, but its contribution to heightened CeA GABA transmission driving their characteristic post-dependent phenotype is largely unknown.We determined Nr3c1 (the gene encoding GR) gene transcription in the CeA in male and female msP and Wistar rats using in situ hybridization and studied acute effects of mifepristone (10 μM) and its interaction with ethanol (44 mM) on pharmacologically isolated spontaneous inhibitory postsynaptic currents (sIPSCs) and electrically evoked inhibitory postsynaptic potentials (eIPSPs) in the CeA using ex vivo slice electrophysiology.Female rats of both genotypes expressed more CeA GRs than males, suggesting a sexually dimorphic GR regulation of CeA activity. Mifepristone reduced sIPSC frequencies (GABA release) and eIPSP amplitudes in msP rats of both sexes, but not in their Wistar counterparts; however, it did not prevent acute ethanol-induced increase in CeA GABA transmission in male rats.In msP rats, GR regulates CeA GABAergic signaling under basal conditions, indicative of intrinsically active GR. Thus, enhanced GR function in the CeA represents a key mechanism contributing to maladaptive behaviors associated with AUD.

Resumo A Educação Interprofissional em Saúde (EIP) constitui-se como uma estratégia para formação de profissionais capazes de trabalhar em equipe por meio de competências colaborativas e da aprendizagem baseada em interações. O Projeto Pedagógico da Universidade Federal de São Paulo (Unifesp) baseia-se nos princípios da EIP para a formação de profissionais, dentre eles, terapeutas ocupacionais. Objetivou-se compreender a influência da formação interprofissional na trajetória profissional de terapeutas ocupacionais egressas da Unifesp. Pela abordagem qualitativa, produziram-se informações por aplicação de formulário on-line semiestruturado, analisado posteriormente com base na Análise Temática. Os resultados revelaram que a formação interprofissional possibilita segurança às profissionais nas práticas colaborativas, no fomento ao trabalho em equipe e no desenvolvimento de ações que visam à integralidade do cuidado. Notou-se, porém, dificuldades no reconhecimento da identidade profissional da Terapia Ocupacional e na ação conjunta com profissionais formados em perspectiva disciplinar.

Abstract The hydrological regime leads to fluctuations in the availability of food resources for fish, which may reflect in their diet variation. Our main goal was to analyze the relationship between water-level fluctuations and the diet and the body condition of Trachelyopterus galeatus in the Upper Paraná River floodplain. The fish used in the study were sampled quarterly along nine years, at nine sampling stations. Our results showed that diet of T. galeatus varied in response to oscillations in the hydrometric level, with the consumption of terrestrial invertebrates being positively related to hydrometric level, while the consumption of aquatic invertebrates was negatively related. The trophic niche breadth and feeding activity were not affected by hydrometric level, but the fish body condition was positively affected. The high food plasticity allows T. galeatus to consume the most available resources (e.g., terrestrial invertebrates in high water levels), which probably contributed to the increase in its body condition in higher hydrometric levels. In summary, the hydrological dynamic is fundamental for the diet variation of omnivorous fish, which can take advantage of the resource availability according to the hydrometric level, increasing its body condition when allochthonous resources as terrestrial invertebrates are supposed to be more available.

12. High glucose induces an activated state of partial epithelial-mesenchymal transition in human primary tubular cell cultures.

Tubulointerstitial fibrosis is observed in diabetic nephropathy. It is still debated whether tubular cells, undergoing epithelial-mesenchymal transition (EMT) in high glucose (HG) conditions, may contribute to interstitial fibrosis development. In this study, we investigated the phenotypic and molecular EMT-like changes and the alteration of inflammatory and fibrogenic secretome induced by HG in human primary tubular cell cultures. Taking advantage of this in vitro cell model composed of proximal and distal tubular cells, we showed that HG-treated tubular cells acquired a fibroblast-like morphology with increased cytoplasmic stress fibers, maintaining the expression of the epithelial markers specific of proximal and distal tubular cells. HG increased Snail1, miRNA210 and Vimentin mesenchymal markers, decreased N-cadherin expression and migration ability of primary tubular cells, while E-cadherin expression and focal adhesion distribution were not affected. Furthermore, HG treatment of tubular cells altered the inflammatory cytokine secretion creating a secretome able to enhance the proliferation and migration of fibroblasts. Our findings show that HG promotes an activated state of partial EMT in human tubular primary cells and induces a pro-inflammatory and pro-fibrogenic microenvironment, supporting the active role of tubular cells in diabetic nephropathy onset.

13. Evaluation of RAS Mutational Status in Liquid Biopsy to Monitor Disease Progression in Metastatic Colorectal Cancer Patients

In this study we evaluated both~ K- and N-RAS mutations in plasma samples from patients with metastatic colorectal cancer by means of the BEAMing technology, and we assessed their diagnostic performance compared to RAS analyses performed on tissue. The sensitivity of BEAMing in identifying KRAS mutations was of 89.5%, with a fair specificity. The agreement with tissue analysis was moderate. The sensitivity for NRAS was high with a good specificity, and the agreement between tissue analysis and BEAMing was fair. Interestingly, significantly higher mutant allele fraction (MAF) levels were detected in patients with G2 tumors, liver metastases, and in those who did not receive surgery. NRAS MAF level was significantly higher in patients with mucinous adenocarcinoma and for those with lung metastases. A sharp increase in the MAF values was observed in patients who moved towards disease progression. More strikingly, molecular progression always anticipated the radiological one in these patients. These observations pave the way to the possibility of using liquid biopsy to monitor patients during treatment, and to enable oncologists to anticipate interventions compared to radiological analyses. This will allow time to be saved and ensure a better management of metastatic patients in the near future.

Abstract: The size of soybean seeds needs further study because its relationships with physiological quality, field establishment, and yield are controversial. The aim of this study was to evaluate the effects of seed size on soybean establishment and seed physiological quality. In the field, a 2x3x3 factorial design was used, with two cultivars (M 5947 IPRO and 59HO124 IPRO), three seed sizes (5.5, 6.0, and 6.5 mm), and three sowing densities (280,000, 320,000, and 360,000 plants.ha-1). Agronomic traits and grain yield were evaluated. After harvest, only seeds from plots with a population of 320,000 plants were classified by size using circular sieves with 5.5-mm, 6.0-mm, 6.5-mm, and 7.0-mm mesh sizes. For the evaluation of physiological quality, a 2x4 factorial design was applied, with two cultivars and four seed sizes. Germination, seedling emergence, accelerated aging, seedling dry matter, and emergence speed index were evaluated. Smaller seeds gave rise to shorter plants with a lower first pod insertion height. Larger seeds had higher physiological quality. The M 5947 IPRO cultivar showed better seed performance and quality.

16. Cultural Heritage and Religious Phenomenon between Urbicide and Cancel Culture: The Other Side of the Russian–Ukrainian Conflict

The Russian–Ukrainian conflict, in addition to causing an unacceptable loss of human life, is straining the integrity of Ukraine’s cultural heritage, despite the fact that both countries involved are parties to the 1954 Hague Convention for the Protection of Cultural Property in the Event of Armed Conflict and its First Protocol. Churches are one of Ukraine’s most important historical assets, as well as symbolic places of Orthodox religious identity common to both the invaders and the invaded. The destruction of these places and their deliberate damage on the part of both sides appear to be part of a more general conflict concerning internal disagreements between Russian and Ukrainian Orthodoxy, which, in turn, reflect two different historical views of the Russian–Ukrainian relationship. A brief reconstruction of relations between the Orthodox Churches operating on the territory of Ukraine demonstrates how religious affiliation has affected the conflict, causing it to become decisive and deeply divisive, so much so that the Patriarchate of Moscow has become an active part of the conflict. This circumstance favours the hypothesis that it is precisely the religious cultural heritage that is most at risk of deliberate destruction. The Russians, by destroying the symbolic places of Ukrainian religious identity (urbicide), affirm the spiritual unity of the Russian and Ukrainian peoples. For their part, the Ukrainians attempt to erase the Russian presence and the common religious cultural roots by destroying buildings of worship dear to the tradition of the Moscow Patriarchate (cancel culture). They reject the imperial traditions of Russia and, at the same time, claim an independent Church. The question arises as to whether the reconstruction process following the war will take into account the original cultural–religious identities, or whether it will take the opportunity to adopt a new (also) religious identity instead, and whether the old and new instruments offered by law are adequate.

El artículo aborda el desarrollo teórico-práctico de los conceptos territorio y comunidad en la Terapia Ocupacional colombiana. El objetivo fue comprender el uso y la elaboración de tales conceptos e identificar cómo se expresan en la práctica. La recolección de datos se realizó en dos etapas: primera, revisión conceptual de la literatura publicada entre 1990 y 2020 en revistas y libros de amplia circulación en América Latina, así como entrevistas a dos autoras recurrentes en las publicaciones seleccionadas; segunda, observación participante de una pasantía profesional orientada a estudiantes de pregrado en Terapia Ocupacional de una universidad colombiana y entrevistas a actores sociales involucrados en esa práctica. Se identifica que los términos territorio y comunidad conforman el vocabulario técnico de las y los terapeutas ocupacionales en Colombia; se refieren históricamente a prácticas realizadas en contextos de pobreza, y adquirieron mayor relevancia con el desarrollo de políticas públicas. Se evidencia, además, una actuación territorial-comunitaria relacionada con el modo de vida comunitario y con la apropiación de los recursos materiales, sociales, culturales y relacionales del territorio, desde una perspectiva colectiva de la vida. Se reitera la necesidad de superar el uso de estos conceptos solo como espacios físicos de actuación profesional.

19. Relationship of Freshwater Fish Recruitment With Distinct Reproductive Strategies and Flood Attributes: A Long-Term View in the Upper Paraná River Floodplain

The flood pulse is the main driving force for communities' structure and functioning in river-floodplain systems. High synchrony exists between the hydrological cycle and reproductive cycle events for several fish species. However, species with different reproductive strategies can respond in different ways to the flood regime. Thus, this study intends to evaluate the relationship between the recruitment of different reproductive guilds of freshwater fish and flood attributes (flood duration, maximum annual water level, and delay of flood) from a time series of 20 years in the Upper Paraná River floodplain, Brazil. The abundance of four guilds was evaluated: (i) long-distance migratory with external fertilization and without parental care (LMEF); (ii) non-migratory or short-distance migratory with external fertilization and without parental care (NEFW); (iii) non-migratory or short-distance migratory with external fertilization and parental care (NEFP); and (iv) non-migratory or short-distance migratory with internal fertilization and without parental care (NIF). Multiple regression analyses were applied between flood attributes and abundance of young-of-the-year or juveniles for each reproductive guild. This study observed a consistent pattern of long-lasting flooding positively influencing the recruitment of all reproductive guilds, while water level intensity and the time of the onset of flooding also influenced some non-migratory strategies. We can conclude that the conservation of fish populations and the maintenance of ecosystem functions and services associated with them need to be considered in the operating protocols of upstream hydroelectric plants, since they are dependent on the flooding controlled by them.

20. Use of inhaled devices during a hospital exacerbation of COPD: a summary of an interdisciplinary audit held at ICS Maugeri Pavia, Italy (March-June 2019)

To date treatment protocols in Respiratory and or Internal departments across Italy for treatment of chronic obstructive pulmonary disease (COPD) patients at hospital admission with relapse due to exacerbation do not find adequate support in current guidelines. Here we describe the results of a recent clinical audit, including a systematic review of practices reported in literature and an open discussion comparing these to current real-life procedures. The process was dived into two 8-hour-audits 3 months apart in order to allow work on the field in between meeting and involved 13 participants (3 nurses, 1 physiotherapist, 2 internists and 7 pulmonologists). This document reports the opinions of the experts and their consensus, leading to a bundle of multidisciplinary statements on the use of inhaled drugs for hospitalized COPD patients. Recommendations and topics addressed include: i) monitoring and diagnosis during the first 24 h after admission; ii) treatment algorithm and options (i.e., short and long acting bronchodilators); iii) bronchodilator dosages when switching device or using spacer; iv) flow measurement systems for shifting to LABA+LAMA within 48 h; v) when nebulizers are recommended; vi) use of SMI to deliver LABA+LAMA when patient needs SABA 30 litres/min; viii) contraindication to use DPI; ix) continuation of LABA-LAMA when patient is already on therapy; x) possible LABA-LAMA dosage increase; xi) use of SABA and/or SAMA in addition to LABA+LABA; xii) use of SABA+SAMA restricted to real need; xiii) reconciliation of drugs in presence of comorbidities; xiv) check of knowledge and skills on inhalation therapy; xv) discharge bundle; xvi) use of MDI and SMI in tracheostomized patients in spontaneous and ventilated breathing.

A equipe do Grupo de Pesquisa Terapia Ocupacional: Memórias, Histórias e Fundamentos (CNPq, 2015) vem trabalhando para organizar e sistematizar materiais e documentos que integrem a história da terapia ocupacional brasileira, ampliando fontes primárias e secundárias nesse âmbito, a fim de apreender, dentre outros objetivos, o processo de institucionalização acadêmica da terapia ocupacional no Brasil. Entre 2015 e 2016, uma parte dessas atividades se voltou para os percursos em torno do evento Encontro Nacional de Docentes de Terapia Ocupacional – ENDTO, que, em linhas gerais, vem promovendo a discussão coletiva do ensino de terapia ocupacional no país e, portanto, reverbera parte da história da referida institucionalização. Em 2016, por ocasião do preparo de uma conferência a ser realizada no XV ENDTO, as autoras se debruçaram sobre os documentos, materiais de divulgação, fotos, textos publicados reunidos e, também, apoiando-se na narrativa de uma história da qual participaram de diversas maneiras e em momentos diferentes, perseguiram o objetivo de contar e registar a trajetória dos 30 anos desde a realização do I ENDTO, em 1986. Neste texto, expõe-se uma narrativa dessa trajetória e o registro imagético de parte das fontes reunidas. Trata-se de um patrimônio coletivo da área de terapia ocupacional no Brasil a ser documentado e partilhado com os que fizeram e com os que continuam fazendo essa história. AbstractThe team of the research group “Occupational Therapy: Memories, Histories and Foundations” (CNPq, 2015) has been working to organize and systematize materials and documents that integrate the Brazilian occupational therapy story, expanding primary and secondary sources. Among other objectives the aim is to comprehend the occupational therapy academic institutionalization in Brazil. Part of the activities, between 2015 and 2016, has been directed to the event “The National Meeting of Occupational Therapy Lecturers”, which, in general lines, promotes a collective discussion about teaching occupational therapy in the country and due to that echoes part of the referred history of its institutionalization. In 2016, preparing a conference held at the XV National Meeting of Occupational Therapy Lecturers, the authors looked over a collection of documents, diffusion material, photos and published texts. They are based on the narrative of a history which they have been witnesses in many different ways and moments. The objective was to present and register the trajectory of the thirty years of the Lecturers National Meetings, since the first edition in 1986. In this paper the narrative of this trajectory and registered images from some sources are exposed. That is a collective treasury of occupational therapy in Brazil which needed to be documented and shared with those who participated and those who still continue making this history.Keywords: Brazil; Higher education; History; Memories; Occupational therapy.

Abstract The objectives of this study were to examine the implication of the genotype × environment interaction (G × E) in the identification of genetically superior soybean [Glycine max (L.) Merr.] progenies; to obtain estimates of genetic and phenotypic parameters for agronomic traits in soybean progenies; and to select genetically superior progenies to obtain lineages. Progenies F3:4 were evaluated in the municipalities of Lavras‐MG and Itutinga‐MG, Brazil, in the agricultural year 2016–2017. Progenies F3:5 were evaluated in the summer of 2017–2018 in the municipalities of Lavras‐MG, Itutinga‐MG, and Ijaci‐MG. The traits of days to flowering, days to full maturity, plant height, bottom pod height, lodging, and grain yield were evaluated. The genetic and phenotypic parameters, expected gain from selection, genetic correlation, correlated response, achieved heritability, and frequency distribution of the adjusted means were estimated. The estimates of the components of variance showed the existence of variability among the progenies, enabling the selection of superior genotypes. There was an effect of the G × E interaction for all traits, and most of the interaction was due to complex interaction. From the analysis of genotypic correlation, significant estimates were observed between the traits. The G × E interaction affected the estimates of genetic and phenotypic parameters in the soybean, and the achieved heritability is a tool to study this interaction.

The fibrotic tissue and the stroma adjacent to cancer cells are characterised by the presence of activated fibroblasts (myofibroblasts) which play a role in creating a supportive tissue characterised by abundant extracellular matrix (ECM) secretion. The myofibroblasts remodel this tissue through secreted molecules and modulation of their cytoskeleton and specialized contractile structures. The non-receptor protein tyrosine kinase Arg (also called Abl2) has the unique ability to bind directly to the actin cytoskeleton, transducing diverse extracellular signals into cytoskeletal rearrangements. In this study we analysed the 1ALCTL and 1BLCTL Arg isoforms in Arg−/− murine embryonal fibroblasts (MEF) cell line, focusing on their capacity to activate fibroblasts and to remodel ECM. The results obtained showed that Arg isoform 1BLCTL has a major role in proliferation, migration/invasion of MEF and in inducing a milieu able to modulate tumour cell morphology, while 1ALCTL isoform has a role in MEF adhesion maintaining active focal adhesions. On the whole, the presence of Arg in MEF supports the proliferation, activation, adhesion, ECM contraction and stiffness, while the absence of Arg affected these myofibroblast features. Introducción: El contexto de desigualdad social en consonancia con las políticas sociales y las discusiones acerca del acceso a la ciudadanía propuso para la Terapia Ocupacional la entrada en una nueva área: lo social. La terapia ocupacional social brasileña proporciona un recorte metodológico específico con acciones para públicos que tengan la vulnerabilidad social como eje de su demanda. Utilizándose de los presupuestos de la terapia ocupacional social brasileña, se he investigado la producción de la Revista Chilena de Terapia Ocupacional, con vistas a identificar la presencia de textos acerca de la área social. Método: Se realizó una encuesta en dos etapas: colección de todos los textos publicados y selección y análisis de los artículos que hacían referencia directa o indirectamente con el ámbito social. Resultados y discusión: Fueron publicados 182 artículos desde 2001 hasta 2014, y se describen sus áreas, sub-áreas y países de los autores. En la segunda fase se seleccionaron 19 textos relacionados en la área social. La clasificación de los trabajos fue realizada de la siguiente manera: discusiones en un enfoque en el nivel individual (07 textos) y en una perspectiva colectiva (12 textos). El debate es el desafío en avanzar de una perspectiva individual al desarrollo de una práctica colectiva y social. Conclusión: La Terapia Ocupacional en Latinoamérica debe desarrollar metodologías y aportaciones teóricas que son singulares a nuestra realidad y dirigir la búsqueda de respuestas a las demandas de la sociedad. Proponemos vínculos más estrechos entre los países de la región para construir una perspectiva en terapia ocupacional socialmente comprometido con nuestro contexto.

This paper presents the challenges of production and dissemination of knowledge in Occupational Therapy in Latin America, its place in the global scenario and the weaving of a political-epistemic network of communication, discussion and exchange. A sense of groupwork, processes and intervention as a collective network is proposed to analyse and understand the social relational practices that make up the political-epistemic network between key-subjects, different types of knowledge, as well as different ways of producing and disseminating them for the institutionality of the field of Occupational Therapy itself and for its outreach in society in general. To this end, we describe and analyse the actions of the Amaru Network as a collective strategy to face the challenges of the production and dissemination of hegemonic and excluding knowledge in Occupational Therapy in the Latin American context. We take as the axis of analysis the idea of collective as a conceptual category that serves to designate and guide the counter-hegemonic processes of knowledge production in the scientific and cosmological reality of the Latin American region. The Amaru Network, in proposing and sustaining a social practice engendered in a collaborative network between Latin American countries, turns its gaze to the broadening of epistemological bases for other possible modes of knowledge production. Este ensaio apresenta os desafios da produção e difusão de conhecimento em Terapia Ocupacional na América Latina, seu lugar no cenário global e a tessitura de uma rede político-epistêmica de comunicação, de discussão e troca. Propõe-se um sentido de trabalho em grupo, de processos e intervenção, enquanto coletivo em rede para analisar e compreender as práticas sociais relacionais que conformam a rede político-epistêmica entre sujeitos-chaves, diferentes tipos de conhecimento, bem como distintos modos de produzi-los e difundi-los para a institucionalidade da própria área de Terapia Ocupacional e para seu alcance na sociedade em geral. Para tanto, são descritas e analisadas ações da Rede Amaru como estratégia coletiva no enfrentamento aos desafios da produção e difusão do conhecimento hegemônico e excludente em Terapia Ocupacional que se colocam ao contexto latino-americano. Tomamos como eixo de análise a ideia de coletivo enquanto uma categoria conceitual que serve para designar e orientar os processos contra-hegemônicos da produção de conhecimento na realidade científica e cosmológica da região latino-americana. A Rede Amaru, ao se propor e sustentar uma prática social engendrada em uma rede colaborativa entre países latino-americanos, volta seu olhar para o alargamento das bases epistemológicas para outros modos possíveis de produção de conhecimento

Clear cell renal cell carcinoma (ccRCC) is fundamentally a metabolic disease. Given the importance of lipids in many cellular processes, in this study we delineated a lipidomic profile of human ccRCC and integrated it with transcriptomic data to connect the variations in cancer lipid metabolism with gene expression changes. Untargeted lipidomic analysis was performed on 20 ccRCC and 20 paired normal tissues, using LC-MS and GC-MS. Different lipid classes were altered in cancer compared to normal tissue. Among the long chain fatty acids (LCFAs), significant accumulations of polyunsaturated fatty acids (PUFAs) were found. Integrated lipidomic and transcriptomic analysis showed that fatty acid desaturation and elongation pathways were enriched in neoplastic tissue. Consistent with these findings, we observed increased expression of stearoyl-CoA desaturase (SCD1) and FA elongase 2 and 5 in ccRCC. Primary renal cancer cells treated with a small molecule SCD1 inhibitor (A939572) proliferated at a slower rate than untreated cancer cells. In addition, after cisplatin treatment, the death rate of tumor cells treated with A939572 was significantly greater than that of untreated cancer cells. In conclusion, our findings delineate a ccRCC lipidomic signature and showed that SCD1 inhibition significantly reduced cancer cell proliferation and increased cisplatin sensitivity, suggesting that this pathway can be involved in ccRCC chemotherapy resistance.

The mechanism upon which human kidneys undergo regeneration is debated, though different lineage-tracing mouse models have tried to explain the cellular types and the mechanisms involved. Different sources of human renal progenitors have been proposed, but it is difficult to argue whether these populations have the same capacities that have been described in mice. Using the nephrosphere (NS) model, we isolated the quiescent population of adult human renal stem-like PKHhigh/CD133+/CD24− cells (RSC). The aim of this study was to deepen the RSC in vitro multipotency capacity. RSC, not expressing endothelial markers, generated secondary nephrospheres containing CD31+/vWf+ cells and cytokeratin positive cells, indicating the coexistence of endothelial and epithelial commitment. RSC cultured on decellularized human renal scaffolds generated endothelial structures together with the proximal and distal tubular structures. CD31+ endothelial committed progenitors sorted from nephrospheres generated spheroids with endothelial-like sprouts in Matrigel. We also demonstrated the double commitment toward endothelial and epithelial lineages of single RSC. The ability of the plastic RSC population to recapitulate the development of tubular epithelial and endothelial renal lineages makes these cells a good tool for the creation of organoids with translational relevance for studying the parenchymal and endothelial cell interactions and developing new therapeutic strategies.

Clear cell Renal Cell Carcinoma (ccRCC) is the most common and metastatic urological cancer. Molecular players of ccRCC progression and metastasis are not completely known. Here, using primary cell cultures from patients' specimens, we found that TGFβ1/Smad signalling is more activated in high versus low grade ccRCC and inversely correlates with Abl2 tyrosine kinase protein expression. TGFβ1 treatment increased ubiquitination and degradation of Abl2 protein in ccRCC cell lines by TGFβ1/Smad pathway activation and reactive oxygen species production. 3D invasion and matrix degradation assays showed that Abl2 promoted TGFβ1-induced ccRCC cell invasion and maturation of invadopodia, a hallmark of tumour invasion and metastasis. Our findings define Abl2 as a new downstream molecule of TGFβ1 signalling and putative target to counteract advanced ccRCC.

O presente capítulo de livro tem como objetivo apresentar os aspectos do abandono afetivo nas relações familiares frente ao ordenamento jurídico. A afetividade ganhou relevância na família contemporânea, destacando-se na legislação brasileira de forma principiológica. Em virtude disso, a legislação impõe, por exemplo, aos pais deveres na relação com seus filhos, como assistir, cuidar, educar e participar de seu desenvolvimento, e em caso de descumprimento, será imputada aos genitores a devida responsabilização pelo dano cometido. A discussão do tema implica na valoração do afeto no cenário jurídico, visto a crescente demanda pela reparação dos prejuízos morais. Como resultado, aponta-se que o dano causado ao descendente pode ser imensurável, todavia, a imputação da responsabilidade civil cumprirá sua função social e trará segurança jurídica na consagração do direito positivo. Nesse sentido, conclui-se que a reparação moral não será em relação ao amor que o genitor demonstra, mas sobre o descumprimento do dever legal de cuidado com a prole. Como metodologia, utilizou-se o método dedutivo, pesquisa bibliográfica, entendimento dos Tribunais, sítios da web, bem como em legislações pátrias.

38. De-intensification of basal-bolus insulin regimen after initiation of a GLP-1 RA improves glycaemic control and promotes weight loss in subjects with type 2 diabetes

To evaluate the impact of adding a glucagon-like peptide-1 receptor agonist (GLP-1 RA) in people with type 2 diabetes (T2D) in basal-bolus (BB) insulin regimen, on insulin requirement, HbA1c, weight loss up to 24 months.Data on subjects with T2D on BB who initiated a GLP-1 RA have been retrospectively collected. HbA1c, body weight, and insulin dose were recorded at baseline, 6, 12, and 24 months after initiation of GLP-1 RA therapy. A linear mixed model for repeated measures was used to evaluate the changes in HbA1c, body weight, and insulin requirement over time.We included 156 subjects (63.5% males; age 62 ± 11 years, HbA1c 70 ± 22.0 mmol/mol; 8.6 ± 4.2%). Compared to baseline, HbA1c and body weight were significantly lower at 6 months after introducing a GLP-1RA and remained stable up to 24 months (all p 0.0001 vs. baseline). At 24 months, 81% of subjects discontinued prandial insulin, while 38.6% discontinued basal insulin as well. Insulin requirement at baseline (aOR 0.144; 95% CI, 0.046-0.456; P = 0.001) was the only significant predictor of prandial insulin discontinuation.Replacing prandial insulin with GLP-1 RA is a valuable strategy to simplify the BB insulin regimen while improving glycaemic control and promoting weight loss in subjects with T2D.

41. Metabolic regulation of GLP-1 and PC1/3 in pancreatic α-cell line.

An intra-islet incretin system has been recently suggested to operate through modulation of the expression and activity of proconvertase 1/3 and 2 (PC1/3, PC2) in pancreatic alpha-cell accounting for local release of GLP-1. Little is known, whether this alpha-cell activity can be affected by the metabolic alterations occurring in type 2 diabetes, such as hyperglycemia, hyperlipidemia or hyperglucagonemia.AlphaTC1/6 cells from a mice pancreatic cell line were incubated in the presence of two glucose (G) concentration (5.5 and 16.7 mM) for 16 h with or without free fatty acid, IL6 or glucagon. GLP-1 secretion was measured by ELISA and expression of PC1/3 and PC2 by RT-PCR and western blot; cell viability was determined by MTT method, Reactive Oxygen Species generation (ROS) by H2DCFDA fluorescence and apoptosis by Annexin staining and Propidium Iodine (PI) fluorescence.Upon 16.7G incubation, GLP-1 secretion (total and active) was significantly increased in parallel with a significant increment in PC1/3 expression, a slight increase in cell viability and ROS generation and by a decrement in PC2 expression with no change in cell apoptosis. When cells were incubated at 5.5mM glucose with FFA, also an increment in GLP-1 secretion and PC1/3 expression was observed together an increment in ROS generation, a decrement in cell viability, and a modest increment in apoptosis. When incubated with 16.7mM glucose with FFA, the increment in GLP-1 secretion was reduced to basal, accompanied by an increment in apoptosis and ROS generation. This was also observed with IL-6, but in this case, no modification in ROS generation or apoptosis was observed when compared to 16.7mM glucose. The presence of glucagon did not modify any of the parameters studied.These data suggest that under hyperglycemic, hyperlipidemia or inflammatory conditions, alpha cells can increase expression PC1/3 and activate GLP-1 secretion, which may contribute protecting both alpha and beta-cells from glucose and lipotoxicity, while this effect seems to be lost in the presence of both pathophysiological conditions.

While clinical and pre-clinical evidence suggests that adolescence is a risk period for the development of addiction, the underlying neural mechanisms are largely unknown. Stress during adolescence has a huge influence on drug addiction. However, little is known about the mechanisms related to the interaction among stress, adolescence and addiction. Studies point to ΔFosB as a possible target for this phenomenon. In the present study, adolescent and adult rats (postnatal day 28 and 60, respectively) were restrained for 2 hours once a day for 7 days. Three days after their last exposure to stress, the animals were challenged with saline or amphetamine (1.0 mg/kg i.p.) and amphetamine-induced locomotion was recorded. Immediately after the behavioral tests, rats were decapitated and the nucleus accumbens was dissected to measure ΔFosB protein levels. We found that repeated restraint stress increased amphetamine-induced locomotion in both adult and adolescent rats. Furthermore, in adult rats, stress-induced locomotor sensitization was associated with increased expression of ΔFosB in the nucleus accumbens. Our data suggest that ΔFosB may be involved in some of the neuronal plasticity changes associated with stress induced-cross sensitization with amphetamine in adult rats.

Since it is unknown whether glucose variability (GV) is increased and whether this is related to worsening of insulin secretion and action in prediabetes, we have assessed insulin secretion and sensitivity, and daily GV in early stages of dysglycemia.Twenty subjects with normal glucose tolerance (NGT; age 45.0 ± 9.5 years; BMI 31.1 ± 6.4 kg/m2), 25 with NGT and 1hrOGTT8.6 mmol/L (1hrOGTT; 45.7 ± 8.5 years; 32.4 ± 7.0 kg/m2), and 59 with isolated impaired glucose tolerance (iIGT; 47.7 ± 11.2 years; 31.3 ± 6.1 kg/m2) underwent OGTT and MMTT. CGM was performed with blinded FreeStyle Libre Pro for 24 h under standard conditions. Parameters of beta-cell function, insulin sensitivity and GV were calculated.Overall insulin secretion and action as well as GV progressively worsened across glucose tolerance categories. On a matrix analysis, GV parameters were inversely related to ISSI-2; r = -0.37 to -0.52; p 0.0001; and IGI; r = -0.28 to -0.48; p 0.0001 for CV, SD, J-index, LI, HBGI and MAGE. Insulin secretion (IGI) and b-cell function (ISSI-2) emerged as independent contributors to GV in early stage of dysglycemia accounting for about 16%-38% of its variability.Our results show that daily GV worsens already with mild impairment of glucose tolerance. The increase in GV is inversely related to insulin secretion and action.

Aims As recommended by the Institute of Medicine (IOM), health practitioners should encourage a healthy nutrition and adequate weight gain during pregnancy in order to ensure favorable pregnancy and fetal outcomes, and to prevent diseases later in life for both mother and child. The purpose of this online survey was to determine the knowledge, attitude, and practice of the 2009 IOM recommendations among healthcare professionals managing nutritional therapy in pregnancies complicated by diabetes in Italy. Methods A cross-sectional survey was conducted by using an online self-administered questionnaire undertaken between October and December 2021. Results Of the 220 participants 89% were diabetologists/endocrinologists/internal medicine specialists and 11% dietitians/nutritionists. The survey found that the 53% of respondents provide a personalized diet to pregnant women with diabetes, while 32% a standard diet plan and only 15% healthy dietary advice. The 69% of the participants investigated for appropriate gestational weight gain, mainly based on pre-pregnancy BMI (96%), gestational weight gain (GWG) at first prenatal visit (80%) and presence of twin pregnancy (58%). Maternal weight gain was evaluated at each regularly scheduled prenatal visit and compared with IOM recommendations for the 87% of healthcare professionals. Diet plan was periodically re-evaluated and/or modified (90% of participants), based on inadequate maternal weight gain and/or fetal growth abnormalities (78%), trimester transition (53%), changes in physical activity and/or a “feel hungry” (50%). Conclusions This survey reported the knowledge and attitude of IOM guidelines and the nutritional knowledge and practice of Italian professionals on the nutritional management of diabetes in pregnancy. The application of these recommendations seemed more feasible in clinics/team dedicated to "Diabetes in Pregnancy".

O estudo teve como objetivo comparar três modalidades de crioterapia em mulheres saudáveis e não grávidas. Trata-se de um ensaio clínico randomizado, não controlado, com 32 alunas do curso de graduação de uma faculdade de enfermagem particular da cidade de São Paulo, divididas em três grupos (gelo água, gelo mole, gelo gel). Foram verificadas as temperaturas (axilar, coxa e das três bolsas de gelo) entre zero e vinte minutos. As temperaturas das bolsas foram: gelo mole de 9°C negativos a 2°C, gelo água de 0°C a 8°C e gelo gel de 11°C negativos a 2°C. Houve diferença significativa entre as médias das temperaturas da coxa com 10 minutos (p=0,007), 15 minutos (p=0,003) e 20 minutos (p=0,005). O gel foi mais eficiente no resfriamento comparado aos outros dois métodos. As três modalidades de crioterapia atingem a temperatura recomendada para analgesia e podem ser aplicadas em puérperas com dor perineal após o parto normal.El estudio tuvo como objetivo comparar tres modalidades de crioterapia en mujeres saludables y no grávidas. Las temperaturas de las bolsas fueron: hielo blando, de -9°C a 2°C; agua helada, de 0°C a 8°C; gel helado, de -11°C a 2°C. Hubo diferencia significativa entre las medias de las temperaturas del muslo tomadas a los 10 minutos (p=0,007), 15 minutos (p=0,003) y 20 minutos (p=0,005). Las tres modalidades de crioterapia alcanzan la temperatura recomendada para la analgesia y pueden ser aplicadas en mujeres con dolor perineal posparto.The objective of the present study was to compare three methods of cryotherapy in healthy non-pregnant women. This is a randomized controlled clinical trial that was conducted by 32 undergraduates of a private nursing college in the city of Sao Paulo, divided into three groups (iced water, soft ice, ice gel). The temperatures were verified (axillary, thigh, of the three ice packs) between zero and twenty minutes. The temperatures of the packs were the following: soft ice, from negative 9°C to 2°C; iced water, from 0°C to 8°C; and ice gel from negative 11°C to 2°C. There was a significant difference between the average thigh temperature values at 10 minutes (p=0.007), 15 minutes (p=0.003) and 20 minutes (p=0.005). The gel was the most efficient cooling method. The three cryotherapy methods achieved the recommended temperature for analgesia and may be tested in women with perineal pain after childbirth.

The discovery of insulin in 1921 – due to the efforts of the Canadian research team based in Toronto – has been a landmark achievement in the history of medicine. Lives of people with diabetes were changed forever, considering that in the pre-insulin era this was a deadly condition. Insulin, right after its discovery, became the first hormone to be purified for human use, the first to be unraveled in its amino acid sequence and to be synthetized by DNA-recombinant technique, the first to be modified in its amino acid sequence to modify its duration of action. As such the discovery of insulin represents a pivotal point in medical history. Since the early days of its production, insulin has been improved in its pharmacokinetic and pharmacodynamic properties in the attempt to faithfully reproduce diurnal physiologic plasma insulin fluctuations. The evolution of insulin molecule has been paralleled by evolution in the way the hormone is administered. Once-weekly insulins will be available soon, and glucose-responsive “smart” insulins start showing their potential in early clinical studies. The first century of insulin as therapy was marked by relentless search for better formulations, a search that has not stopped yet. New technologies may have, indeed, the potential to provide further improvement of safety and efficacy of insulin therapy and, therefore, contribute to improvement of the quality of life of people with diabetes.

Os processos educativos têm exigido e possibilitado delinear e vivenciar experiências de formação e cuidado em resposta aos efeitos da atual pandemia. Em consonância com essa afirmação, o presente trabalho tem por objetivo relatar uma experiência formativa com estudantes do curso de Terapia Ocupacional de uma universidade pública durante a pandemia de Covid-19. A experiência foi analisada em dois momentos, nomeados no relato como “Respiros”. No primeiro momento, em ambiente virtual, foram realizadas rodas de conversa-mosaico sem a presença obrigatória das estudantes; no segundo momento, como um módulo opcional no currículo, foram introduzidas práticas artísticas e corporais como dispositivos para o cuidado, expressão e conexão entre os corpos. As estudantes indicaram a importância de se criar um ambiente confiável e de sustentarmos uma proposta coletiva, dando ensejo à construção de corpos sensíveis, inventivos e críticos, fundamentais no exercício profissional.
